Abstract
本发明涉及生物医学技术领域。本发明提供了PRKD1基因作为非小细胞肺癌诊断或预后标志物的应用。本发明的实验证明,PRKD1在非小细胞肺癌中表达显著下调,提示可以将PRKD1应用于非小细胞肺癌的诊断或治疗;体外细胞实验及体内裸鼠实验证实PRKD1具有显著的抑制非小细胞肺癌细胞增殖、迁移和侵袭的能力,提示PRKD1可以作为药物靶标应用于非小细胞肺癌的临床治疗。本发明还公开了与PRKD1相互作用基因EIF5A,发现PRKD1与EIF5A呈负相关,提示PRKD1可通过调控EIF5A基因的表达水平,从达到控制非小细胞肺癌转移的目的,提供了一种新的治疗思路。
Description
技术领域
本发明涉及生物医学技术领域,尤其涉及PRKD1基因作为非小细胞肺癌诊断或预后标志物的应用。
背景技术
肺癌是最常见的恶性肿瘤,每年在全世界造成177万人死亡。肺癌分为两大类,非小细胞肺癌(Non-Small Cell Lung Cancer,NSCLC)和小细胞肺癌(SCLC)。其中NSCLC占所有肺癌病例的85%。随着外科手术、化疗和放疗的发展,以及广泛的禁烟努力,NSCLC的生存率不断提高,但死亡率仍然是所有恶性肿瘤中最高的。转移是影响NSCLC预后的重要因素,NSCLC转移是指肺原发性恶性肿瘤通过多种途径离开原发部位并向远处生长的过程。最常见的转移部位是脑、骨、淋巴结和肝脏。转移是一个极其复杂的过程,包括肿瘤微环境和干细胞的参与,以及上皮间质转化形成、血管生成和淋巴管生成等多种机制。这些因素与多种影响NSCLC转移的基因以及多种信号通路有关。对NSCLC转移机制的研究旨在寻找更有效的控制转移的方案,并为临床治疗提供指导。
发明内容
本发明的目的在于提供PRKD1基因作为非小细胞肺癌诊断或预后标志物的应用,寻找更有效的控制转移的方案,并为临床治疗提供指导。
为了实现上述发明目的,本发明提供以下技术方案:
本发明提供了PRKD1基因作为非小细胞肺癌诊断或预后标志物的应用。
本发明还提供了PRKD1基因在制备非小细胞肺癌诊断/治疗的产品中的应用。
本发明还提供了PRKD1基因在调控EIF5A基因的表达中的应用。
本发明提供了PRKD1基因作为非小细胞肺癌诊断或预后标志物的应用。本发明的实验证明,PRKD1在非小细胞肺癌中表达显著下调,提示可以将PRKD1应用于非小细胞肺癌的诊断或治疗;体外细胞实验及体内裸鼠实验证实PRKD1具有显著的抑制非小细胞肺癌细胞增殖、迁移和侵袭的能力,提示PRKD1可以作为药物靶标应用于非小细胞肺癌的临床治疗。本发明还公开了与PRKD1相互作用基因EIF5A,发现PRKD1与EIF5A呈负相关,提示PRKD1可通过调控EIF5A基因的表达水平,从达到控制非小细胞肺癌转移的目的,提供了一种新的治疗思路。

具体实施方式
下面结合实施例对本发明提供的技术方案进行详细的说明,但是不能把它们理解为对本发明保护范围的限定。
实验细胞
本研究使用了1种肺上皮细胞系(BEAS-2B)和4种NSCLC细胞系(A549、H1299、PC9、H1975)。A549、H1299和H1975培养在1640培养基,PC9和BEAS-2B培养在DMEM培养基。
实验动物
本研究中的动物实验得到了山东大学齐鲁医院动物伦理委员会的批准和监督,本课题所用的动物为4-5周龄左右、体重为20-22g的雄性BLAB/c裸鼠,购自北京维通利华技术有限公司。裸鼠饲养在山东大学齐鲁医院实验动物管理中心内,所有动物实验操作均遵守《实验动物管理条例》。
实施例1细胞培养
1)当细胞长满至培养皿80%左右时应进行传代,否则由于细胞过多过密会发生凋亡。
2)吸弃培养皿中旧培养基,加入预冷的1×PBS,轻晃培养皿,吸弃PBS,重复此步骤三次。
3)再向培养皿中加入1ml 0.25%胰酶溶液消化细胞,将培养皿放入37℃细胞培养箱中孵育2min。
4)培养皿置于显微镜下观察细胞形态,若细胞变成圆形,并且漂动时,即终止消化;若大部分细胞还为贴壁状态,则可继续放入细胞培养箱中孵育,或者增加胰酶的用量。
5)向培养皿中加入1ml完全培养基,终止消化,吹打培养皿壁,并将细胞悬浮液转移至15ml离心管中。
6)离心机转速设置为1000rpm,离心5min,吸弃上清液。
7)加入完全培养基,吹打混匀,形成细胞悬液。
8)将细胞悬液分至3个培养皿中,每个培养皿中再加入完全培养基。
9)将培养皿放入5%CO2、37℃恒温培养箱中,按时观察、换液。
实施例2转细胞株的筛选
1)在6孔板中接种细胞,待细胞密度达60%时进行转染操作。
2)弃旧培养基,每个孔加入新鲜的1ml完全培养基。按照慢病毒使用说明书,计算每个孔的细胞中所需要加入的病毒量。
3)12h后每个孔补充1ml完全培养基。48h后细胞培养基换成含有2mg/ml嘌呤霉素的完全培养基。后续根据细胞密度进行换液传代处理。
实施例3qRT-PCR
1)将稳转细胞接种于6孔板中,待其长满后,利用TRIzol法提取总RNA。
2)用反转录试剂盒将RNA反转录成cDNA。
3)利用荧光定量PCR仪进行扩增。反应条件为:95℃预变性30s;95℃变性5s,60℃退火30s,共40个循环。
4)以β-actin作为内参,对qRT-PCR结果进行定量分析。实验重复3次,取均值。
实施例4Western blot
1)细胞蛋白提取试剂盒提取总蛋白,BCA法进行总蛋白浓度检测。
2)制胶,每孔加入相应的蛋白样品,80V恒压电泳30min后,100V恒压电泳1.5h。
3)待电泳结束后,湿转法将蛋白电转移到PVDF膜上。
4)5%脱脂奶粉封闭1h后,加入相应的一抗,4℃孵育过夜。
5)第二天,二抗室温孵育1h,利用成像系统成像以及image J分析实验结果,以β-actin为内参,分析目的蛋白表达水平。实验重复3次,取均值。
实施例5CCK8实验
1)取对数生长期细胞接种到96孔细胞培养板中,调整细胞密度为2000个细胞每孔。
2)每孔中加入10μl的CCK8溶液后37℃孵育1h,酶标仪检测450nm波长处每孔的吸光度值(OD值)。实验重复3次,取均值。
实施例6EdU实验
1)取对数生长的细胞,将细胞消化,用PBS洗两次,计数。
2)100ul的细胞悬液加至96孔,20000个细胞/孔,3个复孔,培养过夜;后续按照EdU说明书操作。
3)倒置荧光显微镜镜检,拍照,计数。
实施例7划痕实验
1)在6孔板中接种细胞,待细胞密度达90%时进行划痕操作。
2)用枪头比着直尺,尽量垂至于背后的横线划痕。
3)PBS洗细胞2次,去除划下的细胞,加入无血清培养基。放入37℃,5%CO2培养箱培养。在0和24小时拍照。
实施例8transwell小室实验
1)细胞消化离心,用含1%FBS的培养基重悬细胞,每个小室的上室取100ul细胞悬液,加入3x104个细胞。
2)下室预先加入600μL含有20%FBS的培养基。
3)37℃、5%CO2培养箱培养24h。
4)取出小室,甲醇固定15min;PBS洗2遍,结晶紫染色30min,显微镜下观察计数。
实施例9裸鼠皮下瘤模型
1)培养A549-LV-PRKD1和对照组LV-NC细胞使之处于对数生长期,消化重悬调整至细胞密度为1×108个/ml。
2)将裸鼠随机分成两组,每组5只裸鼠,将上述细胞100ul注射至每只裸鼠的腋窝,建立皮下成瘤模型。
3)实验过程中密切关注动物状态,待瘤体出现后用游标卡尺每5天测量一次,测量指标为瘤体的长径(瘤体最长的直径)和短径(垂直于长泾的直径),根据公式计算瘤体体积V=(长径x短径2)/2,并绘制瘤体的生长曲线。
4)37天后,终止实验,裸鼠以安乐死。剥离皮下瘤并拍照。
5)瘤体称重,标本保存在4%的多聚甲醛中、石蜡包埋后用于常规制作切片,进行HE染色和IHC染色。
实施例10裸鼠转移模型
1)培养A549-LV-PRKD1和对照组LV-NC细胞使之处于对数生长期,消化重悬调整至细胞密度为108个/ml。
2)将裸鼠随机分成两组,每组5只裸鼠,用酒精棉球对注射区域的尾部进行消毒,将上述细胞混匀后100ul注射至每只裸鼠的尾静脉,建立转移瘤模型。
3)每天观察裸鼠状态,8周后终止实验,裸鼠以安乐死。剥离肺并拍照,统计肺部转移灶的个数。将肺部标本保存在4%的多聚甲醛中,用于后续染色。
实施例11HE染色
1)取材固定:取小鼠体内成瘤组织或肺部组织块放入预先配好的固定液(4%多聚甲醛)中
2)脱水透明:依次经过不同浓度的乙醇溶液各脱水1次,二甲苯透明2次,每次5min。
3)浸蜡包埋:将石蜡高温熔化后,将上述取得的组织块缓慢放于己溶化的石蜡中。观察石蜡完全没入组织块后进行包埋。冷却凝固成块即成。
4)切片:将包埋好的蜡块放置于切片机上,切成薄片,放到37℃恒温水浴锅中,用载玻片抄起薄片,使薄片均匀的覆盖在载玻片上。
5)烤片:将含有薄片的切片放入60℃恒温箱中烘干。
6)苏木素染色:配置好苏木素染液,将切片浸入其中3min,自来水洗。
7)分化返蓝:配置好苏木素分化液,将切片浸入其中3s,配置好苏木素返蓝液,自来水冲洗,将切片浸入其中3s,自来水冲洗,镜检细胞核呈淡蓝色,组织背景近乎无色。
8)伊红染色:伊红染液5min。
9)脱水透明:依次经过不同浓度的乙醇溶液脱水1次,二甲苯透明2次,每次5min。
10)使用中性树胶封片,晾干后观察。
实施例12IHC染色
1)烤片:将小鼠体内成瘤组织切片放入60℃恒温箱中烘干。
2)脱蜡水化:使用二甲苯浸泡2次,每次5min,依次经过不同浓度的乙醇溶液脱水1次。
3)抗原修复:在修复盒中加入柠檬酸抗原修复缓冲液,将组织切片置于其中,于微波炉内进行加热,中火8min至沸腾,停火8min保温再转中低火7min。自然冷却后将组织切片用PBS冲洗,在脱色摇床上晃动洗涤3次,每次5min。
4)阻断内源性过氧化物酶:3%双氧水溶液没过切片,室温避光孵育25min,将玻片置于PBS中洗,每次5min。
5)血清封闭:用组化笔画出一定的区域,在区域内滴加3%BSA均匀覆盖组织,室温封闭30min。
6)加一抗:弃掉封闭液,在切片上滴加PBS按一定比例配好的一抗,切片平放于湿盒内4℃孵育过夜。
7)加二抗:将玻片置于PBS中洗3次,每次5min。切片稍甩干后在圈内滴加二抗覆盖组织,室温孵育50min。
8)DAB显色:将玻片置于PBS中洗3次,每次5min。切片稍甩干后在圈内滴加新鲜配制的DAB显色液,显微镜下控制显色时间,阳性为棕黄色,自来水冲洗切片终止显色。
9)苏木素染色:将切片浸入苏木素染液中3min,,自来水洗。
10)分化返蓝:苏木素分化液3s,自来水洗,苏木素返蓝液返蓝3s,自来水冲洗,镜检细胞核呈淡蓝色,组织背景近乎无色。
11)伊红染色:伊红染液5min。
12)脱水透明:依次经过乙醇溶液脱水1次,二甲苯浸泡2次,每次5min。
13)使用中性树胶和盖玻片封片,晾干后观察。
实施例13免疫共沉淀
1)抗原样品制备:弃培养基,1xPBS洗涤细胞2次。用刮棒刮脱细胞,收集至1.5mlEP管内,加入结合缓冲液和蛋白酶抑制剂,混匀后置于冰上10min。14000g,4℃离心10min,置于冰上备用。
2)磁珠预处理:取30ul磁珠悬液放入1.5ml EP管中,200ul结合缓冲液通过磁性分离方式洗涤,重复2次,加入200ul结合缓冲液重悬磁珠备用。
3)抗体结合反应:首先结合缓冲液稀释抗体至30ug/ml,步骤(2)中的磁珠加入200ul稀释后的抗体,然后置于翻转混合仪,15min后磁性分离,弃上清。加入200ul结合缓冲液,轻轻吹打使磁珠-抗体复合物均匀分散,磁性分离后弃上清。再加入结合缓冲液,重复上述步骤2次。
4)抗原沉淀反应:加入步骤(1)的样品,轻轻吹打使抗原与磁珠-抗体复合物均匀分散,置于4℃翻转混合仪过夜。第二天磁性分离后弃上清,加入200ul洗涤缓冲液,轻轻吹打使磁珠-抗体-抗原复合物均匀分散,磁性分离后弃上清,重复2次。加入200ul洗涤缓冲液,将复合物悬液转移至新的1.5mlEP管中,磁性分离后弃上清。
5)抗原洗脱:加入35ul 1x loading buffer混合均匀,95℃加热5min,13000g,室温离心10min,收集上清进行SDS-PAGE检测,或加入20ul洗脱缓冲液混合均匀,室温孵育10min,13000g,4℃离心10min,收集上清至新的EP管,立即加入1ul中和缓冲液,进行质谱检测。
实施例14统计学方法
采用GraphPad Prism 8软件进行统计学处理。计量数据以均数±标准差表示,实验结果采用t检验进行分析。以P<0.05为差异有统计学意义。
结论:
TCGA数据库差异表达分析显示,PRKD1在NSCLC组织中的表达远低于正常肺组织(P<0.001,图1A)。在GSE10072数据集中,与49例正常肺组织相比,PRKD1在58例NSCLC组织中的表达显著下调(P<0.0001,图1B)。GSE19804数据集中60例肺癌和癌旁组织配对分析显示,PRKD1在肺癌组织中明显低表达(P=0.004,图1C);GSE32863数据集中58例NSCLC组织的PRKD1表达水平明显低于58例癌旁组织(P<0.0001,图1D)。在GEPIA数据库483例LUAD组织和486例LUSC组织中,PRKD1的表达均显著低于正常肺组织(P<0.05,图1E)。以上结果表明,PRKD1在NSCLC组织中低表达。
将TCGA数据库中具有明确生存时间的948例NSCLC样本,根据PRKD1中位表达水平,分为PRKD1高表达组(n=474)和PRKD1低表达组(n=474),生存分析表明PRKD1低表达组患者预后更差(P=0.033,图2A)。将TCGA数据库中具有病理分期的966例NSCLC样本(其中I期501例,II期271例,III期162例,IV期32例)进行分组,由于IV期样本较少,分为I-II期和III-IV期两个亚组进行表达差异分析,结果显示PRKD1在III-IV期患者中表达更低(P=0.044,图2B)。在Kaplan-MeierPlotter网站分析PRKD1表达与NSCLC患者预后的关系,发现低表达PRKD1的NSCLC患者有较短的总生存期(Overall Survival,OS)和首次进展生存期(First-Progression Survival,FP),差异具有统计学意义(图2C-D)。
过表达慢病毒LV-PRKD1(LV-NC作为对照)及敲低慢病毒shPRKD1(shNC作为对照)感染A549和H1299细胞后,使用嘌呤霉素筛选出稳定转染的细胞株。应用qRT-PCR和Westernblot检测细胞中PRKD1的过表达和敲低效率,发现转染了LV-PRKD1的细胞中PRKD1表达水平较LV-NC组明显升高(图3A-D),而转染了shPRKD1的细胞中PRKD1表达水平较shNC明显下降(图3E-H),说明PRKD1过表达及敲低的稳转细胞株构建成功。
CCK8和EDU增殖实验可以用于探索PRKD1对NSCLC细胞增殖能力的影响。CCK8检测结果显示,过表达PRKD1能够明显抑制A549和H1299细胞的增殖(图4A-B),而敲低PRKD1的表达则显著促进细胞的增殖(图4C-D)。EDU增殖实验同样显示,PRKD1-OE组细胞增殖率明显低于vector对照组(图5A-B),而siPRKD1组细胞增殖率显著高于siNC组(图5C-D),即PRKD1过表达后细胞增殖能力降低。
划痕实验表明,PRKD1过表达显著抑制A549和H1299细胞的划痕愈合能力(图6A-B),而敲低PRKD1表达则促进了A549和H1299细胞的划痕愈合能力(图6C-D)。Transwell迁移和侵袭实验中,PRKD1-OE组细胞穿过Transwell小室和Matrigel胶的数目明显少于vector对照组(图7A-B),而siPRKD1组细胞穿过Transwell小室和Matrigel胶的数目明显多于siNC对照组(图7C-D)。综上所述,PRKD1能抑制NSCLC细胞的迁移和侵袭。
在裸鼠皮下瘤实验中,肉眼可见(图8A、8D)PRKD1过表达组(LV-PRKD1)皮下瘤明显小于空载组(LV-NC),PRKD1敲低组(shPRKD1)皮下瘤明显大于对照组(shNC)。在肿瘤生长过程中,每5天测量一次瘤体的长径和短经,计算肿瘤体积并分别绘制生长曲线,结果如图8B和图8E所示,LV-PRKD1组裸鼠皮下瘤生长速度明显慢于LV-NC组,shPRKD1组裸鼠皮下瘤生长速度明显快于shNC组。对取下的瘤体进行称重,统计分析显示LV-PRKD1组皮下瘤重量明显轻于LV-NC组(0.6916±0.2504g vs0.1281±0.044g,P<0.01,图8C),shPRKD1组皮下瘤重量明显重于shNC组(0.2542±0.0237g vs 0.4249±0.0954g,P<0.05,图8F)。在尾静脉转移模型中,8周左右处死裸鼠取出肺脏,可见LV-NC组与LV-PRKD1组均有肺转移灶形成(图9A)。对两组肺脏进行HE染色及转移灶数目统计,结果表明与LV-NC组相比,LV-PRKD1组肺部转移灶数目明显较少(图9B、C)。综上所述,在A549细胞中过表达PRKD1不仅能抑制NSCLC生长,还能抑制NSCLC转移。
两组质谱检测取交集后,按肽段覆盖率(鉴定到的氨基酸数目占蛋白质总氨基酸数目的比例)排序,得到可能与PRKDI相结合的基因(图10A-B)。取肽段覆盖率最高的基因EIF5A进行免疫共沉淀实验,发现与anti-lgG抗体相比,anti-Flag抗体可以显著富集到EIF5A,即EIF5A可与PRKD1相结合(图10C)。TCGA和GEO(GSE19804、GSE42127、GSE32863)数据库进一步验证了NSCLC中PRKD1和EIF5A表达的相关性,结果提示PRKD1与EIF5A显著负相关(图10D-G)。
在包含33种肿瘤的泛癌研究中,与正常组织相比,EIF5A在近20种恶性肿瘤中均呈现高表达(图11A)。TCGA、GEO数据库(GSE10072、GSE19804、GSE32863)分析显示:相较于正常肺组织,EIF5A在NSCLC组织中的表达更高(图11B-E)。根据EIF5A中位表达值,将NSCLC患者分为高低表达两组,无论是在TCGA还是GEPIA数据库,我们均发现EIF5A低表达组患者的总体生存率明显优于EIF5A高表达组患者(图12A,TCGA:P=0.007;图12C,GEPIA:P=0.0013)。根据患者临床分期,我们将TCGA中NSCLC患者分为stage I-II期和stage III-IV期两个亚组,发现与stage I-II期患者相比,EIF5A分子在stage III-IV期患者中表达量更高,结果具有统计学差异(图12B,P=0.0023)。Kaplan-MeierPlotter网站分析发现EIF5A高表达患者的OS(图12D)和FP(图12E)均低于EIF5A低表达患者。
qRT-PCR结果显示,EIF5A在4种NSCLC细胞(H1299、A549、H1975和PC9)中的表达量高于肺正常上皮细胞BEAS-2B,且在A549细胞中表达量最高(图13A)。在A549细胞中转染EIF5A siRNA,48h后qRT-PCR和显示,与阴性对照(siNC)相比,siRNA能显著下调EIF5A的mRNA表达(图13B)。CCK8和EDU细胞增殖实验表明,敲低EIF5A表达后细胞的增殖能力被明显抑制(图14A-B)。划痕和Transwell实验表明,EIF5A表达下调后,穿膜细胞数明显减少,即细胞迁移和侵袭能力下降(图15A-B)。
在稳定敲低PRKD1(shPRKD1)的A549细胞中应用siRNA敲低EIF5A的表达,实验分成三组:shNC组,shPRKD1组和shPRKD1+siEIF5A组。CCK8和EDU增殖实验表明,稳定敲低PRKD1(shPRKD1)促进A549细胞增殖,在此基础上低表达EIF5A能削弱shPRKD1对A549细胞增殖的促进作用(图16A-B)。划痕和Transwell实验结果表明,稳定敲低PRKD1(shPRKD1)促进A549细胞迁移和侵袭,在此基础上敲低EIF5A能削弱shPRKD1对A549细胞迁移侵袭的促进作用(图17A-B)。
由以上实施例可知,本发明提供了PRKD1基因作为非小细胞肺癌诊断或预后标志物的应用。本发明的实验证明,PRKD1在NSCLC中表达显著下调,提示可以将PRKD1应用于NSCLC的诊断或治疗;体外细胞实验及体内裸鼠实验证实PRKD1具有显著的抑制NSCLC细胞增殖、迁移和侵袭的能力,提示PRKD1可以作为药物靶标应用于NSCLC的临床治疗。本发明还公开了与PRKD1相互作用基因EIF5A,发现PRKD1与EIF5A呈负相关,提示PRKD1可通过调控EIF5A基因的表达水平,从达到控制NSCLC转移的目的,提供了一种新的治疗思路。
