Abstract
本发明涉及一种天然橡胶乳清发酵产物提取物、其制备方法及用途,尤其涉及一种天然橡胶乳清发酵产物提取物、其制备方法、在制备化妆品组合物中的用途以及相应的化妆品组合物。所述天然橡胶乳清发酵产物提取物在化妆品组合物中的含量为0.01%‑100%。本发明的天然橡胶乳清发酵产物提取物是以天然橡胶乳清和大米为发酵底物,经酵母菌、乳酸菌双菌发酵后获得的,其含有麦角硫因、谷胱甘肽、β‑葡聚糖、氨基酸、白坚木皮醇、果酸、甘油、维生素、矿物质等多种活性成分。

技术领域
本发明属于化妆品原料技术领域,涉及一种天然橡胶乳清发酵产物提取物、其制备方法及用途,尤其涉及一种天然橡胶乳清发酵产物提取物在化妆品中的用途。
背景技术
巴西橡胶树(Hevea brasiliensis),是大戟科橡胶树属植物,原产于南美洲亚马孙森林,在热带、亚热带地区广泛种植。
天然胶乳是产自橡胶树的一种白色乳液,其由约30%的橡胶烃粒子和70%的乳清组成。乳清中含有丰富的活性成分,包括麦角硫因、谷胱甘肽、氨基酸、白坚木皮醇、矿物质等,这些天然活性成分在化妆品领域有着广泛应用。
在橡胶加工厂,胶乳被加工成生胶或浓缩胶乳后,乳清作为废水被处理和排放。在以往的文献中,对乳清的研究集中于废水处理、肥料制备以及乳清中白坚木皮醇提取方法的研究,而对乳清中活性物质开发利用的研究较少。
专利号为US8071106[1]的美国专利,公开了一种利用大戟科、夹竹桃科、菊科、番木瓜科植物乳液开发的制剂,涂抹于局部皮肤上,能够形成胶膜,促进皮肤再生。但对于天然胶乳来讲,其长期存贮通常需要加入多种化学试剂,不适合开发成护肤产品。
Kerche-Silva等[2]进行了胶乳C-乳清(即胶乳经超速离心后所得的清液)的抗氧化活性和细胞毒性研究,结果表明:胶乳C-乳清能有效清除羟基自由基、一氧化氮自由基和过氧化氢,表现出较强的抗氧化活性;乳胶C-乳清的细胞毒性评价证明,对于源自皮肤成纤维细胞的CCD 1059sk细胞,乳胶C-乳清没有细胞毒性。同时,作者也对利用胶乳C-乳清在治疗皮肤疾病方面进行了展望。
国际专利号WO2019/022679[3]的专利,公开了一种生发产品配方,将乳清经不同截留分子量的超滤膜处理后喷雾干燥,获得的喷干粉进一步提纯,最后将提取物和生发药物米诺地尔混合制成生发药膏。
专利号为CN101204362[4]的专利公开了一种天然胶乳提取物的制备方法,乳清经超滤处理后进行喷雾干燥,再利用丙酮溶液提取粉末中的活性成分,该提取物具有美白效果。
专利号为JP18789988A[5]和JP19574088A[6]的日本专利分别公开了利用乳清中白坚木皮醇开发入浴剂和化妆品的方法,并介绍了白坚木皮醇具有保湿、平滑肌肤、增加皮肤弹性的生物活性。
Leite等[7]研究了巴西橡胶树天然乳胶乳清提取物对实验性皮肤擦伤创面的愈合作用,与洗必泰(双氯苯双胍己烷,消毒防腐药)和生理盐水相比,可促进角质形成细胞在体外的迁移/增殖,并显著加速动物表皮损伤模型的伤口愈合。
Vijayakumar等[8][9]的研究结论表明白坚木皮醇可以抑制表皮葡萄球菌和耐甲氧西林金黄色葡萄球菌的生物膜形成和毒力产生。
通过前人研究,目前已经了解到天然橡胶乳清具有抑菌、抗氧化、美白、生发、促进皮肤创伤愈合等生物活性,且对皮肤成纤维细胞没有毒性。这些研究结论为天然橡胶乳清在化妆品以及医药领域的产品开发提供了很好的理论基础。
在这些研究中,其研究思路通常是将乳清中活性成分进行提取,制成粉状或凝胶状成品后进行活性研究,也有用乳清中单一组分(如:白坚木皮醇)进行产品开发的。
发明内容
在橡胶加工厂,天然橡胶乳清被当作废水进行处理,不仅增加了企业负担,而且浪费了资源,橡胶树丰富的次生代谢产物如麦角硫因、谷胱甘肽等没有得到利用。本发明的发明人以天然橡胶乳清作为发酵底物,以大米作为补充碳源,既可以防止因碳源不足导致的发酵液变臭,也可以作为对乳清发酵液的有益补充。通过发酵工艺,将大分子物质转化为小分子活性成分,产生以果酸、甘油为代表的新的活性成分,并去除致敏性蛋白质等有害成分,实现天然橡胶乳清的资源化利用。
本发明第一个方面在于提供一种天然橡胶乳清发酵产物提取物的制备方法,包括:
A.天然橡胶乳清的获取;
B.发酵底物的制备:在天然橡胶乳清中加入碎大米和α-淀粉酶进行液化,随后加入葡糖淀粉酶进行糖化,过滤,得到糖化液;
C.双菌发酵:用酵母菌和乳酸菌发酵步骤B的糖化液,然后灭菌,得到双菌发酵液;
D.天然橡胶乳清发酵产物提取物的制备:双菌发酵液经澄清、脱色后即得到天然橡胶乳清发酵产物提取物。
其中,所述天然橡胶乳清是指天然胶乳中除橡胶粒子以外的其余物质的总称,也包括其浓缩液。天然橡胶乳清可以通过本领域常用的方法获得,例如但不限于离心法、热凝聚法、化学凝聚法、电泳法。
在一些实施方案中,步骤A通过以下方法进行:鲜胶乳经离心处理后获得胶清,任选地用酸溶液将pH调节到5.0-7.0,胶清置于夹层罐中煮沸,捞出凝胶,冷却后过超滤膜,收集滤过液,得到天然橡胶乳清。
或者,在一些实施方案中,步骤A通过以下方法进行:鲜胶乳置于夹层槽中,任选地用酸溶液将pH调节到5.0-7.0,加热使胶乳凝聚,挤压凝胶得到乳清,乳清冷却后过超滤膜,收集滤过液,得到天然橡胶乳清。
在一些实施方案中,所述超滤膜截留分子量为5k-20kDa。
在一些实施方案中,步骤B包括:任选地将天然橡胶乳清pH调节至5.0-7.0,加入碎大米,边搅拌边加入α-淀粉酶,进行液化,在这里,在加入碎大米前检测天然橡胶乳清的pH,若pH在5.0-7.0的范围内,则可以省略此处调节pH的步骤;将液化液的pH调节至4.0-4.8,加入葡糖淀粉酶,进行糖化,然后过滤得到糖化液。
在一些实施方案中,在步骤B中,天然橡胶乳清与碎大米的比例为(20:1)-(2:1),优选(10:1)-(4:1)。在这里,天然橡胶乳清与碎大米的比例为以L:kg计算的体积与质量比。
在一些实施方案中,α-淀粉酶的量为碎大米质量的0.02%-0.05%。
在一些实施方案中,液化在95℃-100℃进行30-120min。
在一些实施方案中,液化后,将液化液冷却至58-64℃后,调节液化液的pH,然后加入0.04%-0.08%碎大米质量的葡糖淀粉酶。
在一些实施方案中,糖化在58-64℃进行12-48h。
在一些实施方案中,在步骤C中,在糖化液中接入0.05%-0.25%碎大米质量的酵母菌以及同等比例的植物乳杆菌,在30-45℃发酵1-14天。发酵完成后,95℃灭菌20-30min,得到双菌发酵液。
或者,在一些实施方案中,步骤C包括:
C1.酵母菌发酵:将糖化液置于发酵罐中,接入0.05%-0.25%碎大米质量的酵母菌,在不超过42℃的温度发酵1-14天后,85℃灭菌20-60min,得到二次发酵底物;和
C2.植物乳杆菌发酵:在二次发酵底物中按植物乳杆菌:酵母菌=1:1的比例接入植物乳杆菌,发酵温度为30-45℃,发酵时间为12-72h。发酵完成后,95℃灭菌20-30min,得到双菌发酵液。
在一些实施方案中,澄清、脱色采用本领域普通技术人员熟知的方式进行,例如但不限于使用活性炭进行。
在一些实施方案中,所述方法还包括进一步浓缩、干燥或浓缩并干燥天然橡胶乳清发酵产物提取物的步骤。
本发明的第二方面提供了一种天然橡胶乳清发酵产物提取物,包含麦角硫因、谷胱甘肽、游离氨基酸、白坚木皮醇、果酸、β-葡聚糖和甘油。在这里,游离氨基酸指的是没有形成肽的氨基酸,并且包括盐形式的氨基酸。
在一些实施方案中,所述天然橡胶乳清发酵产物提取物包含:麦角硫因0.01-200mg/L,谷胱甘肽0.01-100mg/L,总游离氨基酸1000-8000mg/L,白坚木皮醇2-20g/L,以天然橡胶乳清发酵产物提取物的重量计,果酸0.21%-5.0%,β-葡聚糖0.001%-0.1%,甘油0.1-2.0%;优选地,所述天然橡胶乳清发酵产物提取物包含:麦角硫因35mg/L,谷胱甘肽27mg/L,总游离氨基酸2622.1mg/L,白坚木皮醇6.9g/L,果酸2.20%,β-葡聚糖0.016%、甘油0.67%。
在一些实施方案中,所述果酸包含乳酸、苹果酸和柠檬酸;优选地,以所述天然橡胶乳清发酵产物提取物的重量计,所述果酸包含乳酸0.1-5.0%、苹果酸0.01%-2.0%和柠檬酸0.1%-1.0%;最优选地,所述果酸包含乳酸1.22%、苹果酸0.06%和柠檬酸0.92%。
在一些实施方式中,所述天然橡胶乳清发酵产物提取物还包括在此方法基础上通过继续浓缩、干燥获得的浓缩液、浸膏或干粉。
本发明的第三方面提供了上文所述的天然橡胶乳清发酵产物提取物在制备化妆品组合物中的用途。
本发明的第四方面提供了一种化妆品组合物,包含通过本发明第一方面的方法获得的天然橡胶乳清发酵产物提取物,任选地还包含天然橡胶乳清提取物和一种或更多种化妆品领域可接受的辅料。
所述化妆品组合物可以为化妆水、乳/霜、精华液或精华乳、气雾、面膜、洁面乳、沐浴露、洗发水、护发素形式。这些形式可通过本领域技术人员熟知的方法制备。
所述化妆品领域可接受的辅料可以是:增溶剂、防腐剂、保湿剂、脂质体、增稠剂、乳化剂、表面活性剂、螯合剂、香精,以及其他功效添加剂。
在一些实施方案中,本发明的化妆品组合物可用于保湿、控油和美白。

下面通过实施例来描述本申请的实施方式,本领域的技术人员应当认识到,这些具体的实施例仅表明为了达到本申请的目的而选择的实施技术方案,并不是对技术方案的限制。根据本申请的教导,结合现有技术对本申请技术方案的改进是显然的,均属于本申请保护的范围。
其中,在以下实施例中所用的5k Da超滤膜(型号:PT4040C2016)、纳滤膜(型号:DK2540F30)购自SUEZ公司;10k Da超滤膜(型号:4040-UF10)购自MICRODYN-NADIR公司;α-淀粉酶、葡糖淀粉酶购自沧州夏盛酶生物技术有限公司;所用酵母菌购自安琪酵母股份有限公司;植物乳杆菌购自山东中科嘉亿生物工程有限公司。
天然橡胶乳清提取物和白坚木皮醇为自制,天然橡胶乳清提取物是将天然橡胶乳清用5k Da或10k Da超滤膜过滤,滤过液用纳滤膜浓缩,浓缩液过大孔树脂柱,洗去多糖、白坚木皮醇和无机盐后,用乙醇溶液洗脱活性成分,蒸馏去除乙醇,经喷雾干燥后制成干粉。白坚木皮醇是将天然橡胶乳清用1kDa的超滤膜过滤,滤过液纳滤浓缩,浓缩液经过蒸馏、结晶、重结晶后制取的。
如本文使用的,“乳清”指胶乳的分散介质,是胶乳中除橡胶粒子以外的其余物质的总称。
如本文使用的,“胶清”指胶乳通过离心机浓缩后,所得的干胶含量约5%的副产品,即含有5%橡胶烃粒子的乳清。
实施例1
新鲜胶乳经离心分离出胶清,将胶清置于夹层罐内,用10%盐酸溶液调节pH至6.0,加热至沸,捞出凝胶,冷却后用10kDa的超滤膜过滤,滤过液进一步用纳滤膜浓缩至原体积的15%,得到天然橡胶乳清浓缩液。
取100L乳清浓缩液置于夹层罐内,按5:1的比例加入20kg碎大米,搅拌中加入0.05%大米质量的α-淀粉酶10g,加热至95℃以上,并保持60min至液化完成。液化液冷却至60℃,用20%柠檬酸溶液调节pH至4.7,加入0.06%大米质量的葡糖淀粉酶12g,在60℃保持30h,糖化完成,过滤得到初级发酵底物。
糖化液置于发酵罐中,接入0.1%大米质量的酵母菌20g,在室温发酵7天后,85℃灭菌30min,得到二次发酵底物。
在二次发酵底物中接入20g植物乳杆菌,42℃发酵24h后,95℃灭菌20min,得到双菌发酵液。
双菌发酵液澄清后,用1%活性炭脱色,0.1μm膜过滤后得到天然橡胶乳清发酵产物提取物。
对比了发酵前后溶液中活性成分的变化情况,发酵前,天然橡胶乳清浓缩液中麦角硫因含量为38mg/L,谷胱甘肽41mg/L,总游离氨基酸2051.06mg/L,白坚木皮醇7.8g/L。发酵后,天然橡胶乳清发酵产物提取物中麦角硫因含量为35mg/L,谷胱甘肽27mg/L,总游离氨基酸2622.1mg/L,白坚木皮醇6.9g/L。此外,发酵前的天然橡胶乳清浓缩液不含果酸、β-葡聚糖、甘油成分,经过双菌发酵,天然橡胶乳清发酵产物提取物含2.20%的果酸(其中乳酸1.22%,苹果酸:0.06%,柠檬酸0.92%)、0.016%的β-葡聚糖和0.67%的甘油,在这里,果酸、β-葡聚糖和甘油的含量以天然橡胶乳清发酵产物提取物的重量计。
实施例2
鲜胶乳置于夹层槽中,用10%盐酸溶液调节pH至5.5,加热使胶乳凝聚,挤压凝胶得到乳清,乳清冷却后用5kDa的超滤膜过滤,收集滤过液,得到可使用的天然橡胶乳清。
取100L乳清置于夹层罐内,按10:1的比例在乳清中加入10kg碎大米,加入0.02%大米质量的α-淀粉酶2g,加热至95℃以上,并保持120min至液化完成。液化液冷却至60℃,用20%柠檬酸溶液调节pH至4.5,加入0.04%大米质量的葡糖淀粉酶4g,在60℃保持24h,糖化完成,过滤后即得到发酵底物。
发酵底物置于发酵罐中,按0.1%大米质量的比例依次接入酵母菌20g和植物乳杆菌20g,37℃发酵7d后,95℃灭菌30min,得到双菌发酵液。
发酵液澄清后,减压蒸馏浓缩至原体积的50%,用1%活性炭脱色,0.22μm膜过滤后,得到天然橡胶乳清发酵产物提取物的浓缩液。
实施例3 DPPH自由基清除试验
DPPH是一种稳定的自由基,被广泛用于评价天然产物清除自由基的能力。
1试验方法
①取4ml样品溶液于试管中,加入4ml 2x10-4mol/L的DPPH溶液,混合均匀,室温、暗室反应30min后,以无水乙醇调零,在515nm处测定其吸光度Ai。
②取4ml样品溶液于试管中,加入4ml无水乙醇溶液,混合均匀,室温、暗室反应30min后,以无水乙醇调零,在515nm处测定其吸光度Aj。
③取4ml无水乙醇溶液于试管中,加入4ml 2x10-4mol/L的DPPH溶液,混合均匀,室温、暗室反应30min后,以无水乙醇调零,在515nm处测定其吸光度AC。

结果显示,未发酵的天然橡胶乳清对DPPH自由基清除率为80.12%,天然橡胶乳清发酵产物提取物样品对DPPH自由基显示出更好的清除能力,清除率高达99.53%。
实施例4爽肤水的制备
爽肤水1:
一种化妆品组合物,由如下质量百分数的组分组成:根据实施例1得到的天然橡胶乳清发酵产物提取物97%、天然橡胶乳清提取物2.0%、白坚木皮醇1.0%。
将上述组分混合均匀,充分溶解后,即得化妆品组合物。
爽肤水2:
一种化妆品组合物,由如下质量百分数的组分组成:根据实施例1得到的天然橡胶乳清发酵产物提取物97.2%、天然橡胶乳清提取物2.0%、山梨酸0.3%、苯甲酸钠0.3%、羟苯甲酯0.2%。
将上述组分混合均匀,充分溶解后,即得化妆品组合物。
爽肤水1是以植物乳杆菌发酵产生的细菌素、果酸作为天然防腐剂,和爽肤水2一起模拟正常使用,一年后两种爽肤水都没有出现发霉、浑浊等变质情况。
本发明制备的爽肤水不仅含有近年来关注度极高的两种抗氧化剂麦角硫因和谷胱甘肽,而且谷胱甘肽是“美白针”的主要成分,具有美白效果。白坚木皮醇和甘油两种多元醇保湿剂与游离氨基酸一起对皮肤起到保湿、滋润的作用。果酸具有优异的平衡皮肤油脂的作用,高浓度果酸常被用于治疗脸部痤疮,即称为“果酸换肤”的医疗美容项目。这些活性成分赋予了本发明优异的护肤美容属性。

Claims (23)
1.一种天然橡胶乳清发酵产物提取物的制备方法,包括:
A.天然橡胶乳清的获取;
B.发酵底物的制备:在天然橡胶乳清中加入碎大米和α-淀粉酶进行液化,随后加入葡糖淀粉酶进行糖化,过滤,得到糖化液;
C.双菌发酵:用酵母菌和乳酸菌发酵步骤B的所述糖化液,然后灭菌,得到双菌发酵液;和
D.天然橡胶乳清发酵产物提取物的制备:所述双菌发酵液经澄清、脱色后即得到天然橡胶乳清发酵产物提取物。
2.根据权利要求1所述的方法,其中所述天然橡胶乳清通过离心法、热凝聚法、化学凝聚法或电泳法获得。
3.根据权利要求1所述的方法,其中步骤A通过以下方法进行:鲜胶乳经离心处理后获得胶清,任选地用酸溶液将pH调节到5.0-7.0,将所述胶清煮沸,捞出凝胶,待冷却后过超滤膜,收集滤过液,得到天然橡胶乳清。
4.根据权利要求1所述的方法,其中步骤A通过以下方法进行:鲜胶乳任选地用酸溶液将pH调节到5.0-7.0,加热使胶乳凝聚,挤压凝胶得到乳清,待乳清冷却后过超滤膜,收集滤过液,得到天然橡胶乳清。
5.根据权利要求3或4所述的方法,其中所述超滤膜截留分子量为5kDa-20kDa。
6.根据权利要求1所述的方法,其中步骤B包括:任选地将天然橡胶乳清的pH调节至5.0-7.0,加入碎大米,边搅拌边加入α-淀粉酶,进行液化;将液化液的pH调节至4.0-4.8,加入葡糖淀粉酶,进行糖化,然后过滤得到糖化液。
7.根据权利要求6所述的方法,其中在步骤B中,所述天然橡胶乳清与所述碎大米的比例为(20:1)-(2:1),优选(10:1)-(4:1)。
8.根据权利要求6所述的方法,其中所述α-淀粉酶的量为碎大米质量的0.02%-0.05%。
9.根据权利要求6所述的方法,其中液化在95℃-100℃进行30-120min。
10.根据权利要求6所述的方法,其中在液化后,将液化液冷却至58-64℃后,调节液化液的pH,然后加入0.04%-0.08%碎大米质量的葡糖淀粉酶。
11.根据权利要求6所述的方法,其中糖化在58-64℃进行12-48h。
12.根据权利要求1所述的方法,其中在步骤C中,在糖化液中接入0.05%-0.25%碎大米质量的酵母菌以及同等比例的植物乳杆菌,在30-45℃发酵1-14d,发酵完成后,95℃灭菌20-30min,得到双菌发酵液。
13.根据权利要求1所述的方法,其中步骤C包括:
C1.酵母菌发酵:将糖化液置于发酵罐中,接入0.05%-0.25%碎大米质量的酵母菌,在不高于42℃的温度发酵1-14天后,85℃灭菌20-60min,得到二次发酵底物;和
C2.植物乳杆菌发酵:在二次发酵底物中按植物乳杆菌:酵母菌=1:1的比例接入植物乳杆菌,发酵温度为30-45℃,发酵时间为12-72h,发酵完成后,95℃灭菌20-30min,得到双菌发酵液。
14.根据权利要求1所述的方法,其中使用活性炭进行澄清、脱色。
15.根据权利要求1所述的方法,其中所述方法还包括进一步浓缩、干燥或浓缩并干燥天然橡胶乳清发酵产物提取物的步骤。
16.一种天然橡胶乳清发酵产物提取物,其包含麦角硫因、谷胱甘肽、游离氨基酸、白坚木皮醇、果酸、β-葡聚糖和甘油。
17.根据权利要求16所述的天然橡胶乳清发酵产物提取物,其中所述天然橡胶乳清发酵产物提取物包含:麦角硫因0.01-200mg/L,谷胱甘肽0.01-100mg/L,总游离氨基酸1000-8000mg/L,白坚木皮醇2-20g/L,以所述天然橡胶乳清发酵产物提取物的重量计果酸0.21%-5.0%,β-葡聚糖0.001%-0.1%,甘油0.1%-2.0%;优选地,所述天然橡胶乳清发酵产物提取物包含:麦角硫因35mg/L,谷胱甘肽27mg/L,总游离氨基酸2622.1mg/L,白坚木皮醇6.9g/L,果酸2.20%,β-葡聚糖0.016%、甘油0.67%。
18.根据权利要求17所述的天然橡胶乳清发酵产物提取物,其中所述果酸包含乳酸、苹果酸和柠檬酸;优选地,以所述天然橡胶乳清发酵产物提取物的重量计,所述果酸包含乳酸0.1%-5.0%、苹果酸0.01%-2.0%和柠檬酸0.1%-1.0%;最优选地,所述果酸包含乳酸1.22%、苹果酸0.06%和柠檬酸0.92%。
19.根据权利要求16-18中任一项所述的天然橡胶乳清发酵产物提取物,所述天然橡胶乳清发酵产物提取物通过权利要求1-15中任一项所述的方法制备。
20.天然橡胶乳清发酵产物提取物在制备化妆品组合物中的用途,所述天然橡胶乳清发酵产物提取物为根据权利要求16-19中任一项所述的天然橡胶乳清发酵产物提取物。
21.根据权利要求20所述的用途,其中所述天然橡胶乳清发酵产物提取物和所述化妆品组合物可用于保湿、控油和美白。
22.一种化妆品组合物,包含通过权利要求16-19中任一项所述的天然橡胶乳清发酵产物提取物,任选地还包含天然橡胶乳清提取物和一种或更多种辅料,优选地,所述辅料为增溶剂、防腐剂、保湿剂、脂质体、增稠剂、乳化剂、表面活性剂、螯合剂、香精,以及其他功效添加剂。
23.根据权利要求22所述的化妆品组合物,其中按重量计,所述天然橡胶乳清发酵产物提取物占所述化妆品组合物的0.01%-100%。
