Abstract
本发明公开了CD123靶向CAR‑NK细胞的制备及其应用,本发明设计并筛选获得了适用于CAR‑NK细胞制备的优选scFv分子,筛选可提升NK细胞扩增和活性的mIL‑15,通过高效转导体系和稳定冻存体系,可制备对AML以及其他CD123+肿瘤细胞具有显著杀伤活性的“现货型”CAR‑NK细胞,并在小鼠移植瘤模型中证实了抗肿瘤效应,有望为AML以及其他CD123+肿瘤的治疗提供一种更加有效的治疗产品。

技术领域
本发明属于生物技术领域,涉及CD123靶向CAR-NK细胞及其应用。
背景技术
急性髓系白血病(Acute Myelocytic Leukemia,AML)是成人最常见的急性白血病,我国年发病率约为1.62/10万,其中,复发或难治患者(r/r AML)约占10%。目前,AML治疗仍以化疗为主,但仅有不到1/3的患者能达到持续缓解。同种异体造血干细胞移植,是目前治愈AML的唯一手段,但老年患者往往无法耐受,同时,还存在移植物抗宿主病(Graft-Versus-Host Disease,GVHD)等风险。据统计,AML患者5年总生存率(Overall survival,OS)为28.3%,而60岁以上患者5年OS则低至3-8%;r/r AML患者的5年OS为10%,中位生存期为6个月。总之,r/r AML治疗的临床需求远未被满足,亟需研发更加有效的治疗药物。
NK细胞(Natural Killer,NK)作为人体抵御病毒和异常细胞的第一道防线,具有广谱抗癌性,能在第一时间做出反应,被誉为人体内的“宪兵”。早在20年前,NK细胞疗法就被认为是晚期白血病治疗的一种安全、有效的手段。近年来,随着体外扩增培养技术、抗体技术和基因转导技术的进步,CAR-NK细胞已经开始崭露头角。近年来,M.D.安德森癌症中心、Fate Therapeutics、Nkarta Therapeutics等相继公布了惊艳的临床研究数据。与CAR-T疗法相比,CAR-NK细胞具有以下优势:无主要组织相容性复合体(MajorHistocompatibility Complex,MHC)限制,可制备“货架”产品;不会引起细胞因子风暴和神经毒性;通过CAR依赖和非依赖等多机制发挥抗肿瘤效应,对肿瘤实行“全方位、立体式”杀伤;NK对肿瘤免疫抑制微环境的抵抗能力强等。基于上述优势,CAR-NK细胞具有巨大的潜能,有望成为突破实体瘤治疗瓶颈的一大利器。
CD123,又称白细胞介素3受体α链(Interleukin-3Receptorα,IL-3Rα),能特异识别并结合IL-3,介导细胞的增殖与存活。研究报道,高达80%左右的AML病人肿瘤细胞表达CD123。更为重要的是,CD123在白血病干细胞(leukemia Stem Cell,LSCs)和白血病起始细胞(Leukemia-Initiating Cells,LICs)上高表达,且在LSCs上的表达丰度显著高于其它肿瘤细胞。LSCs具有自我更新能力,表型类似于正常造血干细胞(Hematopoietic StemsCells,HSCs),是白血病复发的根源,且LSCs表达CD123与AML的复发、化疗药物耐受等密切相关。因此,靶向CD123有望根除LSC,实现AML“治愈”。与CAR-T相比,CAR-NK细胞体内存活周期较短,不引起长期骨髓抑制效应。因此,靶向CD123的CAR-NK细胞具有良好的成药性,有望为AML及CD123+肿瘤的治疗带来重大突破。逆转录病毒载体和慢病毒载体是目前用于CAR-T和CAR-NK细胞制备的最常见的病毒转导体系,但其对外周血来源NK细胞的基因转导效率低。
发明内容
为了解决现有技术中存在的技术问题,本发明提供了具体的实施方案。
本发明提供了一种靶向CD123的嵌合抗原受体,所述嵌合抗原受体包括N到C端的氨基酸序列,所述氨基酸序列包括靶向CD123的胞外抗体结合结构域CD123 scFv-1、或CD123 scFv-2。
所述CD123 scFv-1与SEQ ID NO:1所示的氨基酸序列具有至少90%以上的序列同一性。
所述CD123 scFv-2与SEQ ID NO:2所示的氨基酸序列具有至少90%以上的序列同一性。
术语“scFv”指一种融合蛋白,其包含至少一个包含轻链可变区的抗体片段和至少一个包含重链可变区的抗体片段,其中轻链可变区和重链可变区借助柔性短多肽接头连续地连接,并且能够表达为单链多肽,并且其中scFv保留衍生它的完整抗体的特异性。除非另外指出,否则如本文所用,scFv可以具有按任何顺序(例如,相对于多肽的N末端和C末端)的VL可变区和VH可变区,scFv可以包含VL-接头-VH或可以包含VH-接头-VL。
术语“同源的”或“同一性”指两个聚合物分子之间(例如,在两个核酸分子(如,两个DNA分子或两个RNA分子之间)或在两个多肽分子之间)的亚单位序列同一性。当这两个分子中的亚单位位置被相同单体性亚单位占据时,例如,若两个DNA分子的每个分子中的某位置被腺嘌呤占据,则它们在该位置是同源或同一的。两个序列之间的同一性直接随匹配位置或同源位置的数目而变化,例如,如果两个序列中一半位置(例如长度为10个亚单位的聚合物中的5个位置)是同源的,则这两个序列是50%同源的;如果90%的位置(例如10个位置中9个位置)是匹配或同源的,则这两个序列是90%同源的。

本发明提供了一种核酸分子,所述核酸分子包括编码前面所述的靶向CD123的嵌合抗原受体的核苷酸序列。
术语“核酸”、“多核苷酸”或“核酸分子”指脱氧核糖核酸(DNA)或核糖核酸(RNA),或其DNA或RNA的组合,及其单链形式或双链形式的聚合物。术语“核酸”包括基因、cDNA或mRNA。在一个实施方案中,核酸分子是合成的(例如化学合成的)或重组的。除非特别限制,否则该术语包括含有天然核苷酸的类似物或衍生物的核酸,所述核酸具有与参考核酸相似的结合特性并且以与天然存在核苷酸相似的方式代谢。除非另外说明,特定核酸序列也内在包括其保守方式修饰的变体(例如简并密码子置换)、等位基因、直向同源物、SNP和互补序列,以及明确指出的序列。

本文所用的术语“载体”是指可用于递送和/或表达核酸分子的任何载体。其可以是如本文所述的转移载体或表达载体。术语“慢病毒”指逆转录病毒科(Retroviridae)的一个属。慢病毒在逆转录病毒当中的独特之处在于能够感染非分裂性细胞;它们可以递送显著量的遗传信息至宿主细胞的DNA,从而它们是基因递送载体的最高效方法之一。

本发明提供了前面所述的嵌合抗原受体细胞在制备哺乳动物中提供抗表达CD123的肿瘤免疫的药物中的应用。

进一步,所述应用中嵌合抗原受体细胞与第二药剂或操作组合施用,所述第二药剂或操作选自化疗、靶向抗癌疗法、溶瘤药物、细胞毒性剂、细胞因子、外科操作、放射操作、共刺激分子的激动剂、免疫检查点分子的抑制剂、疫苗或基于第二CAR的免疫疗法中的一种或多种。

术语“一个”和“一种”指该冠词的一个或多于一个(即,指至少一个)的语法对象。例如,“一个要素”意指一个要素或多于一个要素。
术语“癌症”指异常细胞快速且失控生长为特征的疾病。癌细胞可以局部地或通过血流和淋巴系统扩散到身体其他部分。本文中描述了多种癌症的例子并且它们包括但不限于乳腺癌、前列腺癌、卵巢癌、宫颈癌、皮肤癌、胰腺癌、结直肠癌、肾癌、肝癌、脑癌、淋巴瘤、白血病、肺癌等。术语“肿瘤”和“癌症”在本文中互换地使用,例如,两种术语涵盖实体瘤和液体肿瘤,例如,弥散型或循环型肿瘤。如本文所用,术语“癌症”或“肿瘤”包括恶变前以及恶性癌症和肿瘤。
本发明提供了一种药物组合物,所述药物组合物包括治疗有效量的前面所述的嵌合抗原受体细胞。
进一步,所述药物组合物还包括以下一种或多种药剂组合施用:
A、改善与施用包含嵌合抗原受体核酸/多肽的细胞相关的一种或多种副作用的药剂;
B、增加包含嵌合抗原受体核酸/多肽的细胞的功效的药剂;或
C、治疗与CD123表达相关的疾病的药剂。
如本文所用的术语“药物组合”意指因混合或合并多于一种有效成分产生的产品并且包括有效成分的固定组合和非固定组合。术语“固定组合”意指有效成分,例如本发明的化合物和组合配偶体,均以单一实体或剂量同时地施用至患者。术语“非固定组合”意指有效成分。
术语“有效量”或“治疗有效量”在本文中互换地使用并且指化合物、制剂、材料或组合物如本文所述那样有效实现特定生物学结果的量。

本发明提供了制造嵌合抗原受体细胞的方法,所述方法包括用前面所述的嵌合抗原受体、前面所述的核酸分子、或前面所述的载体引入免疫效应细胞。
如该术语在本文中所用那样,术语“免疫效应细胞”指参与免疫应答,例如参与促进免疫效应子反应的细胞。免疫效应细胞的例子包括T细胞,例如,α/βT细胞和γ/δT细胞、B细胞、天然杀伤(NK)细胞、天然杀伤T(NKT)细胞、肥大细胞、和髓细胞衍生的吞噬细胞。
本发明提供了一种提升前面所述的嵌合抗原受体转导效率的方法,所述方法包括使用含2000U/mL的IL-2和100ng/ml的IL-15的X-VIVOTM15培养基重悬细胞,调整细胞密度至2.0×106细胞/ml,加入促转导试剂为0.1μM的BX795的步骤。
术语“转染”或“转化”或“转导”指借以将外源核酸转移或引入宿主细胞的过程。“转染”或“转化”或“转导的”细胞是已经用外源核酸转染、转化或转导的细胞。细胞包括原代主题细胞和其后代。
本发明的优点和有益效果:
本项目以此为基础,制备靶向CD123,并表达膜mIL-15的CAR-NK细胞,用于CD123+AML的治疗。

实施例1病毒的构建与制备
1、病毒中嵌合抗原受体结构的结构如图1所示,其中CAR的结构为:CD8信号肽+scFv+CD8 hinge+CD8 TM+4-1BB共刺激+CD3ζ。
2、载体构建:
合成靶向CD123的CAR序列,并将其克隆到慢病毒主质粒载体上。通过4质粒包装体系制备获得重组慢病毒JD-LV123-1、JD-LV123-2和JD-LV123-3。
3、各部件经测序鉴定正确,其序列如表1所示。

实施例2重组慢病毒的制备及滴度测定
1、实验方法
1)CAR分子构建到慢病毒主质粒载体后,采用四质粒包装系统,转导293T细胞后获得重组慢病毒JD-LV123-1、JD-LV123-2、JD-LV123-3。进一步经柱层析纯化、稀释后进行滴度检测。
2)收集K562细胞,使用RPMI1640培养基重悬细胞,使用12孔板进行铺板,K562铺板密度为5×105个cells/孔,1mL/孔。按照2倍梯度稀释法加入慢病毒浓缩液,分别为16μl/孔、8μl/孔、4μl/孔、2μl/孔、1μl/孔。感染4~6h后补液(含10% FBS的RPMI1640),1mL/孔。
3)感染72h后,使用流式细胞术进行转导效率检测。
4)根据阳性表达率计算慢病毒滴度。
病毒滴度计算公式如下TU/mL=(P×N/V)×1000,TU/mL为病毒滴度;P=阳性细胞比例(%),N=感染时的细胞数量;V=每孔加入慢病毒的体积;流式检测阳性率的可信范围为10~30%。
2、实验结果
流式检测结果如图2、图3所示,根据实验方法中给出的计算公式:
(1)CAR序列筛选中JD-LV123-1慢病毒滴度为4.9×107TU/mL;JD-LV123-2慢病毒滴度为5.7×107TU/mL。
(2)mIL-15序列确认中JD-LV123-1慢病毒滴度为1.84×108TU/mL;JD-LV123-3慢病毒滴度为2.92×107TU/mL。
实施例3CD123 scFv序列的筛选
1、实验方法
(1)通过分选获得外周血单个核细胞,分选当天诱导NK细胞。
所使用培养基的基础培养基是X-VIVOTM15培养基,诱导NK用培养基为含2000IU/ml的IL-2、100ng/mL的IL-15、100IU/mL的IL-12、5μg/ml的CD137和10%灭活血浆的培养基;
(2)第一次补液:第3天时,向培养基中补充1倍体积的NK培养基(含有5%的灭活血浆、2000IU/mL的IL-2、100ng/mL的IL-15、100IU/mL的IL-12);
(3)第二次补液:第5天时,向培养基中补充第一次补液后总体积1倍体积的NK培养基(含有5%的灭活血浆、2000IU/mL的IL-2、100ng/mL的IL-15、100IU/mL的IL-12);
(4)第7天NK细胞活化过程为:室温,1800rpm离心收集细胞,采用含2000U/mL的IL-2、100ng/mL的IL-15和100U/mL的IL-1β的X-VIVOTM15培养基重悬细胞,将其细胞密度调整为1×106细胞/mL,接种于T75或T175培养瓶中,于37℃、5% CO2、饱和湿度培养箱中继续培养2天。
(5)培养第9天:室温,1800rpm离心10min收集活化的NK细胞,采用含2000U/mL的IL-2和100ng/ml的IL-15的X-VIVOTM15培养基重悬细胞,加入促转导试剂BX795(0.1μM)。按照2.0ml/孔接种至6孔板,加入慢病毒JD-LV123-1、JD-LV123-2。
将其混匀后,放入培养箱中培养4h。然后补加2ml的新鲜完全培养基继续培养。
(6)培养24h后;取出,室温1800rpm离心10min收集细胞,离心收获沉淀;使用CARNK扩增培养基(含2%灭活血浆或5% HPL、2000U/mL IL-2、100ng IL-15、20ng/mL IL-18的X-VIVOTM15培养基)重悬细胞,按照1×10 6细胞/ml的密度进行扩增。
(7)培养至转导后第5天,检测NK细胞的转导效率。
(8)培养至转导后第12天检测细胞活率、细胞数量、细胞纯度、转导效率、杀伤作用。
2、实验结果
结果如图4、图5、图6所示,在相同MOI条件下,JD-LV123-1与JD-LV123-2,的转导效率无显著差异;病毒转导后,各组之间活率无显著影响,而NK细胞的增殖受到一定的抑制,但是,JD-LV123-1转导组的扩增能力强于JD-LV123-2转导组;JD-LV123-1转导的CAR-NK细胞,其对MOLM-13-Luc的杀伤活性高于JD-LV123-2转导组。因此,CD123 scFv-1的结构是制备CAR123-NK细胞的更优结构。
实施例4表达mIL-15对CAR-NK细胞抗肿瘤活性的影响
A、实验结果
1.CAR-NK细胞的制备与检测
(1)通过分选获得外周血单个核细胞,分选当天诱导NK细胞。
所使用培养基的基础培养基是X-VIVOTM15培养基,诱导NK用培养基为含2000IU/ml的IL-2、100ng/mL的IL-15、100IU/mL的IL-12、5μg/ml的CD137和10%灭活血浆的培养基;
(2)第一次补液:第3天时,向培养基中补充1倍体积的NK培养基(含有5%的灭活血浆、2000IU/mL的IL-2、100ng/mL的IL-15、100IU/mL的IL-12);
(3)第二次补液:第5天时,向培养基中补充第一次补液后总体积1倍体积的NK培养基(含有5%的灭活血浆、2000IU/mL的IL-2、100ng/mL的IL-15、100IU/mL的IL-12);
(4)第7天NK细胞活化过程为:室温,1800rpm离心收集细胞,采用含2000U/mL的IL-2、100ng/mL的IL-15和100U/mL的IL-1β的X-VIVOTM15培养基重悬细胞,将其细胞密度调整为1×106细胞/mL,接种于T75或T175培养瓶中,于37℃、5% CO2、饱和湿度培养箱中继续培养2天。
(5)第9天:室温,1800rpm离心10min收集活化的NK细胞,采用含2000U/mL的IL-2和100ng/ml的IL-15的X-VIVOTM15培养基重悬细胞,加入促转导试剂BX795。按照2.0ml/孔接种至6孔板,加入慢病毒JD-LV123-1、JD-LV123-3。
将其混匀后,放入培养箱中培养4h。然后补加2ml的新鲜完全培养基继续培养。
(6)培养24h后;取出,室温1800rpm离心10min收集细胞,离心收获沉淀;使用CAR-NK扩增培养基(含2%灭活血浆或5% HPL、2000U/mL IL-2、100ng IL-15、20ng/mL IL-18的X-VIVOTM15培养基)重悬细胞,按照1×10 6细胞/ml的密度进行扩增,每两天补原有培养基的1-2倍。
(7)培养至转导后第5天,检测NK细胞的转导效率。
(8)转导后第12天检测细胞活率、细胞数量、细胞纯度、转导效率、杀伤作用,并冻存。
(9)冻存:
1)750g,10min,室温离心,收集培养至第12天的CAR-NK细胞。
2)弃上清,100ml 0.9%氯化钠注射液重悬细胞,750g,10min室温离心。
3)弃上清,100ml 0.9%氯化钠注射液重悬细胞,细胞计数。
4)将过筛后的细胞750g,10min室温离心,获得沉淀。
5)用适量预冷的冻存液重悬细胞,AOPI染色并细胞计数,调整密度至5×107cells/mL。将细胞/冻存液混合悬液以1.5ml/管灌装到细胞冻存管中。将冻存管放入到(2-8℃)预冷的异丙醇梯度降温盒中。-80℃条件下异丙醇降温盒放置18-24小时后,将冻存细胞取出并尽快转移到液氮中保存。
2.CAR-NK细胞体外活性评价:
(1)分别采用流式细胞术分析四株AML细胞系Molm-13-Luc(95.98%)、THP-1-Luc(99.05%)、KG-1a(46.99%)及HL-60(0.76%)表面CD123抗原的表达水平,如图9所示,其中,HL-60为阴性对照。
(2)靶细胞标记及效应细胞准备
1)收集对数生长期的靶细胞,500g、5min、室温离心收细胞,弃上清,用RPMI1640基础培养基重悬细胞。台盼蓝染色,记录活细胞数、细胞活率,将靶细胞调整密度为1×106cells/ml。
2)加浓度为1mM的Calcein-AM溶液(7.5uL/mL),避光孵育30min。染色结束后,加入2倍体积的PBS洗两次,500g、5min、室温,加RPMI1640基础培养基重悬细胞。
3)台盼蓝染色,记录活细胞数、细胞活率,调整细胞密度为2×105cells/ml,备用。
4)取足量的效应细胞,500g、10min、室温离心收细胞,弃上清,用RPMI1640基础培养基重悬细胞。按效靶比(2:1、5:1),将效应细胞稀释至相应浓度,备用。
(3)靶细胞与效应细胞共孵育
按照效靶比分别将效应细胞与靶细胞铺至96孔板中,对照组为未感染病毒的NK细胞。同时设置自发孔(只有靶细胞)及最大孔(靶细胞和最大释放剂)。37℃、5% CO2,孵育4小时。孵育3小时的时候最大孔加入释放剂(0.8%Triton),50μL/孔。
(4)收样上机检测
1)孵育结束后,500g、5min、室温离心96孔板,取上清每孔取150μL,酶标仪激发光:485/20、发射光:528/20,检测荧光强度。
2)计算公式:杀伤效率(%)=(实验孔-自发孔)/(最大孔-自发孔)×100%。
3.CAR-NK细胞体内抗肿瘤效应评价:
(1)动物建模:
1)在NCG小鼠体内建立THP-1-Luc人急性单核细胞白血病动物模型,于小鼠尾静脉接种THP-1-Luc细胞悬液200μL,5×105cells/mL细胞浓度。
2)待肿瘤信号值达到1.0E+06时,按肿瘤信号值及体重随机分为5组,每组8只。
(2)供试品准备:
采用流式细胞术对冻存复苏后KA071批次CAR123-NK细胞检测CAR123表达情况。
(3)细胞复苏当天,采用Calcein AM法检测CAR123-NK细胞及对照组NK细胞对THP-1-Luc的杀伤效应。
(4)分组当天开始给药,给药细胞为KA071批次人原代NK细胞转导后冻存复苏的CAR123-NK细胞,给药剂量为1E+07/只,每周给药3次,共给药4次,给药途径为尾静脉。给药期间,每周2次体内成像及体重。
B、实验结果
1、CAR-NK细胞的制备与检测
(1)表达CAR123的同时,通过P2A连接的方式表达mIL-15,不影响NK细胞的转导效率,均能达到40%以上;不影响NK和CAR-NK细胞的活率和增殖能力(JD-LV123-1和JD-LV123-3之间无显著差异),其结果如图7、图8所示。
(2)体外杀伤评价结果显示,JD-LV123-1与JD-LV123-3转导可显著提升NK细胞(CAR-NK细胞)对CD123+的AML细胞的杀伤效果(图10)。
(3)CAR-NK细胞复苏后,CAR分子的表达(图11)与冻存前(图7)相比无显著变化。如图12、图13、图14所示,CAR-NK细胞的杀伤活性无显著改变,体内抗肿瘤效应评价结果显示,NK细胞可降低小鼠肿瘤负荷(荧光值),而JD-LV123-1、JD-LV123-3转导的CAR-NK细胞对肿瘤负荷的抑制作用更加显著,与Vehicle组和NK组相比具有显著统计学差异(p<0.05),同时,小鼠的体重并未被影响,其结果如图15所示。表明,CAR-NK细胞具有显著抗THP-1肿瘤作用。
更为重要的是,在小鼠生存期指标上,JD-LV123-3显示出显著延长荷瘤鼠生存期作用,表明mIL-15表达有助于增强NK细胞的体内存活并增强抗肿瘤效应(图16)。
上述实施例的说明只是用于理解本发明的方法及其核心思想。应当指出,对于本领域的普通技术人员来说,在不脱离本发明原理的前提下,还可以对本发明进行若干改进和修饰,这些改进和修饰也将落入本发明权利要求的保护范围内。

1.一种靶向CD123的嵌合抗原受体,所述嵌合抗原受体包括N到C端的氨基酸序列,所述氨基酸序列包括靶向CD123的胞外抗体结合结构域CD123scFv-1、或CD123 scFv-2;
所述CD123 scFv-1与SEQ ID NO:1所示的氨基酸序列具有至少90%以上的序列同一性;
所述CD123 scFv-2与SEQ ID NO:2所示的氨基酸序列具有至少90%以上的序列同一性。

优选地,所述应用中嵌合抗原受体细胞与第二药剂或操作组合施用,所述第二药剂或操作选自化疗、靶向抗癌疗法、溶瘤药物、细胞毒性剂、细胞因子、外科操作、放射操作、共刺激分子的激动剂、免疫检查点分子的抑制剂、疫苗或基于第二CAR的免疫疗法中的一种或多种。
7.权利要求5所述的嵌合抗原受体细胞在制备哺乳动物中治疗患有与表达CD123相关的疾病的药物中的应用;
优选地,所述哺乳动物包括人或非人哺乳动物;
优选地,所述应用中与表达CD123相关的疾病是:
A、癌前病变疾病,
B、癌症或恶性肿瘤,或
C、与表达CD123相关的非癌症相关适应症;
优选地,所述疾病选自以下一种或多种:缩写为AML的急性髓性白血病、缩写为ALL的急性淋巴母细胞性白血病、B细胞幼淋巴细胞性白血病、慢性淋巴细胞性白血病、缩写为CML的慢性髓性白血病、毛细胞白血病、霍奇金淋巴瘤、组织细胞疾病、肥大细胞疾病、脊髓发育不良、脊髓发育不良综合征、骨髓增生性肿瘤、浆细胞性骨髓瘤、浆细胞样树突细胞肿瘤或它们的组合;
优选地,所述疾病为缩写为AML的急性髓性白血病。
8.一种药物组合物,所述药物组合物包括治疗有效量的权利要求5所述的嵌合抗原受体细胞;
优选地,所述药物组合物还包括以下一种或多种药剂组合施用:
A、改善与施用包含嵌合抗原受体核酸/多肽的细胞相关的一种或多种副作用的药剂;
B、增加包含嵌合抗原受体核酸/多肽的细胞的功效的药剂;或
C、治疗与CD123表达相关的疾病的药剂。
9.制造嵌合抗原受体细胞的方法,所述方法包括用权利要求1或2所述的嵌合抗原受体、权利要求3所述的核酸分子、或权利要求4所述的载体引入免疫效应细胞。
10.一种提升权利要求5所述的嵌合抗原受体转导效率的方法,所述方法包括使用含2000U/mL的IL-2和100ng/ml的IL-15的X-VIVOTM15培养基重悬细胞,调整细胞密度至2.0×106细胞/ml,加入促转导试剂为0.1μM的BX795的步骤。
