Abstract
本发明提供了一种保护胃黏膜的中药组合物及其制法与应用。该中药组合物包含下述重量配比的原料:燕麦1‑40份、小米1‑30份、山药1‑30份、白扁豆1‑20份、猴头菇0.1‑5份、铁皮石斛0.1‑5份、茯苓0.1‑5份、银耳0.1‑5份。该中药组合物具有改善胃黏膜充血水肿、糜烂,显著降低MDA、TNF‑α水平,升高SOD酶活力的作用。
Description
技术领域
本发明涉及中药技术领域,具体涉及一种具有保护胃黏膜功能的中药组合物的配方及其制备方法。
背景技术
当今社会,快节奏的工作与生活让许多人饮食不规律,这也催生了许多健康问题。全球胃、肠病患者从1985年的1.5亿人增至2011年的5亿人,预计到2025年将达7亿人。胃的消化功能主要依靠机械和化学作用来完成的,胃粘膜作为胃内与食物接触最多的部位,容易出问题,正常情况下,胃粘膜分泌的黏液和胃粘膜表面上皮等黏膜保护因子能保护胃黏膜免受机械摩擦、热刺激、胃酸、胃蛋白酶、细菌等黏膜攻击因子的侵害,从而预防胃黏膜损伤,但往往由于现代人工作节奏快﹑精神紧张、大量饮酒、吸烟、长期饮浓茶﹑饮食不规律等因素,导致黏膜攻击因子加强,黏膜保护因子减弱,造成胃黏膜损伤,甚至逐渐发展成为慢性胃炎、胃溃疡等胃部疾病。胃病的发生和发展在一定程度上均与胃黏膜损伤有关。
中医学认为,脾胃在人体的生命活动中发挥着至关重要的作用。脾为后天之本,主统血,主肌肉,主一身水谷精微的运化输布,为气血生化之源。脾胃为人体生命活动的本源,人体的滋养依赖脾胃功能的正常。脾运正常,则可运化饮食中的水谷精微使之转输于全身,以满足人体正常的生理需求,使人精神充足,体力充沛。脾统血功能正常,则血行于脉内而不外溢,使人体气血充足。脾失健运,则纳少、不欲饮食、腹痛腹胀。若水谷精微难以吸收,机体失养,可见面黄肌瘦、肌肉筋骨不充。
很多人会选择服用药物来调节脾胃问题,临床上用于治疗胃黏膜损伤的药品主要有胃酸中和药、H2受体拮抗剂、H+泵抑制剂等,然而,服用这些药物可能存在治疗时间长,愈合缓慢,复发率高,副作用大等问题。
发明内容
有鉴于此,本发明的目的在于提供一种具有保护胃黏膜功效的中药组合物配方及其制备方法。
为了实现上述目的,本发明采用以下技术方案:
本发明首先提供一种中药组合物,其包含下述重量配比的原料:燕麦1-40份、小米1-30份、山药1-30份、白扁豆1-20份、猴头菇0.1-5份、铁皮石斛0.1-5份、茯苓0.1-5份、银耳0.1-5份。本发明的组合物可以直接研磨成粉,也可以是经过本领域常规手段制得的提取物等。本发明组合物中使用的中药也可以以直接研磨成粉、提取物、或者其它加工形态的方式使用。
优选地,所述中药组合物包括:燕麦15-30份、小米10-20份、山药3-12份、白扁豆2-10份、猴头菇0.5-4份、铁皮石斛0.5-4份、茯苓0.5-4份、银耳0.5-4份。
优选地,所述中药组合物包括:燕麦20-25份、小米12-15份、山药5-9份、白扁豆2-6份、猴头菇0.5-3份、铁皮石斛0.5-3份、茯苓0.5-3份、银耳0.5-3份。
优选地,所述中药组合物包括:燕麦20-22份、小米12-14份、山药5-6份、白扁豆2-4份、猴头菇0.5-1份、铁皮石斛0.5-1份、茯苓0.5-1份、银耳0.5-1份。
更优选地,本发明所述的中药组合物包括以下重量份数的组分:燕麦22份、小米14份、山药6份、白扁豆4份、猴头菇1份、铁皮石斛1份、茯苓1份、银耳1份;或燕麦20份、小米12份、山药5份、白扁豆2份、猴头菇0.5份、铁皮石斛0.5份、茯苓0.5份、银耳3份
本发明进一步提供了一种含有如前任一所述中药组合物的药物、保健品或食品。
具体地,所述的药物是任何一种药学上可接受的剂型。
进一步方案,所述的药学上可接受的剂型为颗粒剂、口服溶液剂、片剂、胶囊剂或丸剂。
本发明还提出了如前任一所述中药组合物在制备用于胃粘膜损伤或胃炎的药物的应用。所述用于,可以是包括治疗或预防在内的一切有益于改善患者相应症状的方式。
进一步地,所述胃粘膜损伤可以是酒精引起的胃粘膜损伤。
本发明提出了如前任一所述中药组合物在制备保护胃黏膜的药品、保健品或食品的应用。
本发明进一步提供了一种如前述任一项所述的中药组合物的制备方法,包括下列步骤:
(1)取药材加入8~12倍水回流提取2次,每次1小时,合并提取液,过滤;
(2)将所述提取液浓缩至55~60℃相对密度1.10~1.15,得浓缩液;
(3)将所述浓缩液进行离心过滤,得离心后的浓缩液;
(4)步骤(3)得到的浓缩液进行喷雾干燥,入口温度170-180℃,出口温度不低于75℃,得干浸膏;
(5)步骤(4)所得干浸膏根据需要进行粉碎、过筛,得到所述中药组合物。
所述的中药组合物的制备方法,制备步骤如下:
(1)取药材加入10倍水回流提取2次,每次1小时,合并提取液,过滤;
(2)将所述提取液浓缩至55~60℃相对密度1.10~1.15,得浓缩液;
(3)将所述浓缩液进行离心过滤,得离心后的浓缩液;
(4)步骤(3)得到的浓缩液进行喷雾干燥,入口温度175℃,出口温度不低于75℃,得干浸膏;
(5)步骤(4)所得干浸膏根据需要进行粉碎、过筛,得到所述中药组合物。
本发明提供的中药组合物中,小米味甘咸,健胃除湿、和胃安眠,与燕麦配伍,既温暖脾胃,又养心安神;山药补肺健脾,固肾益精,既可补脾气之虚,又养肾之阴精,可治诸虚百损、疗五劳七伤、除烦热、补心气不足,与茯苓配伍,脾得健运则气得以生,可有效助后天生化之源;白扁豆和中化湿、补脾止泻,可助健脾化湿;猴头菇性味甘平,健胃补虚,配石斛性味甘咸,益胃生津,一方面补水,一方面促进水液运化,相得益彰;银耳性平、味甘、淡,润肺养胃、滋补生津。各组分安全可靠,无毒副作用,也无任何药品依赖性,长期食用安全有效。诸药合用,共奏健脾和胃、保护胃黏膜之效。
通过实验表明,本发明所提出的组合物能显著减轻模型大鼠胃黏膜充血、水肿、出血症状,还能够显著降低MDA、TNF-α水平,提高SOD酶活力,说明对酒精引起的大鼠胃黏膜损伤以及引起的胃炎均具有明显保护作用。
具体实施方式
如前所述,本发明旨在提供一种中药组合物及其制备方法和应用。以下将结合实验例的内容进行具体描述。
本发明提供了一种用于治疗胃阴不足、脾虚食少、脾胃虚寒的中药组合物,所述中药组合物具有健脾和胃、保护胃黏膜等功效。具体地,本发明中提供的中药组合物中包括以下原料药:燕麦、小米、山药、白扁豆、猴头菇、铁皮石斛、茯苓、银耳。其中,燕麦(Avenasativa L.)是禾本科、燕麦属一年生草本植物。裸粒型燕麦的籽粒不带壳,称为裸燕麦(Avena sativa ssp.nuda),是我国的主要栽培品种;小米(粟),是禾本科狗尾草属草本植物,粟的谷粒可食,为中国北方重要粮食作物之一;谷粒去皮后称为“小米”;山药为薯蓣科植物薯蓣的干燥根茎。白扁豆为豆科植物扁豆Dolichos lablab L.的干燥成熟种子。猴头菇(学名:Hericium erinaceus(Bull.)Pers.)是齿菌科、猴头菇属真菌。铁皮石斛为兰科植物铁皮石斛Dendrobium officinale Kimura et Migo的干燥茎。茯苓,为多孔菌科真菌茯苓Poria cocos(Schw.)Wolf的干燥菌核。银耳是一种担子菌门、银耳纲、银耳目、银耳科、银耳属真菌的子实体。本发明如未注明具体条件者,均按照常规条件或制造商建议的条件进行,所用原料药或辅料,以及所用试剂或仪器未注明生产厂商者,均为可以通过市购获得的常规产品。
本发明第一方面公开了一种中药组合物,其包含下述重量配比的原料:燕麦1-40份、小米1-30份、山药1-30份、白扁豆1-20份、猴头菇0.1-5份、铁皮石斛0.1-5份、茯苓0.1-5份、银耳0.1-5份。优选地,所述中药组合物包括:燕麦15-30份、小米10-20份、山药3-12份、白扁豆2-10份、猴头菇0.5-4份、铁皮石斛0.5-4份、茯苓0.5-4份、银耳0.5-4份。
优选地,所述中药组合物包括:燕麦20-25份、小米12-15份、山药5-9份、白扁豆2-6份、猴头菇0.5-3份、铁皮石斛0.5-3份、茯苓0.5-3份、银耳0.5-3份。
优选地,所述中药组合物包括:燕麦20-22份、小米12-14份、山药5-6份、白扁豆2-4份、猴头菇0.5-1份、铁皮石斛0.5-1份、茯苓0.5-1份、银耳0.5-1份。
具体的,本发明所述的中药组合物包括以下重量份数的组分:燕麦22份、小米14份、山药6份、白扁豆4份、猴头菇1份、铁皮石斛1份、茯苓1份、银耳1份;或燕麦20份、小米12份、山药5份、白扁豆2份、猴头菇0.5份、铁皮石斛0.5份、茯苓0.5份、银耳3份。
进一步方案,所述的中药组合物还包括药学上可接受的辅料,从而使得该中药组合物可以是任何一种药学上可接受的剂型,如颗粒剂、口服溶液剂、片剂、胶囊剂或丸剂。
本发明第二方面提供了一种本发明第一方面中任一项所述的中药组合物的制备方法,对原料依次进行水提、浓缩、离心、喷雾干燥、制粒,从而最大限度地从药物组合物中提取出有效组分,制成颗粒剂、口服溶液剂、片剂、胶囊剂或丸剂后仍具有相同的效果。具体的该中药组合物的制备,包括下列步骤:
(1)取药材加入8~12倍水回流提取2次,每次1小时,合并提取液,过滤;
(2)将所述提取液浓缩至55~60℃相对密度1.10~1.15,得浓缩液;
(3)将所述浓缩液进行离心过滤,得离心后的浓缩液;
(4)步骤(3)得到的浓缩液进行喷雾干燥,入口温度170-180℃,出口温度不低于75℃,得干浸膏;
(5)步骤(4)所得干浸膏根据需要进行粉碎、过筛,得到所述中药组合物。
下面结合具体的实施例对本发明的技术方案进行更加完整的说明。
实施例1中药组合物的制备
该中药组合物包含以下原料:燕麦22份、小米14份、山药6份、白扁豆4份、猴头菇1份、铁皮石斛1份、茯苓1份、银耳1份。
具体制备步骤如下:
(1)取药材加入10倍水回流提取2次,每次1小时,合并提取液,过滤;
(2)将所述提取液浓缩至55~60℃相对密度1.10~1.15,得浓缩液;
(3)将所述浓缩液进行离心过滤,得离心后的浓缩液;
(4)步骤(3)得到的浓缩液进行喷雾干燥,入口温度175℃,出口温度不低于75℃,得干浸膏;
(5)步骤(4)所得干浸膏根据需要进行粉碎、过筛,得到所述中药组合物。
实施例2中药组合物的制备
该中药组合物包含以下原料:燕麦20份、小米12份、山药5份、白扁豆2份、猴头菇0.5份、铁皮石斛0.5份、茯苓0.5份、银耳3份。
具体制备步骤如下:
(1)取药材加入10倍水回流提取2次,每次1小时,合并提取液,过滤;
(2)将所述提取液浓缩至55~60℃相对密度1.10~1.15,得浓缩液;
(3)将所述浓缩液进行离心过滤,得离心后的浓缩液;
(4)步骤(3)得到的浓缩液进行喷雾干燥,入口温度175℃,出口温度不低于75℃,得干浸膏;
(5)步骤(4)所得干浸膏根据需要进行粉碎、过筛,得到所述中药组合物。
实验例3本发明对酒精致急性胃粘膜损伤模型的影响
1、实验材料
1.1动物
SD大鼠,SPF级,雄性,体重180~200g。购买自浙江维通利华实验动物技术有限公司,许可证号:SCXK(浙)2019-0001,合格证编号:20220817Aazz0619000382。
1.2药物及试剂
实施例1中的中药组合物(干浸膏粉),江苏康缘药业股份有限公司;奥美拉唑,山西津华晖星制药有限公司,批号:H20045975,规格:20mg/粒。无水乙醇,生工生物工程(上海)股份有限公司,批号H618WA7002;氯化钠注射液,石家庄四药有限公司,规格:500mL/瓶,批号:2003193205;水合氯醛,沪试国药集团化学试剂有限公司,规格:250g/瓶,批号:20180629;丙二醛(MDA)测定试剂盒,南京建成生物工程研究所,批号:221105;超氧化物歧化酶(SOD)测定试剂盒,南京建成生物工程研究所,批号:221207;肿瘤坏死因子(TNF-α)测定试剂盒,美国invitrogen公司,批号:323787-005。
1.3仪器
超低温冰箱(Haier公司,型号:DW-86L728);电子天平(Sartorius科学仪器公司型号:BSA224S-CW);体重秤(上海香川电子衡器有限公司,型号:ACS-SC-DA);电热恒温鼓风干燥箱(上海精宏实验设备有限公司,型号:DHG-9076A)。
2、实验方法
将60只雄性SD大鼠随机分为6组,分别为正常组、模型组、奥美拉唑组(30mg/kg)、中药组合物低、中、高剂量组(4.17g/kg、8.33g/kg、16.67g生药/kg),每组10只。大鼠适应性饲养5d后,开始正式实验。奥美拉唑组、中药组合物组每天灌胃给予相应药物,正常组、模型组给予生理盐水,连续14d。末次给药前大鼠禁食24h,末次给药1h后,正常组给予1mL生理盐水,模型组和各给药组给予1mL无水乙醇,作用1h,造成大鼠急性胃黏膜损伤。
大鼠灌胃无水乙醇1h后,以10%水合氯醛腹腔麻醉,沿腹中线打开腹腔,取胃,沿胃大弯剪开胃壁,0.9%的氯化钠溶液冲洗胃部食物残渣,于立体显微镜下观察胃粘膜的损伤情况,并用游标卡尺测定出血点或出血带的长度和宽度。因宽度所代表损伤的严重性远较长度大,故双倍积分。其评分标准见表1。另留取大鼠部分胃组织,检测组织中GSH、SOD、TNF-α水平的变化。

3、实验结果
正常组大鼠胃黏膜形态及色泽正常、黏膜光滑,而模型组大鼠胃黏膜出血严重,有条索状出血、糜烂,表明造模成功。与模型组比较,中药组合物低剂量组动物胃黏膜有明显充血水肿,条索状出血,有点片状糜烂,胃黏膜出血及糜烂程度稍轻于模型组;中药组合物中、高剂量组动物胃黏膜可见少量充血水肿,花斑样改变,有散在出血、糜烂,但较模型组有明显改善(P<0.05,P<0.01)。见表2。

乙醇是常见的胃黏膜损伤因素,对胃黏膜有很强的腐蚀性,经胃部吸收后生成乙醛及多种自由基,这些自由基在酒精性胃黏膜损伤过程中有重要作用。MDA是氧自由基导致的脂质过氧化产物,在一定程度上反映了组织受自由基破环的程度;SOD是机体内最重要的自由基清除剂,具有极强的抗氧化活性。此外,炎症是粘膜损伤发病机制中的重要因素,而TNF-α是一种关键的炎症因子,可引起炎症反应发生。通过试剂盒检测发现,与正常组比较,模型组胃粘膜组织中MDA、TNF-α水平显著升高(P<0.01,P<0.01),SOD酶活力显著降低(P<0.01);与模型组比较,中药组合物低、中、高剂量组能显著降低MDA、TNF-α水平(P<0.05,P<0.01),升高SOD酶活力(P<0.05,P<0.01)。见表3。

本发明所提出的组合物能显著减轻模型大鼠胃黏膜充血、水肿、出血症状,还能够显著降低MDA、TNF-α水平,提高SOD酶活力,说明对酒精引起的大鼠胃黏膜损伤具有明显保护作用,其能够抑制胃黏膜炎症反应,提高抗氧化及清除自由基能力。
本发明的上述实施例仅仅是为了能够清楚地说明本发明所作的举例,而并非是对本发明的实施方式的限定。对于所属领域的普通技术人员来说,在上述说明的基础上还可以做出其它不同形式的变化或变动。这里无需也无法对所有的实施方式予以穷举。凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明权利要求的保护范围之内。
Claims (10)
1.一种保护胃黏膜的中药组合物,其特征在于,其由下述重量配比的原料制成:燕麦1-40份、小米1-30份、山药1-30份、白扁豆1-20份、猴头菇0.1-5份、铁皮石斛0.1-5份、茯苓0.1-5份、银耳0.1-5份。
2.根据权利要求1所述的中药组合物,其特征在于,所述中药组合物包括:燕麦15-30份、小米10-20份、山药3-12份、白扁豆2-10份、猴头菇0.5-4份、铁皮石斛0.5-4份、茯苓0.5-4份、银耳0.5-4份。
3.根据权利要求1所述的中药组合物,其特征在于,所述中药组合物包括:燕麦20-25份、小米12-15份、山药5-9份、白扁豆2-6份、猴头菇0.5-3份、铁皮石斛0.5-3份、茯苓0.5-3份、银耳0.5-3份。
4.根据权利要求1所述的中药组合物,其特征在于,所述中药组合物包括:燕麦22份、小米14份、山药6份、白扁豆4份、猴头菇1份、铁皮石斛1份、茯苓1份、银耳1份;或燕麦20份、小米12份、山药5份、白扁豆2份、猴头菇0.5份、铁皮石斛0.5份、茯苓0.5份、银耳3份。
5.含有权利要求1-4任一所述中药组合物的药物、保健品或食品。
6.如权利要求5所述的药物,其特征在于,所述的药物是任何一种药学上可接受的剂型。
7.如权利要求5所述的药物,其特征在于,所述药物包括颗粒剂、口服溶液剂、片剂、胶囊剂或丸剂。
8.权利要求1-5任一所述中药组合物在制备用于胃粘膜损伤或胃炎的药物的应用。
9.权利要求8所述的应用,所述胃粘膜损伤是酒精引起的胃粘膜损伤。
10.根据权利要求1-5所述中药组合物的制备方法,其特征在于,包括以下步骤:取药材加入8~12倍水回流提取2次,每次1小时,合并提取液,过滤,将所述提取液浓缩至55~60℃相对密度1.10~1.15,得浓缩液,将所述浓缩液进行离心过滤,得离心后的浓缩液,将浓缩液进行喷雾干燥,入口温度170-180℃,出口温度不低于75℃,得干浸膏,将所得干浸膏根据需要进行粉碎、过筛。
