Description

Traditional Chinese medicine composition for treating cervical precancerous lesions
Technical Field
The invention relates to the field of traditional Chinese medicine compositions, and in particular relates to a traditional Chinese medicine composition for treating cervical precancerous lesions.
Background
Cervical cancer is one of the most common malignancies in women worldwide, second only to breast cancer, and is especially common in developing countries, first to various malignancies. The research on the occurrence and development of cervical cancer changes from quantitative change to qualitative change, and the occurrence and development of the cervical cancer is a continuous process of in-situ cancer, early stage infiltration cancer and infiltration cancer caused by the development of precancerous lesion, generally lasts for 5-10 years, if timely treatment can be found in the early stage, the prognosis of the cervical cancer is good, and the 5-year survival rate of patients in the Ia and Ib stages can reach 80% -100%, so that the prevention of the precancerous lesion has important clinical value for reducing the occurrence of the cervical cancer.
In recent years, three breakthrough advances have been made in the worldwide research on the prevention of cervical cancer: (1) Establishes that high-risk Human Papilloma Virus (HPV) infection is a necessary condition for the generation of cervical cancer; (2) The newly developed thin-layer liquid-based cytology and the new technology (the second generation hybridization capture test) for detecting the HPVDNA remarkably improve the sensitivity of identifying the cervical advanced lesion, greatly reduce the false negative, and can detect more than 98 percent of early patients by reasonably applying a screening method; (3) HPV prophylactic vaccines have been approved for clinical validation and are undergoing phase three clinical trials. The injection vaccine is a main prevention measure for cervical HPV infection in modern medicine, but the vaccine is still in shortage in most areas in China at present, and the vaccine is expensive in price, short in prevention period and needs to be injected repeatedly. For patients in the pre-cervical cancer lesion stage, the main treatment means is injection of antitumor drugs such as interferon and cisplatin and operation modes such as cervical ring-cutting, wherein the antitumor drugs have large toxic and side effects, long treatment course, low cure rate and difficult popularization; the cervical ring incision requires surgery, and the sequelae are severe.
Patients with cervical cancer-related diseases generally do not show obvious symptoms, or only have symptoms of general cervicitis, such as leucorrhea increase, and complaints about leucorrhea blood or little vaginal bleeding after sexual contact. It belongs to the categories of leukorrhagia, metrorrhagia, abdominal mass and the like in the field of traditional Chinese medicine, and is formed by considering that the core pathogenesis of the disease is the deficiency of healthy qi and the accumulation of damp-heat and blood stasis toxin in the uterus subphylum due to the external infection of damp-heat and toxin and dirt. The therapeutic principle should be heat-clearing and detoxicating, promoting blood circulation and resolving hard mass, removing putrefaction and promoting granulation. For example, patent application CN113456730A discloses a traditional Chinese medicine composition for treating cervical precancerous lesion caused by HPV infection, which is prepared from salvia miltiorrhiza, sophora flavescens, radix sileris, verbena, fresh stiff silkworm and fresh centipede through separation and purification technical design, so that the problem of poor stability caused by polypeptide, phenolic acid and alkaloid components is solved, and meanwhile, the formula is mainly prepared from dampness-heat-toxin clearing and stasis-removing herbs, so that the negative conversion rate of HPV infection can be greatly improved. However, the composition contains a lot of cold components, and long-term administration of the composition may damage female health and is not beneficial to disease recovery.

The comparative example provides a traditional Chinese medicine composition for treating cervical precancerous lesions, and compared with example 1, the difference is only that the dragon blood is replaced by the pseudo-ginseng.
Experimental example 1
The activity of the composition for inhibiting the proliferation of Hela cells is examined, and the specific method is as follows:
the cell proliferation inhibition rate of Hela was measured by MTT method (cell proliferation and cytotoxicity assay kit, available from Shanghai Shangbao Biotech Co., ltd., product number 20311). Preparing Hela cells in logarithmic growth phase into single cell suspension, and regulating cell density to 2 × 10 5 200 μ L per well of 96-well culture plate, at 37 deg.C, 5% 2 The culture box is used for culturing for 4 hours.
Serum-free culture medium is added into the blank control group, the positive control group is cisplatin solution with the concentration of 10ug/ml, and the examples 1 to 3 are oral liquid of the examples 1 to 3 respectively; comparative examples 1-2 are oral liquids of comparative examples 1 and 2, respectively. mu.L of the above sample was added to each well, and after culturing for 72 hours, 20. Mu.L of MTT (5 g/L) was added, and the mixture was incubated at 37 ℃ for 4 hours, and the supernatant was discarded, 200. Mu.L of DMSO was added to each well, and the crystals were dissolved by shaking, and the absorbance (A) value at 490nm was measured to calculate the cell growth inhibition ratio.
Cell growth inhibition = (1-sample a value/blank a value) × 100%, the results are shown in table 1.
TABLE 1 inhibition of Hela cell proliferation (%)
Inhibition ratio (%)
Blank control group 0
Positive control group 70.47±3.09
Example 1 79.44±2.73
Example 2 80.03±1.97
Example 3 77.79±2.55
Comparative example 1 62.19±3.17
Comparative example 2 54.73±2.29
The data in Table 1 show that the composition can effectively inhibit the proliferation of Hela cells, has the effect equivalent to or even higher than that of cisplatin, and can replace the cisplatin for treatment so as to avoid the toxic and side effects of the cisplatin. In addition, after the rhizoma atractylodis is replaced by the rhizoma atractylodis macrocephalae and the dragon's blood is replaced by the pseudo-ginseng, the effect of the composition for inhibiting the proliferation of Hela cells is obviously reduced, which shows that the curative effect is poor, so that the components of the composition provided by the invention have a synergistic effect, and the replacement of the components can cause the reduction of the curative effect.
Experimental example 2
The effect of the composition of the present invention in clinical application was examined.
100 cases of cervical cancer pre-lesion patients caused by HPV infection (through liquid-based ultrathin technology, colposcopy and histopathological detection, the pathological result is clear as the cervical cancer pre-lesion) are selected and randomly divided into four groups: example 1, cisplatin group, 50 people each, patients between each group had no significant difference in age, gender (P > 0.05), and were comparable.
Example 1 the oral liquid prepared in example 1 was taken 1 pill a day 3 times a day after meals for 3 months. Cis-platinum group: dissolving with normal saline before application, each time according to body surface area of 20mg/m 2 The administration is repeated for 5 days at 2 weeks intervals. The results are shown in Table 2.
Treatment criteria are as follows:
(1) The method has the following advantages: both TCT and HPV detection were negative. Colposcopy, e.g., no abnormal findings under the scope or no presence of CIN as confirmed by biopathological examination, was also interpreted as effective.
(2) And (4) invalidation: grade CIN1 or grade CIN2 of pathological diagnosis remains unchanged after biopsy.
TABLE 2 clinical results of the compositions of the present application for the treatment of pre-cervical lesions

Example 1 38 12 76%
Cis platinum group 35 15 70%
The data in table 2 show that the effect of the traditional Chinese medicine composition for clinically treating cervical precancerous lesions is equivalent to that of a cisplatin medicine, but compared with the cisplatin medicine, the traditional Chinese medicine composition provided by the invention is high in safety, small in toxic and side effects and wide in application prospect.
The present invention is not limited to the above preferred embodiments, and any modifications, equivalent substitutions, improvements, etc. within the spirit and principle of the present invention should be included in the protection scope of the present invention.
