Abstract
本发明公开了一种评估化学物质对于斑马鱼神经内分泌影响的方法,涉及生态风险评价技术领域。设置不进行化学物质暴露的转neurod1:荧光报告基因的转基因斑马鱼对照组,利用转neurod1:荧光报告基因的转基因斑马鱼在化学物质暴露环境下的肠内分泌细胞的分布水平与转基因斑马鱼对照组的肠内分泌细胞的分布水平的差异进行斑马鱼神经内分泌影响的评估。与传统技术方案相比,本发明提供的方法可在活体条件下直接观测,无需牺牲试验生物,且斑马鱼无需具有自主运动能力之后才能观测,大幅降低了对于观测时间的特定要求。此外,本发明提供的方法简单易行,不需要额外的设备、检测试剂,也无需特定的检测环境。

技术领域
本发明涉及生态风险评价技术领域,具体而言,涉及一种评估化学物质对于斑马鱼神经内分泌影响的方法。
背景技术
神经内分泌干扰效应,是环境污染物、尤其是具有内分泌干扰效应的污染物对生物的重要毒理靶标。目前对神经内分泌干扰效应的评价主要通过神经递质水平、神经行为影响等方式。神经递质主要通过靶向代谢组学或免疫学等手段进行检测,一般需要采集鲜活组织后匀浆,取上清液进行测定;神经行为学测试主要是通过特定软件对动物的行为视频进行分析获得。
其中,组织水平的神经递质检测,不仅需要牺牲试验生物,靶向代谢组学检测需要依赖于大型质谱仪器才能完成,而免疫检测法则需要购买商品化的试剂盒来进行检测,试验成本和劳动消耗均较大。神经行为学测试虽能在活体条件下获得结果,但依赖于专业的视频分析软件才能完成,且试验过程容易受到外界的干扰(外界的声音等对鱼类的行为影响很大),为此一般需要非常安静的试验环境或专用的行为观察箱。此外,行为学必须在斑马鱼具有自主运动能力之后(一般在受精后120小时)才能观测,无法实现更早期的观察。
鉴于此,特提出本发明。
发明内容
本发明的目的在于提供一种评估化学物质对于斑马鱼神经内分泌影响的方法以解决上述技术问题。
本发明是这样实现的:
第一方面,本发明提供了一种评估化学物质对于斑马鱼神经内分泌影响的方法,其包括如下步骤:设置不进行化学物质暴露的转neurod1:荧光报告基因的转基因斑马鱼对照组,利用转neurod1:荧光报告基因的转基因斑马鱼在化学物质暴露环境下的肠内分泌细胞的分布水平与转基因斑马鱼对照组的肠内分泌细胞的分布水平的差异进行斑马鱼神经内分泌影响的评估。
发明人偶然发现,显微观察对照组与转neurod1:荧光报告基因的转基因斑马鱼的在化学物质暴露后肠内分泌细胞的分布水平差异(或变化),可以根据该水平差异或水平变化对化学物质是否具有神经内分泌干扰效应进行预判。经过研究发现,与对照组相比,化学物质暴露后肠内分泌细胞的分布水平的变化与大部分传统神经内分泌干扰效应指标(如神经递质水平、行为学指标等)具有显著的正相关关系,且部分指标相关性系数高于0.7。与传统技术方案相比,本发明提供的方法可在活体条件下直接观测,无需牺牲试验生物,且斑马鱼无需具有自主运动能力之后才能观测,大幅降低了对于观测时间的特定要求。此外,本发明提供的方法简单易行,不需要额外的设备、检测试剂,也无需特定的检测环境(如非常安静的试验环境、专用的行为观察箱)。
在本发明应用较佳的实施方式中,检测转基因斑马鱼对照组和化学物质暴露后的转基因斑马鱼的肠内分泌细胞的荧光强度;
对待测的斑马鱼进行显微拍摄,使用图片分析软件对拍摄的图片进行荧光强度数据提取,选定肠道区域,获得肠内分泌细胞的荧光强度。
直接分析显微图片的方式操作更加简便,不需要额外的设备和检测试剂。图片分析软件包括不限于Image J等软件,只要能对荧光强度数据进行数据提取,从而获得肠内分泌细胞的荧光强度均在本发明的发明构思范围之内。分析步骤如下:
在Image J中将图片转换为单通道(绿色荧光)图片格式,选择肠道区域,之后调整合适的阈值以选择肠内分泌细胞,测量选取范围的灰度值表征荧光强度。
在其他实施方式中,也可直接采用具有显微拍摄和图片分析的设备获得肠内分泌细胞的荧光强度。
在一种可选的实施方式,将要观测的仔鱼在麻醉剂(如三卡因)中麻醉,在荧光显微镜下调整好角度并拍摄照片,每条仔鱼尽量拍摄角度相同的照片。
在一种可选的实施方式,斑马鱼为受精后80h的斑马鱼胚胎和/或斑马鱼。
在本发明应用较佳的实施方式中,方法包括:与转基因斑马鱼对照组的肠内分泌细胞的荧光强度相比,若化学物质暴露后的转基因斑马鱼的肠内分泌细胞的荧光强度显著下降,则评估化学物质对于斑马鱼神经内分泌具有显著抑制作用。分析方法包括不限于利用SPSS 16.0中的单因素方差分析(dunnett检验),对对照组和化学物质处理组的荧光强度进行显著性比较,确定处理组与对照组斑马鱼之间肠内分泌细胞水平是否出现显著性差异。
在本发明应用较佳的实施方式中,化学物质选自环境污染物、药物和水质改良剂中的至少一种。
在本发明应用较佳的实施方式中,环境污染物选自农药、抗生素和化肥中的至少一种。
术语“农药”应理解为是指一种或多种农药可以包含在所述农业化学组合物中。本领域技术人员熟知该类农药,其可参见例如Pesticide Manual,第16版(2013),TheBritish Crop Protection Council,London。
在本发明应用较佳的实施方式中,农药为杀菌剂、杀虫剂、杀线虫剂、除草剂、安全剂、生物农药和/或生长调节剂。
在本发明应用较佳的实施方式中,药物选自抗抑郁药物。
在本发明应用较佳的实施方式中,环境污染物选自双酚类污染物。本发明中,双酚类化合物(bisphenol compounds,BPs)是由碳或硫原子桥连2个羟苯基形成的一系列化合物。
在一种可选的实施方式,双酚类污染物选自双酚A(BPA)、双酚AF(BPAF)、双酚F(BPF)和双酚G(BPG)中的至少一种。
在一种可选的实施方式,当环境污染物为双酚类污染物时,设置化学物质暴露浓度,暴露时间3-6天;
在一种可选的实施方式,暴露浓度为0.05-5000μg/L。例如暴露浓度为0.05μg/L-500μg/L,或0.5μg/L-1000μg/L,或1μg/L-5000μg/L。本领域的技术人员可以根据需要调整暴露浓度。暴露浓度是指化学物质在溶剂或分散介质中的分散浓度。
在本发明应用较佳的实施方式中,荧光报告基因选自编码GFP、RFP、BFP、EGFP、YFP、mCherry、mStrawberry、mApple、mRuby和EosFP荧光蛋白中的至少一种编码基因。
第二方面,本发明还提供了一种转neurod1:荧光报告基因的转基因斑马鱼在评估化学物质对于斑马鱼神经内分泌影响中的应用。
在本发明应用较佳的实施方式中,上述应用包括:设置不进行化学物质暴露的转neurod1:荧光报告基因的转基因斑马鱼对照组,利用转neurod1:荧光报告基因的转基因斑马鱼在化学物质暴露环境下的肠内分泌细胞的分布水平与转基因斑马鱼对照组的肠内分泌细胞的分布水平的差异进行斑马鱼神经内分泌影响的评估。
本发明具有以下有益效果:
本发明提供了一种评估化学物质对于斑马鱼神经内分泌影响的方法,通过显微观察对照组与转neurod1:荧光报告基因的转基因斑马鱼的在化学物质暴露后肠内分泌细胞的分布水平差异(或变化),可以根据该水平差异或水平变化对化学物质是否具有神经内分泌干扰效应进行预判。
经过研究发现,与对照组相比,化学物质暴露后肠内分泌细胞的分布水平的变化与大部分传统神经内分泌干扰效应指标(如神经递质水平、行为学指标等)具有显著的正相关关系,且部分指标相关性系数高于0.7。本发明的提出对于获得传统指标的结果具有较大的指示意义。
与传统技术方案相比,本发明提供的方法可在活体条件下直接观测,无需牺牲试验生物,且斑马鱼无需具有自主运动能力之后才能观测,大幅降低了对于观测时间的特定要求。此外,本发明提供的方法简单易行,不需要额外的设备、检测试剂,也无需特定的检测环境(如非常安静的试验环境、专用的行为观察箱等)。

为使本发明实施例的目的、技术方案和优点更加清楚,下面将对本发明实施例中的技术方案进行清楚、完整地描述。实施例中未注明具体条件者,按照常规条件或制造商建议的条件进行。所用试剂或仪器未注明生产厂商者,均为可以通过市售购买获得的常规产品。
以下结合实施例对本发明的特征和性能作进一步的详细描述。
实施例1
本实施例提供了一种评估双酚A对于斑马鱼神经内分泌影响的方法,其包括如下步骤:
(1)污染物暴露。根据实际需求,确定供试物、暴露浓度(0.5μg/L、50μg/L、500μg/L)和处理时间,对neurod1:GFP转基因斑马鱼成鱼(购买自国家斑马鱼资源中心(http://www.zfish.cn/))配对产下的胚胎进行处理。对照组50条转基因斑马鱼;处理组50条转基因斑马鱼,暴露时间3-6天,在完成暴露后,选择10条转基因斑马鱼,用于后续检测。每个处理和对照组应统计10条斑马鱼。
(2)荧光显微镜观测。将要观测的仔鱼在三卡因中麻醉,在荧光显微镜下调整好角度并拍摄照片,每条仔鱼尽量拍摄角度相同的照片。
(3)荧光强度计算。对拍好的照片,使用图片分析软件(Image J)进行荧光强度数据提取,选定肠道区域,获得肠内分泌细胞的相对荧光强度。
(4)肠内分泌细胞指标统计分析。利用SPSS16.0中的单因素方差分析(dunnett检验),对对照组和污染物处理组的荧光强度进行显著性比较,确定处理组与对照组斑马鱼之间肠内分泌细胞水平是否出现显著性差异。在其他实施方式中,上述统计分析也可使用其它分析软件。
若处理组与对照组斑马鱼之间肠内分泌细胞水平出现显著性差异,则判断双酚A对斑马鱼神经内分泌具有干扰作用。
实施例2
与实施例1相比,区别在于双酚类污染物为双酚AF,其他的步骤相同。
实施例3
与实施例1相比,区别在于双酚类污染物为双酚G,其他的步骤相同。
实验例1
按照实施例1-3的方法分别评估了三种双酚类污染物(双酚A、双酚AF和双酚G)对斑马鱼神经内分泌的干扰作用。
根据评估结果,发现双酚AF和双酚G在高于50μg/L的剂量时,对肠内分泌细胞具有显著抑制作用;双酚A在500μg/L剂量下,对肠内分泌细胞水平具有抑制作用(图1)。
发明人开展了双酚类污染物对斑马鱼神经递质水平和神经行为的验证试验。
神经递质水平检测方法:收集固定数量的仔鱼样品放入离心管中,以30尾仔鱼/离心管作为一个样本,依次加入200μL预冷的10%甲酸甲醇和200μL纯水,将混合物在冰浴上超声处理20min后,转移到1.5ml离心管,在4℃下以15300rpm离心10分钟。离心后收集上清液。而后在45℃下,持续的氮气流下,将上清液蒸发至干,重新溶解在400μL含有0.1%甲酸的甲醇/水(1:1,V/V)溶液中,涡旋直至完全溶解,并在室温下以10,000rpm离心10分钟。取上清液,采用SCIEX QTrap 6500+MS/MS系统(SCIEX,美国)与Xbridge C18色谱柱(4.6mm x150mm,3.5μm;Waters)。梯度洗脱,水相为0.1%甲酸蒸馏水,有机相为0.1%甲酸乙腈。质谱检测采用多反应监测(MRM)模式,配备ESI离子源,在正离子模式下工作。
神经行为试验方法:每个处理组和对照组,选择24尾斑马鱼仔鱼(n=24)进行行为测试。使用DanioVision(荷兰Noldus公司)的行为观察系统在48孔板中记录了6dpf斑马鱼的行为。将48孔板放置于观察室中,经过5分钟的适应期后,使用视频记录斑马鱼20min内的行为。通过EthoVision XT 15.0软件对视频进行分析,获得包括总移动距离、平均速度和活跃时间在内的三个指标。
上述实验发现双酚G和双酚AF可干扰多巴胺等9种神经递质的水平(图2),并抑制斑马鱼的神经行为能力(图3),以上结果验证了双酚类污染物对斑马鱼的神经内分泌干扰效应,且双酚G>双酚AF>双酚A的影响强度顺序与本方案结果一致。
发明人还对本发明的数据结果与传统方案进行了相关性分析,利用SPSS16.0软件,进行Spearman相关性分析,并获得不同指标之间的相关性系数,以p<0.05认为相关性显著。发现肠内分泌细胞(EDC)水平与神经行为和多种神经递质水平呈显著的正相关性(图4)。表明了本发明的方法获得结果对传统指标获得的结果具有较大的指示意义。

Claims (10)
1.一种评估化学物质对于斑马鱼神经内分泌影响的方法,其特征在于,其包括如下步骤:设置不进行化学物质暴露的转neurod1:荧光报告基因的转基因斑马鱼对照组,利用转neurod1:荧光报告基因的转基因斑马鱼在化学物质暴露环境下的肠内分泌细胞的分布水平与转基因斑马鱼对照组的肠内分泌细胞的分布水平的差异进行斑马鱼神经内分泌影响的评估。
2.根据权利要求1所述的方法,其特征在于,检测转基因斑马鱼对照组和化学物质暴露后的转基因斑马鱼的肠内分泌细胞的荧光强度;
优选地,对待测的斑马鱼进行显微拍摄,使用图片分析软件对拍摄的图片进行荧光强度数据提取,选定肠道区域,获得肠内分泌细胞的荧光强度;
优选地,所述斑马鱼为受精后80h的斑马鱼胚胎和/或斑马鱼。
3.根据权利要求2所述的方法,其特征在于,所述方法包括:与转基因斑马鱼对照组的肠内分泌细胞的荧光强度相比,若化学物质暴露后的转基因斑马鱼的肠内分泌细胞的荧光强度显著下降,则评估化学物质对于斑马鱼神经内分泌具有显著抑制作用。
4.根据权利要求3所述的方法,其特征在于,所述化学物质选自环境污染物、药物和水质改良剂中的至少一种。
5.根据权利要求4所述的方法,其特征在于,所述环境污染物选自农药、抗生素和化肥中的至少一种。
6.根据权利要求4所述的方法,其特征在于,所述药物选自抗抑郁药物。
7.根据权利要求4所述的方法,其特征在于,所述环境污染物选自双酚类污染物;
优选地,所述双酚类污染物选自双酚A(BPA)、双酚AF(BPAF)、双酚G(BPG)、双酚F(BPF)中的至少一种;
优选地,当环境污染物为双酚类污染物时,设置化学物质暴露浓度,暴露时间3-6天;
优选地,暴露浓度为0.05-5000μg/L。
8.根据权利要求1所述的方法,其特征在于,所述荧光报告基因选自编码GFP、RFP、BFP、EGFP、YFP、mCherry、mStrawberry、mApple、mRuby和EosFP荧光蛋白中的至少一种编码基因。
9.转neurod1:荧光报告基因的转基因斑马鱼在评估化学物质对于斑马鱼神经内分泌影响中的应用。
10.根据权利要求9所述的应用,其特征在于,所述应用包括:设置不进行化学物质暴露的转neurod1:荧光报告基因的转基因斑马鱼对照组,利用转neurod1:荧光报告基因的转基因斑马鱼在化学物质暴露环境下的肠内分泌细胞的分布水平与转基因斑马鱼对照组的肠内分泌细胞的分布水平的差异进行斑马鱼神经内分泌影响的评估。
