Abstract
本发明公开了一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,包括如下步骤:首先将tarRNA、crRNA和LwaCas13a特异性结合形成三元复合物,利用Cas13a蛋白的旁系切割能力,实现对RNABiotin和6‑FAM双标记探针的非特异性、非靶标性切割;然后以感光微球和发光微球作为检测平台对双标记探针进行特异性结合,完成光信号的检测,实现对靶标序列的定量。本发明采用单链的RNA探针连接发光微球和感光微球,使均相时间分辨荧光免疫分析方法的应用范围更广,而CRISPR/Cas13a的高特异性和高灵敏性也提升了均相时间分辨荧光免疫分析方法的可靠性和检测灵敏度。

RNA作为遗传信使,在细胞和组织的发育以及疾病的进展中起着至关重要的调节作用,对RNA的高灵敏度检测和定量将有助于我们对各类疾病做到即时的预防和治疗,另外也可以阐明它们在细胞功能中所起到的作用。国家和大众迫切需要超灵敏和低成本的RNA诊断,因此及时准确地检测RNA可以有效监测和控制传染病的爆发,并及早发现包括癌症在内的其他病理状况,定量逆转录聚合酶链反应(qRT-PCR)是目前RNA检测的金标准,其灵敏度高,但由于其笨重且昂贵的检测设备、繁琐的操作步骤、高昂的检测成本和较高的假阳性,使其无法进行快速、准确的即时检测和用于大规模的常规检测,其他RNA检测技术,包括微阵列和核糖核酸荧光原位杂交(RNAFISH),在信号特异性、灵敏度和稳定性方面存在局限性。CRISPR/Cas13a作为一种高效而强大的基因编辑工具,被科学界广泛的用于体内基因工程和离体生物检测,基于CRISPR/Cas13a的生物检测的优点在于可编程识别与tarRNA互补配对的crRNA激活Cas13a蛋白,通过设计与tarRNA序列互补的crRNA来实现对tarRNA检测的特异性,在识别tarRNA并将其与crRNA结合后,Cas13a蛋白对反应体系内的单链RNA(ssRNA)切割能力被激活,Cas13a蛋白的高特异性使其能够区分单碱基错配,对tarRNA有更高的灵敏度和特异性,与qRT-PCR等传统方法相比,显著提高了检测准确性。基于CRISPR/Cas13a生物检测的传统方法涉及用共振能量转移(FRET)对标记的报告RNA(reRNA),其中reRNA的切割导致荧光信号的激活。与传统荧光团相关的微弱荧光信号不可避免地需要预扩增过程,但均相时间分辨荧光免疫分析由发光微球产生的强荧光使的CRISPR/Cas13a的检测不需要预扩增减少了实验过程中的污染和提高了检测准确性,例如张峰团队提出的SHERLOCK检测需要CRISPR/Cas13a与重组酶聚合酶扩增相结合,在扩增的过程中其容易出错并且需要精心设计的引物操作繁琐容易污染,因此其方法主要用作定性。而均相时间分辨荧光免疫分析方法,均相是指所有参与反应的试剂或化合物无需经过洗涤处理,反应体系内的各成分互不影响,时间分辨荧光是一种将荧光共振能量转移(1946年由TheodorForster首先提出,该方法是基于两个荧光团之间的能量转移,一个是感光微球,一个是发光微球,当两者接近时,生物分子之间的相互作用可以通过耦合的荧光标记进行检测)与时间分辨荧光(源于Jean-Marie Lehn获得诺贝尔奖的稀土化合物研究,该化合物具有良好的化学和光物理特性,且在时间上特别稳定;该技术利用稀土化合物中的镧系元素,该元素具有不同于普通荧光的半衰期(纳秒级),为毫秒级,因此,经过50μs的时间延迟,普通荧光的信号几乎为0,使得时间分辨荧光背景信号低,能够真实的反应样品的实际情况)相结合的技术,通过采用两种抗体识别目标蛋白,一种抗体与感光微球偶联,另一种抗体与发光微球偶联。如果2个抗体识别到目标蛋白,感光微球和发光微球的基团接近时,引发能量转移,感光微球会诱导发光微球长时间地发射荧光,发光微球激发后产生的荧光便能持续较长时间,这样通过时间分辨就可以区分出荧光共振能量转移的信号,使其易于检测且节省时间、高通量、荧光信号在时间上更稳定。更重要的是,各种各样的病原体,如铜绿假单胞菌,非洲猪瘟病毒,牛病毒性腹泻病毒等等,都已成功通过CRISPR/Cas13检测,但在测量之前不可避免的核酸扩增方案是上述此类方法的共同缺点,微弱的荧光信号、较差的信噪比和较低的灵敏度是基于CRISPR/Cas13a检测的主要挑战。
近年来,CRISPR/Cas13a系统已成为新一代分子诊断工具,其作为对抗病毒感染、肿瘤预防、疾病检测的有力武器受到人们越来越多的关注,随着对分子监测需求的不断升级,CRISPR技术和其他检测仪器通过巧妙的组合相辅相成,进一步改良了核酸检测领域,Cas13a核酸内切酶可以通过荧光以外的各种信号转导使Cas13a检测性能充分发挥作用,如电化学、光学等,用于高灵敏检测肿瘤细胞中的tarRNA,其中由tarRNA激活的Cas13a可以切割设计良好的预引物并触发下游发光反应。这些基于电化学或光转导的实现通常需要复杂的器件设计、多步反应或适当的指示性分子,CRISPR/Cas13a仪器的联用提高了检测的灵敏度但操作复杂、检测成本高、实验设备昂贵。
发明内容
为解决上述问题,本发明提供了一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,将原来连接感光发光微球的抗原换成了单链的RNA探针连接感光和发光微球,使均相时间分辨荧光免疫分析方法的应用范围更广,而CRISPR/Cas13a的高特异性和高灵敏性也提升了均相时间分辨荧光免疫分析方法检测的可靠性和检测灵敏度,提高了检测肿瘤的灵敏度和降低了检测的成本。
为实现上述目的,本发明采取的技术方案为:
一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,首先将靶标RNA(tarRNA)、crRNA和LwaCas13a特异性结合形成三元复合物,利用Cas13a蛋白的旁系切割能力,实现对RNA Biotin和6-FAM双标记探针的非特异性、非靶标性切割(反式切割);然后以偶联链霉亲和素的含感光物质的聚苯乙烯微球(感光微球)和偶联6-FAM抗体的含发光物质的聚苯乙烯微球(发光微球)作为检测平台对双标记探针进行特异性结合,完成光信号的检测,实现对靶标序列的检测。
本发明实施例的一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,具体包括如下步骤:
S1、将LwaCas13a蛋白和crRNA预先混合形成LwaCas13a:crRNA复合物,在这种情况下,crRNA对tarRNA的核苷酸序列具有特异性;
S2、将6-FAM和生物素标记的ssRNA探针引入所述的LwaCas13a:crRNA复合物中,37℃孵育半小时;
S3、将感光微球和CRISPR/Cas13a反应液添加到96孔板中震荡孵育15分钟,再加入发光微球孵育15分钟,通过均相时间分辨荧光免疫分检测仪检测荧光值;当靶RNA存在时与crRNA互补配对激活Cas13a蛋白,切割溶液中的ssRNA探针,在有tarRNA的情况下,被切断的ssRNA俩端与感光发光微球特异性结合,但由于ssRNA被Cas13a蛋白切断,感光微球与发光微球的距离大于200nm(6-FAM和生物素标记的ssRNA探针长度远小于200nm,它代替抗原连接感光微球和发光微球),发光微球接收不活性氧无法产生荧光。在没有靶RNA的情况下,未切割的ssRNA探针被感光和发光微球捕获,感光微球和发光微球的距离小于200nm,激发光将感光微球激活感光微球产生活性氧传递到发光微球,发光微球吸收活性氧产生荧光;
S4、基于ssRNA探针切割与目标浓度的相关曲线,目标浓度与检测信号的关系曲线,得到原始样品中存在的靶标RNA量。
进一步地,所述的步骤S1中,取2μL 500nM的LwaCas13a蛋白和2μL500nM的crRNA预先混合形成Lwa-Cas13a:crRNA复合物。
与现有技术方案相比,本发明具有以下有益效果:
本发明单链的RNA探针替代抗原将感光微球和发光微球连接起来,RNA探针与抗体的结合会导致感光微球和发光微球靠近(小于200nm),当感光微球与发光微球距离小于200nm时,在激光激发情况下,发光微球能够接受周围感光微球释放的活性氧,从而启动发光微球表面的化学发光反应过程并产生光信号(610nm);将CRISPR/Cas13a和均相时间分辨荧光免疫分析方法结合到一起,Cas13a的作用机制是特异性的检测单链RNA,均相时间分辨荧光免疫分析方法是利用发光物质(luminescent material)和感光物质(photoactivesubstance)两种标记,借助RNA探针-抗体间结合实现高能活性氧的传递,诱导光激发化学发光过程,从而实现“免分离”的均相免疫分析,精密度高,检测速度快。同时,在操作模式方面,分析仪器采用微孔八联条,自由选择标本优先或项目优先模式,在考虑常规标本最大检测速度的同时,可实现急诊标本优先测定。基于均相时间分辨荧光免疫分析检测平台,已研发感染免疫血清标志物(含ToRCH)、血清肿瘤标志物、甲状腺功能相关激素、性腺相关激素等数十种常规检测项目。本发明可用于检测细胞裂解液、血液、血清等样品中的RNA水平,可适用于miRNA、LncRNA、环状RNA等。

具体实施方式
下面结合具体实施例对本发明进行详细说明。以下实施例将有助于本领域的技术人员进一步理解本发明,但不以任何形式限制本发明。应当指出的是,对本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进。这些都属于本发明的保护范围。
为了克服原来RNA检测的限制实现超灵敏、低成本、易于使用和快速的RNA定量定性技术,并在资源有限的环境中大规模筛查传染病和其他疾病。本发明实施例提供了一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,将原来连接微球的抗原换成了单链的RNA探针,并将CRISPR/Cas13a和均相时间分辨荧光免疫分析方法结合到一起;其中,均相时间分辨荧光免疫分析方法是一种新型的均相免疫分析系统,全程无需分离游离标记微球和未参加抗原抗体结合的标记物质(非均相免疫分析需要分离去除未参加反应的游离标记物,测定结合标记物的信号强度,而均相时间分辨荧光免疫分析体系中游离发光微球不能获得活性氧能量,不会产生光信号,因此,无需分离洗涤,可实现均相免疫分析)。在均相时间分辨荧光免疫分析方法中,发光物质和感光物质均分布于粒径约200nm的微球表面,并不结合生物分子(抗原或抗体),且其分布和微球使用浓度相关。同时,发光微球或感光微球均为聚苯乙烯微球,比重轻(1g/cm3),且粒径远小于普通化学发光微球(1000nm)的粒径,微球密度接近水溶液,于水溶液中呈悬浮状态。基于上述特性,通过控制两种微球的工作浓度,使得感光微球和发光微球之间的距离大于活性氧最远扩散距离,即使在激光激发情况下,发光微球不能接受活性氧的能量,不能启动化学发光反应即不产生光信号。但是,由于微球表面包被生物分子(抗原或抗体),RNA探针与抗体的结合会导致感光微球和发光微球靠近(小于200nm),当感光微球与发光微球距离小于200nm时,在激光激发情况下,发光微球能够接受周围感光微球释放的活性氧,从而启动发光微球表面的化学发光反应过程并产生光信号(610nm)。但如将上述连接微球的RNA探针切断(靶标RNA与crRNA、Cas13a蛋白结合形成三元复合物被激活产生旁系切割能力)两种微球于液相中分散,当光激发时不满足能量传递条件(感光微球和发光微球距离小于200nm),无光信号产生(整个均相时间分辨荧光免疫分析检测过程分为两个阶段:RNA探针与抗体结合和化学发光过程);而基于CRISPR/Cas13a的生物检测的优点在于可编程识别和靶标依赖性激活,通过设计与tarRNA序列互补的crRNA来实现对靶RNA检测的特异性,在识别靶RNA并将其与crRNA结合后与Cas13a蛋白形成三元复合物,Cas13a对反应体系内单链RNA(ssRNA)切割能力被激活。Cas13a蛋白的高特异性使其能够区分单碱基失配(准确的检测待测物质),与qRT-PCR等传统方法相比,显著提高了检测灵敏度降低假阳性。在本发明中,Cas13a的作用机制是特异性的检测单链RNA,均相时间分辨荧光免疫分析利用发光物质(luminescent material)和感光物质(photoactive substance)两种标记,借助RNA探针-抗体间结合实现高能活性氧的传递,诱导光激发化学发光过程,从而实现“免分离”的均相免疫分析,精密度高,检测速度快。
具体使用时,本发明的一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,包括如下步骤:S1、将LwaCas13a蛋白和crRNA预先混合形成LwaCas13a:crRNA复合物;S2、将待测样品加入所述的LwaCas13a;S3、将生物素6-FAM和生物素标记的ssRNA探针引入所述的LwaCas13a复合物中,37℃孵育半小时;S4、感光微球和CRISPR/Cas13a反应液添加到96孔板中震荡孵育15分钟,再加入发光微球孵育15分钟,通过均相时间分辨荧光免疫分析检测仪检测荧光值;当靶RNA存在时激活Cas13a:crRNA:tarRNA三元复合物,切割溶液中的ssRNA探针,S5、基于ssRNA探针切割与目标浓度的相关曲线,目标浓度与检测信号的关系曲线,得到原始样品中存在的靶标RNA量。由于ssRNA探针切割与目标浓度呈正相关,目标浓度与检测信号呈负相关。因此,信号强度越高,表明原始样品中存在的靶标RNA量越低。
实施例1
链霉亲和素、6-FAM抗体偶联供体微球和受体微球
1.含有感光物质和发光物质的聚苯乙烯微球偶联方法(以偶联6-FAM抗体为例)微球的稀释:I.取1mg微球(固含1%)标记0.1mg 6-FAM抗体,100ul供体微球+900ul标记缓冲液(50mM MES,pH 6.0);II.微球的活化各称取20mg NHS和EDC,用标记缓冲液溶解,现用现配,即20mg/ml NHS和EDC;取10ul NHS,加入到清洗后的微球中,快速混匀;然后再取5ulEDC加入到微球中,快速混匀;室温混匀孵育,20min;III.清洗去除残留EDC将活化后的微球:1.17000rpm,离心20min,第一遍;2.去掉上清,用1000ul标记缓冲液重悬微球;3.17000rpm,离心20min,第二遍;4.去掉上清,用1000ul标记缓冲液重悬微球,备用;IV.微球与6-FAM抗体的偶联取0.1mg抗体于2ml离心管中,加入活化后的微球,快速混匀后,室温混匀孵育,2小时;V.封闭配制20mg/ml的BSA,即称取20mg BSA,用终浓度100mM乙醇胺溶液充分溶解,备用;供体微球标记抗体后,加入100ul上述封闭液(含BSA),室温混匀孵育1小时。VI.去除未结合的抗体,用1000ul稀释液重悬微球,即为抗体-微球标记复合物,4℃放置备用。
2.基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法检测LncRNA H19。
制备含有不同浓度的LncRNA溶液,在37℃下孵育30分钟并将它们应用于其检测(图2),在孵育时间半小时内实现了对1pM LncRNA待测物的检测。
3.临床相关标本中检测低LncRNA浓度的可行性
对患结肠癌的4个患者和20个健康人的血清样本进行了测试和对比,使用常用的RNA纯化试剂盒分离血清样品中的总RNA并在37℃下孵育,通过均相时间分辨荧光免疫分析方法检测荧光值进行对比(图3),通过患者和健康人检验结果对照验证我们检测方法在临床相关标本中检测低LncRNA浓度的可行性。
本发明将过去通过抗原连接受体珠和供体珠的检测方法改良成通过一段含10个核苷酸的RNA探针连接受体珠和供体珠,使均相时间分辨荧光免疫分析方法的应用范围更广,而且CRISPR/Cas13a的高特异性和高灵敏性也提升了均相时间分辨荧光免疫分析检测的可靠性和检测灵敏度,提高了检测肿瘤的灵敏度和降低了检测的成本。与其它发光免疫分析类似,均相时间分辨荧光免疫分析方法同样包括光激发化学发光过程和免疫分析过程,前者赋予此系统高灵敏度,后者保证此系统高特异性,均相时间分辨荧光免疫分析借助“双球”和“双标”的特性,无需分离未参加免疫反应的标记微球,避免复杂的洗涤过程,完美实现均相免疫分析。
以上对本发明的具体实施例进行了描述。需要理解的是,本发明并不局限于上述特定实施方式,本领域技术人员可以在权利要求的范围内做出各种变形或修改,这并不影响本发明的实质内容。
Claims (3)
1.一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,其特征在于:首先将tarRNA、crRNA和LwaCas13a特异性结合形成三元复合物,利用Cas13a蛋白的旁系切割能力,实现对RNA Biotin和6-FAM双标记探针的非特异性、非靶标性切割;然后以偶联链霉亲和素的感光微球和偶联6-FAM抗体的发光微球作为检测平台对双标记探针进行特异性结合,完成光信号的检测,实现对靶标序列的定量。
2.如权利要求1所述的一种基于CRISPR/Cas13a的均相时间分辨荧光免疫分析方法,其特征在于:包括如下步骤:
S1、将LwaCas13a蛋白和crRNA预先混合形成LwaCas13a:crRNA复合物,再加入待测RNA样品;
S2、将6-FAM和生物素标记的ssRNA探针加入所述的LwaCas13a:crRNA复合物中,37℃孵育半小时;
S3、将50μL的感光微球和2μL的CRISPR/Cas13a反应液添加到96孔板中震荡孵育15分钟,再加入50μL的发光微球孵育15分钟,通过均相时间分辨荧光免疫分析检测荧光值;当靶RNA存在时与crRNA互补配对形成Cas13a:crRNA:tarRNA三元复合物激活Cas13a蛋白,切割溶液中的ssRNA探针,在有tarRNA的情况下,被切断的ssRNA俩端与感光发光微球特异性结合,但由于ssRNA被Cas13a蛋白切断,感光微球与发光微球的距离大于200nm,发光微球接收不到活性氧无法产生荧光;在没有靶RNA的情况下,未切割的ssRNA探针被感光和发光微球捕获,感光微球和发光微球的距离小于200nm,激发光将感光微球激活感光微球产生活性氧传递到发光微球,发光微球吸收活性氧产生荧光;
S4、基于ssRNA探针切割与目标浓度的相关曲线,目标浓度与检测信号的关系曲线,得到原始样品中存在的靶标RNA量。
3.如权利要求1所述的一种基于CRI SPR/Cas13a的均相时间分辨荧光免疫分析方法,其特征在于:所述的步骤S1中,取2μL 500nM的LwaCas13a蛋白和2μL 500nM的crRNA预先混合形成LwaCas13a:crRNA复合物。
