针对现有技术中所存在的不足,本发明提供了一种载IR780和氢氧化铝靶向抗原捕获纳米粒及其应用,其提供了一种协同抗原捕获及目标纳米粒的选择性积累和深度肿瘤渗透用以治疗宫颈癌的新方法。
本发明一方面,提供一种载IR780和氢氧化铝靶向抗原捕获纳米粒,包括核壳结构,所述核为携氧全氟戊烷,所述壳为聚乳酸-乙醇酸,所述聚乳酸-乙醇酸上负载有IR780和氢氧化铝。
进一步地,所述载IR780和氢氧化铝靶向抗原捕获纳米粒的流体力学直径为257.133±3.82nm,表面电位为21.97±0.75mV。
进一步地,所述载IR780和氢氧化铝靶向抗原捕获纳米粒中氢氧化铝的包封率为14.19±1.45%,载药量为0.27±0.03%;IR780的包封率为79.53±0.47%,载药量为2.93±0.071%。
本发明另一方面,提供一种载IR780和氢氧化铝靶向抗原捕获纳米粒的制备方法,采用W/O/W双重乳化法进行制备,包括以下步骤:
S1:将聚乳酸-乙醇酸和IR780溶解于二氯甲烷中,再加入携氧全氟戊烷和氢氧化铝水溶液,进行乳化,得到初乳液;
S2:将步骤S1得到初乳液溶解在聚乙烯醇水溶液中,再次乳化,得到混合乳液;将混合乳液搅拌、离心得到载IR780和氢氧化铝靶向抗原捕获纳米粒。
进一步地,按mg/mg/ml计,聚乳酸-乙醇酸、IR780和二氯甲烷的用量比为50-100:1-2:2。
进一步地,氢氧化铝水溶液的浓度为1mg/ml,携氧全氟戊烷、氢氧化铝水溶液、二氯甲烷的体积比为0.2-0.3:1-2:2。
进一步地,聚乙烯醇水溶液的浓度为5%,二氯甲烷与聚乙烯醇水溶液的体积比为2:5-7。
进一步地,步骤S1或步骤S2中,所述乳化为冰浴条件下,超声乳化3-5min。
本发明再一方面,提供一种载IR780和氢氧化铝靶向抗原捕获纳米粒在制备治疗抗肿瘤药物中的应用。
进一步的,所述肿瘤为宫颈癌。
本发明的技术原理为:本发明通过局部PDST和具有捕获抗原潜力的载IR780和氢氧化铝靶向抗原捕获纳米粒(OIRAI-NPs)治疗宫颈癌。
具体来说,本发明使用聚乳酸-乙醇酸(PLGA)作为包封聚合物、IR780作为光敏剂以使全氟戊烷(PFP)作为携带氧的相变造影剂。
氢氧化铝纳米颗粒作为捕获抗原的佐剂,可高效吸附抗原,并且逐渐释放抗原,增加了抗原呈递细胞(APC)对抗原摄取的影响,并引发肿瘤特异性免疫应答。
与传统光敏剂(即吲哚菁绿)相比,IR780显示出更高的光稳定性、荧光强度、ROS产生效率、光热转换和更宽的吸收截面。同时,IR780可以选择性地聚集在肿瘤区域,赋予OIRAI-NPs选择性积累并深入肿瘤组织的能力。进一步,内化的IR780主要集中于线粒体,促进癌症治疗。更进一步,IR780的宽吸收截面使得OIRAI-NPs在近红外(NIR)激光照射下产生了大量的1O2和热量,以促进光疗,从而增强了抗肿瘤治疗,从而引发了TDPA的更高释放。并且,OIRAI-NP具有原位收集和维持TDPA的能力,这增强了抗原呈递细胞(APC)对抗原摄取的影响,并触发肿瘤特异性免疫应答。
相比于现有技术,本发明具有如下有益效果:
(1)本发明通过协同作用来实现癌症治疗,OIRAI-NP不仅可以用作肿瘤治疗的PSDT介质,而且可以捕获治疗期间释放的抗原,以触发抗肿瘤免疫应答,诱导免疫原性细胞死亡(ICD),大大增强了损伤相关分子模式(DAMP)的产生。
(2)本发明中的IR780的固有结构赋予了OIRAI-NPs线粒体靶向能力,氢氧化铝作为抗原捕获剂的OIRAI-NP改善了树突状细胞(DC)对肿瘤抗原的摄取。同时,IR780和氢氧化铝的联合应用具有协同增效作用,共同促进了载IR780和氢氧化铝靶向抗原捕获纳米粒的抗肿瘤效果。
(3)OIRAI-NPs在增强光声(PA)和超声成像作为造影剂方面具有巨大潜力。
综上所述,基于抗癌免疫疗法的核心原理,本发明的联合治疗方法可显著抑制原发性肿瘤生长和肿瘤转移进展,可以成为癌症治疗的潜在纳米平台。该联合策略有望显著提高抗肿瘤免疫能力,实现宫颈癌的诊断与治疗一体化。

具体实施方式
下面结合附图及实施例对本发明中的技术方案进一步说明。纳米级氢氧化铝(Aluminumhydroxidenanoparticles)由西安瑞禧生物科技有限公司(西安,中国)提供。
实施例1载IR780和氢氧化铝靶向抗原捕获纳米粒的制备
通过双重乳化法(水/油/水)制备线粒体靶向的载IR780和氢氧化铝靶向抗原捕获纳米粒(OIRAI-NPs)。具体步骤如下:
S1:将1mgAl(OH)3溶解于1ml双蒸馏水(1mg/ml)中,将50mgPLGA和1mgIR780混合,然后完全溶解于2ml二氯甲烷(DCM)中。之后,向混合物中加入200μlPFP和1ml Al(OH)3(1mg/ml),使用声震仪在冰浴中以35w的功率(5s开和5s关)将混合物乳化3分钟(美国HeatSystemInc)。
S2:将混合物溶解在5ml聚乙烯醇水溶液(5%)中,并在35W的功率下声震仪额外处理3分钟(5son和5soff)。将所得乳液磁力混合搅拌4小时以除去DCM(冰浴),随后,以10000rpm离心5分钟,离心三次,每次离心后用双蒸水洗涤,然后使制备好的纳米粒悬浮在双蒸馏水中。所有的操作都是在完全黑暗的冰浴中进行的。
采用相同方法制备未装载氢氧化铝的纳米粒(OIR-NPs)、未装载IR780的纳米粒(OAI-NPs)和未装载IR780和氢氧化铝的纳米粒(O-NPs)。
透射电镜观察多功能复合纳米粒OIRAI-NPs为均匀一致的球形结构(图1),可均匀分散于双蒸水中。粒度及Zeta电位分析仪测量OIRAI-NPs、OIR-NPs、OAI-NPs、O-NPs的平均粒径和表面电位。装载IR780和氢氧化铝的多功能复合纳米粒OIRAI-NPs流体力学直径为257.133±3.82nm(图2),表面电位为21.97±0.75mV。未装载氢氧化铝的纳米粒(OIR-NPs)平均粒径为213.3±3.90nm,表面电位为-5.74±0.58mV。未装载IR780的纳米粒(OAI-NPs)平均粒径为226.03±0.68nm,表面电位为3.76±0.57mV。未装载IR780和氢氧化铝的纳米粒(O-NPs)平均粒径为209.86±4.49nm,表面电位为-11.77±0.25Mv。
电感耦合等离子体发射光谱仪(ICP-OES法)检测OIRAI-NPs、OAI-NPs中氢氧化铝的包封率和载药量。OIRAI-NPs纳米粒中氢氧化铝的包封率为14.19±1.45%,载药量为0.27±0.03%,纳米粒OAI-NPs中氢氧化铝的包封率为4.67±0.32%,载药量为0.09±0.01%,两种纳米粒的铝含量均符合铝佐剂的安全性限度标准(世界卫生组织及欧盟1.25mg/剂,美国0.85mg/剂,中国0.35-3.0mg/mL-1)。XPS检测进一步证实OIRAI-NPs中氢氧化铝的成功包载(图3)。
紫外分光光度计检测到OIRAI-NPs在780nm波长处可见较强的吸收峰(图4),根据IR780的标准曲线(图5)计算得出OIRAI-NPs中IR780的包封率为79.53±0.47%,载药量为2.93±0.071%;OIR-NPs中IR780的包封率为71.92±0.193%,载药量为2.75±0.01%。根据游离IR780和OIRAI-NPs、OIR-NPs、OAI-NPs、O-NPs的紫外吸收光谱表明,IR780被成功封装在纳米粒中(图6)。
由于IR780不溶于水,纳米粒中较高的包封率可避免IR780的淬灭并增加其稳定性,提高其在生物组织中的应用,如图7所示。由结果可知,14天内,IR780的光学稳定性下降明显,而OIRAI-NPs光学稳定性变化不大。此外,分散在双蒸水中的OIRAI-NPs稳定性较好,4℃放置30内,其粒径无明显改变。
实施例2载IR780和氢氧化铝靶向抗原捕获纳米粒的体外成像
将实施例1制备的纳米粒OIRAI-NPs、OIR-NPs、OAI-NPs及PBS进行体外光声成像研究。经PSDT处理前后的成像如图8所示:
由图8分析可知,经PSDT处理后的PBS组,在B模式、CEUS模式和PA模式中均没有成像增强。经PSDT处理后的OAI-NPs组,在B模式、CEUS模式出现了成像增强现象,但PA模式中没有出现明显的成像增强,这种现象可能是由于经PSDT处理后,经近红外激光照射后轻微产热生成而导致OAI-NPs相变所致。而OIRAI-NPs、OIR-NPs组的超声信号B模式、CEUS模式和PA模式下较其他组显著增强,即为通过近红外光激发IR780引起纳米粒温度升高,超过一定温度(49℃)使纳米粒内的PFP产生大量的汽化,引起多模态成像的增强。
与声致相变(Acousticdropletvaporization,ADV)不同,光致相变(Opticaldroplet vaporization,OVD)利用激光照射纳米粒,其中的光吸收子吸收光能进而转化成热能,最终致使纳米粒内的PFP发生汽化。该实验结果表明相变材料携氧全氟戊烷(PFP)的成功包封,也为后续的双模态成像奠定了基础。
实施例3载IR780和氢氧化铝靶向抗原捕获纳米粒的体外光热效果
激光(辐照条件:1.5W/cm25min)辐照OIRAI-NPs后,OIRAI-NPs的升温速度与浓度呈正相关,而对照组PBS组,温度基本保持不变(图9)。
高效的光热转换剂是进行光致相变(Opticaldropletvaporization,OVD)的前提。实验结果验证了OIRAI-NPs作为光热转换剂的潜能。研究报道,肿瘤细胞持续暴露于高温状态至一定时间后便会发生细胞消融。因此,该实验结果表明OIRAI-NPs具有一定的抗肿瘤潜能。
试验例1载IR780和氢氧化铝靶向抗原捕获纳米粒的体外吞噬和杀伤实验
有效的细胞内摄取OIRAI-NPs纳米粒是提高其杀灭TC-1肿瘤细胞效果的先决条件。将DiI标记的OIRAI-NPs、OIR-NPs、OAI-NPs及PBS与宫颈癌细胞TC-1共孵育4h后,在激光共聚焦扫描显微镜(CLSM)下观察到DiI标记的OIRAI-NPs在TC-1肿瘤细胞内高效积聚(图10)。进一步发现,在OIRAI-NPs浓度在5.0mg/ml内,对肿瘤细胞无毒性作用,如CCK8所示(图11)。
进一步观察了OIRAI-NPs纳米粒对TC-1肿瘤细胞的杀伤效应,用不同的分组处理肿瘤细胞,分组如下:Control组、OIRAI-NPs组、OIR-NPs组、OAI-NPs组、Control+PSDT组、OIRAI-NPs+PSDT组、OIR-NPs+PSDT组OAI-NPs+PSDT组等。Control组、OAI-NPs组几乎对肿瘤细胞无任何杀伤作用,OIR-NPs组对肿瘤细胞具有极小的杀伤作用,而OIRAI-NPs+PSDT组对肿瘤细胞的杀伤作用最大,明显好于OIR-NPs+PSDT组和OAI-NPs+PSDT组。如CCK8(图12)所示。说明IR780和氢氧化铝在肿瘤杀伤方面具有协同增效作用。激光共聚焦扫描显微镜图如图13所示。
试验例2载IR780和氢氧化铝靶向抗原捕获纳米粒的活性氧产生能力
吞噬了纳米粒OIRAI-NPs、OIR-NPs的肿瘤细胞经过PSDT处理后,胞内活性氧显著增加,与2,7-二氯二氢荧光素二乙酸酯(DCFH-DA)探针结合后,可转化为荧光2,7-二氯荧光素(DCF),表现为CLSM下的强绿色荧光。如图14所示,单独的纳米粒(control组)或者PSDT处理细胞(control+PSDT组)内ROS的干扰作用有限。相比之下,OIRAI-NPs+PSDT组和OIR-NPs+PSDT组的ROS水平升高,而OIRAI-NPs+PSDT组的ROS水平升高最为显著,明显高于OIR-NPs+PSDT组,从DCF通道观察到强烈的绿色荧光,也证实了我们的OIRAI-NPs+PSDT组的疗效显著。
试验例3载IR780和氢氧化铝靶向抗原捕获纳米粒的亚细胞损伤水平
分组如下:Control组、OIRAI-NPs组、OIR-NPs组、Control+PSDT组、OIRAI-NPs+PSDT组、OIR-NPs+PSDT组。不同分组诱导的细胞损伤通过JC-1测定试剂盒进一步评估,JC-1是一种膜渗透探针,用于基于监测线粒体膜电位(MMP)分析细胞凋亡状态。线粒体损伤导致MMP去极化后,JC-1聚集体(λem:585nm)可能从细胞器扩散到细胞质中,并分解成JC-1单体(λem:115nm),即为荧光从红色变为绿色。如图15所示,在未经过PSDT处理时,没有观察到线粒体损伤;经过PSDT处理后,Control+PSDT组组几乎没有产生绿色荧光,OIR-NPs+PSDT组产生少量至中量绿色荧光。而OIR-NPs+PSDT组则产生了大量的绿色荧光,表明线粒体膜电位的下降,肿瘤细胞处于凋亡早期。
IR780赋予了OIRAI-NPs选择性在肿瘤组织中聚集和向肿瘤组织深部穿透的能力。此外,在IR780的帮助下,OIRAI-NPs可以进一步在线粒体聚集。
试验例4载IR780和氢氧化铝靶向抗原捕获纳米粒结合PSDT处理促进树突状细胞的成熟
在Transwell实验中,通过对DC细胞的共刺激分子表达来评估捕获肿瘤抗原的纳米粒能否有效促进DC成熟。流式检测结果如图16所示。结果表明,与对照组(Control组)比较,OIRAI-NPs+PSDT组CD80/CD86的表达量显著增加,明显高于OIR-NPs+PSDT组。
APCs对抗原的识别、加工和提呈,是抗肿瘤免疫应答的第一阶段(感应阶段)。树突状细胞(DC)作为专职性抗原提呈细胞,通过捕获和交叉呈递肿瘤细胞释放的抗原并激活T细胞,是获得性免疫反应的关键启动者。由此可见,OIRAI-NPs结合光声治疗可以实现肿瘤抗原的充分暴露,且有效的诱导DC成熟,促进抗原的加工与提呈,从而激活抗肿瘤免疫反应。
对比例
采用CN115192707A中实施例1的方法制备纳米粒PIAO-NPs。按照试验例4的方法评估了PIAO-NPs纳米粒结合PSDT处理对树突状细胞成熟的效果,检测结果显示PIAO-NPs+PSDT组CD80/CD86的表达量为50.8,明显低于OIRAI-NPs+PSDT组的表达量74.8。这说明相对于CN115192707A中的纳米粒PIAO-NPs,实施例1制备的纳米粒OIRAI-NPs具有更强的激活肿瘤免疫应答反应的效果。并且,CN115192707A中的纳米粒使用了吲哚菁绿(ICG)及化疗药物奥沙利铂,虽然吲哚菁绿可用于光声成像和光热治疗,但ICG在水溶液中极易发生聚集和淬灭、稳定性差、无特定的功能团和细胞特异性,除此之外,ICG在体内代谢速度非常快,使其应用受到了限制。而近红外(NIR)染料IR780显示出优异的光敏性能,同时IR780和氢氧化铝的联合应用具有协同增效作用,共同促进了载IR780和氢氧化铝靶向抗原捕获纳米粒的抗肿瘤效果。化疗药物奥沙利铂会出现神经毒性、血液和冒肠道毒性、中性粒细朐减少、恶心和呕吐等副作用,这些副作用限制了其应用范围和治疗效果。而本申请未使用化疗药物,避免了化疗药物的副作用,应用范围更广泛。
最后说明的是,以上实施例仅用以说明本发明的技术方案而非限制,尽管参照较佳实施例对本发明进行了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的宗旨和范围,其均应涵盖在本发明的权利要求范围当中。
Claims (10)
1.一种载IR780和氢氧化铝靶向抗原捕获纳米粒,包括核壳结构,所述核为携氧全氟戊烷,所述壳为聚乳酸-乙醇酸,所述聚乳酸-乙醇酸上负载有IR780和氢氧化铝。
2.如权利要求1所述的一种载IR780和氢氧化铝靶向抗原捕获纳米粒,其特征在于:所述载IR780和氢氧化铝靶向抗原捕获纳米粒的流体力学直径为257.133±3.82nm,表面电位为21.97±0.75mV。
3.如权利要求1所述的一种载IR780和氢氧化铝靶向抗原捕获纳米粒,其特征在于:所述载IR780和氢氧化铝靶向抗原捕获纳米粒中氢氧化铝的包封率为14.19±1.45%,载药量为0.27±0.03%;IR780的包封率为79.53±0.47%,载药量为2.93±0.071%。
4.一种载IR780和氢氧化铝靶向抗原捕获纳米粒的制备方法,其特征在于,采用W/O/W双重乳化法进行制备,包括以下步骤:

6.如权利要求4所述的一种载IR780和氢氧化铝靶向抗原捕获纳米粒的制备方法,其特征在于,氢氧化铝水溶液的浓度为1mg/ml,携氧全氟戊烷、氢氧化铝水溶液、二氯甲烷的体积比为0.2-0.3:1-2:2。
7.如权利要求4所述的一种载IR780和氢氧化铝靶向抗原捕获纳米粒的制备方法,其特征在于,聚乙烯醇水溶液的浓度为5%,二氯甲烷与聚乙烯醇水溶液的体积比为2:5-7。
8.如权利要求4所述的一种载IR780和氢氧化铝靶向抗原捕获纳米粒的制备方法,其特征在于,步骤S1或步骤S2中,所述乳化为冰浴条件下,超声乳化3-5min。
9.权利要求1-3任一项所述的一种载IR780和氢氧化铝靶向抗原捕获纳米粒在制备治疗抗肿瘤药物中的应用。
10.如权利要求9所述的应用,其特征在于,所述肿瘤为宫颈癌。
