Abstract

本发明提供一种人ACE2改造蛋白嵌入的广谱新冠抗体,以新冠抗体作为骨架,稳定性好;以人ACE2来对抗新型冠状病毒变异株,具有广谱性及特异性好等优点。由人ACE2改造蛋白嵌入的抗体可以与以人ACE2作为受体的新冠病毒突变体进行结合,作为广谱的抗体中和持续突变的突变株,弥补单克隆抗体无法中和免疫逃逸的突变株,极具有应用价值。

一种人ACE2改造蛋白嵌入的新型广谱新冠抗体及其应用
技术领域
本发明属于生物技术及免疫技术领域,具体涉及人ACE2蛋白嵌入的广谱新冠抗体及其应用。
背景技术
新型冠状病毒(SARS-CoV-2)造成新冠肺炎(2019-nCoV)的全世界大流行,治疗性的抗体、疫苗、药物以前所未有的速度被研发以抵抗病毒的传播。冠状病毒,如,SARS病毒、新型冠状病毒,通过病毒表面的刺突蛋白(Spike ,S蛋白)结合宿主细胞受体血管紧张素转换酶2(ACE2)分子感染宿主。刺突蛋白由S1和S2两个亚基组成,分别负责宿主细胞受体的识别和入侵时的膜融合。并且,S1亚基的C端(CTD),也称为受体结合区域(RBD),是识别ACE2受体关键的区域。因此,刺突蛋白,特别是RBD区域,对于病毒的侵染非常重要。然而,病毒的流行传播和治疗增加了RBD区域突变的概率。RDB区域的突变可能使病毒传播能力增强,免疫逃逸,造成重复感染风险提高、疫苗和现有治疗药物有效性降低。因此,针对越来越多,形形色色的突变株。迫切需要研发具有广谱作用,或者针对突变株依然有保护性/有效性的治疗性抗体和疫苗。
世界卫生组织(WHO)针对新型冠状病毒的特征选出了五株令人担忧的突变株(variant of concern, VOC),分别是:Alpha株、Beta株、Gamma株、Delta株、Omicron株。其中,Alpha株,特别是Delta株以惊人的速度在全球扩散,成为2021年的主流毒株。而Beta株,则是已知突变中(除新发现的Omicron株)免疫逃逸程度最高的。这些突变株都造成了临床治疗的单克隆抗体和疫苗有效性不同程度的降低。目前新发现的Omicron株,刺突蛋白一共有32个氨基酸突变,已有的单克隆抗体及疫苗很多都无效,令人担忧。值得一提的是,尽管VOC发生了不同程度的免疫逃逸,它们依然利用人ACE2作为受体,而且有些VOC的S蛋白对ACE2的亲和力变得更强[Han P, Li L, Liu S, et al. Receptor binding and complexstructures of human ACE2 to spike RBD from omicron and delta SARS-CoV-2.Cell, 2022]。这提示通过改造ACE2可以实现“以不变应万变”,规避抗原突变导致的抗体逃逸。
发明内容
为了解决上述问题,本发明设计人ACE2蛋白或改造蛋白嵌入的广谱新冠抗体设计,用人ACE2改造蛋白嵌入的抗体作为竞争者,与宿主细胞ACE2受体竞争结合新型冠状病毒突变株,起到保护作用。
本发明人基于在疟疾研究过程中发现,在疟疾病人体内分离到可以和多种恶性疟原虫株结合的共有抗体,经过序列分析表明这些抗体中插入有人的免疫抑制性受体 LAIR1的胞外段序列,插入方式有两种,一种是VDJ插入还有一种是 switch 区域的插入。这些共有抗体可以通过插入的 LAIR1 片段结合被感染红细胞表面的疟原虫蛋白RIFIN,进而发挥抗疟疾功能。这提示我们,人类的免疫系统在进化中,存在可以在抗体V区插入自身编码基因片段的机制,而这种天然存在的插入机制。对此,本发明人根据疟疾患者体内的两种不同形式的共有抗体,尝试改造新冠抗体。CB6单抗是一款靶向新冠肺炎病毒S蛋白RBD的抗体,由微生物研究所高福院士团队和严景华团队联合研发(Rui Shi等,A humanneutralizingantibody targets the receptor binding site of SARS-CoV-2.Nature,2020年5月26日),目前已在美国、欧盟、印度等国家获得紧急使用授权。但有研究报道很多VOC突变株对CB6已产生逃逸。
本发明人把CB6单克隆抗体为例作为骨架,通过两种插入方式把hACE2蛋白嵌入CB6抗体中。第一种,hACE2插入在抗体重链可变区中互补决定区(complementaritydetermining region,CDR)的CDR3,因为互补决定区是抗原结合部位(antigen-bindingsite),hACE2的插入破坏了抗原结合区,使抗体骨架失去结合抗原的能力;第二种,hACE2插入在抗体重链可变区和恒定区之间的switch μ区域,因为没有破坏抗体的抗原结合部位,保留了抗体骨架与抗原的识别能力,而成为双特异性抗体。
因此,本发明的目的在于提供一种人ACE2改造蛋白嵌入的广谱新冠抗体,以新冠抗体作为骨架,稳定性好;以人ACE2来对抗新型冠状病毒变异株,具有广谱性及特异性好等优点。可提供一种新的双特异性抗体构建方法,并用于新型冠状病毒感染者的治疗。
所用新冠抗体骨架可以是靶向新型冠状病毒S蛋白不同抗原位点的任意单抗,而嵌入的ACE2本身可以对抗新型冠状病毒不同变异株,此新型双特异性抗体,可以最大程度提升母源单抗的选择性和中和活性。
进一步地,本发明使用不同突变改造的hACE2蛋白,hACE2-T27F,hACE2-T27F-R273Q。hACE2-T27F,是hACE2蛋白中的氨基酸27T突变成苯丙氨酸(以下简称T27F),以增强彼此之间疏水作用力从而增强彼此之间的亲和力;hACE2-T27F-R273Q,是hACE2蛋白中的氨基酸273R突变成谷氨酰胺 (以下简称R273Q),使改造蛋白仍具有结合新冠病毒的能力,但不再具有降血压等,酶活作用的能力。其次,还设计了全长的hACE2,以及截取hACE2的肽酶结构域(以下简称hACE2-PD)。为此,共设计了13个构建体,分别为:Switch-ACE2、Switch-ACE2-T27F、Switch-ACE2-T27F-R273Q、Switch-ACE2-PD-T27F-R273Q、Switch-L-ACE2-PD-T27F-R273Q、VDJ-ACE2、VDJ-ACE2-T27F、VDJ-ACE2-T27F-R273Q、VDJ-GS-ACE2、VDJ-GS-ACE2-T27F、VDJ-GS-ACE2-T27F-R273Q、VDJ-GS-ACE2-PD-T27F-R273Q、VDJ-ACE2-PD-T27F-R273Q。其中,8个构建体可表达,并挑选Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q进行进一步的功能研究,验证了其效果。
因此,本发明提从人ACE2蛋白嵌入的广谱新冠抗体,其特征在于,以新冠抗体作为骨架,将人ACE2蛋白插入在新冠抗体重链可变区中互补决定区的CDR3,或者插入在抗体重链可变区和恒定区之间的switch μ区域。
优选实施方式,所述新冠抗体是单抗,更优选为以CB6单克隆抗体作为骨架。更具体地,所述单克隆抗体CB6抗体重链的氨基酸序列如SEQ ID NO:2所示。
优选实施方式中,所述人ACE2蛋白是全长野生型人ACE2蛋白、野生型人ACE2蛋白的肽酶结构域,以及氨基酸替换的人ACE2改造蛋白,优选地,所述人ACE2改造蛋白相对于野生型人ACE2蛋白而言,存在T27F突变,或T27F和R273Q突变。更具体地,所述野生型hACE2蛋白的氨基酸序列如SEQ ID NO: 6所示;所述人ACE2改造蛋白的氨基酸序列如SEQ ID NO:8所示,或如SEQ ID NO:10所示;野生型人ACE2蛋白的肽酶结构域的氨基酸序列如SEQ IDNO:6中第1-597位。
优选地,以无间隔地连接的方式将所述人ACE2蛋白插入单克隆抗体CB6抗体重链的第139-140位氨基酸位点;更具体地,其氨基酸序列如SEQ ID NO:12、14、16、18;或者以L柔性铰链区为连接(优选地序列为:LDPGLQPGNFSAD;),再将人ACE2蛋白插入单克隆抗体CB6抗体重链的第139-140位氨基酸位点;更具体地,其氨基酸序列如SEQ ID NO:20所示。
在另一个优选实施方式中,以无间隔地连接的方式将所述人ACE2蛋白插入单克隆抗体CB6抗体重链的第125-126位氨基酸,更具体地,其氨基酸序列如SEQ ID NO:22、24、26、36;或者以GS柔性铰链区(优选地序列为:GGGSGS)为连接,再将人ACE2蛋白插入再插入单克隆抗体CB6抗体重链的第125-126位氨基酸,更具体地,其氨基酸序列如SEQ ID NO:28、30、32、34。
优选地,轻链采用CB6的抗体的轻链,具体地其氨基酸序列如SEQ ID NO:4所示。
本发明还提供所述广谱新冠抗体的编码基因,优选地,所述人ACE2蛋白的编码核苷酸序列如SEQ ID NO:5、7、9所示;所述单克隆抗体CB6抗体重链的编码核苷酸序列如SEQID NO:1所示;
优选地,所述的广谱新冠抗体的重链编码核苷酸序列如SEQ ID NO:11、13、15、17、19、21、23、25、27、29、31、33、35;
另外优选地,所述的广谱新冠抗体的轻链编码核苷酸序列如SEQ ID NO:3所示。
本发明进一步提供含有所述广谱新冠抗体的编码基因的载体,优选地,将抗体的重链、轻链编码基因分别克隆到真核细胞表达载体上,得到不同的抗体构建载体。
本发明还提供所述广谱新冠抗体在制备预防或治疗新冠病毒或SARS病毒引起的疾病的药物中的应用。具体地,预防或治疗由新型冠状病毒野生株(Prototype)、阿尔法突变株(Alpha),贝塔突变株(Beta)、伽马突变株(Gamma)、德尔塔突变株(Delta)、奥密克戎突变株(Omicron)、SARS病毒(SARS-CoV)引起的疾病。
本发明提供的hACE2改造蛋白嵌入的抗体可以与以hACE2作为受体的新冠病毒突变体进行结合,作为广谱的抗体中和持续突变的突变株,弥补单克隆抗体无法中和免疫逃逸的突变株,以“不变应万变”。例如,研究表明Switch-ACE2-T27F-R273Q对VOCs毒株拥有很高的中和活性。

下文将结合具体实施例对本发明的技术方案做更进一步的详细说明。应当理解,下列实施例仅为示例性地说明和解释本发明,而不应被解释为对本发明保护范围的限制。凡基于本发明上述内容所实现的技术均涵盖在本发明旨在保护的范围内。
除非另有说明,以下实施例中使用的原料和试剂均为市售商品,或者可以通过已知方法制备。下列实施例中未注明具体条件的实验方法,通常按照常规条件如Sambrook等人,分子克隆:实验室手册(New York: Cold Spring Harbor Laboratory Press, 第四版)中所述的条件,或按照制造厂商所建议的条件。
实施例1:抗体构建设计
为确保设计抗体可以进行可溶表达并且具有功能活性,本实施例在抗体设计时,设计了大量不同的抗体构建,以期获得具有较好功能活性的可溶表达构建。
本发明人设计了全长的hACE2,以及截短体hACE2-PD(截取hACE2从第1-597位氨基酸为肽酶结构域,以下简称hACE2-PD,氨基酸序列如SEQ ID NO:38所示)。并使用野生型hACE2蛋白,以及氨基酸替换的hACE2改造蛋白,hACE2-T27F,hACE2-T27F-R273Q。单克隆抗体CB6作为骨架,通过两种插入方式把hACE2蛋白嵌入CB6抗体中。

在这类插入模式中,我们将野生型hACE2蛋白(编码核苷酸序列和氨基酸序列分别如SEQ ID NO:5、6所示)和hACE2-T27F蛋白(编码核苷酸序列和氨基酸序列分别如SEQ IDNO:7、8所示),hACE2-T27F-R273Q蛋白(编码核苷酸序列和氨基酸序列分别如SEQ ID NO:9、10所示),hACE2-T27F-R273Q蛋白截取的肽酶结构域hACE2-PD-T27F-R273Q (序列如SEQ IDNO:37所示)的基因序列的基因序列,直接插入单克隆抗体CB6抗体重链(编码核苷酸序列和氨基酸序列分别如SEQ ID NO:1、2所示),可变区和恒定区之间switch μ区域(CB6抗体重链第139-140位氨基酸位点)的基因序列中,无间隔地连接,将得到的序列人工合成(由金斯瑞提供合成服务),得到Switch系列的构建抗体的重链基因序列,轻链采用CB6的抗体轻链基因序列(编码核苷酸序列和氨基酸序列分别如SEQ ID NO:3、4所示)。
将抗体的重链、轻链基因分别克隆到pCAGGS真核细胞表达载体上,得到不同的抗体构建载体。
重链序列为:

此外,还设计了hACE2-PD-T27F-R273Q,利以一小段氨基酸序列(氨基酸序列为LDPGLQPGNFSAD),命名为L柔性铰链区,作为连接hACE2蛋白与抗体之间的柔性铰链区,再插入至重链switch μ区域(抗体重链第139-140位氨基酸)的基因序列中。
重链序列为:

2.VDJ插入模式设计
在这类插入模式中,我们将野生型hACE2蛋白和hACE2-T27F,hACE2-T27F-R273Q,hACE2-PD-T27F-R273Q蛋白的基因序列,直接插入单克隆抗体CB6抗体重链(序列如1、2所示)的可变区的互补决定区CDR3中(在CB6抗体重链第125-126位氨基酸之间插入),无间隔地连接,将得到的序列人工合成(由金斯瑞提供合成服务),得到VDJ系列的构建抗体的重链基因序列。轻链采用CB6的抗体轻链基因序列(序列如SEQ ID NO:3、4所示)。
将抗体的重链、轻链基因分别克隆到pCAGGS真核细胞表达载体上,得到不同的抗体构建载体。
重链序列为:

此外,还设计了野生型hACE2蛋白和hACE2-T27F,hACE2-T27F-R273Q,hACE2-PD-T27F-R273Q蛋白,利用GS柔性铰链区(氨基酸序列为GGGSGS)作为hACE2蛋白与抗体之间的连接,再插入至CB6抗体重链的CDR3区域(CB6抗体重链第126位氨基酸位点)的基因序列中。
重链序列为:

所有构建基因示意图如图1,具体抗体卡通示意图具体组成和预测结合模式图如图2所示。
实施例2:抗体重组表达载体试表达
在含10%FBS的DMEM中培养HEK 293T细胞(ATCC CRL-3216),把上述不同抗体,分别编码抗体重链和轻链的重组表达载体,共转染至六孔板的HEK 293T细胞中。转染4-6小时后,将细胞培养液更换成无血清的DMEM,并且继续培养3天。收集上清,通过蛋白免疫印迹实验(Western Blot)检验目的蛋白是否可溶表达(使用羊抗人抗体孵育),表达结果如图3所示,结果汇总表如图1所示。
13个重组质粒构建体中,8个构建体可溶表达,其中VDJ-ACE2、VDJ-ACE2-T27F、VDJ-ACE2-T27F-R273Q、VDJ-GS-ACE2-PD-T27F-R273Q无法表达。根据实验目的不同,挑选抗体进行进一步的功能研究。
实施例3:抗体表达纯化
根据试表达的结果,选取7种构建进行表达纯化,分别是:Switch-ACE2、Switch-ACE2-T27F、Switch-ACE2-T27F-R273Q、Switch-ACE2-PD-T27F-R273Q、Switch-L-ACE2-PD-T27F-R273Q、VDJ-GS-ACE2、VDJ-GS-ACE2-T27F-R273Q。
将分别编码抗体重链和轻链的重组表达载体,通过阳离子聚合物类型转染试剂(SinoBiology)共转染到悬浮的293F细胞(Thermo)中,细胞密度约为2×106ml。转染后24h、72h添加补料液一次,第5天收集细胞上清。将收集的上清以6500rpm,4摄氏度环境下离心40min,随后用0.22μm滤膜进行过滤,除去细胞碎片。通过蠕动泵,将含有抗体蛋白的细胞上清在4摄氏度环境下装载至Protein A亲和层析柱中。用20mM磷酸钠缓冲液(PH 7.4)洗杂蛋白,直至缓冲液里不含蛋白,以除去非特异结合蛋白。用0.1M甘氨酸洗脱液(PH 2.0)洗脱目的蛋白,收集管里预先加入200ul 1M Tris-HCl(PH 9.0)缓冲液,防止蛋白在过酸的环境下失活,最后换液至PBS缓冲液(137 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 2 mM KH2PO4,PH 7.4)中。
将洗脱抗体产物用100KDa浓缩管进行浓缩,然后通过Superdex200分子筛层析法进行纯化。随后,通过SDS-PAGE(还原性)检测所纯化的目的蛋白结果如图4所示。图4的结果显示,经纯化获得的不同抗体构建蛋白,在还原条件下SDS-PAGE胶中,抗体重链和轻链间的二硫键被打开,从而显示为两个条带,且抗体纯度超过95%。
实施例4:Switch系列双特异性抗体的双位点结合能力评估
利用生物膜光干涉技术(BLI) Octet RED系统,选取Switch-ACE2,验证抗体的CB6、hACE2双位点结合抗原能力。以VDJ-ACE2、hACE2-hFc作为对照,VDJ系列构建因把hACE2蛋白插入至CDR3,破坏了CB6骨架的抗原结合区,使抗体骨架失去结合抗原的能力,只留下hACE2可以结合抗原RBD。Switch系列抗体因把hACE2蛋白插入至Switch μ区域,没有破坏抗体的抗原结合部位,保留了抗体骨架与抗原的识别能力,而成为双特异性抗体。
具体实验步骤如下:
a.首先,把三种蛋白样品(10-15μg/mL)分别固定至抗人 IgG 捕获 (AHC) 生物传感器中。
b.使用500mM 11B11(抗hACE2抗体的Fab片段)封闭抗体hACE2位点。封闭饱和之后,流动500mM 11B11和500mM 新型冠状病毒野生型RBD(Prototype RBD)。如果是双特异性抗体,如Switch- hACE2抗体,CB6抗体骨架的RBD结合位点可Prototype RBD,而VDJ-ACE2、hACE2-hFc则不能结合Prototype RBD。全程操作均在PBST(137 mM NaCl, 2.7 mM KCl, 10mM Na2HPO4, 2 mM KH2PO4 and 0.005% (v/v) Tween 20, pH 7.4)的缓冲液体系中。

实验结果如图5所示:Switch-ACE2作为双特异性抗体,在封闭hACE2位点饱和后,流动PrototypeRBD,结合曲线上升,证明有第二个位点结合RBD,双特异性抗体设计是可行的。相比之下,VDJ-ACE2、hACE2-hFc流动RBD时,曲线依然平直,没有上升,证明没有第二个位点结合RBD,作为Switch-ACE2的阴性对照。
实施例5:抗体与新冠病毒野生株/变异株和SARS病毒RBD结合能力的评估
利用表面等离子共振SPR技术,检测三种抗体(CB6、Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q),一种融合蛋白hACE2--T27F-R273Q-hFc与七种毒株新型冠状病毒野生株(Prototype)、阿尔法突变株(Alpha),贝塔突变株(Beta)、伽马突变株(Gamma)、德尔塔突变株(Delta)、奥密克戎突变株(Omicron)、SARS病毒(SARS-CoV)RBD的亲和力。
具体实验步骤如下:
a.将三种抗体,CB6、Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q,一种融合蛋白hACE2--T27F-R273Q-hFc固定在Protein A芯片的通道(flow cell ,Fc),Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q抗体固定量控制在约1 ,000响应值,CB6、hACE2-T27F-R273Q-hFc 固定量控制在大约400响应值(response units ,RU)。
b.实验缓冲液体系为PBST(137 mM NaCl, 2.7 mM KCl, 10 mM Na2HPO4, 2 mMKH2PO4,PH 7.4),并使用PBST连续倍比稀释RBD蛋白。然后,将连续稀释的RBD蛋白(CB6、hACE2-T27F-R273Q-hFc使用的稀释浓度梯度为3.125、6.25、12.5、25、50 nM,Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q使用的稀释浓度梯度为6.25、12.5、25、50、100 nM)依次通过各通道。
c .记录各抗体结合RBD蛋白的动力学曲线,并利用BIAevaluation software 8K(Biacore,Inc.)软件计算动力学常数。新型冠状病毒野生型、突变株和SARS病毒的RBD与改造蛋白及不同抗体的亲和力动力学曲线如图6,亲和力常数(KD)如下表所示。
结果如图6和上表可知:CB6不与SARS-CoV-2 Beta株、Gamma株、Omicron株、SARS-CoV株的RBD结合。相比之下,Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q弥补了这个缺失,可以与CB6无法结合的突变株结合。与Switch-ACE2-T27F-R273Q相比,VDJ-GS-ACE2-T27F-R273Q与所有RBD的KD值更高。Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q与突变株的KD值均比野生型数值高约2倍,而SARS-CoV毒株的KD值则与Prototype株接近。
实施例6:抗体针对新冠病毒野生株/变异株和SARS病毒假病毒体外中和活性实验
测定三种抗体CB6、Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q在Vero细胞中对七种毒株SARS-CoV-2 Prototype株、Alpha株、Beta株、Gamma株、Delta株、Omicron株、SARS-CoV株的中和效果。
设置了Vero细胞+培养基(阴性对照组),Vero细胞+假病毒稀释液+培养基(阳性对照组)。
抗体原液配制:分别将CB6、Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q抗体在无菌条件下溶解在10%FBS的DMEM培养基中,配制成浓度120μg/mL的溶液。
各抗体原液的梯度稀释液的配制:使用空的深孔板,用10%FBS的DMEM培养基,对各抗体原液依次进行10倍稀释。在起始梯度(即浓度最高)孔中,各抗体浓度均为120μg/ml(加入等体积病毒稀释液后最高浓度为60μg/ml)。对每种蛋白均稀释9个浓度,每个浓度设置3组平行,共3次重复实验。
病毒稀释液:将假病毒病毒液,用10%FBS的DMEM,依照测定病毒滴度稀释至适当的感染数。
具体实验步骤如下:
a.进行假毒中和实验的前一天,在96孔细胞培养板中铺对数生长期的Vero细胞,在培养基为10%FBS的DMEM培养基,5%二氧化碳、37摄氏度的培养箱中培养一晚。24h后细胞汇合密度达到90%以上。
b.取上述各抗体的梯度稀释液,在深孔板中加入等体积的假病毒稀释液,混匀后即为混合工作液,在37℃孵育1h后使用。
c.将96孔细胞培养板小心地弃去上清,用排枪分别缓慢,小心地加入上述混合工作液100μL/孔至Vero细胞孔中。同时,每板细胞设置阴性对照和阳性对照。在培养箱中培养15h。
d.使用CQ1仪器读取每孔荧光数值,通过GraphPad8.0计算半数抑制率IC50,曲线绘图如图7。数值汇总如下表。
结果表明:CB6抗体不能中和Beta株、Gamma株、Omicron株和SARS-CoV株,但是,Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q却能很好地中和所有新冠突变株和SARS病毒毒株。从半数抑制率IC50中和数值反映出,VDJ-GS-ACE2-T27F-R273Q的中和有效性低于Switch-ACE2-T27F-R273Q,IC50数值在150ng/mL以内,针对SARS-CoV-2 Prototype andSARS-CoV毒株降低为300-400 ng/mL。Switch-ACE2-T27F-R273Q能够非常有效地中和所有新冠突变株以及SARS病毒毒株,针对突变株IC50数值在10ng/mL以内,数值高于Prototype。并且针对SARS-CoV株,也拥有很好的中和效果。Switch-ACE2-T27F-R273Q、VDJ-GS-ACE2-T27F-R273Q弥补了突变株对筛选抗体的免疫逃逸,具有很好的广谱性。并且,随着病毒的突变,抗体对突变株产生越来越强或者接近中和Prototype株的中和活性。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。

Claims (12)

1.人ACE2蛋白嵌入的广谱新冠抗体,其特征在于,以新冠抗体作为骨架,将人ACE2蛋白插入在新冠抗体重链可变区中互补决定区的CDR3,或者插入在抗体重链可变区和恒定区之间的switch μ区域。
2.如权利要求1所述的广谱新冠抗体,其特征在于,所述新冠抗体是单抗,尤其是靶向新冠病毒S蛋白的多抗或单抗,例如CB6单克隆抗体。
3.如权利要求2所述的广谱新冠抗体,其特征在于,所述单克隆抗体CB6抗体重链的氨基酸序列如SEQ ID NO:2所示。
4.如权利要求1所述的广谱新冠抗体,其特征在于,所述人ACE2蛋白是全长野生型人ACE2蛋白、野生型人ACE2蛋白的肽酶结构域,以及氨基酸替换的人ACE2改造蛋白,优选地,所述人ACE2改造蛋白相对于野生型人ACE2蛋白而言,存在T27F突变,或T27F和R273Q突变。
5.如权利要求5所述的广谱新冠抗体,其特征在于,所述野生型hACE2蛋白的氨基酸序列如SEQ ID NO: 6所示;所述人ACE2改造蛋白的氨基酸序列如SEQ ID NO:8所示,或如SEQID NO:10所示;所述野生型人ACE2蛋白的肽酶结构域的氨基酸序列如SEQ ID NO:6中第1-597位。
6.如权利要求5所述的广谱新冠抗体,其特征在于,以无间隔地连接的方式将所述人ACE2蛋白插入单克隆抗体CB6抗体重链的第139-140位氨基酸位点;更具体地,其氨基酸序列如SEQ ID NO:12、14、16、18;
或者以L柔性铰链区为连接,再将人ACE2蛋白插入单克隆抗体CB6抗体重链的第139-140位氨基酸位点;优选地,L柔性铰链区序列为:LDPGLQPGNFSAD;更具体地,所述广谱新冠抗体的氨基酸序列如SEQ ID NO:20所示。
7.如权利要求5所述的广谱新冠抗体,其特征在于,以无间隔地连接的方式将所述人ACE2蛋白插入单克隆抗体CB6抗体重链的第125-126位氨基酸,更具体地,其氨基酸序列如SEQ ID NO:22、24、26、36;
或者以GS柔性铰链区为连接,再将人ACE2蛋白插入再插入单克隆抗体CB6抗体重链的第125-126位氨基酸;优选地,所述GS柔性铰链区的氨基酸序列为GGGSGS;更优选地,所述广谱新冠抗体的氨基酸序列如SEQ ID NO:28、30、32、34。
8.如权利要求1至7任一项所述的广谱新冠抗体,其特征在于,轻链采用CB6的抗体的轻链,具体地其氨基酸序列如SEQ ID NO:4所示。
9.如权利要求1-8任一项所述广谱新冠抗体的编码基因。
10.如权利要求9所述的广谱新冠抗体的编码基因,其特征在于,所述人ACE2蛋白的编码核苷酸序列如SEQ ID NO:5、7、9所示;所述单克隆抗体CB6抗体重链的编码核苷酸序列如SEQ ID NO:1所示;
优选地,所述的广谱新冠抗体的重链编码核苷酸序列如SEQ ID NO:11、13、15、17、19、21、23、25、27、29、31、33、35;
更优选地,所述的广谱新冠抗体的轻链编码核苷酸序列如SEQ ID NO:3所示。
11.含有如权利要求8至10任一项所述广谱新冠抗体的编码基因的载体,优选地,将抗体的重链、轻链编码基因分别克隆到真核细胞表达载体上,得到不同的抗体构建载体。
12.如权利要求1-8任一项所述广谱新冠抗体在制备预防或治疗新冠病毒或SARS病毒引起的疾病的药物中的应用,具体地,预防或治疗由新型冠状病毒野生株、阿尔法突变株,贝塔突变株、伽马突变株、德尔塔突变株、奥密克戎突变株、SARS病毒引起的疾病。
