Description

Poultry feed containing bacillus natto for improving immunity of broiler chickens
Technical Field
The invention belongs to the technical field of broiler immunity regulation and poultry feed preparation, and particularly relates to a poultry feed containing bacillus natto for improving broiler immunity.
Background
At present, the intensive broiler raising mode has caused serious stress on broiler chickens and has serious negative influence on the production performance of the broiler chickens. Research shows that high stress can damage intestinal health of broiler chickens, resulting in intestinal villi fracture, impaired mucosal epithelial barrier, epithelial oxidative stress, inflammatory reaction, and microbiota disorder. The damaged intestinal tract not only affects the absorption of nutrients, but also is easy to cause toxin to invade the body, and further affects the production performance of broilers. Therefore, the preparation of a novel microecological preparation capable of regulating the intestinal health of broiler chickens and maintaining the intestinal homeostasis becomes a problem to be solved urgently in the broiler industry.
Research has reported that bacillus natto can produce competitive inhibition effect on harmful microorganisms in intestinal tractsThe bacterium self-metabolite can also increase the acidity in the intestinal tract, promote the production of various amino acids and digestive enzymes, thereby improving the absorption and utilization rate of the feed and enhancing the immunity of organisms. In recent years, feed probiotics are increasingly applied to livestock production, for example, chinese patent CN104381211B (Jiangsu Jiashi) discloses a breeding method for improving chicken immunity, which comprises the following steps of: the chicken feed comprises the following components in parts by weight: 5-10 parts of fish meal, 40-60 parts of corn, 30-50 parts of bean pulp, 10-15 parts of eggshells and 10-20 parts of bacillus natto; prevention and treatment of common chicken diseases: boiling 10-30 parts of dyers woad leaf, 20-25 parts of radix isatidis and 5-10 parts of garlic, drinking the chicken at a frequency of 2-3 times per week, and sterilizing the chicken shed by using a sterilizing liquid at regular intervals, wherein the sterilizing liquid comprises the following components: 25-30% of hydrogen peroxide, 30-40% of sodium chloride, 5-10% of undecylimidazoline and 10-20% of bacillus natto. The feed used by the method contains bacillus natto, and the bacillus natto can be quickly propagated after entering the intestinal canal of the chicken to generate a large amount of vitamins, amino acids and various enzymatic growth factors, so that the feed utilization rate can be improved, pathogenic bacteria propagation can be inhibited, diseases can be reduced, various proteases (especially alkaline proteases), saccharifying enzymes, lipases and amylases can be generated, complex carbohydrates in the plant feed can be degraded, and the conversion utilization rate of the feed can be improved. Meanwhile, the bacillus natto serving as one of the components of the disinfectant can improve the culture environment, reduce the generation of harmful substances in the environment, promote the decomposition of organic pollutants such as feces, residual baits and the like generated in the culture process, and reduce the NH of the harmful substances 3 、NO 2 、H 2 And S, generating. However, bacillus natto is easy to be influenced by environmental factors in the intestinal tracts of broilers after entering the intestinal tracts of broilers, and is unfavorable for mass propagation, so that the effect of bacillus natto cannot be fully exerted if only bacillus natto is added into conventional feed. In order to solve the problem, chinese patent application CN105707451A (Hospital of fertilizer application) provides a special feed for improving immunity of broiler chickens, wherein a complex enzyme preparation, anhydrous betaine, a feed additive,Attapulgite clay and traditional Chinese medicine components of liquorice, cortex moutan and radix scutellariae. Proved by verification, the addition of the substances can provide a more favorable environment for the propagation of bacillus natto in the intestinal tracts of the broiler chickens, so that the efficacy of the bacillus natto is promoted, the body immunity and the digestion function of the broiler chickens are improved, the microecological balance of the intestinal tracts is maintained, and the morbidity and the mortality are reduced. However, the feed provided by the patent application has complex formula composition, traditional Chinese medicine components are easy to cause intestinal stress of broiler chickens, and the use cost of the compound enzyme preparation is higher, so that the compound enzyme preparation is not beneficial to industrialized application.

Test examples
1. Test method
A fully random experiment is adopted, a broiler stress model is established by using lipopolysaccharide stimulation, bacillus natto premix is added or not added in basic ration of the broiler, and the growth performance, the antioxidant capacity and the immune function of the broiler in a stress state are studied. The specific experimental method is as follows: 144 broiler chickens 500 in the age of 1 day with close weight were selected and randomly divided into 3 treatment groups, 6 replicates per treatment, 8 chickens per replicate, and male and female mixed culture.
Control group: basic diet (composition same as example 1) was fed, and physiological saline was injected 1mL on 1, 3, 5 days per week for two weeks from the age of 8 days.
Test 1 group: the control group was fed with basal diet, and 1mL of LPS dissolved in physiological saline was intraperitoneally injected at 500. Mu.g/kg BW on 1, 3, 5 days per week starting at 8 days old, and the injection was continued for two weeks.
Test 2 group: 300 g/ton of Bacillus natto premix (composition same as in example 1) was added to the basic ration from 1 day old, and 1mL of LPS dissolved in physiological saline was intraperitoneally injected at 500. Mu.g/kg BW for 1, 3, 5 days per week from 8 days old for two weeks.
At the initial stage of the experiment, the weight difference of the broilers in each treatment group was not significant. Raising all chickens in an automatic temperature-controlled and ventilated broiler house according to the variety raising manual, and three-layer cage raising with cage area of 1.5m 2 (1.2mX1.25 m) the cage is 0.5m high. The chickens only eat and drink water freely, the light and dark proportion is 23:1, meanwhile, the chickens are immunized according to the conventional broiler chicken immunization program, the feed intake and daily gain of the chickens are counted, the activities/contents of superoxide dismutase (SOD), glutathione peroxidase (GSH-px) and Malondialdehyde (MDA) in serum (the three are important indexes reflecting the oxidation stress level) and the total antioxidant capacity (T-AOC) are detected at 14d and 42d, meanwhile, the contents of ileum mucosa SIgA, IL-6 and TNF-alpha are tested at 14d and 42d, and the influence of the bacillus natto premix on the growth of the broilers is evaluated from three aspects of the broiler chicken growth performance, the antioxidant capacity and the immunity, and the results are shown in figures 1-8.
2. Test results
(1) Influence of bacillus natto premix on growth performance of broiler chickens
As shown in figure 1, the feed intake and average daily gain of the broiler chickens of the test group 2 are higher than those of the broiler chickens of the test group 1 and the control group, and the fact that a small amount of bacillus natto premix is added into basic daily ration is beneficial to improving the growth performance of the broiler chickens, relieving the adverse stress caused by lipopolysaccharide and having a certain forward promotion effect.
(2) Influence of bacillus natto premix on oxidation resistance of broiler chickens
As shown in fig. 2-5, when the broiler chicken is raised to 14d, compared with the control group, the serum superoxide dismutase (SOD) activity of the broiler chicken in the test group 1 has an increasing trend, the glutathione peroxidase (GSH-px) activity is obviously reduced, the Malondialdehyde (MDA) content is obviously increased, and the reduction of the oxidation resistance of the body caused by the lipopolysaccharide-induced stress is indicated. Compared with the test 1, the test 2 has the advantages that the activities of SOD and GSH-px are both increased, and the MDA content is obviously reduced, which indicates that the problem of oxidation resistance reduction caused by lipopolysaccharide induction can be relieved when the bacillus natto premix is fed in the early stage (1-14 d) of feeding. When fed to 42d, GSH-px activity and MDA content of the test group 2 are obviously increased compared with those of the test group 1 and the control group. And, the total antioxidant capacity of the broiler serum of the test 2 group fed with the bacillus natto premix is obviously higher than that of the test 1 group and the control group. The poor stress caused by lipopolysaccharide can reduce the oxidation resistance of broiler chicken organisms, and the oxidation resistance of broiler chicken can be improved to a certain extent by adding a small amount of bacillus natto premix in basic ration.
(3) Influence of bacillus natto premix on immunity of broiler chickens
Intestinal mucosa SIgA is a major immunoglobulin and also an important defense line against various endogenous symbiotic bacteria and exogenous pathogenic bacteria. As shown in fig. 6-8, the ileal intestinal mucosa SIgA content of the broiler chickens of the test group 2 is significantly higher than that of the test group 1 and the control group when fed for 42d, which indicates that the addition of a small amount of bacillus natto premix in the basic ration is beneficial to improving the intestinal barrier function and relieving the inflammatory reaction caused by lipopolysaccharide; when the feed is fed to 14d, the content of IL-6 in the ileum intestinal mucosa of the broiler chicken in the test 1 group is obviously increased, the change of the content of TNF-alpha in the test 2 group is not great, but the content of IL-6 and TNF-alpha in each group is obviously lower than that in the test 1 group and the control group, and when the feed is fed to 42d, the difference of the content of IL-6 and the content of TNF-alpha in each group is not obvious, so that the intestinal inflammatory reaction can be reduced by adding the bacillus natto premix.
In conclusion, the invention utilizes lipopolysaccharide stimulation to establish a stress model, and bacillus natto premix is added or not added into basic ration of broiler chickens, so that the influence on the growth performance, the antioxidation capability and the immune function of broiler chickens in a stress state is studied, and the result shows that the addition of a small amount of bacillus natto premix can effectively reduce inflammatory reaction of intestinal tracts, protect intestinal health and improve the immunity of organisms, thereby promoting healthy growth of broiler chickens.
Although the technical solutions of the present invention have been described in detail in the foregoing general description, the specific embodiments and the experimental examples, it should be noted that the examples and the experimental examples are only for illustrating the technical solutions and the technical effects of the present invention, and should not be construed as limiting the scope of the present invention in any way. Any simple deformation, modification or improvement made based on the technical idea of the present invention belongs to the protection scope of the present invention.
