概述

根据本公开的一个示例性实施方案,一种方法可包括使用光学成像技术测量生殖细胞结构的生物力学性质,和基于测得的生物力学性质评估生殖细胞结构实现成功妊娠的潜力。
该方法可以单独或以它们的任何组合包括一个或多个下列特征。光学成像技术可基于布里渊光谱法测量生物力学性质。生物力学性质可包括弹性模量或粘性模量。生殖细胞结构可包括选自胚胎、桑椹胚、囊胚、原肠胚、合子、卵子和卵母细胞中的一种。

本公开至少部分地基于以下认识:布里渊光谱法可用于量化生殖细胞结构的一种或多种生物力学性质(在本文中也被称为生物力学参数)。在本公开的实施方案中,基于布里渊光谱法的方法和系统可用于监测和/或评估卵子和/或胚胎。使用光学方法,可以以非接触方式、实时和感应地局部检查或评估生殖细胞结构。
布里渊光谱法基于布里渊散射,其中与从外部光源入射的光子相比,非弹性散射的光子(通常是反向散射的光子)发生频移。如下文论述,本公开提供通过测量响应于用询问辐射照射生殖细胞结构而由生殖细胞结构散射的辐射相关的至少一个布里渊频移来确定生殖细胞结构的一种或多种生物力学性质的方法。
体外受精(IVF)涉及一系列复杂的程序以帮助生育力来辅助受孕,同时防止遗传问题。图1显示通过Intracytoplasmic Sperm Injection(卵胞浆内单精子注射)(ICSI)进行单精子注射的IVF程序的示意图,其中将单精子细胞被直接注射到卵母细胞的细胞质中。在一种示例性程序中,从卵巢收集(例如收取)的成熟卵子可在体外(即,在实验室环境中)被精子人工受精。然后可将(一个或多个)受精卵或(一个或多个)胚胎(其可能被称为原胚(proembyo))转移到子宫。
IVF的成功率(例如导致怀孕)取决于许多因素。在Loendersloot等人的研究中,作者发现了怀孕与各种预测因素之间的相关性,包括女性年龄、低生育力的持续时间、基础促卵泡激素(FSH)、收取的健康卵母细胞的数量、转移的胚胎数量和胚胎质量(Loendersloot等人, 2010;Loendersloot等人, 2014)。该发现与其它研究一致,这得出的结论是胚胎质量本身是重要的预测因子,仅次于年龄(McKenzie等人, 2004)。
因此,在IVF中,临床医生和患者经常面临不仅选择最佳卵母细胞来进行受精还要选择最佳胚胎进行转移的挑战。尽管染色体的形态和存在正常,但许多胚胎在植入过程期间失败。为了弥补该失败,临床医生通常在超过60%的病例中转移多个胚胎("NationalSummary Report," 2016)。但是,这可能导致高比率的多胞胎和相关并发症,以及随之而来的风险,如除了预料外的经济负担外,还有不良围产期和母体预后。目前最可靠的方法在给定正确年龄框架下只能在60-70%的时间成功地预测妊娠(Forman等人, 2013)。
已经将大量的努力投入到识别卵母细胞和胚胎二者发育潜力的客观、定量和可靠的标志物。近年来,如延长培养(extended culture)(Forman等人, 2013)、植入前遗传筛选(PGS)(Gardner等人, 2015)和细胞周期的延时成像(VerMilyea等人, 2014)之类的技术已经帮助识别存活胚胎并降低多胎妊娠受孕的发生率,并且有助于选择性单胎移植的更高比率。为了更高的植入率,引入胚胎到囊胚期的延长培养(Jones等人, 1999)。但是,培养时间越长,需要的时间和资源越多,且因此该技术仅用于小部分患者(Forman等人, 2013)。此外,相关的长期风险是不清楚的,并且囊胚转移后单卵双生的风险增加。PGS可以筛选胚胎的染色体异常。这是基于先前的发现,即在自然流产中非整倍性高(Hassold等人, 1980),并且从分裂期到囊胚期观察到染色体异常的减少(Ata等人, 2012)。后来的研究证实非整倍体胚胎是大多数失败的IVF周期的原因,特别是与母体年龄增加相关的那些(Harton等人, 2013)。尽管随机对照试验证实PGS可提高植入和活产的成功率,但其是侵入式的,并且需要训练有素的胚胎学家进行滋养外胚层活检,以致该过程昂贵并且资源需求高。
延时成像能够识别长时间段上的动力学参数并有助于提高胚胎选择的成功率(VerMilyea等人, 2014)。在人类中,发现各种卵母细胞形态学参数与胚胎发育和植入潜力相关,包括透明带厚度、粒度、卵周隙和卵母细胞形状。基于形态学的胚胎选择是临床实践中最广泛使用的方法之一。但是,这些特征是高度主观的,并且它们的预测值是不确定的。结果与染色体状态几乎没有相关性(Werner等人, 2012)。一些研究表明胚胎的命运在发育阶段早期,甚至在受精前确定(Stitzel等人, 2007;Li等人, 2010)。因此,为了产生更高的植入率和活产率并降低不想要的多胎妊娠的发生率,对卵母细胞/胚胎质量的标志物的追求仍然是IVF领域中的主要课题。
力学性质对于在分子水平上调节细胞命运和功能起到重要作用,并且细胞的内部状态可反映在其力学性质中(Xu等人, 2012;Suresh等人, 2005)。生物力学性质对卵母细胞和胚胎功能可以也是关键的。胚胎和卵母细胞的生物力学与人类的妊娠和小鼠的母体年龄相关联,表明力学和生存力之间的联系(Ebner等人, 2003;Murayama等人, 2008;Murayama等人, 2006)。
例如,如图3(其显示合子的结构和受精程序)中所示,随着卵母细胞随其发育成合子通过有丝分裂的不同阶段而转变,透明带(其是硫酸化糖蛋白的网络)可发生显著变化。合子在成熟过程中可变软,这可能是促进精子穿入的机制(Papi等人, 2010)。随后,合子在受精过程中可变硬。这可归因于皮质反应,其中皮质颗粒将它们的内容物释放到卵周隙中。酶介导机制可能够防止多精入卵(Drobnis等人, 1988)。透明带结构和厚度的差异与卵母细胞的生殖能力相关。较薄的透明带或无法区分的内层可指示较低的受孕率或囊胚发育率(Ebner等人, 2010)。如这些发现所示,卵母细胞或胚胎的生物力学性质可以是用于卵母细胞或胚胎选择的有效措施。
透明带物理性质的改变可伴随着在卵泡发生和成熟过程中的卵母细胞细胞质粘度(Krause等人, 2016)。一些研究发现较高粘度或注射漏斗持久性(injection funnelpersistence)与随后的植入前发育中的不良预后之间的相关性(Ebner等人, 2003)。
Yanez等人(2016)研究了基于Zener模型的力学参数(Bausch等人, 1999)对预测在人和小鼠合子中形成囊胚的成功率的潜力。使用分类器预测人胚胎囊胚,他们实现了90%的精确度、95%的特异性和75%的灵敏度。因此,在合子阶段,力学参数可提供关于胚胎生存力的信息。
在培养中可需要额外48小时以收集延时(time-lapse)细胞周期形态学参数。力学和细胞周期参数的最佳组合可实现90%和91%的灵敏度和特异性。作者还显示,存活组与非存活组具有显著不同的转录组。这些结果表明胚胎潜力主要取决于受精前的卵母细胞质量和成熟度,并且还表明可基于力学指标预测卵母细胞生存力的潜力。
致力于开发测量卵母细胞力学性质的技术,主要是使用微流体方法。目前量化卵母细胞和胚胎的力学性质的方法可包括压缩(compression)、压痕(indentation)、抽吸(aspiration)等。
压缩 – 这一领域中最早的工作追溯到早在1970年代,并包括Nakamura等人(1978)和Nemoto等人(1980)进行的研究。这些研究使用平行板压缩和通过微量移液器抽吸施加负压以测量动物模型中的参数,包括细胞刚度、膜表面张力和细胞内压力。Nakahara等人使用力传感器进行透明带的延时顺序机械测量,以观察其硬化(Nakahara等人, 2018)。设计特殊的传感器系统以测量压缩卵母细胞时的阻力。法国的一个团队报道了与卵胞浆内单精子注射(intra-cytoplasmic sperm injection)方案中所用类似的力感测平台,用玻璃压头(glass indenter)代替注射移液器(Gana等人, 2017)。两个被动且线性-磁性弹簧用于测量施加于卵母细胞的纳米力。他们仅提供了来自未成熟卵母细胞(中期I)的初步结果。
压痕 – Liu等人报道了通过亚像素计算机视觉跟踪来量化卵母细胞对外部施加力的阻力的系统(Liu等人, 2010)。使用计算机视觉跟踪系统,成功地将健康小鼠卵母细胞与衰老诱发的细胞缺陷区分开。
抽吸 – 使用微量移液器抽吸(MPA),Evans等人研究了皮质张力,或用于使表面积/体积比最小化的皮层和上覆质膜中的力(Evans等人, 2018)。Yanez等人使用修正模型评估基于Zener模型的力学参数在对预测在人和小鼠合子中形成囊胚的成功率的潜力。
其它方法 – Wang等人设计了用于胚胎内导航和测量的三维磁镊系统(Wang等人,2017)。Wang等人使用该磁镊系统研究了小鼠胚胎。为了对小鼠胚胎的内细胞团(ICM)上的多个位置进行力学表征,通过用已知的力在胚胎内导航磁控微珠来测量胚胎内粘度。在Wang等人的研究中,力被精确施加到胚胎内的5 μm磁珠上。通过(一个或多个)磁珠的3D导航,估算细胞质粘度。作者得出结论是,小鼠胚胎中的粘度是水粘度的8倍。使用母牛和小母牛卵母细胞,用原子力显微术(AFM)来测量透明带的力学性质(Papi等,2010)。在这一研究中,研究人员报道了在卵母细胞成熟过程中透明带弹性的损失。Andolfi等人(2016)将这种方法推向临床研究。他们分析了来自14名患者的卵母细胞并发现具有阴性结果的合适的中期II表现比实现怀孕的卵母细胞更软的外层透明带。Hornick等人使用“刚性移液器(stiffpipette)”方法来研究减数分裂染色体的刚性(Hornick等人, 2015)。他们使用两个移液器来精确地拉伸小鼠模型的分离染色体的两端以测量阻力,并观察到较高年龄组和较低年龄组之间的显著差异,其中较高年龄组表现出较大的力常数和潜在较高的非整倍性概率。

考虑到前述技术需要,本公开的主题提供对生殖细胞结构基于它们的一种或多种生物力学性质的光学测量进行非接触式检查或监测的方法和系统。更具体地,本公开的方面可提供用于测定生殖细胞结构的一种或多种生物力学性质并任选利用该生物力学性质来选择更可能导致成功妊娠的那些生殖细胞结构的光学成像技术。在本文中,生殖细胞结构可包括胚胎、合子、卵子、卵母细胞等。但是,可使用本公开表征的生殖细胞结构不限于本文明确公开的那些,而是可用于在IVF过程中的任何阶段评估生殖细胞结构的生存力。在一些实施方案中,光学成像技术可实施为具有亚细胞分辨率。例如,光学成像技术可基于布里渊光谱法。
在一些实施方案中,光学成像系统可以是用于卵母细胞和胚胎成像的布里渊显微成像系统。该系统可以在亚细胞水平下以改善的精确度(例如约10 MHz或更低)和增强的分辨率(例如约3微米或更低)获得卵母细胞和胚胎的三维布里渊显微成像。布里渊显微成像系统可测量卵母细胞和胚胎的生物力学性质,而不会不利地影响它们用于后续发育和IVF的后续程序。

本公开的方面还包括基于布里渊量度(Brillouin metric)量化卵母细胞和胚胎的生存力。布里渊量度可充当存活生殖细胞结构(例如合子)的生物力学标志物。此外,布里渊测量可用作提供足够灵敏度以检测生物力学性质的量度。
此外,本公开的方面包括表征合子上的布里渊测量值以预测在动物或人类中的实施和活产成功性。例如,根据本公开基于布里渊光谱法导出的量度(metric)可用于预测卵母细胞和胚胎的使用将致使成功活产的可能性。换言之,卵母细胞和/或胚胎的一种或多种生物力学性质的布里渊测量可充当预测活产的生物力学标志物。
举例而言并且如下文更详细论述,在一些实施方案中,根据本公开通过布里渊散射辐射的分析获得的弹性模量可用于评估卵母细胞、合子和/或胚胎的刚度或坚实度(firmness),并且卵母细胞、合子和/或胚胎的刚度或坚实度可与成功妊娠相关联。在一些这样的实施方案中,生物力学测量可预测在动物或人类中的合子阶段的胚胎潜力。
根据本公开的相关方面,本公开可实现IVF的临床管理的显著增强。例如,可以利用基于布里渊的生物力学量度以对存活卵母细胞的选择做出更好的决定。可以更可靠地进行对捐卵的卵母细胞筛选以识别存活卵母细胞,这可增强受精过程。此外,受精前的卵母细胞和受精后的胚胎的定量生物力学评估可进一步改进存生存力和活产率,并可降低不想要的多胎妊娠的发生率。先进的布里渊技术和光学仪器通常可通过推进对卵母细胞和胚胎的力学或生物力学性质的基础理解而使生育诊所获益。此外,卵母细胞和胚胎的布里渊测量可引发对ART领域中的生物力学的更积极的探索。
下面将描述典型的IVF程序并解释本公开的主题在IVF程序过程中的重要性。在典型的IVF中,胚胎可以在受精后在受控实验室环境下生长2至3天。然后可将长成的胚胎转移到女性的子宫中。
为了提高成功率,胚胎可在体外培养直至胚胎准备好植入的稍后阶段。图2显示人胚胎的植入前发育。在受精后约90分钟,合子可分裂成两个细胞并进入双细胞卵裂球状态。其可被认为是受精卵母细胞的最早有丝分裂产物。这些有丝分裂可以继续,并导致被称为卵裂球的一组细胞。当合子含有16至32个细胞时,其可被称为桑椹胚。卵裂球从合子分裂可允许单个可育细胞继续分裂和分化,直至囊胚形式。
当胚胎选择方法是非侵入式的、便宜的、易于使用、对生存力有强预测力并适用于卵母细胞和胚胎两者时,存在许多优点。因此,在一些实施方案中,本公开通过布里渊显微术提供卵母细胞和胚胎的生物力学评估以用于胚胎选择。
如上所述,本公开的主题提供了对生殖细胞结构的一种或多种生物力学性质进行非侵入式测量的方法。特别地,根据本公开的方法可使用布里渊光谱法进行生殖细胞结构的生物力学性质,如它们的弹性模量和/或粘度模量的非侵入式和非接触式表征。因此,本公开的基于布里渊光谱法的方法和系统可允许对卵子和/或胚胎进行实时和感应地局部(例如以亚细胞分辨率)非侵入式检查。
在根据本公开的生殖细胞结构的布里渊光谱法中,入射在生殖细胞结构上的光子可通过与声学声子的相互作用而散射以产生能量略低或略高的光子。散射光子的能量(和因此频率)偏移因此与声学声子的能量有关,后者又可与生殖细胞结构的弹性模量有关。因此,如下文更详细论述,布里渊频移可用于获得生殖细胞结构的弹性模量的估算(例如多个亚细胞值的平均值)。
如图4中所示,可用具有中心光频率v0(以绿色显示)的入射光束照射样本(例如生殖细胞结构)。在与样本相互作用时,光可被划分成弹性散射部分和对应于布里渊散射辐射的非弹性散射部分。可以例如使用一个或多个滤光器消除具有频率v0的未被干扰的弹性散射部分,以展现非弹性散射的布里渊分量,其以蓝色和橙色(或以虚线)显示,分别具有频率v0+ΔvB和v0-ΔvB。可以使用光谱仪测量频移分量∆vB。入射光和散射光(例如激光束)的所得频移可限定布里渊光谱,其与样本探测位置处的纵向弹性模量直接相关。根据光源的频率,可以调谐测量分辨率。此外,可以使用布里渊成像在整个样本上映射出机械性质的空间分布。
在一些实施方案中,辐射源可以是提供具有约400 nm至约800 nm的真空波长和约1 mW至约100 mW的辐射输出功率的辐射的激光器。例如,该激光器可具有约400 nm、约425nm、约450 nm、约475 nm(蓝)、约500 nm、约525 nm(绿)、约550 nm、约575 nm、约600 nm、约625 nm、约650 nm、约675 nm、约700 nm(红)、约725 nm、约750 nm、约775 nm或约780的波长。例如,该激光器可具有约1 mW、约2.5 mW、约5 mW、约7.5 mW、约10 mW、约20 mW、约30mW、约40 mW、约50 mW、约75 mW或约100 mW的功率。
根据本公开的方法测得的布里渊散射偏移∆vB可为约5 GHz至约15 GHz。例如,布里渊散射分量∆vB可为约5 GHz、约6 GHz、约7 GHz、约8 GHz、约9 GHz、约10 GHz、约11GHz、约12 GHz、约13 GHz、约14 GHz或约15 GHz,取决于例如所研究的生殖细胞结构(例如卵母细胞或胚胎)。
根据本公开,布里渊频移可用约0.05 GHz(即50 MHz)或更低的最低检测分辨率测量。例如,检测分辨率可为约10 MHz或更低。由于这样的精确度水平,根据本公开的光学测量技术可提供可应用于人类细胞的可靠测量。
测得的布里渊散射偏移∆vB可通过以下公式与(高频率)纵向储能模量(也被称为“弹性模量”)M'相关联,M'是纵向上的应力与纵向上的应变的比率。
其中n是介质,例如生殖细胞的折射率;λ是源光的真空波长;ρ是介质,例如生殖细胞的密度;且θ是入射光与散射光之间的角度。对于反向散射,θ为约180 deg,且sin(θ/2)变成约1。相应地,纵向模量M'可通过以下公式获得。
纵向储能模量M'又可通过以下公式与杨氏模量E'相关联。

其中M'是储能模量,且M''是损耗模量。储能模量测量储存的能量(弹性部分),并且损耗模量测量作为热耗散的能量,其量化复数纵向模量的粘性部分。自发布里渊光散射来源于光子与声学声子的相互作用。相互作用可被理解为通过在介质中由传播热力学波动的压力波引起的折射率调制而光散射。由于波动是随机的,波具有白谱(即,全频率分量)并以声波速度在所有方向上传播。从相位匹配声波散射的光经历多普勒频移,其幅值等于机械波的频率,通常为5至10 GHz。频率可容易地用布里渊光谱法测量。在一系列求导程序中,本构公式的最终形式可表示为,
如上所述,弹性部分的储能模量是布里渊频移的函数(),且粘性部分的损耗模量是布里渊频移和布里渊峰线宽的函数()。和都可通过布里渊显微测量法测量并随后进行数值分析。使用布里渊光谱仪的布里渊频移和峰线宽测量可受到光谱上的光学分辨率的影响。可通过使用两个虚像相位阵列(VIPA)光谱仪使用测得的系统响应对该光谱进行去卷积而实现这两个变量的精确测量。
如上所述,可通过入射辐射的2D和/或3D扫描和检测来自扫描位置的布里渊散射辐射来获得空间分辨的弹性模量映射。使用空间分辨的和/或代表性的(例如整体或全局)弹性模量,可以评估卵母细胞或胚胎的生存力指数。此外,在一些实施方案中,基于布里渊的生存力指数可以普遍适用。在其它实施方案中,基于布里渊的生存力指数可结合个人因素,如种族、病史等进行评估。
下面将描述根据本公开的测量生殖细胞结构的生物力学性质并使用测得的生物力学性质以识别、选择或筛选存活的生殖细胞结构的方法和系统的示例性实施方案。
图5显示根据本公开的一个示例性实施方案的对生殖细胞结构的生物力学性质进行光学检查或表征的流程图。对生殖细胞结构进行非接触式检查的方法可包括获得(例如收集)生殖细胞结构(S100),和使用光学成像技术测量生殖细胞结构的一个位置处的布里渊频移(S200)。参考图6,布里渊频移的测量(S200)可包括用辐射照射生殖细胞结构(S210),检测散射(例如反向散射)的辐射(S220),和例如使用光谱仪分析检测到的辐射以识别至少一个布里渊频移(S230)。
随后,可将测得的布里渊光谱与生殖细胞结构的生物力学性质相关联(S300)。在一些实施方案中,可以重复步骤S200和S300以获得生物力学性质在整个生殖细胞结构上的2D或3D映射。生物力学性质可进一步与生殖细胞结构的生育力相关联(S400)。基于确定的生育力,可以识别、选择或筛选生殖细胞结构以用于IVF中的进一步程序(S500)。
下面参考图6更详细地描述测量生殖细胞结构的一个位置处的布里渊频移的步骤(S200)。参考图6,可用具有至少一个所需频率分量的辐射照射生殖细胞结构(S210)。在一些实施方案中,照射辐射可以是激光辐射。进而可以检测从生殖细胞结构散射的至少一部分辐射(S220)。随后,可以分析检测到的散射辐射的频谱以识别频谱中的至少一个布里渊频移(例如下移或上移的频率分量)(S230)。在一些实施方案中,可以滤出从生殖细胞结构弹性散射的辐射以促进布里渊频移的检测。
在一些实施方案中,提供生殖细胞结构的步骤(S100)可进一步包括固定生殖细胞结构。例如,可以通过机械保持、电磁保持或流体动力学保持将生殖细胞结构固定以供光学观察。在其它实施方案中,可以移动布里渊光谱仪的样品容器和/或光学组件,使得将所观察的细胞定位在最佳观察位置。例如,可以基于图像识别技术实施软件算法以移动布里渊光谱仪的样品容器和/或光学组件。在这样的实施方案中,图像识别可基于神经网络算法来确认细胞及其范围。一旦被确认,可以使用机动载物台将样品保持在视线中心,或者可以基于样品位置动态更新扫描仪设置。图像识别技术可加速作为测量的第一步骤的样品初始定位。
待测量的生殖细胞结构可包括胚胎、合子、卵子、卵母细胞等。在一些实施方案中,可以测量卵母细胞的生物力学性质以在多个卵母细胞中选择要用雄配子受精的最具生存力的(一个或多个)卵母细胞。在一些实施方案中,可以测量胚胎的生物力学性质以选择最具生存力的(一个或多个)胚胎以转移到子宫。但是,根据本公开的光学检查不限于卵母细胞和胚胎的选择,而是也可在体外受精的各种阶段的过程中应用。
生殖细胞结构的生育力可通过生存力(viability)、活产率、发育潜力等表示。但是,生殖细胞结构的生育力不限于此,并且其可涉及指示成功受孕概率的各种量度。在一些实施方案中,生育力可被量化为生存力指数。例如,当测得的布里渊频移落在约5.0 GHz至约6.2 GHz之间时,可以确定生殖细胞具有高的活产生存力,且因此可将生存力指数评价为最高。生存力指数还可反映在一定时间跨度内作出的一系列测量,例如基于上述布里渊光谱法。生存力指数可进一步反映其它量度,如化学测量以及布里渊光谱法。
如上所述,使用空间分辨的和/或代表性的(例如整体)弹性模量,可以选择表现出最高生存力指数(例如落在预定杨氏模量范围或预定布里渊散射分量范围内)的卵母细胞以继续进行精子注射。例如,致使存活的可能性的杨氏模量范围可为约2 GPa至约6 GPa,其可对应于约25 GHz至约15 GHz的布里渊频移。在一些实施方案中,可以选择表现出最高生存力指数(例如落在预定杨氏模量范围或预定布里渊散射分量范围内)的胚胎以继续进行植入。
此外,在一些实施方案中,基于布里渊的生存力指数可以仅基于测得的生物力学性质应用。在其它实施方案中,基于布里渊的生存力指数可结合个人因素,如父母的年龄、种族、医疗状况/病史等进行评估。例如,在一些实施方案中,常规量度(例如父母的年龄、种族、医疗状况/病史等)可用于缩减卵母细胞到少量,然后可使用根据本公开的布里渊散射测量从通过常规受精参数初始选择的那些卵母细胞中选择卵母细胞。
图7图示根据本公开的一个示例性实施方案的用于对生殖细胞结构的生物力学性质进行光学检查和/或表征的系统10。如图7中所示,根据本公开的一个示例性实施方案的用于对生殖细胞结构进行光学检查的系统10可包括发光源(例如激光器)100,其生成用于照射样品20(例如生殖细胞结构)的辐射。所生成的辐射可被分色镜150反射到光学器件200(在该实施方案中为物镜的形式)上,其可进而将辐射聚焦到样品20上。光接收组件300可接收由生殖细胞结构散射的至少一部分光,所接收的光可通过光纤350传输,且光谱仪400可测量散射光的频移分量。
在一些实施方案中,为了阻挡离焦光,可将针孔250设置在光接收组件300的前面(即上游)。系统10可进一步包括在发光源100的前面(即下游)的另一针孔125。由于针孔250和/或125,系统10可调节焦平面30,且因此可提供用于2D或3D扫描的光学分层和垂直扫描。在一些实施方案中,使用共焦系统,可以以低于约5 μm的分辨率获得垂直扫描。例如,垂直扫描分辨率可为约5 µm、约4 µm、约3 µm、约2 µm或约1 µm。在一些实施方案中,可以使用单模光纤,尽管在其它实施方案中,多模光纤也可用作光纤350。由于其芯直径小,单模光纤可本质上充当针孔,且因此可兼具针孔250、光接收组件300和光纤350的功能。
在分析器700中,可将测得的频移分量与生殖细胞结构的生物力学性质相关联,随后可将生殖细胞结构的生物力学性质与生殖细胞结构的生育力相关联。分析器700可在硬件、软件和/或固件中以如本公开告知的本领域已知的方式实现。例如,分析器可包括处理器和与处理器通信的一个或多个存储器模块。根据本公开的用于将生殖细胞结构的一种或多种测得的生物力学性质与其可致使成功的IVF结果的可能性相关联的指令可存储在分析器的存储器模块中以在运行过程中通过处理器访问。
为了以扫描方式对样品20进行表征,该系统可进一步包括一个或多个致动器500。在图7中,致动器500被显示为耦合到含有样品20的样品支座25,并相对于平台600在水平面上平移样品支座25,以便用询问辐射照射样品20内的各个位置。在使用中,样本支座25可以是用于胚胎学工作流程的各种类型的皮氏培养皿,且致动器500和平台600可被设计为容纳各种类型的样本支座25。
但是,本公开不限于这样的配置,并且致动器500可以平移光学组件以形成光学组件和样品20之间的相对移动,以便照射样品的不同部分。在一些实施方案中,致动器500可以移动物镜200的位置和/或取向以执行扫描。在一些实施方案中,通过引发样品20和系统10的光学组件之间的相对水平移动,可用低于约5µm的分辨率获得水平扫描。例如,水平扫描分辨率可为约5 µm、约4 µm、约3 µm、约2 µm或约1 µm。
如上所述,该系统可进一步包括用于相对于平台600移动样品支座25和/或相对于样品20移动光学组件(例如物镜200)使得将细胞结构定位在最佳观察位置的装置。在一些这样的实施方案中,致动器可基于例如图像识别技术调节细胞结构的相对位置。
测得的频移分量可对应于布里渊光谱。此外,生物力学性质可包括生殖细胞结构的弹性模量或粘性模量。如上所述,用根据本公开的系统表征的生殖细胞结构可包括胚胎、合子、卵子、卵母细胞等。为了使该系统更有效地表征生殖细胞结构,光学系统可具有亚细胞分辨率。因此,可以测量特定位置或位点的生物力学性质,并且该系统可以以扫描方式检查生殖细胞结构,以允许将其生物力学性质在整个生殖细胞结构中映射出。在一些实施方案中,生物力学性质的代表性点测量可用于评估生殖细胞结构,或可采用全局生物力学性质(例如通过局部生物力学性质的空间平均获得的一种)。
如本文所述,本公开的主题提供通过光学表征生殖细胞结构,如卵母细胞和胚胎的一种或多种生物力学性质,如弹性模量和粘性模量来检查生殖细胞结构的能力。因此,可以以非侵入性地和以非接触方式检查生殖细胞结构,以选择最有生存力的样本以进一步继续进行IVF程序,从而潜在改进成功IVF的概率。

Claims (20)
1.一种测量生殖细胞结构的至少一种生物力学性质的方法,所述方法包括:
用辐射照射生殖细胞结构;
检测从受照射的生殖细胞结构散射的至少一部分辐射;
分析检测到的散射辐射的频谱以识别所述频谱中的至少一个布里渊频移;和
基于所述布里渊频移确定至少一种生物力学性质。
2.根据权利要求1所述的方法,其进一步包括:
基于所述至少一种生物力学性质确定所述生殖细胞结构的生存力指数。
3.根据权利要求1所述的方法,其中所述至少一种生物力学性质包括至少一部分生殖细胞结构的弹性模量、粘性模量或两者。
6.根据权利要求1所述的方法,其中所述照射辐射是激光辐射。
7.根据权利要求6所述的方法,其中所述激光辐射包括至少一个与约400 nm至约800nm的真空波长对应的频率分量。
8.根据权利要求1所述的方法,其进一步包括:
滤出从所述生殖细胞结构弹性散射的辐射以促进所述布里渊频移的检测。
9.根据权利要求1所述的方法,其中用亚细胞分辨率测定所述生殖细胞结构的所述至少一种生物力学性质。
10.根据权利要求9所述的方法,其进一步包括:
在所述生殖细胞结构的多个亚细胞位置处测定所述至少一种生物力学性质。
11.根据权利要求1所述的方法,其进一步包括:
利用光谱仪获得散射辐射的频谱。
12.一种用于测定生殖细胞结构的至少一种生物力学性质的系统,其包括:
辐射源,其用于生成照射辐射;
至少一个光学器件,其用于将所述照射辐射引导到至少一部分生殖细胞结构上;
检测器,其用于检测从所述生殖细胞结构散射的至少一部分辐射;
光谱仪,其用于生成检测到的散射辐射的频谱;和
分析器,其用于处理频谱以识别至少一个布里渊频移和利用所述布里渊频移测定所述至少一种生物力学性质。
13.根据权利要求12所述的系统,其进一步包括:
至少一个辐射收集光学器件,其用于通过光纤将所述至少一部分散射辐射引导到所述检测器。
14.根据权利要求13所述的系统,其进一步包括:
针孔,其设置在所述至少一个辐射收集光学器件上游以阻挡离焦光。
15.一种方法,其包括:
使用布里渊光谱法测量生殖细胞结构的至少一种生物力学性质;和
基于测得的至少一种生物力学性质确定生殖细胞结构的生存力指数。
16.根据权利要求15所述的方法,其中所述生物力学性质包括弹性模量、粘性模量或两者。
17.根据权利要求15所述的方法,其中所述生殖细胞结构包括选自胚胎、桑椹胚、囊胚、原肠胚、合子、卵子和卵母细胞中的一种。
18.根据权利要求15所述的方法,其中进行所述方法以选择要用雄配子受精的卵母细胞。
19.根据权利要求15所述的方法,其中进行所述方法以选择合子以进一步继续体外受精。
20.根据权利要求15所述的方法,其中进行所述方法以选择要转移到子宫的胚胎。
