本发明公开了冰川水在制备皮肤外用剂中的应用,及一种含冰川水的皮肤外用剂。本发明首次发现,冰川水可以明显促进成纤维细胞的增殖,提高皮肤成纤维细胞胶原蛋白Ⅰ和弹性蛋白的合成,显著抑制人源表皮黑色素细胞HEM‑m的增殖活性和黑色素的生成。冰川水作为皮肤外用剂中的抗衰老活性成分、美白活性成分和保湿成分中的一种或多种,因而,本发明的含冰川水的皮肤外用剂天然、安全,具有增加皮肤弹性,抗皱,修复受损皮肤,以及减少色素沉着和美白的功效。

技术领域
本发明涉及冰川水在制备皮肤外用剂中的应用及含其的皮肤外用剂。
背景技术
冰川水是地表上长期存在并能自行运动的天然冰体。由大气固体降水经多年积累而成,是地表重要的淡水资源,不同于冬季河湖冻结的水冻冰。新雪降落到地面后,经过一个消融季节未融化的雪叫粒雪。雪逐步密实、经融化再冻结,使晶体合并,晶粒改变其大小和形态,出现定向增长。当其密度达到临界值,晶粒间推动透气性和透水性,便成为冰川冰。许多国家的科学家已经对常喝冰川水有利于提高人类体质、减少疾病、延长寿命等现象进行了系列研究。
冰川水被中外科学界称为“生命之水”。而取水点的不同,深刻影响着冰川水对生命的神奇作用。纵向分析,位于雪线以下地质深层处万年老冰融水处的冰川融水纯度最高,而越往下的冰川融水,一定程度上汇聚新年份冰雪融水甚至生物影响,纯净程度不断减弱;横向分析,山之阴的冰川优于山之阳。水蒸气中较重分子在顺山体上升过程中受重力作用,不断形成降雨降雪落于山间,爬过山顶落至山阴的将是经自然过滤后最轻、最纯净的水体。
有学者研究比较了蒸馏水、自来水和祁连山“七一”冰川的冰川水的pH值、电导率。冰川水的电导率为2.31×10-2s·m-1,为自来水的1/2,与其pH值一样,介于蒸馏水和自来水之间。测定了冰川水的主要矿质元素含量,结果表明,冰川水具有很低的Na+含量及较低的矿化度(101.50mg·L-1)。通过种子萌发实验、植物组织培养的脱分化和再分化过程的比较研究,揭示冰川水的水质纯净,完全可以满足植物组织培养的要求,是地球上非常宝贵的优质水资源。(刘光琇,潘有福,陆卫.祁连山冰川水的生物学效应实验研究.冰川冻土.2004年6期)
有资料显示,一种由天然冰川水、雪莲提取液、芦笋提取液、太白米提取液、淫羊藿提取液和红景天提取液组成的保健冰川水(CN1194120A),其中配有优选的中草药,对人体及生物的健康生长极为有益,具有增加新陈代谢及抗衰老之功效,可成为人们的日常饮用水。
随着年龄的增长,皮肤会出现松弛、干燥粗糙、弹性下降、皱纹增多等老化现象,皮肤衰老主要分为自然衰老和光老化两种形式。所谓自然衰老(intrinsic aging)系指由于机体内在因素的作用(主要为遗传因素)引起,见于暴露部位和非暴露部位,明显特征为皱纹的出现和皮肤的松弛。而光老化(photoaging)则指皮肤衰老过程紫外线损害的积累,是自然老化和紫外线辐射共同作用的结果,表现为皮肤暴露部位粗燥、皱纹加深加粗、结构异常、不规则性色素沉着、血管扩张、表皮角化不良、出现异常增殖、真皮弹力纤维变性及降解产物蓄积等。
一般来说,真皮衰老表现为真皮对外来化学物清除力下降,真皮厚度变薄、胶原蛋白和弹性蛋白合成减少、分解增加,分解酶活性增强。这些现象都与成纤维细胞数减少以及分泌合成功能下降或异常有关。人皮肤成纤维细胞是皮肤真皮网织层中最重要的细胞,是皮肤衰老和细胞受损后的主要修复细胞之一。它不但能够促进表皮细胞的迁移、增殖和分化,还能分泌大量的胶原蛋白、弹性纤维蛋白及多种细胞修复因子,具有强大的自我更新能力,从而修复老化的皮肤。
就光老化中的色素沉着而言,是由皮肤上的黑色素细胞合成色素并将其转移到周围的角质形成细胞所导致的。目前,大多数的美白产品都集中在开发新因素抑制黑色素的形成。
综合上述的文献和专利调研,未见关于冰川水用于制备具有抗衰老和美白功效的皮肤外用剂的专利申请和文献。
发明内容
本发明所解决的技术问题在于提供了冰川水在制备皮肤外用剂中的应用及一种含冰川水的皮肤外用剂。本发明的含冰川水的皮肤外用剂天然、安全,具有增加皮肤弹性,抗皱,修复受损皮肤,以及减少色素沉着和美白的功效。
本发明人通过大量研究发现:1)冰川水可以明显促进成纤维细胞的增殖;2)在冰川水的培养环境下,成纤维细胞的细胞和细胞核均较大,细胞功能活动旺盛;3)冰川水可以提高皮肤成纤维细胞胶原蛋白Ⅰ和弹性蛋白的合成;4)冰川水能显著抑制人源表皮黑色素细胞HEM-m的增殖活性和黑色素的生成;5)冰川水具有良好的保湿作用。
由此,本发明提供了冰川水在制备皮肤外用剂中的应用,所述的冰川水作为所述皮肤外用剂中的抗衰老活性成分、美白活性成分和保湿活性成分中的一种或多种。较佳地,所述的冰川水同时作为所述皮肤外用剂中的抗衰老活性成分和美白活性成分;更佳地,所述的冰川水同时作为所述皮肤外用剂中的抗衰老活性成分、美白活性成分和保湿活性成分。
其中,所述抗衰老活性成分为提高皮肤成纤维细胞胶原蛋白Ⅰ和弹性蛋白的合成的抗衰老活性成分。
其中,所述美白活性成分较佳地为抑制人源表皮黑色素细胞HEM-m的增殖活性和黑色素的生成量的美白活性成分。
其中,所述冰川水的水源地可为任意地区,较佳地为喜马拉雅山。所述冰川水的取水口较佳地为多吉曲登尼玛泉。多吉曲登尼玛泉位于喜马拉雅山北坡,海拔5128米,其泉口温度常年保持在1.5-2℃。
本发明中,所述的冰川水较佳地满足下述条件:所述冰川水的pH值为7.1-7.9,所述冰川水中的氘含量为133-137ppm,且所述冰川水中不含溴酸盐。更佳地,每升所述冰川水含有下述组分:Sr+0.2-0.5mg、K+0.5-10.0mg、Na+1.0-5.0mg、Ca2+14.0-30.0以及Mg2+2.0-10.0;所述冰川水中的溶解性总固体含量为60-150mg/L;最佳地,所述冰川水的pH值为7.4,所述冰川水中的氘含量为133ppm。
具体地,所述冰川水具有促进成纤维细胞的增殖,提高皮肤成纤维细胞胶原蛋白I和弹性蛋白的合成,延缓皮肤衰老的作用;所述冰川水具有抑制人源表皮黑色素细胞HEM-m的增殖活性和抑制黑色素的生成的作用,因而具有美白皮肤的功效;所述的冰川水具有延缓皮肤水分流失的作用,因而具有皮肤保湿的功效。
本发明还提供了一种皮肤外用剂,其含有所述的冰川水,所述冰川水相对于所述皮肤外用剂的重量百分比含量大于10%,且小于等于99%。
其中,所述冰川水的水源地可为任意地区,较佳地为喜马拉雅山。所述冰川水的取水口较佳地为多吉曲登尼玛泉。多吉曲登尼玛泉位于喜马拉雅山北坡,海拔5128米,其泉口温度常年保持在1.5-2℃。
本发明中,所述的冰川水较佳地满足下述条件:所述冰川水的pH值为7.1-7.9,所述冰川水中的氘含量为133-137ppm,且所述冰川水中不含溴酸盐;每升所述冰川水含有下述组分:Sr+0.2-0.5mg、K+0.5-10.0mg、Na+1.0-5.0mg、Ca2+14.0-30.0mg以及Mg2+2.0-10.0mg;所述冰川水中的溶解性总固体含量为60-150mg/L;最佳地,所述冰川水的pH值为7.4,所述冰川水中的氘含量为133ppm。
其中,所述的皮肤外用剂中,所述冰川水相对于所述皮肤外用剂的重量百分比较佳地为67.1-87.2%。
本发明中,所述皮肤外用剂是通常用于皮肤外部的所有成分的统称概念,例如可以是化妆品或药品。所述化妆品中可以是基础化妆品、面部妆容化妆品、身体用化妆品、头部用护理品等,对其剂型无特殊限制,根据不同目的可合理选择。
本发明中,所述的皮肤外用剂还可以包含其他活性成分,所述其他活性成分较佳地为保湿活性成分、抗氧化活性成分、控油活性成分、美白活性成分和抗衰老活性成分中的一种或多种。所述其他活性成分的含量为本领域常规的含量。
其中,所述的保湿活性成分包括天然保湿活性成分和/或合成保湿活性成分,所述天然保湿活性成分可以选自:角鲨烷、霍霍巴油、蜂蜜、灵芝提取液、芦荟提取物、透明质酸、神经酰胺、丝蛋白类保湿活性成分、胶原蛋白等天然物质;所述合成保湿活性成分可以选自:多元醇类保湿成分(甘油、丙二醇、丁二醇、戊二醇、聚乙二醇、聚甘油等)、乳酸钠、葡萄糖衍生物、聚丙烯酸树脂、蓖麻油及其衍生物、甲壳素壳聚糖及其衍生物、吡咯烷酮羧酸钠等化学合成物。
其中,所述的抗氧化活性成分可以选自:黄酮类抗氧化活性成分(水飞蓟素、黄芪苷、芦丁、槲皮素、青兰苷、双氢青兰苷、芒果苷等),鞣质类抗氧化活性成分(表没有食子儿茶素、表没有食子儿茶素没食子酸酯、表儿茶素没食子酸酯、儿茶素等),醌类抗氧化活性成分(紫草素等),二肽类氨基酸(蛋氨酸、色氨酸、苯丙氨酸、脯氨酸等),糖醇类抗氧化活性成分(五碳糖、六碳糖单糖、百合多糖、果糖、糖醇等),多酚类抗氧化活性成分(谷物的胚芽、葡萄酚、迷迭香酚、表迷迭香酚、异迷迭香酚等),生物碱类抗氧化活性成分(川穹嗪、药根碱、木兰碱、海罂粟碱、潘荔枝碱等),维生素类抗氧化活性成分(维生素E、维生素C、类胡萝卜素)和植物提取物类的抗氧化活性成分,如凹顶藻提取物等。
其中,所述的控油活性成分可以选自:收敛剂(氧化锌、硫酸锌、氯化铝、硫酸铝、单宁酸、柠檬酸、乳酸等),清凉剂(酒精和薄荷),维生素类控油活性成分(维生素B6、维生素B3和维生素H)和角质剥离溶解剂(水杨酸、果酸等)。
其中,所述的美白活性成分可以选自:熊果苷及其衍生物、曲酸及其衍生物、壬二酸及其衍生物、亚油酸及其衍生物、维生素C及其衍生物,以及果酸。
其中,所述的抗衰老活性成分可以选自:维生素类抗衰老活性成分(维生素A、维生素B),酶(过氧化歧化酶SOD、谷胱甘肽过氧化酶GPO、过氧化氢酶等)和植物提取物(刺山柑芽提取物、飞机草提取物等)。
按照本领域常识,根据不同的剂型和目的,所述的皮肤外用剂中还可以含有本领域中常规使用的介质或基质赋形剂,只要所述赋形剂为化妆品或药学领域中可接受的赋形剂。所述赋形剂的含量为本领域常规的含量。
本发明中,所述赋形剂的形式可以为水相、油相、凝胶、水包蜡型乳液、水包油型乳液或油包水型乳液的形式。所述水相可以为一种或多种水溶性或分散性组分的混合物,其在室温下可以为液体、半固体或固体。本领域技术人员可以基于本领域技术人员掌握的知识选择合适的赋形剂的形式,以及其中包含的组分。
当所述赋形剂的形式为水相时,所述赋形剂较佳地包括水,或水与至少一种亲水性有机溶剂的混合物。所述的亲水性有机溶剂诸如醇,尤其是含有2-5个碳原子的直链或支链低级一元醇,如乙醇或丙醇;多元醇,如丙二醇、山梨醇、甘油、泛醇或聚乙二醇及其混合物。
当所述赋形剂的形式为乳液时,所述的皮肤外用剂较佳地还可以包括流变改进剂和/或成膜剂。所述流变改进剂可以选自:高分子增稠剂;疏水改性的丙烯酸酯共聚物,例如丙烯酸酯/Steareth-20丙烯酸甲酯共聚物、丙烯酸酯/Beheneth-25丙烯酸酯共聚物;疏水改性的乙氧基化聚胺酯;例如PEG-150/癸醇/SMDI共聚物、PEG-150/硬脂醇/SMDI;疏水改性非离子多羟基化合物,例如PEG-150二硬脂酸盐。

所述成膜剂可为本领域常规使用的成膜聚合物,只要其可溶于或分散于所述赋形剂中。所述成膜剂较佳地为下述中的一种或多种:聚氨基甲酸酯、聚丙烯酸均聚物或共聚物、聚酯、基于烃的树脂和/或硅氧烷树脂。
当所述赋形剂的形式为油相时,所述赋形剂较佳地包括在室温下为液体的油溶性或油分散性组分和/或在室温下为油状或蜡状的物质。

所述在室温下为油状或蜡状的物质较佳地为蜡和/或树胶。所述的蜡较佳地包括下述物质中的一种或多种:基于烃的蜡、氟蜡和/或硅氧烷蜡,并且可以来源于植物、矿物、动物和/或合成来源;所述的蜡更佳地包括下述物质中的一种或多种:蜂蜡、巴西棕榈蜡、小烛树蜡、石蜡、微晶蜡、地蜡、聚乙烯蜡、含有C16-45的硅氧烷蜡。所述的树胶一般为聚二甲基硅氧烷、羧甲基纤维素钠或多糖类,并且半固体物质一般为基于烃的化合物,如羊毛脂及其衍生物。
本发明的皮肤外用剂还可以进一步包含常规的用于化妆品领域中的任何其他组分,例如防腐剂、芳香剂以及亲水性和亲脂性活性剂中的一种或多种。该些其他组分的含量可为本领域常规的含量。
本发明中,所述皮肤外用剂还可以包含颗粒相,所述的颗粒相可以为化妆品组合物中使用的颜料、珠光剂和填充剂中的一种或多种。所述颗粒相的含量可为本领域常规的含量。所述颜料较佳地为无机颜料和/或有机颜料。所述无机颜料较佳地包括氧化钛、氧化锆和氧化铈、氧化锌、氧化铁和铁蓝中的一种或多种。所述有机颜料较佳地包括钡色淀、锶色淀、钙色淀和铝色淀和碳黑。所述珠光剂较佳地为涂覆了氧化钛、氧化铁或天然颜料的云母。所述填充剂较佳地包括滑石粉、二氧化硅、硬脂酸锌、云母、高岭土、尼龙粉末、聚乙烯粉末、特氟龙、淀粉、一氮化硼和共聚物微球中的一种或多种。所述的共聚物微球较佳地为硅氧烷树脂微珠。
本发明的皮肤外用剂还可以进一步包括一种或多种下述成分:抗过敏剂、抗微生物剂、抗氧化剂、螯合剂、着色剂去色素剂、润肤剂、乳化剂、表皮脱落剂、香料、保湿剂、昆虫驱避剂、润滑剂、药物活性剂、增湿剂、耐光剂、防腐剂、护肤剂、皮肤渗透增强剂、防晒剂、稳定剂、表面活性剂、增稠剂、粘度调节剂、维生素或其任意组合。该些成分的含量可为本领域常规的含量。
本发明中,所述的皮肤外用剂的形式可以为任何合适的产品形式,所述的产品形式包括,但不限于气溶胶型喷雾剂、霜剂、乳液、固体、液体、分散体、泡沫、凝胶、化妆水、摩丝、软膏、粉剂、贴剂、润发油、手按泵型喷雾剂、棒状物、面膜和湿纸巾。众所周知地,本发明的皮肤外用剂可为化妆品、皮肤病学或药物局部施用产品,其制备方法可为本领域常规的制备方法。
在本发明的较佳实施方式一中,所述的皮肤外用剂为化妆水,所述的皮肤外用剂包含下述重量百分比的组分:甘油5%,多元醇类A3-6%,甜菜碱1%,泛醇0.5%,甘草酸二钾0.05-0.1%,其他活性成分A0.11-2.0%,聚乙二醇-32 0.5%,表面活性剂A0.15-0.2%,防腐剂A0.4-0.45%,黄原胶0-0.15%,木瓜提取物0-2.0%,香料0.03%和所述冰川水84.27-87.16%;其中,所述的多元醇类A为丁二醇、1,2-戊二醇和双丙甘醇中的一种或多种;所述其他活性成分A为尿囊素、烟酰胺和透明质酸钠中的一种或多种;所述表面活性剂A为羟乙基纤维素、聚山梨醇酯-20和PEG-40氢化蓖麻油中的一种或多种;所述防腐剂A为防腐剂IS-45、羟苯甲酯和苯氧乙醇中的一种或多种。
按照本领域常识,防腐剂IS-45为美国Germall公司生产的防腐剂,其中含有丙二醇、双咪唑烷基脲、羟苯甲酯和碘代丙炔基丁基甲胺酸。
在本发明的较佳实施方式二中,所述的皮肤外用剂为精华液,所述皮肤外用剂包含下述重量百分比的组分:甘油5%,多元醇类B 3-6%,泛醇1%,甘草酸二钾0.1%,尿囊素0.1%,其他活性成分B 2.1-4.3%,表面活性剂B0.05-3.6%,防腐剂B 0.25-0.45%,黄原胶0.2-0.4%,木瓜提取物0-2.0%,丙烯酸树脂Pemulen TR-2 0.3%和香料0.05%;所述皮肤外用剂中还包含氨甲基丙醇,所述氨甲基丙醇的含量以调节至所述皮肤外用剂的pH值为5.5-7为准;所述的皮肤外用剂中,所述的冰川水补足余量至100%;其中,所述的多元醇类B为丁二醇和/或1,3-丙二醇;所述其他活性成分B为烟酰胺、抗坏血酸磷酸酯钠、生育酚乙酸酯和透明质酸钠中的一种或多种;所述表面活性剂B为羟乙基纤维素、异硬脂酸异丙酯、聚甘油-2油酸酯和聚甘油-10油酸酯中的一种或多种;所述防腐剂B为羟苯丙酯、羟苯甲酯和苯氧乙醇中的一种或多种。
按照本领域常识,丙烯酸树脂Pemulen TR-2为一种丙烯酸(酯)类/C10-30烷醇丙烯酸酯交联聚合物,由美国诺誉化工公司生产。
在本发明的较佳实施方式三中,所述的皮肤外用剂为乳液,所述皮肤外用剂包含下述重量百分比的组分:甘油5%,1,3-丙二醇5%,泛醇1%,生育酚乙酸酯0.5%,其他活性成分C 2-7.1%,聚二甲基硅氧烷2%,乳化剂A1652%,异硬脂酸异丙酯3%,油类C 2-3%,鲸蜡硬脂醇0.5%,防腐剂C 0.3%,黄胶原0.5和EDTA二钠0.1%;所述皮肤外用剂中还包含三乙醇胺,所述三乙醇胺的含量以调节所述皮肤外用剂的pH值至5.5-7为准;所述皮肤外用剂中,所述冰川水补足余量至100%;其中,所述其他活性成分C为甘草酸二钾、烟酰胺和氢化聚异丁烯中的一种或多种;所述油类C为矿油和/或异十六烷;所述防腐剂C为羟苯甲酯和/或羟苯丙酯。
在本发明的较佳实施方式四中,所述的皮肤外用剂为乳霜,所述皮肤外用剂包含下述重量百分比的组分:甘油5%,1,3-丙二醇5%,泛醇1%,生育酚乙酸酯0.5%,其他活性成分D 2-7.1%,聚二甲基硅氧烷2%,乳化剂A1652%,异硬脂酸异丙酯3%,油类D 2-8%,鲸蜡硬脂醇2-3%,防腐剂D 0.3%,丙烯酸羟乙酯/丙烯酰二甲基牛磺酸钠共聚物2%和EDTA二钠0.1%;所述皮肤外用剂中还包含三乙醇胺,所述三乙醇胺的含量以调节所述皮肤外用剂的pH值至5.5-7为准;所述皮肤外用剂中,所述冰川水补足余量至100%;其中,所述其他活性成分D为甘草酸二钾、烟酰胺和氢化聚异丁烯中的一种或多种;所述油类D为矿油和/或异十六烷;所述防腐剂D为羟苯甲酯和/或羟苯丙酯。
按照本领域常识,乳化剂A165为市售商品,其含有甘油硬脂酸酯和PEG-100硬脂酸酯。所述丙烯酸羟乙酯/丙烯酰二甲基牛磺酸钠共聚物为赛比克公司生产的牌号为SepinovEMT 10的增稠剂。
在本发明的皮肤外用剂中,较佳地,水的来源仅为所述的冰川水,即是说,所述的皮肤外用剂中不含有其他类型的水,例如超纯水或自来水。
在本发明中,所述的室温为本领域常规意义上的室温,一般指20-30℃。
在符合本领域常识的基础上,上述各优选条件,可任意组合,即得本发明各较佳实例。
本发明所用试剂和原料均市售可得。
本发明的积极进步效果在于:
本发明首次发现冰川水可以明显促进成纤维细胞的增殖,在冰川水的培养环境下,成纤维细胞的细胞和细胞核均较大,细胞功能活动旺盛,可以提高皮肤成纤维细胞胶原蛋白I和弹性蛋白的合成;冰川水能显著抑制人源表皮黑色素细胞HEM-m的增殖活性,即能有效抑制黑色素的生成;含有冰川水的皮肤外用剂具有增加皮肤弹性,抗皱,修复受损皮肤,保湿以及减少色素沉着和美白的功效。

具体实施方式
下面通过实施例的方式进一步说明本发明,但并不因此将本发明限制在所述的实施例范围之中。下列实施例中未注明具体条件的实验方法,按照常规方法和条件,或按照商品说明书选择。
下述实施例中,所使用的冰川水取自喜马拉雅山的多吉曲登尼玛泉(海拔5128米),每升的该冰川水含有下述组分:Sr+0.2-0.5mg、K+0.5-10.0mg、Na+1.0-5.0mg、Ca2+14.0-30.0mg以及Mg2+2.0-10.0mg;溶解性总固体含量为60-150mg/L;该冰川水的pH值为7.4,该冰川水中的氘含量为133ppm。
实施例1人原代成纤维细胞的培养
细胞分离:人体皮肤组织取自于上海市瑞金医院泌尿科切除的人体包皮。将手术切取的组织置于含有青霉素(100kU/L)和链霉素(100mg/L)的D-Hanks缓冲液中反复漂洗后,去除皮下脂肪。在培养皿中用眼科剪将皮肤剪成0.5cm2左右的小块并将表皮层朝下平铺在培养皿中,加入少许的1640培养基和10%的FBS于37℃,5% CO2条件下培养数周后,待成纤维细胞贴壁后取出皮肤组织。传代培养数周后细胞可用于后续实验,每3-4天更换培养基。
细胞传代:原代培养的成纤维细胞生长汇合率达90%时,向培养瓶内加入0.25%的胰蛋白酶,当胞质回缩、细胞变圆、细胞间隙增大后,加入成纤维细胞培养液终止消化,按1∶3传代至新的培养瓶中,置37℃,5%CO2,湿度90%的孵箱中培养。3~5天待细胞生长汇合率达80%以上时即可再次传代。
实施例2MTT法测定细胞的增殖活性
取实施例1中的对数生长期的成纤维细胞消化稀释,调节细胞浓度为2x104/ml。接种于含100μL培养液的96孔培养板中,每孔100μL,每次设3个复孔,分别培养4、6、8、10、12、24小时后,每孔加入50μL MTT溶液继续培养4小时,吸去培养液,每孔加150μL二甲基亚砜(Dimethyl sulfoxide或DMSO),振荡10分钟,选择490nm波长,测定各孔的光密度(OD)值。实验进行3次,结果取平均值。
用冰川水配置的1640培养基处理人原代皮肤成纤维细胞(HSF),以超纯水配置的1640培养基做对照,分别在4、6、8、10、12、24小时用MTT法检测培养液中细胞增殖情况。研究发现,与超纯水相比,冰川水可促进人皮肤成纤维细胞的增殖。24小时差异有显著性(p<0.05)。结果如图1所示。
细胞培养48小时后,在倒置显微镜下观察细胞形态,结果如图2所示。在倒置显微镜下人皮肤成纤维细胞大多呈长梭形细胞,排列成簇状。研究发现,与空白对照组比较,冰川水组成纤维细胞细胞和细胞核较大,轮廓清楚,显示细胞功能活动旺盛。
通过对比天然冰川水和超纯水对人原代成纤维细胞增殖活性以及形态学影响,说明天然冰川水可以明显促进成纤维细胞的增殖,并且在冰川水培养环境下,成纤维细胞细胞和细胞核较大,轮廓清楚,显示细胞功能活动旺盛。
实施例3实验动物和给药
老年C57bl6小鼠由上海交通大学药学院实验动物中心提供。动物生产和使用许可证号码分别为SCXK(沪)2003-0003和SYXK(沪)2007-0025。
随机选取6只小鼠,背部脱毛,每日每只老鼠背部皮肤分左右分别涂抹超纯水和冰川水各3次,连续给药一个月。
实施例4免疫荧光检测法
取实施例3中的连续给药一个月后的小鼠备用。
A.取材、石蜡包埋、切片
1)将小鼠处死后,取下皮肤组织,浸泡在含有4%多聚甲醛的PBS中,4℃固定过夜。
2)洗涤、脱水、浸蜡,用PBS洗去多聚甲醛,以70%乙醇,80%乙醇,90%乙醇,无水乙醇,二甲苯依次浸泡组织1-2小时。最后组织于融化的石蜡中浸泡1-2小时。此步可在自动化的脱水浸蜡机器中进行。
3)包埋、切片,石蜡包埋组织后切片,切片厚度以5-8um为宜。载玻片事先以多聚赖氨酸或者明胶包被,对于细胞建议使用多聚赖氨酸包被,对于组织建议使用明胶包被。
B.脱蜡水化、抗原修复、打孔等
1)脱蜡水化,以二甲苯在60℃脱蜡3次,每次30分钟,再以无水乙醇,90%乙醇,80%乙醇,70%乙醇依次室温处理,每次5分钟。
2)抗原修复,采用高压抗原修复法:
以PBS冲洗干净玻片后,加入0.01M柠檬酸盐缓冲液(pH为6.0),于高压中加热至沸腾,保温5分钟后断电,使其自行冷却到室温。
3)打孔,以含0.3%Triton X-100的PBS室温处理1小时后PBS洗三遍,如果目标蛋白在膜上,则不需要打孔。
C.封闭、孵一抗、孵二抗
1)封闭,以含5%BSA的PBS滴加在组织上室温孵育30分钟,然后甩去BSA,以PBS冲洗一遍。注意要保持BSA完全覆盖组织,防止组织干裂,最好能在湿盒中封闭,下同。
2)孵一抗,在载玻片上滴加一抗,以完全覆盖组织为宜,湿盒中4℃过夜,或者湿盒中室温1小时,然后以PBS洗三遍,每次5分钟,一抗稀释比为1:100
3)孵二抗,在载玻片上滴加FITC标记的羊抗兔二抗,以完全覆盖组织为宜,湿盒中室温1小时,然后以PBS洗三遍,每次5分钟,二抗稀释比为1:100左右。
D.镜检
在组织所在位置滴加50%甘油/PBS,其中含5μg/mL DAPI用于显示细胞核,然后加盖玻片后即可使用荧光显微镜观察。
老年小鼠皮肤组织中胶原蛋白Ⅰ和弹性蛋白的免疫荧光观察
研究结果显示,冰川水组老年小鼠皮肤组织中胶原蛋白I和弹性蛋白表达的光密度要高于超纯水对照组,结果见图3和图4;其中,图3(A)、图4(A)均为低倍DAPI染色情况下的结果图,图3(B)为低倍胶原蛋白I染色情况下的结果图,图4(B)为低倍弹性蛋白染色情况下的结果图,图3(C)、图4(C)均为高倍DAPI染色情况下的结果图,图3(D)为高倍胶原蛋白I染色情况下的结果图,图4(D)为高倍弹性蛋白染色情况下的结果图。
实施例5Western检测蛋白表达变化
取经实施例4处理过的样品备用。
1)蛋白样品制备:将提取到的蛋白加入1/5上清体积的SDS上样缓冲液和1/10总体积的二硫苏糖醇(DTT),沸水浴变性7-8分钟,低速离心30秒,可存放于-20℃保存或直接用于Western blotting。
2)SDS-PAGE电泳:充分洗净玻璃板并烘干,玻璃板对齐,垂直放置于架子上卡紧;先配10%的分离胶,加入TEMED立即摇匀后灌胶。灌胶至胶面达到绿带中间线高度后,双蒸水均匀封闭分离胶的上层。当在水面和胶面间出现一条折射线时,表明胶已凝固,倒去水层,用吸水纸轻轻将水吸干。配5%的浓缩胶,加入TEMED立即摇匀后灌胶,待胶灌满后,将梳子水平插入浓缩胶中。待浓缩胶凝固后,垂直向上轻轻地拔出梳子。组装好电泳装置,并加入1×电泳缓冲液,至少超过内侧的小玻璃板。然后使用微量加样器缓慢在孔道中加入蛋白,蛋白上样量为20μg,同时在另一孔道加入预染蛋白Marker。打开电源,调电压至60V,待跑到浓缩胶与分离胶分界处时将电压调到120V,跑胶到溴酚蓝接近胶底部时终止电泳。
3)转膜(半干法):根据所测蛋白大小与预染Marker相对应的位置切出蛋白所在的凝胶,切割六张与胶大小相同的滤纸和一张PVDF膜。将膜在甲醇中充分浸透20秒,然后将滤纸、硝酸纤维素膜和切好的胶在转移缓冲液中浸泡10分钟。在半干转膜仪的电极板上,按照3层滤纸-膜-胶-3层滤纸的顺序制作三明治夹层,注意不要有气泡产生。盖上转膜仪的上层电极板和盖子,打开电源,以恒流2mA/cm2的条件转膜40-60分钟。转膜完成后,在膜右上角切角以区分正反面。
4)免疫反应:将结合蛋白的PVDF膜浸没入盛有5%脱脂牛奶的平皿中,置于脱色摇床上室温封闭1小时。用TBST稀释一抗,制作合适大小的杂交袋,将用牛奶封闭好的膜取出,滤纸吸去多余液体,放入杂交袋中,在膜正面侧将一抗加入杂交袋,封好,4℃孵育过夜。将膜取出,TBST在脱色摇床上进行洗涤3次,每次10分钟。用相应的HRP标记的二抗室温孵育1小时,TBST洗涤3次,每次10分钟。
5)化学发光,显影,定影及凝胶图象分析:将膜与等体积的A、B混合液充分接触1分钟,之后将膜移到保鲜膜上,包好,放入胶片夹。于暗室中,将显影液和定影液均倒进塑料盘中,在红灯下取出胶片,并将胶片裁剪为适当大小,放置于膜上,关上胶片夹并开始曝光。打开胶片夹取出胶片,迅速完全浸入显影液中进行显影,当出现明显条带时立刻终止显影,立即将胶片充分浸于定影液中定影,待胶片透明即可。用自来水洗去残留定影液后晾干,将胶片进行扫描并使用scanAlyze对目标条带的分子量和净光密度值进行进一步分析。
Western-blotting结果显示,冰川水组的老年小鼠鼠皮肤组织中弹性蛋白和胶原蛋白I表达量高于超纯水对照组,结果见图5和图6。
结果显示,冰川水能提高皮肤成纤维细胞胶原蛋白和弹性蛋白的合成,延缓皮肤衰老。
实施例6DMEM(H)培养基的配制及HEM-m细胞的培养
超纯水(CK),冰川水,DMEM高糖培养基干粉(简称DMEM(H),购自于GIBCO公司,NO.12800-017),以及原代培养人源表皮黑色素细胞(HEM-m)(提取于人表皮组织,于第一代冻存,购自美国ScienCell公司)。
(1)分别用超纯水和冰川水配置DMEM(H)培养基,0.22μm无菌过滤,分别加入胎牛血清(购自GOBCO公司,NO.10099-141),配制成含10%胎牛血清(购自GIBCO公司,货号10099-141)的DMEM(H)培养基。
(2)等培养瓶中的细胞HEM-m长至80%融合状态时,倒去原来的培养液,用PBS洗涤,之后加入胰酶细胞消化液进行消化。胰酶消化过程可以将培养瓶放置在37℃培养箱中。
(3)待贴壁的HEM-m细胞漂浮在液体之中并且相互散开之时,加入2mL胎牛血清终止消化。
(4)吹打并收集细胞,1000r/m,离心3分钟,小心弃去上清液,加入PBS重新悬浮细胞,注意尽量使细胞呈相互分离状态。
(5)用含10%胎牛血清的超纯水配制的DMEM(H)培养基进行传代。
(6)将传代好的细胞置于37℃,5% CO2恒温培养箱中进行培养。
实施例7人表皮黑色素细胞HEM-m增殖抑制率测定
采用MTT法,待细胞培养瓶中HEM-m细胞长至80%融合度时,胰酶消化并细胞计数,吸取细胞,并调整细胞浓度为6×104个/mL,充分混匀,取100μL接入96孔板的孔中。每组设置6个复孔,37℃孵育72小时后。将5×MTT用稀释液稀释为1×MTT。每孔加入50μL 1×MTT,在37℃孵育4小时。吸出上清液,每孔加入150μL的二甲基亚砜(DMSO),放置在平板摇床上10分钟充分摇匀,摇匀后用酶标仪在490nm处检测每孔的光密度(OD490)。
MTT法检测人源表皮黑色素细胞HEM-m增殖活性,结果如图7所示。从图7可以看出,与对照组(CK)相比,冰川水能显著抑制HEM-m细胞增殖活性,图中*号表示与CK比较p<0.05。
结果表明,相比超纯水,冰川水能显著抑制人源表皮黑色素细胞HEM-m的增殖活性。
实施例8人源表皮黑色素细胞HEM-m黑色素含量测定
采用改良的Hideya Ando法,选择对数生长期的选择对数生长期的B-16黑素瘤细胞,以2×104个/mL的密度接种于6孔培养板,每孔2.5mL,置于37℃,5%CO2饱和湿度环境中进行培养。分别用超纯水、冰川水配制的含10%胎牛血清的DMEM(H)培养基培养HEM-m细胞72小时后,分别收集细胞,调整细胞浓度到3×105个/mL,吸取1mL细胞悬液分别置于离心管中,离心去上清,用200μL的PBS缓冲液重悬细胞沉淀后,加入1mL的1:1乙醇乙醚液,室温下放置30分钟,3000r/min离心5分钟后弃上清。加入含10%二甲基亚砜(DMSO)的1mol/L的NaOH溶液,80℃水浴45分钟后于470nm波长处测定吸光度值(OD470)。每组设三个平行对照。
HEM-m细胞黑色素含量的测定结果如图8所示,从图中可以看出,冰川水能显著抑制HEM-m细胞黑色素的生成,图中*号表示与CK比较p<0.05。
结果表明,相比超纯水,冰川水能显著抑制人源表皮黑色素细胞HEM-m的生成量。
实施例9冰川水的保湿活性
测试样品:含5%甘油的去离子水,含5%甘油的冰川水。
受试人群:年龄25-30岁年龄段的女性6名。
6名受试者在恒温恒湿的稳定环境的实验室内,露出待测手前臂静坐15分钟。用水分测试探头测量已被标尺标记部位皮肤的水分初始值,再将不同的待测样品均匀涂抹在标尺标记部位。让受试者在实验室静坐3小时,并于30分钟、60分钟、90分钟、120分钟、180分钟用水分测试探头测量被标记部位的水分值,测试结果见图9。
图9中,点划线A代表的是含5%甘油的去离子水水,而虚线B代表的是含5%甘油的冰川水。从图9中可以看出,在同等条件下,冰川水相比去离子水表现出了更好的保湿性。

Claims (5)
1.冰川水作为抗衰老活性成分在制备皮肤外用剂中的应用;所述抗衰老活性成分为提高皮肤成纤维细胞胶原蛋白Ⅰ和弹性蛋白的合成的抗衰老活性成分;
所述冰川水的取水口为多吉曲登尼玛泉;
所述冰川水的pH值为7.1-7.9,所述冰川水中的氘含量为133-137ppm,且所述冰川水中不含溴酸盐;
每升所述冰川水含有下述组分:Sr+0.2-0.5mg、K+0.5-10.0mg、Na+1.0-5.0mg、Ca2+14.0-30.0mg以及Mg2+2.0-10.0mg;所述冰川水中的溶解性总固体含量为60-150mg/L。
2.一种皮肤外用剂,其特征在于,其含有冰川水,所述冰川水相对于所述皮肤外用剂的重量百分比含量大于10%,且小于等于99%;所述冰川水为权利要求1中所述的冰川水。
3.如权利要求2所述的皮肤外用剂,其特征在于,所述的皮肤外用剂包含其他活性成分,所述其他活性成分为保湿活性成分、抗氧化活性成分、控油活性成分、美白活性成分和抗衰老活性成分中的一种或多种;
和/或,所述的皮肤外用剂含有赋形剂;所述赋形剂的形式为水相、油相、凝胶、水包蜡型乳液、水包油型乳液或油包水型乳液的形式;
和/或,所述的皮肤外用剂包含防腐剂、芳香剂以及亲水性和亲脂性活性剂中的一种或多种;
和/或,所述皮肤外用剂包含颗粒相,所述的颗粒相为颜料、珠光剂和填充剂中的一种或多种。
4.如权利要求3所述的皮肤外用剂,其特征在于,当所述的皮肤外用剂为化妆水时,所述的皮肤外用剂包含下述重量百分比的组分:甘油5%,多元醇类A3-6%,甜菜碱1%,泛醇0.5%,甘草酸二钾0.05-0.1%,其他活性成分A0.11-2.0%,聚乙二醇-32 0.5%,表面活性剂A0.15-0.2%,防腐剂A0.4-0.45%,黄原胶0-0.15%,木瓜提取物0-2.0%,香料0.03%和所述冰川水84.27-87.16%;其中,所述的多元醇类A为丁二醇、1,2-戊二醇和双丙甘醇中的一种或多种;所述其他活性成分A为尿囊素、烟酰胺和透明质酸钠中的一种或多种;所述表面活性剂A为羟乙基纤维素、聚山梨醇酯-20和PEG-40氢化蓖麻油中的一种或多种;所述防腐剂A为防腐剂IS-45、羟苯甲酯和苯氧乙醇中的一种或多种;
当所述的皮肤外用剂为精华液时,所述皮肤外用剂包含下述重量百分比的组分:甘油5%,多元醇类B 3-6%,泛醇1%,甘草酸二钾0.1%,尿囊素0.1%,其他活性成分B 2.1-4.3%,表面活性剂B 0.05-3.6%,防腐剂B 0.25-0.45%,黄原胶0.2-0.4%,木瓜提取物0-2.0%,丙烯酸树脂Pemulen TR-2 0.3%和香料0.05%;所述皮肤外用剂中还包含氨甲基丙醇,所述氨甲基丙醇的含量以调节至所述皮肤外用剂的pH值为5.5-7为准;所述的皮肤外用剂中,所述的冰川水补足余量至100%;其中,所述的多元醇类B为丁二醇和/或1,3-丙二醇;所述其他活性成分B为烟酰胺、抗坏血酸磷酸酯钠、生育酚乙酸酯和透明质酸钠中的一种或多种;所述表面活性剂B为羟乙基纤维素、异硬脂酸异丙酯、聚甘油-2油酸酯和聚甘油-10油酸酯中的一种或多种;所述防腐剂B为羟苯丙酯、羟苯甲酯和苯氧乙醇中的一种或多种;

5.如权利要求2-4中任一项所述的皮肤外用剂,其特征在于,在所述的皮肤外用剂中,水的来源仅为所述的冰川水。
