Abstract
本发明公开了一种具有葡萄糖响应性的载药水凝胶及其制备方法和应用,涉及化学和生物医药技术领域。所述制备方法,包括以下步骤:ε‑聚赖氨酸和2‑甲酰基苯硼酸反应得到苯硼酸的聚合物PL‑PBA;CaCl2溶液和山药多糖反应后,再加入碳酸盐溶液,继续反应得到山药多糖‑CaCO3微球;将所述山药多糖‑CaCO3微球分散在聚乙烯醇溶液中,得到DOP‑CaCO3/PVA混合液,再在所述DOP‑CaCO3/PVA混合液中加入降血糖药物溶液,混合均匀后加入所述PL‑PBA,反应后得到所述载药水凝胶。本发明开发的载药水凝胶具有葡萄糖响应性,能够有效抑制糖尿病伤口的炎症反应,促进伤口愈合,同时具有良好的生物相容性。
Description
技术领域
本发明涉及化学和生物医药技术领域,特别是涉及一种具有葡萄糖响应性的载药水凝胶及其制备方法和应用。
背景技术
慢性糖尿病伤口是糖尿病的一种严重并发症,通常会导致高治疗成本和高截肢率。许多研究试图探索由于伤口生理环境的差异而导致慢性糖尿病伤口不愈合的原因。正常伤口的愈合包括止血、消炎、增殖和重塑;然而,伤口微环境中高浓度的葡萄糖会阻碍慢性糖尿病伤口进入组织重塑阶段。长期的高血糖使晚期糖基化终产物(AGEs)在皮肤组织中积聚,进而导致组织学和细胞学的一系列变化,如细胞功能障碍和多种生长因子的分泌减少,特别是炎症细胞功能障碍,加剧并延长了慢性糖尿病伤口的炎症期。此外,影响糖尿病伤口愈合的另一个因素是去除坏死组织后伤口的不规则形状;由于其不规则的伤口形状,传统的临床敷料非常不合适,进而导致感染风险更高。在糖尿病的治疗中,目前依然以皮下注射胰岛素治疗为主,然而,注射胰岛素的脂溶性差、易失活和高分子量降低了其生物利用度,导致糖尿病慢性并发症的发生,同时,这种给药方式可能会在注射部位带来疼痛和感染,这很容易导致糖尿病患者的心理紧张。水凝胶在胰岛素的输送系统中有着独特的优势,既可实现药物的靶向可控递送,降低药物副作用,又能模仿细胞外基质,作为良好的伤口敷料。基于PBA的葡萄糖响应性水凝胶已得到广泛研究,但是由于苯硼酸pKa值较高,生理条件下,无法与葡萄糖分子竞争性的结合,且在体内,合成的药物载体易被巨噬细胞识别而发生排异反应,无法参与体内循环过程。
因此,有必要开发一种完全适应伤口形状并能调节伤口葡萄糖水平的敷料,以加速慢性糖尿病伤口的愈合。
发明内容
本发明的目的是提供一种具有葡萄糖响应性的载药水凝胶及其制备方法和应用,以解决上述现有技术存在的问题,该载药水凝胶具有葡萄糖响应性,能够有效抑制糖尿病伤口的炎症反应,促进伤口愈合,同时具有良好的生物相容性。
为实现上述目的,本发明提供了如下方案:
本发明提供一种具有葡萄糖响应性的载药水凝胶的制备方法,包括以下步骤:
(1)ε-聚赖氨酸和2-甲酰基苯硼酸反应得到苯硼酸的聚合物PL-PBA;
(2)CaCl2溶液和山药多糖反应后,再加入碳酸盐溶液,继续反应得到山药多糖-CaCO3微球;
(3)将所述山药多糖-CaCO3微球分散在聚乙烯醇溶液中,得到DOP-CaCO3/PVA混合液,再在所述DOP-CaCO3/PVA混合液中加入降血糖药物,混合均匀后加入所述PL-PBA,反应后得到所述载药水凝胶。
进一步地,在步骤(1)中,所述ε-聚赖氨酸和所述2-甲酰基苯硼酸的摩尔比大于或等于2:1。
进一步地,在步骤(1)中,所述反应的pH值为4.5-5.5,温度为80℃,时间为24-72h。
进一步地,在步骤(2)中,所述CaCl2溶液中的Ca2+、所述碳酸盐溶液中的CO3 2-和所述山药多糖的摩尔质量比为(3-8)×105:(3-8)×105:1。
进一步地,在步骤(3)中,所述聚乙烯醇溶液中的聚乙烯醇与所述山药多糖-CaCO3微球的质量为20:(1-4)。
进一步地,在步骤(3)中,所述降血糖药物包括胰岛素。
本发明还提供一种根据上述的制备方法制备得到的载药水凝胶。
本发明还提供上述的载药水凝胶在制备促进糖尿病伤口愈合的药物中的应用。

本发明公开了以下技术效果:
葡萄糖响应性体系可被应用于人体血糖检测和糖尿病治疗药物的可控性释放,随着技术的发展,体系的精确控制越来越为大家所关注。为了达到准确的控制,首先需要体系在人体生理pH7.4下,能够对对葡萄糖做出响应;其次,体系在人体生理pH的响应要足够敏感,与人体血糖水平相适应。针对苯硼酸pKa约为8~9,远高于人体生理pH值和苯硼酸体系糖敏感性低,以及材料生物相容性低的问题,本发明以天然药物山药多糖为基础原料,利用苯硼酸与糖单元形成环酯的交联作用,降低苯硼酸的表观pKa值,设计合成了在人体生理pH7.4下具有极高葡萄糖敏感性、可生物降解和良好的生物相容性的凝胶载药体系。同时,山药多糖是天然药物山药中的有效活性成分之一,具有良好的降血糖活性。
本发明首先利用山药多糖(DOP)合成山药多糖-CaCO3(DOP-CaCO3)微球,并将该微球掺杂在生物基材料聚赖氨酸-聚乙烯醇(PL-PVA)水凝胶中,并将胰岛素加载到水凝胶网络中。苯硼酸官能化的聚赖氨酸(PL)可与PVA在数秒钟之内交联形成稳定水凝胶,并且经过微球的掺杂可明显提高水凝胶的机械性能。动态硼酸酯键的形成赋予了水凝胶葡萄糖响应性性能,在高葡萄糖浓度条件下,胰岛素得到缓慢释放。另外,载药水凝胶根据体内葡萄糖浓度调节释放胰岛素的同时,也可释放具有降血糖活性的DOP以及具有抗炎作用的Ca2+。本发明利用体外细胞实验和红细胞溶血实验证明了DOP-CaCO3掺杂的水凝胶的具有较好的生物相容性,及促进大鼠成纤维(NIH/3T3)细胞的增殖作用,且DOP可与释放的胰岛素协同作用促进成纤维细胞对葡萄糖的消耗,改善细胞由高糖引起的氧化应激,降低细胞内炎症因子(TNF-α和IL-6)的表达。将该水凝胶应用于糖尿病大鼠伤口敷料,结果表明本发明合成的水凝胶可以作为抑制糖尿病大鼠伤口的炎症反应,促进伤口愈合,且安全的水凝胶敷料。

以下实施例中所使用的山药多糖(DOP)为怀山药多糖,纯度大于40%,提取方法参考文献“FoodChemistry,227,64-72”。
实施例1
一种能用于加速糖尿病伤口愈合的具有葡萄糖响应性的山药多糖-CaCO3微球掺杂水凝胶的制备方法,步骤如下:
(1)准确称取600mg(4.68mmol)ε-聚赖氨酸(ε-PL)和351mg(2.34mmol)2-甲酰基苯硼酸(2-FPBA)完全溶解于100mLpH4.5的乙酸/乙酸钠缓冲液中,80℃反应72h。反应结束后,将反应液移置透析带(MW1000)中透析7天,除去小分子杂质,最后透析后的溶液冷冻干燥得到海绵状固体,即为PL-PBA。
其中,ε-PL和2-FPBA的摩尔比需要大于等于2:1,否则合成的PL-PBA水溶性不好,无法进行下步合成。
PL-PBA的红外图谱和核磁共振氢谱如图1所示。
对于ε-PL和PL-PBA,2934cm-1处的吸收峰为ε-PL主链上C-H的伸缩振动峰,1678cm-1处的吸收峰为C=O的拉伸振动产生的吸收峰,位于1568cm-1处的峰值归因于C=N的拉伸振动,而1264cm-1处为PBA上B-O-H键的特征吸收峰。由于PBA中存在苯环,因此在832cm-1、726cm-1处出现苯环面外变形的振动峰。由此表明,PBA成功接枝在ε-PL上。
对ε-PL及PL-PBA在D2O中的1HNMR谱图如5-3所示,3.93ppm(d,2H)处的质子峰是C1及C9上的质子信号,1.86ppm(d,4H)处的质子峰归属于C2及C10上H的信号,1.40ppm(d,4H)处的质子峰归属于C3及C11上H的信号,1.59ppm(d,4H)处的质子峰归属于C4及C12上H的信号,3.24ppm(d,4H)处的质子峰归属于C5及C13上H的信号。与ε-PL相比,在7.52ppm(t,4H)处出现了很明显的特征峰,该峰对于苯环骨架上H的质子峰,在6.02ppm(d,1H)附近也出现了新的质子峰,其为与C-N双键中碳原子上质子信号。
以上结果进一步表明成功合成了PL-PBA。
(2)分别称取Na2CO3和CaCl2,分别溶于10mL二次水中,得到0.33M的Na2CO3溶液和0.33M的CaCl2溶液。首先将CaCl2溶液和10mgDOP混合(nCa:nDOP≈3×105:1),室温下搅拌1min,然后,在磁力搅拌下将Na2CO3溶液迅速注入反应液中,800rpm继续搅拌10min。最后,将反应液离心并用二次水洗涤沉淀,真空干燥后得到DOP-CaCO3微球。
图2显示了CaCO3及DOP-CaCO3微球的XRD光谱,由图可看出,CaCO3为方解石型,DOP-CaCO3微球为方解石和球霰石的混合晶型,其中衍射角2θ=29.40°主要与方解石的(104)晶面相对应,而DOP-CaCO3微球出现了球霰石(110)、(112)、(114)的晶格衍射峰。
通过SEM观察DOP-CaCO3微球的微观形貌。如图3中a和b所示,未加DOP的CaCO3呈立方体结构,而加DOP反应后呈均匀的球型,表明已成功合成了DOP-CaCO3微球。
(3)称取4gPVA(聚乙烯醇,Mw~27000)加入36mL二次水于圆底烧瓶中,60℃搅拌2h后,升温至95℃,继续回流15min,得到10%(W/V)PVA溶液,冷却,然后将步骤(2)中得到的DOP-CaCO3微球1mg加入到200μL的上述10%的PVA溶液中,待微球分散均匀立即加入200μL2%(W/V)的用NaOH溶液调节pH至7(pH7-8可达相同效果)的PL-PBA溶液,迅速搅拌,数秒内即可形成水凝胶(PL-PVA/DOP-CaCO3)。
通过扫描电子显微镜(SEM)观察不同浓度PL-PBA聚合物的PL-PVA/DOP-CaCO3水凝胶的微观结构和孔隙率,如图3中c所示,由于动态硼酸酯键的交联作用,所有PL-PVA/DOP-CaCO3水凝胶可以观察到连续的多孔微结构。此外,随着PL-PBA聚合物浓度的增加,多孔结构变得更致密,孔径减小。
通过流变仪进一步评估水凝胶的力学性能,结果如图4所示。在时间扫描模式下(图4中A),所有水凝胶的弹性模量(G’)均大于粘性模量(G”),且每个水凝胶的G’值和G”值在整个测试时间内保持恒定,表明成功合成了水凝胶且具有良好的机械稳定性。水凝胶的机械强度由G’值反应。用振荡频率扫描进一步检测水凝胶的机械强度,如图4中B所示,PL-PBA的浓度越高,PL-PVA水凝胶的机械强度越高,随着PL-PBA浓度的增加,更多的PBA基团可与PVA化学交联,从而使PL-PVA水凝胶呈现出更强的机械强度。其次,制备不同DOP-CaCO3微球含量的水凝胶来检测微球的掺杂对水凝胶机械性能的影响。如图4中C所示,DOP-CaCO3微球的掺杂可明显增高水凝胶的弹性模量(G’),且当掺杂0.25%(W/V)的微球时,水凝胶的弹性模量(G’)最高,且保持稳定。图4中D显示了水凝胶的自愈合性和拉伸性能。与一些由刚性共价网络形成的水凝胶不同,PL-PVA/DOP-CaCO3水凝胶由于硼酸酯键的形成-断裂循环而具有重塑和自愈性能。当水凝胶被拉伸时,它发生了形变,而不是从皮肤表面剥离,表明该水凝胶作为一种动态网络,可通过变形耗散能量,导致作用在粘合界面上的力降低,从而增强水凝胶的粘合强度。水凝胶的拉伸性能表明在活动关节处水凝胶不会随着关节的活动而脱落,由此可见PL-PVA/DOP-CaCO3水凝胶可用作伤口的良好敷料。图4中E显示了DOP-CaCO3微球的掺杂对PL-PVA水凝胶溶胀性能的影响,PL-PVA水凝胶在90分钟内达到溶胀平衡(≈448%),之后开始出现降解。而PL-PVA/DOP-CaCO3水凝胶在75分钟内达到溶胀平衡,且溶胀率明显升高(≈502%),在180分钟后开始的降解。表明,经DOP-CaCO3微球掺杂后的明显提高了水凝胶的溶胀性能,PL-PVA/DOP-CaCO3水凝胶可较快的吸收伤口渗出物和血液,并可保持稳定的形状和体积,同时保持湿润的愈合环境,加速伤口愈合。
(4)水凝胶对胰岛素的负载:用0.1M的HCl溶解胰岛素并用PBS定容,配置成2mg/mL的胰岛素溶液。载药水凝胶的制备过程与(3)部分相同,在加入PL-PBA溶液之前,在200μLDOP-CaCO3/PVA混合液中滴加100μL的胰岛素溶液,混合均匀后迅速加入200μL的PL-PBA溶液,搅拌均匀,形成载药水凝胶(INS@PL-PVA/DOP-CaCO3)。
水凝胶的体外释放:将载药水凝胶置于2.5mL不同浓度葡萄糖的PBS溶液(0mg/mL、1.0mg/mL和4.0mg/mL)中,37℃下摇晃,并在一定时间间隔抽取250μL介质以检测释放的胰岛素量,同时补充相同体积的新鲜的相应溶液,以保持释放介质体积不变。使用ELISA试剂盒检测释放胰岛素的含量。
PL-PVA/DOP-CaCO3水凝胶在不同葡萄糖浓度下的降解程度如图5中A所示,水凝胶的降解速率可随着葡萄糖浓度的升高而增高。当葡萄糖浓度为4mg/mL时,87%的水凝胶在24h内降解,而在PBS中,只有69%的水凝胶在24h小时内降解。这是由于葡萄糖竞争性的结合硼酸分子,导致水凝胶中的动态硼酸酯键裂解,水凝胶崩塌降解。体外降解结果充分说明了PL-PVA/DOP-CaCO3水凝胶具有良好的葡萄糖响应性,可作为一种可降解性且具有葡萄糖敏感特性的糖尿病伤口愈合敷料。在不同浓度葡萄糖介质中胰岛素的释放曲线如图5中B所示,在无葡萄糖PBS介质中,48小时内胰岛素的累积释放量为53.77±2.43%,可能的原因是水凝胶在PBS中发生了溶胀,孔径变大导致了胰岛素泄露。当葡萄糖浓度从0增加到4mg/mL时,48h内胰岛素的累积释放量从53.77±2.43%增加到91.91±5.27%,在三天内持续缓慢释放,且随着葡萄糖浓度的升高胰岛素的累积释放量逐渐增加,表明在高浓度葡萄糖条件下,葡萄糖竞争性的结合硼酸键使水凝胶的3D网络结构的破坏,水凝胶逐渐崩解使胰岛素持续释放,以响应高血糖水平。
利用水凝胶的细胞毒性可以评估其生物相容性,用MTT法检测PL-PVA/DOP-CaCO3水凝胶(HydrogelⅠ),PL-PVA水凝胶(HydrogelⅡ)及DOP-CaCO3微球对大鼠成纤维细胞(3T3)的细胞存活率的影响,结果如图6所示。与空白组相比,不同浓度的水凝胶的提取液在48h内的细胞存活率均在100%左右,表现出较高的细胞存活率,且hydrogelⅠ的高浓度组细胞存活率高于100%,表明高浓的的hydrogelⅠ可能促进成纤维细胞的增殖。对于DOP-CaCO3组,不同浓度的DOP-CaCO3微球在48h内的细胞存活率均高于95.0%。水凝胶的血液相容性如图6中C所示,与阳性对照组相比,各组水凝胶的溶血率远远低于阳性组(<2%),因此,各水凝胶组的溶血率可忽略不计。由此可以看出该水凝胶具有良好的生物相容性,可作为体内伤口的安全辅料材料。
检测不同葡萄糖浓度对细胞的影响,结果如图7所示,当葡萄糖浓度高于6mg/mL时,细胞存活率开始降低(93.72%),因此选取5mg/mL的葡萄糖浓度作为制造高糖损伤模型。与高糖模型组相比,与INS@HydrogelⅠ水凝胶共培养24h的3T3细胞在5mg/mL葡萄糖条件下的葡萄糖消耗增加了24.56%,而添加了高浓度胰岛素(0.32mg/mL)的3T3细胞的葡萄糖消耗仅增加了13.42%,可能的原因是高浓度的葡萄糖可使水凝胶裂解并缓慢释放胰岛素,且水凝胶包封的胰岛素更具活性,从而促进了细胞的葡萄糖消耗。细胞划痕愈合结果显示,与对照组相比,水凝胶处理的细胞在24h内逐渐愈合,这可能是由于水凝胶的性质可模拟细胞外基质(ECM),进而可以促进细胞的黏附。其中,HydrogelⅠ和INS@HydrogelⅠ组的成纤维细胞在24h内的迁移能力增加,划痕愈合明显。
效果验证例1
水凝胶的体外抗炎效果:首先,将RAW264.7巨噬细胞以每孔2×105个细胞的密度接种在6孔板中,待细胞贴壁后,加入含有1μg/mLLPS的DMEM培养基继续培养24h。然后,将RAW264.7巨噬细胞与含1μg/mLLPS和0.1g/mL水凝胶的DMEM培养基再孵育24h。最后,使用ELISA试剂盒检测每组细胞中TNF-α和IL-6的含量。
持续性高血糖是糖尿病最显著的特征,而长期持续性高血糖使慢性糖尿病伤口更容易发生长期炎症。因此,检测了细胞促炎因子TNF-α和IL-6的表达,以证明水凝胶的抗炎能力。如图8所示,经LPS诱导后,RAW264.7细胞的TNF-α和IL-6的分泌明显增高,经水凝胶处理后,TNF-α和IL-6的分泌均下调。其中,HydrogelⅠ对TNF-α的抑制分泌作用优于HydrogelⅡ,这是因为DOP-CaCO3微球的掺杂提高了凝胶的抗炎作用。与LPS组相比,INS@HydrogelⅠ组TNF-α和IL-6的分泌分别减少了48.35%和46.41%,表现出比游离胰岛素更好的抗炎作用,表明水凝胶由于胰岛素的有效释放而具有更好的抗炎活性。
水凝胶对糖尿病大鼠伤口愈合效果:采用糖尿病大鼠全层皮肤缺损模型来评估水凝胶对体内伤口的愈合过程。SD雄性大鼠,体重200-220g,首先用高糖高脂饲料喂养4周,然后向大鼠腹腔注射链脲佐菌素溶液(STZ,30mg/kgBW,溶于0.01M柠檬酸钠缓冲液,pH4.4)。STZ诱导72小时后,在尾部测量空腹血糖值,选择空腹血糖(FBG)值≥11.1mmol/L的大鼠作为T2DM模型大鼠。血糖稳定7天后建立全层皮肤缺损模型,记录SD大鼠的血糖,直至伤口完全愈合。糖尿病大鼠被随机分为五组(n≥8)。麻醉大鼠并剃毛后,在每只大鼠背部形成一个完整的皮肤缺损伤口(直径7mm)。用生理盐水冲洗后,以不同方式处理大鼠:(1)用纱布包扎伤口(纱布组,Control组),(2)用商业敷料处理伤口(左氧氟沙星凝胶敷料组,Positivecontrol组);(3)用0.4mLPL-PVA/DOP-CaCO3水凝胶(HydrogelⅠ组)处理伤口;(4)用0.4mLPL-PVA水凝胶(HydrogelⅡ组)处理伤口;(5)用0.4mL INS@PL-PVA/DOP-CaCO3水凝胶(INS@HydrogelⅠ组)处理伤口,另外设正常大鼠伤口愈合组(Normal组)。所有水凝胶完全覆盖皮肤缺陷的伤口。在愈合过程中,每48h更换一次水凝胶。使用相机记录伤口区域,期间持续监测血糖水平。在给药0、7、14、21日,分别拍摄各组大鼠创口照片并记录,测量每组创口面积,计算创口愈合速率。在给药第7、14、21日,将动物处死,以创口为中心,直径约1cm取下完整皮层组织及周围组织,并用4%多聚甲醛固定并过夜。大鼠伤口切片用H&E染色并观察拍照。
伤口愈合照片如图9所示,愈合率如图10所示。随着时间的延长,所有治疗组创面均呈现减小趋势,对照组创面恢复相对缓慢。治疗3天后,INS@HydrogelⅠ水凝胶组和左氧氟沙星凝胶敷料组的创面闭合率显著高于对照组,伤口闭合率分别达36.01%和37.89%,其它糖尿病组变化不大。7d后,HydrogelⅠ和INS@HydrogelⅠ组水凝胶创面闭合率在65~75%之间,均显著高于对照组(21.06%),并且结痂自然脱落。可能是因为INS@HydrogelⅠ在糖尿病伤口周围高浓度葡萄糖条件下,释放胰岛素,同时水凝胶发挥细胞外基质特性,及时止血并缓解炎症,促进细胞黏附和增殖,从而促进体内伤口愈合。治疗14天后所有创面均为未完全修复,左氧氟沙星凝胶敷料组伤口愈合率为96.38%,HydrogelⅠ和INS@HydrogelⅠ组伤口愈合率分别为96.18%和99.35%,显著高于对照组,伤口基本长好,并显示出结痂状态。治疗21天后,所有创面均已长好。结果表明负载胰岛素的水凝胶敷料优于商业敷料,并加速了伤口愈合。
从组织学角度评估水凝胶敷料的愈合效果,分别在第7、14、21天收集再生皮肤组织进行H&E染色,如图11所示。在给药第7天时,阳性对照组中观察到大量炎性细胞浸润的肉芽组织,在HydrogelⅠ组和INS@HydrogelⅠ组的伤口组织中有较少的炎性细胞,可能的原因是在慢性糖尿病伤口周围葡萄糖浓度较高,使水凝胶裂解释放胰岛素和多糖,从而发挥抗炎作用。在第14天,与未经处理的对照组相比,HydrogelⅠ组和INS@HydrogelⅠ组处理的伤口出现毛囊组织,且表皮逐渐增厚,与正常组伤口相似。表明生长良好。在第21天,所有组均已形成了完整的上皮,而在三个水凝胶组中都可以清楚地看皮肤附件的形成(红色箭头),表明愈合的组织与健康皮肤高度相似。综上,INS@HydrogelⅠ凝胶处理的糖尿病伤口明显优于其他组的糖尿病创面修复效果,由此可见,负载胰岛素的水凝胶可促进角质形成细胞、内皮细胞和成纤维细胞的增殖、迁移和分泌,加速伤口愈合。
以上所述的实施例仅是对本发明的优选方式进行描述,并非对本发明的范围进行限定,在不脱离本发明设计精神的前提下,本领域普通技术人员对本发明的技术方案做出的各种变形和改进,均应落入本发明权利要求书确定的保护范围内。
Claims (10)
1.一种具有葡萄糖响应性的载药水凝胶的制备方法,其特征在于,包括以下步骤:
(1)ε-聚赖氨酸和2-甲酰基苯硼酸反应得到苯硼酸的聚合物PL-PBA;
(2)CaCl2溶液和山药多糖反应后,再加入碳酸盐溶液,继续反应得到山药多糖-CaCO3微球;
(3)将所述山药多糖-CaCO3微球分散在聚乙烯醇溶液中,得到DOP-CaCO3/PVA混合液,再在所述DOP-CaCO3/PVA混合液中加入降血糖药物,混合均匀后加入所述PL-PBA,反应后得到所述载药水凝胶。
2.根据权利要求1所述的制备方法,其特征在于,在步骤(1)中,所述ε-聚赖氨酸和所述2-甲酰基苯硼酸的摩尔比大于或等于2:1。
3.根据权利要求1所述的制备方法,其特征在于,在步骤(1)中,所述反应的pH值为4.5-5.5,温度为80℃,时间为24-72h。
4.根据权利要求1所述的制备方法,其特征在于,在步骤(2)中,所述CaCl2溶液中的Ca2 +、所述碳酸盐溶液中的CO3 2-和所述山药多糖的摩尔质量比为(3-8)×105:(3-8)×105:1。
5.根据权利要求1所述的制备方法,其特征在于,在步骤(3)中,所述聚乙烯醇溶液中的聚乙烯醇与所述山药多糖-CaCO3微球的质量为20:(1-4)。
6.根据权利要求1所述的制备方法,其特征在于,在步骤(3)中,所述降血糖药物包括胰岛素。
7.一种如权利要求1-6所述的制备方法制备得到的载药水凝胶。
8.一种如权利要求7所述的载药水凝胶在制备促进糖尿病伤口愈合的药物中的应用。
9.一种促进糖尿病伤口愈合的药物,其特征在于,包括权利要求7所述的载药水凝胶。
10.根据权利要求9所述的促进糖尿病伤口愈合的药物,其特征在于,所述药物还包括药学上可接受的辅料。
