Further, the preparation method of the diluted vaccinia virus comprises the following steps:
selecting male rabbits for passage; step two, quarantine observation; step three, inoculating the male rabbits; step four, feeding and observing the inoculated male rabbits; step five, taking testicles of male rabbits; step six, repairing and washing the testicles; seventhly, mashing the adopted testis; step eight, freezing and thawing the mashed testis; step nine, centrifuging the frozen and thawed testicles; step ten, subpackaging the centrifuged testis; and step eleven, diluting the vaccinia virus.
Further, the dilution factor of the vaccinia virus is 1000-fold.
Further, the vaccinia virus after injection dilution is fed to observe the pox occurrence: the number of the pox with the diameter of 1-2 cm is more than 80, and the number of the pox with the diameter of less than 1cm is more than 100.
Furthermore, the rapid freezing adopts the rapid cooling temperature of the rabbit skin of-18 ℃, the adopted rabbit skin needs to be complete, the color is reddish, no putrefactive odor is generated, and the pox is purplish red.
Further, the fifth step, the sixth step, the seventh step, the tenth step and the eleventh step are all performed inside the biosafety cabinet.
(III) advantageous effects
Compared with the prior art, the invention provides a production method of diluted vaccinia virus inflammation-causing rabbit skin, which has the following beneficial effects:
1. after the vaccinia virus seeds are diluted by 1000 times, the inflammatory rabbit skin meeting the quality standard of the inflammatory rabbit skin caused by vaccinia virus can be produced, and the production requirement for the inflammatory rabbit skin can be met only by producing 5 batches of vaccinia virus seeds; 6 batches of vaccinia virus seeds are produced in a small amount, and the operating cost of workshop facilities can be saved by 40 ten thousand yuan; a large amount of labor and plant facility operating cost is saved for a company;
2. if the vaccinia virus seeds can produce the inflammatory rabbit skin meeting the quality standard of the inflammatory rabbit skin of the vaccinia virus seeds after being diluted by 1000 times, 22500 sheets of inflammatory rabbit skin can be produced by each batch of antigen, 67.5 ten thousand sheets of inflammatory rabbit skin can be produced every year, and a solid foundation is laid for the future production expansion and sufficient raw material supply.
Drawings
In order to more clearly illustrate the embodiments of the present invention or the technical solutions in the prior art, the drawings needed in the embodiments will be briefly described below, and it is obvious that the drawings in the following description are only some embodiments of the present invention, and it is obvious for those skilled in the art to obtain other drawings without creative efforts.
FIG. 1 is a schematic flow chart of the inflammatory rabbit skin method of the present invention;
FIG. 2 is a flow chart of the preparation of diluted vaccinia virus according to the invention.
Detailed Description
For further explanation of the various embodiments, the drawings which form a part of the disclosure and which are incorporated in and constitute a part of this specification, illustrate embodiments and, together with the description, serve to explain the principles of operation of the embodiments, and to enable others of ordinary skill in the art to understand the various embodiments and advantages of the invention, and, by reference to these figures, reference is made to the accompanying drawings, which are not to scale and wherein like reference numerals generally refer to like elements.

In one embodiment, the method for preparing a diluted vaccinia virus comprises the steps of:
selecting male rabbits for passage; step two, quarantine observation; step three, inoculating the male rabbits; step four, feeding and observing the inoculated male rabbits; step five, taking testicles of male rabbits; step six, repairing and washing the testicles; seventhly, mashing the adopted testis; step eight, freezing and thawing the mashed testis; step nine, centrifuging the frozen and thawed testicles; step ten, subpackaging the centrifuged testis; and step eleven, diluting the vaccinia virus.
In one embodiment, the vaccinia virus is diluted 1000-fold.
In one embodiment, the diluted vaccinia virus feed is administered to observe pox onset: the number of the pox with the diameter of 1-2 cm is more than 80, and the total number of the pox with the diameter of less than 1cm is more than 100.
In one embodiment, the rapid freezing temperature of the rabbit skin is-18 ℃, the rabbit skin needs to be complete, reddish in color, without putrefactive odor, and purplish red in whelk.
In one embodiment, the fifth step, the sixth step, the seventh step, the tenth step and the eleventh step are all performed inside the biosafety cabinet.
Although the production cost of batches of vaccinia virus seeds is not changed greatly (about 200 yuan/ml), the use cost of each milliliter of vaccinia virus seeds is obviously reduced, if 10 ten thousand of inflammatory rabbit skins are produced in one year, and the current vaccinia virus seeds are diluted by 1; if the vaccinia virus seeds can produce the inflammatory rabbit skin meeting the quality standard of the inflammatory rabbit skin of the vaccinia virus after being diluted by 1000 times, the production requirement for the inflammatory rabbit skin can be met only by producing 5 batches of vaccinia virus seeds; 6 batches of vaccinia virus seeds are produced in a small amount, and the operating cost of workshop facilities can be saved by 40 ten thousand yuan; in addition, only 2 months are needed for producing 5 batches of vaccinia virus seeds, and the workers in the vaccinia virus seed workshop can be distributed to other workshops for 10 months, so that a large amount of labor and plant facility operating cost are saved for a company.
If 30 batches of vaccinia virus seeds are produced at full load in one year, the yield of one batch of vaccinia virus seeds is about 600ml at present, the dilution multiple of the current vaccinia virus seeds is 1; if the vaccinia virus seeds can produce the inflammatory rabbit skin meeting the quality standard of the inflammatory rabbit skin of the vaccinia virus seeds after being diluted by 1000 times, 22500 sheets of inflammatory rabbit skin can be produced by each batch of antigen, 67.5 ten thousand sheets of inflammatory rabbit skin can be produced every year, and a solid foundation is laid for the future production expansion and sufficient raw material supply.
