Abstract
本发明属于生物技术领域,具体涉及一种假病毒高效感染人NK细胞及其他免疫细胞的方法。具体地,本发明所提供的病毒转染系统中的包膜质粒上具有结构为X‑Y‑Z的蛋白,所述X是长臂猿白血病病毒包膜糖蛋白的胞外结构,Y是长臂猿白血病病毒包膜糖蛋白的跨膜结构,Z是鼠病毒基因的胞内段部分。
Description
技术领域
本发明属于生物技术领域,具体涉及一种假病毒高效感染人NK细胞及其他免疫细胞的方法。
背景技术
固有免疫系统中的NK细胞具有优秀的杀伤能力,并且仅表达模式识别受体,因此可以实现异体输注并且没有显著的移植物抗宿主反应(GVHD)。基于NK细胞制备CAR-NK细胞产品能够实现规模化生产,实现异体输注,是下一代的细胞治疗产品,将会替代现有的CAR-T产品。
制备CAR-NK产品,目前主要采用的基因递送方法是病毒转导系统,包括慢病毒系统、逆转录病毒系统,以及少数公司采用转座子/转座酶的方法。以上这些方法无一例外都会遇到一个技术难点,就是基因递送的效率太低且不稳定。大多数CAR-NK细胞产品的CAR阳性率仅有10%左右,达不到细胞类产品的最低要求:CAR阳性率≥30%。并且随着后续CAR-NK细胞的扩增,CAR阳性率还会进一步下降,仅有1-2%的阳性率。因此,优化基因递送的方法,尽可能提高对NK细胞的转导率,获得高比例的CAR-NK产品是目前研发的重点及难点。
另外,根据文献的报道,慢病毒系统采用的VSV-G或MD2.G包膜蛋白能够抑制NK细胞增殖,甚至诱导NK细胞凋亡,导致CAR-NK细胞的阳性率不断下降,最终CAR-NK细胞的阳性率会降低至1-2%,无法达到制备CAR-NK细胞产品的要求。
发明内容
为解决以上技术问题,本发明通过生物信息学分析及实验筛选,构建了一种新的病毒包膜蛋白(Envelope),进而形成一种新的假病毒以及病毒转导体系。这种新的假病毒不再依赖低密度脂蛋白受体(LDLR)感染NK细胞,而是通过另一种在免疫细胞上高表达的受体SLC20A1感染细胞,将目的基因递送至NK细胞内。采用这种假病毒进行基因递送,我们可以获得转导率高达90%的CAR-NK细胞。并且在维持高阳性率的同时不抑制NK细胞的扩增,CAR-NK细胞可以扩增2500倍。
具体地,本发明提供以下技术方案:
蛋白
一方面,本发明提供了一种蛋白,所述蛋白具有X-Y-Z结构,所述X是长臂猿白血病病毒(Gibbon ape leukemia virus,GALV)包膜糖蛋白(Envelope glycoprotein)的胞外结构(ex),Y是长臂猿白血病病毒包膜糖蛋白的跨膜结构(TM),Z是鼠病毒基因的胞内段部分(MEVc)。
优选地,所述长臂猿白血病病毒(Gibbon ape leukemia virus,GALV)包膜糖蛋白(Envelope glycoprotein)的胞外结构与SEQ ID NO.1具有至少80%、至少85%、至少90%、至少91%、至少92%、至少93%、至少94%、至少95%、至少96%、至少97%、至少98%、或至少99%的序列同一性,或者,与SEQ ID NO.1相比具有1个、2个、3个、4个、5个、6个、7个、8个、9个或10个氨基酸的置换、缺失或添加。

更优选地,所述Z的氨基酸序列如SEQ ID NO.3所示。更具体地,本发明所述MEVc是GenBank:AH002056.2(Mus musculus endogenous virus endogenous Moloney-likemurine retrovirus N-myc and env polypeptide genes)基因的胞内段前17个氨基酸。
也即,最优选地,本发明所述蛋白的序列是SEQ ID NO.1、SEQ ID NO.2和SEQ IDNO.3依次连接所组成的蛋白(或称为肽、多肽、肽段)。
本领域熟知,可以改变蛋白质的结构或序列而不对其活性和功能产生不利影响,例如可以在蛋白质氨基酸序列中引入一个或多个保守性氨基酸取代,而不对蛋白质分子的活性和/或三维结构产生不利影响;本领域技术人员清楚保守性氨基酸取代的实例以及实施方式。具体的说,只要取代不导致蛋白质生物活性的失活,则一种氨基酸被其他氨基酸替换的保守取代落在本发明的范围内。另外,本领域熟知,可以从蛋白质的N和/或C末端改变(置换、删除、截短或插入)一或多个氨基酸残基而仍保留其功能活性。
如本文中所使用的,术语“同一性”用于指两个多肽之间或两个核酸之间序列的匹配情况。当两个进行比较的序列中的某个位置都被相同的碱基或氨基酸单体亚单元占据时(例如两个多肽的每一个中的某个位置都被赖氨酸占据),那么各分子在该位置上是同一的。
在一种具体实施例中,将本发明蛋白命名为QMV,以QMV为包膜蛋白的病毒转导系统也称为QMV病毒转导系统。
融合蛋白
另一方面,本发明提供了包含以上蛋白的融合蛋白。
优选地,所述融合蛋白还可以包括其他修饰部分,所述修饰部分包括表位标签(epitope tag)、可检测标记、核定位信号(NLS)序列等。
具体地,所述表位标签是本领域技术人员熟知的,包括但不限于His、V5、FLAG、HA、Myc、VSV-G、Trx等,并且本领域技术人员可以选择任意合适的表位标签(例如,纯化、检测或示踪);所述可检测标记包括报告基因序列,或者,荧光染料,例如FITC或DAPI;所述报告基因序列是本领域技术人员熟知的,其实例包括但不限于GST、HRP、CAT、GFP、HcRed、DsRed、CFP、YFP、BFP等。
本发明的蛋白或融合蛋白不受其产生方式的限定,例如,其可以通过基因工程方法(重组技术)产生,也可以通过化学合成方法产生。
核酸分子
另一方面,本发明提供了一种核酸分子,所述核酸分子包含编码本发明的蛋白或融合蛋白的核酸序列。
更优选的,所述核酸分子是编码本发明蛋白或融合蛋白的核酸序列。
本领域熟知,不同生物在翻译过程中对简并密码子的使用频率存在差异,而在进化的过程中形成了一套与其相适应的常用密码子(密码子偏好性)。密码子优化则是通过基因工程技术,根据宿主的密码子偏好性调整基因的同义密码子,达到消除稀有密码子的目的,并且优化如mRNA二级结构和Motif等相关参数,从而提高翻译效率。为提高在不同宿主中的表达量,对密码子进行优化而得到的编码序列均落入本发明核酸分子的保护范围之内。
在一个实施方式中,所述核酸分子经密码子优化用于在原核细胞中进行表达。在另一个实施方式中,所述核酸分子经密码子优化用于在真核细胞中进行表达。所述真核细胞包括人或其他动物的细胞。
在本说明书中可互换使用的术语“核酸”、“核酸分子”、“核酸序列”、“多核苷酸”、“寡核苷酸”、“编码核酸”和“核苷酸序列”意指精确的核苷酸序列。
载体
另一方面,本发明还提供了一种载体,所述载体表达前述蛋白或融合蛋白、或者,所述蛋白上包含前述核酸分子。
优选地,所述载体上还包含本领域所常见的调控元件。
优选地,所述调控元件包括以下任意一种或多种:增强子、转座子、启动子、终止子、前导序列、多腺苷酸序列、标记基因。
优选地,本发明具体实施例中以CMV为启动子。所述CMV启动子是巨细胞病毒(Cytomegalovirus,CMV)中发现的强启动子,其功能和序列是本领域所熟知的。
在一种具体地实施例中,所述载体可称为病毒转导体系中的包膜质粒,与其他表达病毒基因的载体组合成为病毒转导体系,实现病毒转导功能。
载体组合物
另一方面,本发明还提供了一种载体组合物,所述载体组合物中包括第一载体成分和第二载体成分,所述第一载体成分包括上述载体,所述第二载体成分中的载体或载体组合物上含有编码组装慢病毒所需蛋白的编码核酸,或者,所述第二载体成分表达组装慢病毒所需蛋白。
优选地,所述组装慢病毒所需蛋白包括REV、GAG和POL中至少一种。
优选地,所述组装慢病毒所需蛋白包括REV、GAG和POL。
优选地,所述第二载体成分中还可以包括以下任意一种或多种基因的编码核酸:调控基因tat,辅助基因vpr、vif、vpu和nef,长末端重复序列(long terminal repeat,LTR),病毒包装信号等。
具体地,所述第二载体成分可以是一个质粒或多个质粒,目的在于表达REV、GAG和POL,所述REV、GAG和POL的编码核酸可以连接在同一个载体上或者连接在不同的载体上。
更具体地,所述第二载体成分可以是市售的慢病毒转导产品中的包装质粒。根据慢病毒转导系统的构成,所述第二载体成分的构成是容易确定的。
在一种具体的实施例中,所述第二载体成分包括包装质粒1和包装质粒2,所述包装质粒1上包含GAG和POL的编码核酸,所述包装质粒2上包含REV的编码核酸。
在另一种具体的实施例中,所述第二载体成分是1个质粒,所述质粒上包含REV、GAG和POL的编码核酸。
如本领域所公知的,GAG基因编码病毒的核心蛋白如核衣壳蛋白(p7)、内膜蛋白(p17)和衣壳蛋白(p24),POL基因编码病毒复制相关的酶,REV是一种RNA结合蛋白,保证基因的表达。以上基因的功能和其具体序列是本领域所公知并容易获得的。
在一种具体实施例中,所述载体组合物就可以称为一种系统(病毒系统、病毒转导系统、病毒包装系统),该载体组合物具备产生病毒颗粒的功能。
更优选地,所述载体组合物包括以下质粒:
1)表达质粒(含有目标基因的编码序列或表达目标基因的蛋白)、
2)包装质粒1(含有GAG和POL的编码序列或表达GAG和POL)、
3)包装质粒2(含有REV的编码序列或表达REV)、和、
4)包膜质粒(前述载体)。
优选地,所述包装质粒1、包装质粒2、包膜质粒上各自独立地包括CMV启动子。
组合物
另一方面,本发明提供了一种组合物,所述组合物中包含以上载体组合物以及转染试剂。
具体地,所述转染试剂具有将目标基因(在本发明中可以是病毒载体)导入目标细胞的作用,示例性的,所述转染试剂可以是市售的产品,例如Lipofectamine2000及其相似产品(thermofisher)、Lipo293FTM转染试剂及其相似产品(deyotime)、X-tremeGENETM360DNA Transfection Reagent及其相似产品(Roche)、高效核酸转染试剂盒(L3170,Solarbio)、LipoJetTM转染试剂盒(Ver.II)等。或者,所述转染试剂还可以是各种型号的聚乙烯亚胺(Polyethylenimine,PEI)及其衍生物,所述PEI及其衍生物可以单一成分使用、或者、多种成分混合使用。
宿主细胞
另一方面,本发明还提供了一种宿主细胞,所述宿主细胞中表达以上蛋白或融合蛋白,或者,所述宿主细胞中含有以上核酸分子、载体或载体组合物。
优选地,所述宿主细胞包括原核细胞、真核细胞。
优选地,所述真核细胞包括低等真核细胞和高等真核细胞。
优选地,所述高等真核细胞包括动物细胞,哺乳动物细胞。
优选地,所述哺乳动物细胞包括免疫细胞、干细胞、体细胞、血液细胞等。
优选地,所述免疫细胞包括T细胞(T淋巴细胞)、B细胞(B淋巴细胞)、NK细胞(NK淋巴细胞)、NKT细胞、树突状细胞、浆细胞、粒细胞、肥大细胞、抗原呈递细胞及单核吞噬细胞等。
优选地,所述干细胞包括胚胎干细胞、成体干细胞。
优选地,所述干细胞来源包括胚胎、骨髓、周边血、脐带和脐带血。

本领域公知,被称为“万能细胞”的干细胞,按其所处的发育阶段,可分为胚胎干细胞和成体干细胞。根据干细胞的发育潜能可分为全能干细胞、多能干细胞和单能干细胞。按成体干细胞的功能,可将其分为神经干细胞、造血干细胞、骨髓间质干细胞、皮肤干细胞、脂肪干细胞等。
优选地,所述哺乳动物是人,所述细胞是人的细胞。
在一种具体地实施例中,所述宿主细胞可以是制备病毒液过程中所使用的细胞。所述宿主细胞受病毒转导系统转染后,表达出的病毒颗粒,从而可应用于后续的病毒转导。
在作为所述制备病毒液所使用的宿主细胞时常用的细胞包括各种细胞株,例如293细胞、293T细胞、293FT细胞、293LTV细胞、293F细胞、293EBNA细胞及其他的从293细胞分离的克隆;SW480细胞、u87MG细胞、HOS细胞、C8166细胞、MT-4细胞、Molt-4细胞、HeLa细胞、HT1080细胞、TE671细胞。
优选地,本发明在制备病毒液时所使用的细胞株是HEK293T(又称293T,人胚胎肾细胞源性细胞系)。
制备病毒液的方法
另一方面,本发明提供了一种制备病毒液的方法,所述方法包括将载体组合物转染到细胞(或称宿主细胞)中的步骤:所述载体组合物中包括第一载体成分和第二载体成分,所述第一载体成分是上述载体,所述第二载体成分中的载体或载体组合物上含有编码组装慢病毒所需蛋白的编码核酸,或者,所述第二载体成分表达组装慢病毒所需蛋白。
优选地,所述组装慢病毒所需蛋白包括REV、GAG和POL。
优选地,所述第二载体成分中还可以包括以下任意一种或多种基因的编码核酸:调控基因tat,辅助基因vpr、vif、vpu和nef,长末端重复序列(long terminal repeat,LTR),病毒包装信号等。
具体地,所述第二载体成分可以是一个质粒或多个质粒,目的在于表达出REV、GAG和POL,所述REV、GAG和POL的编码核酸可以连接在同一个载体上或者连接在不同的载体上。
在一种具体的实施例中,所述第二载体成分包括包装质粒1和包装质粒2,所述包装质粒1上包含GAG和POL的编码核酸,所述包装质粒2上包含REV的编码核酸。
在另一种具体的实施例中,所述第二载体成分是1个质粒,所述质粒上包含REV、GAG和POL的编码核酸。
更优选地,所述载体组合物包括以下质粒:
1)表达质粒(含有目标基因的编码序列或表达目标基因的蛋白)、
2)包装质粒1(含有GAG和POL的编码序列或表达GAG和POL)、
3)包装质粒2(含有REV的编码序列或表达REV)、和、
4)包膜质粒。
更优选地,所述方法还包括提纯病毒液的步骤。
更优选地,在制备内含有特定目标基因的病毒液时,还需要向细胞中转染表达目标基因的载体。
优选地,本发明所述目标基因可以是任意一种基因的编码序列,示例性的,所述目标基因包括抗体、嵌合抗原受体(Chimeric Antigen Receptor,CAR)或功能性蛋白的编码核酸。
本领域公知,术语“嵌合抗原受体(CAR)”可以包含抗原结合结构域(例如靶向癌症治疗靶点的抗体)、跨膜结构域、共刺激域及细胞内信号转导结构域。随CAR技术发展,还可以在CAR结构上引入一个或更多个共刺激分子或者其他成分。无论结构如何变化,以上CAR或其相似结构的编码基因都可以作为本发明所述目标基因。
如本领域公知,所述将载体组合物转染细胞所使用的转染方法是常规的。如本发明所使用的转染方法是通过PEI促进转染,PEI的使用方法是本领域所熟知的。具体地,本发明中所使用PEI与载体组合物的体积质量比为3:1,其比例可根据具体PEI类型确定,其确定方式是本领域所公知的。转染方法中可能使用的试剂如前述成品化的转染试剂或自行配制的促进转染的试剂,其配置方法是本领域所熟知的。
病毒颗粒
另一方面,本发明提供了一种病毒颗粒,所述病毒颗粒中包括本发明蛋白和组装慢病毒所需蛋白。
优选地,所述组装慢病毒所需蛋白包括REV、GAG和POL。
优选地,所述组装慢病毒所需蛋白还包括以下任意一种或多种:调控基因tat,辅助基因vpr、vif、vpu和nef等。
优选地,所述病毒颗粒中还含有目标蛋白(目标基因所编码出的蛋白)。
优选地,本发明所述目标基因可以是任意一种基因的编码序列,示例性的,所述目标基因包括抗体、嵌合抗原受体(Chimeric Antigen Receptor,CAR)或功能性蛋白的编码核酸,也即,所述目标蛋白包括抗体、嵌合抗原受体或功能性蛋白。
在一种具体的实施例中,所述病毒颗粒是前述制备病毒液的方法所制备得到的主要产物。
一种制备表达目标基因的细胞的方法
另一方面,本发明提供了一种制备表达目标基因的目标细胞的方法,所述方法包括将载体组合物转染细胞,收集病毒液,然后将病毒液与目标细胞接触的步骤。
在一种具体的实施例中,所述细胞和目标细胞是/不是相同的细胞,其具体的细胞类型可自独立的可选任意一种合适的细胞,选择合适细胞的方式是本领域所熟知的。
所述载体组合物中包括第一载体成分和第二载体成分,所述第一载体成分是上述载体,所述第二载体成分中的载体或载体组合物上含有编码组装慢病毒所需蛋白的编码核酸,或者,所述第二载体成分表达组装慢病毒所需蛋白。
优选地,所述组装慢病毒所需蛋白包括REV、GAG和POL。
优选地,所述第二载体成分中还可以包括以下任意一种或多种基因的编码核酸:调控基因tat,辅助基因vpr、vif、vpu和nef,长末端重复序列(long terminal repeat,LTR),病毒包装信号等。

更优选地,所述表达质粒、包装质粒1、包装质粒2和包膜质粒的质量比是本领域技术人员通过常规方法所容易得到的,在本发明具体实施例中使用的比例为2:1:1:1。
更优选地,所述方法还包括提纯病毒液的步骤。
在一种具体的实施例中,所述方法也可称为前述病毒颗粒的使用方法。
另一方面,本发明提供了上述制备表达目标基因的目标细胞的方法所得到的目标细胞,也即,本发明提供了通过本发明蛋白、病毒载体转导而表达目标基因的目标细胞。
另一方面,本发明提供了上述目标细胞在制备药物中的应用。
优选地,所述药物是治疗癌症的药物,具体地包括免疫疗法、细胞疗法中所使用细胞、药品。
如具体实施例所使用的,本发明制备了表达靶向BCMA的嵌合抗原受体的NK细胞,从而得到了治疗BCMA相关癌症的药物。
应用
另一方面,本发明提供了本发明蛋白、融合蛋白、核酸分子、载体、载体组合物、组合物、宿主细胞在制备表达目标基因的病毒颗粒中的应用。
另一方面,本发明提供了本发明蛋白、融合蛋白、核酸分子、载体、载体组合物、组合物、宿主细胞在通过病毒转导使目标细胞表达目标基因(制备表达目标基因的细胞)中的应用。

本发明所述“抗体”或“嵌合抗原受体(Chimeric Antigen Receptor,CAR)”可以靶向不同位点,血液肿瘤靶点:BCMA、CD19、CD20、CD123、CD22、CD3D、CD3E、CD7CLEC12AGPRC5D、CD138、CD30、CD33、CD38、CD3E、CD79BSLAMF7、CD10、CD117、CD37、CD4、CD5、CD56、CD72、CD79A、CD99、Flt-3、LILRA3、LILRB4、SLAMF3;实体肿瘤靶点:Her2、MSLN、B7-H3、CLDN18、EGFR、GPC3、KRAS(G12D)、CA9、CEA、EGFRvIII、EphA2、ERBB3、ERBB4、FAP、GUCY2C、IL13RA2、MUC1、PD-1、PSMA、VEGFR2、AFP、AXL、CD133、CD147、CD171、CD80、CD86、c-Met、DLL4、EpCAM、Nectin-4、Podoplanin、ROBO1、ROR2、SSTR2;同时靶向实体或非实体肿瘤:FOLR1、ROR1、CD70、NKG2D、PD-L1、SIRP alpha。本发明具体实施例中以靶向(特异性结合)BCMA的单抗为核心构建CAR结构作为目标基因进行验证。
本发明所用的术语“抗体”是指与特异性抗原免疫反应的免疫球蛋白分子,表示用作特异性识别抗原的受体作用的蛋白分子,该术语可包括多克隆抗体、单克隆抗体、全抗体和抗体片段。此外,该术语还可包括嵌合抗体(例如,人源化鼠抗体)、二价或双特异性分子(例如,双特异性抗体)、双抗体、三抗体和四抗体。

具体实施方式
下面结合实施例对本发明做进一步的说明,以下所述,仅是对本发明的较佳实施例而已,并非对本发明做其他形式的限制,任何熟悉本专业的技术人员可能利用上述揭示的技术内容加以变更为同等变化的等效实施例。凡是未脱离本发明方案内容,依据本发明的技术实质对以下实施例所做的任何简单修改或等同变化,均落在本发明的保护范围内。
通用方法-制备病毒液的步骤
1、准备HEK293T在DMEM+10%血清条件下培养汇聚度80%后;
2、使用PEI溶液(上海李记生物科技有限公司Cat:AC04L092)以3:1的体积质量比混合DNA生成转染复合体,4个质粒分别为表达质粒(含有本发明欲表达的目标基因)、包装质粒1、包装质粒2和包膜质粒,质量比为2:1:1:1;所述包装质粒1、包装质粒2是CellBiolabs VPK-206病毒转导产品中的质粒;
3、转染复合体室温下放置10-20分钟后,将转染复合体加入HEK293T,12小时后,去掉转染复合体,更换DMEM+5%血清后继续培养52小时后,收获病毒液,通过浓缩,测量生物滴度后-80度保存。
实施例1、本发明转导系统与第三代慢病毒包装系统的比较
使用proteinatlas.org数据库数据进行分析NK细胞的受体表达情况,分析结果如图1-4所示:Slc20a1是长臂猿白血病病毒受体,LDLR是VSVG包膜的受体,SLC1A5是Baev受体,图1-4数据显示RNA-seq数据,数据库中三次多样本测序结果,横向第一行是来源于6个样本测序,发现Slc20a1的rna表达在NK细胞是平均值50.2nTPM,LDLR表达为平均值2.5nTPM,SLC1A5表达为平均值21,横向第二行来源于4的样本测序,Slc20a1的rna表达在NK细胞是平均值295nTPM,LDLR表达为平均值39.7nTPM,SLC1A5表达为平均值53nTPM。横向第三行来源于105个样本测序,Slc20a1的rna表达在NK细胞是平均值175nTPM,LDLR表达为平均值23.7nTPM,SLC1A5表达为平均值18nTPM。
根据图1-4数据可知长臂猿白血病病毒受体(slc20A1)在各种血液细胞中的表达都明显高于LDLR(VSVG受体),也明显高于Baboon retroviral(baEV)受体(slc1A5)。
因此将SEQ ID NO.1-3依次连接后构建到载体上作为本发明病毒转导系统的包膜质粒,所述病毒转导系统的质粒结构如图5所示。

按照通用方法中的方法制备本发明病毒系统与第三代慢病毒包装系统的病毒液,并进行NK细胞转导,其中,目标基因为表达BCMA抗体的CAR结构(结构图如图6所示)。
NK细胞来源于PBMC,加入8μg/ml polybrene,加入10MOI病毒,16小时后,换新鲜NK培养基,目的是去掉含有病毒的培养基。
在NK细胞感染病毒后,第3、6、9、15天,取出细胞检测细胞活率和阳性率。具体方法是取0.5E6NK细胞,加入PBS清洗2次后,加入MACS buffer 100μl加入1μl BCMA ScFv特异性识别试剂(美天旎order#:130-126-090)室温孵育10分钟,两次洗涤后,使用流式细胞仪检测BCMA阳性细胞比率。
实验结果
图7AB显示,QMV病毒感染的阳性率第三天在80%以上,第六天下降到65%附近,第九天后,阳性率稳定在50%以上。而VSVG病毒感染,阳性率下降很快,第三天阳性率在40%,第六天下降至20%,第9天阳性率10%,第15天阳性率在5%。
同时监控第3、6、9、15天的细胞活率,QMV和VSVG感染的细胞活率在15天培养没有改变,细胞活率统计如图7C所示。
实施例2、向NK细胞和T细胞转染CAR和GFP的比较
为了检测QMV病毒是否是CAR特异性和NK特异性,进一步进行对比实验。
按照实施例1的方法使用VSV-G和QMV病毒向PB-NK、NK92和T细胞中转导BCMA CAR,转导后第9天检测阳性率,检测方法同实施例1。如图8所示,VSVG感染的NK92和PB-NK,BCMA-CAR阳性率在10%左右;QMV感染的NK92和PB-NK,BCMA-CAR阳性率在95%和60%左右。VSVG和QMV病毒感染T细胞,BCMA-CAR阳性率相似,都在83%左右。
按照实施例1的方法使用VSV-G和QMV病毒向PB-NK、NK92和T细胞中转导GFP,转导后第9天检测阳性率,检测方法同实施例1。如图9所示,VSVG感染的NK92和PB-NK,GFP阳性率分别为36%和22%,QMV感染的NK92和PB-NK,GFP阳性细胞比率为60%和50%。VSVG和QMV病毒感染T细胞,GFP阳性率相似,都在96-97%的范围内。
以上结果说明,本发明QMV病毒转导系统适用于各种免疫细胞,在表达不同目标基因时,转导效率高而且稳定。
Claims (11)
1.一种制备表达目标基因的NK细胞的方法,所述方法包括将包含包膜质粒的载体组合物转染细胞,收集病毒液,然后将病毒液与NK细胞接触的步骤,
所述包膜质粒上含有具有X-Y-Z结构的融合蛋白,所述X的氨基酸序列如SEQ ID NO.1所示,所述Y的氨基酸序列如SEQ ID NO.2所示,所述Z的氨基酸序列如SEQ ID NO.3所示。
2.如权利要求1所述方法,所述载体组合物中还含有以下质粒:
1)表达质粒,所述表达质粒含有目标基因的编码序列或表达目标基因的蛋白、
2)包装质粒1,所述包装质粒1含有GAG和POL的编码序列或表达GAG和POL、
3)包装质粒2,所述包装质粒2含有REV的编码序列或表达REV。
3.如权利要求2所述方法,所述表达质粒、包装质粒1、包装质粒2和包膜质粒的质量比为2:1:1:1。
4.如权利要求2所述方法,所述包装质粒1、包装质粒2、包膜质粒上各自独立地包括CMV启动子。
5.如权利要求1所述方法,所述方法还包括分离和/或提纯病毒液的步骤。
6.权利要求1所述方法制备得到的NK细胞。
7.权利要求6所述NK细胞在在制备药物中的应用。
8.包膜质粒或其组成的组合物在制备表达目标基因的NK细胞中的应用,
所述包膜质粒上含有具有X-Y-Z结构的融合蛋白,所述X的氨基酸序列如SEQ ID NO.1所示,所述Y的氨基酸序列如SEQ ID NO.2所示,所述Z的氨基酸序列如SEQ ID NO.3所示。
9.如权利要求8所述应用,所述NK细胞是人的细胞。
10.如权利要求8所述应用,所述目标基因包括抗体、嵌合抗原受体或功能性蛋白。
11.如权利要求10所述应用,所述抗体、嵌合抗原受体靶向以下任意一种或多种位点:BCMA、CD19、CD20、CD123、CD22、CD3D、CD3E、CD7CLEC12AGPRC5D、CD138、CD30、CD33、CD38、CD3E、CD79BSLAMF7、CD10、CD117、CD37、CD4、CD5、CD56、CD72、CD79A、CD99、Flt-3、LILRA3、LILRB4、SLAMF3、Her2、MSLN、B7-H3、CLDN18、EGFR、GPC3、KRAS、CA9、CEA、EGFRvIII、EphA2、ERBB3、ERBB4、FAP、GUCY2C、IL13RA2、MUC1、PD-1、PSMA、VEGFR2、AFP、AXL、CD133、CD147、CD171、CD80、CD86、c-Met、DLL4、EpCAM、Nectin-4、Podoplanin、ROBO1、ROR2、SSTR2、FOLR1、ROR1、CD70、NKG2D、PD-L1、SIRP alpha。
