须药藤是一种木质攀缘藤本植物。它分布在中国的西南省份,即云南,贵州,广西和四川。须药藤的叶和茎在传统上用于治疗感冒,胃痛,气管炎和风湿性疼痛的中药中。
所有或部分须药藤植物可用于本方法。在一个实施方案中,在提取之前部分干燥须药藤植物材料。在另一个实施方案中,在提取之前完全干燥须药藤植物材料。在一些实施方案中,任选地在提取之前研磨该植物材料。

实施例
以下实施例仅出于示例的目的给出,并且不应被解释为对本发明的限制,因为在不脱离本公开的精神和范围的情况下,本发明的许多变化是可能的。
实施例1:须药藤提取物和级分的制备
将须药藤粉末(200g)加载到66mL不锈钢池(2或3个池,取决于材料密度)上。DionexTM ASETM 350加速溶剂提取器(ASE)的条件设置为:压力1500psi;温度100℃;静置时间15min;循环2;冲洗70%;吹扫60s。依次用100%和80%甲醇提取每个池。合并所有提取物,并在旋转蒸发器上蒸发有机溶剂,得到42g萃取物。将旋转蒸发器后的提取物冷冻干燥。
HP-20用于对植物提取物进行分级。在Biotage快速色谱系统上研发了自动HP-20分级方法。简言之,使100g HP-20吸收性树脂上25mm×500mm柱并用甲醇洗涤。HP-20柱用水平衡。将10g植物提取物悬浮于水中并超声处理20min。然后使该悬浮液上柱。使用以下流动相:0%甲醇水溶液350mL,30%甲醇450mL,50%甲醇450mL,70%和100%MeOH550mL。将流速设定为4mL/min。在254nm下监测UV信号以确保分级一致性。每个级分为225mL(240mL收集瓶)。将每两个级分合并,得到最终的五个级分(0,30%,50%,70%,100%MeOH级分)。分级后,将0,30%和50%MeOH级分送至TurboVap以除去溶剂。使用旋转蒸发器蒸发70%和100%MeOH级分。将全部五个级分最终在冷冻干燥器上冻干。
实施例2:来自须药藤的化合物的表征
在Waters ACQUITY H-Class UPLC-Synapt G2-Si系统(Waters,USA)上进行化学分析。在25℃下使用UPLC C18分析柱(2.1mm×150mm,I.D.,1.8μm,ACQUITYHSST3,Waters,USA)。水(A)和乙腈(B)(均包含0.1%甲酸)用作流动相,线性梯度编程如下:0-30min,5%B;30-45min,90%B;45-46min,B%;46-50min,5%B。流速设定为0.8mL.min-1,且注射体积为2μL。质谱仪配备有电喷雾电离(ESI)源,并且通过从m/z 100至1500扫描以正模式获得数据。MS分析如下操作:去溶剂化气体流速,800L/hr;去溶剂化温度300℃;雾化器气流,6.5Bar;毛细管电压,3000V;源温度,80℃;采样锥形电压,40V。扫描时间设定为0.2s。通过在质谱仪中以10μL/min的流速输注400ng/mL亮氨酸-脑啡肽溶液,通过lockspray界面进行内部校准。将lockspray扫描时间和频率分别设定为0.5s和15s。该系统由Masslynx 4.1(Waters)控制。在没有前体选择的情况下,在MSE模式中在m/z范围50-1500内使用在低(10eV)和高(20-120eV斜坡)碰撞能量下0.2s的交替扫描,获得数据。
