Abstract
本发明公开了基因Wisp1在制备调控MSCs抗衰老表型药物中的应用,涉及干细胞衰老表型调控及药物研发技术领域,其技术方案要点是:Wisp1基因能够通过调控细胞增殖、分化和血管骨骼肌系统发育的相关基因,从而精准调控MSCs衰老表型。衰老的MSCs经Wisp1调控后,其衰老表型改善,为开发治疗退行性疾病的药物提供了创新思路。此发明不仅有利于推进老年退行性疾病治疗方式的革新,同时也为退行性疾病治疗的应用提供了新的理论支撑。
Description
技术领域
本发明涉及基因工程技术领域,更具体地说,它涉及基因Wisp1在制备调控MSCs抗衰老表型药物中的应用。
背景技术
Wnt-inducible signaling pathway protein 1(Wisp1),也被称为CCN4或者Elm1,是一种富含半胱氨酸的多细胞蛋白,属于CCN(CYR61/CTGF/NOV)蛋白家族的成员之一。作为Wnt/β-连环蛋白(Wnt/β-catenin)信号通路的下游效应分子,Wisp1由367个氨基酸组成,并富含38个保守的半胱氨酸残基以及4个潜在的N-糖基化位点。在成人期,Wisp1在多个器官如大脑、心脏、肾脏、肺、胰腺、胎盘、卵巢、小肠和脾脏中均有表达。该蛋白负责调控细胞的增殖、分化和凋亡,并可促进神经、血管和骨骼发育。此外,Wisp1与体内的代谢平衡有着密切的关联,并在细胞衰老过程中展现出显著的调控作用。Wisp1不仅在多种器官的上皮细胞中有表达,还在间充质细胞中表达,因此在组织修复和再生方面具有重要的生物学功能。这些特性使Wisp1成为多种疾病治疗和组织工程领域内的一个重要研究目标。
Wisp1作为Wnt通路的下游靶基因,在多种生理和病理条件下具有显著的细胞保护作用,特别是在心肌组织损伤、氧化应激、神经元退行性变等场景下,能够磷酸化Akt,进而通过PI3k/Akt信号通路来抑制细胞凋亡。该通路不仅能够增强神经胶质细胞的生存能力,还能维护血管内皮细胞的结构完整性,防止神经退行性变,促进心肌保护,以及增强细胞对氧化应激的耐受性。近期研究进一步揭示了Wnt蛋白及其下游信号通路在骨髓间充质干细胞(MSCs)的自我更新和分化中的核心作用。作为Wnt/β-catenin信号通路的下游效应分子,Wisp1参与调节细胞黏附和迁移,并可作为一种生长因子,影响细胞的增殖和分化,特别是在人类骨髓间充质干细胞(MSCs)的增殖以及成骨细胞和软骨细胞的分化方面。值得注意的是,Wisp1对p53的活化具有负性调控作用。Wisp1的过表达能导致Akt的磷酸化和抗凋亡蛋白Bcl-xL的表达上调,同时抑制线粒体释放的细胞色素C,从而阻断p53介导的细胞凋亡途径,减少细胞死亡。
也有研究发现,Wnt1诱导的Wisp1是一种由FAPs(Fibro-adipogenicprogenitors)衍生的基质细胞信号分子,该信号在生物衰老过程中逐渐减弱或丧失。Wisp1在肌肉再生机制中起到至关重要的作用,特别是通过Akt信号通路来调控肌细胞的增殖和不对称分化。全身性的Wisp1治疗能够显著提升老年小鼠骨骼肌的肌生成能力,并有效促进骨骼肌组织的再生。相反,Wisp1的缺失或功能失调会导致老年骨骼肌出现明显的肌肉功能障碍。
发明内容
本发明旨在提供Wisp1在间充质干细胞(MSCs)抗衰老调控的应用,Wisp1通过调节与细胞增殖、发育及血管生成相关基因,从而精准地调控MSCs衰老表型,经Wisp1调控后,MSCs衰老表型明显改善,经调控改善后的MSCs具有治疗老年退行性疾病的潜力。
为实现上述技术目标,本发明提供以下技术方案:所述MSCs在Wisp1调控作用下表现出衰老表型的明显改善,可被用作制备治疗退行性疾病的药物。
本发明进一步设置为:所述药物通过Wisp1增强其作用,所述Wisp1基因通过调控细胞增殖和发育过程,从而精准调控MSCs的衰老表型。
本发明进一步设置为:所述药物是使Wisp1基因过表达,从而有效改善老年MSCs的衰老表型。
本发明进一步设置为:所述Wisp1基因能够调控SDF-1、Survivin、Bcl-2、Nqo1、Sod2以及其他免疫调节相关的基因表达,进一步影响MSCs的衰老表型。
本发明进一步设置为:所述药物通过增强Wisp1基因表达,显著提升MSCs的成骨、成脂诱导分化潜能。
综上所述,本发明具有以下有益效果:
本发明在研究中发现,在Wisp1过表达处理的青年和老年小鼠中,SDF-1、Survivin以及Bcl-2蛋白的表达均有显著增加。这一结果暗示Wisp1的过表达可能对增强MSCs的定位归巢、细胞增殖以及抗凋亡功能起到了积极作用。此外,Wisp1的过表达还能显著提升小鼠干细胞的成骨和成脂的诱导分化潜能。当Wisp1基因在青年小鼠MSCS中被沉默后,与细胞增殖、细胞发育和血管骨骼肌系统发育相关的基因表达发生了显著变化。其中,多数有利于细胞增殖和血管骨骼肌系统发育的基因被显著下调。此外,青年小鼠MSCs中Wisp1基因的敲除加速了细胞衰老,而在老年鼠MSCs中过表达Wisp1则能逆转衰老表型,确认Wisp1是MSCs衰老的关键调控基因。因此,Wisp1可以精准调控MSCs衰老表型,并进一步将这些经过调控的MSCs用于治疗退行性疾病。

具体实施方式
以下结合附图1-33对本发明作进一步详细说明。
本发明中所用到的缩略词中英文对照如下:
实施例1:不同年龄小鼠骨髓间充质干细胞原代分离、培养及鉴定
实验动物:选取SPF级别的C57BL/10雄性小鼠,其来源为成都达硕实验动物有限公司。实验分组为三个年龄段:青年小鼠(<4周,MiceY,N=50)、中年小鼠(12周-16周,MiceM,N=50)及老年小鼠(>12个月,MiceO,N=50)。
细胞分离与培养:采用创新改良的全骨髓及密质骨碎片滤过贴壁培养法。首先,小鼠经脱颈处死,75%酒精浸泡5分钟。在超净工作台内,取出双侧胫骨和股骨,去除骨上的肌肉和筋膜。随后,将骨在含α-MEM的培养基中碾碎,收集上清液并通过100μm和40μm滤膜进行双重过滤。离心处理后,用含17% Gibco胎牛血清的α-MEM完全培养基重悬细胞,并调整至1×106/mL。接种至25cm2塑料培养瓶中,并在37℃、5% CO2的条件下培养。48小时后,更换新鲜培养基,并在细胞融合达到80%-90%后,使用Gbico TrypLE胰蛋白酶替代物进行传代。
细胞鉴定:利用流式细胞术对小鼠骨髓MSCs的表面标志物CD29、CD105、CD90及CD45进行鉴定。
结果与讨论:经上述方法培养的MSCs在体外表现为典型的集落样生长,细胞形态为均一的长梭形,符合骨髓MSCs特征。光镜下显示,青年过表达和老年过表达小鼠干细胞表现出较正常对照组相似或更优的细胞形态,而青年沉默的小鼠干细胞形态更加趋于扁平,胞浆颗粒感增强(图2)。荧光显微镜下显示不同处理组干细胞(MiceY-Wisp1KO、MiceY-Wisp1OV以及MiceO-Wisp1OV)在同样的转染条件下(MOI=30,Polybrene=5μg/ml),慢病毒转染率均超过80%,且三组间无显著差异(图3)。流式细胞术结果显示,青年小鼠MSCs的免疫表型为:CD29(99.8±0.22)%;CD105(95.9±0.81)%,CD90(97.6±1.81)%,CD45(5.8±0.61)%;老年小鼠的免疫表型为:CD29(98.5±0.55)%;CD105(16.8±1.76)%,CD90(92.9±1.76)%,CD45(19.3±0.86)%。青年小鼠MSCs的免疫表型与老年小鼠MSCs存在显著差异,尤其在CD105和CD45的表达上(图4、图5),提示供体年龄会影响MSCs的免疫表型。
实施例2:小鼠骨髓MSCs中Wisp1基因的过表达和敲除构建
实验分组:青年小鼠MSCs细胞Wisp1敲除组(MSCsKO);中年小鼠MSCs细胞Wisp1敲除组(MSCsKO);青年小鼠MSCs细胞Wisp1过表达组(MSCsOV);中年小鼠MSCs细胞Wisp1过表达组(MSCsOV);老年小鼠MSCs细胞Wisp1过表达组(MSCsOV)。
细胞准备:将状态良好的小鼠骨髓MSCs细胞接种至24孔板中,细胞浓度调整为1×105/mL细胞,确保病毒感染时的细胞汇合率在50%-70%之间。。
病毒感染:
(1)预处理:细胞接种后,于37℃培养过夜,确保细胞密度约为50%。
(2)感染准备:准备含Polybrene的完全培养基,Polybrene的最终浓度为5μg/mL。移除旧培养基,加入0.5mL的Polybrene/培养基混合液至每孔中(适用于24孔板,其他规格孔板请按比例调整)。
(3)病毒添加:病毒从冰箱取出,在冰上解冻。移除部分培养基,加入1/2体积新鲜培养基,随后加入病毒原液,轻轻混匀。在37℃下进行小体积感染4小时,之后补充培养基至正常体积。(感染时培养基体积表格如下):
(4)感染后处理:感染后24小时,替换含病毒的培养液为新鲜完全培养液,继续在37℃下培养。48小时后,对带有GFP报告基因的病毒,通过荧光显微镜检查GFP的表达效率。对于携带Puromycin抗性基因的病毒,使用含有适当浓度Puromycin的新鲜完全培养液,进行稳定转导细胞的筛选。
实施例3:基因表达定量与样本相关性分析
测序与数据产出:测序交由华大基因(BGI,深圳,中国)完成,测序仪器为BGISEQ-500高通量测序仪。共24个样品,包括MiceY、MiceM、MiceO、MiceY-Wisp1OV、MiceY-Wisp1KO、MiceM-Wisp1OV、MiceM-Wisp1KO、MiceO-Wisp1OV8个组别,每个样品平均产出21.37M数据。样品与参考基因组的平均比对率达到94.36%,与参考基因集的比对率为88.76%。在此基础上,成功检测到18,440个基因。
基因表达定量与深度分析:对测序数据进行基因表达定量分析,并基于基因的表达水平进行了一系列的统计学分析,如主成分分析、样本间的相关性分析以及差异基因筛选。进一步,对筛选出的差异表达基因进行了GO功能显著性富集分析、KEGG路径显著性富集分析、基因聚类分析、蛋白质互作网络分析以及转录因子的相关分析。
样本间相关性分析:为了准确反映样本间的基因表达相关性,我们计算了所有样品间基因表达量的Pearson相关系数,并通过热图直观地展示了这些相关性数据。如图1所示,相关系数普遍高于0.8,其中大多数超过0.92,反映出各实验组样品间的基因表达模式具有很高的一致性。
实施例4:MiceY与MiceY-Wisp1KO组间的差异表达基因分析
在青年小鼠MSCs中,采用慢病毒介导的shRNA技术沉默了Wisp1基因。通过对比MiceY与MiceY-Wisp1KO两组,分析了差异表达基因。以MiceY组为对照,MiceY-Wisp1KO组显著下调的基因设置筛选条件如下:FPKM≥1(排除表达量过低的基因),Qvalue≤0.001,log2︱FC︱≥4。在GO富集中(图6),显著的20个GO通路涉及:与细胞外基质相关的通路,如Extracellular region,Extracellular region part,Extracellular matrix及Proteinaceous extracellular matrix·;与骨骼系统发育相关的通路,如Skeletalsystem development;与血管新生和血管发育相关的通路,如Angiogenesis,Blood vesseldevelopment及Vascsulature development;与细胞增殖和发育有关的通路,如Cellactivation,Multicellular organism development,Positive regulation of cellproliferation。以上结果提示,当Wisp1基因在青年小鼠的MSCs中被沉默后,与细胞增殖、发育及血管骨骼肌系统的相关基因表达发了显著变化,其中,大多数对细胞增殖、血管骨骼肌系统发育有益的基因都显著下调。
基于上述富集到的基因及其表达量,我们绘制了相关图表。在这些基因中,有几个关键基因尤为重要:Gas6能够促进细胞增殖;Figf和Stabl1都能够促进血管新生;而Excr4是Cxcl12(SDF-1)的配体(参见图7)。
在KEGG路径分类分析中,采用了FPKM≥1、Q值≤0.001和|FC|≥2作为筛选标准,对MiceY v与MiceY-Wisp1KO组间下调的DEGs进行了筛选。以MiceY组为对照,发现显著差异表达的基因主要涉及Cell growth and death,Immune system以及Development等与细胞增和、免疫调节相关的通路(图8)。值得注意的是,Cxcl12和Cxcr4两个基因都显示除了显著差异(图9)。
另外,在GO富集分析中,采用FPKM≥1,Qvalue≤0.001,︱FC︱≥2为标准,筛选差异表达基因。在MiceY与MiceY-Wisp1KO的比较中,我们发现了多个与细胞外基质和细胞增殖紧密相关的GO通路,如Cell activation、Intrinsic component of membrane、membranepart、Extracellular region part以及Attachment of spindle microtubules tokinetochore。
更为重要的是,我们还发现了与免疫系统相关的富集通路,例如Immune systemprocess(图10)。此通路中共有87个与免疫调节紧密相关的基因。值得注意的是,当Wisp1被沉默后,多个与免疫调节相关的基因,如CD28、CD4、C1qa、C1qb和Fcgr3等,其表达量都显著下降。同时,随着Wisp1的沉默,化学趋化因子Cxcl12(也称为SDF-1)的表达也呈现出显著的下降趋势。
实施例5:分析MiceY与MiceY-Wisp1OV组间基因表达差异
筛选MiceY和MiceY-Wisp1OV组上调的DEGs,以FPKM≥1(去除表达量过低的基因),Qvalue≤0.001,log2︱FC︱≥2为标准,得到293个基因,移除未在蛋白互作网络中形成相互作用的基因后,进一步进行了GO功能显著性富集分析、KEGG通路显著性富集分析以及聚类等深度分析。
在蛋白互作网络图(图11)中,观察到Wisp1基因的富集。与Wisp1直接相关的基因有3个:Col5a2,Mmp2,Mmp9。这些基因在Wisp1过表达后也表现出显著的上调趋势(图12)。特别地,Col5a2是细胞外基质(ECM)的关键组成部分。ECM是细胞微环境的主要组件,通常处于持续的合成与降解的动态平衡中。在ECM的降解中,基质金属蛋白酶是最关键的酶系,负责调节ECM的动态平衡。这些富集结果暗示Wisp1与细胞外基质的表达有紧密的联系。
基于蛋白网络互作,我们进行了GO注释功能分类分析。在MiceY与MiceY-Wisp1OV组间的显著差异中,我们鉴定了涉及细胞增殖、生长、发育、抗氧化等功能的子类别,如Cellproliferation、Growth、Developmental process和Antioxidant activity(图13)。在抗氧化通路中,主要富集了Nqo1和Sod2两个基因。Sod2是细胞内的关键抗氧化系统组成部分,而Nqo1是重要的抗氧化蛋白,都对维持细胞的氧化-抗氧化平衡起到关键作用。
在蛋白网络互作的基础上,进行GO注释功能分类分析,分子功能、细胞组分和生物过程这三大功能类别在青年小鼠MiceYvs.青年小鼠过表达MiceY-Wisp1OV组间显著差异中覆盖到的子类别包括:Cell proliferation,Growth,Developmental process,Antioxidant activity等与细胞增殖、发育、免疫调节及抗氧化相关的通路(图13)。在抗氧化功能通路(Antioxidant activity)中,我们主要鉴定了两个基因:Nqo1和Sod2。其中,Sod2,也称为超氧化物歧化酶2,主要存在于线粒体中,作为细胞内的核心抗氧化系统成员,其能够降低超氧阴离子等超氧化物的水平,从而维护细胞的氧化-抗氧化平衡。Nqo1是另一种关键的抗氧化蛋白,它可以减少自由基的生成并缓解氧化应激。值得注意的是,我们在Growth通路中也观察到了Wisp1基因的富集。此外,Cell proliferation、Growth和Antioxidant activity通路中富集的基因在蛋白质网络互作图(图14)以及基因表达量柱状图(图15)中都有详细展示。
在KEGG通路分析中,MiceY与MiceY-Wisp1OV组间的显著差异基因涉及了多个与细胞生长、衰老、发育和免疫调节紧密相关的通路,包括Cell growth and death、Immunesystem、Aging和Development(图16)。在Aging通路中,我们鉴定了Hspa1b、Hspa1a、Sod2和Mapk13等基因(图17)。特别地,Hspa1b(也称为HSP70)和Hspa1a均为热应激蛋白,它们对于外部应激因子具有保护性反应,同时也有助于维持细胞的结构完整性和正常的新陈代谢。随着衰老过程,这些蛋白的表达会逐渐降低。在Development通路中,关键基因Src被鉴定出来。由Src基因编码的蛋白在细胞生长、分化、迁移以及免疫反应的调节中起到了核心作用。而在Cell growth and death通路中,我们富集到了BCL2a1d、NGF和TNFaip3等关键基因。其中,NGF能够结合TRKA受体,进而驱动如BCL-2等基因的表达,促进细胞增殖和存活。TNFaip3则具有抑制NF-kappaB激活和抵抗TNF介导的细胞凋亡的功能。更进一步的蛋白互作网络分析显示,Hspa1b、Hspa1a和Sod2之间存在直接的相互作用。
实施例6:对MiceO与MiceO-Wisp1OV组间差异表达基因深入分析
在MiceO与MiceO-Wisp1OV组间,我们筛选了上调的差异表达基因(DEGs),采用以下标准:FPKM≥1,Qvalue≤0.001,以及log2|FC|≥2。在GO富集分析中(图18),我们观察到与免疫系统和免疫调节紧密相关的20个显著富集的GO通路,如Defense response、Immunesystem Process、Response to stress和Regulation of immune system process。
特别地,Defense response通路中鉴定的关键基因包括BMP2、Ang2(血管生成素)、Fcgr1、Fpr2、Vcam1、Cd40、Bst2(B细胞发育)、Slamf8和DHX58。而在Immune system process通路中,关键基因有Csf2、Bcl2a1a、Bcl2a1b、Bcl2a1c、Bcl2a1d、Fgf10、Hspa1a、Sod2和Vegfr。Response to stress通路中,Mapk13被鉴定为关键基因。在GO分类分析(图19)中,富集到了Antioxidant activity通路,其关键基因如Nqo1、Prdx1、Sod2和Prdx5,这些都是细胞内的核心抗氧化酶。进一步的KEGG分类分析(图20)显示,Aging通路中的关键基因包括HSPa1a、Sod2和Mapk13。值得注意的是,MiceO与MiceO-Wisp1OV组的DEGs与MiceY与MiceY-Wisp1OV组的DEGs有很大的重叠,显示出相似的趋势。
实施例7:评估小鼠骨髓MSCs衰老相关基因和端粒酶编码基因表达水平
为评价干细胞转染Wisp1过表达和沉默后对干细胞本身衰老的影响及端粒酶活性的影响,对衰老相关基因P16,P53,P21,以及编码端粒酶逆转录酶的基因TERT进行筛选。随着年龄增长,P16,P53,P21基因的表达量呈现逐渐升高的趋势,即从MiceY到MiceM再到MiceO逐渐升高(图21)。在Wisp1过表达后,P16和P53的MiceOvs.MiceO-Wisp1OV组呈现下降趋势(图22)。TERT的MiceYvs.MiceY-Wisp1OV组和MiceOvs.MiceO-Wisp1OV组都呈现上升趋势(图23、图24)。
实施例8:利用Q-PCR及Western blot技术验证小鼠骨髓MSCs转染后Wisp1表达水平Q-PCR、Western blot结果显示:青年、老年小鼠骨髓干细胞过表达Wisp1后,Wisp1较正常青年、老年小鼠骨髓MSCs显著升高。同时,转染条件能有效地使Wisp1基因过表达或沉默。青年小鼠骨髓干细胞通过慢病毒介导沉默Wisp1后,Wisp1较正常青年小鼠骨髓MSCs显著降低(图25、图26)和转录组测序结果一致。
实施例9:采用Q-PCR及Western Blot技术验证不同年龄小鼠骨髓MSCs的测序数据
Q-PCR以及WB结果显示,随着小鼠年龄的增长,骨髓MSCs衰老相关蛋白P16、P21及P53的表达逐渐增高,而与细胞增殖相关的因子PCNA、Survivin、CDK1;抗凋亡因子Bcl-2以及促进MSCs体内归巢的因子SDF-1逐渐降低。另外,在青年小鼠和老年小鼠Wisp1升高的同时,其SDF-1、Survivin以及Bcl-2也有明显上升,提示Wisp1过表达可能增强MSCs的归巢、增殖以及抗凋亡能力(图27、图28)。
实施例10:不同年龄骨髓MSCs多向分化潜能评价
青年、中年、老年小鼠骨髓MSCs及Wisp1转染后的细胞,在不同培养条件下对其分化能力进行评价:老年组间充质干细胞经诱导后向脂肪细胞、成骨细胞分化的能力发生不同程度下降,其中青年组成骨诱导可见大量红色深染的钙化结节,成脂诱导可见均匀分布的红色圆形脂滴,而相应的老年组钙化结节和脂滴都显著减少;而Wisp1过表达能够提高小鼠干细胞的成骨、成脂诱导分化能力;Wisp1沉默后小鼠的分化诱导能力也相应下降,提示供体年龄能够显著影响小鼠的诱导分化潜能(图29),而Wisp1能够提高衰老细胞的诱导分化能力。
实施例11:评估不同年龄小鼠骨髓MSCs内ROS和细胞死亡水平
在对小鼠骨髓间充质干细胞(MSCs)的ROS水平进行评估时,观察到随着小鼠年龄的增长,其MSCs中的ROS水平逐渐累积。具体地,青年、中年和老年小鼠骨髓MSCs的ROS水平分别为(32103.71±17961.88)、(102646.70±10435.12)和(317882.10±48421.46)。在这三个年龄段中,每两组之间的差异均达到了显著水平。当进行Wisp1转染后,发现青年过表达细胞的ROS水平(23465.10±3498.89)和老年过表达细胞的ROS水平(146677.70±34487.23)均显著降低。相反,青年沉默细胞的ROS水平(283586.90±18890.34)显著增加(图30)。在细胞死亡水平的评估中,采用了罗氏Cell Death Detection ELISA plus检测试剂盒,对不同年龄的小鼠干细胞以及转染后的过表达和沉默小鼠MSCs进行了检测。结果显示,随着小鼠年龄的增长,MSCs的细胞死亡率逐渐增加。具体地,青年、中年和老年小鼠骨髓MSCs的细胞死亡率分别为(0.18±0.02)、(0.372±0.15)和(1.198±0.03)。在这三个年龄段中,每两组之间的差异均为显著。经过Wisp1过表达后,青年小鼠的细胞死亡率(0.134±0.01)有所降低,而沉默后则显著上升至(0.405±0.02)(图31)。
实施例12:评估不同年龄小鼠骨髓MSCs端粒酶活性及端粒酶相对长度
在TeloTAGGGTMTelomerase PCR ELISAPLUS端粒酶活性的检测中,青年、中年和老年小鼠骨髓MSCs的端粒酶活性(450nm处吸光度值)分别为(0.71±0.32),(0.25±0.20)以及(0.18±0.08),随年龄增加不断下降,青年组分别与中年组、老年组比较有统计学意义。而在Wisp1转染后,老年过表达细胞(0.85±0.22)的端粒酶活性水平明显上升,而青年沉默细胞(0.10±0.07)的端粒酶活性水平显著下降(图32)。在Q-PCR端粒酶相对长度检测中,青年、中年和老年小鼠骨髓MSCs的端粒酶相对长度分别为(1.00±0.12),(3.18±0.08)以及(5.95±0.46),青年组分别和中年组、老年组以及中年组和老年组比较有显著差异,随年龄增加不断下降。在Wisp1转染后,青年过表达(10.18±0.29)和老年过表达细胞(7.96±2.12)的端粒酶相对长度都明显上升,而青年沉默细胞(1.11±0.21)的端粒酶相对长度显著下降(图33)。端粒酶作为细胞对DNA损伤反应的衰老指标可以反映小鼠骨髓MSCs的增龄性变化。同时,Wisp1能够显著改善供体年龄相关的端粒酶活性下降和端粒酶长度减少。
实施例13:基于实验结果的深入讨论
在本发明中,青年、中年、老年小鼠MSCs基因转录组测序分析发现Wisp1、SDF-1的表达随年龄增加呈显著下降趋势。当Wisp1基因在年轻小鼠的骨髓间充质干细胞(MSCs)中被沉默以后,细胞增殖、发育和血管骨骼肌系统的基因表达发生显著变化,其中大多数有利于细胞增殖、发育和血管骨骼肌系统发育的基因显著下调。同时,免疫调节相关的基因表达量显著下降,如CD28,CD4,C1qa,C1qb,Fcgr3等。免疫衰老可在系统水平上加重衰老表型,这源于免疫系统障碍导致其无法清除感染因子、感染细胞及癌前细胞。
在青年小鼠MSCs通过慢病毒转染过表达Wisp1基因,与细胞生长、衰老、发育、免疫调节等密切相关通路的基因表达发生了显著变化。如Aging(衰老)通路富集到的基因有Hspa1b,Hspa1a,Sod2,Mapk13。其中,Hspa1b(HSP70)和Hspa1a为热应激蛋白,对外界应激具有一种保护性作用,且有助于机体维持正常的新陈代谢和细胞结构的完整性,衰老导致其表达降低。Development(发育)通路富集到的关键基因有Src,该基因编码的蛋白对于调节细胞生长、分化、迁移和免疫反应中起着重要作用。Cell growth and death(细胞生长与死亡)通路富集到的关键基因有BCL2a1d,NGF,TNFaip3等,NGF可以通过结合TRKA受体来驱动Bcl-2等基因的表达,从而刺激细胞的增殖和存活。同时,随着年龄增长,P16,P53,P21基因的表达量呈现逐渐升高的趋势。在Wisp1过表达后,P16和P53的MiceOvs.MiceO-Wisp1OV组呈现下降趋势。TERT的MiceY vs.MiceY-Wisp1OV组和MiceOvs.MiceO-Wisp1OV组都呈现上升趋势。
测序结果的分析可以从转录组水平解释在本发明中观察到的不同年龄骨髓MSCs生物学特性的差异,以及年龄因素对其免疫表型、增殖、存活以及功能改变的影响。诸如细胞形态上,青年过表达和老年过表达小鼠干细胞表现出较正常青年小鼠和老年小鼠类似或更好的细胞形态,而青年沉默的小鼠干细胞形态更加趋于扁平,胞浆颗粒感增强;Wisp1过表达能够提高小鼠干细胞的成骨、成脂诱导分化能力;Wisp1沉默后小鼠的分化诱导能力也相应下降;青年过表达和老年过表达的ROS水平都有明显下降,而青年沉默的ROS水平显著上升;在细胞死亡水平的检测中也同样发现小鼠MSCs随年龄增长细胞死亡率高,Wisp1过表达后有所降低,沉默后显著上升。同样,不同年龄的小鼠干细胞其端粒酶相对活性和相对长度随年龄增加不断下降。而在Wisp1转染后,青年过表达和老年过表达的端粒酶活性水平都有明显上升,而青年沉默的端粒酶活性水平显著下降。以上结果均表明,随着年龄增长,骨髓间充质干细胞发生了相应的衰老变化,而Wisp1细胞转染能够显著改善老年MSCs的衰老表型。

2.基因Wisp1在制备调控MSCs抗衰老表型药物中的应用,其特征是:所述药物作用于Wisp1基因,所述Wisp1基因通过调控细胞增殖和发育过程,从而精准调控MSCs的衰老表型。
3.根据权利要求1所述的基因Wisp1在制备调控MSCs抗衰老表型药物中的应用,其特征是:所述药物使Wisp1基因过表达,从而有效改善老年MSCs的衰老表型。
4.根据权利要求1所述的基因Wisp1在制备调控MSCs抗衰老表型药物中的应用,其特征是:所述Wisp1基因能够调控SDF-1、Survivin、Bcl-2、Nqo1、Sod2以及其他免疫调节相关的基因表达,进一步影响MSCs的衰老表型。
5.根据权利要求1所述的基因Wisp1在制备调控MSCs抗衰老表型药物中的应用,其特征是:所述药物通过增强Wisp1基因表达,显著提升MSCs的成骨、成脂诱导分化潜能。
