Description

Composition with free radical removal effect and application thereof
Technical Field
The invention relates to the technical field of cosmetics, in particular to a composition with a free radical removal effect and application thereof.
Background
In the metabolic process of the organism, after one electron of oxygen is taken out, an oxygen atom without paired electrons becomes a free radical. The free radical is unstable, has strong oxidizing property and strong electron absorbing capability, and needs to abstract electrons again in cell molecules of body tissues to pair the cell molecules, and when the cell molecules push out new electrons, the cell molecules become free radicals and also need to rob electrons in cell membranes or cell nucleus molecules, so that new free radicals are continuously generated. The free radical can cause protein denaturation and crosslinking, so that a plurality of enzymes and hormones in the body lose biological activity, the immunity, the nerve reflex, the exercise capacity and other system activities of the body are reduced, and simultaneously, the nucleic acid structure can be destroyed, the whole body is caused to be abnormal in metabolism and the like, and finally, the body is diseased.
The oxygen inhaled into human body from air is converted into superoxide anion by enzyme, which is used to kill virus and other exotic invaders, and the excessive superoxide anion becomes free radical for promoting cell aging. The human body can generate endogenous antioxidant substances to catalyze and decompose superoxide anion free radicals, so that the human body reaches a safe balance state. However, with the increase of age, the endogenous antioxidant level of human body is gradually reduced, and at this time, the excessive superoxide anion free radical attacks the unsaturated phospholipids on the cell membrane, so that the cells age prematurely, pigmentation is generated, senile plaque is formed, and the human body is aged.

(3) The DPPH reagent was used to test the radical scavenging ability of the test substance in each of the test group and the control group, and each of the test group and the control group was provided with the following test system A, B, C, D (20 ul of test substance, 180ul of DPPH):
a: the volume of the substance to be detected is replaced by PBS (phosphate buffer solution) without the substance to be detected and only DPPH;
b: the volume of the substance to be detected and DPPH are not added, the volume of the substance to be detected is replaced by PBS, and the volume of the DPPH is replaced by absolute ethyl alcohol;
c: adding the substance to be detected and DPPH;
d: and no DPPH is added, only the substance to be detected is added, and the volume of DPPH is replaced by absolute ethyl alcohol.
The detection steps are carried out in a 96-well plate, at least 3 multiple wells are arranged, required objects to be detected, DPPH, PBS or absolute ethyl alcohol are added, then a row gun is used for mixing uniformly, and the mixture is placed for 30min at room temperature in a dark place.
(4) Absorbance values (0.01 mg/ml EGCG was used as a positive control in this experiment) were measured at 540nm for detection system A, B, C, D to obtain absorbance a, absorbance B, absorbance C, and absorbance D, respectively, and DPPH removal activity (%) was calculated for each experimental group and comparative group.
DPPH scavenging Activity calculation:
