技术领域
本发明涉及山羊精液保存技术领域,尤其涉及褪黑素作为奶山羊常温精液保存制备方法。
背景技术
人工授精是动物遗传改良计划中有价值且必不可少的工具,并且山羊精液保存的成功对于奶山羊人工繁殖技术的使用至关重要。然而,温度降低会对精子细胞造成严重损害。最近对其他物种的研究表明,如果能降低精子免受脂质过氧化(代谢未改变的副作用)风险的增加,那么常温和低温的精液保存是一种可行的替代方法。目前的羊的人工授精用到的主要方式也主要是用鲜精常温稀释使用和低温保存运输使用。
山羊精液的保存在主要分为冷冻保存(-196℃液氮保存)常温保存(15℃-25℃)与低温保存(4℃),常温保存采用的原理是通过弱酸环境抑制精子活动和代谢,减少能量的消耗。而低温保存采用的原理是采用较低的温度使精子处于较低的代谢水平,并在稀释液中添加一定糖分给精子提供能量,需要精子复苏时再将其保存温度缓慢升至37℃。目前山羊精液的保存技术与猪、牛的精液保存质量还存在一定差距,并且存在效果不稳定,母羊配种后受胎率低等问题。尽管很多研究人员在精液稀释保存液上做了很多研究,比如稀释液配方的优化和稀释液保存程序的优化,但是该过程仍然会导致精子超微结构的改变和精子配种受胎率下降。近期报道在一些物种的精液中添加某些促生存因子可以改善精子的保存质量,例如L-肉碱、丙酮酸盐等。因此,有必要研究这些因素在常温维持精子功能的同时,将脂质过氧化作用降至最低并降低ROS的作用,所以,筛选良好的精液稀释液配方对于精液保存质量是至关重要的。
为此,我们提出褪黑素作为奶山羊常温精液保存制备方法。
发明内容
本发明采用了如下技术方案:
褪黑素作为奶山羊常温精液保存制备方法,包括以下步骤:S1、选取原材料;S2、配置基础稀释液;S3、配置褪黑素溶液;S4、基于步骤2与步骤3对山羊精液进行保存。
作为本发明中更进一步的,步骤1中,S1-1、山羊精液保存液原材料包括:柠檬酸、果糖、葡萄糖、三羟甲基氨基甲烷、柠檬酸三钠、青霉素、链霉素、双蒸水。
作为本发明中更进一步的,S1-1-1、原材料具体重份:每100ml包含柠檬酸14g、果糖4g、葡萄糖6g、三羟甲基氨基甲烷27g、柠檬酸三钠2g、青霉素50万单位、链霉素50万单位、双蒸水。
作为本发明中更进一步的,步骤2中,S2-1、称取步骤1-1-1中柠檬酸、果糖、葡萄糖、三羟甲基氨基甲烷与柠檬酸三钠相关组分并倒在在烧杯中。
作为本发明中更进一步的,S2-2、用双蒸水定容至1L,而后加入50万单位青霉素与50万单位链霉素,溶解稀释液成分时可放置于37摄氏度水浴锅中加速溶解。

作为本发明中更进一步的,S4-3、精液稀释完成后添加褪黑素溶液,并将其分装在灭好菌的10ml离心管里,并用18层纱布包裹,包裹后放置在恒温箱中,温度设置为17℃,保存期间每隔24h将精子轻轻摇动,防止精子沉淀在底部。
与现有技术相比,本发明的有益效果为:
本发明采用的褪黑素,能在山羊精液常温保存中提高精液的保存质量,能在第3天活率和活力达到0.6以上,在山羊精液常温保存液中添加褪黑素后,可有效提高精子的质膜完整率和顶体完整率,可以有效缓解氧化应激对精子的损害;
另外本发明提供褪黑素作为奶山羊常温保存稀释液中唯一的抗氧化剂成分和自由基清除成分以及精液保存质量提高的有效成分,无需添加其他活性成分。

下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。
实施例一
褪黑素作为奶山羊常温精液保存制备方法,包括以下步骤:S1、选取原材料:
S1-1、山羊精液保存液原材料包括:柠檬酸、果糖、葡萄糖、三羟甲基氨基甲烷、柠檬酸三钠、青霉素、链霉素、双蒸水。
S1-1-1、原材料具体重份:每100ml包含柠檬酸14g、果糖4g、葡萄糖6g、三羟甲基氨基甲烷27g、柠檬酸三钠2g、青霉素50万单位、链霉素50万单位、双蒸水;
S2、配置基础稀释液:
S2-1、称取步骤1-1-1中柠檬酸、果糖、葡萄糖、三羟甲基氨基甲烷与柠檬酸三钠相关组分并倒在在烧杯中。

实施例二
基于实施例一,常温保存时,测定奶山羊精子的活率,活力,质膜完整率,顶体完整率,评定褪黑素对山羊精液的影响,其中设计五组用于奶山羊精液常温保存的配方,其中基础液中添加褪黑素的浓度分别为0.05g/L,0.07g/L,0.10g/L,0.15g/L,测定方法包括以下步骤:
S1、精液活力的检测:
采用精液自动辅助分析系统(南京松景天伦)检测精子活率和活力。从17℃取出精液样品,从离心管中取10ul精液滴在载玻片上,同时盖上盖玻片,放在37℃加温板上加热1分钟,可统计活力和活率的实验数据,每次试验选取3个视野统计数据测定,结果取平均值;
S2、精子质膜完整率的测定:
准确称取柠檬酸钠0.735g与果糖1.35g,溶于双蒸水中,并定容到100ml,温度为4℃待用,检测时分别将各组保存的稀释液0.1ml和0.9ml低渗溶液混合,而后置于37℃水浴锅中水浴30min;
水浴时轻微晃动试管避免精子沉淀,水浴结束后,吸取10ul样品滴加载玻片上制片,显微镜下统计弯尾精子与正常精子的比率,共观察3个视野,每个视野不少于200个精子;
S3、精子顶体完整率的测定:
采用异硫氰荧光素酶标记的花生凝集素(FITC-PNA)方法来测定精子顶体的完整性。精子顶体的完整率=顶体完整的精子/计数精子总数×100%,检测时取样品30ul均匀涂抹于专用多聚赖氨酸载玻片上自然风干,风干后的载玻片置于无水甲醇中固定10min,自然风干;
最后取30ulFITC-PNA染液均匀涂抹于风干精子上保证全覆盖样品,放于恒温箱内37℃避光孵育30min;
统计时随机选择3个视野,每个视野不少于200个精子,其中精子顶体呈明亮绿色荧光的为顶体完整精子,前部荧光残缺的为顶体破损精子;
S4、测定结果见如下表1、表2、表3所示:

根据实施例操作步骤,测定奶山羊精子活力,质膜完整率,顶体完整率等指标;
如表1所示,在奶山羊常温稀释液中添加褪黑素能显著提高精子保存第三天和第五天的活力(p<0.05),并且最适浓度为0.1g/L;
如表2所示,在奶山羊常温稀释液中添加褪黑素能显著提高精子保存第三天和第五天的精子顶体完整率(p<0.05),并且最适浓度为0.1g/L;
如表3所示,在奶山羊常温稀释液中添加褪黑素能显著提高精子保存第三天和第五天的质膜完整率(p<0.05),并且最适浓度为0.1g/L。

Claims (10)
1.褪黑素作为奶山羊常温精液保存制备方法,包括以下步骤:S1、选取原材料;S2、配置基础稀释液;S3、配置褪黑素溶液;S4、基于步骤2与步骤3对山羊精液进行保存。
2.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,步骤1中,S1-1、山羊精液保存液原材料包括:柠檬酸、果糖、葡萄糖、三羟甲基氨基甲烷、柠檬酸三钠、青霉素、链霉素、双蒸水。
3.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,S1-1-1、原材料具体重份:每100ml包含柠檬酸14g、果糖4g、葡萄糖6g、三羟甲基氨基甲烷27g、柠檬酸三钠2g、青霉素50万单位、链霉素50万单位、双蒸水。
4.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,步骤2中,S2-1、称取步骤1-1-1中柠檬酸、果糖、葡萄糖、三羟甲基氨基甲烷与柠檬酸三钠相关组分并倒在在烧杯中。
5.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,S2-2、用双蒸水定容至1L,而后加入50万单位青霉素与50万单位链霉素,溶解稀释液成分时可放置于37摄氏度水浴锅中加速溶解。
6.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,S2-3、溶解后将稀释液用0.22μm滤器过滤一遍,配置完成后用封口膜密封,放置于4℃冰箱待用。
7.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,步骤3中,S3-1、购买20mg褪黑素标准品,用10ml超纯水溶解后,配置为2mg/ml的褪黑素溶液,并分装成多个小管,并在-20摄氏度保存。
8.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,步骤4中,S4-1、使用假阴道法采集山羊精液,山羊选年龄集中在2-3岁,并且性欲旺盛无特殊疾病;
收集好的精液在光学显微镜下观察其活力,选择气味颜色正常的精液。
9.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,S4-2、稀释精液,按照精液:稀释液为1:8的比例稀释,具体的是稀释分为两步法,首先按照精液:稀释液1:1的比例进行等温稀释,稀释时,应将稀释液沿着管壁缓慢加入,轻轻摇匀,待平衡1分钟后,再将余下稀释液缓慢加入再轻轻摇匀,两步中的稀释液与精液都应保持等温。
10.根据权利要求1所述的褪黑素作为奶山羊常温精液保存制备方法,其特征在于,S4-3、精液稀释完成后添加褪黑素溶液,并将其分装在灭好菌的10ml离心管里,并用18层纱布包裹,包裹后放置在恒温箱中,温度设置为17℃,保存期间每隔24h将精子轻轻摇动,防止精子沉淀在底部。
