Abstract
The invention discloses a traditional Chinese medicine compound for clearing away heat and toxic materials, which comprises the following medicinal materials in parts by weight: 15-25 parts of Chinese yam, 7-13 parts of poria cocos, 25-35 parts of honeysuckle, 15-25 parts of houttuynia cordata, 15-25 parts of reed rhizome, 12-18 parts of ageratum, 4-6 parts of wolfberry and 7-13 parts of liquorice. The invention also discloses a traditional Chinese medicine compound granule for clearing away heat and toxic materials, which comprises the following raw materials: dextrin and the Chinese medicinal compound for clearing away heat and toxic materials. The invention also discloses a preparation method of the traditional Chinese medicine compound granule for clearing away heat and toxic materials, which comprises the following steps: decocting rhizoma Dioscoreae, poria, flos Lonicerae, herba Houttuyniae, rhizoma Phragmitis, herba Agastaches, fructus Lycii and Glycyrrhrizae radix with water to obtain water decoction, concentrating the water decoction to obtain extract A, spray drying the extract A and dextrin, and granulating to obtain the Chinese medicinal compound granule with heat and toxic materials clearing away effects. The invention realizes the effects of strengthening body resistance and eliminating evil, clearing heat and detoxicating by the mutual compatibility of monarch, minister, assistant and guide medicines so as to improve the immunity of the organism and enhance the antiviral ability.
Description
Technical Field
The invention relates to the technical field of traditional Chinese medicine compound and preparation, in particular to a traditional Chinese medicine compound and compound granules for clearing away heat and toxic materials and a preparation method thereof.
Background
Aiming at the prevention and treatment of virus infectious diseases, the western medicine therapy has the problems of vaccine lag, virus variability, western medicine drug resistance and the like. According to the traditional Chinese medicine thought of 'three deficiency' caused epidemic and the prevention and treatment concept of 'strengthening body resistance and eliminating evil' and 'preventing and treating before disease' proposed in 'Huangdi' Nei Jing, su Wen Yi (article of ancient folk research), under the guidance of the traditional Chinese medicine basic theory, eight traditional Chinese medicines of honeysuckle, tuckahoe, wrinkled gianthyssop herb, cordate houttuynia, reed rhizome and liquorice which are homologous in medicine and food are selected for composing prescriptions and compatibility.
Yam, rhizoma Dioscoreae, sweet and neutral, is good at tonifying qi and yin, so it is said from Shen nong Ben Cao Jing that it is effective in treating injury, tonifying deficiency and win, dispelling cold and heat, eliminating pathogenic factors, tonifying middle-jiao and invigorating qi, growing muscle, improving hearing and eyesight, reducing weight and not hunger, and prolonging life.
Poria has mild property, sweet and bland taste, and has effects of invigorating spleen, calming heart, inducing diuresis and eliminating dampness.
In the formula, the Chinese yam and the tuckahoe are monarch drugs which have the effects of tonifying qi and strengthening spleen, eliminating dampness in time, clearing hollow viscera and discharging turbidity and assisting the body to strengthen vital qi; the honeysuckle, the heartleaf houttuynia herb and the reed rhizome all have the functions of clearing heat and removing toxicity and are used as ministerial drugs; in the formula, the agastache rugosus is fragrant and dampness is removed, and the medlar is capable of replenishing vital essence and nourishing blood, and moistening lung to arrest cough, and is an adjuvant drug; gancao, which enjoys the name of "Gulao", regulates the middle warmer and relieves urgency, can coordinate the effects of the drugs, relieve various toxicities, and replenish qi to invigorate the spleen, and is used as a guiding drug. The whole formula emphasizes that the invention strengthens body resistance to eliminate pathogenic factors, clears away heat and toxic materials, can benefit the organism and improve immunity and enhances antiviral ability.
The Chinese medicinal compound for clearing away heat and toxic materials can also contain medicinal adjuvants such as filler, dispersant, lubricant, etc.
Preferably, the Chinese herbal compound preparation is a granule, a decoction, a capsule, a tablet or a sustained-release preparation.
The invention also provides a traditional Chinese medicine compound granule for clearing away heat and toxic materials, which comprises the following raw materials: dextrin and the Chinese medicinal compound prescription for clearing away heat and toxic materials.
In order to improve the drug loading rate and stability, the traditional Chinese medicine compound is prepared into granules, so that the traditional Chinese medicine compound has the advantages of quick absorption, quick response, and convenient taking, carrying and storage stability.
In the preparation process of the traditional Chinese medicine granules, sucrose, starch and the like are generally selected as auxiliary materials. However, sucrose is hygroscopic, easily softened and deteriorated, starch is sticky, and meanwhile, the sugar-containing preparation is not suitable for patients with diabetes, hypertension, obesity and the like. The invention selects dextrin with smaller viscosity than starch as auxiliary material, and combines proper preparation process, so that the granule has better granule shape, good forming rate, fluidity and granule dissolubility, and can reduce hygroscopicity.
The granules prepared by the invention are beautiful and wear-resistant, have high forming rate and good fluidity, and meet the requirements of 2020 edition of Chinese pharmacopoeia on the granules.
The invention also discloses a preparation method of the traditional Chinese medicine compound granule for clearing away heat and toxic materials, which comprises the following steps: decocting rhizoma Dioscoreae, poria, flos Lonicerae, herba Houttuyniae, rhizoma Phragmitis, herba Agastaches, fructus Lycii and Glycyrrhrizae radix with water to obtain water decoction, concentrating the water decoction to obtain extract A, spray drying the extract A and dextrin, and granulating to obtain the Chinese medicinal compound granule with heat and toxic materials clearing away effects.
Preferably, the density of the extract A is 1.1-1.3g/ml.
Preferably, the weight ratio of the extract A to the dextrin is 4-8.
The invention uses a small amount of dextrin and combines a proper process, so that the granules have better granule shape, forming rate and granule dissolubility.
Preferably, during spray drying, the temperature of the extract A is increased to 65-75 ℃.
Preferably, the air inlet temperature of the spray drying is 100-110 ℃, and the air outlet temperature is 60-70 ℃.
Preferably, the atomization pressure of the spray drying is 1.5-2.5MPa, and the spray speed is 30-40Hz.
Preferably, the water is decocted and extracted for 1-3 times, the extraction time is 0.5-1.5h each time, and the water consumption for each time is 8-12 times of the total weight of the Chinese yam, the tuckahoe, the honeysuckle, the heartleaf houttuynia herb, the reed rhizome, the ageratum, the medlar and the liquorice.
Preferably, the water extraction is carried out for 3 times, the time of each water extraction is 0.5h, and the water consumption of each water extraction is 12 times of the total weight of the Chinese yam, the tuckahoe, the honeysuckle, the houttuynia cordata, the reed rhizome, the ageratum, the medlar and the liquorice.
In order to avoid the volatilization loss of the components in the houttuynia cordata and the ageratum, the houttuynia cordata and the ageratum are added at last, and a proper adding time is screened out.
Preferably, the herba Houttuyniae and herba Agastaches are added after decocting with water, and the herba Houttuyniae and herba Agastaches are added 5-15min before decocting and decocting.
The invention selects a proper extraction process, so that the invention has higher cream yield, chlorogenic acid content and ferulic acid content at the same time.

In addition, the invention selects proper auxiliary materials and combines proper granulating process, so that the granules have good granule shape, forming rate, fluidity and granule dissolubility, low hygroscopicity and accord with the requirement of 2020 edition Chinese pharmacopoeia on granules.

Hereinafter, the technical solution of the present invention will be described in detail by specific examples, but these examples should be explicitly proposed for illustration, but should not be construed as limiting the scope of the present invention.
The instrument and material information is as follows:
a T2003 electronic balance (Hangzhou Wante weighing apparatus Co., ltd.), a WB-2000 constant temperature water bath (Zhengzhou Changcheng Kogao Co., ltd.), a DZF-6050 type vacuum drying oven (Shanghai Bocheng Chengyi Co., ltd.), an RXH-7C hot air circulation oven (Nanjing Xuan Jamo pharmaceutical equipments Co., ltd.), a PGL-5B spray granulator (Changzhou Shahua drying equipments Co., ltd.), and an Alliance Waters1525 high performance liquid chromatograph (Watts Co., USA).
Chlorogenic acid standard (batch: 026Y-PD4V, institute for testing and testing Chinese food and drug), and ferulic acid standard (batch: DSTDF008101, lemeitian medicine/Designant biology).
Chinese yam, tuckahoe, honeysuckle, heartleaf houttuynia herb, reed rhizome, liquorice, patchouli and medlar (purchased from Anhui Huaxiu pharmaceutical industry Co., ltd.) and are identified by experts. The rhizoma Dioscoreae is dried rhizome of Dioscorea opposita thumb of Dioscoreaceae; poria cos (Schw.) Wolf is dry sclerotium of Poria cocos (Schw.) Wolf of Polyporaceae family; the flos Lonicerae is dried bud or flower with initial bloom of Lonicera japonica Thunb of Caprifoliaceae; the herba Houttuyniae Houttuynia cordiata Thunb is dried aerial part of Houttuynia cordata Thunb of Saururaceae; the rhizoma Phragmitis is dried rhizome of Phragmitis communis Trin of Gramineae; the Glycyrrhrizae radix is dried root and rhizome of Glycyrrhiza uralensis Fisch of Leguminosae; pogostemon cablin is dry aerial part of Pogostemon cablin (Blanco) Benth, belonging to Labiatae; the fructus Lycii is dried mature fruit of Lycium barbarum L.
Chromatographic grade methanol (batch No.: 21110106G401, european Preson Limited, sweden); chromatographic grade acetonitrile (batch: SHBM1775, merck, USA), other chemical reagents were analytically pure.
Example 1
A traditional Chinese medicine compound for clearing away heat and toxic materials comprises the following medicinal materials in parts by weight: 15g of Chinese yam, 7g of poria cocos, 25g of honeysuckle, 15g of houttuynia cordata, 15g of reed rhizome, 12g of ageratum, 4g of wolfberry and 7g of liquorice.
Example 2
A traditional Chinese medicine compound for clearing away heat and toxic materials comprises the following medicinal materials in parts by weight: 25g of Chinese yam, 13g of poria cocos, 35g of honeysuckle, 25g of houttuynia cordata, 25g of reed rhizome, 18g of ageratum, 6g of Chinese wolfberry and 13g of liquorice.
Example 3
A traditional Chinese medicine compound for clearing away heat and toxic materials comprises the following medicinal materials in parts by weight: 18g of Chinese yam, 9g of poria cocos, 28g of honeysuckle, 18g of houttuynia cordata, 18g of reed rhizome, 14g of ageratum, 4.5g of wolfberry and 9g of liquorice.
Example 4
A traditional Chinese medicine compound for clearing away heat and toxic materials comprises the following medicinal materials in parts by weight: 22g of Chinese yam, 11g of poria cocos, 32g of honeysuckle, 22g of houttuynia cordata, 22g of reed rhizome, 16g of ageratum, 5.5g of wolfberry and 11g of liquorice.
Example 5
A traditional Chinese medicine compound for clearing away heat and toxic materials comprises the following medicinal materials in parts by weight: 20g of Chinese yam, 10g of poria cocos, 30g of honeysuckle, 20g of houttuynia cordata, 20g of reed rhizome, 15g of ageratum, 5g of wolfberry and 10g of liquorice.
Example 6
According to the formula of the embodiment 5, eight medicinal materials including Chinese yam, poria cocos, honeysuckle, houttuynia cordata, reed rhizome, ageratum, wolfberry and liquorice are accurately weighed, the eight medicinal materials are fully soaked in water and then decocted and extracted, the houttuynia cordata and the ageratum are added 10min before the decoction is finished and are continuously decocted until the decoction is finished, dregs are removed through filtration to obtain water decoction, and the water decoction is subjected to reduced pressure concentration and drying to obtain dry paste powder.
Because the honeysuckle and the heartleaf houttuynia herb all contain chlorogenic acid monomer components, the chlorogenic acid has pharmacological actions of antibiosis, antivirus, antiphlogosis and the like, and is an important index of a quality control system of a plurality of medicinal materials and a Chinese medicine compound; the active component of ferulic acid contained in rhizoma phragmitis has broad-spectrum antibacterial effect, and can remarkably inhibit cold virus and respiratory syncytial virus; therefore, the inventor takes the cream yield, the chlorogenic acid content and the ferulic acid content as indexes and inspects the quality of the dry cream powder obtained under different extraction parameters. The results are shown in table 2 and fig. 1.
Fig. 1 is a typical chromatogram of the high performance liquid chromatography in example 6, wherein a is an HPLC chromatogram of a mixed reference of chlorogenic acid and ferulic acid, B is an HPLC chromatogram of the prepared dry paste powder, peak 1 is a characteristic peak of chlorogenic acid, and peak 2 is a characteristic peak of ferulic acid.
Detecting the content of chlorogenic acid and the content of ferulic acid in the dry paste powder by adopting a high performance liquid chromatography, wherein the specific detection conditions are as follows: the chromatographic column is Thermo Acclaim TM 120-C18 (4.6X 250mm,5 μm); the flow rate is 1.0 mL/min -1 The detection wavelength is 325nm, the column temperature is 30 ℃, and the sample injection amount is 10 mu L; the mobile phase A is acetonitrile, the mobile phase B is phosphoric acid water solution with the volume fraction of 0.1 percent, and the gradient elution conditions are shown in the table 1.

Remarking: the weight of the water added each time is based on the total weight of the Chinese yam, the tuckahoe, the honeysuckle, the heartleaf houttuynia herb, the reed rhizome, the ageratum, the medlar and the liquorice.
As can be seen from Table 2, the extraction times are 3 times, the times of adding water each time are 12, and the decoction time is 0.5h each time, so that the cream yield, the chlorogenic acid content and the ferulic acid content are high.
Example 7
Dividing 5g of dry extract powder prepared by the sequence number 3 in the embodiment 6 into 5 groups, preparing 1g of each group into extract A with certain density by using water, adjusting the temperature of the extract A to 65-75 ℃, and carrying out spray drying on the extract A and dextrin in a fluidized bed spray granulator to obtain the heat-clearing and detoxifying traditional Chinese medicine compound granules, wherein the air inlet temperature of the spray drying is 100-110 ℃, the air outlet temperature is 60-70 ℃, the atomization pressure is 1.5-2.5MPa, and the spray speed is 30-40Hz.
The influence of different dextrin dosages and different extract A densities on the particle properties is examined, and the results are shown in table 3 and fig. 2.

As can be seen from Table 3 and FIG. 2, when the density of extract A is 1.1-1.3g/ml and the amount of dextrin is 1g (i.e. the weight ratio of extract A to dextrin is 4-8.

Following the procedure of example 8, 3 groups of granules were prepared in parallel and the granules were examined for particle size, moisture and angle of repose. The results are shown in Table 4.

Example 9
Compound granules, 10g per bag, were prepared according to the formulation of examples 1-5 and the preparation method of example 8 and are referred to as granules 1-5, respectively.
104 patients (50 women, 54 men and age 10-65 years old) who have one or more symptoms of fever (axillary temperature is more than or equal to 37.3 ℃), sneeze, watery nasal discharge, cough, pharyngalgia, chilly fever, headache and dizziness, tired pain of the body, normal or red tongue quality, thin and white tongue coating, acute disease onset, quick transmission, epidemic and the like are screened to be homonymy and randomly divided into 6 groups, and the 6 groups have no statistical difference in aspects of vital signs, disease severity, past history, laboratory examination and the like and have comparability;
