Description
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a cosmetic composition, essence emulsion and a preparation method thereof.
Background
Skin aging is characterized in that oxygen free radicals attack collagen and elastic fibers in skin to cause collagen denaturation and elastic fiber fracture; on the other hand, with age, the proliferation rate of collagen and elastin in skin is slowed down, and damaged parts cannot be compensated in time; meanwhile, along with aging, the skin can generate color spots, and then the skin can develop into senile plaques; the skin gradually loses elasticity and becomes loose, wrinkles and spots are formed. Skin anti-aging cosmetics are increasingly popular with the aged society. Skin aging resistance is a comprehensive process, on one hand, the generation of oxygen free radicals in skin is reduced or the oxygen free radicals are timely removed; on the other hand, it is required to remove wrinkles by accelerating the removal of old skin horns and generating new collagen, and it is also required to remove or suppress the occurrence of color spots. Resveratrol is a natural antioxidant existing in plants, and is mainly extracted from plant polygonum cuspidatum at present mainly by scavenging or inhibiting oxygen free radicals and inhibiting lipid peroxidation; dimethoxy tolyl-4-propyl resorcinol is a phenolic compound, can inhibit the generation of oxygen free radicals, is mainly extracted from plant dianella, and has remarkable freckle-removing and whitening effects at present; the salicyloyl phytosphingosine can induce collagen in dermal papilla layer to smooth skin, and can help to remove old skin cutin and relieve wrinkles generated in skin aging process. Aiming at the skin anti-aging mechanism, the resveratrol, dimethoxy tolyl-4-propyl resorcinol and salicyl phytosphingosine are compounded and all-round anti-aging is realized. However, resveratrol, dimethoxytolyl-4-propylresorcinol and salicyl phytosphingosine are all dissolved in ethanol and are insoluble in water and oil, so that the application is difficult; the resveratrol has strong dimethoxy tolyl-4-propyl resorcinol activity and very poor stability, for example, 40% ethanol solution of resveratrol can be stable for only a few days under the condition of avoiding light, and can not keep activity in cosmetics for a long time; therefore, although the effect is remarkable, it is very difficult to use it in cosmetics.

test case
Samples prepared by the technical schemes of example 1, example 2, example 3 and example 4, and samples prepared by the technical schemes of the comparative schemes were selected and tested as follows:
Anti-inflammatory performance test:
IL-8 is a cytokine secreted by Th1 cells, mainly mediates the generation of immune response auxiliary antibodies related to cytotoxin and local inflammation, participates in the generation of cellular immunity and delayed type hypersensitive inflammation, has the main biological effects of chemotactic and activation of neutrophils, promotes the lysosomal enzyme activity and phagocytosis of the neutrophils, has a certain chemotactic effect on basophils and T cells, has morphological change after the neutrophils are contacted with the IL-8, directionally moves to a reaction part and releases a series of active products, and can lead to local inflammatory reactions of organisms, thereby achieving the purposes of sterilizing and damaging the cells.
Experimental materials: ku812 cells were supplied by the center of chinese academy of cells; RPMI1640, IMDM medium, penicillin, streptomycin were all purchased from Gibco; fetal bovine serum was purchased from HyClone company; DMSO, PMA, and a12387 were purchased from Sigma; IL-8 kits were purchased from R & D systems; mu Q mu ant microplate reader was purchased from BIO-TEK company.
Sample preparation of an experimental method: the resulting cream of examples 1-4 and comparative protocol was formulated with DMSO or double distilled water and diluted to a concentration of 25mg/mL in a medium containing 10% fetal bovine serum.
Ku812 cell density was adjusted to 2.2x10 6/mL cell suspension in IMDM medium containing 10% fetal bovine serum and seeded in 96-well plates at 200 μl per well. Setting a cell blank control group, an example group and a comparison example group, wherein each group is provided with 3 parallel holes, culturing and standing for 10min in a culture box with 5% CO 2 at 37 ℃, adding samples with the final concentration of 50 mug/mL into the example group and the comparison scheme group, pre-incubating in the culture box for 15min, adding a stimulator PMA and A12387 into each group of cells except the cell blank group, so that the final concentration is 5nm and 1 mu m respectively, and adding the same volume of DMSO culture medium with the same concentration into the cell blank group. After 2h, the cells were centrifuged, and the cell supernatant was collected and the IL-8 content of the supernatant was measured. In addition, a model group was set, and the treatment was performed in the same manner as in the example group or the comparative example group, but without adding a sample.
TABLE 1IL-8 assay results
In combination with Table 1, the products prepared in examples 1-4 of the present application have a significant effect of inhibiting skin inflammation compared to the model group and the comparative regimen group. Specifically, the raw materials of the first to fifth active components can solve the problems of skin inflammation such as redness, itching and the like caused by ultraviolet injury, free radical oxidation, environmental pollution, excessive sebum secretion, bacterial infection and the like by inhibiting IL-8 to play a role in inhibiting IV type hypersensitivity reaction (delayed type hypersensitivity reaction), and supposedly play a role in inhibiting the high expression of inflammatory factors, thereby inhibiting the increase of capillary permeability and the occurrence of inflammation and allergy phenomena caused by histamine release.
Anti-aging test: the skin elasticity test was performed on test areas inside the forearms of both hands of a subject using the skin elasticity tester Cutometer Dual MPA580,580 of the germany CK company to reflect the anti-aging effect of the present invention, and the average value of the skin elasticity values of each test area was measured 3 times, the closer to 1 the better. Finally, the average values of the skin elasticity values of the subjects of examples 1 to 4 and the comparative example protocol were counted 30.
TABLE 2 skin elasticity values
/>
As can be seen from Table 2, the products prepared in examples 1-4 according to the present invention have skin elasticity values ranging from 0.87 to 0.92, with a better significant difference in elasticity parameters compared to the comparative examples. On the basis of the prior art, a plurality of active ingredients are introduced, and nutrition ingredients are reserved as much as possible in the preparation method, so that the essence emulsion permeates into the skin, and the excellent performance is shown by the synergy among all parameter conditions.
