Example 1:
a traditional Chinese medicine composition with an anti-alcohol function is prepared from the following raw materials in parts by weight: 10 parts of kudzu root, 10 parts of reed rhizome, 5 parts of mulberry leaf, 5 parts of chrysanthemum, 5 parts of lophatherum gracile, 10 parts of lily, 10 parts of polygonatum, 15 parts of Chinese yam, 5 parts of raw gardenia and 5 parts of mint.
The preparation method of the traditional Chinese medicine composition comprises the following steps:
step one, weighing a certain proportion of kudzu root, reed rhizome, mulberry leaf, chrysanthemum, lophatherum gracile, lily, polygonatum, chinese yam, raw gardenia and mint according to a weight ratio, and crushing and sieving;
weighing raw materials in a certain proportion, crushing and sieving the raw materials, adding a solvent with the volume of 8-10 times of that of the raw materials, extracting, concentrating under reduced pressure, drying, crushing for the second time, and sieving.
The solvent is one of water, ethanol or a mixture of water and ethanol, and preferably water.
Step two, uniformly mixing the sieved raw materials with sorbitol to obtain raw material mixed powder;
step three, carrying out wet granulation on the raw material mixed powder by using alcohol and carrying out drying treatment;

The preparation method of the traditional Chinese medicine composition is the same as that of example 1, wherein the drying temperature, the drying time and the selected solvent are the same as those of example 1.
Example 3:
a traditional Chinese medicine composition with an anti-alcohol function is prepared from the following raw materials in parts by weight: 30 parts of kudzuvine root, 30 parts of reed rhizome, 10 parts of mulberry leaf, 15 parts of chrysanthemum, 10 parts of lophatherum gracile, 30 parts of lily, 30 parts of fragrant solomonseal rhizome, 40 parts of Chinese yam, 15 parts of raw gardenia and 15 parts of mint.
The preparation method of the traditional Chinese medicine composition is the same as that of example 1, wherein the drying temperature, the drying time and the selected solvent are the same as those of example 1.
The preparation method in the embodiment can quickly prepare the traditional Chinese medicine composition with the function of relieving alcoholism according to the requirement of the dosage form, is simple and efficient, and is suitable for batch production and application of the traditional Chinese medicine composition.
The traditional Chinese medicine composition prepared by the embodiment can reduce the uncomfortable drinking feeling, protect gastric mucosa, reduce liver injury, improve the sobering-up speed and has high safety.
Animal experiments are carried out by taking the composition obtained in the example 1 as an experimental sample, and the hangover alleviating effect and the protective effect on alcoholic liver injury of the composition after drinking are verified.
1 materials of the experiment
1.1 Experimental animals
C57BL/6 mice, SPF grade, 21-24g, purchased from Zhuhai Baidiantong Biotech, inc.
1.2 drugs and reagents
Haiwang Jinzun tablets, the composition prepared in example 1, glutamic-oxaloacetic transaminase (AST) and glutamic-pyruvic transaminase (ALT) determination kit (Nanjing institute of bioengineering Co., ltd.), and tissue Alcohol Dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) determination kit (Nanjing institute of bioengineering Co., ltd.).
1.3 instruments
Electronic balance, constant temperature metal bath, low temperature centrifuge.
2 method of experiment
2.1 determination of alcohol consumption
9 mice were randomly assigned, 3 per group. After fasting for 12 hours, the mice with 56-degree red star Erguotou gavage are respectively given three doses of 0.2ml/20g, 0.3ml/20g and 0.4ml/20g of body weight and are respectively marked as a small-drinking group, a medium-drinking group and a large-drinking group.
After the stomach is filled, the drunkenness state of the mouse is observed, drunkenness is judged according to disappearance of righting reflex, and drunkenness and sobering time and death number of the mouse in 24 hours are recorded.
Calculating the drunkenness rate and the death rate according to the following formula:

As can be seen from Table 1, the small-drinking group did not die but the intoxication rate was low (33%), the large-drinking group all died, the medium-drinking group all drunk and did not die within 24h, and the stomach-feeding amount of the medium-drinking group of 0.3ml/20g was supposed to be the drinking dose in the subsequent experiments.
2.2 determination of antialcoholism and hepatoprotective action of drugs
Blank control group: when the drug is administrated, distilled water is given, and the stomach of the mouse is irrigated according to the weight of 0.3ml/20 g; after 30min, the mice were gavaged with distilled water at a volume of 0.3ml/20 g.
Model control group: when the drug is administrated, distilled water is given, and the stomach of the mouse is irrigated according to the weight of 0.3ml/20 g; after 30min, the mice are administrated to 56-degree Hongxing Erguotou intragastric mice according to the drinking dose measurement of the mice and the weight of 0.3ml/20 g.
Positive control group: pulverizing Haiwang Jinzun tablets, dissolving and diluting to 83.3mg/ml liquid medicine according to the weight and the mouse administration conversion coefficient of 9.1, and gavage the mouse according to 0.3ml/20g of the weight; after 30min, the mice are administrated to 56-degree Hongxing Erguotou intragastric mice according to the drinking dose measurement of the mice and the weight of 0.3ml/20 g.
Administration group: according to the calculation of the body weight and the mouse administration conversion factor of 9.1, the composition obtained in the example 1 is dissolved and diluted, the liquid medicines of the low, medium and high dose groups are respectively 83.3mg/ml,166.7mg/ml and 333.3mg/ml, and the mice are gavaged according to the body weight of 0.3ml/20 g; after 30min, the mice are administrated to 56-degree Hongxing Erguotou intragastric mice according to the drinking dose measurement of the mice and the weight of 0.3ml/20 g. The low dose group, the medium dose group and the high dose group were designated, respectively.
30 mice were randomly assigned, 5 per group. After fasting for 12h without water prohibition, the stomach-irrigation administration and drinking are carried out according to the method. After 8h of intragastric administration and drinking, blood and liver were sacrificed for testing. The liver was weighed, homogenized in 10-fold 0.9% physiological saline, centrifuged at 10000rpm, and the supernatant was used to determine the activity of ADH and ALDH enzymes. Standing whole blood at room temperature for 1 hr, centrifuging at low temperature 3000rpm for 20min, collecting serum, and measuring ALT and AST enzyme activities in serum.

As can be seen from Table 2 and FIG. 1, the level of Alcohol Dehydrogenase (ADH) activity in the liver of mice after drinking alcohol increased significantly (P < 0.01), and the composition increased the level of Alcohol Dehydrogenase (ADH) activity significantly compared to the model group, and increased the gradient with the increase of the dosage. Acetaldehyde dehydrogenase (ALDH) activity level did not change significantly in all groups, and ALDH activity was higher than ADH activity, and accumulation of acetaldehyde, an intermediate product of alcohol metabolism, did not occur. Experiments show that the composition can obviously improve the alcohol metabolism level after drinking, and is beneficial to sobering up.
After drinking, the level of aspartate Aminotransferase (AST) in the serum of a mouse is obviously increased (P is less than 0.01), compared with a model group, the AST level is obviously reduced by the composition, the effect is improved along with the administration dosage, and the effect is most obvious in a high-dosage group (P is less than 0.001). The level of glutamic-pyruvic transaminase (ALT) is obviously increased (P is less than 0.05) after drinking, and compared with a model group, the level of ALT in a high dose is obviously reduced (P is less than 0.05).
In serum transaminase indexes, alanine Aminotransferase (ALT) is very sensitive under liver injury, and the ALT level is doubled after every 1% of hepatic cell injury; in alcoholic liver injury, the rise of aspartate Aminotransferase (AST) is obvious, and the rise of alanine Aminotransferase (ALT) is not obvious or unchanged. In the experiment, AST is obviously increased after drinking wine, and ALT is not changed greatly (less than 1 time), which indicates that alcohol causes certain damage to the liver after drinking wine by the model group of mice; after the composition is taken, the AST level is obviously reduced, which shows that the composition has obvious inhibiting effect on alcoholic liver injury and has protective effect on liver.

The traditional Chinese medicine composition with the hangover alleviating function according to claim 1, wherein the traditional Chinese medicine composition is prepared from the following raw materials in parts by weight: 10-30 parts of kudzuvine root, 10-30 parts of reed rhizome, 5-10 parts of mulberry leaf, 5-15 parts of chrysanthemum, 5-10 parts of lophatherum gracile, 10-30 parts of lily, 10-30 parts of fragrant solomonseal rhizome, 15-40 parts of Chinese yam, 5-15 parts of raw gardenia and 5-15 parts of mint.
3. The traditional Chinese medicine composition with the hangover alleviating function according to claim 1, which is prepared from the following raw materials in parts by weight: 20 parts of kudzu root, 20 parts of reed rhizome, 8 parts of mulberry leaf, 10 parts of chrysanthemum, 8 parts of lophatherum gracile, 8 parts of lily, 20 parts of polygonatum, 25 parts of Chinese yam, 10 parts of raw gardenia and 10 parts of mint.
4. The traditional Chinese medicine composition with the hangover alleviating function according to claim 1, further comprising pharmaceutically or food acceptable excipients or additives.
5. The traditional Chinese medicine composition with an anti-alcohol function according to claim 4, wherein the auxiliary materials or additives are one or more of erythritol, sorbitol, microcrystalline cellulose, strawberry fruit powder, maltodextrin, stevioside, magnesium stearate and starch.
6. The preparation method of the traditional Chinese medicine composition with the hangover alleviating function according to any one of claims 1 to 5, wherein the preparation method comprises the following steps:
step one, weighing a certain proportion of kudzu root, reed rhizome, mulberry leaf, chrysanthemum, lophatherum gracile, lily, polygonatum, chinese yam, raw gardenia and mint, and crushing and sieving;
step two, uniformly mixing the sieved raw materials with sorbitol to obtain raw material mixed powder;
step three, carrying out wet granulation on the raw material mixed powder by using alcohol and drying;
step four, mixing the dried granules with magnesium stearate and uniformly stirring to obtain target mixed powder;
and step five, carrying out dosage form treatment on the target mixed powder to obtain the traditional Chinese medicine composition of the target dosage form.
7. The preparation method of the Chinese medicinal composition with the hangover alleviating function according to claim 6, wherein the drying temperature in the third step is 60 ℃, the drying time is 60min, and the moisture content in the dried granules is below 5%.
