Preferably, the auxiliary material is selected from at least one of yeast powder, skim milk powder, maltodextrin and sugar alcohol, and is further preferably yeast powder and skim milk powder. The inventor finds that in the preferable embodiment, the yeast powder contains antioxidant, so that the liver can be protected, a certain detoxifying effect is achieved, and minerals such as selenium and chromium in the yeast powder can resist aging, resist tumors, prevent arteriosclerosis and improve the immunity of a human body; the skim milk powder has low fat content, is easy to store, and is not easy to generate oxidation.
According to the present invention, the amount of the yeast powder and the skim milk powder is not particularly limited, and may be selected conventionally in the art, and in order to further improve the mouthfeel of the probiotic composition, it is preferable that the weight ratio of the yeast powder to the skim milk powder is 1:48-52.
The above substances are all commercially available.
The invention provides a preparation method of a probiotic composition for relieving postpartum depression, which comprises the following steps: mixing probiotics, prebiotics and active ingredients to obtain a mixture; the probiotic contains Lactobacillus acidophilus, lactobacillus bulgaricus, lactobacillus reuteri and Lactobacillus helveticus; the active ingredients comprise beta-cryptoxanthin and N-acetylneuraminic acid.
According to the invention, preferably, the prebiotics are present in an amount of 1.5 to 5 parts by weight and the active ingredient is present in an amount of 0.2 to 0.5 part by weight, relative to 10 parts by weight of the probiotic.
According to the present invention, preferably, the weight ratio of said lactobacillus acidophilus, said lactobacillus bulgaricus, said lactobacillus reuteri and said lactobacillus helveticus is 1-5:1-5:1-2:1.

According to the invention, preferably, the weight ratio of the auxiliary material to the probiotic bacteria is 0.1-0.25:1, specifically, it may be 0.1: 1. 0.15: 1. 0.2: 1. 0.25:1, or any value in between the two values.
According to the present invention, preferably, the auxiliary material is selected from at least one of yeast powder, skim milk powder, maltodextrin, and sugar alcohol.
According to the invention, the prebiotic and the adjuvant may be dried separately before being mixed. The drying may be carried out by any conventional drying means, such as hot air drying, freeze drying, etc. Preferably, the drying conditions include: the temperature is 35-45 deg.C, specifically 35 deg.C, 40 deg.C, 45 deg.C, or any value between the above two values; the time is 30-60min, specifically 30min, 40min, 50min, 60min, or any value between the above two values.
According to the present invention, the mixing method is not particularly limited, and conventional selection in the art can be adopted, and preferably, the rotation speed is 50 to 100rpm, specifically, 50rpm, 60rpm, 70rpm, 80rpm, 90rpm, 100rpm, or any value between the two values; the time is 5-10min, specifically 5min, 6min, 7min, 8min, 9min, 10min, or any value therebetween.
In the present invention, the probiotic composition is preferably dispensed and sealed after it has been obtained. The method of dispensing and sealing is not particularly limited, and a method of dispensing and sealing conventional in the art may be employed.
In a third aspect, the present invention provides a use of the probiotic composition of the first aspect and/or the probiotic composition obtained by the preparation method of the second aspect in preparing a medicament and/or food for relieving postpartum depression.
According to a particularly preferred embodiment of the present invention, there is provided a method for preparing a probiotic composition for relieving postpartum depression, comprising the steps of:
(1) Respectively drying prebiotics and adjuvants at 35-45 deg.C for 30-60min;
(2) Mixing probiotics, prebiotics, active ingredients and auxiliary materials, and stirring for 5-10min under the condition that the rotating speed is 50-100rpm to obtain the probiotic composition;
the probiotic bacteria comprise Lactobacillus acidophilus, lactobacillus bulgaricus, lactobacillus reuteri, lactobacillus helveticus, bifidobacterium longum, lactobacillus rhamnosus and Bifidobacterium breve; the active ingredients comprise beta-cryptoxanthin and N-acetylneuraminic acid;
relative to 10 parts by weight of the probiotics, the content of the prebiotics is 1.5-5 parts by weight, the content of the active ingredients is 0.2-0.5 part by weight, and the content of the auxiliary materials is 1-2.5 parts by weight; the weight ratio of the lactobacillus acidophilus, the lactobacillus bulgaricus, the lactobacillus reuteri, the bifidobacterium longum, the lactobacillus rhamnosus, the bifidobacterium breve to the lactobacillus helveticus is 1-5:1-5:1-2:0.2-1:0.5-2:0.2-1:1; the weight ratio of the beta-cryptoxanthin to the N-acetylneuraminic acid is 1:1-4; the prebiotics are fructo-oligosaccharide, galacto-oligosaccharide, inulin and isomaltooligosaccharide; the weight ratio of the fructo-oligosaccharide, the galacto-oligosaccharide, the inulin and the isomalto-oligosaccharide is 1:0.5-2:1-4:1-4; the auxiliary materials are yeast powder and skim milk powder.
In the above particularly preferred embodiment, the probiotics, prebiotics and active ingredients are reasonably matched, and under the combined action, the ecology of the intestinal flora can be adjusted, abnormal mood can be relieved, and the effect of relieving postpartum depression can be achieved; and the probiotic composition has the advantages of simple production process, convenient operation and low production cost.
The present invention will be described in detail below by way of examples.
In the following examples and comparative examples, lactobacillus acidophilus was obtained from Shannon Biotech (Suzhou) Inc. under the model AL12; lactobacillus bulgaricus was purchased from Shangen Biotech (Suzhou) Inc. under model number LB10; lactobacillus reuteri was obtained from shann kang biotechnology (suzhou) limited as model RL17; lactobacillus helveticus was purchased from shangkang biotechnology (suzhou) limited, model number HL32; bifidobacterium longum is available from Shangen Biotechnology (Suzhou) Inc. with model number LB32; lactobacillus rhamnosus was purchased from shangkang biotechnology (suzhou) ltd, model No. NJ551; bifidobacterium breve was purchased from Shangen Biotechnology (Suzhou) Inc. under model number BB28; beta-cryptoxanthin was purchased from Shanghai Allantin Biotechnology, inc. under the model number C339418; n-acetylneuraminic acid was purchased from Shanghai Michelin Biotechnology Ltd, model No. N801293; other materials were commercially available unless otherwise specified.
Example 1
(1) Respectively placing fructo-oligosaccharide, galacto-oligosaccharide, inulin, isomaltooligosaccharide, yeast powder and skimmed milk powder into an oven, and drying at 40 deg.C for 40min;
(2) Weighing 1g of fructo-oligosaccharide, 1g of galacto-oligosaccharide, 1.5g of inulin, 1.5g of isomaltooligosaccharide, 0.06g of yeast powder and 3g of skim milk powder obtained after drying in the step (1), putting into a container, and stirring for 5min under the condition that the rotating speed is 80rpm to obtain a mixture;
(3) And (3) adding 4g of lactobacillus acidophilus, 4g of lactobacillus bulgaricus, 3g of lactobacillus reuteri, 2g of lactobacillus helveticus, 1g of bifidobacterium longum, 2g of lactobacillus rhamnosus, 1g of bifidobacterium breve, 0.2g of beta-cryptoxanthin and 0.4g N-acetylneuraminic acid into the mixture obtained in the step (2), and stirring for 5min at the rotating speed of 80rpm to obtain the probiotic composition.
Example 2
(1) Respectively placing fructo-oligosaccharide, galacto-oligosaccharide, inulin, isomaltooligosaccharide, yeast powder and skimmed milk powder into an oven, and drying at 35 deg.C for 60min;
(2) Weighing 1g of fructo-oligosaccharide obtained after drying in the step (1), 0.5g of galacto-oligosaccharide, 1g of inulin, 1g of isomaltooligosaccharide, 0.1g of yeast powder and 5g of skim milk powder, putting into a container, and stirring for 5min under the condition that the rotation speed is 50rpm to obtain a mixture;
(3) And (3) adding 10g of lactobacillus acidophilus, 2g of lactobacillus bulgaricus, 4g of lactobacillus reuteri, 2g of lactobacillus helveticus, 2g of bifidobacterium longum, 1g of lactobacillus rhamnosus, 0.4g of bifidobacterium breve, 0.1g of beta-cryptoxanthin and 0.4g N-acetylneuraminic acid into the mixture obtained in the step (2), and stirring for 5min at the rotation speed of 50rpm to obtain the probiotic composition.
Example 3
(1) Respectively placing fructo-oligosaccharide, galacto-oligosaccharide, inulin, isomaltooligosaccharide, yeast powder and skimmed milk powder into an oven, and drying at 45 deg.C for 30min;
(2) Weighing 1g of fructo-oligosaccharide, 2g of galacto-oligosaccharide, 4g of inulin, 4g of isomaltooligosaccharide, 0.05g of yeast powder and 2.5g of skim milk powder obtained after drying in the step (1), putting into a container, and stirring for 5min under the condition that the rotation speed is 100rpm to obtain a mixture;
(3) And (3) adding 2g of lactobacillus acidophilus, 10g of lactobacillus bulgaricus, 2g of lactobacillus reuteri, 2g of lactobacillus helveticus, 0.4g of bifidobacterium longum, 4g of lactobacillus rhamnosus, 2g of bifidobacterium breve, 0.5g of beta-cryptoxanthin and 0.5g N-acetylneuraminic acid into the mixture obtained in the step (2), and stirring for 5min at the rotating speed of 100rpm to obtain the probiotic composition.
Example 4
A probiotic composition was prepared according to the method of example 1, except that the amount of N-acetylneuraminic acid was replaced with 2g.
Example 5
A probiotic composition was prepared according to the method of example 1, except that the amount of lactobacillus acidophilus was replaced with 1g, the amount of lactobacillus bulgaricus was replaced with 1g, and the amount of lactobacillus reuteri was replaced with 9g.
Example 6
(1) Placing galactooligosaccharide into oven, and drying at 40 deg.C for 40min;
(2) Mixing 5g of galactooligosaccharide obtained after drying in the step (1) with 4g of lactobacillus acidophilus, 4g of lactobacillus bulgaricus, 3g of lactobacillus reuteri, 2g of lactobacillus helveticus, 1g of bifidobacterium longum, 2g of lactobacillus rhamnosus, 1g of bifidobacterium breve, 0.2g of beta-cryptoxanthin and 0.4g N-acetylneuraminic acid, and stirring for 5min under the condition of the rotation speed of 50rpm to obtain the probiotic composition.
Example 7
A probiotic composition was prepared according to the method of example 1, except that step (3) was replaced by:
(3) And (3) adding 4g of lactobacillus acidophilus, 4g of lactobacillus bulgaricus, 3g of lactobacillus reuteri, 2g of lactobacillus helveticus, 0.2g of beta-cryptoxanthin and 0.4g N-acetylneuraminic acid into the mixture obtained in the step (2), and stirring for 5min at the rotation speed of 50rpm to obtain the probiotic composition.
Example 8
A probiotic composition was prepared according to the method of example 1, except that step (2) was replaced by:
(2) Weighing 1g of fructooligosaccharide obtained after drying in the step (1), 1g of galactooligosaccharide, 1.5g of inulin and 1.5g of isomaltooligosaccharide, putting into a container, and stirring for 5min at the rotation speed of 50rpm to obtain a mixture.
Example 9
A probiotic composition was prepared according to the method of example 1, except that the amount of β -cryptoxanthin was replaced with 0.05g and the amount of n-acetylneuraminic acid was replaced with 0.1g.
Comparative example 1
A probiotic composition was prepared according to the method of example 1, except that step (3) was replaced by:
(3) And (3) adding 4g of lactobacillus acidophilus, 4g of lactobacillus bulgaricus, 3g of lactobacillus reuteri, 2g of lactobacillus helveticus, 1g of bifidobacterium longum, 2g of lactobacillus rhamnosus and 1g of bifidobacterium breve into the mixture obtained in the step (2), and stirring for 5min at the rotation speed of 50rpm to obtain the probiotic composition.
Comparative example 2
A probiotic composition was prepared according to the method of example 1, except that step (3) was replaced by:
(3) And (3) adding 4g of lactobacillus acidophilus, 4g of lactobacillus bulgaricus, 3g of lactobacillus reuteri, 2g of lactobacillus helveticus, 1g of bifidobacterium longum, 2g of lactobacillus rhamnosus, 1g of bifidobacterium breve and 0.2g of beta-cryptoxanthin into the mixture obtained in the step (2), and stirring for 5min at the rotation speed of 50rpm to obtain the probiotic composition.
Comparative example 3
A probiotic composition was prepared according to the method of example 1, except that step (3) was replaced by:
(3) And (3) adding 4g of lactobacillus bulgaricus, 3g of lactobacillus reuteri, 2g of lactobacillus helveticus, 1g of bifidobacterium longum, 2g of lactobacillus rhamnosus, 1g of bifidobacterium breve, 0.2g of beta-cryptoxanthin and 0.4g N-acetylneuraminic acid into the mixture obtained in the step (2), and stirring for 5min at the rotating speed of 50rpm to obtain the probiotic composition.
Test example
The mouse tail suspension test and the forced swimming test are routine animal experiments for verifying whether the drug has obvious functions of relieving anxiety and depression after administration. ICR mice, 18-22g in each male and female half, after 1 week of adaptive feeding, were randomly divided into 13 groups, 6 mice in each group were fed with the probiotic compositions prepared in examples 1-9 and comparative examples 1-3, respectively, at an amount of 0.1g/kg, the drug was formulated into a suspension with physiological saline and administered by intragastric administration, and the blank control group was given an equal volume of physiological saline during administration. The medicine is administered for 7 days by continuous gavage 1 time a day, and the tail suspension experiment and forced swimming experiment of mice are carried out after the last administration for 1 h.

Claims (10)

1. A probiotic composition for relieving postpartum depression, wherein the probiotic composition contains probiotics, prebiotics and active ingredients;
the probiotic contains Lactobacillus acidophilus, lactobacillus bulgaricus, lactobacillus reuteri and Lactobacillus helveticus; the active ingredients comprise beta-cryptoxanthin and N-acetylneuraminic acid.
2. The probiotic composition according to claim 1, characterized in that the prebiotic is present in an amount of 1.5 to 5 parts by weight and the active ingredient is present in an amount of 0.2 to 0.5 parts by weight, relative to 10 parts by weight of the probiotic.
3. The probiotic composition according to claim 2, characterized in that the weight ratio of said lactobacillus acidophilus, said lactobacillus bulgaricus, said lactobacillus reuteri and said lactobacillus helveticus is between 1 and 5:1-5:1-2:1;
preferably, the weight ratio of the β -cryptoxanthin to the N-acetylneuraminic acid is 1:1-4.
4. The probiotic composition according to any one of claims 1 to 3, characterized in that the probiotic also contains Bifidobacterium longum, lactobacillus rhamnosus and Bifidobacterium breve;
preferably, the weight ratio of bifidobacterium longum, lactobacillus rhamnosus, bifidobacterium breve to lactobacillus helveticus is 0.2-1:0.5-2:0.2-1:1.

7. A preparation method of a probiotic composition for relieving postpartum depression is characterized by comprising the following steps: mixing probiotics, prebiotics and active ingredients to obtain a mixture;
the probiotic bacteria comprise Lactobacillus acidophilus, lactobacillus bulgaricus, lactobacillus reuteri and Lactobacillus helveticus; the active ingredients contain beta-cryptoxanthin and N-acetylneuraminic acid.
8. The method according to claim 7, wherein the prebiotic is contained in an amount of 1.5 to 5 parts by weight and the active ingredient is contained in an amount of 0.2 to 0.5 part by weight, relative to 10 parts by weight of the probiotic bacteria;
preferably, the weight ratio of said lactobacillus acidophilus, said lactobacillus bulgaricus, said lactobacillus reuteri and said lactobacillus helveticus is 1-5:1-5:1-2:1;

preferably, the auxiliary material is selected from at least one of yeast powder, skim milk powder, maltodextrin and sugar alcohol.
10. Use of a probiotic composition according to any one of claims 1 to 6 and/or obtained by a method according to any one of claims 7 to 9, for the preparation of a medicament and/or a food product for alleviating postpartum depression.
