Abstract
一种抗菌纳米颗粒、溶致液晶前体溶液喷雾敷料及其制备方法,所述抗菌纳米颗粒由树枝状抗菌肽和金纳米粒反应得到;所述树枝状抗菌肽为用树枝状聚酰胺‑胺支链化的聚赖氨酸,其具有如下式(I)所示结构,其中,n为6‑20,m为4‑8。该溶致液晶前体溶液喷雾敷料由包括如下组分的原料制备而成:抗菌活性成分、溶致液晶基质材料、有机溶剂、晶格调节剂、水;所述抗菌活性成分为所述的树枝状抗菌肽和/或抗菌纳米颗粒。该抗菌纳米颗粒可有效突破生物被膜渗透屏障,抗菌效果优异;溶致液晶前体溶液喷雾敷料在为伤口提供有效屏障的同时,可以实现局部药物高浓度,杀灭致病菌,最终促进感染伤口快速愈合。

技术领域
本发明涉及药物制剂领域,特别是涉及一种抗菌纳米颗粒、溶致液晶前体溶液喷雾敷料及其制备方法。
背景技术
伤口愈合是一个复杂且受高度调控的过程,通常分为四个阶段:止血、炎症、增殖和重塑。一部分伤口在炎症阶段后进展缓慢,未能在“合理”的时间范围内愈合,被归类为慢性伤口。慢性伤口内的感染是一个复杂因素,不仅会导致患者痛苦增加,而且随着全球人口老龄化,久坐不动以及肥胖和糖尿病等并发症增加,慢性伤口也随之增加。
感染是伤口延迟愈合的主要原因,60%的慢性伤口感染由生物被膜细菌造成。由于皮肤上存在复杂的微生物群落,细菌微生物侵入皮肤伤口,定植于皮肤组织引发感染,细菌定植并且持续存在是引发感染重要因素。慢性伤口内的细菌微生物群落以生物被膜的形式存在。
自青霉素等抗生素被成功纯化,抗生素已经成为细菌感染相关疾病的首要治疗方法。临床常用抗生素包括β-内酰胺类、磺胺类、喹诺酮类、氨基糖苷类及大环内酯类等,其主要通过与细菌胞内特异性靶点相互作用,阻碍或破坏细菌内部关键细胞功能(如抑制核酸复制、转录,抑制蛋白质合成,降低胞内酶活性等)而实现抗菌效果。
然而,随着抗生素在医学和农业方面的滥用、泛用,致病菌产生灭活酶、主动外排系统过度表达、膜靶点蛋白改变、16s核糖体RNA甲基化等改变而产生耐药性,导致抗生素疗效骤降。例如,几乎所有可用的β-内酰胺类药物对耐甲氧西林金黄葡萄球菌(Methicillin-resistant Staphylococcus aureus,MRSA)均无效。欧洲每年死于多重耐药菌导致的感染性疾病的患者超过25000人,细菌耐药性已然成为感染性疾病治疗的最大威胁,预计到2050年,全球将会有超过1000万人死于耐药菌相关感染性疾病,累计将会造成超过100万亿美元的损失。据中国耐药细菌监测网报道,2019年临床检出的所有SA菌株中,MRSA占比已高达30.2%,其中江苏省的MRSA检出率已然高至45.5%。因此研发针对非特异性靶点的新型抗菌药物以杀灭耐药致病菌是治疗慢性伤口感染的先决条件。
此外,致病菌可附着在慢性伤口表面,大量繁殖,集聚形成三维、多细胞社群,形成生物被膜。生物被膜是指微菌落嵌入自身分泌的胞外聚合物(Extracellular PolymericSubstances,EPS)内形成的三维结构聚膜样物。它在起始粘附聚集、生物被膜成熟以及细菌脱落释放三个阶段中循环往复(如图1所示),其对细菌的庇护作用表现在低药物可及性与高细菌耐药性两个方面。一方面,EPS形成的致密天然屏障,产生渗透障碍,严重降低了抗菌药物对膜内致病菌的可及性。同时生物被膜内高密度分布的微菌落之间存在的丰富的菌间物质信息交流,提高了致病菌对抗菌药物的耐药性,引发顽固性、复发性和持续性感染。因而杀灭耐药致病菌的难点在于有效突破生物被膜。
因此,针对非特异性靶点杀灭致病菌、有效攻克生物被膜病理屏障的新型抗菌药物研发已然成为杀灭耐药致病菌、治疗慢性伤口感染的重点。
针对慢性伤口部位的皮肤缺损,覆盖伤口暂时维持皮肤屏障保护作用,防止伤口部位水分、蛋白质大量流失,隔离环境致病菌,同时缓冲意外外力冲击,为伤口愈合创造适宜微环境是促进慢性伤口愈合的必要条件。近年来研究人员认为类生理状态的湿性环境有助于伤口愈合,其原理主要包括:1)保持伤口处的温湿度,保留创面渗液释放,激活多种酶以及酶的活化因子(如蛋白酶和尿激酶等),从而促进坏死组织和纤维蛋白的溶解和吸收,避免干痂形成;2)避免神经末梢在空气中暴露,减轻疼痛;3)加速表皮细胞移行,迅速缩小创面,促进肉芽组织新生,促进创面再上皮化;4)增强白细胞生理功能,从而提高伤口抗感染能力。
因此,由一种或多种亲水聚合物交联而成,具有良好吸水性的水凝胶因其可提供湿润微环境,在伤口治疗领域备受关注。常用聚合物材料主要有海藻酸钠、聚乙烯醇、卡拉胶、明胶、纤维素等,但水凝胶的机械强度普遍较低,稳定性差,遭受外力冲击后,难以复原。随着伤口周围皮肤的弯曲、扭转、拉伸或压缩时极易发生结构坍塌,从而机械性能骤降,无法持续作为有效屏障抵御外力冲击。这一过程也可能会导致凝胶内药物的大量释放,进一步引起蛋白多肽类药物的迅速降解失活。同时水凝胶贴片难以完全适配伤口形状更加剧了其对伤口力学保护的不足。因此适宜的伤口递药系统除可为伤口愈合提供湿润微环境外,还应提供适宜的力学微环境,抵御意外外力冲击,并适配于伤口周围皮肤的牵拉,而不造成患者不适。可见,构建具有力学响应特性的伤口递药系统是为伤口部位提供有效屏障,促进手术感染切口等愈合的关键。
因此,针对慢性伤口感染局部治疗的两大瓶颈:如何有效杀灭致病菌与如何为病灶提供有效屏障,需要提供一种新型抗菌药物,其可有效突破生物被膜渗透屏障,且可作用于非特异性靶点而杀灭细菌,此外还需构建一种能够响应外力刺激的新型递药系统以运载该抗菌药物,以期在为伤口提供有效屏障的同时,实现局部药物高浓度,杀灭致病菌,最终促进感染伤口快速愈合。
发明内容
基于此,本发明针对慢性伤口感染局部治疗的两大瓶颈:如何有效杀灭致病菌与如何为病灶提供有效屏障,本发明提供了一种新型抗菌纳米颗粒以及包含该抗菌纳米颗粒的溶致液晶前体溶液喷雾敷料。该抗菌纳米颗粒可有效突破生物被膜渗透屏障,且可作用于非特异性靶点而杀灭细菌,抗菌效果优异。该溶致液晶前体溶液喷雾敷料在为伤口提供有效屏障的同时,可以实现局部药物高浓度,杀灭致病菌,最终促进感染伤口快速愈合。
本发明具体包括如下技术方案。
一种抗菌纳米颗粒,由树枝状抗菌肽和金纳米粒反应得到,所述树枝状抗菌肽为用树枝状聚酰胺-胺支链化的聚赖氨酸,具有如下式(I)所示结构:

在其中一些实施例中,所述树枝状抗菌肽的制备方法包括如下步骤:
1)将N6-苄氧羰基-L-赖氨酸环内酸酐与G0PAMAM或G1PAMAM分别溶于无水N,N-二甲基甲酰胺中,N2保护下,混合并冰浴搅拌反应20小时-28小时;
2)将适量正丁醇加入至步骤1)的反应液中搅拌0.8h-1.2h,除去未完全反应的N6-苄氧羰基-L-赖氨酸环内酸酐,反应液真空浓缩,转移至乙醚中洗涤沉淀,离心浓缩,取沉淀真空干燥;
3)将步骤2)干燥后的沉淀溶解于三氟乙酸,加入氢溴酸水溶液,室温搅拌反应1.5h-2.5h,然后将反应溶液转移至乙醚中,洗涤,离心分离沉淀物;
4)将步骤3)所得沉淀物溶解于稀盐酸,转移至透析袋,用水透析,冻干后即得所述树枝状抗菌肽。
在其中一些实施例中,步骤3)包括:将步骤2)干燥后的沉淀按每克沉淀4mL-6mL溶解于三氟乙酸,再按每克沉淀8mL-12mL加入氢溴酸水溶液,室温搅拌反应1.5h-2.5h,然后将反应溶液转移至乙醚中,洗涤,离心分离沉淀物;所述氢溴酸水溶液的质量浓度为28-40%。

将所述树枝状抗菌肽配置成浓度为8mg/mL-12mg/mL的水溶液,取所述水溶液加入至浓度为0.04mg/mL-0.06mg/mL的金纳米粒水溶液中,振摇,即得所述抗菌纳米颗粒。
上述的树枝状抗菌肽和/或抗菌纳米颗粒在制备抗菌药物中的应用。

将所述溶致液晶基质材料熔融后和有机溶剂混合均匀,加入或者不加所述晶格调节剂,涡旋过程中缓慢滴入所述药物溶液,搅拌,将得到的均一前体溶液灌装至喷雾容器中,即得溶致液晶前体溶液喷雾敷料。
本发明的抗菌纳米颗粒和溶致液晶前体溶液喷雾敷料的相关原理如下:
聚赖氨酸(ε-Poly-lysine,PLL)是由微生物发酵分离纯化得到的具有25~35个赖氨酸氨残基的聚合物。其表现出明显抑菌活性,对革兰氏阴性菌、革兰氏阳性菌、霉菌、酵母菌等都有良好抑制效果,常被用作食品防腐剂。PLL是由低疏水性的赖氨酸构成的多肽,从而对哺乳动物细胞安全性较高,作用机制为非特异性靶点,因此几乎不产生耐药性。同时其于体内可被生物降解,转变为人体必需氨基酸之一的赖氨酸,被机体吸收,安全、无毒。本发明选择正电荷强且疏水性弱的PLL进一步进行结构改造,以聚酰胺-胺(Polyamidoamine,PAMAM)为核,制备得到了树枝状的PAMAM-PLL,提高了其正电荷密度,缩短了侧链长度,从而提高了PLL的抗菌活性;进一步地,将树枝状的PAMAM-PLL连接到金纳米粒(AuNPs)的表面,金纳米粒具有粒径可控、无毒性和非免疫原性,同时AuNPs可作用于细菌细胞内,通过干扰能量代谢、诱导活性氧产生、导致氧化损伤等多重机制杀死细菌,与树枝状PLL产生协同抗菌作用,从而可以进一步提高所得药物抗菌活性。因此,本发明将PAMAM-PLL与AuNPs共价连接,构建得到的PAMAM-PLL@AuNPs具有非常优异的快速抗菌活性。AuNPs具有生物组织高渗透性与灵活运动轨迹,能够引导PAMAM-PLL突破生物被膜渗透屏障,具有不同抗菌机制的PAMAM-PLL和AuNPs可发挥协同作用,快速瓦解微菌落节点,进而破除生物被膜,杀灭耐药菌,治疗顽固性、复发性和持续性皮肤创面感染。
溶致液晶主要是由分散在有机溶剂中的液晶材料分子,在遇到溶剂之后(通常是水),发生溶剂交换,导致溶致液晶材料分子在水中发生分散和自组装形成的。溶致液晶应用最为广泛的一个特点是其独特的相转变特点,即溶致液晶材料分子在未遇到水分子之前为溶液状态,在与水分子接触后发生自组装,形成固态的溶致液晶凝胶,完成由液态向固态的相转变。发有明采用将溶致液晶材料分子与有机溶剂混合的方式进行应用,即前体溶液的方式。前体溶液的应用方式具有诸多优点,如其粘度小,可以十分方便地以液态的形式进行推注或者喷雾;在给药部位吸收生理环境中的水分子后可原位形成凝胶并进行滞留,完成药物释放等生理功能;液态向固态的通常较为敏感,相转变迅速;液态的给药方式具有使用的顺应性好等优点。具体在皮肤创面的愈合中,由于皮肤创面在受损后难以维持平整,而是多以凹凸不齐的状态存在,因此溶致液晶前体溶液喷雾的形式可以充分与伤口进行贴合,分布满创面的各个部位,充分发挥敷料的功能。
发明人在大量实验研究的基础上,首次制备得到了一种新型抗菌纳米颗粒PAMAM-PLL@AuNPs,并且将其与溶致液晶材料等组分制备成溶致液晶前体溶液喷雾敷料。前体溶液敷料中以溶致液晶材料为基质,在应用时为溶液状态,有利于敷料的喷洒以及对伤口部位紧密且完整的覆盖,前体溶液吸收水分后发生相转变形成具有一定晶格结构的凝胶,贴附于伤口处发挥保护与隔离的作用。克服了传统敷料以固态的形式进行涂抹或者贴附时带来的顺应性差、难以与伤口表面形成完整贴附、给予敷料剂量不可控等的问题;敷料前体溶液体系以作用于非特异性靶点的阳离子抗菌肽聚赖氨酸(PLL)为基础,以聚酰胺-胺(PAMAM)为内核,对其进行树枝状支链化结构改造,合成PAMAM-PLL,通过提高正电荷密度来提高其抗菌活性。并将PAMAM-PLL连接至高渗透性的金纳米粒(AuNPs)表面,所得PAMAM-PLL@AuNPs可以突破生物被膜渗透屏障并通过多重机制提高杀菌活性。进一步地将PAMAM-PLL@AuNPs装载于溶致液晶晶格结构中,利用其自发相转变与晶格结构的力学响应特性构建具有自愈特性的皮肤递药系统,在清除伤口致病菌的同时,形成屏障保护伤口,并为其提供适宜的愈合环境,促进伤口愈合。
因此,本发明的抗菌纳米颗粒、溶致液晶前体溶液喷雾敷料及其制备方法具有如下有益效果:
本发明制备的抗菌纳米颗粒具有抗菌活性高,起效快,能够有效抑制生物被膜的形成和清除生物被膜,其抗菌活性和抑制生物被膜形成的能力远远好于单独的聚赖氨酸,并且好于以聚酰胺-胺(PAMAM)为内核进行树枝状支链化结构改造的PAMAM-PLL。
本发明所制备的溶致液晶前体溶液喷雾敷料均一稳定,粘度低,有利于应用过程中的喷洒;吸水性能良好,可对伤口处的组织渗出液以及伤口清创后残留的液体进行迅速的吸收,营造出适合伤口恢复的低氧湿润环境;前体溶液触变性高,所形成的凝胶模量与皮肤接近,能够在拥有良好生物相容性的基础上在伤口处进行贴附,抵抗外力磨损或者冲击并维持完整状态,发挥保护皮肤的功能;药物释放迅速,可在敷料更换周期内完成。该溶致液晶前体溶液喷雾敷料在为伤口提供有效屏障的同时,可以实现局部药物高浓度,杀灭致病菌,最终促进感染伤口快速愈合。
本发明的抗菌纳米颗粒和溶致液晶前体溶液喷雾敷料的制备方法工艺简单,生产成本较低,制备和存储稳定性高,有利于工业化生产。对于慢性伤口的抗菌以及愈合有重要意义,具有良好的应用前景,有利于推进溶致液晶的工业化应用与发展。

以下为具体实施例。
本发明的溶致液晶前体溶液喷雾敷料的制备、所载药物的结构改造方法。通过氨基酸酸酐开环聚合法,控制PAMAM与PLL投料量,合成不同支链氨基酸数目的树枝状抗菌肽PAMAM-PLL,引入高渗透性的AuNPs,将两者共价连接,构建PAMAM-PLL@AuNPs。将药物溶解于水,得到水相,将其添加入溶致液晶材料中,并在1000rpm转速下进行搅拌,得到均一的层状前体溶液;所得前体溶液喷雾敷料均匀覆盖伤口表面在遇伤口渗出液可进行自组装并发生相转变,形成立方液晶凝胶,在慢性伤口感染部位释放抗菌药物,AuNPs具有生物组织高渗透性与灵活活动轨迹,能引导PAMAM-PLL突破生物被膜渗透屏障,协同发挥作用,清除致病菌,控制感染,凝胶具有良好的吸水性与持水性为伤口提供湿润微环境,促进伤口愈合。
实施例1:不同支链长度PAMAM-PLL的合成、表征与抗菌性能评价
本实施例以G0代和G1代的树枝状聚酰胺-胺(PAMAM)为内核与引发剂,通过控制N6-苄氧羰基-L-赖氨酸环内酸酐(Lys-NCA)投料量为180mg—800mg,使用氨基酸酸酐开环聚合法合成了具有不同支链长度及不同支链数量的树枝状抗菌肽PAMAM-PLL。以耐甲氧西林金黄葡萄球菌(MRSA)为试验菌,以最小抑菌浓度(MIC)、最低杀菌浓度(MBC)为指标,对比线性PLL(聚赖氨酸),初步评价了PAMAM-PLL的抗菌性能。
1.1PAMAM-PLL的具体合成步骤如下所示:
1)将适量Lys-NCA、与G0PAMAM或G1PAMAM(投料量如表1所示)分别溶于无水N,N-二甲基甲酰胺(DMF),N2保护下,混合并冰浴搅拌反应24h;
2)将0.5mL正丁醇加入至上述反应液搅拌1h,除去未完全反应的Lys-NCA单体,反应液真空浓缩,转移至乙醚(40mL)中洗涤沉淀3次,离心浓缩,取沉淀真空干燥12h(0.1mbar);
3)将步骤2)干燥后的沉淀溶解于三氟乙酸(TFA)(5mL/g沉淀),加入质量浓度为33%的氢溴酸水溶液(10mL/g沉淀),室温搅拌反应2h,溶液转移至乙醚中,洗涤3次,离心分离沉淀物;
4)将步骤3)所得沉淀物溶解于稀盐酸(0.2mol/L,0.2mL/mg沉淀物),转移至透析袋(3.5kda),超纯水透析24h*4次,冻干后即得目标产物PAMAM-PLL。

1.2对本实施例制备的PAMAM-PLL的结构进行如下检测和表征
1H NMR:取适量样品溶解于500μLD2O,转移至核磁管,采用超导核磁共振波谱仪测定聚合物的氢原子化学位移,计算PAMAM中氢原子与聚赖氨酸侧链亚甲基氢原子比值,分析PAMAM-PLL的支链长度。结果如图3中的A所示,PAMAM的特征峰在3.0~3.4ppm与2.2~2.8ppm范围内,PAMAM-PLL的1HNMR图谱在1.0~2.0ppm处出现两个新的特征峰,对应于赖氨酸残基中的三个亚甲基(图3中的B)。由PAMAM与赖氨酸亚甲基的峰面积比例计算可得PAMAM-PLL的实际聚合度,结合所选用PAMAM的末端氨基数目,可进一步计算得到PAMAM-PLL的单条聚赖氨酸支链的聚合度。
结果如表1所示:G0和G1与不同投料量的Lys-NCA合成的PAMAM-PLL产物中支链数量分别为4、8,单条聚赖氨酸支链的聚合度(支链长度)分别为8、12;所得产物分别记为PAMAM4-PLL8(P1),PAMAM4-PLL12(P2),PAMAM8-PLL8(P3)、PAMAM8-PLL12(P4),其中4和8代表支链数目,8和12代表支链长度。即PAMAM-PLL的结构式如下式(I)所示,P1和P2中的m为4,n分别为8和12;P3和P4中的m为8,n分别为8和12。
Zeta电位:取适量样品溶解于超纯水,配置50μg/mL样品溶液,采用ZetasizerNano ZS90测定样品电位。如图3中的C所示,支链数目相同时,随支链长度的增加其Zeta电位逐步增大,支链长度相同时,随支链数目增加其Zeta电位逐步增大。
1.3采用MIC和MBC对不同结构PAMAM-PLL进行抗菌性能评价
MIC:采用肉汤稀释法分别测定了P1~P4的MIC与MBC,取对数生长期的MRSA悬液,测定浊度,采用MHB培养基稀释调整细菌密度为5×105CFU/mL。取适量P1~P4溶解于超纯水,倍比稀释成500-12.5μg/mL的稀释液,每孔加入菌液50μL,共设6个平行孔,以MHB培养基处理组作为阳性对照。将96孔板置于37℃培养箱孵育16h,测定600nm下的吸光度。酶标仪测定结果为抑制细菌生长的最小浓度MIC。
MBC:在上述测定后的96孔板中各孔分别取100μL悬液,涂布于MHA培养基,重复3次,置于37℃培养箱孵育16h,以未见明显菌落生长的最低药物浓度作为该药物的MBC。
结果如图4所示。所有结构均表现出明显的抗菌活性,其MIC与MBC从P1-P4呈现出明显的浓度依赖性,随支链数目增加、支链长度增加而降低。P4具有8条支链,每条支链有12个赖氨酸,即12个带正电的伯胺基团,共96个正电荷,其对于MRSA的MIC和MBC分别为12.5μg/mL与25μg/mL。
实施例2:时间杀菌曲线
本实验通过平板菌落计数法对比了P3、P4与线型PLL在同等浓度下的时间杀菌曲线。取对数生长期的MRSA悬液,测定浊度,采用MHB培养基稀释调整细菌密度为5×105CFU/mL。将适量上述组别药物分散于超纯水并倍比稀释,与MRSA培养液1:1混合,置于37℃培养箱150rpm振摇0,1,2,3,4,6,8,10,12h后取出,进行菌落计数。以时间为横坐标,菌落数的对数值为纵坐标绘制时间杀菌曲线。
结果如图5所示,未经药物处理的对照组MRSA在前6h快速增殖,随后逐渐到达平台期,而抗菌肽处理组细菌增殖被抑制。线型PLL、P3与MRSA作用的第一个小时中,MRSA仍继续增殖但增殖速度低于对照组,此后逐步杀灭MRSA,抑制其继续增殖,其中线型PLL在12h内仅能抑制MRSA增殖,P3处理10h后无细菌存活,P4给药之后立刻抑制细菌增殖,并且在1h即杀死全部细菌。
实施例3:PAMAM-PLL@AuNPs的合成与表征
本实施例通过配体交换法将实施例1中合成的PAMAM-PLL与金纳米粒(AuNPs)表面连接,通过紫外吸收光谱UV-vis、Zeta电位的测定、微观形貌观察及元素分析,对P4@AuNPs进行体外结构与性能表征。
2.1 PAMAM-PLL@AuNPs具体合成步骤如下
将实施例1制备的PAMAM-PLL配置成浓度为10mg/mL的水溶液,备用,取适量水溶液加入至1mL 0.05mg/mL的AuNPs(粒径5nm)水溶液中,剧烈振摇,进行配体交换。本实验设置了P4:AuNPs(w/w)不同投料比的处方,P4:AuNPs(w/w)为2:1-8:1。P4加入AuNPs溶液,剧烈振摇时,体系逐渐由AuNPs溶液的酒红色向淡紫色转变,最终可观察到明显的紫色,配体交换成功。
2.2 PAMAM-PLL@AuNPs的体外表征
紫外吸收广谱:采用紫外可见光吸收光谱仪在400~700nm波长范围内,对样品溶液进行光谱扫描;如图6中的A,PAMAM-PLL无紫外吸收,AuNPs在516nm波长处有明显紫外吸收。相对于AuNPs,P4@AuNPs(4:1连接)的紫外吸收发生红移(由516nm转变到542nm左右),这是由于引入PAMAM-PLL酰胺键上羰基的碳氧双键的超共轭效应所致,表明PAMAM-PLL成功连接至AuNPs表面。
Zeta电位:取样品溶解于超纯水,配置50μg/mL样品溶液,采用Zetasizer NanoZS90测定样品电位;如图6中的B,P4连接至AuNPs表面后,其Zeta电位略有降低,但仍保持正电位,可与负电细菌细胞膜相互作用。
微观形貌及元素分析:采用透射电子显微镜(TEM)观察样品外观形貌。样品浓度为200μg/mL,镊子夹取铜网,反复蘸取样品溶液,自然干燥后,放入样品。加速电压设置为120kV,随后,采用TEM的能谱仪附件(EDS)分析样品表面的C、N、O、Au元素组成。采用TEM观察P4@AuNPs的外观形貌。由图6中的C可知,P4连接至AuNPs表面后,AuNPs仍保持原有的球形,粒径无显著变化,仍为5nm左右,且依旧为单分散存在,表明P4@AuNPs稳定性高,不易团聚。采用EDS分别分析了AuNPs与P4@AuNPs(4:1连接)粒子表面C,N,O,Au四种元素的含量,C,N,O的出峰位置分别为0.277,0.392与0.525keV,Au的出峰位置为9.713与2.123keV,谱图如6中的D所示。相对于AuNPs,树枝状抗菌肽连接后的P4@AuNPs中N/Au的原子比例明显上升(从0.61升高至1.19),进一步证明PAMAM-PLL在金纳米粒表面的成功连接(图6中的E)。
实施例4:PAMAM-PLL@AuNPs的体外抗菌性能评价
取对数生长期的MRSA悬液,测定浊度,采用MHB培养基稀释调整细菌密度为5×105CFU/mL。将不同组别及浓度的药物溶液50μL与MRSA50μL培养液1:1混合,分组:PBS组,AuNPs组(6.25μg/mL),P4(12.5μg/mL)组、P4(25μg/mL)组、P4(50μg/mL)组,P4@AuNPs(12.5μg/mL:6.25μg/mL)组,P4@AuNPs(25μg/mL:6.25μg/mL)组,P4@AuNPs(50μg/mL:6.25μg/mL)组,置于37℃培养箱150rpm振摇0,1,2,3,4,6,8,10,12h后取出,进行菌落计数。以时间为横坐标,菌落数的对数值为纵坐标绘制时间杀菌曲线。考察P4@AuNPs的时间杀菌动力学。通过时间杀菌动力学实验评价AuNPs接入前后的杀菌速率。
结果表明AuNPs单独使用时几乎与空白对照组无明显差别,但当PAMAM-PLL引入AuNPs后,杀菌速率明显提高,并且P4@AuNPs的抗菌速率显著高于单独的P4(图7)。当P4@AuNPs中PAMAM:AuNPs浓度为50μg/mL:6.25μg/mL时,8:1接枝的P4@AuNPs的时间杀菌曲线与相同浓度的P4完全一致,杀菌速率未见提高,这可能是由于高浓度下的P4即可快速杀灭细菌,并且,AuNPs的接枝比例不高,从而对其抗菌速率的提高并不明显;当P4@AuNPs中PAMAM:AuNPs浓度为25μg/mL:6.25μg/mL时,4:1接枝时P4@AuNPs的杀菌速率提高至单独P4的2.67倍,于3h即可杀灭全部细菌,但相同浓度下的P4需要8h才能达到同样效果。12.5μg/mL浓度下的P4仅能抑制细菌增殖,而不能完全杀灭细菌,但当P4@AuNPs中PAMAM:AuNPs浓度为12.5μg/mL:6.25μg/mL以2:1的比例接枝后,即可于8h时杀灭所有细菌。
实施例5:PAMAM-PLL@AuNPs的体外抗生物被膜活性评价
生物被膜生物量评价:取对数生长期的MRSA,测定菌液浊度,采用MHB培养基稀释调整菌液密度为2×107CFU/mL,对照组:PBS,加药组:线型PLL、P4、AuNPs、P4@AuNPs(P4:AuNPs=4:1(w/w)),终浓度分别为100、100、25与125μg/mL,接种于96孔板,每孔加入混合菌液200μL,各设6个平行孔,以PBS为对照组。置于37℃培养箱培养48h或72h,弃上清,PBS轻轻冲洗3次,风干后,每孔加入200μL1%(w/w)结晶紫溶液染色,染色10min后,PBS轻轻冲洗3次以除去残余结晶紫,加入200μL无水乙醇溶解附着于生物被膜的结晶紫,静置20min后,在595nm处测定吸光度。结果如图8中的A所示,线型PLL、P4与P4@AuNPs均可抑制MRSA生物被膜形成,且呈现剂量依赖性,随药物浓度升高,抑制能力增强。经支链化结构改造后的P4抑制生物被膜形成的能力显著高于线型PLL。62.5μg/mL浓度下,48h内P4可抑制超过80%的生物被膜形成,而同浓度下线型PLL的抑制率仅为44.15±2.58%。此外,相对于P4,P4@AuNPs抑制生物被膜形成的能力提高了14.93%,较低浓度下P4@AuNPs也可显著抑制MRSA生物被膜的形成。
生物被膜结构观察:采用CLSM观察药物处理后MRSA生物被膜的形成情况可视化生物被膜结构变化。MRSA菌液用MHB培养基稀释至2×107CFU/mL,分别与PBS、线型PLL、P4、AuNPs、P4@AuNPs(P4:AuNPs=4:1(w/w))溶液以1:1混合,使得线型PLL、P4、AuNPs与P4@AuNPs的终浓度分别为100、100、25与125μg/mL。各取1mL混合菌液加入CLSM专用24孔玻璃板中,37℃培养48h。吸除上清液,PBS轻柔冲洗1次,每孔加入SYTOTM9/PI双染染料(SYTOTM930μM,PI 5μM)或FITC-conA染料(500μg/mL)500μL,室温避光染色30min,染色完成后,PBS轻轻冲洗3次,除去残余染料,每孔加入500μL PBS溶液,采用CLSM拍摄图像并进行三维重构。SYTOTM9,PI,FITC-conA激发波长分别为514nm,488nm与488nm。结果如图8中的B所示,分别为CLSM二维图片,和三维重构后的生物被膜。PBS对照组与AuNPs组可见密集的绿色荧光,表明两组中MRSA快速增殖,形成的生物被膜结构致密。FITC-conA对EPS基质进行染色,结果如图8中的C所示,SYTOTM9/PI以及FITC-conA的CLSM图中荧光强度由强到弱顺序为PBS,AuNPs,线型PLL,P4与P4@AuNPs。P4@AuNPs组中仅可见少量绿色荧光,表明其处理后细菌大量死亡,仅可见极为稀疏的生物被膜结构和少量存活细菌,P4@AuNPs具有高效的MRSA生物被膜形成抑制效果,且抑制能力强于P4与线型PLL。
实施例6:PAMAM-PLL@AuNPs对细菌形貌的影响
将P4@AuNPs(P4:AuNPs=4:1(w/w))分散于超纯水,配制浓度为250μg/mL的P4@AuNPs溶液(同法配制浓度为200μg/mL的线型PLL、P4溶液,以及浓度为50μg/mL的AuNPs溶液),将其与浓度为2×108CFU/mL的MRSA培养液1:1混合,37℃,150rpm培养箱振摇2h后取出,10000rpm离心5min收集菌体,PBS溶液洗涤离心3次,收集菌体沉淀。随后加入2.5%的戊二醛溶液固定菌体,样品经多次乙醇梯度脱水,收集菌体,加入无水乙醇重悬菌体,滴加至载玻片,风干后采用SEM观察并拍摄图片。
如图9所示,PBS处理的MRSA为典型球状结构,边界清晰,细菌表面光滑、圆润、结构完整,无破损。线型PLL、P4与P4@AuNPs处理后,可见MRSA细胞膜出现不同程度的皱缩、破损,其中P4与P4@AuNPs处理后,菌体明显干瘪皱缩变形,细胞膜表面有不规则内陷甚至孔洞出现,有细胞碎片产生。
实施例7:溶致液晶递药系统的构建与评价
7.1载药溶致液晶前体溶液喷雾敷料及其制备
7.1.1将实施例3制备的P4@AuNPs(P4:AuNPs=4:1(w/w))溶于处方量水中,得药物溶液;以单油酸甘油酯(GMO)为溶致液晶基质,放置于45℃水浴锅中加热至熔融状态,GMO与无水乙醇分别以6:4-8:2(w/w)的比例均匀混合,加入PEG 400(或者不加)作为晶格调节剂,涡旋过程中再逐滴加入处方量的药物溶液(其中所含药物P4@AuNPs在所得载药溶致液晶前体溶液喷雾敷料中的浓度为2mg/g),并在1000rpm转速下进行搅拌,得到均一的前体溶液(处方组成如表2所示),灌装至喷雾容器中即得载药溶致液晶前体溶液喷雾敷料。

注:P4@AuNP:LLCP表示P4@AuNPs在所得载药溶致液晶前体溶液喷雾敷料中的浓度。
采用马尔文旋转流变仪对所制备基质进行粘度检测。如图10中的A所示,所制备处方均呈现出剪切变稀的特性,即在剪切速率较低时具有较高的粘度,在剪切速率增大时,粘度逐渐下降。喷雾剂在喷洒时,外界对前体溶液瞬时施加较大的应力,剪切速率在短时间内上升,假塑性流体的粘度因此下降,可以使前体溶液更加顺利地从容器中喷出。
7.1.2载药溶致液晶前体溶液喷雾敷料(载药LLCP前体溶液)粘度与胶凝时间测定
LLCP前体溶液的粘度对其喷雾效果至关重要,是处方筛选过程中的重要指标。前体溶液自喷雾容器中喷出的剪切速率约为10s-1,以10s-1的剪切速率模拟喷雾自喷瓶喷出的剪切过程,采用流变仪测定此时各处方的剪切粘度,测量夹具型号为CP1/60SR0909SS,测定温度为25±0.5℃。具体结果如表3所示。所有处方的剪切粘度均小于可顺利喷雾的临界值50mPa·s,前体溶液呈现良好的流动状态,且喷雾雾滴小,雾化效果佳,有利于前体溶液喷雾在伤口表面的均匀分布。此外,结果显示处方粘度随处方中GMO含量减少,乙醇含量的增加而降低。
将GMO、乙醇、PEG400、药物溶液按照表2中制备处方F1~F9,取1mL液晶前体加入水浴预热至33±0.5℃的去离子水中,观察前体溶液遇水是否能够形成凝胶并观察其外观变化以判断胶凝度。采用秒表计时,记录从前体溶液加入水中到其完全形成凝胶所需的时间。随着有机溶剂比例的升高,其胶凝时间也逐步延长。F5和F6的胶凝时间均大于5s,胶凝时间过长会导致前体喷雾在伤口部位的流失,且形成的凝胶为松散絮状结构,力学性能较差,轻轻摇动即可分散,结构被破坏。因此综合前体溶液粘度、胶凝时间以及凝胶结构3个因素,处方F1~F4制备的载药溶致液晶前体溶液喷雾敷料的性能更好。

7.2溶致液晶前体溶液喷雾敷料的体外释放行为评价
本实施例以7.1中所制备的溶致液晶前体溶液喷雾敷料作为主体滴入释放介质后即立即相变形成凝胶,浮于液面,单面与释放介质接触,并未直接沉入释放介质溶液中,这种释放方式与前体施加于皮肤伤口表面,组织渗出液缓慢渗入凝胶相似,因此本实施例选择无膜溶出法考察溶质液晶体系的释放行为。
标准曲线的建立:采用样品稀释液倍比稀释试剂盒BSA标准品(2mg/mL),配置12.5~2000μg/mL的系列标准溶液。以50:1的比例混合试剂A与试剂B,配置检测工作液。取各浓度标准溶液加入96孔板,加入0.2mL工作液,充分混合,37℃孵育30min后冷却至室温,在562nm波长下,检测吸光度,每个浓度平行三份。将经过空白校正的吸光值对浓度作图10中的B,绘制标准曲线,并进行线性拟合。
体外释放行为评价:采用无膜溶出法评价前体的体外释放行为,体系恒温于33±0.5℃,振摇频率为100rpm。PBS预热至33±0.5℃,加入5mL至EP管作为释放介质,重量记为m1;吸取约0.15g溶致液晶前体溶液喷雾敷料滴入释放介质中,形成凝胶,重量记为m2。根据差重法,m2与m1相减即得滴入载药LLCP前体溶液的质量,通过载药量即可计算总投药量。
分别于1,3,6,12,24,48h时取出所有释放介质,并加入5mL空白释放介质。取出的释放样品暂存于-20℃冰箱,待全部取样完成后,样品解冻恢复至室温,稀释后采用BCA试剂盒测定浓度Ct,计算累积释放量Mt。
释放行为如图10中的C所示,F2~F4在48h内的累积释放率分别为97.87±3.95%、89.89±3.76%和100.0±4.19%,显著高于F1(62.53±3.75%)。这可能是由于F1中GMO比例较高,形成凝胶后,其晶格结构较为致密,阻滞了药物的释放。前期(0~3h)各处方均快速释放,保证伤口处的初始有效杀菌浓度,3h时,F2~F4的累积释放率分别为64.75%,75.80%与78.46%。然而F3在6~48h几乎无继续释放行为,F4在6~12h呈现较为快速的释放,12h时累积释放率达到了94.43%,但在12~48h几乎无药物释放。在6~48h,F2呈现平稳释放状态,缓慢释放药物以维持创面药物浓度,避免伤口处细菌再度增殖,同时抵御环境致病菌入侵。因此选择前期药物快速释放,随后缓释药物至48h的F2(GMO:EtOH:PEG400:药物溶液=54:13.5:25:7.5,w/w/w/w)作为最优处方。
各处方之间释放行为的差异可能是由于F2和F4中添加了亲水性PEG400,促进液晶晶格向负曲率化转变,扩大晶格,从而促进了药物的释放,二者的释放速率均较高于未添加PEG400的F1与F3。
实施例8:溶致液晶前体溶液喷雾敷料体内药效学研究
构建慢性感染模型对本发明的溶致液晶前体溶液喷雾敷料进行体内药效学评价。以伤口形态、伤口收缩率、伤口细菌残余量、伤口皮肤病理情况以及炎症程度为指标评价了本发明制备的溶致液晶前体溶液喷雾敷料对慢性伤口感染的治疗效果。
8.1空白溶致液晶前体促进伤口愈合性能探究
通过小鼠全层皮肤缺损模型探究了溶致液晶前体喷雾(根据F2处方制备的没有添加药物的空白溶致液晶前体)促进伤口愈合的能力。腹腔注射1%戊巴比妥钠溶液麻醉小鼠(75mg/kg),采用小动物剃毛刀除去小鼠背部和尾部之间的毛发,在剪毛区域涂抹脱毛膏脱去残余毛发,以湿润纱布擦净皮肤。脱毛后,采用碘伏与75%乙醇消毒背部皮肤。用直径为8mm的皮肤打孔器在小鼠背部造成两个对称的圆形伤口,创面间隔5mm左右。将Balb/C小鼠随机分为溶致液晶组与PBS对照组,每组12只。如图11所示,构建小鼠全层皮肤缺损模型,消毒后,将100μL LLCP或PBS溶液轻轻喷雾至伤口,每2天换药1次。分别于给药后1,3,7,10d后,拍照小鼠伤口,并采用Image Pro Plus软件计算伤口面积,按公式2计算伤口收缩率。

如图12中的A所示,给药后,LLCP组小鼠伤口在观察期间内都保持湿润状态,未形成干痂,触之柔软湿润,凝胶呈淡黄色,可能是由于其优异的吸水及保水性,给药后可快速吸收伤口组织渗出液与清创残余生理盐水,并进一步为伤口提供湿润微环境。而PBS对照组中,伤口水分迅速蒸发,导致干痂形成。如图12中的B所示,相对于PBS,LLCP可显著促进伤口愈合,各时间点的伤口愈合率均高于对照组。在1,3及7天时,LLCP组伤口愈合率分别为27.12%,46.57%以及93.32%,在10天时,伤口全部愈合。
8.2溶致液晶前体溶液喷雾敷料促慢性感染伤口愈合性能评价
通过小鼠慢性感染伤口模型探究本发明制备的溶致液晶前体溶液喷雾敷料促进伤口愈合的能力。腹腔注射1%戊巴比妥钠溶液麻醉小鼠(75mg/kg),采用小动物剃毛刀除去小鼠背部和尾部之间的毛发,在剪毛区域涂抹脱毛膏脱去残余毛发,以湿润纱布擦净皮肤。脱毛后,采用碘伏与75%乙醇消毒背部皮肤。用直径为8mm的皮肤打孔器在小鼠背部造成两个对称的圆形伤口,创面间隔5mm左右。将50μL 105CFU/mL的MRSA悬液滴加至伤口表面。待菌液被吸收后,采用无菌薄膜敷料覆盖伤口24h。将动物随机分为4组,每组16只,常规清创消毒后,采用喷雾方式给药,每2天换药1次,每次给药100μL,给药组分别为PBS组、PLLLLCP组(PLL:LLCP=10mg/g,以PLL及其浓度代替P4@AuNP及其浓度,按实施例7处方2制备)、P4LLCP组(P4:LLCP=2mg/g,以P4代替P4@AuNP,按实施例7处方2制备)、P4@AuNP LLCP组(P4@AuNP:LLCP=2mg/g,按实施例7处方2制备)(如图13所示)。
伤口形态学观察:分别于给药后1,3,7天拍照小鼠伤口,对比不同组别、不同时间伤口的愈合情况。使用游标卡尺测量小鼠伤口的长径和短径,计算伤口面积,计算伤口收缩率。细菌感染会引发伤口的过度炎症反应并阻滞伤口愈合,这也是其会转变为慢性伤口的主要原因之一。因此清除病原菌,同时促进伤口愈合是治疗术后感染伤口的关键。
创面愈合情况与伤口收缩率如图14所示,PBS组与PLL LLCP组伤口在第1天时出现严重感染,伤口红肿,在第3天时,伤口表面出现大量脓液,外观凸起,结痂隆起明显,伤口几乎无收缩,呈迟滞性愈合状态。而P4 LLCP组与P4@AuNPs LLCP组第1天时,伤口并未出现严重细菌感染,伤口平滑柔软,第3天时出现少量脓液,被LLC凝胶吸收,伤口收缩。第7天时,P4LLCP组与P4@AuNPs LLCP组伤口面积大幅减小,边缘处可见新生肉芽组织与新生表皮。而PBS组伤口边缘形成大量凝血块,炎症较为严重,伤口仍呈现明显突起。结果表明,针对手术伤口感染,给药初期强效抗菌,有助于快速杀灭伤口致病菌,为降低伤口处炎症水平奠定基础,避免由于细菌感染导致的伤口迟滞性愈合,提高伤口愈合速度,实验结果显示P4@AuNPsLLCP组的伤口愈合率比P4 LLCP组更高,说明P4@AuNPs的抗菌效果比P4更好。
伤口细菌计数:分别于给药后1与7天处死小鼠,剥离伤口及周围1mm皮肤,加入2mL无菌PBS溶液匀浆,十倍稀释至不同浓度样品,取10μL稀释液滴至MHA培养基平板,待液体挥干后,倒置平板于37℃培养箱培养至肉眼可见单菌落,进行计数。按以下公式计算细菌存活率:
结果如图15中所示,与PBS对照组相比,PLL LLCP、P4 LLCP以及P4@AuNPs LLCP组中的残余MRSA数量均大幅降低,P4@AuNPs LLCP组效果最佳,并且P4@AuNPs LLCP组7天内细菌未再度增殖。给药前期,LLC递药系统可快速释放抗菌颗粒,迅速杀灭伤口表面大部分细菌,后期缓慢释放抗菌颗粒,长期起效,避免细菌再度增殖,在手术切口感染的治疗过程中起到了有效的屏障保护作用。此外,P4@AuNPs LLCP组单次给药与第7天时的细菌数量均明显低于P4 LLCP与PLL LLCP组,证明对线型PLL的支链化改造,有效提高了其抗菌活性,树枝状PAMAM-PLL连接至AuNPs表面后,提高了抗菌颗粒的渗透性,同时实现多重机制杀菌,进一步提高了其抗菌活性和抗生物被膜的活性。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。

1.一种抗菌纳米颗粒,其特征在于,由树枝状抗菌肽和金纳米粒反应得到;所述树枝状抗菌肽为用树枝状聚酰胺-胺支链化的聚赖氨酸,其具有如下式(I)所示结构:

所述树枝状抗菌肽和金纳米粒的质量比为1-6:1。

树枝状聚酰胺-胺与N6-苄氧羰基-L-赖氨酸环内酸酐在有机溶剂中以及惰性气体或者氮气保护下反应,得聚合产物,再将所得聚合产物脱去苄氧羰基保护基,即得。
7.根据权利要求6所述的抗菌纳米颗粒,其特征在于,所述树枝状聚酰胺-胺与N6-苄氧羰基-L-赖氨酸环内酸酐的质量比为1:9-42;和/或,
所述有机溶剂为N,N-二甲基甲酰胺;和/或,
所述反应在冰浴下进行,所述反应的时间为20小时-28小时。
8.根据权利要求6所述的抗菌纳米颗粒,其特征在于,所述树枝状抗菌肽的制备方法包括如下步骤:
1)将N6-苄氧羰基-L-赖氨酸环内酸酐与G1 PAMAM分别溶于无水N,N-二甲基甲酰胺中,N2保护下,混合并冰浴搅拌反应20小时-28小时;
2)将适量正丁醇加入至步骤1)的反应液中搅拌0.8h-1.2h,除去未完全反应的N6-苄氧羰基-L-赖氨酸环内酸酐,反应液真空浓缩,转移至乙醚中洗涤沉淀,离心浓缩,取沉淀真空干燥;
3)将步骤2)干燥后的沉淀溶解于三氟乙酸,加入氢溴酸水溶液,室温搅拌反应1.5h-2.5h,然后将反应溶液转移至乙醚中,洗涤,离心分离沉淀物;
4)将步骤3)所得沉淀物溶解于稀盐酸,转移至透析袋,用水透析,冻干后即得所述树枝状抗菌肽。
9.根据权利要求8所述的抗菌纳米颗粒,其特征在于,步骤3)包括:将步骤2)干燥后的沉淀按每克沉淀4mL-6mL溶解于三氟乙酸,再按每克沉淀8mL-12mL加入氢溴酸水溶液,室温搅拌反应1.5h-2.5h,然后将反应溶液转移至乙醚中,洗涤,离心分离沉淀物;所述氢溴酸水溶液的质量浓度为28-40%;和/或,
步骤4)所述稀盐酸的浓度为0.15mol/L-0.25mol/L;所述沉淀物与稀盐酸的配比为1mg:0.15mL-0.25mL;所述透析袋的分子量为3kda-4kda。

12.一种溶致液晶前体溶液喷雾敷料,其特征在于,以重量百分比计,由包括如下组分的原料制备而成:
溶致液晶基质材料40.5%-74%
有机溶剂10%-37%
晶格调节剂0-30%
药物溶液5-10%;
所述溶致液晶基质材料与有机溶剂的质量比为6:4-8:2;
所述药物溶液为含有抗菌活性成分的水溶液;
所述抗菌活性成分为权利要求1-9任一项中所述的抗菌纳米颗粒;
所述晶格调节剂为聚乙二醇-400。
13.根据权利要求12所述的溶致液晶前体溶液喷雾敷料,其特征在于,以重量百分比计,由包括如下组分的原料制备而成:
溶致液晶基质材料47%-74%
有机溶剂12%-28%
晶格调节剂0-28%
药物溶液7-8%。
14.根据权利要求13所述的溶致液晶前体溶液喷雾敷料,其特征在于,以重量百分比计,所述溶致液晶前体溶液喷雾敷料由包括如下组分的原料制备而成:
溶致液晶基质材料53%-55%
有机溶剂13%-14%
晶格调节剂25%
药物溶液7-8%。
15.根据权利要求12-14任一项所述的溶致液晶前体溶液喷雾敷料,其特征在于,
所述抗菌活性成分在所述溶致液晶前体溶液喷雾敷料中的含量为0.1-5mg/g;和/或,
所述溶致液晶基质材料为单油酸甘油酯;和/或,
所述有机溶剂选自二甲基乙酰胺、N-甲基吡咯烷酮、丙二醇、聚乙二醇400、无水乙醇、乙酸乙酯、异丙醇中的至少一种。
16.根据权利要求15所述的溶致液晶前体溶液喷雾敷料,其特征在于,所述抗菌活性成分在所述溶致液晶前体溶液喷雾敷料中的含量为1-3mg/g。
17.一种权利要求12-16任一项所述的溶致液晶前体溶液喷雾敷料的制备方法,其特征在于,包括如下步骤:
将所述抗菌活性成分溶解于水中,得药物溶液;
将所述溶致液晶基质材料熔融后和有机溶剂混合均匀,加入或者不加所述晶格调节剂,涡旋过程中缓慢滴入所述药物溶液,搅拌,将得到的均一前体溶液灌装至喷雾容器中,即得溶致液晶前体溶液喷雾敷料。
