本发明涉及植物源农药技术领域,具体地,涉及一种芳姜黄酮植物源农药在防治纹枯病中的应用。
背景技术
水稻纹枯病病原菌(Rhizoctonia solani)是禾本科植物特别是水稻生产中发生普遍而且严重的一种病害,在世界水稻种植区均有发生,是限制水稻高产的主要病害之一。目前生产上一般用75%戊唑·嘧菌酯可湿性粉剂等化学合成农药防治,然而由于药剂长期使用,已导致靶标产生了抗药性,目前急需新的生物农药来控制该病害。
申请公开号为CN108935465A的中国发明专利,公开一种红没药烷型倍半萜类化合物在防治白粉病中的应用,其中公开了姜黄作为中药材在我国的使用历史源远流长,其对环境无污染,对人畜无害,根据姜黄粗提液继续深入研发的植物源农药可用作天然环保绿色的植物源杀菌剂。防治白粉病的化合物提取自姜黄植物,具有对环境更友好、低毒或者无毒、低残留等优点,为研发新型植物源农药提供了基础。但是至今尚未发现将姜黄提取物应用在防治纹枯病中的相关报道。
发明内容
针对现有技术中的缺陷,本发明的目的是提供一种芳姜黄酮植物源农药在防治纹枯病中的应用。

具体实施方式
下面结合具体实施例对本发明进行详细说明。以下实施例将有助于本领域的技术人员进一步理解本发明,但不以任何形式限制本发明。应当指出的是,对本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进。这些都属于本发明的保护范围。
本发明实施例中提供芳姜黄酮植物源农药在防治纹枯病中的应用,芳姜黄酮植物源农药中包含化合物芳姜黄酮。
其中,芳姜黄酮是从姜黄植物中提取得到的,采用的方法包括薄板层析制备法、柱层析法、高效液相色谱制备法中的一种或几种方法,选用溶剂从姜黄粗提液中分离提取。姜黄植物优选姜黄根茎。
从姜黄植物提取芳姜黄酮包括以下步骤:
S1:利用有机或无机溶剂对姜黄植物进行浸泡、提取所得的物质,即姜黄植物粗提液。
S2:对姜黄植物粗提液采用薄板层析制备法、柱层析法、高效液相色谱制备法中任一种或多种进行分离提取,得到芳姜黄酮。
上述采用薄板层析法分离提取是指:选用溶剂,将上述得到的姜黄植物粗提液置于平铺硅胶的薄板上进行层析分离。
上述采用柱层析法分离提取具体是指:选用溶剂,将上述得到姜黄植物粗提液置于装有硅胶作为固定相的玻璃柱中进行分离提取。
上述采用高效液相色谱法分离提取具体是指:选用溶剂,将通过薄板层析和柱层析已初步分离的物质置于色谱柱中,使用高压对其进行分离提取制备。
上述溶剂可以采用石油醚、乙酸乙酯、甲醇、二氯甲烷和水中的一种或多种混合溶剂。
上述混合溶剂是由石油醚与乙酸乙酯按体积比为1:0-0:1比例混合得到;或由乙酸乙酯与甲醇按体积比1:0-0:1的比例混合得到;或由甲醇与水按体积比为1:0-0:1的比例混合得到;或由二氯甲烷与水按体积比为2:1的比例混合得到。
芳姜黄酮的化学结构式如图1所示。
姜黄根茎提取物中包括芳姜黄酮,采用姜黄根茎提取物作为芳姜黄酮植物源农药;关于姜黄根茎提取物的制备方法和芳姜黄酮的鉴定,具体可参考WJ Fu,J Liu,MZhang,JQ Li,JF Hu,LR Xu,GH Dai(2018).Isolation,purification andidentification ofthe active compound ofturmeric and its potential applicationto control cucumber powdery mildew.The Journal ofAgricultural Science 156,358-366。
芳姜黄酮植物源农药被配制成芳姜黄酮乳油(即姜黄根茎提取物乳油制剂),具体地,芳姜黄酮乳油的组分以质量百分含量计包括30-70%姜黄根茎提取物、10-35%乳化剂、15-50%溶剂。通过将特定质量配比的姜黄根茎提取物、乳化剂、溶剂在内的组分进行复配制备一种以姜黄根茎提取物为功效成分的用于防治纹枯病的新型乳油制剂,其制备工艺简单、使用便捷,具有优异的高温、低温稳定性,能使姜黄根茎提取物更稳定持久地发挥其纹枯病防治功效。相比于化学合成农药,姜黄根茎提取物来源于自然植物,不会在环境中富集破坏环境,而是会较快地在环境中降解;低毒;同时,相较于成分单一的化学合成农药,本发明实施例制得的姜黄根茎提取物成分复杂,起到防治纹枯病作用的是多种功效成分的相互配合共同起效的结果,其他一些非功效成分也会起到辅助作用,因此靶标不容易产生抗药性。
上述实施例中,姜黄根茎提取物的质量百分含量可以为30%、35%、40%、45%、50%、55%、60%、65%、70%等;乳化剂的质量百分含量可以为10%、15%、20%、25%、30%、35%等;溶剂的质量百分含量可以为15%、20%、25%、30%、35%、40%、45%、50%等。上述各项数值范围内的具体点值均可选择,在此便不再一一赘述。
在一些优选的实施方式中,芳姜黄酮乳油的组分以质量百分含量计包括40-60%姜黄根茎提取物、20-30%乳化剂、15-35%溶剂。当姜黄根茎提取物、乳化剂、溶剂三种组分以上述更加优选的质量配比进行组合时,产品具有更加优异的高温、低温、常温稳定性,能使姜黄根茎提取物更稳定持久地发挥其纹枯病防治功效。
在一些优选的实施方式中,溶剂包括油酸甲酯、大豆油、玉米油、菜籽油或松基油中的任意一种或至少两种的组合。
本发明实施例采用上述生物降解性好、污染小的绿色环保溶剂,减轻了对环境的污染,对该乳油制剂产品的应用更有利。
在一些实施方式中,乳化剂包括非离子型乳化剂和阴离子型乳化剂。优选地,非离子型乳化剂包括三苯乙烯酚聚氧乙烯醚、山梨醇酐三油酸酯聚氧乙烯醚、三苯乙烯基酚-乙氧基-丙氧基化物或蓖麻油聚二醇醚酯中的任意一种或至少两种的组合。阴离子型乳化剂选自二苯基磺酸盐、α-烯烃磺酸盐、木质素磺酸盐、乙氧基化烷基酚的磺酸盐、烷氧基化芳基酚的磺酸盐、缩合萘的磺酸盐、十二烷基苯或十三烷基苯的磺酸盐、萘或烷基萘的磺酸盐或磺基琥珀酸盐中的任意一种或至少两种的组合。
本发明实施例优化了乳化剂的配方,使其更好地与溶剂和姜黄根茎提取物进行配合,从而使体系更加稳定,保障了姜黄根茎提取物药效持久稳定的发挥。
在一些优选的实施方式中,乳化剂以质量百分含量计包括30-40%三苯乙烯酚聚氧乙烯醚、30-40%山梨醇酐三油酸酯聚氧乙烯醚和25-35%十二烷基苯磺酸钙。三苯乙烯酚聚氧乙烯醚的质量百分含量可以选择30%、32%、33%、35%、36%、38%、40%等;山梨醇酐三油酸酯聚氧乙烯醚的质量百分含量可以选择30%、32%、33%、35%、36%、38%、40%等;十二烷基苯磺酸钙的质量百分含量可以选择25%、27%、28%、30%、32%、35%、40%等。上述各项数值范围内的具体点值均可选择,在此便不再一一赘述。
本发明实施例通过对溶剂和乳化剂类型的优化选择,使得该乳油制剂低毒、安全、环保、稳定性好、乳化性能好,持效性优秀。
在一些优选的实施方式中,芳姜黄酮乳油的质量浓度(质量百分比)为4.9-8.1%,能够起到良好的纹枯病防治效果。
以下结合本发明上述的芳姜黄酮植物源农药,采用实验来验证其在防治纹枯病中的应用。
应用例1
按照NY/T 1156.2-2006农药室内生物测定试验准则杀菌剂第2部分:抑制病原真菌菌丝生长试验平皿法,进行芳姜黄酮对水稻纹枯病菌抑菌效果试验,具体如下:
1、试验条件
1.1供试靶标
水稻纹枯病:立枯丝核菌(R.solani Kuhn),由市售途径获取。
1.2培养条件
培养基为PDA培养基,培养温度:25±1℃。
1.3供试药剂:

2.1试验准则
NY/T 1156.2-2006农药室内生物测定试验准则杀菌剂第2部分:抑制病原真菌菌丝生长试验平皿法。
2.2浓度设计
供试药剂芳姜黄酮浓度(mg/L),即上述芳姜黄酮乳油的浓度,设计12、6、3、1.5、0.75共5个浓度。对照药剂250克/升吡唑醚菌酯乳油有效浓度(mg/L)为4、3、1.5、0.75、0.375共5个浓度,设无菌水空白对照。共11个处理,每处理4次重复。
3、试验方法
3.1试验处理方法
将熔好的培养基冷却至45℃~55℃,按所设浓度加入定量药剂,制成含有不同药量的含毒培养基,待其充分冷却后接种直径0.5cm的供试病原菌菌片,另设空白对照,放置培养箱中培养(25℃±1℃)。
3.2结果调查及药效计算方法
在培养箱中培养48小时调查。调查时分别测量每个处理的供试病原菌菌落生长直径。根据菌落直径的测量大小,计算各处理菌丝生长抑制率。菌丝生长抑制率计算方法如下:
菌丝生长抑制率(%)=(1-药剂处理的菌落直径-0.5空白对照菌落直径-0.5)×100。
3.3数据处理方法
EC50值的计算:使用DPS V16.05软件计算药剂的毒力回归方程、有效抑制中浓度EC50值以及相关系数(r),并对结果进行分析评价。
4、结果与分析
4.1试验结果
试验结果表明:6.5%±1.6%(即4.9-8.1%)芳姜黄酮乳油对水稻纹枯病菌具有较好的抑菌活性,在药剂浓度为12、6、3、1.5、0.75mg/L时,对水稻纹枯病菌的抑菌效果分别为71.13%、65.46%、61.34%、42.27%、27.32%,其EC50值为2.4274mg/L,毒力回归方程y=4.6272+0.9680x,略低于对照药剂250克/升吡唑醚菌酯乳油对水稻纹枯病菌的抑菌效果(EC50值为1.2488mg/L),毒力回归方程y=4.8451+1.6055x。
需要说明,若芳姜黄酮乳油的质量浓度低于4.9-8.1%,可能无法起到有效防治水稻纹枯病菌的效果;若高于该浓度,则可能会产生药害问题,同时带来成本增加的问题。
通过上述芳姜黄酮乳油对水稻纹枯病菌室内生物活性测定结果,明确该药剂对该病原菌的抑菌活性,为产品开发和在植物病害防治中的推广应用提供依据。
应用例2
根据芳姜黄酮对水稻纹枯病菌抑菌效果试验,进行芳姜黄酮防治水稻纹枯病田间药效试验,通过使用芳姜黄酮防治水稻纹枯病,观察防治效果,以实现减少稻田化学农药使用量。具体如下:
1、试验条件
1.1试验对象和品种的选择
试验对象:水稻
品种:松香粳1018
1.2环境或栽培条件
试验地点在上海市松江区泖港镇胡光村。
2、试验设计和安排
2.1药剂
2.1.1试验药剂
供试药剂:6.5%芳姜黄酮乳油
对照药剂:75%戊唑·嘧菌酯可湿性粉剂
2.1.2药剂用量与编号
以A表示芳姜黄酮乳油150ml/亩,B表示芳姜黄酮乳油250ml/亩,C表示芳姜黄酮乳油350ml/亩;CK表示空白对照;常规表示75%戊唑·嘧菌酯可湿性粉剂15g/亩。
2.2小区面积和重复
本试验为小区试验,3次重复,共15个试验处理,每个小区面积为60m2。小区试验的行列排布以及药剂用量见表2。

浙江欧森机械有限公司OS-30机动喷雾器喷雾,工作压力0.1-0.4MPa。

7月28日和8月5日进行纹枯病防治。
2.3.4防治其他病虫害的药剂资料
试验阶段杀虫剂参照面上常规防治,不使用其他杀菌剂。

(1)纹枯病调查方法
根据水稻叶鞘和叶片为害症状程度分级,以株为单位,定点调查,每小区对角线五点取样,每点调查相连5丛,共25丛,记录总株数、病株数和病级数。
分级方法(以叶片为单位)
0级:无病斑;
1级:第四叶片及其以下各鞘、叶片发病(以顶叶为第一片叶);
3级:第三叶片及其以下各鞘、叶片发病;
5级:第二叶片及其以下各鞘、叶片发病;
7级:剑叶叶片及其以下各鞘、叶片发病;
9级:全株发病,提早枯死。
3.2.3药效计算方法
(1)纹枯病防效计算方法
发病率(%)=发病株数/调查总株数×100;
病情指数=[∑(各级病株数×各级代表值)/调查总株数×最高级代表值]×100;
病株防效(%)=(CK-PT)/CK×100;
病指防效(%)={1-(CK0×PT1)/(CK1×PT0)}×100;
上述公式中,CK0-空白对照区施药前病情指数,CK1-空白对照区施药后病情指数,PT0-药剂处理区施药前病情指数,PT1-药剂处理区施药后病情指数,CK-空白对照区发病率,PT-药剂处理区发病率。
3.3对作物及其他生物的直接影响

4、结果与分析
4.1试验结果
(1)8月5日(纹枯病第1次用药后防效调查):6.5%芳姜黄酮乳油亩用150ml、250ml、350ml以及常规药剂防治对水稻纹枯病的病株防效分为15.60%、54.27%、61.29%、51.52%;指数防效分别为43.31%、53.46%、60.60%、50.66%。
(2)8月12日(纹枯病第2次用药后防效调查):6.5%芳姜黄酮乳油亩用150ml、250ml、350ml以及常规药剂防治对水稻纹枯病的病株防效分为29.15%、58.99%、69.66%、63.61%;指数防效分别为38.32%、60.30%、73.58%、68.32%。

4.2试验评价
由表4看出,6.5%芳姜黄酮乳油亩用250ml、350ml对水稻纹枯病具有良好防治效果,中、高剂量的6.5%芳姜黄酮乳油防治水稻纹枯病效果接近或高于面上常规药剂防治。此外需要说明,2022年因受持续极端高温和少雨天气影响,不利于水稻纹枯病发病,水稻纹枯病整体发病率低,发病程度轻,以低级别发病为主,因此本次试验针对水稻纹枯病的防效效果偏低。在其他一些应用例如中高级别的水稻纹枯病发病情况,中、高剂量的芳姜黄酮乳油对纹枯病的防效具有更佳的防治效果。
上述实施例中,植物源农药姜黄根茎提取物(主要成分是芳姜黄酮)通过室内离体生测和田间药效试验,结果表明药剂中高浓度对该病害控制效果类似或优于化学对照药剂,植物源农药作为一种植物提取物混合物,能够减少农田化学农药使用量,缓解靶标抗药性问题,环境中易降解,对陆生环境生物低毒,具有一定的经济效益和生态效益。
应用例3
按照NY/T 1156.2-2006农药室内生物测定试验准则杀菌剂第2部分:抑制病原真菌菌丝生长试验平皿法,进行芳姜黄酮对小麦纹枯病菌抑菌效果试验,具体如下:

小麦纹枯病:丝核菌Rhizoctonia cereadis Vander Hoeven水。
1.2培养条件
培养基为PDA培养基,培养温度:25±1℃。
1.3供试药剂:6.5%芳姜黄酮乳油、250克/升吡唑醚菌酯乳油。
1.4仪器设备:恒温培养箱,无菌室,电子天平,超净工作台,移液枪等。
2、试验处理
试验靶标及处理浓度见表5,另设无菌水空白对照。

3.1试验处理方法

4、结果与分析
4.1试验结果
试验结果表明:6.5%芳姜黄酮乳油对小麦纹枯病菌具有较好的抑菌活性,在药剂浓度为12、6、3、1.5、0.75mg/L时,对小麦纹枯病菌的防效分别为69.31%、63.37%、58.42%、37.62%、22.77%,其EC50值为3.0040mg/L,略低于对照药剂250克/升吡唑醚菌酯乳油对小麦纹枯病菌的抑菌效果(EC50值为1.7471mg/L)。详细结果见表6。

4.2分析及评价
试验结果表明:6.5%芳姜黄酮乳油对供试小麦纹枯病菌具有较好的抑菌活性,其EC50值为3.0040mg/L,略低于对照药剂250克/升吡唑醚菌酯乳油对小麦纹枯病菌的抑菌效果(EC50值为1.7471mg/L)。按照上述方法进行6.5%±1.6%(即4.9-8.1%)芳姜黄酮乳油对小麦纹枯病菌的抑菌效果试验,均可取得良好的抑菌效果。植物源农药作为生物农药,可以起到略低于或者基本相当于化学合成农药的小麦纹枯病菌防治效果,考虑植物源农药本身具有的生态效益以及对靶标抗药性的缓解等方面的优点,上述芳姜黄酮植物源农药在防治小麦纹枯病菌方面具有更好的优势。
本发明实施例中的芳姜黄酮植物源农药还可以用于防治谷子纹枯病,其实施方法与上述水稻纹枯病实施例的实施方法相同,对谷子纹枯病的控制效果类似或优于化学对照药剂,此处不再详述。
以上对本发明的具体实施例进行了描述。需要理解的是,本发明并不局限于上述特定实施方式,本领域技术人员可以在权利要求的范围内做出各种变形或修改,这并不影响本发明的实质内容。上述各优选特征在互不冲突的情况下,可以任意组合使用。

2.根据权利要求1所述的应用,其特征在于,所述芳姜黄酮植物源农药被配制成芳姜黄酮乳油,所述芳姜黄酮乳油的质量浓度为4.9-8.1%。
3.根据权利要求2所述的应用,其特征在于,所述芳姜黄酮乳油施于患纹枯病植株的叶面。
4.根据权利要求2所述的应用,其特征在于,所述芳姜黄酮乳油的用量为250-350ml/亩。
5.根据权利要求1所述的应用,其特征在于,所述纹枯病由立枯丝核菌引起。
6.根据权利要求1所述的应用,其特征在于,所述纹枯病为水稻纹枯病、小麦纹枯病和谷子纹枯病中的任意一种。
