Description
Technical Field
The invention belongs to the field of agricultural bactericides, and particularly relates to a bactericidal composition for preventing and treating wheat scab and application thereof.
Background
Wheat Scab (Fusarium head blight, FHB or Scab for short) is a fungal disease with strong hazard mainly caused by fusarium graminearum (Fusarium graminearum), is the most common epidemic disease in wheat producing areas in China and even worldwide, and becomes one of the most important diseases which currently restrict the wheat production safety and the wheat food quality safety in China. The disease not only causes serious loss of wheat yield, but also worsens quality of various mycotoxins contained in the disease-sensitive grains, seriously influences food safety and endangers human and animal health.
Because the wheat scab resistant germplasm resources are limited, the resistance mechanism is complex, and the variety with high scab resistance is not obtained in production, the demand of the resistant variety in production cannot be met. At present, the main measure for preventing and controlling the gibberellic disease is to carry out medicament prevention and control in the period from the wheat snapping stage to the flowering stage. Because the wheat scab pathogenic bacteria group has resistance to traditional medicaments such as carbendazim, compound preparations thereof and the like, the problem of prevention effect reduction and even prevention failure is caused, a batch of bactericides with different action mechanisms such as tebuconazole, cyhalofop-butyl, prothioconazole, fluxapyroxad and the like are developed on the market.
The perennial use of the chemical agent can cause the gibberella wheat germ to generate drug resistance to the bactericide, so that the control effect of the chemical agent is reduced, and even the control fails. In order to improve the control effect, the usage amount of chemical pesticides is increased, so that not only can chemical pesticide residues be accumulated to influence the health of human bodies, but also ecological balance can be destroyed to cause environmental pollution. With the increasing importance of people on the problems of environmental protection, ecological balance, food safety and the like, the research and development of a technology for comprehensively preventing and treating wheat scab mainly by using efficient, safe and nuisance-free microorganism compound pesticides is urgent. Biological control is safe to people and livestock, has the advantages of being green, environment-friendly, long in acting time, not easy to generate drug resistance of bacteria and the like, can reduce the use of chemical pesticides, improve the quality, and can also reduce the pollution to ecology and environment, and is increasingly popular as an environment-friendly control technology.
Bacillus amyloliquefaciens is a bacterium with high relativity with bacillus subtilis and is widely distributed in nature. The bacillus amyloliquefaciens can produce a series of metabolites with the activity of widely inhibiting fungi and bacteria in the growth process, has wide application prospect in the aspect of biological control, and gradually becomes a microorganism with the development potential of biological pesticides. The bacillus amyloliquefaciens WHIG is obtained by separating rice disease control subject components of the institute of plant protection and agricultural product quality safety of Anhui province, has broad-spectrum antibacterial activity and has good inhibition effect on various pathogenic bacteria. The strain is preserved in China general microbiological culture Collection center (China Committee for culture Collection) for 15 days 04 month 2015, and has an address of No.1, no. 3, and a preservation number of CGMCC No.10722. The strain is named WHIG, and the strain is named in a classification mode: bacillus amyloliquefaciens (cf. CN 107136120A).
The prothioconazole is a sterol demethylation inhibitor (DMIs) bactericide developed by Bayer crop science company, is mainly used for preventing and treating various diseases on grains and beans, and has the advantages of wide sterilization range, good prevention and treatment effect, obvious yield increasing effect and the like compared with the traditional triazole bactericide. In China, prothioconazole was registered in 2019 for prevention and control of wheat scab, powdery mildew and rust. The prothioconazole serving as a novel chemical component has good control effect on wheat scab, but has great resistance risk after long-term independent use.
The control strategy combining biological control and chemical control is utilized, so that the defects of slow drug effect, poor control effect and poor stability of biological control can be overcome, the application amount of chemical agents can be reduced, the effect of effectively controlling diseases can be achieved, and no related report on the combined use of bacillus amyloliquefaciens WHIG and prothioconazole exists in the prior art. The applicant researches find that the combination of bacillus amyloliquefaciens WHIG and prothioconazole has a good synergistic effect on wheat scab.
Disclosure of Invention
The invention utilizes the control strategy of combining biological control and chemical control, not only can reduce the application amount of chemical agents, but also can improve the stability of biological control, thereby achieving the aim of effectively controlling diseases, and the composition shows remarkable synergy.
The technical scheme of the invention is as follows:
the bactericidal composition for preventing and treating wheat scab is characterized by comprising bacillus amyloliquefaciens WHIG microbial inoculum and prothioconazole; the mass ratio of the bacillus amyloliquefaciens WHIG microbial inoculum to the prothioconazole is 80:1-1:8; the bacillus amyloliquefaciens WHIG is preserved in the China general microbiological culture Collection center (China Committee for culture Collection) with the address of 1 # 3 of North West Lu of the Korean area of Beijing city and the preservation number of 10722; bacillus amyloliquefaciens WHIG microbial inoculum concentration 1×10 6 -1×10 10 cfu/mL。
Preferably, the concentration of the bacillus amyloliquefaciens WHIG microbial inoculum is 1 multiplied by 10 7 -1×10 9 cfu/mL; the mass ratio of the bacillus amyloliquefaciens WHIG microbial inoculum to the prothioconazole is 50:1-5:1. Further preferably, the concentration of the bacillus amyloliquefaciens WHIG microbial inoculum is 1×10 9 cfu/mL; the mass ratio of the bacillus amyloliquefaciens WHIG microbial inoculum to the prothioconazole is 50:1-5:1.
The bactericidal composition also comprises pesticide auxiliary ingredients, and is prepared into a pesticide preparation. The pesticide preparation is suspending agent, wettable powder, water dispersible granule, aqueous emulsion and dispersible oil suspending agent. The auxiliary materials are prepared by adopting a conventional preparation method by selecting conventional auxiliary material components in the field.
The bactericidal composition can be used for preventing and controlling crop diseases, and the crop diseases are wheat scab.
By adopting the technical scheme, the invention has the following beneficial effects:
the invention utilizes the control strategy of combining biological control and chemical control, not only can reduce the application amount of chemical agents, but also can improve the stability of biological control, thereby achieving the effect of effectively controlling diseases.
When the mass ratio of the WH1G bacterial suspension to the prothioconazole is 50:1-5:1, the composition has obvious synergistic effect on wheat scab, so that the dosage of chemical agents can be effectively reduced, the generation of pathogen resistance can be delayed, and the wheat scab can be effectively prevented and treated.

2.2 test methods
And (3) manually inoculating the wheat with the gibberellic disease spore liquid at the early stage of flowering. The agents with different concentrations are uniformly sprayed on the wheat ears respectively 24 hours after inoculation. A blank control was set. Each treatment was repeated 3 times, 4 square meters per cell. Investigation was started 25d after administration. Each repeated diagonal line is sampled at 5 points, 100 ears are investigated at each point, the number of disease stages and the total ears are recorded at each point, and the grading standard is referred to the pesticide field efficacy test criterion of the agricultural industry standard: the bactericide is used for preventing and treating wheat scab (NY/T1464.14-2007) and is evaluated by the ratio (S) of the area of the disease spike to the area of the whole spike.
Disease severity grading criteria:
level 0: the whole plant is free from diseases;
stage 1: s is less than 1/4;
3 stages: s is more than or equal to 1/4 and less than 1/2;
5 stages: s is more than or equal to 1/2 and less than 3/4;
7 stages: s is more than or equal to 3/4.
Disease index = Σ [ (number of disease plants at each stage×number of relative disease stages)/(total number of investigation×number of highest disease stages) ]100
Control effect (%) = [ (disease index of blank control area-disease index of treatment area)/disease index of blank control area ] ×100
