背景技术

传统预防和治疗嗜神经病毒成分包括疫苗和抗病毒小分子药物,它们以促进宿主抗病毒天然免疫和适应性免疫以清除病毒粒子。根据以往的研究,嗜神经病毒的疫苗已得到广泛开发,包括DNA疫苗、纯化灭活病毒疫苗、mRNA疫苗和病毒载体疫苗。疫苗对大多数病毒性疾病都有很好的治疗效果,因为疫苗中的病毒免疫原蛋白可以刺激宿主产生中和抗体来消灭病毒。抗病毒化合物,例如利巴韦林、瑞德西韦和法匹拉韦,是广谱抗病毒药物,可通过作用于RNA聚合酶有效抑制病毒复制。然而,常规疫苗接种以及抗病毒药物治疗在预防和治疗嗜神经病毒感染中,由于体内递送效率的限制仍然存在一些局限性和挑战。对于传统方法的体内应用来说,病毒疫苗的抗原稳定性、药物的组织递送效率以及病毒靶向性递送效率都很大程度上限制了药物效果。
此外,除了传统预防和治疗嗜神经病毒感染的手段,目前比较常见的处理方式还有脑靶向性治疗,研究表明,狂犬病病毒糖蛋白膜外区具有一段29个氨基酸的多肽(Rabiesvirus glycopeptide,RVG,其序列为N端-YTIWMPENPRPGTPCDIFTNSRGKRASNG-C端)能够识别神经元细胞表面的乙酰胆碱受体,经过该多肽修饰的纳米颗粒在脑肿瘤的治疗中能够从外周血顺利进入中枢神经系统并靶向肿瘤细胞。鉴于RVG的此种特性,目前有技术构建了介孔二氧化硅、RVG、特异性适配体(靶向病毒)以及纳米金棒颗粒的复合纳米颗粒体系,该复合纳米颗粒体系如图1所示:该纳米颗粒由一层介孔二氧化硅包裹内层纳米金棒组成,其介孔硅表层修饰特异性适配体(靶向病毒),RVG多肽(提高脑内递送效率)。并进行了小鼠模型中的靶向光热治疗研究,如图2所示,通过在小鼠尾静脉注射纳米颗粒,待递送至脑部后,通过近红外激光激发纳米颗粒的光热效应,产生热能,灭活病毒,在该研究中成功通过RVG的修饰提高了纳米颗粒的脑部递送效率,实现了靶向治疗。但是该技术也存在明显的缺点:
第一:其光热效应虽然能一定程度灭活病毒,但产生的热量会损伤大脑组织,包括神经元、胶质细胞等。脑内细胞经历此类损伤后,其造成的影响不可逆。
第二:纳米金棒其内部由金元素组成,属于难以代谢消化的成分,经历不断累积之后会损伤体内肝肾功能,同时损伤脑组织。
第三:纳米金棒的光热效应无法引起长时间多次的热能转化,也就无法长时间持续灭活脑内病毒。若微量病毒未被灭活,其经历复制后病毒载量迅速增加同样会引起疾病。
发明内容
针对现有技术中的上述问题,本发明提供一种T-705@MSN-RVG脑内药物递送载体及其制备方法,以解决现有技术损伤大脑组织、金元素累积损伤身体器官以及容易复发的技术难题。
本发明采用的技术方案如下:
一种T-705@MSN-RVG脑内药物递送载体,包括羧基介孔硅及包覆在羧基介孔硅内的法匹拉韦,所述羧基介孔硅的表面修饰有RVG多肽。
T-705@MSN脑内药物递送载体的制备方法,包括如下步骤:
(1)将法匹拉韦溶于乙醇中形成法匹拉韦溶液;
(2)将羧基介孔硅分散于法匹拉韦溶液中,并在35~40℃震荡过夜,然后离心去除上清,并采用pH5.5的MES缓冲液清洗溶剂,得到T-705@MSN;
(3)将T-705@MSN和RVG多肽溶液混合,加入现配的EDC溶液,室温反应过夜,离心去除上清,并洗涤所得沉淀,即得到T-705@MSN-RVG脑内药物递送载体。
更优地,步骤(1)中,法匹拉韦溶解过程中还加有DMSO用于助溶。
更优地,步骤(2)中,采用的MES缓冲液的浓度为0.02mM。
更优地,步骤(3)中,所述RVG多肽溶液的浓度为5mg/mL,溶剂为pH5.5的MES缓冲液。
更优地,步骤(3)中,所述EDC溶液的浓度为10mg/mL。
综上所述,相比于现有技术,本发明具有如下优点及有益效果:
1、本发明通过介孔硅微球包裹T-705药物,能够使药物在脑内进行缓释,提高了脑内不同时间点内药物在细胞内的浓度,在体内能够实现缓慢并持续释放药物,从而长时间地抑制脑内病毒的复制,控制脑内病毒载量;
2、本发明通过构建此药物载体,突破传统抗病毒预防与治疗中药物递送效率低下的科学问题,提供一种高效脑内递送的药物载体,用于脑内嗜神经病毒感染的抗病毒治疗;
3、本发明纳米颗粒释放药物抑制病毒复制,其细胞损伤的程度远低于3.3中纳米金棒所述,对脑内细胞来说,表现出更强的生物安全性;
4、本发明T-705@MSN-RVG药物载体,未使用任何重金属或难以代谢消化的金属元素,介孔二氧化硅本身在体内会被缓慢代谢消化,从而减轻了肝肾的负担。

为了使本发明的目的、技术方案及优点更加清楚明白,以下结合附图以及各实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,并不用于限定本发明,即所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。
在这里专用的词“实施例”,作为“示例性”所说明的任何实施例不必解释为优于或好于其它实施例。本法实施例中性能指标测试,除非特别说明,采用本领域常规试验方法。本发明中所述的术语仅仅是为描述特别的实施方式,并非用于限制本发明公开的内容。
除非另有说明,否则本文使用的技术和科学术语具有本发明所属技术领域的普通技术人员通常理解的相同含义;作为本发明中的其它未特别注明的原材料、试剂、试验方法和技术手段均指本领域内普通技术人员通常使用的原材料和试剂,以及通常采用的实验方法和技术手段。
如图3所示,为本发明提供的T-705@MSN-RVG药物载体的合成以及作用示意图,T-705@MSN-RVG药物载体的合成步骤:包括介孔硅MSN的合成、药物包裹、RVG多肽的表面修饰。T-705@MSN-RVG药物载体的作用方式:通过静脉注射至小鼠体内,其药物载体被递送至脑内感染细胞处,释放药物,抑制病毒的复制。
本发明实施例合成过程中采用的试剂和仪器如表1所示。

实施例
本实施例提供的T-705@MSN-RVG药物载体,其合成过程按照如下步骤进行操作:
1)称取13.2mg法匹拉韦(T-705)粉末,使用660uL无水乙醇溶解(加入10uL DMSO助溶)
2)取2mL,2.5%浓度的羧基介孔硅(MSN)溶液(分散于无水乙醇中),10000rpm离心10min去除多余的乙醇溶液。
3)将法匹拉韦溶液加入羧基介孔硅沉淀中,并水浴超声分散使介孔硅分散于药物溶液中,300rpm 37℃震荡孵育过夜,孵育完成的溶液10000rpm离心10min去除上清(保存全部上清用于载药量测试),并使用pH5.5的0.02mM MES缓冲液离心清洗三次以去除多余的有机溶剂
4)使用pH5.5的0.02mM MES缓冲液配制5mg/mL的多肽(Fpp)溶液共600uL,加入载有法匹拉韦的介孔硅溶液中,另10mg/mL的EDC溶液(现配现用),取250uL(2.5mg)加入介孔硅与多肽混合溶液中,室温孵育过夜。多肽(Fpp)序列:N端-YTIWMPENPRPGTPCDIFTNSRGKRASNG-C端。
5)孵育结束后采用离心去除游离的多肽,以10000rpm的转速离心10min,洗涤3次,由于多肽带有荧光,肉眼可见的多肽均修饰于介孔硅上
6)将最终的产品分散在纯水中定容至5mL、10mg/mL。
验证过程如下:
1、T-705@MSN-RVG药物载体的体外抗病毒实验
建立小鼠N2a细胞感染模型:N2a细胞感染模型:于24孔细胞培养板中接种N2a细胞3×105(细胞/孔)所用培养液为含有10%血清的DMEM培养基,过夜培养后使其生长成为单层细胞。弃去培养基,使用PBS清洗3遍,使用VSV病毒以MOI=0.01的感染复数接种细胞并在37℃下孵育1h。弃去病毒液,使用PBS清洗3遍,每孔加入500μL维持液,所用维持液为含有2%血清、1%低熔点琼脂糖、3μg/mLT-705@MSN-RVG药物载体的DMEM培养基,37℃孵育48h。使用PBS清洗3遍,进行细胞固定与细胞穿孔后,通过DAPI染色液进行细胞核染色。
模型原理:在MOI=0.01的感染复数下,病毒一步生长曲线包括潜伏期、裂解期、平稳期。在药物干预的情况下,病毒生长各阶段皆受到抑制作用,进而更易于体现出药物抑制效果。其次,细胞通过低熔点琼脂糖固定后,病毒感染只能从感染细胞至临近细胞之间传递入侵,故当检测时,通过病毒荧光斑的大小可以确定病毒感染以及复制的程度。
检测方式通过荧光显微镜观察蓝色与绿色荧光。蓝色荧光代表细胞核所在的位置,其作用在于荧光视角下指示所有细胞存在的位置。绿色荧光代表病毒所在的位置,绿色荧光的大小代表了病毒感染细胞的程度。
相关数据见图4。图4中DAPI、VSV代表两个检测通道,分别检测蓝色、绿色荧光。Merge代表蓝绿色的重叠图。DAPI为蓝色荧光,代表细胞核所在的位置,其作用在于荧光视角下指示所有细胞存在的位置。VSV代表绿色荧光,代表病毒所在的位置,绿色荧光的大小代表了病毒感染细胞的程度。Merge代表本视角是前面所有视角叠加在一起的重叠图,图4中为蓝色与绿色荧光的重叠图。图4中纵向排列的三个词,代表分组情况,从上到下:病毒感染+药物载体抑制组;病毒感染对照;无病毒感染无药物组(Mock)。
如图4所示,在小鼠N2a细胞感染模型中,以MOI=0.01的感染复数对N2a细胞感染VSV(水疱性口炎)病毒。感染24h后,经过T-705@MSN-RVG药物载体处理的N2a细胞,其感染的荧光斑相对于VSV病毒对照组显著减小。证明T-705@MSN-RVG药物载体释放T-705药物,并抑制了VSV病毒在N2a细胞模型中的复制水平。
2、T-705@MSN-RVG药物载体的体内抗病毒实验
建立小鼠活体感染模型:
小鼠感染模型:准备6周龄Balb/c雌性小鼠,将小鼠随机分为3组,分别为VSV组(只注射病毒对照组)、VSV+T-705@MSN-RVG组(注射病毒+注射药物载体组)、T-705@MSN-RVG组(未注射病毒+注射药物载体组),每组13只小鼠。
对VSV组以及VSV+T-705@MSN-RVG组脑内注射20μL的VSV病毒液(5FFU/μL),T-705@MSN-RVG组脑内注射20μL PBS作为对照。对VSV+T-705@MSN-RVG组以及T-705@MSN-RVG组尾静脉注射50μL的T-705@MSN-RVG(2μg/μL),对VSV组尾静脉注射50μLPBS作为对照。
连续饲喂21日,记录每组的死亡数,计算21日后3组小鼠存活率曲线。
模型原理:脑内注射100FFU的VSV病毒会导致6周龄Balb/c小鼠死亡80-100%之间。此攻毒剂量刚好使大部分6周龄Balb/c小鼠死亡。在此剂量下进行药物载体的抗病毒实验,根据感染小鼠给药后存活情况评价药物载体的抗病毒效果。
计算的相关数据:饲喂21日后,T-705@MSN-RVG组全体存活(存活率100%),说明药物载体安全性高,不会导致小鼠意外死亡。VSV组存活23%(共13只,存活3只)。VSV+T-705@MSN-RVG组存活77%(共13只,存活10只)。
如图5所示,在小鼠活体感染模型中,6周龄的Balb/c小鼠(n=13)通过i.c.注射VSV病毒后仅存约23%小鼠存活。当T-705@MSN-RVG药物载体注射小鼠后,其存活率提升至约77%。证明T-705@MSN-RVG药物载体的小鼠体内实验显著提升了小鼠感染VSV病毒后的存活率。其中,存活率100%的是药物载体对照组,即未注射病毒+注射药物载体组,证明药物无毒副作用。
3、T-705@MSN-RVG药物载体的细胞安全性实验
实验过程:使用Vybrant MTT细胞增殖分析试剂盒(Sigma)进行细胞活力的测定。将N2a细胞在96孔版中接种1×105(细胞/孔)后,于37℃下培养过夜,待长成单层细胞后,用PBS清洗3次。每孔加入100μL维持液,所用维持液为含有2%血清、梯度稀释的T-705@MSN-RVG(浓度梯度设置:0.03125、0.0625、0.125、0.25、0.5、1、2、4mg/mL,同时设置等量DMEM培养液作为对照)。加入维持液后,在37℃下培养48h。PBS清洗3次后,每孔加入100μL维持液(含10μL,12mM的MTT原液)。在37℃下培养4h检测450nm处的吸光度值。图6纵坐标表示为样品的OD450值与DMEM对照OD450平均值的比值,横坐标表示为样品的浓度梯度。
如图6所示,当使用极高浓度药物载体进行细胞实验时(2mg/mL),细胞活性未表现出显著的下降。说明此药物载体安全性高。
药物成份检测显示该药物载体主要由C、O、Si元素组成,不含有任何重金属元素成分,相比纳米金棒,本药物载体更易于代谢消化,不存在长期难以代谢消化的元素。
4、对T-705@MSN-RVG药物载体在小鼠脑内持续抑制病毒复制的验证实验
实验过程:准备6周龄Balb/c雌性小鼠,将小鼠随机分为2组,分别为VSV组(只注射病毒对照组)、VSV+T-705@MSN-RVG组(注射病毒+注射药物载体组),每组15只小鼠。
对VSV组以及VSV+T-705@MSN-RVG组脑内注射20μL的VSV病毒液(5FFU/μL)。对VSV+T-705@MSN-RVG组尾静脉注射50μL的T-705@MSN-RVG(2μg/μL),对VSV组尾静脉注射50μLPBS作为对照。连续饲喂6日,分别于第2,4,6日,每天每组取5只小鼠鼠脑,用于检测脑内病毒基因组RNA水平。
病毒基因组RNA的检测:收集的鼠脑通过JX-FSTPRP-64匀浆器(Jingxin)匀浆,以10000rpm离心5min。取上清液用TRIZol试剂(Invitrogen)提取病毒基因组RNA。通过HiScript逆转录酶试剂盒(Vazyme)转录成病毒基因组cDNA用于荧光定量qPCR检测。
病毒基因组检测引物:(正向引物5'-AGTCTAGCTTCCAGCTTCTGA-3',反向引物5'-ACAGGATATTAGTTGTTCGAAAGGC-3')。
扩增体系:2×SYBR green master mix(Biorad)5μL,正向引物(0.75μmol/L)1μL,反向引物(0.75μmol/L)1μL,cDNA1μL,ddH2O 2μL。
荧光定量qPCR扩增程序:95℃预变性2min;95℃30s,60℃30s,40个循环;荧光检测通道为SYBR。
结果计算:检查扩增曲线和各样品Ct值,将所得Ct值代入标准曲线计算公式(拷贝数=(Ct-53.717)/-5.195),计算样品中每μg鼠脑总RNA中所含的病毒基因组RNA拷贝数,并作为纵坐标作图,统计分析2,4,6日不同组小鼠脑内的病毒基因组RNA水平差异。
如图7所示,通过持续监控小鼠脑内的VSV病毒基因组RNA水平,证明了注射T-705@MSN-RVG药物载体后,小鼠脑内VSV病毒基因组RNA水平受到持续抑制,分别在感染后4、6天,脑内病毒水平显著低于病毒对照组。图中ns代表无显著性差异。*代表有显著性差异,一颗*代表差异P值<0.05,两颗*代表差异P值<0.01,三颗*代表差异P值<0.001,四颗*代表差异P值<0.0001。*数越多代表差异越大。
以上所述实施例仅表达了本申请的具体实施方式,其描述较为具体和详细,但并不能因此而理解为对本申请保护范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本申请技术方案构思的前提下,还可以做出若干变形和改进,这些都属于本申请的保护范围。
Claims (6)
1.一种T-705@MSN-RVG脑内药物递送载体,其特征在于,包括羧基介孔硅及包覆在羧基介孔硅内的法匹拉韦,所述羧基介孔硅的表面修饰有RVG多肽。
2.如权利要求1所述的T-705@MSN脑内药物递送载体的制备方法,其特征在于,包括如下步骤:
(1)将法匹拉韦溶于乙醇中形成法匹拉韦溶液;
(2)将羧基介孔硅分散于法匹拉韦溶液中,并在35~40℃震荡过夜,然后离心去除上清,并采用pH5.5的MES缓冲液清洗溶剂,得到T-705@MSN;
(3)将T-705@MSN和RVG多肽溶液混合,加入现配的EDC溶液,室温反应过夜,离心去除上清,并洗涤所得沉淀,即得到T-705@MSN-RVG脑内药物递送载体。
3.如权利要求2所述的T-705@MSN脑内药物递送载体的制备方法,其特征在于,步骤(1)中,法匹拉韦溶解过程中还加有DMSO用于助溶。
4.如权利要求2所述的T-705@MSN脑内药物递送载体的制备方法,其特征在于,步骤(2)中,采用的MES缓冲液的浓度为0.02mM。
5.如权利要求2所述的T-705@MSN脑内药物递送载体的制备方法,其特征在于,步骤(3)中,所述RVG多肽溶液的浓度为5mg/mL,溶剂为pH5.5的MES缓冲液。
6.如权利要求2所述的T-705@MSN脑内药物递送载体的制备方法,其特征在于,步骤(3)中,所述EDC溶液的浓度为10mg/mL。
