Detailed Description
In order that the invention may be more readily understood, a further description of the invention will be rendered by reference to specific embodiments that are intended to be illustrative rather than limiting.
A herba Houttuyniae green tea composition with anticariogenic and antibacterial effects is prepared from herba Houttuyniae extract and green tea extract, or herba Houttuyniae extract and tea polyphenols by mixing and applying to preparation of anticariogenic and antibacterial products.
The bacteria used in the invention are all international standard strains, streptococcus mutans (Streptcoccus mutans, ATCC 25175), and the typical culture medium used for the bacteria is brain heart leaching broth (BHI for short). The preparation method of the BHI liquid culture medium comprises the following steps: dissolving 18.5g of BHI powder (Solarbio, LA 036) with 500mL of triple distilled water, stirring for dissolving, sterilizing at high temperature and high pressure (121.3 ℃ C., 103.4 kPa) for 30min, cooling, and sealing for later use. The preparation method of the BHI solid culture medium comprises the following steps: 500mL of triple distilled water was used to dissolve 18.5g of BHI powder (Solarbio, LA 036) and 7.5g of agar powder (Solarbio, A8190), the solution was stirred and sterilized at high temperature and high pressure (121.3 ℃ C., 103.4 kPa) for 30min, and after cooling slightly, the mixture was poured into a microbial culture dish and cooled for use. The culture conditions of the streptococcus mutans are as follows: constant temperature of 37 ℃,85% N 2 、5%CO 2 And 10% H 2 Anaerobic conditions of the mixed gas, and taking the streptococcus mutans suspension cultured to the logarithmic phase for all antibacterial experiment investigation.
Example 1:
the effect of the three extracts of houttuynia cordata, green tea and tea polyphenol on the growth of streptococcus mutans under the independent action is examined to determine the inhibition activity of different extracts on the streptococcus mutans.
100. Mu.L of final concentration 10 was added to each well of a 96-well plate 5 ~10 6 Bacterial suspensions of CFU/mL, sterile houttuynia cordata extract, green tea extract and tea polyphenols dissolved in BHI broth at different concentrations, and negative (BHI broth) and positive (0.06% chlorhexidine gluconate) controls were added and incubated for 24h under anaerobic conditions. After each hole is diluted according to a certain proportion, 5 mu L of the mixture is dripped on a BHI solid culture medium, and the mixture is cultured for 24 hours under anaerobic condition, and compared with a negative control group and a positive control group, the inhibition effect of different extracts on streptococcus mutans when the extracts act independently is observed. As shown in FIG. 1 and Table 1, A, B and C in FIG. 1 are respectively the effects of 10-50 mg/mL houttuynia cordata extract, 10-50 mg/mL green tea extract and 2.5-30 mg/mL tea polyphenol on the growth of Streptococcus mutans under the separate action, and by combining the records of the growth conditions of Streptococcus mutans in Table 1, 20mg/mL houttuynia cordata extract, 40mg/mL green tea extract and 5mg/mL tea polyphenol can obviously inhibit the growth of Streptococcus mutans. The 20mg/mL houttuynia cordata extract, 40mg/mL green tea extract and 5mg/mL tea polyphenol are determined to be the minimum inhibitory concentration, so that the subsequent scientific and reasonable preparation of the composition is facilitated.

Example 2:
investigation of mass ratio 1:1 (0.5, 1, 2, 3, 4, 5 mg/mL) and green tea extract (0.5, 1, 2, 3, 4, 5 mg/mL), and the inhibition effect of different compositions of houttuynia cordata extract (0.25, 0.5, 1, 2, 4, 5 mg/mL) and tea polyphenol (0.25, 0.5, 1, 2, 4, 5 mg/mL) in mass ratio of 1:1.
As shown in FIG. 2 and Table 2, A, B of FIG. 2 is the 1:1 composition of the herba Houttuyniae extract and the green tea extract, and the effect of 0.25-5 mg/mL of the 1:1 composition of the herba Houttuyniae extract and the tea polyphenol on the growth of Streptococcus mutans, and by combining the records of the growth of Streptococcus mutans in Table 2, it is seen that the 1:1 composition of 4mg/mL of the herba Houttuyniae extract and 4mg/mL of the green tea extract and the 1:1 composition of 4mg/mL of the herba Houttuyniae extract and 4mg/mL of the tea polyphenol can significantly inhibit the growth of Streptococcus mutans, and the inhibition rates are all close to 100%. The composition of houttuynia cordata-green tea extract in a mass ratio of 1:1 is in the range of 4 to 5mg/mL and the composition of houttuynia cordata-tea polyphenol in a mass ratio of 1:1 is in the range of 4 to 5mg/mL, compared with the independent action of the different extracts in example 1, has more remarkable mutans streptococcus inhibitory activity.

Example 3:
the inhibition effect of different compositions of houttuynia cordata extract (0.6, 1.2, 2.5, 5, 10, 20 mg/mL) and green tea extract (0.6, 1.2, 2.5, 5, 10, 20 mg/mL) in mass ratio of 1:1 on streptococcus mutans was examined. As shown in FIG. 3A and Table 3, it was found that 1:1 combination of 2.5mg/mL houttuynia cordata extract and 2.5mg/mL green tea extract significantly inhibited Streptococcus mutans growth with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-green tea extract composition with the mass ratio of 1:1 can obviously inhibit streptococcus mutans in the range of 2.5-20 mg/mL.

Example 4:
the inhibition effect of different compositions of houttuynia cordata extract (0.3, 0.6, 1.2, 2.5, 5, 10, 20 mg/mL) and green tea extract (0.6, 1.2, 2.5, 5, 10, 20, 40 mg/mL) in mass ratio of 1:2 on mutans streptococcus was examined. As shown in FIG. 3B and Table 3, it was found that 1:2 combination of 5mg/mL houttuynia cordata extract and 10mg/mL green tea extract significantly inhibited S.mutans growth with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-green tea extract composition with the mass ratio of 1:2 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 5-20 mg/mL and the green tea extract is 10-40 mg/mL.
Example 5:
the inhibition effect of different compositions of houttuynia cordata extract (0.6, 1.2, 2.5, 5, 10, 20 mg/mL) and green tea extract (0.3, 0.6, 1.2, 2.5, 5, 10 mg/mL) in mass ratio of 1:0.5 on Streptococcus mutans was examined. As shown in FIG. 3C and Table 3, it was found that 1:0.5 composition of 5mg/mL houttuynia cordata extract and 2.5mg/mL green tea extract significantly inhibited Streptococcus mutans growth with an inhibition rate approaching 100%. Therefore, the composition of the houttuynia cordata-green tea extract with the mass ratio of 1:0.5 can obviously inhibit the streptococcus mutans when the houttuynia cordata extract is 5-20 mg/mL and the green tea extract is 2.5-10 mg/mL.
Example 6:
the inhibition effect of different compositions of houttuynia cordata extracts (0.12, 0.25, 0.5, 1, 2, 3 mg/mL) and tea polyphenol (0.12, 0.25, 0.5, 1, 2, 3 mg/mL) in a mass ratio of 1:1 on streptococcus mutans was examined. As shown in FIG. 4A and Table 4, it was found that 1:1 combination of 3mg/mL houttuynia cordata extract and 3mg/mL tea polyphenol significantly inhibited Streptococcus mutans growth with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:1 can remarkably inhibit streptococcus mutans when the houttuynia cordata extract is 3mg/mL and the tea polyphenol is 3 mg/mL.

Example 7:
the inhibition effect of different compositions of houttuynia cordata extract (0.06, 0.12, 0.25, 0.5, 1, 1.5 mg/mL) and tea polyphenol (0.12, 0.25, 0.5, 1, 2, 3 mg/mL) in mass ratio of 1:2 on mutans streptococcus was examined. The results are shown in FIG. 4B and Table 4, and can be seen at 1mg/mL houttuynia cordata extract with 1 of tea polyphenols at 2 mg/mL: the composition 2 can obviously inhibit the growth of the streptococcus mutans, and the inhibition rate is close to 100%. Therefore, the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:2 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 1-1.5 mg/mL and the tea polyphenol is 2-3 mg/mL.
Example 8:
the inhibition effect of different compositions of cordate houttuynia extract (0.25, 0.5, 1, 2, 4, 6 mg/mL) and tea polyphenol (0.12, 0.25, 0.5, 1, 2, 3 mg/mL) with the mass ratio of 1:0.5 on streptococcus mutans was examined. As shown in FIG. 4C and Table 4, it was found that 1:0.5 composition of 4mg/mL houttuynia cordata extract and 2mg/mL tea polyphenol significantly inhibited the growth of Streptococcus mutans with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:0.5 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 4-6 mg/mL and the tea polyphenol is 2-3 mg/mL.
Example 9:
preparing herba Houttuyniae-green tea extract composition and herba Houttuyniae-tea polyphenols composition at mass ratio of 1:1, 1:2 and 1:0.5. Wherein, the houttuynia cordata extract concentration in the houttuynia cordata-green tea extract composition is 2-20 mg/mL, and the green tea extract concentration is 2-20 mg/mL in the houttuynia cordata-green tea extract composition with the mass ratio of 1:1; the houttuynia cordata extract concentration in the houttuynia cordata-green tea extract composition is 3-20 mg/mL, and the green tea extract concentration is 6-40 mg/mL in a mass ratio of 1:2; the houttuynia cordata extract concentration in the houttuynia cordata-green tea extract composition is 4-20 mg/mL, and the green tea extract concentration is 2-10 mg/mL in the mass ratio of 1:0.5; the concentration of the houttuynia cordata extract in the houttuynia cordata-tea polyphenol composition is 2.5-10 mg/mL, and the concentration of the tea polyphenol is 2.5-10 mg/mL in the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:1; the concentration of the houttuynia cordata extract in the houttuynia cordata-tea polyphenol composition is 1-10 mg/mL, and the concentration of the tea polyphenol is 2-20 mg/mL in the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:2; the houttuynia cordata extract concentration in the houttuynia cordata-tea polyphenol composition is 3-12 mg/mL, and the tea polyphenol concentration is 1.5-6 mg/mL in the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:0.5. Observing the dissolution and color of solutions of different compositions can facilitate selection of suitable compositions based on the location of the product, e.g., lower concentration compositions may be considered if cost is a priority.
Example 10:
investigation of mass ratio 1:1 (2, 3, 4, 5, 6, 7, 8, 9, 10 mg/mL) and green tea extract (2, 3, 4, 5, 6, 7, 8, 9, 10 mg/mL). As shown in FIG. 6A and Table 5, it was found that 1:1 combination of 2mg/mL houttuynia cordata extract and 2mg/mL green tea extract significantly inhibited S.mutans growth with an inhibition rate approaching 100%. Therefore, the composition of the houttuynia cordata-green tea extract with the mass ratio of 1:1 can obviously inhibit the streptococcus mutans in the range of 2-10 mg/mL.

Example 11:
the inhibition effect of different compositions of houttuynia cordata extract (3, 4, 5, 6, 7, 8, 9, 10 mg/mL) and green tea extract (6, 8, 10, 12, 14, 16, 18, 20 mg/mL) in mass ratio of 1:2 on streptococcus mutans was examined. As shown in FIG. 6B and Table 5, it was found that 1:2 combination of 3mg/mL houttuynia cordata extract and 6mg/mL green tea extract significantly inhibited S.mutans growth with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-green tea extract composition with the mass ratio of 1:2 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 3-10 mg/mL and the green tea extract is 6-20 mg/mL.
Example 12:
the inhibition effect of different compositions of houttuynia cordata extract (4, 5, 6, 7, 8, 9, 10, 20 mg/mL) and green tea extract (2, 2.5, 3, 3.5, 4, 4.5, 5, 10 mg/mL) in mass ratio of 1:0.5 on mutans streptococcus was examined. As shown in FIG. 6C and Table 5, it was found that 1:0.5 composition of 4mg/mL houttuynia cordata extract and 2mg/mL green tea extract significantly inhibited S.mutans growth with an inhibition rate approaching 100%. Therefore, the composition of the houttuynia cordata-green tea extract with the mass ratio of 1:0.5 can obviously inhibit the streptococcus mutans when the houttuynia cordata extract is 4-20 mg/mL and the green tea extract is 2-10 mg/mL.
Example 13:
the inhibition effect of different compositions of houttuynia cordata extract (2.5, 3, 4, 5, 6, 7, 8, 9, 10 mg/mL) and tea polyphenol (2.5, 3, 4, 5, 6, 7, 8, 9, 10 mg/mL) in mass ratio of 1:1 on mutans streptococcus was examined. As shown in FIG. 7A and Table 6, it was found that 1:1 combination of 2.5mg/mL houttuynia cordata extract and 2.5mg/mL tea polyphenol significantly inhibited Streptococcus mutans growth with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:1 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 2.5-10 mg/mL and the tea polyphenol is 2.5-10 mg/mL.

Example 14:
the inhibition effect of different compositions of houttuynia cordata extracts (1, 1.5, 2, 3, 4, 5, 6, 7, 8 mg/mL) and tea polyphenols (2, 3, 4, 6, 8, 10, 12, 14, 16 mg/mL) in mass ratio of 1:2 on mutans streptococcus was examined. As shown in FIG. 7B and Table 6, it was found that 1:2 combination of 1mg/mL houttuynia cordata extract and 2mg/mL tea polyphenol significantly inhibited S.mutans growth with an inhibition rate approaching 100%. Therefore, the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:2 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 1-8 mg/mL and the tea polyphenol is 2-16 mg/mL.
Example 15:
the inhibition effect of different compositions of houttuynia cordata extracts (3, 4, 5, 6, 7, 8, 9, 10, 12 mg/mL) and tea polyphenols (1.5, 2, 2.5, 3, 3.5, 4, 4.5, 5, 6 mg/mL) in mass ratio of 1:0.5 on mutans streptococcus was examined. As shown in FIG. 7C and Table 6, it was found that 1:0.5 composition of 3mg/mL houttuynia cordata extract and 1.5mg/mL tea polyphenol significantly inhibited the growth of Streptococcus mutans with an inhibition rate of approximately 100%. Therefore, the houttuynia cordata-tea polyphenol composition with the mass ratio of 1:0.5 can obviously inhibit streptococcus mutans when the houttuynia cordata extract is 3-12 mg/mL and the tea polyphenol is 1.5-6 mg/mL.
Based on all the above examples, it is finally preferred that:
the houttuynia cordata green tea composition with caries preventing and bacteriostasis activity comprises a composition of houttuynia cordata extract and green tea extract in the mass ratio of 1:0.5-2 or a composition of houttuynia cordata extract and tea polyphenol in the mass ratio of 1:0.5-2.
More preferably, when the cordate houttuynia extract and the green tea extract are mixed according to the mass ratio of 1:1, the preferred concentrations are: houttuynia cordata extract: 2. 2.5, 3, 4, 5, 6, 7, 8, 9, 10, 15mg/mL; green tea extract: 2. 2.5, 3, 4, 5, 6, 7, 8, 9, 10, 15mg/mL; when the cordate houttuynia extract and the green tea extract are mixed according to the mass ratio of 1:2, the preferred concentrations are: houttuynia cordata extract: 3.4, 5, 6, 7, 8, 9, 10mg/mL; green tea extract: 6. 8, 10, 12, 14, 16, 18, 20mg/mL; when the cordate houttuynia extract and the green tea extract are combined according to the mass ratio of 1:0.5, the preferable concentration is as follows: houttuynia cordata extract: 4.5, 6, 7, 8, 9, 10, 20mg/mL; green tea extract: 2. 2.5, 3, 3.5, 4, 4.5, 5, 10mg/mL; when the cordate houttuynia extract and tea polyphenol are mixed according to the mass ratio of 1:1, the preferred concentrations are: houttuynia cordata extract: 2.5, 3, 4, 5, 6, 7, 8, 9, 10mg/mL; green tea extract: 2.5, 3, 4, 5, 6, 7, 8, 9, 10mg/mL; when the cordate houttuynia extract and tea polyphenol are combined according to the mass ratio of 1:2, the preferable concentration is as follows: houttuynia cordata extract: 1. 1.5, 2, 3, 4, 5, 6, 7, 8, 10mg/mL; green tea extract: 2.5, 3, 4, 5, 6, 7, 8, 9, 10, 20mg/mL; when the cordate houttuynia extract and tea polyphenol are combined according to the mass ratio of 1:0.5, the preferable concentration is as follows: houttuynia cordata extract: 3.4, 5, 6, 7, 8, 10, 12mg/mL; green tea extract: 1.5, 2, 2.5, 3, 3.5, 4, 4.5, 5, 6mg/mL.
Meanwhile, the quality requirements of the cordate houttuynia extract, the green tea extract and the tea polyphenol are standardized, and the scientificity of the composition formula composition and the efficacy evaluation is greatly improved. The composition has the advantages of homology of medicine and food, natural and harmless property, single raw material, low price, economy and safety, has the characteristic of obviously inhibiting main pathogenic bacteria of caries-streptococcus mutans, can be applied to research and development of caries prevention and bacteria inhibition related pharmaceutical preparations, oral cavity washing or nursing and other products, and is particularly suitable for meeting urgent requirements of effective, safe and long-term use in aspects of oral cavity health care and nursing.
The quality requirement of the adopted houttuynia cordata extract is 10:1 proportion of extract, brown powder, 80 meshes of particles, no more than 5 percent of drying weight loss, no more than 7 percent of ash, no more than 20ppm of heavy metal and no more than 2ppm of pesticide residue; the quality requirements of the adopted green tea extract are 10:1 proportion of extract, yellow brown powder, granularity of 80 meshes, loss on drying of no more than 5%, ash content of no more than 7%, heavy metal of no more than 20ppm and pesticide residue of no more than 2ppm; the quality of the adopted tea polyphenol is required to be yellow-brown powder, the tea polyphenol has peculiar smell, the polyphenol content is not less than 98.0%, the total catechin of the characteristic components is not less than 80.0%, the epigallocatechin gallate is not less than 40.0%, the granularity is 80 meshes, the moisture is not more than 5%, the heavy metal is not more than 10ppm, the lead is not more than 2.0ppm, and the arsenic is not more than 1.0ppm. This is the quality requirement of the relevant component.
The houttuynia cordata extract, the green tea extract and the tea polyphenol can be extracted from houttuynia cordata and green tea by a solvent extraction method, an ultrasonic extraction method, a supercritical extraction method or other methods, so long as the quality requirements can be met.
The above embodiments are merely preferred embodiments of the present invention, which are used to illustrate the technical solution of the present invention, but not to limit it in any way. All other embodiments obtained by those skilled in the art without creative efforts or modifications or substitutions made by the technical spirit of the invention, such as modifications to the technical solutions described in the foregoing embodiments, equivalent substitutions of some technical features thereof, etc., still belong to the protection scope of the technical solutions of the present invention.

Claims (10)

1. The houttuynia cordata green tea composition with caries preventing and antibacterial activity is characterized by comprising an A component and a B component, wherein 1-20 mg/mL of the A component is combined with the B component according to the mass ratio of 1:0.5-2, the A component is houttuynia cordata extract, and the B component is green tea related extract and comprises one or more of green tea extract and tea polyphenol.
2. 3. The green tea composition of claim 1, wherein the concentrations of the extract of houttuynia cordata and the extract of green tea are 2-20 mg/mL, respectively, when the extract of houttuynia cordata and the extract of green tea are combined according to a mass ratio of 1:1.
4. The green tea composition of claim 1, wherein the concentrations of the houttuynia cordata extract and the tea polyphenol are respectively 2.5-10 mg/mL when the houttuynia cordata extract and the tea polyphenol are combined according to a mass ratio of 1:1.
5. Use of the houttuynia cordata green tea composition according to any one of claims 1-4 for preparing anticarious antibacterial drugs.
6. Use of a houttuynia cordata green tea composition as claimed in any one of claims 1-4 for the preparation of anticarious antibacterial oral wash or care products.
7. The use according to claim 5, wherein the houttuynia cordata green tea composition is formulated as a pharmaceutical preparation with pharmaceutically acceptable excipients as an active ingredient.
8. The use according to claim 6, wherein the houttuynia cordata green tea composition is formulated into an oral wash or care product as an active ingredient together with an auxiliary material acceptable on daily chemical products and on disinfection products.
9. The use according to claim 7, wherein the pharmaceutical formulation is a solution, spray, gel, lozenge or injection.
10. The use according to claim 8, wherein the oral wash or care product is a mouthwash, toothpaste, tooth powder, tooth gel, chewing gum or an oral spray.
