Abstract
本发明提供了一种双靶向性金属离子网络水凝胶及其制备方法与应用,属于生物医药技术领域。本发明的双靶向性金属离子网络水凝胶的制备方法包括以下步骤:(1)以金属有机骨架ZIF‑8与LHERHLNNN肽混合,制备得到负载LHERHLNNN肽的ZIF‑8;(2)以镁基金属有机骨架Mg‑MOF与WYRGRL肽混合,制备得到负载WYRGRL肽的Mg‑MOF;(3)将GelMA水凝胶溶液与步骤(1)中负载LHERHLNNN肽的ZIF‑8和步骤(2)中负载WYRGRL肽的Mg‑MOF混合,经光交联固化,即得所述双靶向性金属离子网络水凝胶。本发明的双靶向金属离子网络水凝胶支架具有促进受损肌腱‑骨界面多组织同步再生的潜力,为其在梯度结构综合重建中的应用提供了良好的前景。

技术领域
本发明属于生物医药技术领域,具体涉及一种双靶向性金属离子网络水凝胶及其制备方法与应用。
背景技术
肌腱-骨界面具有复杂的梯度结构,对于运动过程中的应力传递和压力缓冲至关重要。然而,肌腱和软骨组织的损伤往往阻碍了梯度结构的完全恢复,因此对于此类组织损伤的修复而言,往往需要很长的时间,且疗效较差。
肩关节是人体最灵活的关节,但稳定性相对较差。在运动损伤后,肩关节的功能可能会减弱甚至丧失,严重影响患者的生活质量。肩关节内天然的肌腱-骨止点由骨、软骨和肌腱组成,逐渐过渡并紧密相互连接。肩关节各种组织的不同结构表现出多样的机械性能。值得注意的是,纤维软骨在促进从坚硬的骨组织到柔软的肌腱组织的力传递方面起着关键作用。在自然条件下,纤维软骨在再生方面面临挑战,导致梯度结构被瘢痕组织取代。这种替代导致结构的拉伸强度和变形能力显著降低,从而增加再次撕裂的风险。因此,探索促进纤维软骨再生的方法,促进梯度结构内不同组织的同步重建,特别是关于肌腱-骨止点的愈合,具有至关重要的意义。
研究表明,人体内各种类型的微量金属离子在组织修复和生物代谢中起着至关重要的作用。铜离子诱导胶原成熟并增强VEGF促进新血管生成,从而促进骨形成。
锌离子增强肌腱细胞对一型胶原蛋白的分泌,可以减少肌腱瘢痕愈合。此外,锌离子具有抗菌作用,降低手术部位感染的风险。金属离子转运蛋白SLC39A10介导巨噬细胞对锌离子的摄取,显示其在炎症和免疫调节中的重要作用。镁离子招募间充质干细胞,增加它们向软骨细胞的分化,并促进纤维软骨再生。金属离子作为活性因子的潜在应用已进行过研究。
金属有机骨架材料(MOFs)是一种药物传递系统,通过金属离子和有机配体的协同作用形成特定的框架结构,既能保持金属离子的活性,又能保持有机配体的灵活性。MOF材料中金属中心的选择涵盖了所有金属元素,产生多样的材料并扩大了MOFs的应用范围。MOF材料已用于药物递送,然而,如何有效地将所需的金属-有机框架有效地传递到不同的组织中并高效发挥疗效,仍然面临着诸多困难。
靶向肽是一类具有特异性和低毒性的肽,具有与药物结合以进行靶向诊断和特定部位药物传递的独特能力,在治疗各种疾病方面具有巨大的潜力。最近,由陈教授领导的研究小组利用胶原混合肽(CHP)对聚乳酸-聚乙二醇酸(PLGA)纳米颗粒进行了靶向修饰。他们的研究表明,这些肽可以有效地靶向病理性胶原,通过抑制mTOR信号通路减少肌腱异位骨化,从而促进损伤后的肌腱愈合。软骨富含II型胶原。通过利用靶向肽WYRGRL对II型胶原的特异结合性,将该靶向肽与药物结合可实现对软骨的靶向传递。另一方面,肌腱富含I型胶原。崔宏刚团队设计了一种肽两性分子,其中包含一种I型胶原结合肽LHERHLNNN,有助于其与I型胶原蛋白的结合。现有研究已着眼于将靶向肽与金属有机骨架结合来实现靶向肽的递送和释放。
然而,如何选择MOF材料并在其表面上修饰不同的靶向肽,能够使得MOFs精确地传递到肌腱或软骨,现有技术中并未开展过相应研究。
另一方面,虽然靶向MOFs可以增强关节腔内金属离子的浓度,但关节腔狭窄使得纳米粒容易丢失,从而阻碍了长期治疗效果的产生。组织工程中的再生支架在肌腱-骨连接点的愈合过程中起着重要作用。然而,这些支架的制备是复杂的,材料的选择与其效果也是难以预期的,如化学合成的支架虽然有优异的机械性能和药物负载能力;但是这些支架易于在肌腱-骨连接点产生纤维化,在肩袖损伤修复中的应用仍然受到较大限制。
因此,基于肌腱-骨界面复杂的梯度结构,针对于肩袖损伤修复方面,现有的组织工程支架材料难以在体内发挥很好的效果,且容易产生纤维化。而基于靶向肽递送的金属有机骨架材料虽然能够实现体内靶向给药,但是材料的选择与药物分子的组合,以及靶向部位的组织结构,存在难以将特定的金属离子或靶向药物准确递送到不同的组织如肌腱和软骨中,无法实现梯度组织的一体化再生,因而限制了该类材料在肩袖损伤中的修复效果。上述问题亟待得到解决。
发明内容
本发明就是为了解决上述技术问题,从而提供一种双靶向性金属离子网络水凝胶及其制备方法与在促进肩袖损伤修复方面的应用。本发明的技术目的在于:提供一种能够精准靶向肌腱和软骨的水凝胶递送系统,实现肌腱和软骨的双靶向,同时能够克服肌腱-骨界面复杂的梯度结构,将不同的金属离子准确递送到不同部位,并在梯度结构中实现精准释放金属离子,通过金属离子的方向迁移形成动态梯度,从而实现梯度组织的一体化再生。
为了实现上述目的,本发明采用的技术方案如下:
本发明首先提供了一种双靶向性金属离子网络水凝胶的制备方法,包括以下步骤:
(1)以金属有机骨架ZIF-8与LHERHLNNN肽混合,制备得到负载LHERHLNNN肽的ZIF-8;
(2)以镁基金属有机骨架Mg-MOF与WYRGRL肽混合,制备得到负载WYRGRL肽的Mg-MOF;
(3)将GelMA水凝胶溶液与步骤(1)中负载LHERHLNNN肽的ZIF-8和步骤(2)中负载WYRGRL肽的Mg-MOF混合,经光交联固化,即得所述双靶向性金属离子网络水凝胶。
本发明通过上述制备方法,首次成功构建了一种具有靶向多组织能力并能很好在组织梯度结构中形成动态梯度治疗的金属离子网络水凝胶支架,本发明人发现,上述制备的水凝胶支架能够在体内促进不同金属离子形成不同方向的迁移,能够形成动态梯度,从而很好实现了梯度组织的一体化再生。其中,水凝胶包裹负载靶向肽的纳米颗粒能够将药物递送到受损部位,进一步随着水凝胶的降解,负载LHERHLNNN肽的ZIF-8选择性地迁移到肌腱,释放锌离子以增强一型胶原蛋白分泌,促进肌腱修复;与此同时,负载WYRGRL肽的Mg-MOF选择性地迁移到软骨,释放镁离子诱导细胞分化,促进软骨再生。总的来说,这种双靶向金属离子网络水凝胶支架具有促进受损肌腱-骨界面多组织同步再生的潜力,为其在具有梯度结构的一体化重建中的应用提供了有利的前景。
总的来说,本发明上述制备方法第一步先将肌腱靶向肽(LHERHLNNN)和软骨靶向肽(WYRGRL)通过静电吸附分别固定在ZIF-8和Mg-MOF上,分别合成了具有生物靶向性的两种纳米颗粒(LZIF-8和WMg-MOF),然后,通过紫外诱导的光聚合,LZIF-8和WMg-MOF被固定在甲基丙烯酰胺明胶内,最终形成了本发明的水凝胶支架(LZIF-8/WMg-MOF@GEL)。本发明制备的两种金属-有机框架通过靶向释放,促使组织内形成不断更新的金属离子动态梯度。在体外实验中,锌离子对I型胶原蛋白的分泌具有促进作用,而镁离子诱导间充质干细胞分化。将上述水凝胶梯度材料植入大鼠体内后,结果显示该水凝胶支架具有促进腱骨止点再生的潜力。可见,这种创新的双靶向金属-有机框架水凝胶支架将在梯度结构组织损伤治疗和梯度结构重建中具有重要意义。

具体的,是关于上述双靶向性金属离子网络水凝胶在制备促进肩袖损伤修复的药物方面的应用。
本发明的有益效果如下:
(1)本发明利用LHERHLNNN肽修饰的含锌金属有机骨架(LZIF-8)和WYRGRL肽修饰的含镁金属有机骨架(WMg-MOF)在明胶水凝胶支架内的光聚合作用,成功构建了具有靶向多组织能力的金属离子网络水凝胶支架,能够促进金属离子定向迁移形成动态梯度,从而实现梯度组织的综合再生。LZIF-8选择性地迁移到肌腱,释放锌离子,增强胶原蛋白分泌,促进肌腱修复。同时,WMg-MOF迁移到软骨,释放镁离子诱导细胞分化,促进软骨再生。
(2)体内磁共振以及Micro-CT成像实验清晰地证明了本发明制备的金属离子网络水凝胶支架对大鼠肩袖损伤部位的治疗作用。免疫组化实验也证明了该水凝胶支架处理后大鼠肩袖部位Coll-I和Coll-II两种特征性蛋白的显著表达。该双靶向金属离子网络水凝胶支架具有促进受损肌腱-骨界面多组织同步再生的潜力,为其在梯度结构综合重建中的应用提供了良好的前景。

具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例对本发明进行具体描述,有必要指出的是,以下实施例仅用于对本发明进行解释和说明,并不用于限定本发明。本领域技术人员根据上述发明内容所做出的一些非本质的改进和调整,仍属于本发明的保护范围。
实施例1
一、实验材料与方法
1、合成ZIF-8和LZIF-8
将150mg六水硝酸锌溶解于5mL甲醇中,330mg 2-甲基咪唑溶解于10mL甲醇中。然后,将硝酸锌溶液在涡流下分散到2-甲基咪唑溶液中。1分钟后,出现乳白色的不溶性物质,表明ZIF-8纳米颗粒形成。然后将溶液以13000rpm离心10分钟,得到ZIF-8。随后,用甲醇和H2O(每次20mL)洗涤三次,以完全去除任何剩余的未反应试剂。最后,将固体ZIF-8颗粒在真空下干燥保存制备LZIF-8,取ZIF-8100mg溶于100mL水中,加入LHERHLNNN肽1mg。磁力搅拌12小时,13000rpm离心10分钟,得到LZIF-8。再用H2O洗涤三次(每次5ml)。最后,将固体LZIF-8颗粒在真空下干燥保存。
2、Mg-MOF和WMg-MOF的合成
将2,5-二羟基对苯二甲酸(0.1mmol)、Mg(NO3)2·6H2O(0.2mmol)和聚乙烯吡罗烷酮(PVP,10mg)加入N,N-二甲基甲酰胺(DMF,6mL)和H2O(0.5mL)中形成标准溶液。在标准溶液中加入0.05mL NH3·H2O,密封于内衬聚四氟乙烯的不锈钢容器中。标准溶液在393K下加热8h,离心收集黄色沉淀,命名为Mg-MOF。所有产品用DMF和乙醇洗涤3次,然后冷冻干燥保存制备WMg-MOF,取Mg-MOF 100mg溶于100mL水中,加入WYRGRL肽1mg。将溶液磁力搅拌12小时,13000rpm离心10分钟,得到WMg-MOF。再用H2O洗涤三次(每次5ml)。最后,将固体WMg-MOF颗粒在真空下干燥保存。
3、GelMA的合成
20克明胶溶解在200毫升PBS中,在60℃下连续搅拌。随后,以每分钟0.25mL的速率将16mL的丙烯酸甲酯(MA)缓慢添加到明胶溶液中。两小时后,加入200ml PBS终止反应。然后使用透析袋(MWCO 3500)对溶液进行透析过夜,收集上清液进行进一步透析和纯化三天。随后,将溶液通过0.22μm过滤膜过滤,以实现对GelMA的额外纯化。最后,收集滤液并在真空下干燥储存。
4、LZIF-8/WMg-MOF@GEL的合成
首先,将100mg的GelMA和光交联剂10mg完全溶解在1ml的PBS中,超声下得到GelMA溶液。随后将LZIF-8和WMg-MOF加入到GelMA溶液中充分混合。然后将具有LZIF-8/WMg-MOF的均匀分散的GelMA溶液置于模具中,在紫外光下交联,形成LZIF-8/WMg-MOF@GEL。使用前,所有水凝胶用紫外线灯和酒精消毒,并用PBS溶液清洗。
二、实验结果与表征
首先,制备了ZIF-8(图1中A)和Mg-MOF(图1中C),并采用扫描电镜对其形貌进行表征。我们合成了肌腱靶向肽LHERHLNNN和软骨靶向肽WYRGRL,并采用高效液相色谱和质谱对其纯度和分子量进行表征。然后我们通过静电作用将靶向肽分别固定在ZIF-8和Mg-MOF表面,形成LZIF-8(图1中B)和WMg-MOF(图1中D),并采用扫描电镜对其形貌进行表征。
在不含任何肽的情况下,ZIF-8呈十二面体菱形(图1中A)。它们的平均尺寸约为43.56±1.84nm。同样,不含肽的Mg-MOF呈不规则球形(图1中C),平均尺寸约为316.87±12.41nm。添加多肽后,ZIF-8的形状保持不变,但粒径增加到773.61±62.89nm(图1中B)。相反,添加多肽后,Mg-MOF的形状变成了菜花状(图1中D)。
上述研究表明,肽分子渗入MOF内部孔隙后,在孔隙表面凝聚,形成较大的簇或聚集体,从而增加了MOF的尺寸。
进一步使用能谱仪检测了LZIF-8或WMg-MOF中的元素分布。结果表明,晶体中的元素分布均匀,LZIF-8含有锌元素(图1中E),WMg-MOF含有镁元素(图1中F)。进一步采用X射线衍射仪研究MOF晶体的结晶特性,结果显示LZIF-8和ZIF-8在12.7°(112)、17.8°(222)和14.6°(022)处均出现了峰值(图1中G),而WMg-MOF和Mg-MOF均在11.8°(300)处出现了峰值(图1中H),这表明肽修饰后晶体衍射峰的角度保持不变,表明肽不会破坏MOFs的晶体结构。
为了验证多肽的成功修饰,采用傅立叶变换红外光谱法对MOF表面的基团进行研究。红外光谱图显示,LZIF-8的峰值(约1576cm-1)与肽酰胺的峰值(约1543cm-1)非常相似,这证实了LZIF-8表面含有肽分子(图1中I)。Mg-MOF(~2962cm-1)峰与肽甲基(~2960cm-1)峰之间的距离很近,这证实了Mg-MOF晶体中含有肽分子(图1中I)。与肽的光谱相比,由于MOF与肽之间的相互作用,观察到了峰值的红移。
紧接着,制备了一系列水凝胶支架。在紫外线(UV)照射下,GelMA分子会发生光引发自由基聚合反应。在自然光下,水凝胶呈现出弹性、轻微浑浊和中等透明度(图2中A)。有趣的是,在紫外线照射下,含有Mg-MOF的GelMA水凝胶呈现黄色(图2中B)。这种颜色源于Mg-MOF的固有颜色。这种现象表明Mg-MOF在水凝胶中分布均匀。
GelMA水凝胶多孔支架具有可控的孔径和孔隙率。因此,我们用扫描电子显微镜分析了GelMA、LZIF-8@GEL、WMg-MOF@GEL和LZIF-8/WMg-MOF@GEL水凝胶的微观结构(图2中C-F)。GelMA水凝胶具有大而不均匀的孔隙,有利于药物的负载和细胞的粘附。药物释放实验模拟药物在体内的释放过程,研究药物释放的动力学和机制。在图2中G中,我们观察到一个以Zn快速释放为特征的初始阶段,随后是一个持续和渐进的释放过程。金属离子的控释可能受到水凝胶降解和纳米材料特性的影响。从结果来看,LZIF-8@GEL的控释效果优于LZIF-8。在图2中H中,我们同样观察到,与WMg-MOF相比,WMg-MOF@GEL具有更强的延缓镁离子释放的能力。
为了研究水凝胶支架的生物降解性能,我们将GEL和LZIF-8/WMg-MOF@GEL浸入胶原酶I或PBS溶液中。如图2中I所示,在PBS溶液中浸泡25天后,GEL和LZIF-8/WMg-MOF@GEL的降解率分别为56.4%和48.1%。从图2中J中可以看出,在胶原酶I溶液中7天后,GEL和LZIF-8/WMg-MOF@GEL的降解率分别达到84.1%和77.3%。胶原酶溶液模拟了肩袖插入点的生理环境,实验结果表明水凝胶支架具有良好的生物降解性。
为了评估水凝胶的机械性能,我们测量了储存模量(G')和损耗模量(G″),以评估其凝胶动态。与单独的GelMA组相比,加载MOF颗粒的GelMA组的G′值略有下降,这表明MOF颗粒的引入使GelMA的硬度有了适度的提高。在GEL和LZIF-8/WMg-MOF@GEL中,G′和G″在固凝胶向液凝胶转变过程中维持的临界应变值分别约为250%和248%(图2中K-L)。此外,我们还通过振荡频率测量评估了0.01至20Hz范围内的频率相关流变行为(图2中M)。据观察,水凝胶在整个频率范围内始终保持胶体形态。如图2中N所示,对于LZIF-8/WMg-MOF@GEL,G'在低应变时保持在1198Pa左右,但在高应变时降至约150Pa。然而,当恢复到低应变时,水凝胶的G'又迅速恢复到990Pa。这表明LZIF-8/WMg-MOF@GEL具有出色的形变能力和自愈能力。
为了评估水凝胶支架LZIF-8@GEL对肌腱细胞的生物相容性,我们通过活死染色法和MTT试验进行了检验。如图3中A所示,绝大多数肌腱细胞在3天的培养过程中表现出持续的活力,同时细胞密度逐渐增加。图3中B中的MTT结果证实了这些发现,显示随着时间的推移,肌腱细胞的数量逐渐增加,各实验组之间没有明显差异。总之,活体/死体染色和MTT检测的结果为LZIF-8@GEL的良好生物相容性提供了令人信服的证据,从而为后续的体内研究奠定了重要的先决条件。
紧接着,通过细胞摄取实验来评估水凝胶支架的靶向能力。采用吲哚菁绿(ICG)对纳米颗粒的细胞摄取活性进行荧光定位和跟踪。纳米材料ZIF-8-ICG@GEL和LZIF-8-ICG@GEL与肌腱成纤维细胞共培养6小时或24小时。利用LHERHLNNN对I型胶原蛋白的靶向特性,评估了细胞对ICG的摄取情况,以评估LZIF-8-ICG的靶向能力。此外,在6小时(图3中C)或24小时(图3中E),LZIF-8-ICG@GEL组比ZIF-8-ICG@GEL组显示出更强的红色荧光,表明两组之间存在显著的统计学差异(图3中D和F)。这表明LHERHLNNN肽的存在增强了细胞摄取能力,从而证实了MOFs的靶向能力。胶原蛋白在维持肌腱组织的结构和强度方面起着关键作用。
I型胶原蛋白以其机械韧性和坚固性著称,可提供坚固的框架,而III型胶原蛋白的坚固性相对较差,通常与疤痕组织有关。为了评估LZIF-8@GEL对肌腱的修复作用,我们利用I型胶原蛋白的表达作为代表性指标。免疫荧光利用特异性抗体和荧光染料,通过荧光显微镜观察和定位目标蛋白质。免疫荧光染色显示,与其他组相比,LZIF-8@GEL组的Ⅰ型胶原蛋白荧光表达最强(图3中G-H),表明LZIF-8@GEL能有效促进Ⅰ型胶原蛋白的分泌,有利于肌腱的修复。
为了评估水凝胶支架WMg-MOF@GEL对软骨细胞的生物相容性,我们也通过活死染色法和MTT试验进行了检验。在图4中A中,根据活死染色的结果,可以看出不同组间ATDC5细胞的形态和细胞密度相似。此外,根据MTT检测结果,培养1天、2天和3天的ATDC5细胞的细胞活力和增殖活性在不同组间没有统计学差异(图4中B)。总之,活/死染色和MTT检测结果表明WMg-MOF@GEL具有良好的生物相容性。与肌腱细胞的体外细胞摄取能力实验类似,利用WYRGRL对II型胶原蛋白的靶向特性,通过定量分析ATDC5细胞对吲哚菁绿(ICG)的摄取,验证了WMg-MOF-ICG@GEL的靶向能力。Mg-MOF-ICG@GEL和WMg-MOF-ICG@GEL与ATDC5细胞共培养6小时或24小时。结果表明,无论时间长短,WMg-MOF-ICG@GEL组的红色荧光强度均强于Mg-MOF-ICG@GEL组(图4中C、E),两组间差异有统计学意义(图4中D、F)。这些发现表明,WYRGRL肽的存在增强了细胞摄取能力,从而证实了MOF的靶向能力。
镁离子(Mg2+)在增强间充质干细胞聚集方面表现出显著的潜力,从而促进了纤维软骨的再生过程。据报道,II型胶原蛋白在软骨组织的形成过程中起着关键作用。与对照组相比,WMg-MOF@GEL组的翠绿荧光大幅增加(图4中G-H),表明水凝胶释放的Mg2+有效刺激了II型胶原蛋白的表达。总之,WMg-MOF@GEL通过释放镁离子有效地促进了BMSCs向软骨细胞的分化。
为了验证ZIF-8@GEL和WMg-MOF@GEL的体内靶向能力,我们采用小动物活体成像技术研究了ICG在体内的保留情况。与体外细胞摄取实验类似,我们利用ICG荧光标记MOFs。图5中A中,Mg-MOF@GEL组在注射8天后荧光消失,而WMg-MOF@GEL组可检测到荧光。如图5中B,ZIF-8@GEL组在置入5天后荧光消失,而LZIF-8@GEL组在第8天仍能检测到荧光。这些结果表明,LZIF-8@GEL或WMg-MOF@GEL释放的MOFs在肌腱骨界面停留的时间更长(图5中C、D)。
LZIF-8/WMg-MOF@GEL在体外显示出良好的治疗潜力,但体内和体外环境的新陈代谢可能存在差异。因此,我们建立了大鼠肩袖撕裂模型,以进一步评估LZIF-8/WMg-MOF@GEL的修复性能。我们直接缝合肌腱骨界面,或在缝合前在界面处插入GEL或LZIF-8/WMg-MOF@GEL。8周后,我们进行动物磁共振成像(MRI),检查肩袖内新形成的肌腱组织。随后,对大鼠实施安乐死,并对肱骨进行显微CT扫描,以评估骨再生特征(图6中B)。根据核磁共振成像结果,在T2加权序列中,LZIF-8/WMg-MOF@GEL组的肌腱看起来更接近对照组的健康肌腱,显示出较低的信号强度。然而,其他组的再生肌腱表现出较高的信号强度,这可能与组织炎症和水肿有关(图6中A)。
同时,我们还进行了生物力学实验,以观察肌腱-骨界面在不同外力作用下的承载能力和机械性能。生物力学实验证实,LZIF-8/WMg-MOF@GEL组与其他组相比,肌腱骨界面的最大载荷和刚度值明显更高(图6中C、D)。LZIF-8/WMg-MOF@GEL组的再生肌腱表现出较高的强度和良好的韧性。这些发现表明,LZIF-8/WMg-MOF@GEL可促进肌腱修复,使其达到更正常的组织状态。我们还进行了显微CT观察,以评估软骨下骨的再生情况。有趣的是,结果显示与其他组相比,LZIF-8/WMg-MOF@GEL组表现出更好、更完整的骨再生(图6中B)。如图6中E和F,结果显示在第8周时,LZIF-8/WMg-MOF@GEL组的BV/TV值和BMD值均高于其他对照组,表明其具有出色的骨再生能力。
为了观察再生肌腱骨组织的微观结构,我们对组织切片进行了染色和分析。血色素和伊红(H&E)染色是观察样本的常用方法。在图7中A显示,术后8周时,与其他两组相比,LZIF-8/WMg-MOF@GEL组肌腱中的纤维和细胞排列更有序,炎性细胞浸润减少。采用Masson三色染色法研究肩袖愈合过程中胶原纤维的新生情况。结果表明,LZIF-8/WMg-MOF@GEL促进了胶原纤维蛋白再生和规则排列。甲苯胺蓝(TB)染色法用于检测纤维软骨的含量和分布。蓝色染料会选择性地与软骨中的糖胺聚糖结合,从而在特定区域形成明显而强烈的颜色。术后8周,TB染色显示,与其他对照组相比,LZIF-8/WMg-MOF@GEL组的染色区域更明显,显示出统计学差异(图7中C)。上述结果表明,LZIF-8/WMg-MOF@GEL具有卓越的纤维软骨再生和促进肌腱修复能力。
为了评估Coll I和Coll II的分布和再生情况,我们进行了免疫组化分析(图7中B)。LZIF-8/WMg-MOF@GEL组的Coll I表达面积更大(图7中B和D)。随后,我们进一步研究了Coll II的表达,发现与其他两组相比,LZIF-8/WMg-MOF@GEL组的胶原纤维分布较多(图7中B、E)。这些结果表明,LZIF-8/WMg-MOF@GEL有助于胶原蛋白的再生和排列,从而有利于肌腱骨界面梯度结构的重建。此外,为了进一步验证材料的生物相容性,我们在材料植入两个月后采集了大鼠的心、肝、脾、肺和肾样本,然后进行了组织切片和H&E染色。结果显示,材料植入后对大鼠的其他组织没有明显的有害影响。综上所述,该双靶点金属离子网络水凝胶支架在促进受损肌腱-骨界面同步再生方面具有巨大潜力,在梯度结构特征的综合重建中的应用前景广阔(如图8)。
Claims (10)
1.一种双靶向性金属离子网络水凝胶的制备方法,其特征在于,包括以下步骤:
(1)以金属有机骨架ZIF-8与LHERHLNNN肽混合,制备得到负载LHERHLNNN肽的ZIF-8;
(2)以镁基金属有机骨架Mg-MOF与WYRGRL肽混合,制备得到负载WYRGRL肽的Mg-MOF;
(3)将GelMA水凝胶溶液与步骤(1)中负载LHERHLNNN肽的ZIF-8和步骤(2)中负载WYRGRL肽的Mg-MOF混合,经光交联固化,即得所述双靶向性金属离子网络水凝胶。
2.根据权利要求1所述的制备方法,其特征在于,所述步骤(1)中ZIF-8与LHERHLNNN肽的重量比为100:1。
3.根据权利要求1所述的制备方法,其特征在于,所述步骤(2)中Mg-MOF与WYRGRL肽的重量比为100:1。
4.根据权利要求1所述的制备方法,其特征在于,所述步骤(3)中GelMA水凝胶溶液、负载LHERHLNNN肽的ZIF-8和负载WYRGRL肽的Mg-MOF的重量比为500:1:1。
5.根据权利要求1所述的方法,其特征在于,
步骤(1)中金属有机骨架ZIF-8的制备方法包括:将六水合硝酸锌与2-甲基咪唑按重量比15:33混合于溶剂中进行反应,所得纳米颗粒物经离心后洗涤,真空干燥,即得ZIF-8。
6.根据权利要求5所述的方法,其特征在于,所述溶剂为甲醇,所述离心的条件为13000rpm离心10分钟。
7.根据权利要求1所述的方法,其特征在于,步骤(2)中镁基金属有机骨架Mg-MOF的制备方法包括:将2,5-二羟基对苯二甲酸与硝酸镁按照摩尔比1:2混合于N,N-二甲基甲酰胺中,加入氨水进行反应,制备得到Mg-MOF。
8.由权利要求1-7任一项所述的方法制备得到的双靶向性金属离子网络水凝胶。
9.如权利要求8所述的双靶向性金属离子网络水凝胶在制备促进肌腱-软骨损伤修复的药物方面的应用。
10.根据权利要求9所述的应用,其特征在于,是在制备促进肩袖损伤修复的药物方面的应用。
