Abstract
The present invention provides a fixed bed bioreactor comprising: the device comprises a tank body, a centrifugal stirrer, a top cover and a disc type carrier; the disc type carrier is arranged in the tank body, a through hole is formed in the circle center of the disc type carrier, and one end of a stirring shaft of the centrifugal stirrer penetrates through the through hole of the disc type carrier. The disc-type carriers are directly arranged in the tank body, the internal space of the tank body can be fully utilized, and the disc-type carriers are not stacked together, so that the resistance of the flowing nutrient solution is small, the flow velocity of the nutrient solution is fixed, the flow direction of the nutrient solution is consistent, no turbulent flow is formed, the nutrient substances are uniformly distributed, and the growth state of cells loaded on the disc-type carriers is consistent. In addition, when the cells are digested, the pancreatin can also uniformly flow through the disc carrier along with the nutrient solution, thereby facilitating the separation of the cells from the disc carrier.

Technical Field
The invention relates to the technical field of bioreactors, in particular to a fixed bed bioreactor.
Background
According to the cell characteristics, adherent cells and suspension cells are generally divided, wherein the adherent cells need to be attached to a carrier for growth, and the suspension cells can independently grow. The carriers for adherent cell culture are generally classified into spherical carriers and sheet carriers. In order to meet the requirements of carrier surface area, cell density, cell activity and the like, the spherical carrier is generally a 100-120 mu m small sphere, and the sheet-shaped carrier is generally a strip-shaped or round paper sheet.
In the existing fixed bed bioreactor, a fixed hanging basket is formed by two mesh baffles and a tank body, a flaky carrier is arranged in the fixed hanging basket, adherent cells and a culture medium flow through the fixed hanging basket through a bioreactor stirring system, the adherent cells are adsorbed on the flaky carrier in the fixed hanging basket due to the adherent capacity of the adherent cells, and the culture medium flows through the flaky carrier through the bioreactor stirring system and provides nutrition for the cells, so that the growth and proliferation of the cells are maintained. However, in the prior art, the sheet-shaped carriers are limited in arrangement in the fixed hanging basket, and can only be stacked together in multiple layers, and the sheet-shaped carriers are tightly extruded, so that the resistance of cells to be cultured and a culture medium is large when the cells to be cultured and the culture medium flow through the sheet-shaped carriers, the flow path is short, and the cells and the culture medium are difficult to be uniformly distributed inside and outside. Meanwhile, because adherent cells are adsorbed on the closely stacked sheet-shaped carriers, the cells are difficult to separate from the carriers during digestion. In addition, the conventional fixed-bed bioreactor has the problems that the carrier loading is inconvenient and the cells are difficult to expand again. The common defects of the spherical carrier and the flaky carrier are higher cost, poor repeated use effect and complex digestion and amplification operation procedures.
Disclosure of Invention
In view of the above, the invention provides a fixed bed bioreactor, and the invention optimizes the arrangement of carriers by designing a tank body and a disc type carrier with a brand new structure, thereby optimizing the growth of adherent cells on the carriers and the function of digesting the cells by the fixed bed bioreactor.

For a further understanding of the invention, reference will now be made to the preferred embodiments of the invention in conjunction with the following examples, but it will be understood that the description is intended to illustrate the features and advantages of the invention further, and not to limit the invention.
The fixed-bed bioreactor of the present invention is further described below with reference to the accompanying drawings.
In a first aspect, the present invention provides a fixed bed bioreactor, as shown in fig. 1 and 2, comprising: a tank body 1, a centrifugal stirrer 2, a top cover 3 and a disc type carrier 4. The tank body 1 is cylindrical, and the top of the tank body 1 is provided with an upward opening; the top cover 3 covers the opening, the top cover 3 is connected with the tank body 1 in a sealing mode, and a mounting hole is formed in the top cover 3; the centrifugal stirrer 2 comprises a stirring shaft and stirring blades, the stirring blades are arranged on the stirring shaft, one end of the stirring shaft extends into the tank body 1 from the mounting hole, and one end of the stirring shaft and the stirring blades are positioned in the tank body 1; the disc-type carriers 4 may have a net-shaped structure or a pore-shaped structure to increase a contact area, promote the flow of a nutrient solution, and facilitate the uniform growth of cells. Specifically, unlike the conventional fixed bed bioreactor in which a fixed basket is provided and a disc-shaped carrier is placed in the fixed basket, the present invention provides a cylindrical tank 1 and a disc-shaped carrier 4 corresponding to the shape of the tank 1. The disc carriers 4 of the present invention are directly installed inside the can body 1, the inner space of the can body 1 can be fully utilized, and the disc carriers 4 are not stacked together. The centrifugal stirrer 2 can drive liquid in the tank body to flow when being started, the liquid can be selected as nutrient solution, and the disc type carriers 4 in the invention are directly arranged in the tank body 1 and the disc type carriers 4 are not stacked together, so that the resistance of the flowing nutrient solution is small, the fixed flow speed and the consistent flow direction of the nutrient solution can be ensured, the turbulent flow can not be formed, the uniform distribution of nutrient substances can be ensured, and the consistent growth state of cells loaded on the disc type carriers 4 can be ensured. In addition, when digesting cells, pancreatin put in the tank body 1 can also flow through the disc carrier 4 uniformly with the nutrient solution, thereby facilitating the separation of the cells from the disc carrier 4. Finally, the fixed-bed bioreactor of the present invention also facilitates cell expansion.
According to some embodiments of the invention, the stirring blade is located above the disc support 4. The stirring paddle of the present invention circulates the nutrient solution by centrifugal stirring so that the cells on the disc support 4 can contact fresh nutrients.
According to some embodiments of the present invention, the number of the disc carriers 4 is multiple, multiple disc carriers 4 are sequentially arranged along the length direction of the tank, two adjacent disc carriers 4 can be spaced apart or contacted, and the spacing distance between two adjacent disc carriers 4 can be adjusted, so as to be adjusted as required to meet the needs of actual conditions. According to the invention, the inner wall of the tank body 1 is provided with a plurality of bayonets, the disc-type carriers 4 are fixed in the tank body 1 through the bayonets, and a plurality of disc-type carriers 4 can be arranged in the tank body 1 according to the length of the tank body 1 and the spacing distance between the disc-type carriers 4, so that adherent cells can be cultured in a large quantity.

The nutrient solution 5 of the invention circulates in the tank body 1 under the drive of the centrifugal stirrer 2 and flows through the disc-type carrier 4.
According to some embodiments of the invention, the centrifugal stirrer 2 further comprises: a power mechanism 21, a connecting piece 22 and a sealing piece 23; at least part of the sealing element 23 passes through the mounting hole and seals a gap between the inner side wall of the mounting hole and the stirring shaft; the lower end of the connecting piece 22 is connected with the other end of the stirring shaft, and the upper end of the connecting piece 22 is connected with the power mechanism 21. The centrifugal stirrer 2 comprises a power mechanism 21, a connecting piece 22 and a sealing piece 23 besides a stirring shaft and a stirring blade, wherein the power mechanism 21 provides stirring power for the centrifugal stirrer 2, and the sealing piece 23 realizes the sealing performance of the top cover 3.

The diversion holes 41 on two adjacent disc carriers 4 can be arranged in a staggered way, so that the uniform flow of the nutrient solution 5 can be promoted.
Specifically, as shown in fig. 3, the disc-type carriers 4 are provided with a plurality of guiding holes 41, and the guiding holes 41 are provided to reduce the resistance of the nutrient solution 5 flowing between the disc-type carriers 4 and ensure the uniform circulation of the nutrient solution 5. 5. A fixed bed bioreactor as set forth in claim 1 wherein said centrifugal agitator further comprises:
a power mechanism, a connecting piece and a sealing piece; at least part of the sealing element penetrates through the mounting hole and seals a gap between the inner side wall of the mounting hole and the stirring shaft;
the lower end of the connecting piece is connected with the other end of the stirring shaft, and the upper end of the connecting piece is connected with the power mechanism.
6. A fixed bed bioreactor as claimed in claim 1 wherein the disc support is provided with a plurality of flow directing holes extending through the disc support in the axial direction of the disc support.
7. A fixed bed bioreactor as claimed in claim 6 wherein a plurality of said deflector holes are provided at intervals along the circumference of said through holes of said disc carrier; and/or
