Abstract
本申请涉及干细胞外泌体在治疗衰老的药物中的应用。本申请通过衰老细胞模型发现,干细胞外泌体能够使周期阻滞的衰老细胞重新返回细胞周期,即使衰老细胞重新分裂增殖。此发现正打破目前了“细胞衰老不可逆”的学说。基于此,本申请提供了干细胞外泌体在治疗衰老的药物中的应用以及干细胞外泌体在制备恢复衰老细胞的细胞周期的产品中的应用。进一步地,提供了干细胞外泌体能够用于预防和治疗由于细胞衰老所致的年龄相关性疾病。
Description
技术领域
本申请属于生物医药领域,具体涉及干细胞外泌体在预防或治疗衰老的药物中的应用。
背景技术
衰老分为个体衰老、器官衰老、细胞衰老几个水平,个体与器官的衰老最终反应在细胞衰老上。而细胞衰老指“细胞周期(cell cycle)的永久性阻滞”,也就是说细胞衰老是不可逆的,如果能逆转细胞衰老,也意味着“逆龄”。随着人类生活质量的提高,衰老问题已日渐成受到关注,研发具有抗衰老的新药既具有重大社会意义又有巨大商业价值。
干细胞是具有在一定条件下无限自我更新与增殖分化能力的一类细胞,能够产生表现型与基因型和自己完全相同的子细胞,也能产生组成机体组织、器官的已特化的细胞,同时还能分化为祖细胞。干细胞存在多种分类方法,根据发育阶段的不同,干细胞可以分为胚胎干细胞和成体干细胞。其中胚胎干细胞是早期胚胎或原始性腺中分离出来的一类细胞,具有体外培养无限增殖、自我更新和多向分化的特性。根据分化潜能的不同,干细胞可以分为全能干细胞、多能干细胞和单能干细胞。
外泌体(Exosome,exo)是指由细胞分泌的、直径为30nm~150nm、包含了复杂RNA和蛋白质的小囊泡。1983年,外泌体首次于绵羊网织红细胞中被发现,1987年Johnstone将其命名为“exosome”。多种细胞在正常及病理状态下均可分泌外泌体,且外泌体天然存在于体液中,包括血液、唾液、尿液、脑脊液和乳汁中。其形态为杯状结构或一侧凹陷的半球型,具有明显的膜结构。外泌体的膜结构和细胞膜一样为磷脂双分子层,膜上分布着跨膜蛋白以及一些脂质和配体,包扩4次跨膜蛋白家族成员CD9、CD63和CD81等,囊泡内含有十分丰富的蛋白质、DNA和RNA。大量研究表明外泌体可以介导细胞间的通讯。那么其介导通讯的方式主要有以下三种:一是外泌体膜蛋白可以与靶细胞膜蛋白结合,进而激活靶细胞细胞内的信号通路。二是在细胞外基质中,外泌体膜蛋白可以被蛋白酶剪切,剪切的碎片可以作为配体与细胞膜上的受体结合,从而激活细胞内的信号通路。三是外泌体膜可以与靶细胞膜直接融合,非选择性的释放其所含的蛋白质、mRNA以及miRNA等。外泌体由于荷载着许多蛋白,核酸和脂质,因此具有内在的疾病治疗潜能,特别是来源于干细胞的外泌体在血管再生,抑制凋亡,免疫信号传递,组织再生等方面均有重要作用。对于胚胎干细胞来说,外泌体既可以发挥出媲美干细胞的治疗效果又规避了胚胎干细胞伦理以及致瘤性的问题,因此具有比较突出的应用价值。
发明内容
基于此,本申请有必要提供了一种干细胞外泌体在预防或治疗衰老的药物中的应用,所述药物可以逆转衰老进程,恢复衰老细胞的细胞周期和增殖能力。
具体技术方案如下:

发明人研究初期构建了LO2-P21-YFP阳性细胞系,使用Doxorubicin诱导LO2-P21-YFP细胞系衰老,使用YFP进行流式荧光分选后获得衰老细胞模型。进一步地,将上述衰老细胞模型用于筛选抗衰老药物,插入YFP基因片段的目的为了高效筛选抗衰老药物,只通过检测YFP荧光就可以判断候选药物是否可以作为抗衰老药物。在药物筛选过程中发现胚胎干细胞(ES)外泌体可以逆转衰老细胞的衰老进程,恢复已经衰老的细胞重新进入细胞周期,并恢复其增殖能力,在动物体内也进一步证实。
目前,衰老细胞定义为细胞周期永久终止,而本申请发现衰老细胞并非细胞周期永久终止,而是暂时性停止,可以通过外源因子重新恢复细胞周期,此发现作为一项颠覆性结果,这将对衰老这一亟待解决重大问题带来新思路以及新的潜在药物。
因此,本申请一实施方式提供了一种干细胞外泌体在预防或治疗衰老的药物中的应用。
在一个具体示例中,衰老包括个体衰老、器官衰老和细胞衰老中的一种或多种。

具体的,细胞培养上清于低速低温离心机中,离心3分钟~6分钟除去死细胞;1500g~2500g、低温离心15分钟~20分钟以去除细胞碎片;10000g低温离心20分钟~30分钟进一步去除大囊泡;而后于100,000g、低温离心60分钟~90分钟。离心后的沉淀用PBS重悬,再次于100,000g、低温离心60分钟~90分钟纯化外泌体,加入适量PBS重悬外泌体,过滤除菌,测定浓度分装低温保存。

在一个具体示例中,该药物可以与其他治疗衰老的药物联合使用,所述联合使用的方法包括在所述干细胞外泌体上包载有其他治疗抗衰老的药物。可选的,其他用于治疗衰老的药物包括抑制衰老的蛋白或者RNA,或者促进衰老的蛋白或RNA的抑制剂,可选抑制剂包括能敲低或敲除蛋白或RNA表达或活性的物质,例如siRNA或shRNA,当然还包括其他具有相似功能的物质。
在一个具体示例中,该药物还包括药学上可接受的辅料。

下述的具体实施例中,涉及原料组分的量度参数,如无特别说明,可能存在称量精度范围内的细微偏差。涉及温度和时间参数,允许仪器测试精度或操作精度导致的可接受的偏差。
实施例1Doxorubicin诱导LO2-p21-YFP细胞系制备细胞衰老模型
发明人研究初期,利用CRISPR/Cas9技术,借助Cas9、gRNA、以及p21-YFP donor进行基因编辑。具体地,将编码YFP基因片段定点插入LO2细胞系的p21的终止子密码子的位置,并筛选出LO2-P21-YFP阳性细胞系。
具体的,构建CRISPR/Cas9载体:利用gRNA序列构建CRISPR/Cas9载体,所述gRNA的序列为GGCTTCCTGTGGGCGGATTA,SEQ ID No:1。
合成p21-YFP donor质粒:首先,将YFP编码基因的序列(SEQ.ID No.2所示的第3536~第4252位置的碱基)插入到Venus-P2A-Neo质粒中,制备YFP-P2A-Neo质粒。以LO2细胞基因组DNA和YFP-P2A-Neo质粒为模板,使用表1中的引物,扩增Vector片段、p21Upstream片段、YFP-NeoR片段、p21 Downstream片段,使用Gibson反应体系将上述四个片段组装成p21-YFP donor质粒(donor质粒序列如SEQ.ID No.2所示),上述实验均由生工生物完成。

进一步的,将含有gRNA的CRISPR/Cas9载体和p21-YFP donor质粒共转染LO2细胞中,筛选LO2-P21-YFP阳性细胞系。
更进一步的,使用Doxorubicin诱导LO2-P21-YFP细胞系衰老,使用YFP进行流式荧光分选后,获得衰老细胞模型。
P21(cyclin-dependent kinase inhibitor p21,CDKN1A)是一个重要的衰老标志物,P21是磷酸化P53的下游CDKI,通过结合CDK2阻断细胞周期进入G1/S一直细胞周期。P21的高表达与衰老相关。
Doxorubicin是DNA拓扑异构酶抑制剂,常用于肿瘤治疗,在低浓度50nM下能够诱导细胞衰老。由于Doxorubicin分子中蒽环配基上的醌-氢醌结构,具有接受电子与提供中子的能力,插入DNA相邻碱基对之间,产生活性自由基,使DNA双股螺旋解旋,DNA链断裂,抑制核酸的模板活性,干扰转录过程,抑制mRNA合成。另外也可能引起细胞膜破裂,呈现细胞毒性作用,属细胞周期非特异性药物,对各期细胞均有作用,但对S期的早期最为敏感,M期次之,而对G1期最不敏感,对G1、S和G2期有延缓作用。
具体的,Doxorubicin诱导p21-YFP-Neo细胞系衰老的步骤包括如下:
(1)将0.3x106个LO2-p21-YFP细胞接种到35mm激光共聚焦细胞培养皿,培养12hour。
(2)细胞贴壁之后换液,加入含有50nM的Doxorubicin培养基。
(3)24hour之后,换成正常培养基。
(4)培养4day之后,使用YFP进行流式荧光分选,观察分选后的细胞中p21-YFP的表达情况。
(5)进行衰老相关β-半乳糖苷酶染色。
其中,β-半乳糖苷酶染色方法,包括如下步骤:
将10X Fixative Solution稀释成1X,Staining Solution在37℃水浴中彻底溶解稀释成1X;将20mg的X-gal溶解于DMF中,避光储存于-20℃;配置染色液,6孔板每个孔加入1mL的染色液,930μL 1X Staining Solution、10μL 100XStaining Solution A、10μL100XStaining Solution B和50μL 20mg/mLX-gal配成1mL染色液,并调节pH使pH=6.0;去除细胞培养上清,加入PBS清洗两次,6孔板每个空加入1mL 1XFixative Solution室温固定15min;去除固定液,PBS清洗一次,加入染色液,放入无二氧化碳的培养箱37℃染色12小时;将培养皿置于显微镜下观察染色的情况。
通过FITC荧光设门,选取YFP阳性细胞进行分选,结果如图1所示。
将分选后的细胞铺板,每孔接种0.3x106个细胞接种于35mm细胞培养皿中,通过荧光显微镜成像如图2所示,Doxorubicin能够诱导p21-YFP的表达。
β-半乳糖苷酶染色结果如图3所示,表明加入Doxorubicin能够诱导LO2-p21-YFP细胞系发生衰老,说明成功构建细胞衰老模型。
实施例2胚胎干细胞外泌体分离与鉴定
1.胚胎干细胞(ES)培养:
将人胚胎干细胞h9细胞铺于基质胶包被好的培养皿中,培养基为hESC-ncTarget。每天换液,当细胞长满时,加入EDTA细胞传代工作液消化细胞,按1:8传代。
2.外泌体提取:
(1)收集培养上清液用于外泌体提取。
(2)使用梯度离心的方案提取外泌体。细胞培养上清于300g、4℃离心5分钟除去死细胞;2000g、4℃离心20分钟以去除细胞碎片;10000g、4℃离心30分钟进一步大囊泡;而后于100,000g、4℃超速离心90分钟。
(3)超速离心后的沉淀用50ml PBS重悬,再次于100,000×g、4℃超速离心90分钟进一步纯化外泌体。
(4)加入适量PBS重悬外泌体,使用0.22μm滤器过滤除菌,BrandFord法测浓度后分装,于-80℃保存。
3.外泌体鉴定:
采用透射电镜检测观察外泌体的形态,结果如图4中的A所示,提取的ES外泌体为典型的双层膜囊泡,并具有清晰的杯状结构。
粒度分析检测结果如图4中B-C所示,ES外泌体的平均粒径分别为133.161nm,且主要分布于50~200nm,呈相对正态分布。
采用免疫印迹鉴定外泌体标志Calnexin(阴性)、TSG101、CD63。结果如图4中的D所示,分离获得的ES外泌体表达外泌体标志蛋白CD63和TSG101,同时阴性蛋白Calnexin无表达。
实施例3胚胎干细胞外泌体引起LO2衰老细胞重新具备增殖能力
根据实施例1经流式细胞术筛选出Dox诱导的衰老细胞。将衰老细胞铺板,每孔接种3x105细胞,Dox-Exos组中每孔加入10微升的2mg/mL Exos,处理1d;Dox-PBS中每孔加入10微升的PBS溶液,处理1d,并设置LO2正常细胞组(Con)做对照,收集细胞进行细胞增殖检测。ES外泌体后意外的发现衰老细胞数量随培养时间增多,见图5,显示出衰老细胞可能发生了增殖。进一步检测了增殖信号,结果如图6所示,Dox加外泌体组衰老细胞除P21荧光强度下降以外,有大量细胞有Ki67阳性,也即作为增殖标志物信号增加,同时P21作为衰老标志物表达量下降。再进一步地,进行了β-gal染色,如图7所示,正常LO2细胞衰老细胞比例低于5%,而衰老细胞组超过80%细胞衰老,加入ES外泌体组约15%细胞衰老,明显降低了衰老细胞比例。
为了进一步验证衰老细胞加入ES外泌体是否产生了增殖现象,我们通过高内涵激光共聚焦细胞成像分析系统(Opera Phenix,珀金埃尔默企业管理有限公司)追踪单个细胞长时间变化,结果如图8所示,追踪到加入ES外泌体组的单个细胞发生增殖现象。细胞衰老的定义是永久性的细胞周期终止状态,这也意味着衰老细胞定义将被改写,也即,衰老细胞并非细胞周期永久终止,而是暂时性停止,可以通过外源因子重新恢复细胞周期。
进一步地,我们进行了三组细胞的转录组测序,结果如图9所示,其中差异较大的关键基因与细胞周期直接相关,如TP53/MYC/PCNA/P21等。进一步地,我们验证了其中一些关键基因的表达,结果如图10所示。其中图10中的A图为衰老标记物P16,P21,P53的qPCR结果,可见衰老组中衰老标记物表达显著提高,ES外泌体组中衰老标记物表达显著恢复。图10中的B图为炎症相关指标的qPCR结果,衰老组中炎症相关指标表达显著提高,加入ES外泌体组中炎症相关指标也显著恢复。我们进一步检测了衰老标记物P16,P21蛋白水平表达如图10中的C-D所示,与RNA水平一致,衰老组显著上调,加入ES外泌体组显著恢复。

进一步地,我们进行了细胞周期检测。具体地,胰酶替代物消化细胞,1000xg离心5min。用1mL预冷的生理盐水清洗细胞,1000xg离心5min,小心吸取上清,吸取的时候残留50μl的上清,避免将细胞吸走,轻弹离心管底部以适当分散细胞,避免细胞成团。加入1mL的预冷的70%乙醇,轻轻吹打混匀,4℃固定24小时。1000xg离心5min,弃掉上清,吸取的时候残留50μl的上清,避免将细胞吸走。加入1mL预冷的生理盐水清洗细胞一次。每管样品中加入0.5mL的PI染色液,缓慢充分重悬细胞,37℃避光染色30min。染色完成后,用流式细胞仪进行检测,激发光波长为488nm。
其中,PI染色液的配制:染色缓冲液500μl,PI染色液(20×)25μl,RNaseA 10μl,总体积535μl。
结果如图11所示,LO2正常细胞约有32%处于S期,LO2细胞经Dox处理后加入PBS后约有8%处于S期,LO2细胞经Dox处理后加入胚胎干细胞外泌体后约有18%处于S期,显著高于加入PBS组。说明胚胎干细胞外泌体可以使已经衰老的细胞重新进入细胞周期,进而逆转衰老。
实施例4胚胎干细胞外泌体处理后单细胞测序分析
具体的操作流程参阅图12所示,将LO2-P21-YFP细胞,经50nM Dox诱导5天,流式细胞术筛选出诱导的衰老细胞;
将衰老细胞铺板,每孔接种0.3x106个细胞于35mm细胞培养皿中,每孔加入10微升的2mg/mL Exos,处理1d;收集细胞进行单细胞测序分析。
其中,单细胞测序样品的制备和测序:将制备的单细胞悬液和含有barcodes的凝胶珠和酶的混合液混合,单后被位于微流体“单十字”连接中的油滴包裹,形成GEMs(GelBead-In-Emulsion),随后在GEMs内进行细胞裂解和逆转录反应。本实验采用10×Genomics系统进行单细胞转录组测序,测序深度为150G,原始图像经GASAVA碱基识别后转化为Rawdata,单细胞测序由上海派森诺生物科技股份有限公司完成。
单细胞数据分析:单细胞测序获取的Raw data按照10×Genomics公司提供的分析流程进行分析(https://www.10xgenomics.com/support/single-cell-gene-expression),主要包括质量控制、数据整合、细胞聚类、细胞注释、差异基因富集分析、拟时序分析等,测序数据主要由R softeware中Seurat、monocles3、CellChat等软件包完成。
数据质控和标准化:单细胞测序的数据较为高纬、稀疏,且存在大量噪声,良好的质量控制是进行有效数据分析的基础。利用Seurat包中Read10函数读取测序数据,merge函数不同组的数据整合到一起,然后添加线粒体和核糖体信息。我们根据以下3种参数进行质量控制:
(1)每个细胞测到的基因数目与序列数。这两条标准保证了进行分析的每个细胞检测到的基因表达的丰度和丰富度,该值越高对细胞表达的情况要求越严格,默认使用较为宽松的阈值,两个指标的最低阈值均为100,最高值不限。本实验设定200<nFeature_RNA<6000。
(2)测序得到的线粒体和核糖体基因的比例。过高的线粒体基因表达量代表着该细胞在裂解前处于濒死状态,其核基因表达会偏向于与细胞活性相关通路的基因(如细胞凋亡),如果不去除这类细胞分析后可能会由于细胞状态较差而导致错误的结论,本实验设定线粒体基因比例<5%。核糖体的基因表达比例则代表着细胞rRNA污染的比例,该值较高代表着mRNA没有得到足够的富集,合格的10×试剂盒及实验操作步骤不会导致mRNA富集程度较低的情况,因此该指标默认的阈值为100,即不使用该指标,但可以根据实验目的设定阈值。
(3)细胞周期。细胞周期是一个受到严格控制的DNA复制和有丝分裂事件,单细胞测序数据中的噪声污染来源之一就是细胞周期。同一种细胞处于不同的状态,由于细胞周期的不同,其表达基因会有巨大差异,这种差异会干扰细胞注释聚类算法的性能,导致相同细胞分到不同的细胞群中。根据Tirosh设计的评分策略,我们对S期和G2M期基因表达情况进行打分,预测每个细胞所处的周期阶段,依据评分结果,在对基因UMIs数据缩放时移除细胞周期异质性。利用Seurat函数获取细胞周期,利用G2M和S期标志物为细胞群分配细胞周期分数。
不同阶段(phase)的标志物的表达量应该表现为负相关,且不表达G2M和S期标志物的细胞判定处于G1期。
Exos干预的细胞处于S期的细胞比例上升,如图13所示。
降维聚类并分群:dim=1:30,resolution=0.6,共得到12clusters,利用增殖标记物PCNA和MKI67以及衰老标记物p21和p53作为marker gene对所有clusters进行注释,如图14所示。
以表达增殖标记物注释较为年轻细胞,以表达衰老标记物注释衰老细胞对细胞进行注释后,分析衰老状态细胞(LO2-O)、年轻细胞(LO2-Y)以及中间状态细胞(LO2-M)三个不同种类细胞群的比例,如图15所示,ES外泌体组(Rejuvenation组)年轻状态细胞比例约25.2%高于对照组15.3%,衰老状态细胞比例约14.7%,低于对照组30.7%。
进一步地,对三类细胞群进行热图分析,并list出top表达基因;对top表达基因进行富集分析,如图16所示。
实施例6胚胎干细胞外泌体逆转小鼠衰老
治疗策略流程参见图17所示,使用20月龄C57BL/6雄性小鼠每组20只,尾静脉分别给与外泌体(30μg的外泌体溶解于200μl的PBS中)和PBS(200μl)处理,每周2次,持续1个月。其中,15只用于观察小鼠存活期,如图18所示,结果显示正常小鼠存活期中位数约25个月,外泌体尾静脉给药组存活期中位数约30个月,最长可达39个月,显著增加小鼠寿命。
结果如图19所示,相较于老年鼠组(30月龄),给与胚胎干细胞外泌体的老年小鼠弓背、脱毛现象明显缓解,进一步提示了胚胎干细胞外泌体在动物体内具有逆转衰老能力。进一步地,在小鼠第29-30月龄进行了转棒实验,如图20-A所示,小鼠置于转棒上速度从8rpm在300s增加到80rpm,小鼠掉落或跳落终止计时,统计小鼠在转棒上时间,小鼠经过4次训练(每两天一次)后第五次开始外泌体治疗组相较于对照组具有显著性。进一步地,保持转棒以25rpm速度,统计小鼠在转棒上时间,结果如图20-B所示,外泌体治疗组约持续55s,显著高于对照组约32s的时间。进一步地,进行了抓力测试,三次抓力结果取平均后除以体重,每两天测一次,结果如图20-C显示外泌体治疗组均优于对照组。以上结果显示ES外泌体治疗组衰老明显缓解。
老年鼠往往伴随有膀胱过度活动症状(OAB),OAB表现为尿频、尿急伴有或不伴有尿失禁现象。老年小鼠排尿往往排尿少量多次,在滤纸上可见小斑点,而年轻鼠以大斑点为主,结果如图21所示,ES外泌体治疗后OAB现象明显缓解。
进一步地,对小鼠多个器官病理切片进行了β-gal染色,检测组织衰老水平,如图22所示,衰老鼠(30月龄)相较于年轻鼠(12月龄)在肾脏/肝脏/肺脏/脾脏等多个器官出现β-gal染色阳性,ES外泌体治疗后显著缓解。
实施例7小鼠PBMC单细胞测序
小鼠PBMC获取,PBMC分离方法如下:
提前将淋巴细胞分离液从4℃取出,恢复至室温,取4mL转移至15mL试管中备用。小鼠眼球取血1mL血液样本置于EDTA钠采血管中,取1mL血液用DPBS 1:1稀释,用无菌吸管吸取稀释后的血液,沿管壁缓慢加入至人淋巴细胞分离液液面上。将离心管缓缓地不要晃荡,保持界面清晰,放入水平离心机,800g,4℃离心20min。离心完毕后,小心取出15mL试管,离心后管内可分为三层,在上、中层界面处有一层以单个核细胞为主的白色云雾层狭窄带,即为我们所需要的单个核细胞。用无菌吸管吸取白色云雾层狭窄带的单个核细胞,并置于另一个15mL离心管中,加入5mL DPBS+2%FBS,1500rpm,4℃离心5min。弃去上清,沉淀加入红细胞裂解液,室温静置裂解2min,800rpm离心5min。弃去上清,沉淀加入5mL DPBS+2%FBS,500rpm,4℃离心5min。重复该步骤一次。弃去上清,加入200μL DPBS+2%FBS重悬细胞,进行细胞计数和活力测定,吸取适量体积细胞悬液进行后续实验。

单细胞数据分析方法参见实施例4。
利用Seurat包对不同样本的细胞周期阶段进行打分,判定所处的细胞周期阶段,如图23所示。
统计不同细胞周期阶段的细胞比例并量化,如图24所示,Ageing组G1期和G2M期比例上升,提示Ageing组PBMC中细胞发生周期阻滞;Exos处理抬升了S期PBMC中细胞比例。
实施例8小鼠皮肤单细胞测序
实验分组:12月龄、30月龄、30月龄ES外泌体治疗组分别取3只进行皮肤组织单细胞测序。
取材:背侧皮肤用解剖剪刀沿筋膜面分离,进一步修剪皮下脂肪并用冷PBS冲洗5次。用眼科剪继续将皮肤剪碎,用Liberase DL酶(罗氏,0.5mg/mL)酶解1小时,并用40μm尼龙滤网过滤。
单细胞样品制备和测序、样品准备、测序上机和分析过程参见实施例7。
结果见图25。按照表面标志物将皮肤细胞分为不同类群(图25-A),差异基因见图25-B,细胞周期分析见图25-C-D,可见30月龄鼠皮肤约20%处于S期,12月龄鼠约40%处于S期,30月龄加ES外泌体治疗鼠约32%处于S期,即治疗后皮肤细胞增殖能力显著提高。进一步对CCGs亚群差异基因进行了分析(图25-E),并对G1/S与G2/M进行了分析,结果显示外泌体治疗后处于S期细胞变多(图25-F-G)。进一步地对细胞周期相关基因进行了分析,统计差异基因并进行功能分析(图25-H-I)。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本申请的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本申请构思的前提下,还可以做出若干变形和改进,这些都属于本申请的保护范围。因此,本申请专利的保护范围应以所附权利要求为准,说明书及附图可以用于解释权利要求的内容。
Claims (11)
1.干细胞外泌体在预防或治疗衰老的药物中的应用。
2.根据权利要求1所述的应用,其特征在于,所述衰老包括个体衰老、器官衰老和细胞衰老中的一种或多种。
3.干细胞外泌体在制备恢复衰老细胞的细胞周期的产品中的应用。
4.根据权利要求3所述的应用,其特征在于,所述产品具有促进衰老细胞重新进入细胞周期S期的特征。
5.干细胞外泌体在恢复或促进衰老细胞的增殖的产品中的应用。
6.干细胞外泌体在预防或治疗细胞衰老所致的相关性疾病的产品中的应用。
7.根据权利要求1~6任一项所述的应用,其特征在于,所述干细胞外泌体包括胚胎干细胞外泌体、间充质干细胞外泌体及诱导性多能干细胞外泌体中的至少一种。
8.根据权利要求1~6任一项所述的应用,其特征在于,所述干细胞外泌体为人源干细胞外泌体或其他哺乳动物来源干细胞外泌体。
9.根据权利要求1~6任一项所述的应用,其特征在于,所述产品包括药物或制剂;
可选的,所述药物的活性成分为干细胞外泌体。
10.根据权利要求9所述的应用,其特征在于,所述药物与其他治疗衰老的药物联合使用;
可选的,联合使用方式包括在所述干细胞外泌体内包载有其他治疗衰老的药物;
可选的,所述其他用于治疗衰老的药物包括抑制衰老的蛋白或者RNA,或者促进衰老的蛋白或RNA的抑制剂。
11.根据权利要求10所述的应用,其特征在于,所述药物还包括药学上可接受的辅料。
