慢性肾脏疾病(CKD)的背景:

阿尔茨海默病(AD)的背景:

进行性MS是疾病的最新阶段,其特征是症状逐渐恶化而无缓解。严重的神经损伤极大地降低了个体的生活质量,这主要归因于影响运动功能的皮质病变的扩大。病理学上,广泛存在轴突变性和灰质神经病变。据报道,弥漫性白质和灰质炎症部分与整体小胶质细胞活化以及T细胞、B细胞和富含髓鞘的巨噬细胞的存在有关。此外,OPC总体上不能有效地使受损的白质和灰质区域髓鞘再生,从而大大降低了恢复的可能性(Chang,A.,et al.(2002)The New England journal of medicine 346.165-173)。Klotho促进OPC成熟为成熟的少突胶质细胞(Chen,C.D.,et al.(2013)The Journal of neuroscience 33,1927-1939)。
帕金森病(PD)的背景:
PD是一种进行性神经退行性疾病,临床上以静止性震颤、运动迟缓、齿轮样强直和姿势不稳定为主要症状。对L-3,4-二羟基苯丙氨酸(L-DOPA)的响应能力和脑成像将PD与其他疾病区分开来。PD的病理特征是黑质、致密部多巴胺能细胞的丧失和随后纹状体多巴胺神经支配的丧失。运动症状是这种黑质纹状体神经变性最明显的后果。然而,不仅基底神经节,而且中枢神经系统的其他部分以及自主神经系统都受到影响。由此产生的各种非运动症状会影响患者的生活质量。还有一个广泛的共识是,PD的神经退行性过程在临床症状实际出现前许多年就开始了。家族性和特发性PD之间的表型重叠足以剖析通常涉及的途径。这些包括线粒体功能障碍、氧化应激、蛋白质错误折叠、蛋白质降解、蛋白质聚集和炎症。
Kosakai等人证明,在klotho缺乏小鼠中,在黑质、致密部、腹侧节段区中的酪氨酸羟化酶阳性多巴胺能神经元的数量和纹状体多巴胺水平以年龄依赖性方式显著减少。这些表型特征被维生素D限制完全挽救,表明Klotho不足引起的活性维生素D生物合成的异常增加诱导了多巴胺能神经元的变性(Kosakai,A.,et al.(2011)Brain research 1382,109-117)。
由于Klotho对各种信号通路的贡献及其在许多器官系统中的重要功能,并且基于在各种疾病中蛋白质水平降低的观察,包括施用Klotho在内的新治疗可能代表有希望的治疗方法。例如,WO2016/135295提供了间充质干细胞,其可以施用给患者以能够治疗上述疾病,以及施用Klotho对个体有益的其他病症。
除了已知Klotho水平降低的疾病的可能治疗之外,蛋白质本身也被认为是生物体健康和良好状态的某种标志。虽然确定特定疾病的生物标志物是诊断领域中的一个通常概念,但是对可以表明健康状况良好或至少在各种主要健康方面的健康状况的分子的监测一直被忽视。Fantuzzi(frontiers in IMMUNOLOGY,July 2014,Volume 5,Article 351)专注于“健康的声音”,或者实际上,迄今为止尚未充分找到共鸣的健康的沉默。他报告说,相反,痛苦的信息比可以被认为是生物体代谢平衡和缺乏痛苦的标志的分子更受青睐。Fantuzzi提到Klotho是这种候选的健康信使之一,因为据观察,在健康和没有痛苦的状态下,这种分子以最佳形式产生、释放和具有生物活性,而任何种类的压力都会导致Klotho水平降低。人的最佳Klotho水平可以是良好健康的重要指标,例如当在定期的健康筛查中确定时。
虽然,特别是对于更严重的病况,施用产生Klotho的间充质干细胞提供了蛋白质表达并达到可被认为是治愈性的蛋白质水平,但在某些情况下,个体中Klotho水平的预防性和治疗性增加也可通过体育锻炼来实现。例如Matsubara等人(AM J Physiol HeartCirc Physiol。306:H348-H355,2014)报道了绝经后妇女在有氧运动训练后血浆Klotho水平的增加和动脉僵硬度的降低。Ji等(Experimental and Therapeutic Medicine 16:3511-3517,2018)也预测了由于有氧运动和伴随而来的Klotho表达的促进对衰老和衰老相关疾病的积极作用。此外,Tan等人(Journal of Circulating Biomarkers,Volume 7:1-7,2018)观察到健康志愿者在高强度体育锻炼后Klotho血清水平升高。甚至单次训练也被发现会诱发Klotho的形成。最后,Avin等人(frontiers in Physiology,Volume 5,Article189,June 2014)总结了早期发表的效果,并令人印象深刻地证明了运动(尤其是骨骼肌)对Klotho表达的影响,以及由此导致的血浆中Klotho水平的增加。
这些观察将体育锻炼的寿命延长效果,特别是在患有某些疾病如糖尿病的患者中,与Klotho的存在和血浆水平联系起来。Santos-Dias等人(Br J Sports Med 20160∶1-2)显示,即使单次锻炼也能显著诱导男性和女性的血清Klotho水平升高。Mostafidi等人(Nephro Urol Mon.,2016Jan;8(1):e30245)报道了与非运动员组相比,运动员组的血浆中游离Klotho的浓度显著更高,而血清的其他成分在两组之间没有显著差异。
有氧运动尤其是肌肉力量训练可以增加血清Klotho水平的观察结果是一个非常有希望的发现。这些结果表明,通过体育训练和身体锻炼可以极大地改善某些状况以及总体健康,并且进一步的结果还表明,特别是在老年人或患有神经退行性疾病的人中,提高血浆Klotho水平可以改善认知状况或至少减缓认知衰退。(Shardell et al.,J GerontologA Biol Sci Med Sci,2015,1-6;Cheng et al.,Acta Neurobiol Exp 2015,75:60-71)。
毫无疑问,Klotho水平是健康或疾病的重要指标,并且可以通过将过低的水平提高到最佳值来帮助实现个体的长寿。然而,目前情况的一个缺点是需要对血液样品进行Klotho水平的测量,血液样品通常在医疗实践中抽取并在临床实验室中分析。确定Klotho水平不仅对于初始诊断是重要的,而且需要在用常用药物或通过施用Klotho治疗疾病期间或之后,或者在通过体育锻炼改善人的健康的过程中进行监测。特别是对于个体中Klotho水平的定期观察,目前的情况并不令人满意,本发明的一个目的是提供一种以成本有效且易于应用的方式测定个体中Klotho水平的改进方法,该方法最好甚至可以在体育锻炼期间或之后立即进行,以监测Klotho水平。
发明内容
在第一方面,本发明涉及通过测量体液中Klotho的含量来确定和/或监测个体健康状况的方法,其中所述体液选自唾液和泪液。
在第二方面,本发明涉及一种检测试剂盒,其特征在于包含进行免疫测定所需的试剂。
具体实施方式
本发明通过提供一种测定患者体内Klotho水平的改进方法解决了上述目的,并且它特别以测试试剂盒的形式提供了用于执行该诊断方法的方法和手段。这些测试可以很容易地由个人或患者自己在任何合适的时间进行。
本发明方法基于发明人的惊人发现,即特别是唾液中的Klotho水平与血清样品中测定的Klotho水平非常好地成比例对应。因此,本发明的方法优选用唾液进行。
本发明的方法实现了提供无痛、廉价、与具有分子诊断影响的基于血清或尿液的方法相比更可靠、更容易和更安全的测试的目的。将本方法与HPLC技术进行了比较,从而发现了可比较的结果。已经存在的结果的令人惊讶之处在于患者和健康志愿者之间的血清和唾液中的Klotho水平存在显著差异。
虽然在同一个体和同一组人中血清和唾液的实际值在一定程度上不同,但是可以确定血清和其他体液,特别是唾液中Klotho水平之间的可靠关系(参见图1至5)。这一观察结果证明,Klotho蛋白在唾液中以显著和有意义的水平存在,这与许多其他蛋白的情况不同(Tékus et al.,Acta Biol Hung,2012;63(Suppl1):89-98;Ellis et al.,N Z Med JK,2012;125(1353):47-58;Volkery et al.,Vet Rec,2012;171(8):195;Holten-Andersonet al.,Scand J Gastroenterol,2012;47(10):1234-41;Rahnama et al.,Endokr Res,2012Aug 15epub;Pant Pai et al.,Lancet Infect Dis,2012;12(5):373-80;Mahboobiet al.,J Oral Pathol Med,2012;41:505-16;Yen Bee Ng et al.,FEMS Immunol MedMicrobiol,2007;49(2):252-60;Castagnola et al.,Acta Otorinologica Italica,2011;31:347-57):
利用这种观察结果,本发明首次提供了一种用于监测个体的健康状况的可靠的检测方法和检测试剂盒。在本发明的某些实施方案中,这种健康状况被认为包括与低或降低的Klotho水平相关的现有的或先前的疾病或病况,或这种疾病或病况的倾向。伴有相对较低Klotho水平的此类疾病或病况已在现有技术中进行了描述并且在本说明书的介绍部分中进行了详细描述。然而,这并不排除Klotho水平下降可能只会在未来才能被识别的其他疾病。因此,通过本发明的方法检测到相对较低的Klotho水平也可能是人的健康状况不令人满意的迹象,即使尚未观察到特定的疾病症状。因此,可以应用进一步的诊断方法来检测或排除严重疾病。
在优选的实施方案中,与健康人相比伴随或导致Klotho水平降低的疾病或病况选自癌症、炎性疾病、慢性肾病(CKD)、神经退行性疾病、慢性心脏病、器质性纤维化、动脉粥样硬化、痴呆、糖尿病、勃起功能障碍、自身免疫性疾病或与自身免疫相关的疾病、脓毒症以及早衰和其他年龄相关疾病中的至少一种。
由本发明的方法测定的低Klotho水平可以指示的神经退行性疾病以及神经退行性疾病的先决条件(precondition)包括阿尔茨海默病、帕金森病、肌萎缩侧索硬化(ALS)和亨廷顿病。在进一步优选的实施方案中,人的健康状况受到自身免疫性疾病或与自身免疫相关的疾病的影响,包括1型糖尿病、类风湿性关节炎(RA)、多发性硬化(MS)、系统性红斑狼疮、自身免疫性脑脊髓炎、自身免疫性肾炎如狼疮性肾炎和IgA肾病、骨关节炎或激活患者炎症小体或其成分的其他疾病。
本发明的方法可以用于检测这些疾病存在的风险,或者用于监测疾病的进展和/或药物治疗的效果。
特别是对于在现有或先前疾病的情况下使用本发明方法的情况,优选在治疗之前、期间和/或之后监测和确定Klotho水平。在本文中,治疗包括施用Klotho以及施用其他治疗剂或其组合。如上所述,Klotho可以作为蛋白质应用,或者可以通过例如施用表达Klotho的干细胞来促进Klotho的形成。如果在治疗期间可以提高Klotho水平,则该人的健康状况明显改善。
在其他方面健康的人中发现低水平的Klotho也可以被认为是这个人缺乏足够的体育运动的强烈暗示。如果这个人不改变他或她的生活方式,这种缺乏体育锻炼可能会导致或多或少的严重健康问题和过早衰老。因此,本发明进一步优选的实施方案是体外方法,其中测定Klotho含量以监测体育运动对个体健康状况的影响。如上所述,体育运动,尤其是身体训练、肌肉训练和耐力训练已经显示出增加Klotho水平,这也反映了健康状况,尤其是个体认知能力的增加。
本发明的体外方法也可用于确定个体的生物学年龄。同样,确定这样的生物学年龄可以帮助解决可能的健康问题和不健康的行为,并因此通过随后采取导致增加和调整Klotho水平的所需行动来延长寿命。
对于任何测定形式,在使用血清与唾液的免疫测定中获得的值之间的实际相关性可以很容易地确定或验证。例如,当通过TRF方法(时间相关荧光免疫测定)测量时,血清和唾液中的Klotho之间的关系已被确定为3.6:1(图1)。因此,测试方法的校准提供了健康人在例如唾液中与血清中的“正常”值的关系。
“正常”值也可以通过提供适当相似的人群的测试结果来确定,这些人群被认为是健康的,或者其Klotho水平在与人群平均值的某个可接受的偏差范围内。群组可以从相似年龄、相同性别、相似生活环境等的人中选择。此外,在某些情况下,文献中报道的值可用于选择“正常”值。
如果检测到的Klotho水平低于某一阈值,则本发明的体外方法将优选伴有至少一种治疗措施,该阈值可以例如如上所述获得。治疗措施包括给个体施用Klotho,或者增加已确定Klotho水平的个体的Klotho水平,基于如上所述的适当关系,该水平对应于低于100pg/ml的血清值。如果Klotho水平对应于低于200pg/ml的血清水平,最优选低于300pg/ml的水平,则优选地应该已经启用提高Klotho水平的治疗措施。因此,唾液中测定的、必须考虑采取治疗措施的Klotho水平降低了3.6倍。
优选地,本发明背景中Klotho的施用包括施用治疗有效数量的产生Klotho的间充质干细胞,或治疗有效数量的含有核酸载体的细胞,所述核酸载体包含编码Klotho蛋白的区域,所述区域与启动子或启动子/增强子组合可操作地连接,其中所述含有载体的细胞表达Klotho。此外,在本发明的上下文中可以考虑产生Klotho的病毒载体或通过CRISPR-Cas9基因编辑诱导Klotho表达,以提高个体中的Klotho水平。在其他实施方案中,Klotho蛋白被直接递送或施用给患者,其中Klotho蛋白优选通过生物技术方法重组产生。
Klotho的施用量取决于在个体或患者中测定的实际水平,并且,优选地该施用量适于将该水平提高到当在血清中测定时超过340pg/ml以及,相应地,提高到当在唾液中测定时超过85pg/ml,优选超过100pg/ml。
在另一个优选的实施方案中,本发明还包括让个体增加体育活动,而不是对患者施用Klotho蛋白或通过如上所述的生物技术方法在患者中诱导其表达。在这类患者中尤其指的是进行体育训练,特别是肌肉训练,如果所确定的Klotho水平低于在健康人群中确定的“正常”水平对应的值,尤其是上述值。
根据本发明,Klotho水平优选通过包括至少一种特异性结合Klotho的抗体或适体的免疫测定来确定。本发明的方法优选包括通过双抗体夹心测定法或竞争性测定法测定Klotho水平。这两种测定最优选以侧向流动分析进行。
分析的具体设计不是决定性的,技术人员可以获得合适的测试形式。下面将更详细地描述一些具体的优选实施方案,因为本发明的另一个主题是用于实施本发明方法的试剂盒。这种试剂盒包含进行免疫测定所需的反应物。
原则上,任何免疫测定形式都可以用于实施本发明的方法。在优选的实施方案中,该测试以双抗体夹心测定法或竞争性测定法进行。用于这种方法中的包括固相、抗体以及标记物的测定环境也是技术人员熟知的,并且对可应用的测试形式没有限制。然而,在本发明的一个非常优选的实施方案中,由本发明的检测试剂盒提供的检测形式适用于侧向流动分析形式。这种类型的测试和相应的测试试剂盒更详细地描述如下:
侧向流动分析
如上所述,用于实施本文公开的体外方法的试剂盒可以基于不同的原理。一种优选的原理被称为侧向流动免疫层析分析。患者/运动员可以容易地进行这种侧向流动免疫层析分析,而无需医生或其他受过医学训练的人的帮助。
侧向流动测试是简单的装置,旨在检测目标分析物样品的存在(或不存在),而不需要专门和昂贵的设备,尽管存在许多基于实验室的应用,其由读数设备支持。通常,这些测试用于家庭测试、护理点测试(POC)或实验室使用的医学诊断。一个广泛传播和众所周知的应用是家庭怀孕测试。
该技术基于一系列毛细管床,如多孔纸片或烧结聚合物片。这些元件中的每一个都具有自发输送流体(例如唾液)的能力,第一元件(样品垫)充当海绵并容纳过量的样品流体。一旦被浸透,流体就迁移到第二元件(缀合物垫),在该第二元件中,制造商已经储存了所谓的缀合物,优选为生物活性颗粒的干燥形式(见下文),优选在盐-糖基质中,该基质包含一切以保证靶分子(例如Klotho)和已经固定在颗粒表面上的其化学伴侣(例如抗体)之间的优化化学反应。当样品流体溶解盐-糖基质时,它也溶解颗粒,并且在一个组合输送动作中,样品和缀合物混合,同时流过多孔结构。这样,分析物结合到颗粒上,同时进一步迁移通过第三毛细管床。这种材料有一个或多个区域(反应或捕获区,通常称为条带),其中第三分子已被制造商固定。当样品-缀合物混合物到达这些条带时,分析物已经结合在颗粒上,并且第三“捕获”分子结合复合物。一段时间后,当越来越多的液体通过条带时,颗粒会积聚,条带区域会变色。通常至少有两个条带:一个(对照区)捕获任何颗粒,从而显示反应条件和技术运行良好,第二个包含特定的捕获分子,并且仅捕获那些其上已经固定了分析物分子的颗粒。在通过这些反应区之后,流体进入最终的多孔材料,即仅仅作为废物容器的吸液芯(或废物储器)。
这种测试设计可以根据使用测试试剂盒进行的免疫测定的性质进行调整。侧向流动分析可以竞争性或夹心分析进行操作。
原则上,任何有色颗粒都可以用于缀合物,然而,乳胶(蓝色)或纳米尺寸的金颗粒(红色/黑色)是最常用的。由于局部表面等离子体共振,金颗粒呈红色。也可以使用荧光或磁性标记的颗粒,但是这些需要使用电子阅读器来评估测试结果。
样品首先遇到缀合物或试剂垫中的有色颗粒,这些颗粒被针对目标分析物的抗体标记。捕获区中的测试线也将包含针对相同目标的抗体,尽管它可能结合到分析物上的不同表位。在阳性样品中,测试线将显示为一条彩色带。夹心测定的一个例子是夹心ELISA。虽然不是严格必需的,但是大多数测试试剂盒优选地结合了第二条线,该线包含拾取自由粒子如乳胶/金的抗体,以便确认测试已经正确操作。
在一个优选的实施方案中,侧向流动分析的单个组分以这样一种方式进行调整,即只有当样品中存在超过一定阈值的klotho时,才表明klotho的存在。
本发明优选的检测试剂盒由以下成分组成:
1.作为样品垫,将吸收垫应用到测试样品(唾液、泪液、尿液)上
2.缀合物垫或试剂垫–这包含对目标分析物Klotho特异的抗体,并与标记的,优选是有色的颗粒(通常是胶体金颗粒或乳胶微球)缀合
3.反应区或捕获区:膜——通常为疏水性硝酸纤维素或醋酸纤维素膜,其上抗目标分析物抗体被固定在横跨膜的线上,作为捕获区或测试线(也可能存在对照区,含有对结合抗体特异的抗体)
4.吸液芯或废物区或储器–另一个吸收垫,设计用于通过毛细作用将样品吸过反应膜并收集它。
测试条的成分通常被固定在惰性背衬材料上,并且可以以简单的试纸形式存在,或者存在于具有显示捕获区和对照区的样品口和反应窗口的塑料外壳内。
在本发明的方法中使用的测试试剂盒(侧向流动免疫分析)有两个优选的实施方案:
a.双抗体夹心分析
在这种形式中,样品从样品垫迁移通过缀合物垫,其中任何作为目标分析物存在的Klotho将与缀合物结合。然后样品继续迁移穿过膜,直到它到达捕获区,在那里靶标/缀合物复合物将结合到固定的抗体上,在膜上产生可见的线,然后样品进一步沿着条带迁移,直到它到达对照区,在那里过量的缀合物将结合并在膜上产生第二条可见的线。这条对照线表明样品已经如预期的那样迁移穿过膜。膜上两条清晰的线显示阳性结果。对照区中的单线是阴性结果。双抗体夹心分析最适用于具有多个抗原位点的较大分析物,如细菌病原体和病毒。对于本发明,选择与Klotho上的不同表位结合的合适的抗体对。
这种抗体可以从市售产品中选择,或者通过由本领域已知的方法生成针对不同Klotho表位的抗体,特别是单克隆抗体来产生。当适用于实施这种方法的测试方法或试剂盒使用特异性结合Klotho的抗体时,术语“抗体”不仅指例如通过对实验室动物如兔、绵羊或山羊进行免疫而人工产生的抗体。在一个优选的实施方案中,它还包括根据杂交瘤技术产生的单克隆抗体。此外,术语“抗体”还包括抗体的抗原结合片段,例如重组产生的抗原结合片段。这种构建体可以例如通过噬菌体展示和由其衍生的技术产生。
b.竞争分析
竞争性分析主要用于测试小分子,与双抗体夹心形式的不同之处在于,缀合物垫包含已经与目标分析物或其类似物结合的抗体。如果样品中存在目标分析物,那么它将不会与缀合物结合,并将保持未标记状态。当样品沿膜迁移并到达捕获区时,过量的未标记的分析物将与固定的抗体结合,并阻止缀合物的捕获,从而不产生可见的线。然后,未结合的缀合物将与对照区中的抗体结合,产生可见的对照线。膜上的单一对照线是阳性结果。捕获区和对照区中的两条可见线是阴性结果。然而,如果不存在过量的未标记的目标分析物,则可能在捕获区中产生弱线,指示不确定的结果。竞争性检测最适合检测小分子,如不能同时结合一种以上抗体的真菌毒素。
考虑到可用于根据本发明的方法的这些不同的测试形式,该测试试剂盒的特征在于包含固相,该固相包含:
a)用于施加待测样品的样品垫,
b)缀合物垫或试剂垫,其包含Klotho特异性抗体的缀合物,该缀合物包含可检测的标记,或者这种抗体/标记缀合物和Klotho的缀合物,
c)反应或捕获区,其上固定有Klotho蛋白特异性抗体,和
d)用于收集样品流体和样品组分以及剩余试剂或缀合物的芯或废物储器,并且可选地进一步,
e)对照区,其包含与步骤b)的缀合物但不与Klotho特异性结合的抗体。
侧向流动技术有多种变体。膜上的捕获区可能包含固定的抗原或酶—取决于目标分析物—而不是抗体。还可以应用多个捕获区域来创建多重测试。
在本发明的背景中,术语抗体意味着也包括结合Klotho的抗体片段或适体。因此,在替代的优选实施方案中,如上所述的实施方案还可以在如上所述的区域b)、c)和e)中包括这种抗体片段或适体。
未受过训练的操作人员可以简单地使用侧向流动免疫分析,并且通常在15分钟内产生结果。它们非常稳定耐用,保质期长,通常不需要冷藏。它们的生产成本也相对低廉。这些特性使它们非常适合在护理点使用,以及在现场和实验室测试样品。然而,如果没有额外的浓缩或培养程序,它们的灵敏度是有限的。有可用的定量测试,但是本发明的目的优选地是对一定范围内的唾液或泪液的定性测试。在这种情况下,优选的测试试剂盒被调整为仅当Klotho的浓度高于某一浓度时才测量出Klotho。低于这样的浓度,检测试剂盒将显示阴性结果。

图4显示了慕尼黑数据与慢性肾病III期和IV期患者以及透析患者的Klotho血清水平之间的对比。在患者中,发明人以及其他人(例如,Wang et al.,BioMed ResearchInternational,Vol 2018,Article ID9481475;Rotondi et al.,International Journalof Endocrinology,Vol 2015,Article ID872193;and Pedersen et al.;ClinicalBiochemistry,46(2013)1079-1083)发现肾功能和Klotho在外周血中的浓度之间有很强的相关性,如下表所示:

Claims (15)
1.通过测量唾液或泪液中Klotho的含量来确定和/或监测个体健康状况的体外方法。
2.根据权利要求1所述的体外方法,其中所述个体的健康状况包括与低或降低的Klotho水平相关的现有或先前的疾病或病况或对所述疾病或病况的倾向。
3.根据权利要求2所述的体外方法,其中所述疾病或病况选自癌症、炎性疾病、慢性肾病(CKD)、神经退行性疾病、慢性心脏病、器官纤维化、动脉硬化、痴呆、糖尿病、勃起功能障碍、自身免疫性疾病或与自身免疫相关的疾病、脓毒症、早衰和其他年龄相关疾病中的至少一种。
4.根据权利要求3所述的体外方法,其中所述神经退行性疾病包括阿尔茨海默病、帕金森病、肌萎缩性侧索硬化(ALS)和亨廷顿氏病。
5.根据权利要求3所述的体外方法,其中所述自身免疫性疾病或与自身免疫相关的疾病包括1型糖尿病、类风湿性关节炎(RA)、多发性硬化(MS)、系统性红斑狼疮、自身免疫性脑脊髓炎、自身免疫性肾炎如狼疮性肾炎和IgA肾病、骨关节炎或激活患者或其组分中炎症小体的其他疾病。
6.根据权利要求1至5中任一项所述的体外方法,其中在疾病的治疗处理的情况下,通过在治疗之前、期间和/或之后测量Klotho的含量来进行个体健康状况的确定和/或监测。
7.根据权利要求1所述的体外方法,其中确定所述Klotho含量以监测体育活动,尤其是体育训练、肌肉训练和/或耐力训练对个体健康状况的影响。
8.根据权利要求1所述的体外方法,其用于确定个体的生物学年龄。
9.根据权利要求1至6中任一项所述的体外方法,其包括对所述个体施用Klotho,如果所测定的Klotho水平低于对应于在血清中测定的100pg/ml的值,优选低于对应于在血清中测定的200pg/ml的值。
10.根据权利要求9所述的体外方法,其中施用Klotho包括施用治疗有效数量的产生Klotho的间充质干细胞,或治疗有效数量的含有核酸载体的细胞,所述核酸载体包含编码Klotho蛋白的区域,所述区域与启动子或启动子/增强子组合可操作地连接,并且所述含有载体的细胞表达Klotho。
11.根据权利要求1至6中任一项所述的体外方法,其包括使所述个体增加体育活动,尤其是体育训练,例如肌肉训练和/或耐力训练,如果所测定的Klotho水平低于对应于在血清中测定的100pg/ml的值,优选低于对应于在血清中测定的200pg/ml的值。
12.根据权利要求1至11中任一项所述的体外方法,其中所述体液是唾液。
13.根据权利要求1至12中任一项所述的体外方法,其中所述Klotho含量经由包括至少一种特异性结合Klotho的抗体或适体的免疫分析来确定,优选地,其中所述免疫分析是双抗体夹心分析或作为竞争性分析,优选地作为侧向流动分析来进行。
14.用于实施根据权利要求1至13中任一项所述的方法的测试试剂盒,其特征在于,其为免疫分析测试试剂盒,优选地,其中所述测试以双抗体夹心分析或竞争性分析进行。
15.根据权利要求14至16中任一项所述的检测试剂盒,其特征在于,所述试剂盒适用于侧向流动分析,并且包含固相,所述固相包含:
a)用于施加待测样品的样品垫,
b)缀合物垫或试剂垫,其包含Klotho特异性抗体的缀合物,该缀合物包含可检测的标记,或者这种抗体/标记缀合物和Klotho的缀合物,
c)反应区或捕获区,其上固定有Klotho蛋白特异性抗体,其优选地以横跨固相宽度的线提供,和
d)用于收集样品流体和样品组分以及剩余试剂或缀合物的芯或废物储器,并且可选地进一步,
e)对照区,其包含与步骤b)的缀合物但不与Klotho特异性结合的抗体。
