Abstract
本发明公开了乙酸薄荷酯在驱避红火蚁中的应用。本发明从昆虫体内提取的化合物中筛选和开发的乙酸薄荷酯对红火蚁工蚁、有性雌蚁和雄蚁均具有较好的驱避效果。乙酸薄荷酯可以作为一种天然的红火蚁驱避剂使用,对人体和环境无害,因此具有较高的市场应用价值。

技术领域
本发明属于红火蚁高效监测和综合防控技术领域,具体涉及乙酸薄荷酯在驱避红火蚁中的应用。
背景技术
红火蚁(Solenopsis invicta)是最具危险性的外来入侵物种之一,入侵后不仅会捕食或攻击其他物种,还会啃食农作物,导致农作物产量降低,对当地人畜安全、生物多样性造成巨大威胁。
使用驱避剂可以防止红火蚁在房前屋后等地方筑巢,还可以涂抹在皮肤上防止在野外被红火蚁叮咬。由于驱避剂可能与人类直接接触,所以相较于化学合成药剂,天然产物更能体现其安全性并被公众所接受。同时使用驱避剂结合引诱剂和杀虫剂对红火蚁进行处理,可以对红火蚁起到很好的防治效果。
发明内容
本发明的目的是针对目前红火蚁在全国范围内泛滥成灾、不断危害公共安全、人畜健康但目前市面上针对红火蚁的驱避剂种类较少等问题,提供一种对红火蚁不同品级的蚂蚁均具有较强驱避作用的天然昆虫源化合物药剂及其应用。
本发明通过使用气相色谱仪(GC-MS)、昆虫触角电位仪(EAG)以及诺达思昆虫行为轨迹跟踪仪EthovisionXT等仪器,发现并证实了乙酸薄荷酯对红火蚁工蚁、有性雌蚁和雄蚁均具有较强的驱避作用。
因此,本发明的第一个目的是提供乙酸薄荷酯在防治蚁类害虫中的应用。
优选,所述的应用为乙酸薄荷酯在驱避蚁类害虫中的应用。
优选,所述的蚁类害虫为红火蚁。
优选,所述的红火蚁为红火蚁工蚁、有性雌蚁和雄蚁。
本发明的第二个目的是提供一种驱避红火蚁的方法,其包括将乙酸薄荷酯或乙酸薄荷酯的溶液喷洒在待驱避红火蚁的区域的步骤。这样可以有效地减少红火蚁的出现和攻击。
优选,所述的乙酸薄荷酯的溶液,其浓度为1 µg/mL以上。
本发明筛选的乙酸薄荷酯对红火蚁工蚁、有性雌蚁和雄蚁均具有较好的驱避效果。因此,本发明提供了一种用于防治重大入侵性害虫红火蚁及其他蚂蚁的驱避剂和驱避方法,可以用于红火蚁的驱避剂开发及防治应用。相比较于现阶段其它针对红火蚁的驱避剂,乙酸薄荷酯是昆虫天然产生的化合物,更加绿色和环保;而且其对红火蚁不同品级的蚂蚁均具有驱避性,可以作为一种天然的红火蚁驱避剂使用,对人体和环境无害,因此具有较高的市场应用价值。

具体实施方式
以下实施例是对本发明的进一步说明,而不是对本发明的限制。
实施例1:乙酸薄荷酯的鉴定
利用气相色谱-质谱联用仪(型号:安捷伦8890-5977B GC-MS)对乙酸薄荷酯进行分析,具体条件:色谱柱为HP-5ms (30 m, 0.25 mm内径, 0.25 μm膜厚),载气为高纯氮气,进样口温度250℃,FID检测器温度320℃,离子源电压EI 70 eV,柱接口温度为280℃,扫描离子范围为50~550 m/Z。柱箱升温程序为50℃,保留时间为2 min,随后以15℃/min升至200℃并保持5 min,再以15℃/min升至280℃并保持5 min。每次取样1 μL进行手动进样。
图1结果表明,乙酸薄荷酯标准品保留时间为9.184 min,其中,特征离子碎片m/z分别为55、67、81、95、109和123,分子离子m/z为138。
实施例2:昆虫触角电位(EAG)试验测定红火蚁对乙酸薄荷酯的触角电位反应
使用GC-EAD测试红火蚁大型工蚁、有性雌蚁、雄蚁对乙酸薄荷酯的触角电生理反应。使用维纳斯剪沿红火蚁触角基部剪下触角,并用刀片将触角尖端切掉一小节(约1 mm)。触角的基部和尖端分别连接两根充满0.9%生理盐水的玻璃毛细管(直径1.1 mm),并通过银丝连接到参考电极和记录电极。使用正己烷将乙酸薄荷酯稀释至1 µg/mL。气相色谱型号为安捷伦7890B,色谱条件为:HP-5ms (30 m, 0.25 mm内径, 0.25 μm膜厚) 毛细管色谱柱,载气为高纯氮气,进样口温度250℃,FID检测器温度320℃。柱箱升温程序为初始温度50℃,保留时间为2 min,随后以15℃/min升至200℃并保持5 min,再以15℃/min升至280℃并保持5 min。GC采用手动进样及不分流模式,进样量为1 µL,GC和EAG的样品分流比为1:1,每次实验重复6根触角以上。
图2结果表明,乙酸薄荷酯可以引起红火蚁大型工蚁、有性雌蚁、雄蚁强烈的电生理反应,1 µg/mL的乙酸薄荷酯所引起的大型工蚁、有性雌蚁和雄蚁的触角电位变化值分别为0.076 ± 0.003 mV、0.074 ± 0.003 mV和0.073 ± 0.001 mV,说明乙酸薄荷酯可能是对红火蚁具有较强生理活性的物质。
实施例3:乙酸薄荷酯对单只红火蚁的驱避性测试
采用Ethovision XT11.5软件记录乙酸薄荷酯对单只红火蚁(大型工蚁、有性雌蚁、雄蚁)的驱避行为特性。在内壁涂有聚四氟乙烯和滑石粉的圆形玻璃碗(10 cm ×5 cm)的两处对角处分别放置半径为1 cm的圆形滤纸片,分别为对照区和处理区。将单只红火蚁放置在玻璃碗内,先不做处理,让其在容器内活动5 min以适应环境。5 min后,往处理区滤纸片上滴2 µL经正己烷稀释至浓度为1 µg/mL的乙酸薄荷酯溶液(Menthyl acetate),另外一片滤纸滴加2 µL正己烷(Hexane)作为对照。利用Ethovision XT自带的视频软件拍摄记录红火蚁在玻璃碗内的活动时间20 min,并自动统计红火蚁在培养皿内两个不同区域的活动时间和活动轨迹,利用SPSSt检验对红火蚁在不同区域的活动时间进行差异显著性比较(P= 0.05)。三种品级的红火蚁均重复10次试验。
图3结果表明乙酸薄荷酯对红火蚁工蚁、有性雌蚁、雄蚁均具有较强的驱避作用,三个品级的红火蚁停留在对照区域的时间显著高于停留在处理区域的时间。其中乙酸薄荷酯对雄蚁的驱避作用最为显著,在处理区域的停留时间占比为32.79%,对照区域为67.21%。(t = -3.352,df = 9,p=0.004),其次为有性雌蚁,在处理区域的停留时间占比为33.63%,对照区域为66.37%。(t = -3.476,df = 9,p = 0.003),最后为工蚁,在处理区域的停留时间占比为36.75%,对照区域为63.25%(t = -3.246,df = 9,p = 0.005)。

4.一种驱避红火蚁的方法,其特征在于,包括将乙酸薄荷酯或乙酸薄荷酯的溶液喷洒在待驱避红火蚁的区域的步骤。
5. 根据权利要求4所述的方法,其特征在于,所述的乙酸薄荷酯的溶液,其浓度为1 µg/mL以上。
