技术领域
本发明涉及表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及CCL19的免疫活性细胞、含有所述免疫活性细胞的抗癌剂、用于制备所述免疫活性细胞的表达载体。
背景技术
癌症是全世界罹患者众多的疾病,通常广泛采用化学疗法、放射线疗法或外科疗法。然而,存在产生副作用、部分功能丧失、难以对转移进行治疗等多种问题。
因此,为了进一步维持患者高的生活质量(QOL),近年来持续进行了免疫疗法的开发。在该免疫疗法中,免疫细胞疗法为从患者采集免疫活性细胞、将所述免疫活性细胞以提高其免疫功能的方式进行处置并扩增后再度移入患者的疗法。具体地,已知从患者采集T细胞、向所述T细胞中导入编码CAR的基因并扩增后再度移入患者的疗法(参见非专利文献1)。该疗法目前正在全世界进行临床试验,已获得了其在白血病、淋巴瘤等造血器官恶性肿瘤等中显示有效性的结果。
另外,作为T细胞等免疫活性细胞的免疫功能调控因子,已知细胞因子、趋化因子、信号调控蛋白质等至少数百种因子。其中,白细胞介素7(IL-7)是对于T细胞的存活而言必要的细胞因子,已知其由骨髓、胸腺、淋巴器官/组织的基质细胞等非造血细胞所产生。作为利用所述IL-7的功能的T细胞,已公开了表达融合IL-7和IL-7Rα而成的嵌合细胞因子受体的T细胞(参见专利文献1)。然而,所述T细胞中的嵌合细胞因子受体作为一种融合蛋白质,仅限于在所导入T细胞的膜表面表达,只是对于自身的细胞以非配体依赖性的方式传导IL-7R等细胞因子信号,而无法提高未导入上述受体的T细胞的功能。
另外,已公开了SIRPα变异小鼠的脾脏中T细胞区维持缺陷的原因是CCL19、CCL21、IL-7的表达降低(参见非专利文献2),CCL19、CCL21、IL-7在次级淋巴组织(脾脏、淋巴结)中具有维持T细胞的稳态的作用(参见非专利文献3)。然而,上述非专利文献2、3显示的是对于以稳态存在于次级淋巴组织的T细胞区的非活化T细胞的作用,而并没有显示与抗肿瘤免疫应答的直接相关性。并且,上述非专利文献2、3中的CCL19、CCL21、IL-7表达细胞并非T细胞,而是存在于次级淋巴组织中的网状内皮系统的细胞。
另一方面,T细胞受体(T cell receptor,以下也称为“TCR”)是在T细胞的细胞膜上表达的抗原受体分子。已知其以包含α链和β链、或γ链和δ链的异二聚体的形式存在,通过识别已与主要组织相容性基因复合体(MHC)分子结合的抗原分子而使T细胞活化。
目前正在开发对所述TCR的功能加以应用、将可识别在癌细胞中表达的肿瘤抗原的TCR基因导入获自癌症患者的T细胞中并在扩增后再度移入患者的免疫疗法。具体地,已公开了一种脊膜瘤治疗用药物组合物,其含有表达特异性识别WT1表达细胞的TCR的细胞(参见专利文献2)。
在上述技术的一部分中,尽管某些情况下观察到对于造血器官恶性肿瘤的抗肿瘤效果,但尚无对实体瘤显示显著效果的实例。对此,考虑到所移入的免疫活性细胞在生物体内的存活效率低,或者由所移入的免疫活性细胞诱导的内源性免疫活性细胞的活化、向肿瘤局部的集聚不充分这样的问题,需要开发解决上述问题的技术。

发明内容
发明要解决的课题
在用于现有免疫疗法的免疫活性细胞中,未充分地增强内源性免疫活性细胞的免疫诱导效果、免疫活性细胞的增殖能力、存活能力、或T细胞的集聚能力。因此,本发明的课题在于提供在免疫活性细胞中表达免疫活性细胞的免疫功能调控因子且兼具增殖能力、存活能力、及T细胞的集聚能力的免疫活性细胞,以及提供用于制备所述免疫活性细胞的免疫功能调控因子表达载体。
用于解决课题的手段
本申请的发明人以在利用免疫活性细胞的癌症免疫疗法中实现更优异的免疫诱导效果和抗肿瘤活性为目的,试图改良表达免疫功能调控因子的细胞。在这一过程之中,着眼于作为调控免疫活性细胞的免疫功能的因子的细胞因子、趋化因子、信号调控蛋白质,构建了表达调控前述免疫活性细胞的免疫功能的因子的载体。将所述表达载体导入免疫活性细胞中时,发现可由此制备具有比现有免疫活性细胞更优异的免疫诱导效果、增殖能力、存活能力以及T细胞集聚能力的免疫活性细胞,从而完成了本发明。
即,本发明如以下的(1)~(9)所公开。
(1)免疫活性细胞,其表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及CCL19。
(2)如上述(1)所述的免疫活性细胞,其特征在于,特异性识别癌抗原的细胞表面分子为特异性识别癌抗原的T细胞受体。
(3)如上述(1)或(2)所述的免疫活性细胞,其特征在于,免疫活性细胞为T细胞。
(4)如上述(1)~(3)中任一项所述的免疫活性细胞,其特征在于,癌抗原为WT1、MART-1、NY-ESO-1、MAGE-A1、MAGE-A3、MAGE-A4、磷脂酰肌醇蛋白聚糖-3(Glypican-3)、KIF20A、存活蛋白(Survivin)、AFP-1、gp100、MUC1、PAP-10、PAP-5、TRP2-1、SART-1、VEGFR1、VEGFR2、NEIL3、MPHOSPH1、DEPDC1、FOXM1、CDH3、TTK、TOMM34、URLC10、KOC1、UBE2T、TOPK、ECT2、间皮素(MESOTHELIN)、NKG2D、P1A、GD2、或GM2。

发明的效果
若使用本发明的表达特异性识别癌抗原的细胞表面分子、IL-7、及CCL19的免疫活性细胞(以下也称为“IL-7×CCL19表达免疫活性细胞”),则具有抗肿瘤活性,可抑制由具有细胞表面分子特异性识别的抗原的癌细胞形成的肿瘤所导致的存活率下降。另外,若使用本发明的表达载体,则可制备兼具增殖能力、存活能力及T细胞集聚能力的免疫活性细胞。

具体实施方式
本发明的IL-7×CCL19表达免疫活性细胞只要表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及CCL19即可,没有特别限制,也可以还表达IL-15、CCL21、IL-2、IL-4、IL-12、IL-13、IL-17、IL-18、IP-10、CCL4、Flt3L、干扰素-γ、MIP-1α、GM-CSF、M-CSF、TGF-β、TNF-α等其他免疫功能调控因子。
癌抗原是指在癌细胞中较之正常细胞更高水平地表达、或者在癌细胞中特异性表达的蛋白质、糖脂质等物质,作为所述癌抗原,可举出肿瘤相关抗原、癌睾丸抗原、血管新生相关抗原、因基因变异而产生的癌症新生抗原(新抗原)的表位肽,具体可举出WT1、MART-1、NY-ESO-1、MAGE-A1、MAGE-A3、MAGE-A4、磷脂酰肌醇蛋白聚糖-3、KIF20A、存活蛋白、AFP-1、gp100、MUC1、PAP-10、PAP-5、TRP2-1、SART-1、VEGFR1、VEGFR2、NEIL3、MPHOSPH1、DEPDC1、FOXM1、CDH3、TTK、TOMM34、URLC10、KOC1、UBE2T、TOPK、ECT2、间皮素、NKG2D、P1A等蛋白质、GD2、GM2等糖脂质,但不限于此。
作为特异性识别癌抗原的细胞表面分子,可举出特异性识别癌抗原的细胞表面受体、人工受体、粘附因子,可优选举出特异性识别癌抗原的T细胞受体、特异性识别癌抗原的嵌合抗原受体(CAR)等通过在细胞表面表达而赋予特异性识别癌症的能力的分子,可更优选举出TCR。作为TCR,可以为特异性识别癌抗原的包含α链及β链的异二聚体(α·βTCR)或包含γ链及δ链的异二聚体(γ·δTCR)。需要说明的是,特异性识别癌抗原的细胞表面分子若对癌抗原的识别是特异的,则也可以间接地识别。例如,与本发明的免疫活性细胞同时或连续地向对象施予特异性识别癌抗原的抗体等分子,通过识别所述抗体等分子或识别抗体等分子上所标记的标签,由此本发明的免疫活性细胞可间接地对癌抗原进行特异性识别。作为识别抗体的情况的实例,可举出作为细胞表面分子的CD16等,作为对抗体等的分子进行标记的标签的实例,可举出FITC等。
作为本发明的IL-7×CCL19表达免疫活性细胞中的免疫活性细胞的种类,只要是参与免疫应答的细胞即可,可举出T细胞、自然杀伤细胞(NK细胞)、B细胞等淋巴细胞系细胞,单核细胞、巨噬细胞、树突状细胞等抗原呈递细胞,嗜中性粒细胞、嗜酸性粒细胞、嗜碱性粒细胞、肥大细胞等粒细胞;可优选举出来自人、犬、猫、猪、小鼠等哺乳动物的T细胞(优选来自人的T细胞)。另外,T细胞可从血液、骨髓液等体液,脾脏、胸腺、淋巴结等组织,或者浸润于原发肿瘤、转移性肿瘤、癌症性腹水等癌症组织中的免疫细胞分离、纯化从而得到,另外也可利用由ES细胞、iPS细胞制备得到的T细胞。作为所述T细胞,可举出α·βT细胞、γ·δT细胞、CD8+T细胞、CD4+T细胞、肿瘤浸润T细胞、记忆T细胞、初始T细胞、NKT细胞。
作为本发明的IL-7×CCL19表达免疫活性细胞的制备方法,可举出将下文所述的本发明的表达载体导入免疫活性细胞中而制备的方法。或者,也可列举下述方法:对于受精卵、ES细胞、或iPS细胞,将表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及/或CCL19的载体导入后进行诱导而制备的方法;向从通过基因导入而表达了特异性识别癌抗原的细胞表面分子的转基因哺乳动物分离得到的免疫活性细胞中,进一步根据需要导入表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及/或CCL19的载体,由此进行制备的方法。
作为将下文所述的本发明的表达载体导入上述免疫活性细胞中而制备的方法,没有特别限定,可列举通过病毒感染法、磷酸钙法、脂质体转染法、显微注射法、电穿孔法等已知方法导入的方法,可优选举出通过病毒感染法导入的方法。
作为病毒感染法,可举出以下方法:使本发明的表达载体与包装质粒转染至GP2-293细胞(TAKARA BIO公司制造)、Plat-GP细胞(COSMOBIO公司制造)、PG13细胞(ATCC CRL-10686)、PA317细胞(ATCC CRL-9078)等包装细胞,制备重组病毒,将所述重组病毒感染至免疫活性细胞。可使用Retrovirus packagin Kit Eco(TAKARA BIO公司制造)等市售试剂盒进行。
另外,本发明的免疫活性细胞可通过如下方法制备:使用已知的基因编辑技术,以在适当的启动子的调控下能够表达的方式,将包含编码特异性识别癌抗原的细胞表面分子、IL-7、及CCL19的碱基序列的多聚核苷酸并入细胞的基因组中。作为已知的基因编辑技术,可举出使用锌指核酸酶、TALEN(类转录活化效应子核酸酶)、CRISPR(规则间隔的短小重复回文序列,Clustered Regularly Interspaced Short Palindromic Repeat)-Cas系统等内切核酸酶的技术。在于本发明的免疫活性细胞中表达其他外来蛋白质的情况下,也可同样地使用基因编辑技术,以在适当启动子的调控下能够表达的方式将包含编码其他外来蛋白质的碱基序列的多聚核苷酸并入细胞的基因组中。作为以在适当的启动子的调控下能够表达的方式将多聚核苷酸并入细胞基因组的方法,可列举:将在适当启动子的下游功能性地连接有编码特异性识别癌抗原的细胞表面分子、IL-7、及CCL19(或其他蛋白质)的碱基序列的多聚核苷酸(即,以在所述启动子的调控下能够表达的方式连接有编码序列的多聚核苷酸)并入细胞基因组的非编码区等中的方法;将包含编码特异性识别癌抗原的细胞表面分子、IL-7、及CCL19(或其他蛋白质)的碱基序列的多聚核苷酸并入细胞基因组的内源性启动子的下游的方法等。作为内源性启动子,例如可举出TCRα、TCRβ等启动子。
另外,也可使本发明的IL-7×CCL19表达免疫活性细胞中表达下述单纯疱疹病毒的胸苷激酶(HSV-TK)、诱导性胱天蛋白酶9(inducible caspase 9)。
本发明的IL-7×CCL19表达免疫活性细胞由于表达特异性识别癌抗原的细胞表面分子、IL-7、及CCL19,因此增殖能力、存活能力、及内源性T细胞的集聚能力强,可应用于各种使用免疫活性细胞的过继免疫疗法中。作为过继免疫疗法的实例,可举出树突状细胞疗法、NK细胞疗法、γ·δT细胞疗法、α·βT细胞疗法、CTL疗法、TIL疗法等,但不限于此。可列举如下方法:向自患者采集的免疫活性细胞中导入下述本发明的表达载体进行扩增,并施予患者。以下举出了具体的实例,但不限于此。树突状细胞疗法包括以下步骤:向由自患者采集的单核细胞分化的树突状细胞中导入通过手术摘出的癌症组织或其溶解物,并向患者体内施予的步骤;也可包括将本发明的表达载体导入树突状细胞的步骤。在此,也可以人工合成并利用癌抗原分子的表位肽来代替上述癌症组织、溶解物。NK细胞疗法包括以下步骤:针对自患者采集的淋巴细胞而利用IL-2等多种刺激物质对NK细胞进行活化并增殖、并将其施予患者的步骤;也可包括将本发明的载体导入NK细胞的步骤。需要说明的是,通过将针对癌症的抗体药物与经活化的NK细胞联用,可期待高效率地攻击癌细胞的效果。γ·δT细胞疗法包括以下步骤:利用IL-2或唑来膦酸对自患者采集的淋巴细胞进行培养刺激,使γ·δT细胞增殖,并将其施予患者的步骤;也可包括将本发明的表达载体导入γ·δT细胞的步骤。α·βT细胞疗法包括利用抗CD3抗体或IL-2培养自患者采集的淋巴细胞、将经活化的α·βT细胞施予患者的步骤,也可包括将本发明的表达载体导入α·βT细胞中的步骤。CTL疗法包括以下步骤:利用自患者采集的癌细胞刺激自患者采集的淋巴细胞,添加抗CD3抗体或IL-2进行培养,使对癌细胞具有特异性的CTL增殖并向患者施予的步骤;也可包括将本发明的表达载体导入CTL的步骤。另外,作为上述癌细胞的替代,也可使用呈递癌抗原表位肽的抗原呈递细胞。TIL疗法包括从采自患者的癌症组织中采集淋巴细胞并用IL-2等刺激·培养、并将其施予患者的步骤,也可包括将本发明的表达载体导入淋巴细胞的步骤。
本发明的表达载体为用于制备上述本发明的IL-7×CCL19表达免疫活性细胞的以下(a)~(e)中的任一者。
(a)含有编码特异性识别癌抗原的细胞表面分子的核酸、编码IL-7的核酸及编码CCL19的核酸的表达载体;
(b)以下的(b-1)及(b-2)这两种表达载体:

本发明的表达载体可进一步含有编码IL-15、CCL21、IL-2、IL-4、IL-12、IL-13、IL-17、IL-18、IP-10、CCL4、Flt3L、干扰素-γ、MIP-1α、GM-CSF、M-CSF、TGF-β、TNF-α等其他免疫功能调控因子的核酸。
对于上述编码特异性识别癌抗原的细胞表面分子的核酸、编码白细胞介素7(IL-7)的核酸及编码CCL19的核酸,可分别举出源自哺乳动物的核酸,可优选举出来自人的核酸。上述各种核酸可根据导入本发明的表达载体的细胞种类作出适当选择,所述各种核酸的序列信息可通过检索已知文献或NCBI(http://www.ncbi.nlm.nih.gov/guide/)等数据库而适当地获得。
作为上述编码特异性识别癌抗原的细胞表面分子的核酸,可优选举出来自人的核酸。所述编码特异性识别癌抗原的细胞表面分子的核酸可举出编码T细胞受体(TCR)的核酸或编码嵌合抗原受体(CAR)的核酸,可以是天然来源的核酸或人工合成的核酸,可根据导入本发明的表达载体的细胞的种类作出适当选择,序列信息可通过检索已知文献或NCBI(http://www.ncbi.nlm.nih.gov/guide/)等数据库而适当地获得。
对于编码特异性识别癌抗原的细胞表面分子的核酸、编码IL-7的核酸及编码CCL19的核酸,可以基于其各自的编码核酸的碱基序列的信息,通过化学合成方法、利用PCR扩增的方法等已知技术进行制备。需要说明的是,所选择的用于编码氨基酸的密码子可为了使目标宿主细胞中的核酸发生最适表达而加以变更。
作为上述编码TCR的核酸中的TCR,可以是包含α链及β链的异二聚体(α·βTCR)或包含γ链及δ链的异二聚体(γ·δTCR)。需要说明的是,在编码α·βTCR的核酸中包含编码TCR的α链的核酸与编码β链的核酸这两者,在编码γ·δTCR的核酸中包含编码TCR的γ链的核酸与编码δ链的核酸这两者。
关于上述编码TCR的核酸的序列信息,可使用本技术领域中的已知方法,从而由作为使用特定抗原肽诱导的CTL的TCR亚单位的α链及β链的核酸进行鉴定(国际公开第2007/032255号公报及Morgan等,J Immunol,171,3288(2003))。例如,为了分析TCR,优选使用PCR法。用于分析的PCR引物例如可以是作为5’端引物的5'-R引物(5'-gtctaccaggcattcgcttcat-3':序列号3)、及作为3’端引物的对于TCRα链C区具有特异性的3-TRa-C引物(5'-tcagctggaccacagccgcagcgt-3':序列号4)、对于TCRβ链C1区具有特异性的3-TRb-C1引物(5'-tcagaaatcctttctcttgac-3':序列号5)、或对于TCRβ链C2区具有特异性的3-TRβ-C2引物(5'-ctagcctctggaatcctttctctt-3':序列号6),但不限于此。TCR衍生物能够以高结合力与呈递抗原肽的靶细胞结合,且可任意地在活体内及活体外介导呈递抗原肽的靶细胞的高效杀伤。
作为上述编码TCR的核酸,只要可识别编码例如MART1特异性的TCR(CancerRes.54,5265-5268(1994))、MAGE-A3特异性的TCR(Anticancer Res.,20,1793-1799(2000))、gp100特异性的TCR(J.Immunol.170,2186-2194(2003))、NY-ESO-1特异性的TCR(J.Immunol.,174、4415-4423(2005))、WT1特异性的TCR(Blood,106,470-476(2005))、MAGE-A1特异性的TCR(Int.Immunol.,8,1463-1466(1996))、P1A特异性的TCR(Sarma,S.,Y.Guo,Y.Guilloux,C.Lee,X.-F.Bai,Y.Liu.1999.Cytotoxic T lymphocytes to anunmutated tumor antigen P1A:normal development but restrained effectorfunction.J.Exp.Med.189:811.)等TCR的核酸、已与MHC分子结合的抗原分子、并且活化T细胞即可,可为与编码上述文献中记载的TCR的碱基序列具有80%以上、优选85%以上、更优选90%以上、进一步优选95%以上、最优选98%以上的同源性的碱基序列。此外,也可以为下述碱基序列:在编码上述文献中记载的TCR的碱基序列中确定编码CDR的序列,维持所述编码CDR的序列,并且在编码CDR的序列以外的序列中,与编码上述文献中记载的TCR的碱基序列具有60%以上、优选70%以上、更优选80%以上、进一步优选90%以上、最优选95%以上的同源性的碱基序列。
作为编码IL-7的核酸,可举出编码序列号1所示氨基酸序列的碱基序列,只要具有IL-7中的细胞增殖率或细胞存活率的亢进作用,则也可为与编码序列号1所示氨基酸序列的碱基序列具有80%以上、优选85%以上、更优选90%以上、进一步优选95%以上、最优选98%以上的同源性的碱基序列。作为编码CCL19的核酸,可举出编码序列号2所示氨基酸序列的碱基序列,只要具有CCL19中的细胞的迁移作用,则也可使用与编码序列号2所示氨基酸序列的碱基序列具有80%以上、优选85%以上、更优选90%以上、进一步优选95%以上、最优选98%以上的同源性的碱基序列。
另外,在本发明的表达载体中可包含编码自杀基因的核酸。所谓自杀基因,是指具有以下功能的基因:通过进行表达而直接地、或者二次地诱导具有细胞毒性的物质,使自身的细胞死亡。通过使本发明的表达载体含有编码自杀基因的核酸,根据癌症的治疗过程,例如在肿瘤消失的情况下施予使自杀基因功能活化的药剂,由此能够调控生物体内的免疫活性细胞。另外,IL-7或CCL19与其他细胞因子不同,引起作为副作用的细胞因子释放综合征或基因导入细胞的肿瘤化的可能性低。然而,由于导入了本发明的表达载体的免疫活性细胞的功能得以提高,因此在攻击目标癌症组织时释放的细胞因子等可能出乎意料地对周围组织产生影响。在所述情况下,通过使本发明的表达载体含有编码自杀基因的核酸,可切实地减少成为细胞因子释放综合征的风险。

本发明的载体中的(a)含有编码特异性识别癌抗原的细胞表面分子的核酸、编码IL-7的核酸及编码CCL19的核酸的表达载体中,任意核酸可配置于任意的上游或下游。具体地,以含有编码TCR的核酸作为编码特异性识别癌抗原的细胞表面分子的核酸的情况为例,按照从上游开始的顺序可为编码TCR的核酸、编码IL-7的核酸及编码CCL19的核酸,或可为编码TCR的核酸、编码CCL19的核酸及编码IL-7的核酸,或可为编码IL-7的核酸、编码CCL19的核酸及编码TCR的核酸,或可为编码IL-7的核酸、编码TCR的核酸及编码CCL19的核酸,或可为编码CCL19的核酸、编码TCR的核酸及编码IL-7的核酸,或可为编码CCL19的核酸、编码IL-7的核酸及编码TCR的核酸。
本发明的载体中的(b-2)含有编码IL-7的核酸及编码CCL19的核酸的表达载体中,编码IL-7的核酸及编码CCL19的核酸的配置没有特别限制,相对于编码IL-7的核酸而言,编码CCL19的核酸可配置于上游或配置于下游。
本发明的载体中的(c-1)含有编码特异性识别癌抗原的细胞表面分子的核酸及编码IL-7的核酸的表达载体中,编码特异性识别癌抗原的细胞表面分子的核酸及编码IL-7的核酸的配置没有特别限制,相对于编码特异性识别癌抗原的细胞表面分子的核酸而言,编码IL-7的核酸可配置于上游或配置于下游。
本发明的载体中的(d-2)含有编码特异性识别癌抗原的细胞表面分子的核酸及编码CCL19的核酸的表达载体中,编码特异性识别癌抗原的细胞表面分子的核酸及编码CCL19的核酸的配置没有特别限制,相对于编码特异性识别癌抗原的细胞表面分子的核酸,编码CCL19的核酸可配置于上游或配置于下游。
需要说明的是,编码特异性识别癌抗原的细胞表面分子的核酸、编码IL-7的核酸及编码CCL19的核酸可利用各自不同的启动子转录,也可使用内部核糖体进入位点部位(IRES:internal ribozyme entry site)或自剪切型2A肽利用一个启动子来转录。
在使用内部核糖体进入位点部位(IRES)或自剪切型2A肽而利用一个启动子来转录编码IL-7的核酸及编码CCL19的核酸的情况下的上述各核酸之间、在包含上述编码特异性识别癌抗原的细胞表面分子的核酸的情况下的该核酸与编码IL-7的核酸及编码CCL19的核酸之间、在包含编码α·βTCR的核酸的情况下的编码α链的核酸与编码β链的核酸之间、在包含编码γ·δTCR的核酸的情况下的编码γ链的核酸与编码δ链的核酸之间,可含有任意的核酸,只要能够表达各核酸即可,但优选介由编码自剪切型肽(2A肽)或IRES的序列连接,优选介由编码2A肽的序列连接。通过使用所述序列进行连接,可高效率地表达各核酸。
另外,在含有编码自杀基因的核酸的情况下,自杀基因的位置没有特别限制,例如,在用于表达编码特异性识别癌抗原的细胞表面分子的核酸、编码IL-7的核酸、或编码CCL19的核酸的启动子的下游,可介由编码2A肽或IRES的序列而配置于上述各核酸的上游或下游,也可配置于其他启动子的下游。
所谓2A肽,是指来自病毒的自剪切型肽,其具有如下特征:在如序列号7所示的氨基酸序列中的G-P之间(自C末端起1个残基的位置)由内质网切断(Szymczak等,ExpertOpin.Biol.Ther.5(5):627-638(2005))。因此,介由2A肽而并入其前后的核酸在细胞内相互独立地表达。
作为前述2A肽,优选为来自小核糖核酸病毒、轮状病毒、昆虫病毒、口疮病毒或锥虫病毒的2A肽,更优选为如序列号8所示的来自小核糖核酸病毒的2A肽(F2A)。
作为本发明的表达载体中的载体,可为直链状或环状,可为质粒等非病毒载体、病毒载体、基于转位子的载体。另外,在所述载体中,可含有启动子、终止子等调控序列、药物耐性基因、报道基因等选择标记物序列。通过在启动子序列下游可操作地配置编码IL-7的核酸及编码CCL19的核酸,由此可高效率地转录各核酸。
作为上述启动子,可列举:逆转录病毒的LTR启动子、SV40早期启动子、巨细胞病毒启动子、单纯疱疹病毒的胸苷激酶启动子等来自病毒的启动子;磷酸甘油酸激酶(PGK)启动子、Xist启动子、β-肌动蛋白启动子、RNA聚合酶II启动子等来自哺乳类的启动子。另外,也可使用由四环素诱导的四环素应答型启动子、由干扰素诱导的Mx1启动子等。本发明的表达载体中,通过使用由上述特定物质诱导的启动子,可对应于癌症的治疗过程来调控IL-7及CCL19的表达的诱导。
作为前述病毒载体,可举出逆转录病毒载体、慢病毒载体、腺病毒载体、腺相关病毒载体,可优选举出逆转录病毒载体,可更优选举出pMSGV载体(Tamada k等,Clin CancerRes 18:6436-6445(2002))、pMSCV载体(TAKARA BIO公司制造)。若使用逆转录病毒载体,则导入基因被并入宿主细胞的基因组中,因此可长期间地并且稳定地表达。
为确认免疫活性细胞中含有本发明的表达载体、例如在含有编码TCR的核酸的情况下,可通过以下方法调查TCR的表达:流式细胞仪;北方墨点法、南方墨点法、RT-PCR等PCR方法;ELISA、西方墨点法。在本发明的表达载体含有标记物基因的情况下,可通过调查向该表达载体中插入的标记物基因的表达而确认。
在本发明的IL-7×CCL19表达免疫活性细胞中所含有的表达载体中含有编码TCR的核酸的情况下,所表达的TCR的可变区位于细胞外,通过具有所述TCR可变区,TCR表达免疫活性细胞可识别已与MHC分子结合的抗原分子。
本发明的抗癌剂只要含有本发明的IL-7×CCL19表达免疫活性细胞和药学上允许的添加剂即可,没有特别限制。作为前述添加剂,可举出生理盐水、缓冲生理盐水、细胞培养培养基、右旋糖、注射用水、甘油、乙醇及它们的组合、稳定剂、助溶剂及表面活性剂、缓冲剂及防腐剂、等渗剂、填充剂、以及润滑剂。
本发明的抗癌剂可使用本领域技术人员已知的方法向需要癌症治疗的受试者施予,作为施予方法,可举出静脉内、肿瘤内、皮内、皮下、肌肉内、腹腔内、动脉内、髓内、心脏内、关节内、滑液囊内、颅内、髓腔内、及蛛网膜下(脑脊液)注射。
所施予的抗癌剂中含有的本发明的IL-7×CCL19表达免疫活性细胞的量,可根据癌症的种类、位置、严重程度、接受治疗的受试者的年龄、体重及状态等作出适当调整,但优选地,可举出在每次施予中为1×104~1×1010个、优选1×105~1×109个、更优选5×106~5×108个。
对于所施予的抗癌剂而言,可举出1天4次、3次、2次或1次、每隔1天、每隔2天、每隔3天、每隔4天、每隔5天、每周1次、每隔7天、每隔8天、每隔9天、每周2次、每月1次或每月2次独立施予的方法。
作为本发明的抗癌剂、下述癌症治疗方法所针对的癌症,可为实体瘤或血液癌症,可举出腺癌、鳞状上皮癌、腺鳞状上皮癌、未分化癌、大细胞癌、小细胞癌、皮肤癌、乳腺癌、前列腺癌、膀胱癌、阴道癌、颈癌、子宫癌、肝癌、肾癌、胰脏癌、脾脏癌、肺癌、气管癌、支气管癌、结肠癌、小肠癌、胃癌、食道癌、胆囊癌、睾丸癌、卵巢癌等癌症;骨组织、软骨组织、脂肪组织、肌组织、血管组织及造血组织的癌症;以及软骨肉瘤、尤因氏肉瘤、恶性血管内皮瘤、恶性神经鞘瘤、骨肉瘤、软组织肉瘤等肉瘤;肝母细胞瘤、髓母细胞瘤、肾母细胞瘤、神经母细胞瘤、胰母细胞瘤、胸膜肺母细胞瘤、视网膜母细胞瘤等母细胞瘤、胚细胞瘤、淋巴瘤、白血病。
本发明的抗癌剂可与其他抗癌剂联用。作为其他抗癌剂,可举出:环磷酰胺、苯达莫司汀、异环磷酰胺、达卡巴嗪等烷基化药物;喷司他丁、氟达拉滨、克拉屈滨、甲胺蝶呤、5-氟尿嘧啶、6-巯基嘌呤、依诺他滨等代谢拮抗药;利妥昔单抗、西妥昔单抗、曲妥珠单抗等分子靶向药;伊马替尼、吉非替尼、厄洛替尼、阿法替尼、达沙替尼、舒尼替尼、曲美替尼等激酶抑制剂;硼替佐米等蛋白酶体抑制剂、环孢素、他克莫司等钙调神经磷酸酶抑制剂;伊达比星、多柔比星、丝裂霉素C等抗癌性抗生素;伊立替康、依托泊苷等植物生物碱;顺铂、奥沙利铂、卡铂等铂制剂;他莫昔芬、比卡鲁胺等激素疗法药;干扰素、纳武单抗、帕博利珠单抗等免疫调节药。可优选举出烷基化药物或代谢拮抗药。
作为上述“本发明的抗癌剂与其他抗癌剂联用”的方法,可举出:使用其他抗癌剂进行处理后使用本发明的抗癌剂的方法;同时使用本发明的抗癌剂和其他抗癌剂的方法;使用本发明的抗癌剂进行处理后使用其他抗癌剂的方法。可优选举出使用其他抗癌剂进行处理后使用本发明的抗癌剂的方法。另外,在本发明的抗癌剂与其他抗癌剂联用的情况下,能进一步提高癌症的治疗效果,同时减少各抗癌剂的施予次数或施予量,由此可减少各抗癌剂引起的副作用。另外,本发明的抗癌剂中也可包含上述其他抗癌剂。
作为本发明的另一方案1,可举出:1)癌症的治疗方法,其特征在于,将表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及CCL19的免疫活性细胞施予需要癌症治疗的患者;2)免疫活性细胞,其用作抗癌剂,且表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)及CCL19;3)表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及CCL19的免疫活性细胞在抗癌剂的制备中的用途。
此外,作为本发明的另一方面2,可举出具有上述本发明的表达载体的、用于制备表达特异性识别癌抗原的细胞表面分子、白细胞介素7(IL-7)、及CCL19的免疫活性细胞的试剂盒,作为所述试剂盒,只要具有本发明的表达载体则没有特别限制,也可包含用于制备本发明的IL-7×CCL19表达免疫活性细胞的说明书、用于将本发明的表达载体导入免疫活性细胞的试剂。
实施例1
(免疫功能调控因子的选择)
可调控T细胞的功能的分子在生物体内至少存在数百种。本申请的发明人基于迄今为止的知识和经验,首先从大量组合中选择IL-7和CCL19作为用于进一步提高免疫活性细胞中的免疫功能调控效果的调控分子,而且,并非各自单独地选择而是选择两者的组合、即IL-7与CCL19的组合,制备了表达所述的免疫活性细胞的免疫功能调控因子的载体。
(表达IL-7及CCL19的载体的制备-1)
人工合成编码包含抗FITC scFv、小鼠CD8跨膜区、小鼠CD28-4-1BB-CD3δ细胞内信号基序(motif)的抗FITC CAR的抗FITC CAR DNA片段(序列号9)、编码序列号8所示的2A肽(F2A)与继该肽之后的限制性内切酶位点(MCS)的F2A-MCS DNA片段(序列号10)、编码小鼠IL-7(无终止密码子)与继其之后的F2A及小鼠CCL19的IL-7-F2A-CCL19 DNA片段(序列号11)(Life Technology公司制造)。
为制备表达IL-7及CCL19的载体,将上述抗FITC CAR DNA片段与上述F2A-MCS DNA片段连接,制备抗FITC CAR-F2A-MCS构建体(construct)。接下来,将所制备的构建体克隆至pMSGV逆转录病毒表达载体(Tamada k等,Clin Cancer Res 18:6436-6445(2002))中,制备包含抗FITC CAR-F2A-MCS的pMSGV载体。通过限制性内切酶(NsiI及SalI)处理及接合而将上述IL-7-F2A-CCL19 DNA片段插入所述pMSGV载体的MCS中,得到包含抗FITC CAR-F2A-IL-7-F2A-CCL19的pMSGV载体(IL-7×CCL19表达载体(1))。所得载体的遗传图如图1所示。另外,作为对照,将上述抗FITC CAR DNA片段克隆至上述pMSGV逆转录病毒表达载体中,制备不含IL-7及CCL19的pMSGV载体(对照载体(1))。
(导入了IL-7×CCL19表达载体的逆转录病毒的制备)
为进行小鼠T细胞的转导,制备逆转录病毒。使用Lipofectamine2000或3000(LifeTechnology公司制造)将上述IL-7×CCL19表达载体(1)或对照载体(1)、和pCL-Eco质粒(Imgenex公司制造)转染至GP2-293包装细胞株(TAKARA BIO公司制造),由此制备导入了IL-7×CCL19表达载体(1)或对照载体(1)的逆转录病毒。
使用添加有10%FCS、100U/ml的青霉素、100mg/ml的链霉素的DMEM作为前述GP2-293细胞的培养液。另外,使用添加有10%FCS、100U/ml的青霉素、100mg/ml的链霉素、50mM的2-巯基乙醇、2mM的L-谷氨酰胺的RPMI-1640作为后述实施例所用的T细胞的培养液。
(小鼠T细胞的转导)
为进行小鼠T细胞的转导,利用经固相化的抗CD3mAb(3μg/ml)及IL-2(100IU/ml)对来自脾脏及淋巴结的3×106个经纯化的小鼠T细胞进行48小时活化。接下来,将含有上文中制备的导入了IL-7×CCL19表达载体(1)或对照载体(1)的逆转录病毒的上清液、与在经25μg/ml的Retro Nectin(注册商标:TAKARA BIO公司制造)涂覆的培养板中活化后的上述小鼠T细胞(1×106个细胞/ml)混合,以1500rpm离心2小时后,在IL-2(100IU/ml)的存在下培养6小时。为了自培养液中除去逆转录病毒,回收小鼠T细胞,并转移至含有IL-2(100IU/ml)的新的增殖培养液(RPMI)中,再培养42小时,获得导入了IL-7×CCL19表达载体(1)的小鼠T细胞(IL-7/CCL19表达T细胞(1))或导入了对照载体(1)的小鼠T细胞(对照T细胞(1))。
(表达IL-7及CCL19的表达载体的制备-2)
在上述IL-7×CCL19表达载体(1)的制备中,将序列号9所示序列中所含的抗FITCscFv区域的序列置换为由Life Technology公司基于利妥昔单抗序列合成的抗人CD20scFv(序列号12)的序列,除此之外,利用与上述“表达IL-7及CCL19的表达载体的制备-1”相同的方法制备包含抗人CD20 CAR-F2A-IL-7-F2A-CCL19的pMSGV载体(IL-7×CCL19表达载体(2))。同样地,在上述对照载体(1)的制备中,将序列号9所示序列中所含的抗FITC scFv区域的序列置换为上述抗人CD20 scFv(序列号12)的序列,除此之外,利用与上述“表达IL-7及CCL19的表达载体的制备-1”相同的方法制备不含IL-7及CCL19的pMSGV载体(对照载体(2))。利用与上述相同的方法,使用逆转录病毒向小鼠T细胞中导入所述IL-7×CCL19表达载体(2)或对照载体(2),制备IL-7/CCL19表达T细胞(2)或对照T细胞(2)。
实施例2
(IL-7/CCL19表达T细胞的细胞数及存活率)
对于由IL-7/CCL19表达T细胞产生的IL-7、CCL19是否发挥生物功能并显示免疫诱导效果进行了研究。将含有所制备的IL-7/CCL19表达T细胞(2)(4×105个)或对照T细胞(2)的样品培养5天。为了排除人CD20 CAR对IL-7及CCL19的表达产生的影响,在不存在由CD20引起的抗原刺激的情况下进行上述培养。接下来,利用台盼蓝测定细胞数和存活率。结果如图2A、图2B所示。图2A为细胞数,图2B为存活率,黑色柱表示IL-7/CCL19表达T细胞,白色柱表示对照T细胞。
(结果)
如图2A、图2B所示,与对照T细胞(2)相比,在IL-7/CCL19表达T细胞(2)中细胞数约高达其5倍,存活率约高达其2倍。因此可明确,通过使用将本发明的表达载体导入T细胞而获得的IL-7/CCL19表达T细胞,从而IL-7、CCL19发挥生物功能并显示免疫诱导效果。
实施例3
[T细胞迁移试验]
(利用IL-7/CCL19表达T细胞进行的T细胞迁移试验)
通过使用Transwell进行的细胞迁移试验,研究对CCL19的迁移引发效果。对于应答侧T细胞的迁移性而言,使用96孔Transwell(注册商标)小室(Corning Costar公司制造),使其通过孔径5μm的聚碳酸酯过滤器来进行迁移,由此进行测定。具体地,在小室的下层培养IL-7/CCL19表达T细胞(1)或对照T细胞(1)。为了排除FITC CAR对于IL-7及CCL19的表达产生的影响,在不存在由FITC引起的抗体刺激的情况下进行上述培养。通过对MACS(Miltenyi Biotech公司制造)进行阴性选择,从而由脾脏、淋巴结制备应答侧T细胞。用CytoTell blue(AAT Bioquest公司制造)对应答侧T细胞进行标记,在上层培养3小时。通过流式细胞仪(EC800:索尼公司制造)调查由小室的上层向下层的迁移,使用FlowJo软件(Tree Star公司制造)进行数据分析。结果如图3所示。图3中,黑色柱表示IL-7/CCL19表达T细胞(1),白色柱表示对照T细胞(1),纵轴表示迁移至下层小室的应答侧T细胞的绝对数量。另外,通过学生t检验研究了统计学上的显著性差异。
(结果)
如图3所示,与对照T细胞(1)相比,IL-7/CCL19表达T细胞(1)使约1.8倍的T细胞迁移至下层。在T细胞等淋巴细胞移入疗法中,因所施予的T细胞引起的癌细胞损伤固然是重要的,但除此之外,通过活化癌症患者中原本存在的内源性T细胞(=宿主侧免疫细胞)而将其调动为攻击癌细胞的细胞也是重要的。为此,从提高免疫治疗效果的观点考虑,优选不仅仅单纯地从外部移入具有抗肿瘤活性的淋巴细胞,而且还通过某些手段引发移入的T细胞与内源性T细胞之间的积极的相互作用,使得内源性T细胞聚集于癌症局部。由图3的结果可明确,IL-7/CCL19表达T细胞(1)具有使内源性T细胞聚集的能力,因此可诱导所移入的T细胞与内源性T细胞之间的积极的相互作用。
另外,由图2A、图2B、图3的结果可明确,表达IL-7及CCL19的T细胞通过IL-7而有效地增殖,存活率高,且具备由CCL19使T细胞聚集这一对免疫诱导而言不可或缺的重要效果,具有优异的免疫诱导效果。即,可明确在免疫活性细胞中,通过使“IL-7”和“CCL19”这两种调控分子表达,可提高所述免疫活性细胞的增殖能力、存活率、免疫诱导效果。进而,如上所述,表达IL-7及CCL19的T细胞由于兼具增殖能力、存活能力、及T细胞集聚能力,因此表明了在癌症组织中具有T细胞、树突状细胞的渗透效果、肿瘤增殖抑制效果的可能性。
实施例4
[IL-7×CCL19×HSV-TK表达载体的制备]
在pMSGV1载体的多克隆位点克隆以下碱基序列:使编码IL-7、CCL19及作为自杀基因的HSV-TK的各基因的碱基序列夹着编码作为自剪切型肽的2A肽的碱基序列而串联形成的碱基序列。由此,可制备表达IL-7、CCL19、及HSV-TK的载体。所述载体的遗传图如图4所示。
对于导入了如上文所述地制备的IL-7×CCL19×HSV-TK表达载体的免疫活性细胞而言,通过向施予了上述免疫活性细胞的受试者施予更昔洛韦,可调控受试者内的免疫活性细胞。
实施例5
[TCR×IL-7×CCL19表达载体的制备]
在pMSGV1载体的多克隆位点克隆以下碱基序列:使编码TCR、IL-7及CCL19的各基因的碱基序列夹着编码作为自剪切型肽的2A肽的碱基序列而串联形成的碱基序列。由此,可制备表达TCR、IL-7及CCL19的载体。所述载体的遗传图如图5所示。
导入了如上文所述地制备的TCR×IL-7×CCL19表达载体的免疫活性细胞不仅能够与存在于癌细胞表面的癌抗原特异性结合,而且能够与来自癌细胞内的癌抗原的肽经MHC呈递而得的复合体特异性结合,由此能够针对更广范围的肿瘤相关靶分子诱导特异性T细胞。
实施例6
[表达IL-7、CCL19及eGFP的表达载体的制备]
人工合成编码小鼠IL-7(无终止密码子)与继其之后的F2A、小鼠CCL19的IL-7-F2A-CCL19 DNA片段(Life Technology公司制造)。
为了制备表达IL-7、CCL19及eGFP的载体,通过限制性内切酶(NCOI及ECORI)处理及接合而将上文中合成的IL-7-F2A-CCL19DNA片段插入具有F2A-eGFP序列的pMSGV逆转录病毒表达载体(Tamada K等,Clin Cancer Res 18:6436-6445(2002))的MCS中,获得含有IL-7-F2A-CCL19-F2A-eGFP DNA片段(序列号13)的pMSGV载体(IL-7×CCL19表达载体(3))。所得载体的遗传图如图6所示。另外,作为对照,制备了含有eGFP而不含IL-7及CCL19的pMSGV载体(对照载体(3))。需要说明的是,序列号13中,第1~462位碱基为IL-7(第1~75位碱基为IL-7的信号序列),第463~537位碱基为F2A,第538~861位碱基为CCL19(第538~612位碱基为CCL19的信号序列),第868~942号为F2A,第946~1662位碱基为编码eGFP的核酸,第1663~1665位碱基为终止密码子。另外,与上述碱基序列13对应的氨基酸序列如序列号14所示。需要说明的是,为了使用限制性内切酶NcoI,将序列号13中第4位碱基胸腺嘧啶(t)替换为鸟嘌呤(g)(将序列号14中第2位氨基酸苯丙氨酸(F)替换为缬氨酸(V))。

自转基因小鼠(自Y.Liu获得,其为表达H-2Ld限制性P815肿瘤抗原P1A特异性的TCR的转基因小鼠(Sarma,S.,Y.Guo,Y.Guilloux,C.Lee,X.-F.Bai,Y.Liu.1999.J.Exp.Med.189:811.))采集脾脏细胞,获得表达源自脾脏细胞的P815肿瘤抗原P1A特异性的TCR的小鼠T细胞(P1A特异性的TCR-T细胞)。接下来,利用与实施例1同样的方法制备导入了IL-7×CCL19表达载体(3)及对照载体(3)的逆转录病毒,转导至用P1A肽使含有上述P1A特异性的TCR-T细胞的脾脏细胞(3×106个/孔)活化48小时而得到的细胞中,得到P1A特异性的TCR/IL-7/CCL19/eGFP表达T细胞或P1A特异性的TCR/eGFP表达T细胞。通过流式细胞仪分析检测作为替代标记的eGFP,确认各表达载体的转导。所得各T细胞的eGFP的表达水平在任一实验中均为70~80%。
在第0天,对6~10周龄的雄性DBA/2小鼠(n=30)于侧腹皮下接种悬浮于0.1ml的HBSS中的5×105个P815肥大细胞瘤。在第6天,对小鼠进行亚致死量(3-5Gy)的照射,以进行预处理。在第7天,将小鼠(n=10)分为3组,分别静脉施予1×106个P1A特异性的TCR/IL-7/CCL19/eGFP表达T细胞、或P1A特异性的TCR/eGFP表达T细胞(任一细胞均为70~80%eGFP阳性)。然后,分析各小鼠的存活率,并测定死亡小鼠的肿瘤体积。各小鼠的存活率的分析结果如图7所示,死亡小鼠的肿瘤体积的测定结果如图8所示。
图7中,▲为未处理小鼠的结果,■为施予P1A特异性的TCR/eGFP表达T细胞的小鼠的结果,●为施予P1A特异性的TCR/IL-7/CCL19/eGFP表达T细胞的小鼠的结果,横轴为自皮下接种P815肥大细胞瘤起的天数(天),纵轴为存活率(%)。施予P1A特异性的TCR/IL-7/CCL19/eGFP表达T细胞的小鼠在第60天仍有80%存活,超过100天仍有50%存活。由此可明确,通过使用表达P1A特异性的TCR、IL-7及CCL19的免疫活性细胞,发挥了抗肿瘤效果,抑制因肿瘤导致的存活率降低。
另外,图8中,横轴为自皮下接种P815肥大细胞瘤起的天数(天),纵轴为肿瘤体积(mm3)。由图8可明确,在施予P1A特异性的TCR/IL-7/CCL19/eGFP表达T细胞的小鼠中显著抑制了肿瘤体积的增加,由此可确认,P1A特异性的TCR/IL-7/CCL19/eGFP表达T细胞具有优异的抗肿瘤活性,并且能对实体瘤发挥治疗效果。
产业上的可利用性
本发明的IL-7×CCL19表达免疫活性细胞由于兼具增殖能力、存活能力及淋巴细胞集聚能力,因此可用于免疫疗法的领域中。
Claims (13)
1.编码白细胞介素7(IL-7)的核酸及编码CCL19的核酸用于导入表达T细胞受体(TCR)的免疫活性细胞而制备表达TCR、IL-7、及CCL19的免疫活性细胞的用途。
2.如权利要求1所述的用途,其特征在于,使用表达载体将编码IL-7的核酸、及编码CCL19的核酸导入表达TCR的免疫活性细胞。
3.如权利要求2所述的用途,其特征在于,使用含有编码IL-7的核酸、及编码CCL19的核酸的表达载体导入表达TCR的免疫活性细胞。
4.如权利要求2或3所述的用途,其特征在于,所述表达载体中含有编码自杀基因的核酸。
5.如权利要求1~3中任一项所述的用途,其特征在于,免疫活性细胞为淋巴细胞、抗原呈递细胞、或粒细胞。
6.如权利要求1~3中任一项所述的用途,其特征在于,免疫活性细胞为T细胞。
7.如权利要求1~3中任一项所述的用途,其特征在于,TCR为识别癌抗原的TCR,所述癌抗原为WT1、MART-1、NY-ESO-1、MAGE-A1、MAGE-A3、MAGE-A4、磷脂酰肌醇蛋白聚糖-3、KIF20A、存活蛋白、AFP-1、gp100、MUC1、PAP、TRP2、SART-1、VEGFR1、VEGFR2、NEIL3、MPHOSPH1、DEPDC1、FOXM1、CDH3、TTK、TOMM34、URLC10、KOC1、UBE2T、TOPK、ECT2、间皮素、NKG2D、P1A、GD2、或GM2。
8.表达T细胞受体(TCR)、白细胞介素7(IL-7)、及CCL19的免疫活性细胞的制备方法,其特征在于,将编码IL-7的核酸、及编码CCL19的核酸导入表达TCR的免疫活性细胞。
9.如权利要求8所述的制备方法,其特征在于,使用表达载体将编码IL-7的核酸、及编码CCL19的核酸的导入表达TCR的免疫活性细胞。
10.如权利要求8或9所述的制备方法,其特征在于,免疫活性细胞为T细胞。
11.编码白细胞介素7(IL-7)的核酸及编码CCL19的核酸用于导入表达嵌合抗原受体(CAR)的免疫活性细胞而制备表达CAR、IL-7、及CCL19的免疫活性细胞的用途。
12.如权利要求11所述的用途,其特征在于,免疫活性细胞为T细胞。
13.表达嵌合抗原受体(CAR)、白细胞介素7(IL-7)、及CCL19的免疫活性细胞的制备方法,其特征在于,将编码IL-7的核酸、及编码CCL19的核酸导入表达CAR的免疫活性细胞。
