Abstract
本发明属于超声杀菌技术领域,涉及一种杂化膜囊泡及其制备方法、含其抗菌剂与应用。本发明提供了一种杂化膜囊泡,其通过姜黄衍生纳米囊泡与革兰氏阴性菌的外膜囊泡融合而得,其中所述姜黄衍生纳米囊泡与革兰氏阴性菌的蛋白质质量比小于1:0.1。本发明提供的杂化膜囊泡具有逃避巨噬细胞吞噬的作用从而延长在感染部位滞留时间,同时通过细菌同源靶向性以及膜融合性能,结合在细菌表面或进入细菌内部,从而通过超声达到高效杀菌的效果。

技术领域
本发明属于超声杀菌技术领域,具体涉及一种杂化膜囊泡及其制备方法和抗菌剂应用。
背景技术
肺炎克雷伯菌(Klebsiella pneumoniae)是一种条件致病性革兰氏阴性肠杆菌,常定植于人体的胃肠道、皮肤和鼻咽部,是引起院内感染的最常见病原体之一,尤其是在重症监护病房(ICU)的危重患者中,经常引起肺炎、脑膜炎、菌血症和尿路感染等疾病。高毒力和碳青霉烯类耐药性是肺炎克雷伯菌的两个不同的进化方向,然而由于质粒介导的耐药性或毒力的水平转移,最近在世界范围内的流行克隆中出现整合了这两种表型的肺炎克雷伯菌,形成高毒力和碳青霉烯类耐药的肺炎克雷伯菌(CR-hvKP),如序列类型ST11和ST258。先前流行的荚膜型(KL47)已逐渐被KL64取代,ST11-KL64型的碳青霉烯耐药性肺炎克雷伯菌(CRKP)感染患者在30天内的死亡率明显高于其它CRKP感染患者。这对临床环境构成了极大的威胁。因此,迫切需要开发安全有效的疗法来对抗CR-hvKP的感染。
抗生素是对目前临床抗细菌感染的主要策略,但抗生素耐药问题日趋严峻,耐药细菌的出现对公共卫生造成迫切威胁。
中国专利申请202210588905.X公开了一株多药耐药肺炎克雷伯菌噬菌体及其应用。肺炎克雷伯菌噬菌体Kp_phage_507潜伏期短,爆发量大,能快速杀死培养基中的宿主菌,且对温度和酸碱耐受范围广,宿主谱较宽,具有良好的亲水性,易制成注射剂。经动物试验证实,该噬菌体毒副作用小,安全性高,对多药耐药肺炎克雷伯菌感染有很好的治疗效果。
中国专利申请201811541684.0公开了一种可以显著抑制肺炎克雷伯氏菌生长繁殖的灭菌剂,其主要成分是利普司他汀。包括下列成分:利普司他汀、EDTA二钠盐、NaOH和纯净水;利普司他汀、EDTA二钠盐、NaOH的质量分数分别是:5-13%、0.5-2%和0.5-2%。所述灭菌剂可以用于室内消毒使用,特别是医院等肺炎克雷伯氏菌容易引起传播的公共场所,对抑制肺炎克雷伯氏菌的生长繁殖具有很好效果。
为解决抗生素耐药问题,亟需找到能够替代抗生素的其它疗法。提高药物在体内的循环滞留时间,增强药物在感染部位的聚集,从而实现高效的抗菌与良好的生物安全性,是解决的重要思路。
荚膜是覆盖在多种细菌表面的多糖基质,是肺炎克雷伯菌的典型特征,可通过多种机制防止宿主产生免疫应答,包括抑制免疫细胞吞噬作用,防止早期免疫反应的激活,以及消除补体和抗菌肽的裂解(Paczosa M K,et al.Klebsiella pneumoniae:Going on theOffense with aStrong Defense[J].Microbiol Mol Biol Rev,2016)。与无荚膜的菌株相比,荚膜菌株可以诱导较低水平的促炎细胞因子TNF-α和IL-6以及更高水平的抗炎细胞因子IL-10(Yoshida K,etal.Role of bacterial capsule in local and systemicinflammatory responses of mice during pulmonary infection with Klebsiellapneumoniae[J].J Med Microbiol,2000;Yoshida K,et al.Induction of interleukin-10and down-regulation of cytokine production by Klebsiella pneumoniae capsulein mice with pulmonary infection[J].J Med Microbiol,2001)。
细菌外膜囊泡(OMVs)是细菌外膜向外出芽形成的,并从细菌外膜和周质中获得蛋白质等生物大分子(Huang W L,et al.Bacterial outer membrane vesicles aspotential biological nanomaterials for antibacterial therapy[J].ActaBiomaterialia,2022;Avila-Calderon E D,et al.Outer Membrane Vesicles of Gram-Negative Bacteria:An Outlook on Biogenesis[J].Frontiers in Microbiology,2021)。OMVs保持与母细菌相似的膜组成和结构,因此它们能与靶菌株的外膜融合,将其包载的物质递送进细菌内部,可作为天然的药物递送材料(Gan Y Y,et al.Fight bacteriawith bacteria:Bacterial membrane vesicles as vaccines an deliverynanocarriers against bacterial infections[J].Nanomedicine-NanotechnologyBiology and Medicine,2021)。在抗菌领域,OMVs被用于包封抗生素,载药纳米粒或作为抗菌疫苗等(Solanki K S,et al.Non-infectious outer membrane vesicles derivedfrom Brucella abortus S19 Delta per as an alternative acellular vaccineprotects mice against virulent challenge[J].International Immunopharmacology,2021;Huang W W,et al.Development of novel nanoantibiotics using an outermembrane vesicle-based drug efflux mechanism[J].Journal of ControlledRelease,2020;Gao F,et al.Kill the Real with the Fake:Eliminate IntracellularStaphylococcus aureus Using Nanoparticle Coated with Its ExtracellularVesicle Membrane as Active-Targeting Drug Carrier[J].A Infectious Diseases,2019)。革兰氏阴性菌大肠杆菌的OMVs能够用于包被载有抗生素利福平的纳米粒子,通过细菌同源靶向性和较好的膜融合性能实现抗生素的高效菌内递送,从而增强利福平的抗菌作用(Wu S,et al.Bacterial Outer Membrane-Coated Mesoporous Silica Nanoparticlesfor Targeted Delivery of Antibiotic Rifampicin against Gram-NegativeBacterial Infection In Vivo[J].Advanced Functional Materials,2021)。
目前本技术领域尚无利用细菌外膜囊泡递送非抗生素类抗菌物质的技术方案,此类方案有望实现对耐药性型肺炎克雷伯氏菌感染的靶向性治疗作用。
发明内容
为了解决上述技术问题,本发明提供一种杂化膜囊泡及其制备方法,和包含所述杂化膜囊泡的杀菌剂,以及其在靶向超声杀菌中的应用。本发明提供的杂化膜囊泡具有逃避巨噬细胞吞噬的作用从而延长在感染部位滞留时间,同时通过细菌同源靶向性以及膜融合性能,结合在细菌表面或进入细菌内部,从而通过超声产生活性氧达到高效杀菌的效果。
为了实现上述目的,本发明通过以下几个方面实现:
第一方面,本发明提出了一种杂化膜囊泡,其通过姜黄衍生纳米囊泡与革兰氏阴性菌的外膜囊泡融合而得,其中所述姜黄衍生纳米囊泡与革兰氏阴性菌的蛋白质质量比小于1:0.1,其中质量以μg计。
优选地,所述革兰氏阴性菌选自肺炎克雷伯杆菌、大肠杆菌、铜绿假单胞菌和鲍曼不动杆菌中的至少一种。
更优选地,所述革兰氏阴性菌为具有荚膜结构的上述革兰氏阴性菌。
最优选地,所述革兰氏阴性菌为肺炎克雷伯杆菌。
克雷伯氏菌属(Klebsiella)为肠杆菌科中一类有荚膜的革兰氏阴性杆菌,克雷伯氏菌为较短粗的杆菌,大小(0.5-0.8)×(1-2)μm,单独、成双或短链状排列。无芽胞,无鞭毛,有较厚的荚膜,多数有菌毛。
优选地,所述姜黄衍生纳米囊泡与革兰氏阴性菌的蛋白质质量比约小于1:0.5,其中质量以g计。
更优选地,所述姜黄衍生纳米囊泡与革兰氏阴性菌的蛋白质质量比可以约为1:0.5、1:0.6、1:0.7、1:0.8、1:0.9、1:1.0、1:1.1、1:1.2、1:1.3、1:1.4、1:1.5、1:1.6、1:1.7、1:1.8、1:1.9或1:2。
最优选地,所述姜黄衍生纳米囊泡与革兰氏阴性菌的蛋白质质量比为1:2。

第二方面,本发明提出了一种制备第一方面所述的杂化膜囊泡的方法,所述方法包括以下步骤:
(1)从姜黄中提取衍生纳米囊泡TNVs(Turmeric-derived nanovesicles),得到姜黄衍生纳米囊泡;
(2)从革兰氏阴性菌中提取其外膜囊泡,得到革兰氏阴性菌外膜囊泡;
(3)将所述姜黄纳米囊泡TNVs与所述革兰氏阴性菌外膜囊泡混合后超声融合,得到杂化膜囊泡。
优选地,所述超声条件为功率50W,2s开,3s关,超声10分钟。
优选地,所述步骤(1)包括以下步骤:
1)提取姜黄汁液,低速离心后,得上清液;
2)将所述上清液超速离心后,得沉淀;
3)将所述沉淀重悬后进行蔗糖密度梯度离心,收集上层样品和下层样品;
其中上层样品为姜黄纳米囊泡TNVs的第一组分,下层样品为姜黄纳米囊泡TNVs的第二组分。
优选地,将纳米囊泡TNVs的第二组分与革兰氏阴性菌外膜囊泡融合。
优选地,所述蔗糖密度为8%-45%。
第三方面,本发明提出了一种靶向抗菌剂,其包含有效抗菌含量的根据第一方面所述的杂化膜囊泡和/或根据第二方面所述的方法制备得到的杂化膜囊泡,和药学上可接受的辅料。
优选地,所述辅料选自稀释剂、黏合剂、崩解剂、润滑剂、助流剂、抗结块剂、包衣剂、增塑剂、表面活性剂、栓剂基质、助悬剂、增稠剂、软膏基、络合剂、保湿剂、成膜剂、冻干保护剂、干粉吸入剂、乳化剂、释放调节剂、压敏胶黏剂和硬化剂中的至少一种。
优选地,所述靶向抗菌剂的剂型选自注射剂、片剂、丸剂、胶囊、悬浮和乳剂中的至少一种。
第四方面,本发明提出了一种根据第一方面所述的杂化膜囊泡、根据第二方面所述的方法制备得到的杂化膜囊泡和/或根据第三方面所述的靶向抗菌剂在靶向超声抗菌中的应用。
优选地,所述菌为革兰氏阴性菌。

第五方面,本发明提出了一种根据第一方面所述的杂化膜囊泡、根据第二方面所述的方法制备得到的杂化膜囊泡和/或根据第三方面所述的靶向抗菌剂在制备预防和/或治疗革兰氏阴性菌感染的药物中的应用。
优选地,所述革兰氏阴性菌选自肺炎克雷伯杆菌、大肠杆菌、铜绿假单胞菌、幽门螺旋杆菌和鲍曼不动杆菌中的至少一种。
更优选地,所述革兰氏阴性菌为具有荚膜结构的上述革兰氏阴性菌。
最优选地,所述革兰氏阴性菌为肺炎克雷伯杆菌。
本发明提供的杂化膜囊泡和/或含其的靶向抗菌剂至少具有以下有益效果:
(1)具有逃避巨噬细胞吞噬的作用,从而延长在感染部位滞留时间,可在感染部位滞留6h以上;
(2)较佳的同源靶向性以及膜融合性能,可靶向宿主革兰氏阴性杆菌并与其外膜融合,从而结合在细菌表面或进入细菌内部,增强其包载的抗菌物质与细菌的相互作用;
(3)良好的生物膜穿透能力,杂化膜囊泡结合超声处理,可清除宿主56%的生物被膜;
(4)高效的超声抗菌能力,在TNVs与OMVs比例为1:2时,几乎完全清除细菌。

植物衍生外泌体(Plant-derived nanovesicles)含有多种生物活性成分,已有研究者在姜黄衍生外膜囊泡中鉴定出特定的脂质、蛋白质和生物活性成分姜黄素,且在结肠炎模型中表现出优异的抗炎和抗氧化潜力(Liu C,et al.Oral administration ofturmeric-derived exosome-like nanovesicles with anti-inflammatory and pro-resolving bioactions for murine colitis therapy[J].Journal ofNanobiotechnology,2022)。而姜黄素是一种从植物姜黄中提取的天然多酚,也是一种天然声敏剂,它可以破坏细菌细胞膜,抑制细菌毒力因子产生,诱导氧化应激等途径发挥抑菌作用。
本发明首次发现将姜黄衍生纳米囊泡(TNVs)和肺炎克雷伯菌外膜囊泡(OMVs)超声融合,获得了杂化膜囊泡(TNVs-OMVs),其具备姜黄外泌体的声动力抗菌作用,同时保留了亲本细菌的荚膜多糖。这使得该杂化膜囊泡具有逃避巨噬细胞吞噬的作用从而延长在感染部位滞留时间,同时通过细菌同源靶向性以及膜融合性能,结合在细菌表面或进入细菌内部,从而通过超声(US,ultrasonic)作用下产生ROS达到高效杀菌的效果。在体内肺部感染模型中显示出良好的治疗效果(图1)。
为使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明进行进一步的详细说明。此处所描述的具体实施例仅用于解释本发明,并不用于构成对本发明的任何限制。此外,在以下说明中,省略了对公知结构和技术的描述,以避免不必要地混淆本公开的概念。这样的结构和技术在许多出版物中也进行了描述。
本发明所使用的材料与试剂:
姜黄购于安庆乐图商贸有限公司;
本发明使用的肺炎克雷伯菌株015134(B0菌株)是2017年4月从中国华西医院ICU一名患者的直肠拭子中分离出来的ST11,产生KPC-2菌株,代表中国医院中占主导地位的ST11-KL64-blaKPC-2克隆。015134荚膜基因突变株B1-1_ΔmshA(简写成B1-1)以及荚膜基因回补株B1-1::pmshA,均来源于四川大学华西医院;其中荚膜基因突变株B1-1,相对于野生型部分荚膜基因缺失,荚膜含量减少;荚膜基因回补株B1-1::pmshA是在突变株的基础上导入缺失的荚膜基因,荚膜含量比突变株有所增多,与野生型菌株相当。
本发明使用的RAW264.7细胞系购自武汉普诺赛生命科技有限公司;
本发明采用Balb/c小鼠(6-8周,雌性,体重21-23克,SPF级)作为动物实验对象,购买自深圳市迈科生物科技有限公司,动物生产许可证号:SCXK(粤)2020-0051,动物合格证编号:44822700016573。实验期间小鼠可自由饮水,饲养于洁净区。
结晶紫染色方法:
取10μL的B0细菌冻存液,加入10mL LB培养基中过夜培养。将细菌培养液稀释至OD600为0.1(即细菌密度约为108CFU mL-1),以1mL/孔的体积加入12孔板中,在恒温培养箱培养24h后,移去孔板中的上清液,再加入新鲜LB培养基,再培养24h。取出孔板,去掉培养基。以1mL/孔的体积加入用LB分散的各样品溶液,超声后(功率15%,频率1.0mHz,占空比50%,4min),去掉培养基,按1mL/孔体积加入0.02%的结晶紫溶液,放入恒温培养箱孵育30min。去掉结晶紫溶液,按1mL/孔体积加入去离子水清洗。去掉去离子水,加入无水乙醇(1mL/孔),放入摇床孵育15min。使用酶标仪吸光度测定OD570处的吸光度。
流式细胞术分析:
样品与2μM DiO于37℃孵育样品1h,离心(15000rpm,4℃,1h),弃上清,PBS重悬样品,4℃保存待用。
用DMEM(含10%FBS,1%P/S)培养Raw 264.7细胞,以每孔5×105个细胞的密度接种到12孔板中,待细胞贴壁后,按照20μg/mL加入荧光标记的各样品,孵育2或6小时(5%CO2,37℃),PBS作为对照,随后,通过流式细胞术分析所有样品。
实施例1姜黄衍生纳米囊泡的制备与表征
姜黄衍生纳米囊泡的制备过程如下:
将姜黄(购于安庆乐图商贸有限公司)表面附着的泥渍洗净,削皮称重,按1mL/g加入PBS,榨汁,用无菌纱布过滤榨汁所得汁液,除去植物粗纤维,得到姜黄匀浆液。先低速离心(3000g,4℃,30min),取上清液。再离心(10000g,4℃,1h),取上清液超速离心(100000g,4℃,1h),收集沉淀,重悬于PBS中。进一步纯化TNVs,用保存在4℃的20mM,pH 7.4Tris-HCl配置8%、30%、45%蔗糖梯度溶液,超离管中按照蔗糖浓度从低到高的顺序,依次从底部加入蔗糖溶液,最上层沿管壁加入1mL超速离心所得样品溶液,超速离心(100000g,4℃,1h),收集不同密度分界处的样品,得到8-30%、30-45%上下两层,第一组分TNVs 1和第二组分TNVs 2(图2)。将两种组分转移至100kDa超滤管中离心(4℃,3000g,30min),除去蔗糖,用PBS收集姜黄衍生纳米囊泡,-80℃保存。
透射电镜图像显示,8-30%、30-45%上下两层的TNVs 1和TNVs 2均具有外泌体样囊泡样形态(图3),水合粒径分别为187.0nm和228.3nm,zeta电位分别为-12.93和-15.09,姜黄素含量为3.68%和3.64%,见表1。

随后,将1,3-二苯基异苯并呋喃(DPBF)单线态氧指示探针与TNVs 2孵育后分别超声0min、1min、3min、5min后检测紫外-可见吸收光谱,在410nm出现的吸收峰表明单线态氧的产生。将SOSG单线态氧绿色荧光探针与TNVs 2孵育后分别超声0min、1min、3min、5min的检测荧光光谱,在526nm的特征峰强度的提高表明单线态氧的产量提高。两者结果均显示,在超声条件下,姜黄衍生纳米囊泡TNVs可以产生单线态氧这一类的活性氧(图4和图5),从而可利用超声作用下产生的活性氧发挥抗菌作用。
为了验证这一假设,发明人在K.pneumoniae中分别加入TNVs 1和TNVs 2,超声(功率1.5W/cm2,频率1.0mHz,占空比50%,4min)后取菌液稀释104倍,涂布于LB琼脂平板,37℃培养20h后,对平板上的菌落进行拍照与计数(图6和图7)。与US组相比时,TNVs 1组与TNVs2组细菌存活率都更低,表明TNVs 1与TNVs 2都具有一定抗菌效果。相同浓度下,TNVs 1与TNVs 2的抗菌效果均成剂量依赖特性,并且TNVs 1组与TNVs 2组相比,TNVs 2组引起了K.pneumoniae菌落显著减少,表明TNVs 2具有更好的抗菌效果,故后续研究都使用TNVs 2。
实施例2杂化膜囊泡的制备与表征
杂化膜囊泡的制备过程如下:
分别提取肺炎克雷伯菌的外膜囊泡(Outer membrane vesicles,OMVs),肺炎克雷伯菌荚膜突变株外膜囊泡B1-1-OMVs以及肺炎克雷伯菌荚膜回补株外膜囊泡B1-1::pmshA-OMVs。获OMVs的方法为10mL LB液体培养基中加入10μL菌株冻存液,过夜培养,将10mL培养基全部转移至800mL LB液体培养基中培养24h(37℃,220rpm)。先低速离心去除细菌(4500g,4℃,10min)。取上清超速离心(100000g,4℃,1h),收集沉淀,并用PBS洗涤,分散于PBS,-80℃保存。随后,发明人将TNVs和OMVs以1:2的蛋白质质量比混合,通过探头超声(功率50W,2s开,3s关,10min)融合形成TNVs-OMVs。其中蛋白质质量比通过普通的蛋白定量计算。
透射电镜图像显示,杂化形成的三种囊泡形态相似(图8)。B1-1-OMVs、B1-1::pmshA-OMVs、OMVs以及三种杂化后的TNVs-OMVs的水合粒径测定,显示杂化后TNVs-OMVs的水合粒径为250.67nm,水合粒径不同于单独的TNVs与OMVs,表明两种膜能够融合成功(图9)。为了进一步验证两种膜能否融合,发明人利用1,1’-二十八烷基-3,3,3’,3’-四甲基碳菁高氯酸盐(DiI,红色)与3,3’-二十八烷基沙碳氰酸高氯酸盐(DiO,绿色)作为具有共振能量转移(FRET)特性的染料对,对TNVs进行标记,然后以不同蛋白质质量比与未标记的OMVs进行融合。随着OMVs所占比例的增加,DiO的荧光发射增加,DiI的荧光发射减少,这是由于OMVs的插入使TNVs上的FRET特性减弱,也表明OMVs和TNVs的成功融合(图10)。此外,发明人也通过共聚焦显微镜(图11)和超分辨共聚焦显微镜(图12)观察膜融合情况,用DiI标记TNVs,DiO标记OMVs,通过超声形成的TNVs-OMVs的红色和绿色荧光重叠形成黄色区域,表明膜融合成功。
随后,通过DPBF和SOSG溶液与TNVs-OMVs共孵育得到的谱图(图13和图14),表明在超声条件下杂化膜囊泡TNVs-OMVs能够产生活性氧,发挥抗菌作用。在探究不同比例制备得到的杂化膜囊泡抗菌实验中,杂化膜囊泡的抗菌效果优于单纯的姜黄衍生纳米囊泡TNVs,且TNVs与OMVs比例为1:2时的细菌存活率最低,抗菌效果最佳(图15)。这是由于OMVs的存在,促进了TNVs-OMVs被细菌摄取或结合在细菌表面,从而通过声动力产生活性氧,增强对细菌的杀伤作用。杂化膜囊泡中,OMVs比例的提高可以提高抗菌效果。
实施例3巨噬细胞对杂化膜囊泡的摄取探究
实施例2制备的OMVs同样具有肺炎克雷伯菌外膜上的荚膜成分,荚膜可帮助肺炎克雷伯菌逃避免疫清除。为了验证TNVs-OMVs是否保留OMVs的这一特性,发明人将TNVs-OMVs(DiO标记)与巨噬细胞Raw 264.7共孵育。
将样品与2μM DiO于37℃孵育样品1h,离心(15000rpm,4℃,1h),弃上清,PBS重悬样品,4℃保存待用。用DMEM(含10%FBS,1%P/S)培养Raw 264.7细胞,以每孔5×105个细胞的密度接种到12孔板中,待细胞贴壁后,按照20μg/mL加入荧光标记的各样品,孵育2或6小时(5% CO2,37℃),PBS作为对照,随后,通过流式细胞术分析所有样品。
共聚焦激光扫描显微镜(CLSM)图像显示,在2h可观察到Hoechst标记的细胞核周围具有较强的TNVs-B1-1-OMVs的DiO荧光(图16)。而TNVs-OMVs组以及荚膜回补株的B1-1::pmshA-OMVs制备得到的TNVs-B1-1::pmshA-OMVs组还不能观察到明显的荧光。在6h的共聚焦图像中才在TNVs-OMVs组观察到了较弱的绿色荧光(图16)。表明荚膜有助于TNVs-OMVs逃避巨噬细胞的摄取,从而避免被巨噬细胞清除。为了定量与细胞结合或者摄取的纳米囊泡,发明人采用了流式细胞术定量不同样品在巨噬细胞中的荧光强度。与CLSM结果一致,TNVs-OMVs组中的DiO(标记TNVs-OMVs)的平均荧光强度明显低于TNVs-B1-1-OMVs组(图17),表明荚膜使得TNVs-OMVs具有逃避巨噬细胞摄取的能力。
此外,发明人通过酶联免疫吸附试验(ELISA)检测巨噬细胞Raw 264.7(购自武汉普诺赛生命科技有限公司)分泌的促炎细胞因子白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)水平,可用于验证TNVs-OMVs是否能激活巨噬细胞,促进炎症的发生进而激活固有免疫(图18和图19)。实验结果表明,OMVs组的IL-6和TNF-a水平显著低于B1-1-OMVs组,而杂化后TNVs-OMVs组的IL-6和TNF-a水平较低于TNVs-B1-1-OMVs,且与TNVs-B1-1::pmshA-OMVs组水平相当。综上所述,荚膜赋予了TNVs-OMVs逃避巨噬细胞吞噬的能力,具有能够逃避固有免疫,从而延长作用于细菌感染部位时间的潜力。
实施例4同源细菌对杂化膜囊泡的摄取研究
由于膜成分相似,由E.coli衍生的OMVs包裹的纳米颗粒被证明可以被E.coli有效内化(Solanki K S,et al.Non-infectious outer membrane vesicles derived fromBrucella abortus S19 Delta per as an alternative acellular vaccine protectsmice against virulent challenge[J].International Immunopharmacology,2021)。为了验证TNVs-OMVs是否保留了OMVs的这一特性,发明人将TNVs-OMVs(DiO标记)分别与革兰氏阳性菌S.aureus和革兰氏阴性菌E.coli,K.pneumoniae孵育。CLSM图像显示K.pneumoniae内部具有较强的TNVs-OMVs的DiO荧光,而S.aureus内部的DiO荧光较弱,表明TNVs-OMVs更容易被同源细菌摄取,具有同源细菌靶向作用(图20和图21)。同时,在革兰氏阴性菌E.coli内部也可观察到明显的TNVs-OMVs荧光,说明E.coli亦可有效摄取TNVs-OMVs,这是因为革兰氏阴性菌之间具有相似的细胞膜成分与结构。
接下来,发明人通过流式细胞术测量了TNVs-OMVs在不同细菌中的平均荧光强度(图22)。结果显示,K.pneumoniae对TNVs-OMVs的摄取最强,这表明TNVs-OMVs可以选择性地靶向特定的细菌。此外,发明人也通过CLSM比较了TNVs-B1-1-OMVs、TNVs-B1-1::pmshA-OMVs与TNVs-OMVs的同源细菌靶向能力(图23)。与TNVs-OMVs组的强荧光不同,K.pneumoniae中TNVs-B1-1-OMVs和TNVs-B1-1::pmshA-OMVs的荧光难以被观察到,表明OMVs对于同源细菌的靶向性最强,流式细胞术也证实了这一结论(图24和图25),这可能是由于OMVs相较TNVs-B1-1-OMVs与B1-1::pmshA-OMVs,其膜结构更接近野生型细菌的外膜。
实施例5杂化膜囊泡的体外抗菌探究
根据前述实验结果,TNVs-OMVs可以促进同源细菌的摄取,这有利于TNVs-OMVs进入K.pneumoniae或与K.pneumoniae表面结合,从而增强TNVs-OMVs的体外抗菌作用。为了验证这一假设,发明人分别将含不同药物TNVs、TNVs-B1-1-OMVs、TNVs-B1-1::pmshA-OMVs和TNVs-OMVs与K.pneumoniae共孵育。然后进行超声,随后稀释不同倍数进行涂板培养20h后,对琼脂平板上细菌菌落拍照和计数来考察不同样品的对浮游K.pneumoniae的抗菌活性(图26和图27)。与超声后的TNVs组相比,TNVs-B1-1-OMVs组和TNVs-B1-1::pmshA-OMVs组引起了K.pneumoniae菌落的减少,而TNVs-OMVs组中未观察到K.pneumoniae菌落,这可能与不同样品的同源细菌摄取情况的不同有关。这些结果均表明,杂化膜囊米囊泡TNVs-OMVs具有良好的体外抗游离菌效果。
生物膜是由细菌及其由胞外聚合物质(extracellular polymeric substances)组成,可以降低免疫系统和药物对细菌的杀菌作用。清除已形成的生物膜通常比清除浮游细菌更困难,故破坏生物膜的能力也成为了判断抗菌药物是否具有临床应用潜力的重要指标。TNVs-OMVs已显示出良好的体外抗游离菌效果,发明人进一步使用活/死染色试剂盒对生物膜进行染色,并用激光共聚焦显微镜拍摄经样品与超声处理后的生物膜Z-stack图像,以观察生物膜完整性(图28),超声可以对生物膜造成有限的破坏,与对照组相比,超声结合TNVs-OMVs处理,导致了最大程度生物膜中的细菌溶解,生物膜的破坏。横断面观察到的红色荧光,表明通过声动力学过程产生的ROS可以穿透生物膜,杀死生物膜深处的细菌。同时通过结晶紫染色法考察其对K.pneumoniae生物膜的清除能力(图29),TNVs-OMVs结合超声处理造成了56%的生物被膜清除。以上结果说明,TNVs-OMVs在超声作用下的抗生物膜活性最高。
实施例6杂化膜囊泡的体内治疗研究
为了评估TNVs-OMVs体内治疗K.pneumoniae感染的效果,将健康的Balb/c小鼠随机分为7组(Control组、US组、OMVs组、TNVs组、TNVs+US组、TNVs-OMVs组和TNVs-OMVs+US组),每组5只小鼠。发明人通过气管内雾化注射肺炎克雷伯菌(1×108CFU/20g,即每20g小鼠注射1×108CFU肺炎克雷伯菌)建立肺部感染小鼠模型,在感染24h后气管内雾化注射不同浓度的TNVs-OMVs(10μg/g,即每g小鼠注射10μg的TNVs-OMVs),超声处理(功率15%,频率1.0mHz,占空比50%,4min)后处死小鼠,提取小鼠肺部,称重,加入5mL PBS,使用组织研磨仪于冰浴条件下研磨,获得肺部混悬液,PBS稀释一定倍数(101、103、105倍)后后涂布于LB琼脂平板,培养20h后,对平板上的菌落进行计数(图30)。
结果显示,细菌的清除能力与药物之间呈剂量依赖性,且给予TNVs-OMVs的剂量为10μg/g时,对细菌的清除效果最佳(图31)。随后对比不同处理组的肺部的菌落数量(图32),尽管TNVs+US组能实现了一定程度的细菌清除(相对于PBS组其菌落数降低0.51-lg CFU),但TNVs-OMVs+US组显示出更强的抗菌能力(相对于PBS组其菌落数降低2.06-lg CFU),说明杂化膜囊泡TNVs-OMVs在体内也能发挥显著声动力抗菌功能。TNVs-OMVs相较于单独的姜黄衍生纳米囊泡TNVs具有更强的体内杀菌能力,这可能主要是由于OMVs的靶向作用。以上结果证明,将TNVs与OMVs杂化后形成的杂化膜粒,针对体内耐碳青霉烯肺炎克雷伯菌造成的肺部感染具有较好的治疗效果。
最后应当说明的是,以上内容仅用以说明本发明的技术方案,而非对本发明保护范围的限制,本领域的普通技术人员对本发明的技术方案进行的简单修改或者等同替换,均不脱离本发明技术方案的实质和范围。
Claims (10)
1.一种杂化膜囊泡,其特征在于,其通过姜黄衍生纳米囊泡与革兰氏阴性菌的外膜囊泡融合而得,其中所述姜黄衍生纳米囊泡与革兰氏阴性菌的蛋白质质量比小于1:0.1。
2.根据权利要求1所述的杂化膜囊泡,其特征在于,所述革兰氏阴性菌选自肺炎克雷伯杆菌、大肠杆菌、铜绿假单胞菌和鲍曼不动杆菌中的至少一种;
较佳地,所述革兰氏阴性菌为肺炎克雷伯杆菌。
3.根据权利要求1或2所述的杂化膜囊泡,其特征在于,所述革兰氏阴性菌具有荚膜结构。
4.根据权利要求3所述的杂化膜囊泡,其特征在于,所述姜黄衍生纳米囊泡与革兰氏阴性菌的外膜囊泡的蛋白质质量比小于1:0.5;
较佳地,所述姜黄衍生纳米囊泡与革兰氏阴性菌的外膜囊泡的蛋白质质量比为1:2。
5.根据权利要求3所述的杂化膜囊泡,其特征在于,所述姜黄衍生纳米囊泡的水合粒径为170nm-260nm;
和/或,所述姜黄衍生纳米囊泡的zeta电位为-15mv~-13mv;
和/或,所述姜黄衍生纳米囊泡中姜黄素含量为1%-7%;
较佳地,所述姜黄衍生纳米囊泡中姜黄素含量为3%-4%。
6.根据权利要求3所述的杂化膜囊泡,其特征在于,所述杂化膜囊泡的水合粒径为100nm-300nm;
较佳地,所述杂化膜囊泡的水合粒径为200nm-300nm。
7.一种制备根据权利要求1-6中任一所述的杂化膜囊泡的方法,其特征在于,所述方法包括以下步骤:
(1)从姜黄中提取衍生纳米囊泡TNVs,得到姜黄衍生纳米囊泡;
(2)从革兰氏阴性菌中提取其外膜囊泡,得到革兰氏阴性菌外膜囊泡;
(3)将所述姜黄衍生纳米囊泡TNVs与所述革兰氏阴性菌外膜囊泡混合后超声融合,得到杂化膜囊泡。
8.一种靶向抗菌剂,其特征在于,包含有效抗菌含量的根据权利要求1-6中任一所述的杂化膜囊泡和/或根据权利要求7所述的方法制备得到的杂化膜囊泡,和药学上可接受的辅料;
较佳地,所述辅料选自稀释剂、黏合剂、崩解剂、润滑剂、助流剂、抗结块剂、包衣剂、增塑剂、表面活性剂、栓剂基质、助悬剂、增稠剂、软膏基、络合剂、保湿剂、成膜剂、冻干保护剂、干粉吸入剂、乳化剂、释放调节剂、压敏胶黏剂和硬化剂中的至少一种;
较佳地,所述靶向抗菌剂的剂型选自注射剂、片剂、丸剂、胶囊、悬浮和乳剂中的至少一种。
9.根据权利要求1-6中任一所述的杂化膜囊泡、根据权利要求7中所述的方法制备得到的杂化膜囊泡和/或根据权利要求8所述的靶向抗菌剂在靶向超声抗菌中的应用。
10.根据权利要求1-6中任一所述的杂化膜囊泡、根据权利要求7所述的方法制备得到的杂化膜囊泡和/或根据权利要求8所述的靶向抗菌剂在制备预防和/或治疗革兰氏阴性菌感染的药物中的应用。
