Example 1 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 2 hours, concentrating and drying an extracting solution after the extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent is ethanol with the volume fraction of 95%.
Example 2 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 2 hours, concentrating and drying an extracting solution after the extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent consists of n-butanol and acetone in a volume ratio of 3:1.
Example 3 preparation of the composition
The raw materials comprise the following components in parts by weight: 20 parts of moringa seed, 20 parts of kudzuvine root, 15 parts of kudzuvine flower, 15 parts of hawthorn and 3 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 1h, concentrating and drying an extracting solution after extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 20mL; the organic solvent consists of n-butanol and acetone in a volume ratio of 4:1.
Example 4 preparation of the composition
The raw materials comprise the following components in parts by weight: 40 parts of moringa seed, 8 parts of kudzuvine root, 30 parts of kudzuvine flower, 30 parts of hawthorn and 10 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 3 hours, concentrating and drying an extracting solution after extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 10mL; the organic solvent consists of n-butanol and acetone in a volume ratio of 2:1.
Example 5 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps:
(1) Mixing Moringa seed, radix Puerariae, flos Puerariae Lobatae, fructus crataegi and Ginseng radix, adding organic solvent, heating to boiling state, reflux-extracting for 2 hr, concentrating and drying the extractive solution to obtain organic solvent extract; wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent consists of n-butanol and acetone in a volume ratio of 3:1;
(2) Loading the organic solvent extract onto macroporous resin column (macroporous resin column is filled with macroporous resin of HP-20, the weight of macroporous resin of HP-20 is 40 times of that of the organic solvent extract), eluting with ethanol water solution with volume fraction of 13% and 4 times of column volume to remove impurities; then eluting with ethanol water solution with volume fraction of 27% and 5 times of column volume, collecting eluate eluted with ethanol water solution with volume fraction of 27%, concentrating, and drying to obtain the composition.
Example 6 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 2 hours, concentrating and drying an extracting solution after the extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent is acetone.
Example 7 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 2 hours, concentrating and drying an extracting solution after the extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent is n-butanol.
Example 8 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 2 hours, concentrating and drying an extracting solution after the extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent is methanol with volume fraction.
Example 9 preparation of the composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps: mixing moringa seed, radix puerariae, flos puerariae lobatae, hawthorns and ginseng, adding an organic solvent, heating to a boiling state, carrying out heating reflux extraction for 2 hours, concentrating and drying an extracting solution after the extraction is finished to obtain an organic solvent extract, and obtaining the composition;
wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent consists of n-butanol and methanol in a volume ratio of 3:1.
Preparation of the composition of comparative example 1
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps:
(1) Mixing Moringa seed, radix Puerariae, flos Puerariae Lobatae, fructus crataegi and Ginseng radix, adding organic solvent, heating to boiling state, reflux-extracting for 2 hr, concentrating and drying the extractive solution to obtain organic solvent extract; wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent consists of n-butanol and acetone in a volume ratio of 3:1;
(2) Loading the organic solvent extract onto macroporous resin column (macroporous resin column is filled with macroporous resin of HP-20, the weight of macroporous resin of HP-20 is 40 times of that of the organic solvent extract), eluting with ethanol water solution with volume fraction of 5% and 4 times of column volume to remove impurities; then eluting with ethanol water solution with volume fraction of 15% and 5 times of column volume, collecting eluate eluted with ethanol water solution with volume fraction of 15%, concentrating, and drying to obtain the composition.
Comparative example 1 differs from example 5 in that the elution conditions of the macroporous resin column are different, and the elution conditions of comparative example 1 are: eluting with ethanol water solution with volume fraction of 5% to remove impurities; then eluting with 15% ethanol water solution; whereas the elution conditions for example 5 were: eluting with 13% ethanol water solution to remove impurities; then eluting with 27% ethanol water solution.
Preparation of comparative example 2 composition
The raw materials comprise the following components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng;
the preparation method comprises the following steps:
(1) Mixing Moringa seed, radix Puerariae, flos Puerariae Lobatae, fructus crataegi and Ginseng radix, adding organic solvent, heating to boiling state, reflux-extracting for 2 hr, concentrating and drying the extractive solution to obtain organic solvent extract; wherein the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1g to 15mL; the organic solvent consists of n-butanol and acetone in a volume ratio of 3:1;
(2) Loading the organic solvent extract onto macroporous resin column (macroporous resin column is filled with macroporous resin of HP-20, the weight of macroporous resin of HP-20 is 40 times of that of the organic solvent extract), eluting with ethanol water solution with volume fraction of 20% and 4 times of column volume to remove impurities; then eluting with ethanol water solution with volume fraction of 40% and 5 times of column volume, collecting eluate eluted with ethanol water solution with volume fraction of 40%, concentrating, and drying to obtain the composition.
Comparative example 2 differs from example 5 in that the elution conditions of the macroporous resin column are different, and the elution conditions of comparative example 2 are: eluting with 20% ethanol water solution to remove impurities; then eluting with 40% ethanol water solution; whereas the elution conditions for example 5 were: eluting with 13% ethanol water solution to remove impurities; then eluting with 27% ethanol water solution.
Experimental example 1
Taking male SD rats with the weight of 300 g+/-30 g, and dividing the male SD rats into 12 groups of 10 rats; wherein 11 groups are experimental groups, and 1 group is a model group; the experimental group irrigates the stomach with the compositions prepared in examples 1 to 9 of the invention or the compositions prepared in comparative examples 1 to 2 according to the dosage of 10mg/kg 1h before the gastric lavage of alcohol; filling the model group with distilled water with the same amount; after the administration of the lavage, the experimental group and the model group are filled with 38% alcohol according to the volume fraction of 1 ml/kg. 2 hours after stomach alcohol infusion, testing the alcohol content in the blood of the rat; the test results are shown in Table 1;
wherein, experimental groups 1 to 9 were respectively perfused with the compositions prepared in examples 1 to 9; the compositions prepared in comparative examples 1 to 2 were respectively intragastric in experimental groups 10 to 11.

As can be seen from the experimental data in table 1, the blood alcohol concentration in rats of experimental group 1 is significantly lower than that of the model group, which indicates that: the composition prepared by compounding the kudzuvine root, the kudzuvine flower, the hawthorn and the ginseng with the moringa seed can accelerate alcohol metabolism and has the effect of dispelling alcohol effects.
From the experimental data in table 1, it can be seen that, compared with the model group, the blood alcohol concentration in the rats in the experimental groups 2 to 4 is substantially smaller than that in the model group, and is also substantially smaller than that in the experimental group 1; in addition, the blood alcohol concentration in rats of experimental groups 6 to 9 is equivalent to that of experimental group 1; the blood alcohol concentration in rats of experimental groups 2-4 is also substantially less than that of experimental groups 6-9. This illustrates: the moringa seed, the kudzuvine root, the kudzuvine flower, the hawthorn and the ginseng are taken as raw materials, and different extraction solvents play an important role in the speed of alcohol metabolism of the prepared extract. In the invention, moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng are taken as raw materials, and the extract obtained by extracting the moringa seed, the kudzuvine flower, the hawthorn and the ginseng by adopting a mixed organic solvent consisting of n-butyl alcohol and acetone has a higher alcohol metabolism speed than that of the extract obtained by adopting the conventional ethanol extraction, and is also higher than that of the extract obtained by adopting other organic solvents or a combination of the organic solvents.
As can be seen from the experimental data in table 1, the blood alcohol concentration in rats of experimental group 5 is further substantially less than that of experimental group 2; this illustrates: the composition prepared under the macroporous resin elution condition can further greatly improve the metabolism speed of alcohol on the basis of an extract obtained by extracting moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng serving as raw materials by adopting a mixed organic solvent consisting of n-butyl alcohol and acetone.
As can be seen from the experimental data in table 1, the blood alcohol concentration in rats of experimental groups 10 to 11 was not significantly reduced compared with that of experimental group 2; this shows that the macroporous resin elution conditions of the invention are very critical, and the effective components obtained by enrichment have great difference on alcohol metabolism speed under different macroporous resin elution conditions; on the basis of taking moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng as raw materials and adopting an extract obtained by extracting a mixed organic solvent consisting of n-butyl alcohol and acetone, the composition prepared by adopting any macroporous resin elution condition can not be used for further improving or greatly improving the metabolism speed of alcohol; only under the condition of the macroporous resin elution (namely eluting with 12-15% ethanol water solution by volume fraction and then eluting with 26-28% ethanol water solution) can the composition prepared by the invention further greatly improve the metabolism rate of alcohol, but the composition prepared under other macroporous resin elution conditions can not further greatly improve the metabolism rate of alcohol.
Claims (6)
1. The composition with the anti-alcohol effect is characterized by being prepared from the following raw material components in parts by weight: 20-40 parts of moringa seed, 8-20 parts of kudzuvine root, 15-30 parts of kudzuvine flower, 15-30 parts of hawthorn and 3-10 parts of ginseng;
a method of preparing the composition comprising the steps of:
mixing Moringa seed, radix Puerariae, flos Puerariae Lobatae, fructus crataegi and Ginseng radix, adding organic solvent, extracting, concentrating and drying the extractive solution to obtain organic solvent extract;
loading the organic solvent extract on a macroporous resin column, eluting with an ethanol aqueous solution with the volume fraction of 12-15% and the volume of 3-5 times of the column volume to remove impurities; then eluting with an ethanol water solution with the volume fraction of 26-28% and 4-6 times of the column volume, collecting an eluent eluted from the ethanol water solution with the volume fraction of 26-28%, concentrating and drying to obtain the composition;
the extraction is heating reflux extraction, and the extraction time is 1-3 hours;
the organic solvent is a mixed organic solvent consisting of n-butanol and acetone; wherein the volume ratio of n-butanol to acetone is 2-4:1;
the dosage ratio of the total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn and ginseng to the organic solvent is 1 g:10-20 mL.
2. The composition according to claim 1, which is prepared from the following raw material components in parts by weight: 20-30 parts of moringa seed, 10-15 parts of kudzuvine root, 20-25 parts of kudzuvine flower, 20-25 parts of hawthorn and 5-8 parts of ginseng.
3. The composition according to claim 1, which is prepared from the following raw material components in parts by weight: 22 parts of moringa seed, 11 parts of kudzuvine root, 22 parts of kudzuvine flower, 22 parts of hawthorn and 6 parts of ginseng.
4. The composition of claim 1, wherein the volume ratio of n-butanol to acetone is 3:1.
5. The composition of claim 1, wherein the ratio of total weight of moringa seed, kudzuvine root, kudzuvine flower, hawthorn fruit and ginseng to organic solvent is 1g to 15ml.
6. Use of a composition according to any one of claims 1 to 5 for the preparation of a product having anti-hangover effect, characterized in that said product is a pharmaceutical.
