Abstract

技术领域
本发明涉及能同种异体给药的来自人T细胞来源的iPS细胞的细胞毒性T细胞及其制造方法。
背景技术
抗原特异性的细胞毒性T细胞(CTL)经由存在于其细胞表面上的T细胞受体(TCR),识别与抗原呈递细胞的class 1主要组织相容抗原(MHC I类、HLA I类)一起呈递的、来自病毒、肿瘤等的抗原肽,对于呈递作为异物的该抗原肽的细胞,特异性地发挥细胞毒性并攻击。另外,CTL的一部分成为长期生存型记忆T细胞,维持着对异物的细胞毒性而存储在宿主内,接下来,在曝露于异物的情况下可以进行应对。因此,CTL作为病毒感染患者、癌症疾病中的免疫细胞疗法的细胞而受到期待。
在慢性病毒感染患者、癌症患者中,由于慢性地曝露在抗原中,因此,T细胞会耗竭、老化,无法发挥效果,因此,在很多情况下无法得到T细胞疗法的效果。因此,利用iPS技术使耗竭的T细胞在功能上恢复活力,将得到的恢复活力的T细胞向患者给药的来自iPS细胞的恢复活力的T细胞疗法被期待成为提高癌症治疗效果的有效手段。作为使用该恢复活力的T细胞疗法的得到CTL的手段,开发了:由具有抗原特异性的T细胞建立来自T细胞的iPS细胞(T-iPS细胞),保持原本的T细胞的TCR基因的改造结构并再次分化诱导为CTL、嵌合抗原受体T细胞(CART)等的方法(专利文献1)。
但是,在这些方法中,存在制作需要5个月,且制作成本高的课题。对此,如果预先备份来自同种异体CTL的iPS细胞,制作恢复活力的T细胞(rejT),则可以更迅速地向患者给药,可以削减每一位患者的成本,但是,如果HLA不一致就会被排斥,产生抗肿瘤效果减弱的问题。
为了解决该问题,如果制作使HLA I类表达消失、不被患者CD8+T细胞排斥的同种异体rejT,则可以维持抗原特异性的CTL的强力的抗肿瘤效果,能对大量重症患者迅速地给药。为了能使将来自健康人供体的同种细胞验证备份过的iPS细胞向大量患者给药,通过HLA基因组编辑而敲除B2M,从而使HLA I类抗原消失的方法是有希望的,在该情况下,需要抑制NK细胞的missing-self应答。作为该手段,据报道,已有为了避免NK细胞的missing-self应答的若干编辑方法:分化诱导使iPS细胞的HLA I类消失而仅表达HLA-E的细胞,避免来自NK细胞的攻击的手段(非专利文献1);使HLA I类和I类I消失,在此之上使PD-L1,HLA-G和“don’t-eat me”signal CD47表达的手段(非专利文献2);使HLA-A和B消失,保持HLA-C的手段(非专利文献3)等。

发明内容
但是,在任一手段中,均无法充分满足在NK细胞的missing-self应答的避免和原本所具有的抗原特异性的CTL活性的保持的平衡这一方面。
因此,本发明的课题是提供一种来自人T细胞来源的iPS细胞的细胞毒性T细胞,其可以维持抗原特异性的CTL的强力的抗肿瘤效果,避免NK细胞的missing-self应答,能进行同种异体给药;以及其制造方法。
因此,本发明人发现,对于人T细胞来源的iPS细胞,通过使CTL的抗原表位的HLA限制性的I类分子(例如,如果是HLA-A24限制性的CTL则为HLA-A24,如果是HLA-A02限制性的CTL则为HLA-A02)和HLA-E表达,从而可以更强力地抑制NK细胞的活性,由此完成了本发明。
即,本发明提供以下[1]~[4]。
[1]一种来自人T细胞来源的iPS细胞的细胞毒性T细胞,其表达CTL的抗原表位的HLA限制性的I类分子和HLA-E的HLA I类。
[2]根据[1]所述的来自人T细胞来源的iPS细胞的细胞毒性T细胞,其中,CTL的抗原表位的HLA限制性的I类分子为HLA-A24或HLA-A02。
[3]一种表达CTL的抗原表位的HLA限制性的I类分子和HLA-E的2种HLA I类的、来自人T细胞来源的iPS细胞的细胞毒性T细胞的制造方法,其特征在于,包含:将人T细胞来源的iPS细胞的HLA I类全部敲除的工序;在HLA I类被全部敲除的T细胞中,导入CTL的抗原表位的HLA限制性的I类分子和HLA-E的基因的工序;以及,将导入了上述基因的T-iPS细胞再分化为CD8单阳性T细胞的工序。

根据本发明,可以稳定地提供一种来自人T细胞来源的iPS细胞的细胞毒性T细胞,其可以维持抗原特异性的CTL的强力的抗肿瘤效果,避免NK细胞的missing-self应答,能进行同种异体给药;以及其制造方法。

图3为表示107a检测中,HLA-A24+HLA-E表达HPV-rejT的NK细胞毒性抑制效果强的图。K562细胞株不表达I类,因此,NK细胞通过共培养而攻击K562,高表达107a。用作阳性对照组。Positive Ctrl对NK细胞施加Cell Stimulation Cocktail而刺激,因此,NK细胞高表达107a。同样地表示阳性对照组。HLA-E KI表示在HLA I类消失后仅敲入HLA-E的HPV-rejT。HLA-A24 KI表示在HLA I类消失后仅敲入HLA-A24的HPV-rejT。HL-A24+E KI表示在HLA I类消失后入HLA-A24和HLA-E这两者的HPV-rejT。Negative Ctrl表示仅以NK细胞培养的阴性对照组。
图4表示对于宫颈癌患癌小鼠的HLA编辑HPV-rejT的生存时间延长效果。非处置表示非处置组,原始CTL表示原始CTL处置组,WTrejT表示rejT处置组,EXrejT表示本发明HPV-rejT处置组。
图5表示HLA编辑HPV-rejT的NK细胞同时给药时的生物体内的耐久性。HPV-rejT中的括号内表示HLA I类编辑。
具体实施方式
本发明的细胞毒性T细胞为使人T细胞来源的iPS细胞(T-iPS细胞)的HLA表达CTL的抗原表位的HLA限制性的I类分子和HLA-E的HLA I类的细胞毒性T细胞。
另外,本发明的细胞毒性T细胞中,除了CTL的抗原表位的HLA限制性的I类分子和HLA-E以外,进而也可以表达HLA-G、HLA-C等HLA I类,进而还可以表达CD47、PD-L1、iCaspase9等。其中,如果考虑到NK细胞活性的抑制的观点、以及通过HLA的多型而使供体与HLA结合的观点,则优选为表达CTL的抗原表位的HLA限制性的I类分子和HLA-E的2种的HLAI类的、来自人T细胞来源的iPS细胞的细胞毒性T细胞。
用作原料的人T细胞来源的iPS细胞可通过将人T细胞诱导为iPS细胞(T-iPS细胞)而得到。该T-iPS细胞的制造方法优选通过上述专利文献1所述的方法进行。
首先,对由人T细胞向iPS细胞的诱导手段进行说明。
使用的T细胞优选为人T细胞。作为该T细胞的起源、即人,可以是患有病毒感染症、恶性肿瘤等的人,但是,考虑到在制造治疗用同种抗原特异性的细胞毒性T细胞后,在基因组编辑后储存化而向大量的人给药的点,优选为健康人。另外,作为T细胞的起源而优选的人与应给药使用通过本发明而制造的T-iPS细胞而制造的再生CTL、CART细胞的患者的HLA的类型无需完全一致。
本发明中,诱导为T-iPS细胞的T细胞优选为具有抗原特异性的T细胞。例如为表达CD3和CD8的T细胞,具体来说可举出作为CD8阳性细胞的CTL。另外,例如为表达CD3和CD4的T细胞,具体来说可举出作为CD4阳性细胞的T细胞。应予说明,T细胞中的抗原特异性是通过抗原特异性的重组的TCR基因而得到的。应予说明,从制造效率的观点出发,没有特别限定,为了得到抗原特异性的CD8阳性细胞,作为可以诱导为T-iPS细胞的人T细胞,优选使用抗原特异性的CD8阳性T细胞;为了得到抗原特异性的CD4阳性细胞,作为可以诱导为T-iPS细胞的人T细胞,优选使用抗原特异性的CD4阳性T细胞。另外,在实施免疫疗法的情况下,由iPS细胞分化的人T细胞优选与可以诱导为iPS细胞的人T细胞的抗原特异性相同或实质上相同。另外,作为诱导T-iPS细胞的T细胞,也包含没有抗原特异性的T细胞。具体来说,可举出CART细胞或者TCR-T细胞等转基因T细胞。
这样的T细胞例如可以由人的组织通过公知的手法而单离。
作为人的组织,可举出包含上述T细胞的组织,例如外周血、淋巴结、骨髓、胸腺、脾脏、脐带血、病变组织。在这些中,从对人的侵入性低、且制备容易的观点出发,优选为外周血。在分离肿瘤浸润淋巴细胞(TIL)时,可以从肿瘤组织或者外周血分离。作为用于单离人T细胞的公知的手法,例如可举出使用细胞分离用磁珠等的磁性选择,使用对于CD4或CD8等的细胞表面标记的抗体、以及细胞分选仪的流式细胞法,使用抗CD3抗体、抗CD28抗体的活化T细胞诱导法等。另外,能够以细胞因子的分泌、功能性分子的表达、或者PD-1等信号分子为指标,单离期望的T细胞。另外,可以将颗粒酶、穿孔素等的分泌或产生作为指标,单离细胞毒性T细胞(CTL)。进而,在从包含具有抗原特异性的T细胞的人的组织进行单离的情况下,可以使用使结合了期望的抗原的MHC(主要组织相容基因复合体)多聚化而得的物质(例如,“MHC四聚体”、“Pro 5(注册商标)MHC I类五聚体”),从人的组织纯化具有期望的抗原特异性的T细胞。
本发明中,为了使T细胞成为iPS细胞而导入的基因优选为(a)Oct3/4基因、(b)c-Myc基因、(c)Sox2基因、(d)Klf4基因、(e)NANOG基因、和(f)LIN28基因等中至少4种的基因的组合。
本发明中,作为将上述基因簇导入T细胞的方法,没有特别限定,可以适当选择公知的手法来使用。例如,在以编码上述基因簇的核酸的形态导入到上述T细胞的情况下,将编码上述基因簇的核酸(例如,cDNA、RNA)插入包含在T细胞发挥功能的启动子的适当的表达载体中,将该表达载体以感染、脂质体转染法、脂质体法、电穿孔法、磷酸钙共沉淀法、DEAE葡聚糖法、显微注射法、电穿孔法而导入细胞。
在这样的表达载体中,考虑到癌症化的危险性的降低、导入效率的点,更优选使用包含上述基因簇的隐形RNA表达载体。
隐形RNA表达载体是指设计为避免载体进入染色体,且在细胞质内而非核内持续且稳定地表达基因的载体。具有隐形性,即,可以进行1万3000碱基对以上的大的基因的导入,也可以进行10个基因的同时导入,对细胞没有伤害,在不需要导入基因时能够除去,且细胞不会将载体识别为异物。
作为这样的隐形RNA表达载体,可举出由包含下述(1)~(8)的RNA序列的负单链RNA(A)、单链RNA结合蛋白质(B)、以及RNA依赖性RNA合成酶形成,且没有使自然免疫结构活化的复合体。
(1)相对于上述基因簇的RNA序列;
(2)构成非编码区域的来自人mRNA的RNA序列;
(3)上述RNA依赖性RNA合成酶识别的转录起始信号序列;
(4)上述RNA依赖性RNA合成酶识别的转录终止信号序列;
(5)上述RNA依赖性RNA合成酶识别的包含复制起点的RNA序列;
(6)编码上述RNA依赖性RNA合成酶的RNA序列;
(7)编码调节上述RNA依赖性RNA合成酶的活性的蛋白质的RNA序列;
(8)编码上述单链RNA结合蛋白质的RNA序列。
另外,在建立T-iPS细胞时,上述T细胞在上述基因簇的导入前,在白细胞介素-2(IL-2)或者白细胞介素-7(IL-7)和白细胞介素-15(IL-15)的存在下,优选通过抗CD3抗体和抗CD28抗体而刺激并活化,也可以通过选自植物血凝素(PHA)、白细胞介素-2(IL-2)、同种抗原表达细胞、抗CD3抗体、和抗CD28抗体、CD3和CD28激动剂中的至少1种物质而刺激并活化。该刺激例如可以在培养基中添加PHA、IL-2、抗CD3抗体和/或抗CD28抗体等,培养上述T细胞一定时间,从而进行。另外,抗CD3抗体和抗CD28抗体也可以与磁珠等结合,进而,代替在培养基中添加这些抗体,也可以在表面结合有抗CD3抗体和抗CD28抗体的培养皿上培养上述T细胞一定时间,从而给予刺激。进而,另外还可以通过将上述T细胞(例如,人T细胞)识别的抗原肽和饲养细胞一起添加到培养基中,从而给予刺激。
为了将该刺激给予上述T细胞,则培养基中添加的PHA的浓度,没有特别限定,优选为1~100μg/mL。另外,作为培养基中添加的IL-2的浓度,没有特别限定,优选为1~200ng/mL。
进而,作为培养基中添加的抗CD3抗体和抗CD28抗体的浓度,没有特别限定,优选为上述T细胞的培养量的1~10倍量。另外,为了将该刺激给予上述T细胞,作为结合于培养皿的表面上的抗CD3抗体和抗CD28抗体的浓度,没有特别限定,涂布时的浓度在抗CD3抗体的情况下为0.1~100μg/mL,优选为1~100μg/mL,在抗CD28抗体的情况下优选为0.1~10μg/mL。
另外,用于进行该刺激的培养期间只要是将该刺激给予上述T细胞的充分的期间、且是使T细胞增殖到上述4基因的导入所需的细胞数为止的期间,就没有特别限定,通常为2~7日,从基因导入效率的观点出发,优选为3~5日。通过在15mL管内使T细胞与载体混合从而感染、或者提高基因导入效率的观点出发,优选在涂布有纤维连接蛋白(RetroNectin)的培养皿上进行培养。
作为培养上述T细胞,添加PHA、IL-2、抗CD3抗体和/或抗CD28抗体等的培养基,例如可以使用适合上述T细胞的培养的公知的培养基(更具体来说,包含其它细胞因子类、人血清、罗斯威尔公园纪念研究所(RPMI)1640培养基、AIM VTM medium、NS-A2。培养基中,除了PHA、IL-2、抗CD3抗体和/或抗CD28抗体以外,还可以添加培养所必需的氨基酸(例如,L-谷氨酰胺)、抗生物质(例如,链霉素、青霉素)。另外,优选代替IL-2而在培养基中添加IL-7和IL-15。作为IL-7和IL-15的添加浓度,没有特别限定,分别优选为1~100ng/mL。
另外,作为在将上述4基因导入上述T细胞时或其后的条件,没有特别限定,导入上述4基因的上述T细胞优选在无饲养层条件下培养。例如,可举出作为层粘连蛋白511E8片段的iMatrix-511溶液、或者以玻连蛋白涂布的孔。也能建立在饲养细胞条件下的培养,作为饲养细胞,例如可举出通过放射线的照射、抗生物质处理而使细胞分裂停止的小鼠胎儿成纤维细胞(MEF)、STO细胞、SNL细胞。
进而,在由上述T细胞诱导为T-iPS细胞的过程中,优选从次日起添加iPS细胞的培养基。其后,优选每隔1天进行一半量的培养基交换,慢慢地从T细胞培养基置换为iPS培养基。
另外,优选:配合从上述T细胞向iPS细胞的转移,由适合上述T细胞的培养的公知的培养基慢慢地置换为适合iPS细胞的培养的培养基,并进行培养。作为适合该iPS细胞的培养的培养基,可以适当选择公知的培养基使用,例如在涂布iMatrix的情况下,在以StemFit AK03N或者玻连蛋白进行涂布的情况下,期望在Essential 8Medium、MEF细胞等的饲养细胞上含有敲除血清代替物、L-谷氨酰胺、非必需氨基酸、2-巯基乙醇、和b-FGF等的Dulbecco改良Eagle培养基/F12培养基(人iPS细胞培养基)。
如此,T-iPS细胞的选择可以通过适当选择公知的手法来进行。作为该公知的手法,例如,可举出在显微镜下观察ES细胞/iPS细胞样菌落的形态而进行选择的方法。另一方面,在由单细胞的CTL克隆建立的T-iPS的情况下,性质大多相似,因此,也有不选择T-iPS细胞的各菌落,而是将建立的菌落全部直接传代的方法。
如此选择的细胞为T-iPS细胞的确认例如可以通过如下方法而进行:通过免疫染色、RT-PCR等而检测选择的细胞中的未分化细胞特异性的标记(ALP、SSEA-4、Tra-1-60、和Tra-1-81等)的表达的方法;将选择的细胞移植到小鼠,观察其畸胎瘤形成的方法而进行。另外,如此选择的细胞来自上述T细胞的确认可以通过基因组PCR来检测TCR基因重组的状态,从而进行。
选择这些细胞并回收的时期优选为边观察菌落的生育状态边进行回收。一般来说,从将包含上述4基因的基因簇导入上述T细胞起为10~40日,优选为14日~28日。作为培养环境,只要没有特别说明,优选为:5%CO2、35~38℃的条件,更优选为5%CO2、37℃的条件。
为了由如上所述而得到的来自人T细胞的iPS细胞来制造表达CTL的抗原表位的HLA限制性的I类基因和HLA-E的HLA I类的T细胞,例如,可举出包含如下工序的方法:将来自人T细胞的iPS细胞的HLA I类全部敲除的工序(a),和将CTL的抗原表位的HLA限制性的I类基因和HLA-E的基因导入HLA I类被全部敲除的T―iPS细胞中的工序(b)。
上述敲除工序(a)和基因导入工序(b)可以用各种方法实施,CRISPR-Cas9基因组编辑法为其中一个选项。
首先,为了通过CRISPR-Cas9基因组编辑法而将来自人T细胞的iPS细胞的HLA I类全部敲除,首先敲除β2微球蛋白(B2M)。用敲除用质粒和引导RNA各5μg,对细胞剥离后的2×105左右的T-iPSC进行电穿孔。其后接种于6孔板的3孔,进行培养。敲除质粒中放入用于第2次的编辑的引导RNA的靶点序列、GFP以及CD8、CD19等选择标记。在约10日后,在接种于3孔的细胞汇合的时刻,进行选择标记的阳性选择。选择后,通过薄接种T-iPSC,对iPS细胞进行单细胞克隆。将GFP强阳性细胞提取后培养,进行基因分型。用PCR鉴定标记进入biallelic中的克隆后,扩大培养,进入下个步骤。
接下来,在利用CRISPR-Cas9基因组编辑法将HLA限制性的class 1基因和HLA-E的基因导入HLA I类被全部敲除的T-iPS细胞的工序(b)中,用2种敲入质粒分别各2.5μg和引导RNA5μg,对在最初的编辑中敲除了B2M的T-iPSC于细胞剥离后进行电穿孔。在电穿孔后,接种于6孔板的3孔,进行培养。在约7日后,在接种于3孔的细胞汇合的时刻,进行选择标记的阴性选择。选择后,通过薄接种T-iPS细胞,对iPS细胞进行单细胞克隆。将GFP阴性细胞提取后培养,进行基因分型。用PCR鉴定标记进入biallelic中的克隆后,扩大培养。另外,通过与上述同样的手段,除了CTL的抗原表位的HLA限制性的I类分子和HLA-E以外,进而也可以表达HLA-G、HLA-C等HLA I类,进而还可以表达CD47、PD-L1、iCaspase9等。
接下来,由基因组编辑后的T-iPS细胞分化诱导CTL细胞。
作为该再分化诱导方法,优选将T-iPS细胞分化为CD8+单阳性T细胞的方法,更优选将T-iPS细胞分化为CD4/CD8双阴性T细胞,接下来,使该CD4/CD8双阴性T细胞分化为CD8+单阳性T细胞的方法。
进而,如上述专利文献1所述,优选通过下述方法而得到:使T-iPS细胞分化为CD4/CD8双阴性细胞,添加刺激T细胞受体的物质,给予CD4/CD8双阴性细胞刺激,接下来,将给予T细胞受体刺激的上述CD4/CD8双阴性细胞于IL-7和IL-15的细胞因子的存在下分化为CD8单阳性T细胞。
为了使T-iPS细胞分化为CD4/CD8双阴性细胞,优选在含有细胞因子、血清(例如,胎牛血清(FBS))、胰岛素、转铁蛋白、亚硒酸钠、L-谷氨酰胺、α-单硫代甘油、抗坏血酸等的培养基中,在饲养细胞(优选为小鼠基质细胞)上培养T-iPS细胞。
作为使用的基质细胞,优选为实施了放射线照射等处理的OP9细胞、10T1/2细胞(C3H10T1/2细胞)。添加到培养基的细胞因子优选为选自VEGF、SCF、TPO、SCF和FLT3L中的至少1种的细胞因子,更优选为VEGF、SCF和TPO,或,VEGF、SCF和FLT3L。另外,作为培养基,例如可举出X-VIVO培养基、Iskoff改良Dulbecco培养基(IMDM培养基)、α-MEM、DMEM,但考虑到易于形成T-iPS sack(含有造血祖细胞的袋状的结构物)的点,优选为IMDM培养基。作为该T-iPS细胞的培养期间,优选为从T-iPS细胞的培养开始起8~14日、更优选为10~14日。作为培养环境,没有特别限定,优选为:5%CO2、35~38℃的条件,更优选为5%CO2、37℃的条件。另外,更优选在低氧浓度条件(氧浓度:例如,5~20%)下培养1周左右。
为了使T-iPS细胞分化为CD4/CD8双阴性细胞,进一步优选:将上述包含T-iPSsack的细胞在含有细胞因子、血清(例如,FBS)等的培养基中,在饲养细胞(优选为基质细胞、更优选为人基质细胞)上培养。存在于T-iPS sack的内部的细胞例如可以通过灭菌过的筛状器具(例如,细胞过滤器等),从而分离。作为用于该培养的基质细胞,从进行经由notch信号而向T淋巴细胞的分化诱导的观点出发,优选为实施了放射线照射等处理的OP9-DL1细胞、OP9-DL4细胞、10T1/2/DL4细胞、10T1/2/DL1细胞。作为添加到培养基中的细胞因子,例如可举出IL-7、FLT3L、VEGF、SCF、TPO、IL-2、和IL-15。作为培养基,例如可举出α-MEM培养基、DMEM培养基、IMDM培养基,优选为α-MEM培养基。另外,培养基中,除了IL-7和FLT3L以外,还可以添加培养所必需的氨基酸(例如,L-谷氨酰胺)、抗生物质(例如,链霉素、青霉素)。
作为含有该T-iPS sack的细胞的培养期间,优选为直到T细胞受体(TCR)在如此分化而得到的CD4/CD8双阴性细胞的细胞表面上表达为止的期间,优选从含有该T-iPS sack的细胞的培养开始起14~28日。作为培养环境,没有特别限定,优选为:5%CO2、35~38℃的条件,更优选为5%CO2、37℃的条件。
应予说明,T细胞受体(TCR)是否在CD4/CD8双阴性细胞的细胞表面上表达可以通过使用抗TCRαβ抗体、抗CD3抗体、抗CD4抗体和抗CD8抗体的流式细胞术来评价。
在具有抗原特异性的人CD8单阳性细胞的制造方法中,通过将来自T-iPS细胞的CD4/CD8双阴性细胞经由在该细胞表面上表达的TCR而进行刺激,从而可以抑制TCRA基因的进一步的重组,而且,在再分化而得到的CD8单阳性细胞中,可以使具有与原本的人T细胞相同的TCR基因的重组模式的T细胞的出現频率极高。
作为对来自T-iPS细胞的CD4/CD8双阴性细胞的T细胞受体给予刺激的方法,优选为使来自T-iPS细胞的CD4/CD8双阴性细胞与选自下述中至少1种物质接触的方法:与作为抗CD3抗体、抗CD28抗体、T-iPS细胞的来源的人T细胞特异性地结合的抗原肽,表达与对于上述T细胞受体显示限制性的HLA的复合体的细胞,以及,与该抗原肽结合的MHC多量体;从给予生理刺激的观点出发,更优选为使特异肽/HLA复合体表达细胞接触的方法。另外,从重视刺激的均匀性的观点出发,更优选为与抗体、试剂接触的方法。
接触的方法例如可以在培养基中添加PHA等,培养上述T细胞一定时间,从而进行。另外,抗CD3抗体和抗CD28抗体也可以与磁珠等结合,进而,代替在培养基中添加这些抗体,也可以在表面结合有抗CD3抗体和抗CD28抗体的培养皿上培养上述T细胞一定时间,从而给予刺激。进而,另外还可以通过将上述抗原肽和饲养细胞一起添加到培养基中,从而给予刺激。
为了刺激CD4/CD8双阴性细胞的TCR,作为培养基中添加的PHA的浓度,优选为1~100μg/ml。另外,作为培养基中添加的抗CD3抗体和抗CD28抗体的浓度,优选为上述T细胞的培养量的1~10倍量。另外,为了刺激CD4/CD8双阴性细胞的TCR,作为结合于培养皿的表面上的抗CD3抗体和抗CD28抗体的浓度,涂布时的浓度在抗CD3抗体的情况下为0.1~100μg/ml,在抗CD28抗体的情况下为0.1~10μg/ml。
作为含有该T-iPS sack的细胞的培养期间,优选包含T细胞受体(TCR)在如此分化而得到的CD4/CD8双阴性细胞的细胞表面上表达所必需的期间,优选从含有该T-iPS sack的细胞的培养开始起7~29日。作为培养环境,优选为:5%CO2、35~38℃的条件,更优选为5%CO2、37℃的条件。
本发明中,为了使给予T细胞受体刺激的CD4/CD8双阴性细胞分化为CD8单阳性细胞,CD4/CD8双阴性细胞优选在含有细胞因子、血清(例如,人血清)等的培养基中进行培养。作为添加到培养基的细胞因子,只要能使CD4/CD8双阴性细胞分化为CD8单阳性细胞即可,例如可举出IL-7、IL-15。在这些中,从向CD8单阳性细胞的分化中选择CD8谱系且易于产生记忆型CD8+T细胞生成的观点出发,优选组合IL-7和IL-15而添加。作为IL-7和IL-15的添加浓度,优选为1~20ng/ml。作为培养基,例如可举出RPMI-1640培养基、X-VIVO培养基、DMEM培养基、α-MEM培养基,优选为RPMI-1640培养基或X-VIVO培养基。另外,培养基中,除了IL-7、IL-15等以外,还可以添加培养所必需的氨基酸(例如,L-谷氨酰胺)、抗生物质(例如,链霉素、青霉素)、除IL-7、IL-15等以外的细胞因子。
该培养中,可以将CD4/CD8双阴性细胞与饲养细胞共培养。作为饲养细胞,优选为外周血单个核细胞(PBMC)。作为该PBMC,优选与CD4/CD8双阴性细胞为同种异体(allo)的关系。另外,从持续刺激TCR、且持续抑制TCR的进一步的重组的观点出发,更优选使用呈递与作为CD4/CD8双阴性细胞的来源的人T细胞特异性地结合的抗原肽的外周血单个核细胞。
作为用于使该CD4/CD8双阴性细胞分化为CD8单阳性细胞的培养期间,优选为2~4周。作为培养环境,优选为:5%CO2、35~38℃的条件,更优选为5%CO2、37℃的条件。
如此分化诱导的CD8单阳性细胞来自于T-iPS细胞,且来自于作为该T-iPS细胞的来源的T细胞的确认例如可以通过用基因组PCR检测TCR基因重组的状态,从而进行。
另外,如此得到的CD8单阳性细胞可以适当选择公知的手法而单离。作为该公知的手法,例如可举出使用对于CD8的细胞表面标记的抗体和细胞分选仪的流式细胞术。例如,在CD8单阳性细胞的情况下,可以采用如下方法:使用将作为CD8单阳性细胞的来源的T细胞识别的抗原固定化后的亲和柱等,进行纯化的方法;使用该抗原结合的MHC多量体(例如,MHC四聚体)而进行纯化的方法。
另外,由本发明而得到的CD8单阳性细胞不表达PD-1,与此相对,表达CCR7以及代表中央记忆T细胞的表型的CD27和CD28,并且,端粒也比作为来源的T细胞长,具有高的自我复制能力。因此,根据本发明,可以制造一种细胞,其为具有与原本的T细胞相同的TCR基因的重组模式的CD8单阳性T细胞,不表达PD-1,且表达CD27、CD28和CCR7。此外,从人采取T细胞在会表达PD-1且不成熟的记忆表型的比例少的点上,与得到的T细胞不同。
为了维持如此得到的CD8单阳性细胞,可以每1~2周对该细胞给予刺激。作为该刺激,可举出与选自抗CD3抗体、抗CD28抗体、IL-2、IL-7、IL-15、该CD8SP细胞识别的抗原、该抗原结合的MHC多量体、与该CD8单阳性细胞为同种异体关系的饲养细胞和与该CD8单阳性细胞为自体关系的饲养细胞中至少1种物质的接触。
如此得到的本发明的表达HLA-A24和HLA-E的HLA I类的、来自人T细胞来源的iPS细胞的细胞毒性T细胞能够维持用作原料的人T细胞的抗原特异性的细胞毒性,并能避免NK细胞的missing-self应答,作为能同种异体给药的来自人T细胞来源的iPS细胞的细胞毒性T细胞是有用的。本发明的细胞毒性T细胞的NK细胞的missing-self应答性与仅表达HLA-A24或HLA-E的T细胞相比显著降低。
实施例
以下,举出实施例对本发明进行进一步详细的说明,但本发明不限定于这些实施例。
实施例1
利用人乳头瘤病毒(HPV)特异性CTL克隆,建立使用仙台病毒载体的T-iPS细胞。
1)从健康人外周血分离外周血单个核细胞后,以抗原呈递为目的,诱导树突状细胞。7日后,向诱导的树突状细胞添加HPV抗原肽(HPV16-E6,A2402),开始与外周血单个核细胞共培养。约8~10日后,为了进行HPV特异性的CTL检测,在用MHC四聚体对CTL进行染色后,用流式细胞术确认四聚体阳性率。确认HPV特异性的CTL后,进行单细胞分选或者四聚体/PE珠选择后,进行有限稀释法,对50Gy的X射线照射后的PBMC施加IL2、PHA,进行刺激。
2)进行约3~6周后上升的菌落的四聚体染色,用流式细胞术进行CTL克隆建立的确认。在建立确认后,对CTL克隆进行CD3/28刺激,然后,使用以下的A)的2种载体,进行基因导入。将基因导入后的CTL移动到用iMatrix编码的6孔板,以CTL介质为培养基,在CO2培养箱中开始培养。
A)SeV4因子载体+SV40 large T抗原
3)在SeV基因导入次日,等量加入iPS介质(StemFitAK03N),其后每隔1天置换一半量的Stem FitAK03N。
4)7日后,可以观察T-iPS细胞的菌落,其后进行菌落提取。
实施例2
将来自HPV抗原特异性的CTL的iPS细胞的HLA I类全部敲除。
首先敲除β2微球蛋白(B2M)。用敲除用质粒和引导RNA各5μg,对细胞剥离后的T-iPSC使用LONZA 4-D Nucleofector,进行电穿孔。其后接种于6孔板的3孔,进行培养。由于第2次的编辑中使用的引导RNA的靶点序列与GFP和CD8的选择标记进入敲除质粒,因此,在约10日后,接种到3孔的细胞汇合的时刻,进行CD8的MACS磁珠阳性选择。选择后,通过薄接种T-iPSC,对iPS细胞进行单细胞克隆。提取GFP强阳性细胞后进行培养,进行基因分型,用PCR鉴定标记进入biallelic中的克隆后,扩大培养,进入下个步骤。
实施例3
导入HLA-A24和HLA-E的基因。
由于A2402限制性的表位,因此,将HLA-A2402敲入B2M敲除后的T-iPSC。并且,还同时制作HLA-E三聚体结构的敲入质粒,敲入HLA-E。进而,还制作同时敲入HLA-A2402和HLA-E各一半量的质粒。在细胞剥离后,使用LONZA 4-D Nucleofector,对在最初的编辑中敲除B2M的T-iPSC进行电穿孔。在电穿孔后,接种于6孔板的3孔,进行培养。在约7日后,在接种于3孔的细胞汇合的时刻,使用MACS磁珠,进行CD8的阴性选择。选择后,通过薄接种T-iPSC,对iPS细胞进行单细胞克隆。提取GFP阴性细胞后进行培养,进行基因分型,鉴定克隆后扩大培养。
实施例4
由基因组编辑后的T-iPS细胞再分化为CD8单阳性T细胞。
将实施例3中得到的基因组编辑后的T-iPS细胞的团块转移到C3H10T1/2细胞上,在20ng/mL VEGF、50ng/mL SCF和50ng/mL FLT-3L存在下,用EB培养基进行共培养。在培养第14日,回收包含T-iPS sack的造血细胞,将这些细胞转移到DL1/4表达C3H10T1/2细胞上的细胞上,在10ng/mL FLT-3L和1ng/mL IL-7存在下,于OP9培养基内,使造血细胞分化为T谱系细胞。
然后,在培养第42日,通过将α-CD3/CD28磁珠或5μg/ml PHA添加到上述OP9培养基,从而进行刺激。
接下来,回收该T谱系细胞,在10ng/mL IL-7和10ng/mL IL-15的存在下,与放射线照射过的PMBCs一起在CTL培养基中培养。
然后,在培养第56日,确认到CD8/四聚体阳性细胞。用FACS解析研究这些细胞,其结果是,敲入的HLA基因(HLA-A24、HLA-E、或者两者)得到表达。
实施例5
本发明的CD8单阳性细胞的抗原特异性。
对于实施例4中得到的再分化细胞,研究其在基因组编辑后,是否与作为来源的T细胞具有相同的抗原特异性。其结果是明确了:得到的再分化CD8单阳性细胞与作为来源的HPV―T细胞克隆的抗原特异性一致(图1)。
为了研究在仅表达HLA-A24、或者仅表达HLA-E、或表达这两者的恢复活力的CTL(rejuvenated CTL;rejT)的分化诱导成功后,是否能够避免NK细胞的missing-self反应,进行铬检测和CD107a检测。在仅表达HLA-A24、仅表达HLA-E的HPV-rejT中,抑制NK活性不充分,在表达这两者的HPV-rejT中,可以显著地抑制NK细胞的细胞毒性。可以证明,使CTL的抗原表位的HLA限制性HLA I类分子和HLA-E这两者表达对于抑制NK活性是重要的(图2、图3)。
实施例6
对于宫颈癌患癌小鼠的HLA编辑HPV-rejT的生存时间延长效果。
(方法)
对于免疫缺陷小鼠(NOG小鼠),将宫颈癌细胞株SiHa移植到腹腔内后,分为无治疗对照组和3个治疗组(原本的HPV-CTL克隆、Wild type(WT)HPV-rejT、或HLA编辑后的HPV-rejT),以每周1次、腹腔内注射2.5x106个的T细胞的方式,进行3次给药。以确认无治疗对照组与各治疗组的小鼠的治疗效果为目的,比较生存时间。
(结果)
宫颈癌患癌小鼠的生存时间示于图4。
如图4所示,与原始CTL给药组相比,宫颈癌患癌小鼠的生存时间在WT rejT给药组和HLA编辑的HPV-rejT(EXrejT)给药组中显著延长。
另外,在生存6个月以上的治疗后小鼠的病理观察结果中,在肺、肝脏、肠道、脾脏、子宫中未确认到肿瘤残存。
实施例7
HLA编辑HPV-rejT的NK细胞同时给药时的生物体内的耐久性。
(方法)
对于免疫缺陷小鼠(NOG小鼠)将宫颈癌细胞株SiHa在第4天移植到腹腔内后,分为3组。
分为1.HLA编辑FFluc-rejT+NK细胞(HLA-A24+)
2.HLA编辑FFluc-rejT+NK细胞(HLA-A24-)
3.HLA编辑FFluc-rejT,
在第0天,以腹腔内注射,向组1和2给药2.5x106个的rejT细胞+2.5×106个的NK细胞,向组3给药2.5x106个的rejT细胞。用IVIS监测各组的rejT的体内的增殖、残存,并比较。
(结果)
将结果示于图5。在第7天,在未给药NK细胞的组3中,荧光标记HPV-rejT在生物体内高效地增殖并残存,与此相对,在组2中,其被不具有HLA-A24的NK细胞排除,在第7天无法检测到rejT。另一方面,确认到:在组1中,荧光标记HPV-rejT没有被具有HLA-A24的NK细胞排除,在生物体内高效地增殖并残存(左图)。以信号确认到第7天的荧光标记HPV-rejT的小鼠生物体内的残存,其结果是,组1与组2相比较,信号显著提高。另一方面,与组3没有显著差异(右图)。
Claims (4)
1.一种来自人T细胞来源的iPS细胞的细胞毒性T细胞,表达CTL的抗原表位的HLA限制性的I类和HLA-E的HLAI类。
2.根据权利要求1所述的来自人T细胞来源的iPS细胞的细胞毒性T细胞,其中,CTL的抗原表位的HLA限制性的I类分子为HLA-A24或HLA-A02。
3.一种表达CTL的抗原表位的HLA限制性的I类和HLA-E这2种HLA I类的、来自人T细胞来源的iPS细胞的细胞毒性T细胞的制造方法,其特征在于,包括:将人T细胞来源的iPS细胞的HLA I类全部敲除的工序;向HLA I类被全部敲除的T-iPS细胞导入CTL的抗原表位的HLA限制性的HLA I类和HLA-E的基因的工序;以及将所述基因导入后的T-iPS细胞再分化成CD8单阳性T细胞的工序。
4.根据权利要求3所述的制造方法,其中,CTL的抗原表位的HLA限制性的I类分子为HLA-A24或HLA-A02。
