技术领域
本发明涉及新型毛乳头细胞分离和培养方法。具体地,本发明的分离和培养方法通过在低氧条件下培养从头皮分离的毛乳头组织(dermal papilla)来使从毛乳头组织脱离的毛乳头细胞迅速附着在培养板,从而具有缩短毛乳头细胞的确立(获得)时间的效果,分离及培养的毛乳头细胞具有免疫相容性。
背景技术
人类的头发为约100000根单独的毛发的集合体,毛发通过毛囊(hair follicle)生成。毛囊起到能够生成构成毛囊自身、上皮及皮脂腺所需的所有细胞株的干细胞的储存场作用。毛囊的基底部存在毛囊球(hair follicle bulb),毛囊球中增殖的基质细胞(matrix cells)成为毛发。毛囊球包括真皮鞘帽(dermal sheath cup;DSC)细胞及毛乳头细胞(dermal papilla cell;DPC)。毛乳头细胞为位于毛囊的基底部的间充质来源的成纤维细胞(mesenchymally-derived fibroblasts),为承担毛发的产生及生长的核心细胞。美国授权专利公报第8227244号公开了从毛发分离毛乳头细胞的方法。
毛发经过生长期(anagen)、退化期(catagen)及休止期(telogen)三阶段的周期来生长、保持及脱落。在成人的情况下,已知作为毛发生长期间的生长期每天平均生长0.3mm左右的毛发,一个月生长1cm左右,通常持续3年至7年。通常在生长期后,10天至14天期间发生全面的毛囊细胞的细胞凋亡(apoptosis),经过作为毛囊收缩阶段的退化期、平均3个月的为下个生长期做准备的阶段的休止期后毛发脱落,之所以毛发的长度随部位的不同而不同,是因为各部位的毛囊所固有特征的生长期的持续时间互不相同。
如上所述,正因为人的毛发具有规定的生长周期(hair growth cycle),因此可以保持一定数量的毛发。然而,若发生脱发,则存在于毛根的毛乳头细胞变小,若毛乳头细胞变小,则头发粗度变小,同时毛发的生长周期也变短。因此,随着脱发的发展,头发变得很细,毛发生长周期变得更短,头发生长一点即脱落。
已知脱发的原因是因遗传原因、除雄性激素的作用以外的内分泌疾病、营养不良、药物使用、生育、发热、手术等严重的身体、精神压力等因素的复合作用。近来,不仅是男性脱发,由于饮食生活的变化、社会环境等压力的增加,女性的脱发人数也呈增长的趋势,发病年龄降低。
现在韩国国内常用的脱发治疗剂有非那雄胺(finasteride;商品名:)、度他雄胺(dutasteride;商品名:/>)、米诺地尔(minoxidil;商品名:/>或)等。然而,上述治疗剂表现出性欲减退、勃起功能障碍、支配及执行能力丧失、涂敷部位瘙痒、红斑、皮肤刺激、眼的刺激等副作用,还在头部以外的其他身体部位观察到所不希望的毛发的生长。并且,近来以重症脱发患者为对象尝试毛发移植手术,但却因高昂的费用及术后副作用的局限而备受指责。
另一方面,除直接向皮肤涂敷药物的方式以外,作为脱发的治疗方法,还尝试在生物体外培养毛囊细胞来移植的技术,但将毛乳头细胞用作脱发治疗剂仍存在多个难点。首先,难以从头皮分离,培养条件苛刻,而且难以培养充分数量的细胞,不仅如此,若多次传代培养,则具有毛发再生能力显著降低的限制。概括标准的毛乳头细胞的分离过程(参照图1,Archives of Dermatological Research301(5):357-65)如下:从头皮组织分离毛球(hairbulb)后,分离存在毛乳头细胞多的毛乳头组织的步骤;(a→b),从毛乳头组织使毛乳头细胞附着(attachment)在培养板的步骤;(b→c),传代培养的步骤(c→d)。可以从通过此获得的毛乳头细胞分离细胞治疗剂或利用细胞的外泌体等。其中,使毛乳头细胞从毛乳头组织附着到培养板的时间平均需要12天以上,引起通过毛乳头细胞的治疗剂生产时间及生产成本上升的问题,因此,细胞治疗剂开发中急需从毛乳头组织分离并附着的毛乳头细胞的附着率的增加和附着时间的减少。
于是,毛乳头细胞的分离及培养中,本发明人发现在从毛乳头组织中确立毛乳头细胞的过程中通过低氧(0.5~5%的氧饱和度)培养可以增加毛乳头细胞在培养板的附着并缩短毛乳头细胞的确立时间的新型分离毛乳头细胞的方法,从而完成本发明。

技术方案
本发明提供毛乳头细胞分离和培养方法,包括在低氧条件下培养从头皮分离的毛乳头组织来使毛乳头细胞附着在培养板的步骤。
本发明的低氧条件可具有0.5%至5%的氧饱和度,优选地,可具有2%的氧饱和度。
与在正常条件下培养上述毛乳头组织相比,本发明的毛乳头细胞分离和培养方法可以使毛乳头细胞附着在培养板所需的时间缩短5天以上。
根据本发明分离及培养的毛乳头细胞可以为免疫相容性毛乳头细胞,上述免疫相容性毛乳头细胞可以抑制与免疫反应相关的选自由HLA-A、HLA-B、CD55、IL-1RA及CCL2组成的组中的一种以上信使核糖核酸(mRNA)的表达。
本发明提供根据毛乳头细胞分离和培养方法获得的毛乳头细胞,上述方法包括在低氧条件下培养从头皮分离的毛乳头组织来使毛乳头细胞附着在培养板的步骤,上述毛乳头细胞可以为免疫相容性毛乳头细胞。
本发明提供包含根据毛乳头细胞分离和培养方法获得的毛乳头细胞的用于预防或治疗脱发的药物组合物,上述方法包括在低氧条件下培养从头皮分离的毛乳头组织来使毛乳头细胞附着在培养板的步骤。
本发明提供包含从根据毛乳头细胞分离和培养方法获得的毛乳头细胞获得的细胞培养液或外泌体的用于预防或治疗脱发的药物组合物,上述方法包括在低氧条件下培养从头皮分离的毛乳头组织来使毛乳头细胞附着在培养板的步骤。
发明的效果
根据本发明的新型毛乳头细胞分离和培养方法,在从头皮组织分离毛乳头细胞的过程中,在低氧条件(0.5~5%,优选地,2%的氧饱和度)培养毛乳头组织时,使从毛乳头组织脱离的毛乳头细胞迅速附着(attachment)到培养板,从而可以迅速确立毛乳头细胞的原代细胞(primary cell)。分离毛乳头细胞的原代细胞后,可以进行通过传代培养的毛乳头细胞的大量生产,从而可以缩短细胞治疗剂的生产时间。并且,可以确保脱发治疗过程中所需的充分的毛乳头细胞,从而可以在脱发治疗中有效使用。
并且,根据本发明分离及培养的毛乳头细胞具有免疫相容性,减少毛乳头细胞的同种移植时可能发生的免疫反应,从而可以在通过移植毛乳头细胞的脱发治疗中使用。

图1示出标准的从头皮分离毛乳头细胞的过程(Archives of DermatologicalResearch 301(5):357-65)。
图2示出在正常条件(20%的氧饱和度)及低氧条件(2%的氧饱和度)下从毛乳头组织脱离的毛乳头细胞附着到培养板的水平。

具体实施方式
以下,参照附图详细说明本申请的实施方式及实施例,以便于本发明所属技术领域的普通技术人员可以轻松实施本发明。但本申请可以实现为多种形态,不限定于在此说明的实施方式及实施例。
在本说明书全文中,当提及某部分“包含”某结构要素时,若无特别反对的记载,则表示还可以包含其他结构要素,而不是排除其他结构要素。
本发明提供新型毛乳头细胞分离和培养方法。
本发明中的“毛乳头细胞(dermal papilla cell;DPC)”为位于毛囊基底部的间充质来源的成纤维细胞(mesenchymally-derived fibroblasts),是承担毛发的发生和生长的核心细胞。
本发明中使用的术语“脱发”是指与病因无关地在本应正常存在毛发的部位没有毛发的状态,可以为斑秃、遗传性雄激素性脱发、休止期脱发、创伤性脱发、拔毛癖、压力性脱发、生长期脱发、糠秕样脱发、梅毒性脱发、脂溢性脱发、症状性脱发、瘢痕性脱发或先天性脱发,但不限定于此。
本发明中使用的术语“治疗”是指通过给药组合物延迟或停止脱发进程来使脱发症状好转,或者使毛发变长或数量增多等促进生发及育发等的使脱发症状变更为痊愈的所有行为。
本发明中使用的“流式细胞仪(flow cytometry)”是为分析细胞或粒子的特性而广泛使用的基于激光的技术,可以测定细胞的大小、复杂度、细胞数量、细胞周期等。可以向所要测定的细胞流动的流体中照射激光并使细胞通过,或者也可以检测散射的光线并分析来在某集团中仅分离特定细胞。
以下,通过实施例更为详细地说明本发明,但下述实施例仅用于说明的目的,而不是限定本发明的范畴。
[实施例1]
组织的分离
将20mL的包含4%胎牛血清(FBS)的分离/培养基放入灭菌的100mm培养皿(petridish)底面上来浸泡从捐赠者采集的头皮组织,以不至头皮组织干燥的方式保管。将组织放到培养皿底面后利用一次性手术刀切为条(strip)形状以不使毛囊折叠,在保管组织的装有分利用培养基的培养皿中保管切开的组织条。将切开的组织条放到新的培养皿底面后,利用一次性手术刀逐个分离毛囊并放入装有新的培养基的培养皿中保管。
[实施例2]
从毛球中分离毛乳头组织
将150mm培养皿放到实体显微镜的载物台并放上1个上述实施例1中分离的毛囊后,为不至干燥,使用1mL注射器(syringe)滴入一滴分离用培养基。利用一次性手术刀切开毛囊的真皮鞘帽(DSC;dermal sheath cup)与上部真皮鞘(UDS;upper dermal sheath)的界限。补充一滴分离用培养基使切开的真皮鞘帽不至干燥后,利用两个1mL注射器,一个固定真皮鞘帽,另一个轻轻敲击真皮鞘帽最尖端部分的帽的底部使帽成为翻过来的形态。若从翻过来的真皮鞘帽的内部伸出长长的菱形或水滴形状的毛乳头组织(dermal papilla,DP),则利用注射器只切出毛乳头组织。
[实施例3]
毛乳头细胞从毛乳头组织向培养板附着
将上述实施例2中分离的毛乳头组织(DP)放到注射器针头端部来移到6孔(well)培养板(CellBind表面细胞培养皿(CellBind surface))。每孔接种12个毛乳头组织,以不使培养板晃动的方式放入37℃的正常条件(5%的二氧化碳(CO2)∶21%的氧气(O2))及低氧条件(5%的二氧化碳∶2%的氧气)培养器中培养5天至10天后,2~3天更换一次培养基,更换2次(p0)直至细胞汇合(confluency)达60%。分离(p0)及至5次传代(p5)的培养基使用CellCorTM(XCELL therapeutics公司)或毛囊真皮乳头细胞生长培养基(Follicledermalpapilla cell growth medium)(Promaocell公司)/4%的胎牛血清(Fetal bovine serum)(Hyclone公司)/1%的青霉素链霉素(penicillin streptomycin)(赛默飞世尔公司(Thermo Fisher scientific))。
[实施例4]
毛乳头细胞的传代培养
当上述实施例3中接种的从毛乳头组织脱离的毛乳头细胞在每孔内满至60%左右时,使用T75烧瓶(flask)进行传代培养。扔掉6孔培养板的培养基并使用1x DPBS洗涤一次后,向每孔加入0.5mL的细胞消化液(Accutase)并在各条件的培养器中反应10分钟。然后,向每孔分注2mL的分离用培养基并使用50mL的试管回收全部后,在常温及1300rpm的条件下离心分离3分钟来去除上清液。使用1mL的培养培养基使留在下层的细胞悬浮后,通过台盼蓝(trypan blue)染色来计数细胞数量。根据收集的细胞数量确定所要传代培养的T75烧瓶的个数(2.5×105cell/Flask)。向T75烧瓶放入15mL新的培养培养基并计算悬浮的细胞溶液来分注后,小心地旋转烧瓶3次以上来使细胞均匀地混合。然后,在正常条件(5%的二氧化碳∶21%的氧气)及低氧条件(5%的二氧化碳∶2%的氧气)的培养器中培养4天后,以4天为间隔以1∶5的比例进行从一次传代到5次传代(p1~p5)的传代培养。
[实验例1]
比较毛乳头细胞的培养板附着水平
以与上述实施例3相同的方式在低氧条件(5%的二氧化碳∶2%的氧气)和正常条件(5%的二氧化碳∶21%的氧气)下培养从头皮分离的毛乳头组织后,比较第5~10天的毛乳头细胞的培养板附着水平,结果如图2所示。
如图2所示,比较培养后第6天的附着率的结果,确认到与正常条件相比,在低氧条件下培养的附着于培养板的毛乳头细胞的数量大幅增加。这表示将正常条件(20%的氧饱和度)下平均经12天以上的毛乳头细胞的原代细胞的确立(获得)时间平均缩短5天左右的结果,在传代培养时还可以缩短追加的生产工序。
[实验例2]
分析毛乳头细胞的附着机制
如在上述实验例1中确认的,可知根据本发明分离及培养的毛乳头细胞对培养板的附着水平增加。于是,为了确认上述毛乳头细胞附着到培养板的机制,进行如下分析。
2-1.分析碱基序列(NGS)及本体(Ontology)
分析本发明的在低氧条件下分离及培养的毛乳头细胞的基因的碱基序列及本体。结果,如图3所示,确认到低氧条件给细胞外基质(ECM)蛋白带来影响,确认到增加其中的Col18A1、Col13A1及Col23A1的胶原蛋白表达量。上述胶原蛋白给毛乳头细胞的活性带来影响(参照TGF-β2and collagen play pivotal roles in the spheroid formation andantiaging of human dermal papilla cells.Kim H,Choi N,Kim DY,Kim SY,Song SY,Sung JH.Aging(Albany NY).2021Aug17;13(16):19978 19995.),通过此可知在低氧条件下得到增加的上述胶原蛋白的表达给毛乳头细胞向培养板的附着带来影响。
2-2.分析细胞外基质及附着分子的信使核糖核酸表达
分析本发明的在低氧条件分离及培养的毛乳头细胞的细胞外基质及附着分子的信使核糖核酸表达水平。结果,如图4所示,确认到多种基因中的MMP1、MMP3、MMP11、MMP12及LAMA1的表达减少。尤其,基质金属蛋白酶(MMP,matrix metalloproteinase)为分解胶原蛋白的酶,通过此可知由低氧条件减少的MMP表达抑制胶原蛋白的分解来给毛乳头细胞向培养板的附着带来影响。
[实验例3]
评估毛乳头细胞分离及工序缩短时间
为了评估根据本发明分离及培养的毛乳头细胞的确立(获得)时间,进行下述实验。
将从毛囊分离的毛乳头组织(DP)接种到培养板并在低氧条件(5%的二氧化碳∶2%的氧气)及正常条件(5%的二氧化碳∶21%的氧气)下培养后,在3次传代中使用细胞原液储存(填充)。将6孔培养板中每孔接种的总毛乳头组织中附着80%以上所需的时间设为“分离→附着”,细胞密度到达1孔的60%以上来进行一次传代培养所需的时间设为“分离→一次传代”,将一次传代培养后至细胞密度到达80%以上来进行二次传代培养所需的时间设为“分离→二次传代”,将二次传代培养后至细胞密度到达80%以上来进行三次传代培养所需的时间设为“分离→三次传代”,将三次传代培养后至使用原液填充的时间设为“分离→填充”,以此来评估所需时间。结果如下述表1所示。

如上述表1所示,测定从毛乳头组织被分离至被填充所需的时间的结果,确认到正常条件下培养需要41天,而在低氧条件下培养需要26天,缩短约15天。并且,从分离的毛乳头组织到毛乳头细胞附着所需的时间,在低氧条件下培养时,也缩短5天,示出显著缩短。
因此,在毛乳头细胞的确立(获得)中,与在正常条件下培养分离的毛乳头组织的情况相比,在低氧条件下培养时需要显著缩短的时间,可知到最终填充时为止缩短15天以上。
[实验例4]
评估毛乳头细胞标记物的表达
为了确认根据本发明分离及培养的细胞是否为毛乳头细胞,利用毛乳头细胞的表面抗原结合的抗体通过流式细胞仪进行分析。
使用DPBS洗涤裂解在培养液中的各传代的毛乳头细胞2次。每1×106个细胞使用500μl的细胞染色缓冲液(cell staining buffer)(包含1%的牛血清白蛋白的DPBS(1%BSA in DPBS))使细胞再悬浮后,每个试验组试管中分注100μl的细胞悬浮液。向各试验组分注1μl的相关抗体后,在4℃的温度下遮光反应15分钟。使用DPBS洗涤利用抗体染色的试验组1次后,使用100μl的DPBS使细胞再悬浮并通过流式细胞仪进行分析,结果如下述表2所示。
与各试验组相关的抗体如下:同型抗体对照组(APC小鼠IgG1_κ同型对照抗体(APCMouse IgG1_κIsotype Ctrl Antibody))、表达评估染色组(毛乳头细胞确认用:APC抗人CD34抗体(APC anti-human CD34 Antibody)、APC抗人CD45抗体(APC anti-human CD45Antibody))、纯度评估染色组(APC抗人CD44抗体(APC anti-human CD44 Antibody)、APC抗人CD90抗体(APCanti-human CD90 Antibody))。
在对照组的分布率小于0.1%的门(gate)中,在确认用表达评估染色组的分布率为90%以上且纯度评估染色组的分布率小于10%的情况下,评估为适合。

如上述表2所示,在各传代(p)的毛乳头细胞中评估标记物表达的结果,所有传代的毛乳头细胞中纯度用抗体表达小于1%,确认用抗体表达为90%以上。因此,确认根据本发明分离及培养的细胞为毛乳头细胞。
[实验例5]
评估毛乳头细胞的免疫反应活性基因表达
为了评估根据本发明分离及培养的细胞的免疫反应活性相关基因的表达程度,进行如下实验。
在20多岁及30多岁的男性捐献者的毛囊(毛乳头组织)中分离毛乳头细胞并在低氧条件(5%的二氧化碳∶2%的氧气)和正常条件(5%的二氧化碳∶20%的氧气)下培养。三次传代培养后提取培养的毛乳头细胞的核糖核酸来分析与T细胞的免疫反应活性相关基因的信使核糖核酸表达水平,结果如图5所示。使用皮肤成纤维细胞(信使核糖核酸表达水平:1)作为对照组。
如图5所示,通过毛乳头细胞的核糖核酸分析免疫反应活性相关基因的表达程度的结果,确认到与正常条件相比,在低氧条件下分离及培养毛乳头细胞时,HLA-A、HLA-B、CD55、IL-1RA及CCL2的表达显著减少。尤其,HLA-A、HLA-B及CD55的表达显著减少,可知在低氧条件下分离及培养的毛乳头细胞可以通过此减少因T细胞引起的免疫反应活性。
因此,可知根据本发明在低氧条件下分离及培养毛乳头细胞时,可以通过提高毛乳头细胞的免疫特权,即,免疫调节(immune modulation)功能来减少毛乳头细胞的同种移植时可能发生的免疫反应。最终,可知根据本发明分离及培养的毛乳头细胞具有免疫相容性。
Claims (7)
1.一种毛乳头细胞分离和培养方法,其特征在于,包括在低氧条件下培养从头皮分离的毛乳头组织来使毛乳头细胞附着在培养板的步骤。
2.根据权利要求1所述的毛乳头细胞分离和培养方法,其特征在于,上述低氧条件具有0.5%至5%的氧饱和度。
3.根据权利要求1所述的毛乳头细胞分离和培养方法,其特征在于,上述低氧条件具有2%的氧饱和度。
4.根据权利要求1所述的毛乳头细胞分离和培养方法,其特征在于,与在正常条件下培养上述毛乳头组织相比,使毛乳头细胞附着在培养板所需的时间缩短5天以上。
5.根据权利要求1所述的毛乳头细胞分离和培养方法,其特征在于,上述毛乳头细胞为免疫相容性毛乳头细胞。
6.根据权利要求5所述的毛乳头细胞分离和培养方法,其特征在于,上述免疫相容性毛乳头细胞抑制选自由HLA-A、HLA-B、CD55、IL-1RA及CCL2组成的组中的一种以上信使核糖核酸的表达。
7.一种免疫相容性毛乳头细胞,其特征在于,通过权利要求1所述的分离和培养方法制备。
