Description
Technical Field
The invention relates to the field of rapid propagation of ornamental peony, in particular to a method for promoting the root section cutting of the ornamental peony to generate adventitious buds, and belongs to the field of vegetative propagation of the ornamental peony.
Background
Paeonia lactiflora Pall is a perennial root herbaceous flower of Paeoniaceae and Paeonia, has large and bright flower and rich flower types, and is highly favored by people. Because the seed propagation can not stably keep the good characters of the female parent and the breeding period is long, the new variety of the Chinese herbaceous peony is mainly propagated by an asexual propagation method.
In the asexual propagation method, the plantlet is the most common one in the production of the Chinese herbaceous peony, and the survival rate is higher, but the method has the defects of low propagation coefficient and long period, and is difficult to meet the requirement of expanding propagation of new Chinese herbaceous peony varieties in production. For peony cutting propagation, peony stem segments are generally adopted as cutting slips, and the stem segments need to be collected in the vigorous growth period of the peony, so that the growth and development of plants are seriously influenced (Yiweiling, research on peony stem cutting propagation technology and rooting mechanism, Shandong university of agriculture, 2009; patent CN103493677B of the invention).
In addition, the peony root segment can be used as a cutting shoot, and the cutting shoot not only has high propagation coefficient, but also has small influence on the growth and development of plants. The invention patent CN201910148432.X discloses a peony root section cuttage rapid propagation method, which adopts ABT rooting powder to treat peony root sections. The main components of the ABT rooting powder are potassium indolebutyrate and sodium naphthaleneacetate, which belong to auxin, and the influence of other hormones on the adventitious bud generated by the cuttage of the peony root segment is rarely reported.
Disclosure of Invention
The purpose of the invention is as follows: the technical problem to be solved by the invention is to provide the method for promoting the generation of the adventitious bud by applying 6-BA in the cuttage process of the peony root segment, the method is simple, convenient and easy to implement, short in period and high in adventitious bud generation rate, and can be applied to the cuttage of the peony root segment.
The technical scheme is as follows: in order to solve the technical problems, the invention provides a method for promoting ornamental peony root segment cuttage to generate adventitious buds, which comprises the following steps:
1) root segment selection: selecting radix Paeoniae root segments with substantially consistent growth conditions, and selecting root segments with thickness of 1-2cm and length of 15-20 cm;
2) disinfecting the root section; disinfecting the root segments for 5-15 s by using a carbendazim solution;
3) cutting root sections; cutting the root segment into upper, middle and lower root segments from the morphological upper end to the lower end, wherein the upper part of each root segment is cut flatly, the lower part is cut obliquely into a horseshoe shape, and the length of each root segment is 5-7 cm;
4) root segment soaking: soaking the root segments in 30-60 mg/L6-BA for 1.5-2 h;
5) root section cuttage: cutting the root segment in the wet river sand, wherein the relative humidity of the river sand is kept to be 60% -80% and the covering thickness is 2-4 cm;
6) and after the cuttage is finished, spraying management is carried out by adopting an automatic spraying device.
Wherein the peony variety in the step 1) is ornamental peony variety 'Hangzhou peony'.
Wherein, the selection time of the root section in the step 1) is from 10 middle to last.
Wherein the concentration of the carbendazim solution in the step 2) is 0.1%.
Wherein the carbendazim soaking time in the step 2) is 10 s.
Wherein the concentration of 6-BA in the step 4) is 40 mg/L.
Wherein, the river sand is disinfected with 0.1 percent carbendazim solution overnight before cuttage in the step 5), and is dried in the air the next day.
Wherein, the spraying management time of the step 6) is set to be 1d twice, each time is 20s, after 1 month, the spraying management time is set to be 1 time every day, each time is 20s, and 0.1 percent carbendazim solution is sprayed every 7 d.
Has the advantages that: compared with the prior art, the invention has the following advantages: the method has the advantages of simple operation, low technical requirement, easy popularization and utilization, and the like, and can be applied to the production of the Chinese herbaceous peony. The method has remarkable effect and high adventitious bud production rate, and particularly the adventitious bud production rate of the upper root segment can exceed 90%.
Drawings
FIG. 1 shows the condition of adventitious buds generated in the middle root section of Paeonia lactiflora under distilled water treatment;
FIG. 2 shows the adventitious bud formation in the middle root section of Paeonia lactiflora under distilled water treatment;
FIG. 3 shows the growth of adventitious buds from the middle root segment of peony treated with 40 mg/L6-BA.
The specific implementation mode is as follows:
the technical solutions of the present invention are further described in detail by the following specific examples, but it should be noted that the following examples are only used for describing the content of the present invention and should not be construed as limiting the scope of the present invention.
Example 1:
the research is carried out by taking ornamental peony variety 'Hangzhou peony' planted in the national peony germplasm resource library of Yangzhou university as a material.
6-BA: purchased from Solebao science and technology Co., Ltd. (Beijing), IBA was dissolved in 0.1mol/L NaOH solution at the time of use, and finally prepared into 40mg/L with distilled water, and all the solutions were ready to use.
Carbendazim: the carbendazim reagent used in the invention is purchased from national photo-agriculture chemical Limited liability company (Sichuan), and when in use, the carbendazim is prepared into 0.1 percent by using distilled water, and all solutions are prepared for use.
River sand is sterilized with 0.1% carbendazim solution overnight before cuttage, and is used after being dried in the air the next day.
Selecting 100 peony root segments with basically consistent growth conditions in 10 th ten days, and selecting root segments with the thickness of 1-2cm and the length of 15-20 cm.
After sterilizing with 0.1% carbendazim solution for 10s, the root segments are cut into upper, middle and lower root segments from the morphological upper end to the morphological lower end, the upper part of each root segment is cut flatly, the lower part is cut obliquely into a horseshoe shape, and the length of each root segment is 5-7 cm.
Soaking the root segment in 40mg/L6-BA for 2h, and treating the root segment with distilled water as control.
Cutting in shade, cutting radix Paeoniae root segment in sterilized river sand, and covering 3cm river sand above the root segment.
After the cuttage is finished, an automatic spraying device is adopted, the time is set to be 1d twice, 20s each time, after 1 month, the time is set to be 1 time every day, 20s each time, and 0.1% carbendazim solution is sprayed every 7 d.
And (3) observing the growth condition of the adventitious buds: at the beginning of 4 months in the next year, the whole root segment is taken out for observation. The middle root section of the peony treated by the distilled water can not generate adventitious buds (figure 1), and as can be seen from figure 2, the 40mg/L6-BA treatment can effectively promote the root section to generate the adventitious buds by cutting.
