The Sanhuang eye-protecting candy tablet product prepared by the invention has good taste, easy chewing, high content of eye-protecting core component, good activity, convenient and quick eating and can be used for people with different degrees of uncomfortable symptoms such as dry eyes, eye astringents, eye swelling, eye fatigue, night blindness, mosquito and the like.

s1, extracting marigold particles by adopting a subcritical extraction method with R134a as an extraction solvent to obtain a fat-soluble extract rich in provitamin A; enriching and purifying the fat-soluble extract rich in provitamin A by adopting a medium-pressure preparation chromatographic system to obtain eluent rich in provitamin A; concentrating the eluent rich in provitamin A under reduced pressure, and vacuum freeze-drying to obtain provitamin A;
s2, taking the provitamin A obtained in the step S1 as a core material, taking embedding agent and maltodextrin as wall materials, and adding other main materials, other flavoring agents and fillers, and emulsifying, homogenizing and spray-drying to obtain microcapsule powder;
s3, adding the microcapsule powder obtained in the step S2 into a lubricant, and tabletting and forming to obtain the Sanhuang eye-protecting candy tablet.
Preferably, the marigold particles in S1 have a size of 10mm; the subcritical extraction method has the extraction time of 40-60 min, the extraction pressure of 1.0-1.2 MPa, the extraction temperature of 45-50 ℃ and the extraction times of 4 times.
Preferably, the content of provitamin A in the fat-soluble extract rich in provitamin A in S1 is more than or equal to 30%.
Preferably, the enrichment purification of the medium pressure preparative chromatography system described in S1 is: dissolving fat-soluble extract rich in vitamin A and absolute ethyl alcohol according to a mass-volume ratio of 1:5, loading into a chromatographic column, and performing linear gradient elution at a flow rate of 20mL/min, absolute ethyl alcohol as phase A, water as phase B and a detection wavelength of 445nm for 40min, wherein the initial gradient of elution is 10% B, and the elution is carried out for 40min to 90% B; the chromatographic column is Dubhe ODS-C18 medium pressure chromatographic column, and the column temperature of the chromatographic column is 30 ℃.
Preferably, the temperature of the reduced pressure concentration in S1 is 25-40 ℃, the pressure is-0.08 to-0.06 Mpa, and the rotating speed is 20-40 rpm.

3. The invention adopts marigold, carrot, pumpkin and the like which are recognized as 'eye protection golden partner', is rich in carotenoid bioactive substances such as lutein, zeaxanthin, beta-carotene and the like, and can realize the functions of maintaining normal visual functions, preventing myopia, relieving age-related eye diseases such as age-related macular degeneration, cataract and the like by quenching free radicals, filtering harmful blue light and reducing macular damage.

s1, extracting marigold particles with the size of 10mm by adopting a subcritical extraction method with R134a as an extraction solvent, wherein the extraction time of the subcritical extraction method is 60min, the extraction pressure is 1.0MPa, the extraction temperature is 47 ℃, and the extraction times are 4 times, so as to obtain a fat-soluble extract rich in provitamin A; enriching and purifying the fat-soluble extract rich in provitamin A by a chromatographic column of a medium-pressure preparation chromatographic system, wherein the enriching and purifying is as follows: dissolving 2g of fat-soluble extract rich in provitamin A with 10mL of absolute ethyl alcohol, loading into Dubhe ODS-C18 medium pressure chromatographic column, eluting with linear gradient of detection wavelength 445nm with absolute ethyl alcohol and water as phase A and phase B at flow rate of 20mL/min at 30deg.C for 40min with initial gradient of 10% B and eluting for 40min to 90% B to obtain provitamin A-rich eluent; concentrating the eluent rich in provitamin A under reduced pressure at 25 ℃, under the pressure of-0.07 Mpa and the rotating speed of 20rpm, wherein the content of solids in the concentrated solution obtained after the reduced pressure concentration is 40%, and obtaining provitamin A after vacuum freeze-drying;
s2, taking the provitamin A obtained in the step S1 as a core material, taking embedding agent and maltodextrin as wall materials, and adding other main materials, other flavoring agents and fillers, and emulsifying by adding water, homogenizing and spray drying to obtain microcapsule powder;
the homogenizing pressure is 35MPa; the content of solid in the mixed solution obtained after emulsification and homogenization is 40 percent; the peristaltic pump of spray drying has the rotating speed of 50rpm, the feeding speed of 13mL/min, the air inlet temperature of 225 ℃ and the air outlet temperature of 75 ℃; the consumption of the provitamin A is 2 parts by weight, and the other main materials are inulin, carrot powder, pumpkin powder, sea buckthorn fruit powder and blueberry fruit powder; the inulin comprises 12.5 parts by weight of inulin, 3 parts by weight of carrot powder, 4 parts by weight of pumpkin powder, 3 parts by weight of sea buckthorn fruit powder and 2 parts by weight of blueberry fruit powder; the dosage of the maltodextrin is 38 parts by weight; the dosage of the other flavoring agents is 21 parts by weight, and the other flavoring agents comprise 19 parts of sugarless sweetener, 1 part of acidulant and 1 part of flavoring agent; the total dosage of the filler and the embedding agent is 5.5 parts by weight; the water consumption of the emulsification is 30% of the total weight of the provitamin A, other main materials, maltodextrin, other flavoring agents, embedding agents and filling agents; the maltodextrin is also used as a flavoring agent;

Through detection, the content of provitamin A in the fat-soluble extract rich in provitamin A in S1 is 30.7g/100g, the purity of provitamin A is 86.7g/100g, and the content of lutein ester is 89.5g/100g; the lutein ester content in the Sanhuang eye-protecting candy tablet is 13.4mg/g.
Example 2
The preparation method of the Sanhuang eye-protecting candy tablet comprises the following steps:
s1, extracting marigold particles with the size of 10mm by adopting a subcritical extraction method with R134a as an extraction solvent, wherein the extraction time of the subcritical extraction method is 50min, the extraction pressure is 1.1MPa, the extraction temperature is 50 ℃, and the extraction times are 4 times, so as to obtain a fat-soluble extract rich in provitamin A; enriching and purifying the fat-soluble extract rich in provitamin A by a chromatographic column of a medium-pressure preparation chromatographic system, wherein the enriching and purifying is as follows: dissolving 2g of fat-soluble extract rich in provitamin A with 10mL of absolute ethyl alcohol, loading into Dubhe ODS-C18 medium pressure chromatographic column, eluting with linear gradient of detection wavelength 445nm with absolute ethyl alcohol and water as phase A and phase B at flow rate of 20mL/min at 30deg.C for 40min with initial gradient of 10% B and eluting for 40min to 90% B to obtain provitamin A-rich eluent; concentrating the eluent rich in provitamin A under reduced pressure at 35 ℃ under-0.08 Mpa at 30rpm to obtain concentrated solution with 40% of solid content, and vacuum lyophilizing to obtain provitamin A;
s2, taking the provitamin A obtained in the step S1 as a core material, taking embedding agent and maltodextrin as wall materials, and adding other main materials, flavoring agents and fillers, and emulsifying by adding water, homogenizing and spray drying to obtain microcapsule powder;
the homogenizing pressure is 45MPa; the content of solid in the mixed solution obtained after emulsification and homogenization is 45 percent; the peristaltic pump of spray drying has the rotating speed of 50rpm, the feeding speed of 15mL/min, the air inlet temperature of 230 ℃ and the air outlet temperature of 80 ℃; the consumption of the provitamin A is 2.2 parts by weight, and the other main materials are inulin, carrot powder, pumpkin powder, sea buckthorn fruit powder and blueberry fruit powder; 13.5 parts of inulin, 3 parts of carrot powder, 4 parts of pumpkin powder, 3 parts of sea buckthorn fruit powder and 2 parts of blueberry fruit powder in parts by weight; the maltodextrin is used in an amount of 40 parts by weight, the other flavoring agents are used in an amount of 22.3 parts by weight, and the other flavoring agents comprise 19 parts of sugarless sweetener, 1.8 parts of acidulant and 1.5 parts of flavoring agent; the total dosage of the filler and the embedding agent is 5 parts by weight; the water consumption of the emulsification is 35% of the total weight of the provitamin A, other main materials, maltodextrin, other flavoring agents, embedding agents and filling agents; the maltodextrin is also used as a flavoring agent;
s3, adding the microcapsule powder obtained in the S2 into a lubricant, and tabletting and forming by adopting a tablet press under the condition of 75KN pressure to obtain a Sanhuang eye-protecting candy tablet; the weight portion of the lubricant is 1.3 portions.
Through detection, the content of provitamin A in the fat-soluble extract rich in provitamin A in S1 is 32.3g/100g, the purity of provitamin A is 88.5g/100g, and the content of lutein ester is 91.3g/100g; the lutein ester content in the Sanhuang eye-protecting candy tablet is 15.1mg/g.
The three-yellow eye-protecting candy tablet prepared in the embodiment is used as a sample for crowd trial feeding experiments, and the actual effect of the product on relieving asthenopia and improving the eye health condition is evaluated.
Screening of the tested group: 30 subjects and testers with eye fatigue who voluntarily participated in the project study were selected by means of a questionnaire.
Dosage and method of use of the sample: the sample administration method comprises orally taking two tablets each day for 4 weeks.
Efficacy criterion:
(1) In Table 1 below, 3 of 8 symptoms of eye distention, eye ache, photophobia, blurred vision, dry eyes, foreign body sensation, lacrimation and general discomfort (decrease in symptom integral) were improved, and the improvement of the symptoms was judged without deterioration of the other symptoms. The effective rate (%) of symptom improvement was calculated as the number of symptom improvement cases/the number of test cases×100%.

Results of crowd feeding questionnaire:
after the Sanhuang eye-protecting candy pieces are taken for 4 weeks continuously, the symptom improvement conditions of the tested population are counted, the number of symptom improvement cases is 25, the effective rate of symptom improvement reaches 83.3%, and the Sanhuang eye-protecting candy pieces prepared by the embodiment have obvious effect of improving asthenopia.
For details, see table 2 below, the eye fatigue related data were tested and analyzed in a hospital in lanzhou, gansu, before the test and after the 4 week test.

The measurement results of the near points of the two-eye sets before and after the three-yellow eye-protection candy pieces are shown in fig. 1, the near points of the two-eye sets before the three-yellow eye-protection candy pieces are 14.12, the prompt set is insufficient, the average value of the near points of the two-eye sets after 4 weeks of use is 14.08, the three-yellow eye-protection candy pieces are reduced by 0.04 compared with the three-yellow eye-protection candy pieces before use, and the adjustment capability of test eyes after use is slightly enhanced.

The results of monocular measurement of the adjustment sensitivity before and after the three yellow eye-protecting candy pieces are shown in fig. 2, the average value of the monocular measurement of the adjustment sensitivity after the eye-protecting candy pieces are used for 4 weeks is 14.58, the adjustment sensitivity is improved by 3.37 compared with that before the use, and the monocular adjustment sensitivity is obviously enhanced after the use (P < 0.001).
The results of the binocular measurement of the adjustment sensitivity of the three yellow eye-protecting candy pieces before and after the test person uses the three yellow eye-protecting candy pieces are shown in fig. 3, the average value of the monocular measurement of the adjustment sensitivity of the eye-protecting candy pieces after 4 weeks is 14.12, the adjustment sensitivity is improved by 1.7 compared with that before the test person uses the eye-protecting candy pieces, and the adjustment sensitivity of the eyes is obviously enhanced after the test person uses the eye-protecting candy pieces (P < 0.001).

The results of the positive and negative binocular relative adjustment measurements before and after the test person used the three-yellow eye-protection candy pieces are shown in fig. 4, and the difference between the binocular relative adjustment measurements increases after the eye-protection candy pieces are used for 4 weeks, showing the enhancement of the adjustment (reserve) ability in the second test.

The test results show that the tear secretion of most of the subjects increases after the test is finished, which suggests that the tear secretion function is enhanced. Wherein, the secretion of tears can not be measured due to the shrinkage of eyeballs of the right eyes of 1 person, and 23 persons with increased secretion of the right tear eyes in 29 persons occupy 79.3% of the number of effective tested persons; 25 people with increased lacrimal secretion in left eye account for 83.3% of total subjects. The three-yellow eye-protecting candy tablet prepared by the embodiment has obvious regulating effect on preventing xerophthalmia symptoms caused by asthenopia.
In conclusion, according to a test feeding experiment, the Sanhuang eye-protecting candy tablet prepared by the embodiment has a certain eye-protecting effect.
Example 3

s1, extracting marigold particles with the size of 10mm by adopting a subcritical extraction method with R134a as an extraction solvent, wherein the extraction time of the subcritical extraction method is 40min, the extraction pressure is 1.2MPa, the extraction temperature is 45 ℃, and the extraction times are 4 times, so as to obtain a fat-soluble extract rich in provitamin A; enriching and purifying the fat-soluble extract rich in provitamin A by a chromatographic column of a medium-pressure preparation chromatographic system, wherein the enriching and purifying is as follows: dissolving 2g of fat-soluble extract rich in provitamin A with 10mL of absolute ethyl alcohol, loading into Dubhe ODS-C18 medium pressure chromatographic column, eluting with linear gradient of detection wavelength 445nm with absolute ethyl alcohol and water as phase A and phase B at flow rate of 20mL/min at 30deg.C for 40min with initial gradient of 10% B and eluting for 40min to 90% B to obtain provitamin A-rich eluent; concentrating the eluent rich in provitamin A under reduced pressure at 40 ℃, wherein the pressure is-0.06 Mpa, the rotating speed is 40rpm, the solid content in the concentrated solution obtained after the reduced pressure concentration is 40%, and obtaining provitamin A after vacuum freeze-drying;
s2, taking the provitamin A obtained in the step S1 as a core material, taking embedding agent and maltodextrin as wall materials, and adding other main materials, flavoring agents and fillers, and emulsifying by adding water, homogenizing and spray drying to obtain microcapsule powder;
the homogenizing pressure is 45MPa; the content of solid in the mixed solution obtained after emulsification and homogenization is 35 percent; the peristaltic pump of spray drying has the rotating speed of 50rpm, the feeding speed of 10mL/min, the air inlet temperature of 220 ℃ and the air outlet temperature of 70 ℃; the consumption of the provitamin A is 2.5 parts by weight, and the other main materials are inulin, carrot powder, pumpkin powder, sea buckthorn fruit powder and blueberry fruit powder; 14 parts of inulin, 4 parts of carrot powder, 5 parts of pumpkin powder, 4 parts of sea buckthorn fruit powder and 3 parts of blueberry fruit powder in parts by weight; the maltodextrin is used in an amount of 43 parts by weight, the other flavoring agents are used in an amount of 25 parts by weight, and the other flavoring agents comprise 21 parts of sugarless sweetener, 2 parts of sour agent and 2 parts of aromatic agent; the total dosage of the filler and the embedding agent is 6 parts by weight; the water consumption of the emulsification is 40% of the total weight of the provitamin A, other main materials, maltodextrin, other flavoring agents, embedding agents and filling agents; the maltodextrin is also used as a flavoring agent;
s3, adding the microcapsule powder obtained in the S2 into a lubricant, and tabletting and forming by adopting a tabletting machine under the condition of 80KN pressure to obtain a Sanhuang eye-protecting candy tablet; the weight portion of the lubricant is 2 portions.
According to detection, the content of provitamin A in the fat-soluble extract rich in provitamin A in S1 is 29.7g/100g, the purity of provitamin A is 82.3g/100g, and the content of lutein ester is 82.2g/100g; the lutein ester content in the Sanhuang eye-protecting candy tablet is 12.9mg/g.

s3, adding the microcapsule powder obtained in the step S2 into a lubricant, and tabletting and forming to obtain the Sanhuang eye-protecting candy tablet.
