技术领域

背景技术
聚糖存在于多种不同的蛋白质上,在蛋白上它们对蛋白质运输、稳定性和折叠有影响,最终改变蛋白质的生物化学和生物物理性质。此外,聚糖可介导蛋白水解模式或直接介导配体-受体相互作用、致癌信号转导、免疫识别、迁移以及细胞-细胞和细胞-基质粘附两者。因此,特定聚糖可对肿瘤细胞发挥选择性优势。因此,可将特定聚糖的存在或特定蛋白质上特定聚糖的存在用作,例如用于癌症诊断的生物标志物。
可以通过使用特异性结合特定聚糖结构的结合分子来分析聚糖结构。除了对聚糖结构特异的抗体之外,也可以使用凝集素。凝集素是对其它分子的一部分的糖基高度特异的碳水化合物结合蛋白。这些结合分子可以用在如酶联免疫吸附测定(ELISA)、酶联免疫凝集素测定(ELLA)、磁性ELLA(MELLA)的测定中以分析特定聚糖结构的存在与否。
然而,通过应用这些方法获得的信号需要通过(阳性)对照或标准品进行相对化,以允许有效陈述所述特定聚糖结构的存在与否或在样品中对其定量。可商购的糖蛋白标准品仅含有一种聚糖结构,其与目标聚糖结构不相同,即这些标准品不能用于评估目标蛋白质上与疾病相关的聚糖结构的存在与否。因此,包含特定聚糖结构的蛋白质标准品不容易获得。获得这种对照品的一种可能性是将目标聚糖结构与目标蛋白质缀合。然而,这是复杂的、劳动密集的且昂贵的。此外,对于每种新的目标聚糖结构和目标蛋白质的组合,必须重复该过程。

如果可以获得,现有技术中使用的标准品通常是目标蛋白,这些目标蛋白被怀疑在目标蛋白上的(目标)聚糖结构(A)标记。因此,现有技术中的标准品是包含聚糖结构(A)的目标蛋白本身。然而,合成此类标准品是复杂的、耗时的且昂贵的。因此,本发明旨在提供一种可靠的、廉价的和通用的聚糖结构或糖蛋白标准品,以用于使从确定被怀疑存在于目标蛋白上的聚糖结构中获得的信号相对化。

如本文所用的“拟糖蛋白”涉及链霉亲和素分子,至少1个且至多4个,即1、2、3或4个,优选4个生物素化的聚糖结构(A)通过生物素-链霉亲和素相互作用非共价地结合到该链霉亲和素分子上。因此,拟糖蛋白通过生物素-链霉亲和素相互作用结合。因此,链霉亲和素充当支架,其可以与期望的聚糖结构(A)偶联。因此,可以提供高度通用的并且可快速获得的拟糖蛋白标准品。因此,优选地,以过量,如至少4:1(聚糖结构(A):链霉亲和素)、至少5:1、至少7.5:1或至少10:1的摩尔比将聚糖结构(A)加入链霉亲和素中。优选地,以4.5:1至5.5:1的摩尔比,最优选以5:1的摩尔比将聚糖结构(A)加入到链霉亲和素中。

通常,当在本文中使用时,在两个或更多个核酸或多肽序列的上下文中,术语“百分比(%)同一性的”或“百分比(%)同一性”是指两个或更多个序列或子序列相同的程度。如果两个序列在被比较的区域上具有相同的氨基酸或核苷酸序列,则它们是“相同的”。当在比较窗口或指定区域上比较和比对最大对应性时,如果两个序列具有指定百分比的相同氨基酸残基或核苷酸(即,在指定区域上,或当未指定时,在整个序列上,60%同一性,任选地65%、70%、75%、80%、85%、90%、95%或99%同一性),则两个序列是“基本上同一的”,如使用以下序列比较算法之一或通过手动比对和目视检查所测量。任选地,同一性存在于长度为至少约30个核苷酸(或10个氨基酸)的区域上,或更优选地存在于长度为100至500或1000或更多个核苷酸(或20、50、200或更多个氨基酸)的区域上。
可以采用例如程序BLASTP,blastp 2.2.5版(2002年11月16日)(参见Altschul等,Nucleic Acids Res,1997)测定序列同源性或序列同一性百分比。在本实施方案中,同源性百分比基于包括前肽序列的完整多肽序列比对(矩阵:BLOSUM 62;间隙罚分:11.1;截止值设置为10-3),优选地在配对比较中使用野生型蛋白支架作为参考。作为BLASTP程序输出的结果表示的“阳性”(同源氨基酸)数除以程序选定的用于比对的氨基酸总数来计算百分比。
“聚糖”或“聚糖结构(A)”涉及由糖苷连接的单糖组成的化合物,并且还可以指糖缀合物的碳水化合物部分,比如糖蛋白、糖脂、糖RNA(参见,例如Flynn等,Cell,184(12):3109-3124)或蛋白聚糖,即使碳水化合物仅为单糖或低聚糖。

如本文所用的“信号”涉及通过确定(被怀疑存在在)目标蛋白上的聚糖结构(A)获得的信息。该信息通常是例如在特定波长处的吸收或在特定波长处的荧光发射的信号强度。优选地,信号强度取决于存在于目标蛋白上的聚糖结构(A)的量。因此,信号也可以看作提供了关于目标蛋白的糖普表征的信息。

可以将ELISA看作是各种相似或ELISA-样测定的起点。ELISA基于免疫球蛋白与目标分子(比如被怀疑存在于目标蛋白上的聚糖结构(A))的特异性相互作用,其中该种特异性相互作用允许产生这样一种信号,该信号仅在目标分子存在时产生,并且信号(强度)对应于分析样品中目标分子的浓度。如ELISA、ELLA和MELLA的测定是本领域技术人员已知的。各种ELISA变形,比如夹心ELISA、竞争性ELISA和反向ELISA也是本领域已知的。在本发明的上下文中,初级免疫球蛋白,即结合目标分子的免疫球蛋白,结合被怀疑存在于目标蛋白上的聚糖结构(A)。因此,初级免疫球蛋白,优选抗体或其片段,特异性结合目标聚糖结构(A)。在ELLA中,初级免疫球蛋白被特异性结合聚糖结构(A)的凝集素替代。磁性ELLA(MELLA)是WO2019/185515A1中描述的ELLA的进一步发展,在此通过引用以其整体并入。可以将这些测定的读数,例如信号强度,看作本发明上下文中的信号(1)或信号(2)。通常,读出是偶联至第二免疫球蛋白的酶的酶促反应的结果,所述第二免疫球蛋白结合至初级免疫球蛋白、目标蛋白(目标糖蛋白的蛋白质骨架)或凝集素。或者,酶可以(直接)与凝集素偶联。经常使用的读数包括但不限于变为琥珀色以检测经常用作缀合蛋白的HRP(辣根过氧化物酶)的OPD(邻苯二胺二盐酸盐);在检测HRP时变蓝,加入硫酸或磷酸后变黄的TMB(3,3',5,5'-四甲基联苯胺);在检测HRP时变成绿色的ABTS(2,2-连氮基-双-(3-乙基苯并二氢噻唑啉-6-磺酸)二铵盐);或在检测碱性磷酸酶时变成黄色的PNPP(p-硝基苯基磷酸,二钠盐)。除了这些比色读数之外,也可以使用放射性标记、荧光标记、电化学标记、化学发光标记、比色法或甚至无标记形式(例如SPR)。另一个实例是10-乙酰基-3,7-二羟基吩噁嗪(红试剂)检测过氧化氢(H2O2)或过氧化物酶活性。在过氧化物酶存在的情况下,/>红色试剂与H2O2以1:1的化学计量反应以产生红色荧光氧化产物:试卤灵。试卤灵具有约571nm和585nm的最大激发和发射。
当用于本文时术语“凝集素”指碳水化合物结合蛋白。凝集素通常对一种或多种碳水化合物部分是高度特异性的(例如,其特异性地与其它分子的末端糖苷残基反应,所述其它分子例如糖蛋白的一种或多种聚糖(例如糖蛋白的支链糖分子,例如本发明含义内的靶多肽和本文中表1所述的生物标志物)。凝集素是本领域公知的,技术人员容易获得以确定哪种凝集素可以用于结合一种或多种目标碳水化合物部分,如附着于蛋白质的聚糖的一种或多种碳水化合物部分。本文描述了本发明上下文中应用的优选凝集素。术语“凝集素”也包括Siglecs(结合唾液酸的免疫球蛋白样凝集素),半乳凝素(特异性结合含β-半乳糖苷的聚糖的凝集素)和选择素(结合唾液酸化路易斯X(SLex)决定簇NeuAcα2-3Galβ1-4(Fucα1-3)GlcNAc和相关的唾液酸化的岩藻糖化的聚糖)。值得注意的是,本文使用的术语“凝集素”也指聚糖结合抗体。因此,当在本文使用时,术语“凝集素”也可包括凝集素、Siglec、半乳凝素、选择素等,以及聚糖结合抗体。凝集素还可以包括识别聚糖的DNA/RNA适体。
凝集素可以从豆科植物的种子获得,也可以从其它植物和动物来源获得。凝集素可以含有特定单糖和低聚糖(例如糖蛋白的聚糖)的结合位点。它们可以通过与膜糖蛋白中的特定糖残基结合而凝集细胞。优选地,本发明的凝集素选自:朝鲜槐凝集素II(MAA II);刀豆凝集素A(Con A);橙黄网孢盘菌凝集素(AAL);西洋接骨木(SNA-I)凝集素;如本文所定义的多花紫藤凝集素(WFL)。

朝鲜槐凝集素II(MAA II)是来自朝鲜槐种子的血凝素同工凝集素。其是识别含有通过α2-3键与相邻半乳糖残基连接的末端唾液酸的低聚糖的唾液酸结合凝集素。结合三糖序列Neu5Acα2-3-Gal-β-1-4-GlcNAc。优选地,MAAII具有SEQ ID NO:2(或其成熟形式)。
刀豆凝集素A(Con A)是一种最初从刀豆-矮性刀豆(Canavalia ensiformis)提取的D-甘露糖特异性凝集素。优选地,Con A具有SEQ ID NO:3或SEQ ID NO:4(Con A,成熟形式)。

未缀合的链霉亲和素(~55kDa,未糖基化)、MAA-II(~130kDa)和抗-链霉亲和素抗体(~150kDa)购自美国Vector Labs。聚糖3'-唾液酸化乳糖胺-PEG3-生物素(单臂,~1100Da)获自加拿大Sussex Research。脱盐柱(MWCO=7000Da)购自美国Thermo。所有常用化学品,例如缓冲液成分等,购自美国Sigma Merck。对于所有实验,使用超纯去离子水(G=0.055μS)。所有缓冲液在使用前用0.22μm无菌过滤器过滤。
表面等离子体共振(SPR)
用于SPR的所有试剂都购自GE Healthcare,包括HBS-P+(缓冲液10×;BR-1006-71)、偶联试剂盒(BR100050)、EDC(0.4M)、NHS(0.1M)、乙醇胺盐酸盐(1M;pH 8.5)。为了再生,使用NaOH(50mM;BR-1003-58)和为了偶联,使用pH 4.0的乙酸盐缓冲液(BR-1003-51)。SPR测定在BiacoreX100(GE Healthcare)上在25℃下以30μL/min的恒定流速进行,所述BiacoreX100采用传感器芯片CM5(29-1496-04)或用11-巯基十一烷酸(UV/VIS乙醇中5mM,美国Sigma Merck;在RT下温育过夜)修饰的Au芯片。使用原始SW Biacore X100对照软件运行仪器。
纳米颗粒示踪分析(NTA)
将所有样品在0.1M PB(pH 7.4,0.22μm过滤)中稀释,并在连续流动下测量(制备500μl的总体积以用于单次运行)。如所建议的,通过预测试理想的颗粒/帧(frame)值(20–100颗粒/帧)来发现测量浓度。根据NanoSight NS300的制造商软件手册设置以下设置:确定检测阈值包括尽可能多的颗粒,计数10–100红十字限制。蓝色交叉计数被限制到最小。调节自动聚焦以避免不清楚的颗粒。对于每次测量,在以下条件下捕获五个60s视频:池温度:25℃;注射器速度:10μl/min。在捕获之后,采用检测阈值=10,通过内置NanoSight软件NTA3.1Build 3.1.46分析视频。
结果和讨论
链霉亲和素-聚糖拟糖蛋白制备
将冻干的链霉亲和素重悬于无菌的0.1M PB(pH 7.4)中以获得1mg.ml-1的浓度。对于已知浓度的生物素化聚糖,在37℃下采用温和的混合(500rpm)将这两种溶液以1+5分子比率混合1小时。最后,使用具有7k MWCO的Zeba Spin脱盐柱(预先用PB平衡)在3000rpm下将拟糖蛋白从多余的聚糖中纯化1分钟。在c=~0.9mg.ml-1下获得拟糖蛋白,并将其储存在4℃下以用于实验工作的其余部分。
SPR结合分析。在第一次运行中,使用羧甲基葡聚糖CM5 SPR芯片来发现最佳的固定pH值,随后用于将配体固定在传感器表面上。基于在不同pH值的传感器界面上配体预浓缩期间的斜率值,所有情况的最佳pH是4.0(图2A)。链霉亲和素和MAA-II凝集素的pI值分别是5.0和4.7,而对于拟糖蛋白也预测了类似的值。在所有三种情况下,即对于链霉亲和素、拟糖蛋白和MAA-II凝集素,使用用于活化和乙醇胺封闭的胺偶联(EDC/NHS方案)(图2B)。为了完成夹心构型,抗-链霉亲和素抗体(Ab)和MAA-II凝集素应同时结合拟糖蛋白。然而,可能由于空间障碍、相当高的负电荷密度和棱镜/金界面的层厚度/距离(以及这些因素的组合),在单循环动力学(SCK)期间仅可观察到Ab结合至链霉亲和素修饰的CM5芯片(图3)。对于其余的实验,SCK期间样品的浓度越高,通常阴性反应(negative response)越强-甚至在使用拟糖蛋白修饰的芯片的凝集素结合期间(具有MAA-II、SNA-I和WGA,所有这些都应结合唾液酸化的/带负电荷的聚糖部分)也是如此。
为了确认成功制备了夹心(MAA-II/聚糖-链霉亲和素/抗体),通过将裸Au芯片浸入5mM 11-巯基十一烷酸的乙醇溶液(RT,黑暗,过夜)制备2D芯片。这种方法的优点是基本上所有的负电荷(羧基)都位于表面上,并且在固定/封闭后被除去。整个夹心制备以及测定工作流程示于图5A中。
在下表2中汇总了应用于每个步骤的条件的详细概述。
表2:使用SPR结合分析,用于夹心制备的各个步骤的概述,即分子类型、条件和结果,如图4所示。所有R.U.都是在从检测池(FC2)中减去空白(流动池1,FC1)之后计算的。尽管表面的再生是任选的,并且不应用于GlycanosticsMELLBA测定,仅使用50mM NaOH不能完全再生表面。
MNP修饰和富集过程的NTA分析
为了进行NTA分析以观察磁性纳米颗粒(MNP,130nm COOH封端的,葡聚糖包被的纳米标签)的修饰,所有三个样品,即裸MNP,Ab修饰的MNP(MNP+Ab)和富含拟糖蛋白的Ab修饰的MNP(MNP+Ab+C)被同等处理,在空白的情况下无论使用特定的化学/成分还是使用PB,所有三个样品因此以相同的方式/相同的次数分离。在这种情况下,使用用于生物识别的球形界面产生更高的信号,因为配体密度随着更长的连接体分子而降低,从而与用于上述SPR实验的平面表面相比降低了对MELLBA的空间阻碍(图6A和B;假说,然而在别处公开)。NTA的结果示于图6C。在修饰/富集之后,由于修饰的MNP的直径增加,它们更可能在试管底部形成沉淀。对于NTA分析,将每个样品在无菌PB中稀释500x。
未修饰的(但分离相等次数)MNP几乎只产生一个单峰,d≤140nm,而在Ab缀合(胺偶联,10分钟,RT)后有两个主要级分–(i)≤120nm(与未修饰的颗粒相比稍弱)和约20%的(ii)≤150nm。在将这些颗粒用纯化的拟糖蛋白孵育之后,可以观察到至少四个不同的级分–甚至更大的聚集体在~220nm处出现。如果在固定化过程中MNP的结合能力不饱和(假设),则通过将固定化的ab周围的葡聚糖基质“压缩(compressing)”来引起过程1(ab固定化)中流体动力学直径(dH)的降低。否则,如图7所示,dH增加。该实验的一个重要方面是在过程2中dH增加~20nm–这一值受聚糖实际情况(三糖以及PEG3连接体是高度水合的且相当柔性的)的影响。
检测聚糖含量的MALDI-TOF MS和ELLBA
使用不同的链霉亲和素+生物素化-聚糖比率(即,每体积分子数的1+0、1+1、1+2、1+3、1+4、1+5、1+6和1+7比率)来制备完整糖基化的蛋白标准品。如前面其它地方所述,使用MALDI-TOF质谱测定法(Bruker,USA)和先前优化的方案(使用2,5-二羟基苯乙酮(DHA)基质)来检测电离的样品组分的质量片段(m/z参数)。在图9中,详细显示了质谱,即所有样品(A)中的链霉亲和素片段,差异为~13kDa,即~55kDa同源四聚体中一个亚基的质量。最强的峰也总是在~13kDa(图8),而其它峰具有低得多的强度。只有先前与生物素化-聚糖孵育的样品(除了1+0,即裸露的链霉亲和素之外的所有样品,图9B)也显示存在m/z=~1300Da的峰,3'-唾液酸化的聚糖衍生物(图9C)。此外,该峰的强度随着聚糖/链霉亲和素比率的增加而增加(图9D)。然而,由于链霉亲和素仅可结合至多4个生物素,因此重要的是使用适当的聚糖/链霉亲和素比率以维持蛋白标准品被这些聚糖完全饱和。
为此目的,使用酶联凝集素结合测定(ELLBA)来找出完全饱和比率。在图9E左中描绘了该测定构型–将蛋白标准品固定在ELISA板孔的底部,用未缀合的MAA-II(2,3-唾液酸/Gal特异的,其可有效封闭所有可用的聚糖表位),随后用生物素化的MAA-II孵育,所述MAA-II携带若干生物素分子并结合至不饱和链霉亲和素标准品上的游离生物素结合位点。随后,将链霉亲和素-过氧化物酶用于产生光学信号(OPD/过氧化氢)。链霉亲和素-HRP仅与生物素化MAA-II上存在的游离生物素分子结合,因此与链霉亲和素上存在的聚糖分子的量成反比关系(图9E右)。
结论
通过将生物素化的聚糖附着至链霉亲和素制备的拟糖蛋白(蛋白标准品)同时被MAA-II凝集素和抗-链霉亲和素抗体识别。这一意外发现表明,这种拟糖蛋白可作为ELISA样分析形式(包括MELLA)的蛋白标准品应用。
实施例2
实验工作显示,当在4℃下储存时,糖蛋白标准品–在本文中也称为拟糖蛋白(标准品)或本发明的标准品,例如如上文实施例1中应用的标准品稳定至少1周,这种标准可以以高度可再现的方式制备,即对于在18周内以17个独立制备批次制备糖蛋白标准品,RSD为8.17%。
Claims (15)

其中相对化包括将信号(1)与来自标准品的信号(2)进行比较,从而信号(2)允许将通过信号(1)获得的信息置于相关,
从而使所述信号(1)针对所述信号(2)相对化,反之亦然。
2.根据权利要求1所述的方法,其中相对化包括
