Abstract
A traditional Chinese medicine composition with sleep improving effect and a preparation method and application thereof relate to the technical field of traditional Chinese medicines. The traditional Chinese medicine composition disclosed by the application comprises jujube kernel, poria cocos, lily, huai wheat, jujube, mulberry concentrated juice and strawberry concentrated juice. The preparation method comprises the following steps: weighing semen Ziziphi Spinosae, crushing, decocting with Poria, bulbus Lilii, fructus Tritici Levis and fructus Jujubae in water for multiple times, concentrating the filtrate under reduced pressure, mixing with Mori fructus concentrated juice and strawberry concentrated juice, and filtering with ceramic membrane to obtain Chinese medicinal composition. The application combines the sub-health characteristics of the crowd in China and the characteristics of traditional Chinese medicines, takes the "inheritance-development-innovation" of the traditional Chinese medicine as the thought, carries out scientific and reasonable compatibility of the components, and examines the preparation process and the drug effect of the components, the obtained traditional Chinese medicine composition has the effects of reinforcing the purgation, reinforcing the benefit, clearing and reinforcing the benefit, can harmonize the stomach with the heat of the liver and yin, and purging the heart fire, and fundamentally improves the insomnia state of the organism, and the preparation process of the application is easy to industrialize.

Background

At present, the main means for improving sleep include western medicine intervention, traditional Chinese medicine intervention, health care products and the like, wherein the western medicine for improving sleep is mainly sedative hypnotic drugs including barbiturates, benzodiazepines, non-benzodiazepines and the like, but the drugs have side effects of common insomnia rebound, addiction, tolerance and the like. The health care product mainly contains melatonin, which can prolong the total sleeping time, but can not improve the sleeping quality. Therefore, combining the sub-health characteristics of the population in China and the characteristics of traditional Chinese medicines, developing the sub-health state of insomnia improvement by taking traditional Chinese medicines as main means is imperative.
Disclosure of Invention
In order to more effectively and safely improve the insomnia state of the organism from the root, the application provides a traditional Chinese medicine composition with the function of improving sleep, and a preparation method and application thereof.

s1, weighing spina date seeds, and crushing to obtain a crushed spina date seed A;
s2, weighing poria cocos, lily, common yam rhizome, wheat and jujube, soaking the poria cocos, the lily, the common yam rhizome and the jujube and the crushed spina date seed A in an extraction solvent for a certain time, heating and decocting, and concentrating filtrate under reduced pressure to obtain a mixed extract B;
s3, mixing the mulberry concentrated juice, the strawberry concentrated juice and the mixed extracting solution B to obtain a mixed extracting solution C;
s4, filtering the mixed extract C through a ceramic membrane to obtain the traditional Chinese medicine composition.

Chou, and has effects in nourishing heart, nourishing liver, tranquilizing mind, relieving restlessness, insomnia, palpitation, dreaminess, asthenia, hyperhidrosis, and thirst due to body fluid injury. Is mainly used for treating heart deficiency, mental confusion, terror and convulsion, frequent absentmindedness, easy forgetfulness, restlessness, dreaminess, and heart diseases. Modern pharmacological researches show that the semen zizyphi spinosae has the effects of tranquilizing, hypnotizing, improving learning and memory, protecting brain, enhancing immunity and the like, and can play a role in improving sleep.
Poria is dry sclerotium of Polyporaceae fungus Poria penia cocos (Schw.) Wolf, has sweet, light and flat taste, has effects of promoting water penetration, removing dampness, invigorating spleen, tranquilizing, and calming heart, and can be used for treating phlegm retention palpitation, spleen deficiency, anorexia, heart mind restlessness, palpitation, and insomnia. "Shennong Ben Cao Jing" is: the long-term taking of the medicine can calm the spirit, nourish the spirit, and allay hunger and prolong the life. The leaf angel stands for: spleen is the acquired root, spleen is healthy and allaying hunger, and qi and feet are prolonged. Modern pharmacological researches show that Poria has effects of tranquilizing, regulating immunity, protecting liver, and protecting central nerve, immune system, and digestive system, so as to improve sleep.
The lily is dried fleshy scaly leaf of lily plant Paeonia ostii Lilium lancifolium thunder, lily Lilium brownii F.E.Brownvar. Virdulum Baker or Lilium pumilum DC. The Japanese herbal medicine is: can be used for relieving heart, fixing gallbladder, nourishing mind, and nourishing viscera. Modern pharmacological researches have shown that lily has the effects of tranquilizing, hypnotizing, anti-depression, anti-stress injury, relieving cough, eliminating phlegm, regulating immunity, resisting oxidation, reducing blood sugar and the like.
Huai wheat is sweet in taste, slightly cold in nature, and can nourish heart, relieve restlessness, promote urination, stop bleeding, quench thirst and recover sweat, and can tonify deficiency, nourish qi, aid five viscera and strengthen intestines and stomach. "Su Wen" cloud: the cereal of the fire and heart of wheat. It is combined with Zao to exert the actions of nourishing heart, tranquilizing mind, and regulating middle energizer.
Fructus Jujubae is a dried mature fruit of Zizyphus jujuba Mill. Has effects of invigorating spleen, replenishing qi, nourishing blood, and tranquilizing, and can be used for treating spleen deficiency diarrhea, insomnia, night sweat, palpitation, etc. "Benjing" is: pathogenic qi in the heart and abdomen, spleen and stomach are nourished, twelve meridians are helped, stomach qi is calmed, nine orifices are led, less qi is supplemented, body fluids are reduced, the body is deficient, the person is frightened, the limbs are heavy, and the medicine is filled. Modern pharmacological researches have shown that jujube has the effects of enhancing immunity, resisting oxidation, improving cardiovascular system, repairing liver injury, resisting fatigue, improving intestinal tract and the like.
The Mori fructus is dried fruit ear of Morus alba L. Has effects of replenishing blood, nourishing yin, promoting salivation, and moistening dryness, and can be used for treating palpitation, insomnia, dizziness, tinnitus, premature gray hair, and thirst due to fluid consumption. "Bencaojing Shu" is: mulberry, mori fructus, sweet in flavor and cold in flavor, benefits blood and yin, and is a herb for cooling blood and tonifying yin, and its five zang organs belong to yin and benefit yin to benefit five zang organs. When yin is removed, liver and heart are not fire, so spirit is calm and clear, spirit is smart and internal, and yin is complex and white. Modern pharmacological researches have shown that mulberry has the effects of regulating immunity, resisting oxidation, reducing blood sugar, reducing blood lipid and the like.
Compared with the prior art, the application has the following specific beneficial effects:

Lily can nourish yin, moisten lung, clear heart and tranquilize mind, and can "calm heart, calm gallbladder, benefit mind and nourish five viscera". Huai wheat Gan Liang, which has the effects of nourishing heart, relieving urgency, relieving restlessness and soothing nerves; jujube sweet Wen Zhirun, has effects of invigorating qi, regulating middle warmer, moistening dryness and relieving urgency; the two herbs are combined to achieve the effects of nourishing heart, soothing the nerves and regulating middle energizer and relieving urgency, so that they are in good agreement with the theory of Su Wen and zang-qi methods, called "liver-bitter and urgent, and rapid and sweet and slow food". Mulberry can enrich blood, nourish yin, promote the production of body fluid and moisten dryness, and can nourish liver and kidney, remove heat and nourish yin. The medicines are combined together to play the roles of nourishing heart, soothing nerves, clearing heat and relieving restlessness. Experiments and comparison prove that the application can prolong the sleep time and can also realize the remarkable shortening of the sleep latency time, thereby improving the sleep quality.
2. The traditional Chinese medicine composition provided by the application has the characteristics of scientific formula, green herb, natural health and zero addition, has a simple preparation process, is easy to operate, has low cost, is easy to industrialize, and can provide a high-level product with guaranteed quality and safety for insomnia people.

filtering the mixed extract B with 200nm ceramic membrane to obtain the Chinese medicinal composition.
Example 2.
Weighing 7.5 parts of spina date seeds, slightly crushing, and breaking skin to obtain a crushed spina date seed A;
weighing 7.5 parts of poria cocos, 5 parts of lily, 20 parts of common wheat, 12 parts of Chinese date, soaking for 60min together with the crushed wild jujube seed A, heating and decocting the mixture for 2 times by taking 8 times of water as an extraction solvent, filtering each time by a 200-mesh filter screen, and concentrating the filtrate under reduced pressure (60-80 ℃) to 500mL to obtain a mixed extract B;
weighing 30 parts of mulberry concentrated juice and 25 parts of strawberry concentrated juice, adding the mulberry concentrated juice and the strawberry concentrated juice into the mixed extract B, and uniformly mixing to obtain a mixed solution C;
filtering the mixed solution C with 200nm ceramic membrane to obtain the Chinese medicinal composition.
Example 3.
Weighing 7.5 parts of spina date seeds, slightly crushing, and breaking skin to obtain a crushed spina date seed A;
weighing 7.5 parts of poria cocos, 5 parts of lily, 20 parts of common wheat, 12 parts of Chinese date, soaking for 60min together with the crushed wild jujube seed A, heating and decocting the mixture for 3 times by taking 10 times of water as an extraction solvent, filtering each time by a 200-mesh filter screen, and concentrating the filtrate under reduced pressure (60-80 ℃) to 500mL to obtain a mixed extract B;
weighing 30 parts of mulberry concentrated juice and 25 parts of strawberry concentrated juice, adding the mulberry concentrated juice and the strawberry concentrated juice into the mixed extract B, and uniformly mixing to obtain a mixed solution C;
filtering the mixed solution C with 200nm ceramic membrane to obtain the Chinese medicinal composition.
Effect example 1.
40 ICR mice of grade 18 g-25 g SPF were adaptively kept in a natural circadian rhythm illumination environment at a temperature of 24+ -2 deg.C, a relative humidity of 60+ -5%, and a natural circadian rhythm illumination environment for 1 week, and then were divided into a blank group, an example 1 group, an example 2 group and an example 3 group, each group of 10 mice was respectively given drinking water (8:00 to 15:00 per day) and a corresponding Chinese medicinal composition liquid (15:00 per day to 8:00 per day) for 30 days in a test period; on day 31, each group of mice was fed with 25mL/kg of the corresponding concentrate of the Chinese medicinal composition (concentrated to 200 mL).
And (3) carrying out a barbital sodium sleep latency experiment, observing whether each traditional Chinese medicine composition can shorten the sleep latency on the basis of the barbital sodium sleep, and if the sleep latency is shortened, indicating that the traditional Chinese medicine composition has a synergistic effect with the barbital sodium. Pre-experiments are carried out before formal experiments, and the dosage of barbital sodium (200 mg/kg-300 mg/kg) which enables the animal to fall asleep 100% but does not enable the animal to fall asleep for too long is determined, and the dosage is used for formal experiments.
After the last administration for 15min, the animals of each group were intraperitoneally injected with 0.2ml/20g barbital sodium, and the effect of each Chinese medicinal composition on the sleep latency of barbital sodium was observed with the disappearance of everlasting reflection as an index.
The results of the experiments are shown in Table 1 and FIG. 2, and the sleep latency of the mice in the blank group was 7.57min. The mice of each example had a reduced sleep latency of 3.71min (P < 0.05), 2.74min (P < 0.05) and 3.94min (P < 0.01), respectively, compared to the blank, and exhibited significant differences (P < 0.05). The results show that the traditional Chinese medicine composition in each embodiment has good effect of reducing sleep latency.

Effect example 2.
And (3) carrying out an experiment of prolonging the sleeping time of the pentobarbital sodium, observing whether each traditional Chinese medicine composition can prolong the sleeping time on the basis of hypnotizing the pentobarbital sodium, and if the sleeping time is prolonged, indicating that the traditional Chinese medicine composition has a synergistic effect with the pentobarbital sodium. Preliminary experiments are carried out before formal experiments are carried out, and the dosage (30-60 mg/kg) of pentobarbital sodium which enables the animal to fall asleep 100% but does not enable the animal to fall asleep for too long is determined, and the dosage is used for formal experiments.
After the last administration for 15min, the animals of each group are injected with pentobarbital sodium in the abdominal cavity with the injection amount of 0.2ml/20g, and whether the sleep time of the pentobarbital sodium can be prolonged or not by observing whether the normal reflection disappears or not by taking the normal reflection disappearance as an index.
The experimental results are shown in Table 2 and FIG. 3, and the sleep time of the mice in the blank group is generally 30.5min. The mice of each example had an increased sleep time compared to the blank, respectively increased by 9.33min (P < 0.05), 8.50min (P < 0.05) and 11.33min (P < 0.05), with a significant difference (P < 0.05). The results show that the traditional Chinese medicine composition in each embodiment has good effect of prolonging the sleeping time of mice.

In comprehensive comparison, each group of traditional Chinese medicine composition has the effect of improving sleep, and the group of the embodiment 3 > the group of the embodiment 1 > the group of the embodiment 2 shows that the formula and the process of the application are optimal.
In the foregoing, the present application is merely preferred embodiments, which are based on different implementations of the overall concept of the application, and the protection scope of the application is not limited thereto, and any changes or substitutions easily come within the technical scope of the present application as those skilled in the art should not fall within the protection scope of the present application. s1, weighing spina date seeds, and crushing to obtain a crushed spina date seed A;
s2, weighing poria cocos, lily, common yam rhizome, wheat and jujube, soaking the poria cocos, the lily, the common yam rhizome and the jujube and the crushed spina date seed A in an extraction solvent for a certain time, heating and decocting, and concentrating filtrate under reduced pressure to obtain a mixed extract B;
