Abstract
本发明提供了一种协同稳定的淀粉基皮克林乳液及其在医药、食品领域的应用,属于乳化技术领域、医药配制品、化妆品配制品和食品技术领域。本发明协同稳定的淀粉基皮克林乳液,经过植酸改性交联并干燥后得到改性淀粉颗粒,将淀粉颗粒糊化后的悬浮液、蛋白质分散液与油相混合并高速剪切乳化,得到植酸交联淀粉‑蛋白协同稳定的皮克林乳液。本发明的淀粉基皮克林乳液,其制备工艺简单、原料成本低且生物可降解;制备的可食用的非小分子表面活性剂稳定的皮克林乳液,制备方法简单,可广泛应用于药剂、化妆品以及食品领域。
Description
技术领域
本发明涉及一种协同稳定的淀粉基皮克林乳液及其在医药、食品领域的应用,属于乳化技术领域、医药配制品和化妆品配制品和食品技术领域。
背景技术
近年来,人们对使用食品级或食品相容颗粒或纳米颗粒来稳定皮克林(Pickering)乳液的兴趣日益浓厚。目前对于皮克林乳液的研究大多集中在环境友好型的固体颗粒(如几丁质、淀粉、脂肪、类黄酮、蛋白质和纤维素等)。比如,目前有文献报道了醇溶性蛋白质稳定的皮克林乳液的基本制备方法(i)颗粒制备后用油乳化;(ii)蛋白溶液与油混合后使用乳化剂。现有技术显示,不同蛋白的种类及浓度(2~10wt%,极少数<0.5wt%的蛋白质需要热聚集或其他修饰剂来稳定)显著影响着体系的长期稳定性,同时,蛋白基皮克林乳液在封装抗氧化活性分子方面有较广泛应用。Zhao等人也综述了其他水溶性蛋白基皮克林乳液,类似地,蛋白质浓度多为2wt%的情况下才能使得整个体系更加稳定。然而,如何在复杂体系(例如,在其它表面活性剂、高离子强度、食品成分存在下)提高蛋白基皮克林乳液稳定性仍有待确定。总之,从现有报道的蛋白质稳定的皮克林乳液来看,虽然蛋白质具有种类多、乳化性及生物相容性好等特点,但蛋白质需要提取的成本高,还存在对pH敏感、抗酶解效果差、致敏性等缺陷。因此,降低蛋白质稳定的皮克林乳液中蛋白质的用量,是亟待解决的问题。
与此同时,在众多天然生物源材料中,淀粉具有易获取、可再生、可降解、低致敏性及安全性高等特点,已成为生物基皮克林乳液颗粒乳化剂构建领域的研究热点。有报道显示,淀粉基皮克林乳液由于具备较厚的吸附层和较快的界面吸附而表现出更大的稳定性,将其用于功能因子的输送和包封可以有效改善活性成分的生物可及性,对姜黄素的包封率高达80%以上,并可将其靶向运送至大肠,因此选用淀粉作为载体材料具有很大优势。但是,天然淀粉不耐高温、水溶及乳化性差,在乳液体系方面的应用受到很大限制。目前,已经报道了辛烯基琥珀酸酐(OSA)与淀粉的酯化反应化学改性,增强了淀粉的两亲性、改善其润湿性能。然而,美国食品和药物管理局允许的最大OSA添加水平为3.0%,取代度(DS)小于0.02;此外,OSA在水中的溶解度低,在淀粉颗粒中分布不均匀,渗入颗粒内部受到阻碍,导致酯化反应仅限于淀粉表面。基于以上原因,OSA作用于淀粉改性的取代度及效果仍存在一定局限性,因此,需要寻找其他更安全的小分子物质在更大程度上对淀粉进行疏水化改性,使其能获得较大淀粉取代度的同时又确保体系的稳定性及润湿特性。
其次,许多淀粉基皮克林乳液的稳定也常需借用于各种小分子乳化剂来稳定油水界面,但小分子乳化剂携带的潜在异味可能会影响食品感官,除了HLB值稍高的非离子型,加入亲水基团可能还会对人体有一些损害;因此,它们在食品和制药行业的应用也有一定局限性。基于以上原因,急需取代小分子乳化剂的同时又能够显著提高淀粉基皮克林乳液稳定性的新方法。
综上,有必要开发能降低蛋白质稳定的皮克林乳液中蛋白质的用量、能取代小分子乳化剂的同时又能够显著提高稳定性的淀粉基皮克林乳液,这对扩大其在医药、化妆品、食品等领域的应用具有重要意义。
发明内容
[技术问题]
本发明主要是为了解决如下至少一个问题:(1)现有蛋白质稳定的皮克林乳液中蛋白质含量有待降低的问题;(2)现有淀粉基皮克林乳液制备方法存在复杂、成本高,甚至存在安全性方面的隐患的问题;(3)现有的一些淀粉基乳液体系存在离不开小分子乳化剂在油水界面发挥的稳定效果的问题。
为此,本发明提供了植酸改性淀粉及蛋白质复合胶体颗粒稳定的皮克林乳液。该皮克林乳液的制备,仅通过一步乳化即可,简单、便捷,耗时短、耗能低;在高效率制备的同时避免引入合成乳化剂或小分子表面活性剂,也极大地降低了皮克林乳液中所需的蛋白质含量。本发明的方法简单、绿色、无污染、低能耗,所得的皮克林乳液具有良好的营养素释放特性,可以适用于医药、化妆品、食品等领域。
[技术方案]
本发明提供了一种植酸改性淀粉及蛋白质复合胶体颗粒稳定的淀粉基皮克林乳液及其制备方法。本发明制备的皮克林乳液具有良好的稳定性,制备方法简单,利于实现工业化生产,扩大皮克林乳液在医药、化妆品及食品工业上的应用。
本发明淀粉基皮克林乳液,水相中含有经过植酸改性后淀粉颗粒、蛋白质,与油相混合后,通过高能乳化法制备。
本发明的淀粉基皮克林乳液,其制备方法包括如下:
(1)利用植酸改性淀粉:淀粉加入水中制备原淀粉分散液(即淀粉颗粒悬浮液),在原淀粉分散液中加入植酸进行反应;植酸的质量是淀粉干基质量的0.01~4wt%;一段时间后终止反应,得到植酸改性淀粉分散液;
(2)制备植酸改性淀粉颗粒:将上一步得到的分散液进行离心、洗涤,沉淀物烘干后粉碎,获得磷酸化的淀粉颗粒粉末,即植酸改性淀粉颗粒;
(3)制备淀粉基皮克林乳液:将步骤(2)得到的植酸改性淀粉颗粒和蛋白质分别分散于水中,并将植酸改性淀粉颗粒分散液进行糊化处理;水相为植酸改性淀粉颗粒分散液和蛋白质分散液的混合液,水相中植酸改性淀粉颗粒分散液和蛋白质分散液的体积比为10:9~10:0.1;将油相与水相混合,通过高能乳化法制备协同稳定的淀粉基皮克林乳液;其中,植酸改性淀粉颗粒分散液中的浓度为0.5~7wt%,蛋白质分散液的浓度为0.5~7wt%。
在一种实施方式中,所述步骤(1)中原淀粉分散液为20~60wt%。
在一种实施方式中,所述步骤(1)中,淀粉分散液的制备是将淀粉加入去离子水中,搅拌至充分溶解;可选地,淀粉和去离子水的质量体积比为20~60g:100mL;进一步地,用NaOH溶液将淀粉分散液的pH调至7~7.5;进一步可选地,将淀粉分散液置于恒温水浴锅内磁力搅拌加热,磁力搅拌速度为300~600rpm。
在一种实施方式中,所述步骤(1)中,植酸是通过逐滴加入的方式加入到淀粉分散液中。
在一种实施方式中,所述步骤(1)中,反应体系温度为40~55℃。
在一种实施方式中,所述步骤(1)中,反应的过程中控制体系的pH恒定在7~7.5之间。可选地,是通过1mol/L的NaOH溶液控制pH。
在一种实施方式中,所述步骤(1)中,反应的时间为1-9h。
在一种实施方式中,所述步骤(1)中,终止反应是通过调节pH值至6.5以终止。可选地,使用1mol/L的稀盐酸来调节pH。
在一种实施方式中,所述步骤(2)中,植酸改性淀粉分散液于3500rpm离心15min;可选地,用水和乙醇洗涤充分沉淀物,以去除杂质。
在一种实施方式中,所述步骤(2)中,沉淀物于40-55℃烘干12~24h并研磨粉碎。

在一种实施方式中,所述淀粉包括木薯淀粉、玉米淀粉、大米淀粉、豌豆淀粉、莲藕淀粉、马铃薯淀粉等不同晶型淀粉的一种或几种。
在一种实施方式中,所述蛋白质为乳清分离蛋白、乳铁蛋白、酪蛋白、血清蛋白、卵蛋白、鱼糜蛋白、明胶、胶原蛋白、大豆蛋白、花生蛋白中的一种或多种。
在一种实施方式中,所述油为玉米油、芝麻油、大豆油、花生油、菜籽油、橄榄油、中链甘油三酯、亚麻籽油、葵花籽油、棉籽油、瓜子油中的一种或多种。
本发明的第二个目的是提供包封有因子的本发明的淀粉基皮克林乳液,可以用于医药、化妆品及食品工业。
在一种实施方式中,所述应用,是在水相或者油相中添加对应的需要包封的因子,然后再通过高能乳化法制备乳液。
所述因子,可以是功能性成分或者营养成分及药品等。
在一种实施方式中,所述因子,可以是百里香酚(2-异丙基-5-甲基苯酚)、磷酸奥司他韦、布洛芬、阿霉素等用于生物医药领域。
在一种实施方式中,所述因子,可以是水溶性物质,添加在水相中,比如花青素、茶多酚、维生素C;也可以是脂溶性物质,添加在油相中,比如姜黄素、白藜芦醇、虾青素、肉桂精油,用于食品领域。
在一种实施方式中,所述因子,可以是褪黑激素、生育乙酸酯、生育酚、维生素E,用于防晒、美白、抗衰老、皮肤护理等化妆品领域。
本发明中,用于淀粉改性的植酸(Phytic acid),是一种天然小分子化合物,易溶于水,由一个肌醇和六个磷酸基团组成。作为食品添加剂,植酸具有纯天然、环保和价格低廉等优势,其安全性大于食盐。在酸性条件下,植酸可与带正电的蛋白质形成不溶性的静电复合物,增强乳液液滴之间的空间位阻及物理稳定性。此外,由于植酸具有多个可酯化、络合、螯合的官能团,将其作为磷酸化试剂,对淀粉进行改性,能与淀粉中的羟基共价交联生成磷酸二酯键,可以避免OSA仅作用于淀粉表面的局限性。本发明提出了利用植酸改性淀粉颗粒稳定淀粉基皮克林乳液,植酸对淀粉及蛋白质的双交联作用具有蛋白质取代淀粉基皮克林乳液中小分子乳化剂并维持体系稳定的能力,这对扩大其在食品、医药基化妆品领域的应用具有重要意义。
本发明中利用植酸改性淀粉颗粒与蛋白质相互协同作用,稳定淀粉基皮克林溶液;和已有技术相比,其技术进步是显著的。本发明制备的淀粉基皮克林溶液能够在O/W液滴表面进行吸附,通过形成致密的界面膜抑制液滴的聚结和Ostwald熟化,利于皮克林乳液的稳定(7天)。本发明中制备的可食用的非小分子表面活性剂稳定的皮克林乳液粒径大小为0.3~0.6μm,制备方法简单,可广泛应用于食品、化妆品以及药剂领域。
[有益效果]
本发明利用植酸与淀粉及蛋白质三者之间的相互作用,改性淀粉颗粒紧密吸附在油水界面,阻碍液滴聚结。将该乳液在25℃储藏7天,乳液仍能保持粒径大小为3.3~9.8μm的液滴大小,在同样的储存条件下,空白对照组中粒径增加至5~10μm,说明植酸改性淀粉颗粒大大提高了淀粉基皮克林乳液的稳定协同稳定及油水之间的界面平衡,克服了现有技术中对于利用传统淀粉稳定乳液效果差的缺陷。
(2)本发明构建的淀粉基皮克林乳液具有较强的稳定性,可以使用极低浓度的蛋白质(0.2%左右),避免合成或小分子表面活性剂的使用,制备条件简单温和,原料绿色环保等特点,并且稳定性好,这增加了淀粉基食品的安全性及应用范围。

具体实施方式
以下对本发明的优选实施例进行说明,应当理解实施例是为了更好地解释本发明,不用于限制本发明。
测试方法:
微观结构:将10μL皮克林乳液样品小心置于玻璃片上,覆以盖玻片,利用光学显微镜在40×下观察。
粒度大小:用蒸馏水稀释200倍。使用动态光散射(DLS)和纳米粒度仪测定样品的平均粒径,所有测量均进行三次。
激光共聚焦显微镜:用生物激光共聚焦显微镜观察水凝胶或乳液凝胶的微观分布。尼罗红和FITC分别用作油相和蛋白质相的荧光染料。将皮克林乳液(3μL)转移到载玻片上。将尼罗红(0.1wt%,DMSO)和FITC(0.1wt%,丙酮)按1:1的比例添加到每个样品中,盖上盖玻片。在633nm和488nm的激发波长下,用40×放大镜拍摄每个样品代表区域的图像,由He-Ne激光器提供。
实施例1:协同稳定的皮克林乳液及其制备
一种植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液及其制备方法,包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加水制备不同浓度(1wt%、3wt%、5wt%)的植酸改性淀粉颗粒分散液并于90℃糊化30min,和3wt%的乳清分离蛋白分散液,将糊化处理后的植酸改性淀粉颗粒分散液、乳清分离蛋白分散液按10:1混合获得水相;
(4)将油相与水相按3:7混合,所得混合物立即通过高速均质机进行均质处理,制备得到不同植酸改性淀粉颗粒添加量的皮克林乳液。
将获得的皮克林乳液,在室温静置一段时间后,观察其是否发生分层现象,以判断其稳定性。结果如表1和图2所示。

由图2可知,将皮克林乳液贮藏7天,发现植酸改性淀粉颗粒浓度1wt%制备的皮克林乳液,在均质后静置仅12h就已看到明显分层,贮藏七天后分层更为显著。与此相反,3%的植酸改性淀粉颗粒制备的皮克林乳液在七天后才可以观察到极细微的分层状态,远高于1%淀粉浓度的乳液稳定性,这表明植酸改性淀粉浓度与皮克林乳液的稳定性密切相关。
实施例2:协同稳定的皮克林乳液及其制备
一种植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液及其制备方法,包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加入蒸馏水,配置含有3wt%植酸改性淀粉颗粒的植酸改性淀粉颗粒分散液及3wt%的乳清分离蛋白分散液;将糊化处理(90℃糊化30min)后的植酸改性淀粉颗粒分散液、乳清分离蛋白分散液,按照体积比10:9~10:0.1的比例混合获得水相;
(4)将油相与含有植酸改性淀粉颗粒及蛋白质的水相按3:7混合,所得混合物立即通过高速均质机进行均质处理,制备得到皮克林乳液。
将获得的皮克林乳液,进行光学显微镜及激光共聚焦扫描电镜观察,结果如表2和图3所示。

经过纳米粒度仪测定,发现淀粉与乳清分离蛋白的浓度比为10:1时的实施例2在高速乳化后的液滴大小为3.3μm,远小于10:7(4.2μm)、10:0.7(4.2μm)和10:0.1(9.8μm)时的液滴大小。
实施例3:协同稳定的皮克林乳液及其制备
一种植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液及其制备方法,包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加入蒸馏水,配置含有3wt%植酸改性淀粉颗粒的植酸改性淀粉颗粒分散液及3wt%的乳清分离蛋白分散液;将植酸改性淀粉颗粒分散液于90℃糊化30min后与乳清分离蛋白分散液,按照体积比10:1的比例混合获得水相;
(4)将油相与含有植酸改性淀粉颗粒及蛋白质的水相按不同的体积比混合,所得混合物立即通过高速均质机进行均质处理,制备得到皮克林乳液。
比较不同油水比制备的皮克林乳液的稳定性,结果如表3和图4所示。

从图4光学显微镜的结果,可以观察到的油水比也会影响乳液的界面稳定性,油水比例为3:7时的效果对粒径大小及乳液微观稳定性最为显著。
实施例4:包封有百里香酚(2-异丙基-5-甲基苯酚)的协同稳定的皮克林乳液
包封有2-异丙基-5-甲基苯酚的植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液,其制备包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加入蒸馏水,配置含有3wt%植酸改性淀粉颗粒的植酸改性淀粉颗粒分散液及3wt%的乳清分离蛋白分散液;将植酸改性淀粉颗粒分散液于90℃糊化30min后与乳清分离蛋白分散液,按照体积比10:1的比例混合获得水相;
(4)将500mg/mL的百里香酚在90℃下添加至橄榄油后搅拌一小时作为油相,并与含有植酸改性淀粉颗粒及蛋白质的水相按3:7的体积比混合,所得混合物立即通过高速均质机进行均质处理,制备得到包封有百里香酚的皮克林乳液。
该包封有百里香酚的皮克林乳液,可以递送疏水性抗真菌化合物,来治疗口腔念珠菌病带来的真菌感染等,降低细胞毒性。
实施例5:包封有磷酸奥司他韦的协同稳定的皮克林乳液
包封有磷酸奥司他韦的植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液,其制备包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加入蒸馏水,配置含有3wt%植酸改性淀粉颗粒的植酸改性淀粉颗粒分散液及3wt%的乳清分离蛋白分散液;将植酸改性淀粉颗粒分散液于90℃糊化30min后与乳清分离蛋白分散液,按照体积比10:1的比例混合获得水相;
(4)将0.6mg/mL的磷酸奥司他韦添加至玉米油作为油相,并与含有植酸改性淀粉颗粒及蛋白质的水相按3:7的体积比混合,所得混合物立即通过高速均质机进行均质处理,制备得到包封有磷酸奥司他韦的皮克林乳液。
该包封有磷酸奥司他韦的淀粉基皮克林乳液,在30天的延长释放期内可持续缓慢释放,发挥靶向抗癌活性,调节人胰腺癌细胞活力,在医药领域有巨大的发展潜力。
实施例6:包封有生育酚乙酸酯的协同稳定的皮克林乳液
包封有生育酚乙酸酯的植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液,其制备包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加入蒸馏水,配置含有3wt%植酸改性淀粉颗粒的植酸改性淀粉颗粒分散液及3wt%的乳清分离蛋白分散液;将植酸改性淀粉颗粒分散液于90℃糊化30min后与乳清分离蛋白分散液,按照体积比10:1的比例混合获得水相;
(4)将300mg/mL的生育酚乙酸酯添加至玉米油作为油相,并与含有植酸改性淀粉颗粒及蛋白质的水相按3:7的体积比混合,所得混合物立即通过高速均质机进行均质处理,制备得到包封有生育酚乙酸酯的皮克林乳液。
该包封有生育酚乙酸酯的淀粉基皮克林乳液,包封效率可达到98%以上,具有较强的紫外光照射及贮藏稳定性,可有效降低皮肤的氧化应激,用于化妆品的研制与开发。
实施例7:包封有姜黄素的协同稳定的皮克林乳液
包封有姜黄素的植酸改性淀粉-蛋白质复合物颗粒协同稳定的皮克林乳液,其制备包括如下步骤:
(1)称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后,将分散液的pH调至7.0;加入植酸,在600rpm转速下搅拌5h,利用0.1M NaOH和0.1M HCl维持体系的pH7.0保持恒定,最后将体系的pH值调节至6.5以终止反应。
(2)将植酸改性后的淀粉分散液于3500rpm离心15min,将沉淀物用水和乙醇洗涤充分,直至上清液与AgNO3反应后无白色沉淀,以确保除去植酸改性淀粉中的氯离子等杂质;将沉淀物于50℃烘干24h并研磨粉碎,获得磷酸化的木薯淀粉颗粒,即植酸改性淀粉颗粒。
(3)分别加入蒸馏水,配置含有3wt%植酸改性淀粉颗粒分散液及3wt%的乳清分离蛋白分散液;将植酸改性淀粉颗粒分散液于90℃糊化30min后与乳清分离蛋白分散液,按照体积比10:1的比例混合获得水相;
(4)将1.0mg/mL的姜黄素添加至玉米油作为油相,并与含有植酸改性淀粉颗粒及蛋白质的水相按3:7的体积比混合,所得混合物立即通过高速均质机进行均质处理,制备得到包封有姜黄素的皮克林乳液。
该包封有姜黄素的皮克林乳液,包封率可达91.7%,在消化过程中有更高的生物可及性,可以用于脂肪替代、作为脂溶性活性分子的新型运载工具。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。
Claims (12)
1.一种协同稳定的淀粉基皮克林乳液,其特征在于,所述淀粉基皮克林乳液的制备方法包括;
(1)制备植酸改性淀粉:称取木薯淀粉制备30wt%的淀粉分散液,搅拌至均匀后;加入植酸,植酸的质量是淀粉干基质量的0.01~4wt%;搅拌均匀得到植酸改性淀粉分散液,维持体系的pH 7.0;
(2)制备植酸改性淀粉颗粒:对步骤(1)得到的分散液进行离心、洗涤,烘干沉淀物,研磨粉碎,获得植酸改性淀粉颗粒;
(3)制备淀粉基皮克林乳液:将步骤(2)得到的植酸改性淀粉颗粒和蛋白质分别分散于水中,配制3wt%的淀粉颗粒分散液和3wt%的蛋白分散液,并对植酸改性淀粉颗粒分散液进行糊化处理;植酸改性淀粉颗粒分散液和蛋白质分散液以10:1的体积比进行混合得到水相混合液,再以3:7的比例将油相与水相混合,通过高速搅拌、高压均质或超声方法制备协同稳定的淀粉基皮克林乳液。
2.包封有因子的权利要求1的淀粉基皮克林乳液,其特征在于,在水相或者油相中添加对应的需要包封的因子,然后再通过高能乳化法制备淀粉基皮克林乳液。
3.根据权利要求2所述的淀粉基皮克林乳液,其特征在于,所述因子,包括磷酸奥司他韦、布洛芬、阿霉素、褪黑激素、百里香酚(2-异丙基-5-甲基苯酚)、姜黄素、白藜芦醇、虾青素、茶多酚、花青素、维生素C、维生素E、肉桂精油、生育乙酸酯。

5.权利要求1的淀粉基皮克林乳液在制备食品时的应用。
6.权利要求1的淀粉基皮克林乳液在制备化妆品中的应用。
7.权利要求2的淀粉基皮克林乳液在制备药剂时的应用。
8.权利要求2的淀粉基皮克林乳液在制备食品时的应用。
9.权利要求2的淀粉基皮克林乳液在制备化妆品中的应用。
10.权利要求3的淀粉基皮克林乳液在制备药剂时的应用。
11.权利要求3的淀粉基皮克林乳液在制备食品时的应用。
12.权利要求3的淀粉基皮克林乳液在制备化妆品中的应用。
