The mice were collected from the eyeballs and blood was collected, placed at room temperature for about 1 hour, centrifuged at 3000rpm for 15min to separate serum, stored at-80℃and assayed for serum cytokine (IgG, igA, IL-6, TNF-a) content according to the assay kit instructions.
Index of spleen and thymus
Mice were sacrificed after blood collection, spleen and thymus were dissected after intact uterus was dissected, the viscera surface blood was blotted with filter paper, weighed, and spleen (thymus) index calculated according to the formula, spleen (thymus) index = spleen (thymus) mass (mg)/body weight (g).
Female fertility and fetal mouse mass:
after blood collection, the whole uterus is dissected and further dissected, the total weight of the fossa and the total weight of the placenta are weighed by an analytical balance, the number of mice is recorded (the number of live fetuses, dead fetuses and absorbed fetuses are recorded), and the physique of the mice (the weight, the body length, the tail length and the appearance of the live mice) is recorded.
g) Action detection index on mice
After the rest pregnant mice are naturally produced, offspring observation experiments are carried out, each group of female mice is subjected to gastric lavage for 21 days, the gastric lavage volume is 20mL/kg, the blank control group is subjected to daily administration of equal volume of distilled water, and 15 mice in each group are indirectly interfered with the mice through feeding milk through the female mice.
Body constitution observation of mice
Observing physical indexes of the mice in 1, 7, 14 and 21 days: body weight, length, tail length, shape, etc.
Behavioral observations of mice
The mice were observed for day 21 behavioural behavior in the same manner as under item "4.6.2".
Mice immunity index:
blood is taken, the separated serum is preserved at the temperature of minus 80 ℃, the content of serum cytokines (IgG, igA, IL-6) is detected, then spleen and thymus are dissected, the surface blood stain of the viscera is sucked by filter paper, the spleen (thymus) index is calculated according to a formula, and the spleen (thymus) index=spleen (thymus) quality (mg)/body weight (g).
1.1. Brain tissue index of mice
Brain index
The brains of the mice were weighed with an analytical balance and the brain index = brain mass (mg)/body weight (g) was calculated according to the formula.
Antioxidant index in brain tissue
The brain tissue of the mice is weighed, and then the left hemisphere of the mice is uniformly taken and stored at the temperature of minus 80 ℃ for detecting the oxidation resistance index, and the superoxide dismutase (SOD) activity and the Malondialdehyde (MDA) content are detected according to the instruction of the kit.
Activity detection of ChAT and AchE in cerebral cortex
The rat brain tissue is weighed, then the isolated brain right hemisphere cortex tissue is preserved at-80 ℃ and used for detecting the activities of the Chut and the AchE in the cortex, and the activities of the Chut and the AchE in the brain cortex are detected according to the instruction book of the kit.
1.4 data processing
Experimental data were statistically processed by GraphPad Prism 9.2 biometric software: the metering data are expressed in mean±sd and are analyzed using analysis of variance in combination with Dunnett's multiple comparison.
1.5 experimental results
a) Influence of the sample on the weight and general status of pregnant mice
The animal groups have good mental state, smooth fur color, free movement, uniform breathing, no obvious abnormality of food intake and feces, and no abnormal secretion in the mouth and nose during the experimental period. From Table 1, it can be seen that the animals of each group had a normal weight gain during each period of feeding, and that the pregnant mice of each sample group had a significant weight gain (P < 0.01) compared to the blank group.

b) Behavioral effects of test samples on pregnant mice
As can be seen from table 2, the number of erection times of pregnant mice in the formula bird's nest high dose group is increased (p < 0.05) to a certain extent compared with that in the blank control group.
The results of the 18d elevated maze of pregnant female mice are shown in Table 3, which shows an increase in the number of times and residence time of the high dose group of formula bird's nest into the open arm (p < 0.05) compared to the blank control group.
The results of the mother pregnancy 18d tail suspension experiments show that the formula bird's nest high dose group has a certain increase (p < 0.05) relative to the rest time compared with the blank control group as shown in table 4.

c) Influence of the test sample on fertility of the female mouse
The total weight of the littermates and the total weight of the placenta of the pregnant female mice were 19d, and as can be seen from table 5, the total weight of the placenta was increased (p < 0.05) in the formula bird's nest high dose group compared to the blank control group.
As seen from table 6, the number of live fetuses of each sample group is not significantly different (p > 0.05) from that of the blank control group, and the number of dead fetuses and the number of absorbed fetuses of the dose group in the formula bird's nest and the high-dose group in the formula bird's nest are reduced to a certain extent (p < 0.05).
As can be seen from table 7, compared with the blank control group, the average body weight of the bird's nest high-dose group child is significantly increased (P < 0.05), the average body length of the bird's nest high-dose group child is significantly increased (P < 0.01), and no significant difference (P > 0.05) is seen in the rat tail length of each sample group.

d) Influence of the test sample on the immunocompetence of the female mouse
As shown in Table 8 and Table 9, the serum of pregnant female mice has a tendency of increasing the IgG content (P < 0.05), the serum of female mice has a tendency of increasing the IL-6 and TNF-alpha content, and the serum of female mice has a tendency of increasing the IL-6 and TNF-alpha content (P < 0.05).
As can be seen from table 10, the spleen index of the mice in the d. Formula bird's nest high dose group tended to increase (P < 0.05) compared to the blank group, and the thymus index of the mice in each sample group did not significantly differ (P > 0.05).

e) Influence of the test sample on the constitution of mice
As shown in Table 11, the weight of the mice in the dosage group 1d and 7d in the formula bird's nest tends to increase (P < 0.05) compared with the blank control group, and the weight of the mice in the high dosage group 1d, 7d, 14d and 21d in the formula bird's nest tends to increase (P < 0.05).
As a result of the body length of the mice in 1d to 21d, it can be seen from Table 12 that the body lengths of the mice in the dosage group 7d and 21d in the formula bird's nest are increased (P < 0.05) and the body lengths of the mice in the high dosage group 7d, 14d and 21d in the formula bird's nest are increased (P < 0.05) compared with the blank control group.
From Table 13, it can be seen that the tail lengths of the mice 7d and 21d in the high-dose group of formula bird's nest have an increasing tendency (P < 0.05) compared with the blank group.

f) Effects of test samples on mouse behaviours
As shown in table 14, compared with the blank control group, d. The formula bird's nest high dose group mice have a trend of increasing residence time in the central area of open field, the number of times of entering the central area of open field and the number of erection times (P < 0.05).
The experimental results of 21d overhead maze of the born mice show that compared with a blank control group, the frequency, the residence time and the duty ratio of the open arms of the mice in the formula bird's nest high-dose group are all increased, and the frequency and the residence time duty ratio of the open arms of the mice in the formula bird's nest in the formula group are increased (P is less than 0.05).
The results of 21d tail suspension experiments performed on mice born are shown in Table 16, and compared with the blank control group, the B.common bird's nest group and the D.formula bird's nest high dose group have a reduced tendency (P < 0.05) relative to the rest time.

g) Influence of the test sample on the immunity of the mice
As shown in Table 17, the serum of the mice born with 22d mice has a tendency of increasing the content of IgG, igA and IL-6 (p < 0.05) in the serum of the mice in the high-dose group of formula nidus Collocaliae, and the serum of the mice in the dose group of formula nidus Collocaliae has a tendency of increasing the content of IL-6 (p < 0.05) compared with the serum of the mice in the blank group.
The results of immune organs of the mice born at 22d are shown in Table 18, compared with a blank control group, the thymus of the mice in the common bird's nest group and the dosage group in the formula bird's nest group are increased, and the spleen index and thymus of the mice in the formula bird's nest high dosage group are both increased (p is less than 0.05).

h) Influence of the sample on the brain tissue of the mice
The brain index results of the 22d mice born are shown in table 19. It can be seen that the brain index of the d-formula bird's nest high dose group mice is significantly increased (p < 0.05) compared to the blank control group.
As shown in Table 20, compared with the blank control group, the MDA content in brain tissue of the mice in the formula bird's nest dose group and the formula bird's nest high dose group has a tendency to decrease (p < 0.05), and the SOD activity of brain tissue of the mice in the formula bird's nest high dose group has a tendency (p < 0.05).
Acetylcholine transferase (ChAT) and acetylcholinesterase (AchE) are two important enzymes in brain for synthesis and hydrolysis of the neurotransmitter acetylcholine, whose activity directly affects the content of acetylcholine in the brain. From table 21, it can be seen that the activity of acetylcholine transferase (ChAT) and acetylcholinesterase (AchE) in the cerebral cortex of the newborn mice at birth 22d is increased (p < 0.05) in the c-formulation bird's nest dose group and d-formulation bird's nest high-dose group in the cerebral cortex tissue of the newborn mice compared to the blank control group; B. the AChE activity in rat brain cortex tissues of common bird's nest group and D. formulated bird's nest high dose group has a reduced tendency (p < 0.05).

Conclusion: under the experimental conditions, the formula bird nest can obviously increase the body length and the body weight of the pregnant mice in the pregnant mice with 19 days, improve the open field, the overhead cross maze and the tail suspension behavioural index of the pregnant mice to a certain extent, increase the serum immune index IgG, IL-6, TNF-alpha content and immune organ spleen index of the pregnant mice, and improve the immunity of the pregnant mice to a certain extent. Influence of formula bird's nest on offspring: the bird's nest has the tendency of increasing the body weight, the body length and the tail length of the mice, has a certain effect on improving the constitution of the mice, has a certain improvement on the open field, the overhead cross maze and the tail suspension behavioural index of the mice, has a certain tendency of increasing serum immune index IgG, IL-6 content and immune organ spleen and thymus index of the mice, has a certain improvement on the immunity of the mice, has a remarkable increase on the brain index of the mice, has a tendency of reducing MDA content and improving SOD activity, has a tendency of increasing ChAT enzyme activity and reducing AchE activity, and has a certain improvement effect on the development of the brain of the mice.
Claims (15)
1. An instant bird's nest product suitable for consumption by a pregnant woman, comprising: 1-25 parts of bird's nest and 1-30 parts of bird's nest mate; the bird's nest mate comprises the following components: at least one of sugar, probiotics, vitamin A, vitamin B1, vitamin B2, vitamin B3, vitamin B6, folic acid, vitamin B12, vitamin C, vitamin D and vitamin E.

The instant bird's nest product suitable for pregnant women according to claim 1, wherein the bird's nest mate comprises the following components in parts by weight: 30 parts of sugar, 12 parts of probiotics, 4 parts of vitamin A,4 parts of vitamin B1,4 parts of vitamin B2,4 parts of vitamin B3,4 parts of vitamin B6,2 parts of folic acid, 4 parts of vitamin B12,4 parts of vitamin C,4 parts of vitamin D and 8 parts of vitamin E.
