Abstract
为克服现有防脱、生发产品治疗效率低、具有毒副作用的问题,本发明提供了一种防脱生发的药物配方,包括以下组份:侧柏叶、川芎、红茶、丁香花蕾、墨旱莲、菟丝子、女贞子、地榆根、红花、人参、地黄根、八角茴香果、蒲公英、何首乌、紫苏叶、马齿苋、黑桑果、姜根、当归根、丹参、银杏叶。本发明还提供了包括上述防脱生发的药物配方的微针贴及其制备方法。根据本申请提供的防脱生发的药物配方,从中医病因病机辨证入手,组方配伍君臣佐使齐全,具有调补肝肾,养血活血,祛风通络、除湿利气等功效,能够刺激毛囊再生,具有疏通经络、刺激气血运行的作用,达到防止脱发及生发之功效。

技术领域
本发明属于生发用药物配方技术领域,具体涉及一种防脱生发的药物配方、微针贴及其制备方法。
背景技术
脱发长期以来危害人们的身心健康,现代医学认为,脱发与神经系统功能紊乱和人体免疫力下降有关,通常多由于忧虑过度,心血亏虚,造成血瘀毛窍,血不养发,出现头发脱落;传统中医认为:“发为血之余,发为肾之华”,头发的生长与人体内精、气、血都有密切的关联,而精、气、血又和心、肝、脾、肺、肾密切相关,肾主藏精,精血同源,心主血、肝藏血,所以脱发不仅是头发本身的问题,而是机体亚健康状态的间接体现;若长期忧虑过度,久病体虚,精血亏虚,气血不畅,使得头发失养而导致脱发。
随着社会飞速的发展,人们生活节奏的加快,由于生活、学习、工作压力、饮食、环境的污染等多方面因素影响,很多人出现脱发的现象,并且年轻化趋势日益明显,在生活水平不断提高和社交活动频繁的同时,给我们带来极大苦恼。目前市场上也出现一些有关治疗脱发的产品,西医治疗脱发通常采用激素类药,但效果也不是很理想,并且对身体有副作用;中医治疗脱发也取得了一定的成效,同时也受到广大脱发患者的青睐,但仍存在总体效率较低的问题。同时现有生发产品则大多数为精华液、乳液,配方中需要添加大量溶剂(如水、乙醇等)、乳化剂及防腐剂,外用有效成分的吸收较差,还可能会引起皮肤过敏、发炎等不适症状,导致脱发问题更佳严重。
因此亟需寻求一种高安全、高效、无毒副作用的生发产品来解决上述问题。
发明内容
针对现有防脱、生发产品治疗效率低、具有毒副作用的问题,本发明提供了一种防脱生发的药物配方、微针贴及其制备方法。

川芎:具有活血行气,祛风止痛的功效,《日华子本草》:治一切风,一切气,一切劳损,一切血,补五劳,壮筋骨,调众脉,破症结宿血,养新血,长肉,鼻洪,吐血及溺血,痔瘘,脑痈发背,瘰疬瘿赘,疮疥,及排脓消瘀血。
红茶:具有抗菌消炎的作用,可用于清除自由基能够抑制病毒。
丁香花蕾:具有清热解毒,消肿散结,利尿通淋的作用。
墨旱莲:凉血止血,补肾益阴,乌须发,益肾阴。
菟丝子:性甘、温,归肾、肝、脾经,具有滋补肝肾、固精缩尿之功效,始载《神农本草经》,被列为上品。
女贞子:选自《中华药典》:归肝肾经,滋补肝肾,明目乌发,用于肝肾阴虚,眩晕耳鸣,腰膝酸软,须发早白,目暗不明,内热消渴,骨蒸潮热。
地榆根:性味酸、寒。归肝、大肠经,具有清热解毒,凉血止血之功效。
红花:选自《本草纲目》:活血润燥,止痛散肿,通经,可活血通经,祛瘀止痛,促进头部血液循环。
人参:选自《中国药典》功能与主治:补脾益肺,生津养血,安神益智。用于脾虚食少,肺虚喘咳,津伤口渴,气血亏虚。
地黄根:生津润燥,清热凉血。
八角茴香果:润肾补肾。
蒲公英:具有清热解毒,消肿散结,利尿通淋的作用。
何首乌:选自《中华药典》:归肝、肾经,补肝肾,益精血,乌须发,用于精血亏损、须发早白。
紫苏叶:选自《中国药典》解表散寒,行气和胃。
马齿苋:性味酸、寒,归肝、大肠经,具有清热解毒,凉血止血之功效。
黑桑果:选自《中国药典》功能与主治为滋阴补血,生津润燥。用于肝肾阴虚,眩晕耳鸣,心悸失眠,须发早白,津伤口渴,内热消渴,肠燥便秘。
姜根:姜辣素、姜烯油等成分,可使头部皮肤血液循环正常化,促进头皮新陈代谢,帮助药物到达病灶。
当归根:补血,活血,润燥。
丹参:选自《本草纲目》:活血,通心包络,治疝痛。可活血祛瘀,通经止痛,清心除烦,凉血消痈。
银杏叶:选自《中国药典》清虚热,除疳热用于阴虚发热,骨蒸劳热。
以侧柏叶、菟丝子、女贞子、墨旱莲、何首乌为君药,主滋养补肾,辅以臣药--人参、八角茴香果、黑桑果,佐药--地榆根、马齿苋、地黄根、红花、丁香花蕾、蒲公英、当归、丹参、紫苏叶、银杏叶,使药--川芎、姜根,君臣佐使各成分之间协同作用,其综合作用调补肝肾,疏通经络、刺激气血,刺激毛囊再生,起到防脱、生发疗效。

所述硫酸锌用于头皮抗菌、控油抗脂溢;
所述二氨基嘧啶氧化物为化妆品批准用原料,安全性高,可用于软化胶原蛋白,抑制毛囊周围胶原蛋白生成,避免纤维化,可用于治疗由于发根过早损耗造成的脱发,坚固发根,促进头发生长;
所述生物素是一种维持人体自然生长、发育和正常人体机能健康必要的营养素,可提高血液在皮肤中的循环速度,对防止落发及头顶见光颇见功效,还能预防少年白头。
与上述中药组份配合使用,进一步增强防脱生发效果。
另一方面,本发明还提供了一种微针贴,包括基体以及固定在所述基体上的微针层,所述微针层包括针体,所述针体包括可溶性高分子材料和如上所述的防脱生发的药物配方。
以可溶性高分子材料搭载本申请防脱生发药物配方制成微针贴,所述针体可通过在皮肤角质层形成大量微孔道从而增加药物的渗入,所述可溶性高分子材料具有生物降解性,可直接与搭载药物溶解并注入肌肤表皮层,能够完全被皮肤吸收,可避免现有无法降解的微针在使用时会断裂于皮肤内带来难以处理的问题。
进一步的,优选所述基体上靠近所述微针层的表面设有胶粘层,所述胶粘层位于所述基体的外沿并与所述微针层并排设置,使得本申请微针贴在使用时,可直接将胶粘层敷贴于患者头皮处,按压微针层,使得微针层上的微针刺穿角质层,将药物注入皮肤,相比于现有外用喷涂等的方式,本申请以微针形式导入药物,极大增加了人体对药物的吸收率,从而提高药物的防脱生发效果,使用简单易操作,具有广泛地应用前景。
在优选的实施例中,所述胶粘层上远离所述基体的表面设置有离型层,用于在存储过程中保护胶粘层,使用前,先将离型层剥离。
在一些实施例中,所述可溶性高分子材料与所述防脱生发的药物配方的质量比为17:3~9:1。
在上述范围内的微针贴能够在较低成本下提高防脱生发效果。
在一些实施例中,所述可溶性高分子材料选自透明质酸、海藻糖、明胶、羟乙基纤维素中的至少一种。
所述可溶性高分子材料为具有生物降解性的材料,可直接被皮肤完全吸收,而不产生毒副作用,同时能够搭载上述药物配方制备得到粒径要求范围内的微针体。
在一些实施例中,所述基体选自无纺布、水胶体贴片、医药级胶带、印花布中的至少一种。
具体的所述基体需要具备一定的柔软性,使得微针贴能够在最大程度贴合人贴头皮,进而确保药物治疗效果。
所述微针贴的形状可以为圆形、矩形、异形等,在此不作特别的限定。
另一方面,本发明还提供了上述微针贴的制备方法,包括以下步骤:
S1:按比例将侧柏叶、川芎、红茶、丁香花蕾、墨旱莲、菟丝子、女贞子、地榆根、红花、人参、地黄根、八角茴香果、蒲公英、何首乌、紫苏叶、马齿苋、黑桑果、姜根、当归根、丹参、银杏叶的提取物进行混合;
S2:将上述提取物与硫酸锌、二氨基嘧啶氧化物、生物素和可溶性高分子材料按比例进行混合,通入微针模具,干燥后得到所述针体。

具体的,步骤S2中,通入微针模具后可采用离心或者抽真空的方式将混合溶液填充到微针模具的微针腔体内,然后通过常温或者低温干燥,使可溶性高分子材料固化成形,所述制备方法简单易操作,绿色经济。

实施例1
本实施例用于说明本发明公开的防脱生发的药物配方、微针贴、及其制备方法。
防脱生发药物配方各组分及其含量如表1所示

S1:将侧柏叶、川芎、红茶、丁香花蕾、墨旱莲、菟丝子、女贞子、地榆根、红花、人参、地黄根、八角茴香果、蒲公英、何首乌、紫苏叶、马齿苋、黑桑果、姜根、当归根、丹参、银杏叶分别进行粉碎,酒精提取、浓缩、浸膏、溶剂萃取、萃取液浓缩浸膏、干燥、粉碎过筛得到100目粉末,按表1比例复配各提取物;
S2:将步骤S1复配得到混合提取物与86.3份的透明质酸钠混合,经微针成针技术制备得到若干针体;
S3:将上述针体附着于直径为2.2cm的圆形无纺布上,得到所述微针贴。
测试例:脱发患者试验
选择50例有脱发困扰的志愿者,使用实施例1提供的微针贴,每次3贴,剂量:21mg/贴;每周使用3次;选取脱发面积:4cm2;使用时间:45天;治疗效果如下表2所示:

根据表2的测试结果可以看出,实施例1提供的微针贴对于脂溢性脱发、斑秃性脱发及精神性脱发均具有不同程度的改善效果,对三者的总有效率可达65%以上,其中对于脂溢性脱发及精神性脱发治疗效果最佳,总有效率可达70%以上。
注:头发生长率≥70%的治疗效果称为显效;70%>头发生长率≥30%的治疗效果称为有效;头发生长率<30%的治疗效果称为无效;
头发生长率的计算方法:选取患者脱发部位2cm×2cm面积(用药部位),使用前记录该面积头发数量,治疗周期结束后,计算相同部位头发数量,即:
头发生长率=(使用后该部位头发数量-使用前该部位头发数量)÷使用前该部位头发数量×100%。
总有效率=(显效人数+有效人数)/病例总数×100%。

不进行微针成针技术,并且步骤S2为:将步骤S1制得的粉末按比例与表7中的硫酸锌、二氨基嘧啶氧化物、生物素混合,制成液态产品。
测试例:脱发患者试验
选择50名有脱发困扰的志愿者,使用对比例1中的液态脱发产品,每次使用5ml,将液态产品喷涂于头发稀疏处,按摩吸收;每周使用3次;选取脱发面积:4cm2;使用时间:45天;治疗效果如下表9所示:

根据表9的测试结果可以看出,对比例1提供的液态产品,与实施例4提供的微针贴相比,对比例1对于脂溢性脱发、斑秃性脱发及精神性脱发患者的治疗总有效率均下降了10%及以上,说明本申请防脱生发药物配方配合微针技术制成微针贴,能够进一步提高脱发治疗效果。
以下通过具体的治疗实施例对本发明进行进一步的说明。
实施例5
黄先生,42岁,脂溢性脱发,脱发5年,每周使用实施例1中的微针贴3次,每次使用3贴,将微针贴贴敷于头发稀疏处,按压吸收40min,揭去微针贴,连续治疗30天,脱发现象明显改善,脱发部位有绒毛长出,头发生长率达到96%以上,头发变粗壮。
实施例6
曾先生,50岁,遗传性脱发,脱发7年,每周使用实施例1中的微针贴4次,每次使用3贴,将微针贴贴敷于头发稀疏处,按压吸收35min,揭去微针贴,连续治疗50天,脱发现象明显改善,头发生长率达到77%以上。
实施例7
陈先生,48岁,遗传性脱发,脱发10多年,每周使用实施例1中的微针贴3次,每次使用4贴,将微针贴贴敷于头发稀疏处,按压吸收30min,揭去微针贴,连续治疗45天,脱发现象明显改善,头发生长率达到80%以上。

李先生,41岁,脂溢性脱发,脱发4年,每周使用对比例1中的液态脱发产品4次,每次使用5ml,将液态产品喷涂于头发稀疏处,按摩吸收30min,连续治疗45天,脱发现象有改善,头发生长率达到75%。
根据实施例5~11的治疗结果看出,本申请防脱生发的药物配方能够有效改善脂溢性脱发、遗传性脱发、压力性脱发等常见脱发症状,头发生长率能达到70%以上,同时对比实施例10和实施例11的治疗结果可以看出,实施例10采用微针贴透皮给药技术,而实施例11直接采用涂抹于皮肤表面的给药技术,两者治疗结果看出,微针形式能够提高皮肤对药物的吸收率,从而提高头发生长率,说明本申请防脱生发的药物配方配合微针技术,能够进一步改善脱发症状,具有良好的应用前景。

1.一种微针贴,其特征在于,包括基体以及固定在所述基体上的微针层,所述微针层包括针体,所述针体包括可溶性高分子材料和防脱生发的药物组合物,所述防脱生发的药物组合物为:
侧柏叶提取物1~8份、川芎提取物0.2~3份、红茶提取物0.2~4份、丁香花蕾提取物0.1~2份、墨旱莲提取物0.17~2.5份、菟丝子提取物0.17~2份、女贞子提取物0.5~5份、地榆根提取物0.5~5.5份、红花提取物0.1~2份、人参提取物0.1~2.5份、地黄根提取物0.1~1.5份、八角茴香果提取物0.1~2份、蒲公英提取物0.2~3份、何首乌提取物0.3~2.5份、紫苏叶提取物0.1~3份、马齿苋提取物0.2~2份、黑桑果提取物0.1~1.5份、姜根提取物0.1~3份、当归根提取物0.5~8份、丹参提取物0.1~2份、银杏叶提取物0.5~5份、硫酸锌0.1~3份、二氨基嘧啶氧化物0.2~2份、生物素0.2~2份;
所述可溶性高分子材料选自透明质酸、海藻糖、明胶、羟乙基纤维素中的至少一种;
所述基体选自无纺布、水胶体贴片、医药级胶带、印花布中的至少一种;
所述可溶性高分子材料与所述防脱生发的药物组合物的质量比为17:3~9:1;
所述的微针贴的制备方法,包括以下步骤:
S1:按比例将侧柏叶、川芎、红茶、丁香花蕾、墨旱莲、菟丝子、女贞子、地榆根、红花、人参、地黄根、八角茴香果、蒲公英、何首乌、紫苏叶、马齿苋、黑桑果、姜根、当归根、丹参、银杏叶的提取物进行混合;
S2:将上述提取物与硫酸锌、二氨基嘧啶氧化物、生物素和可溶性高分子材料按比例进行混合,通入微针模具,干燥后得到所述针体;
S3:将所述针体附着于基体上,得到所述微针贴。
