背景技术
晚期糖基化终末产物(AGEs)是美拉德反应(Maillard reaction),如蛋白质、脂肪和核酸的非酶糖基化/氧化以降低糖水平。AGEs的堆积会造成糖尿病、心脏病、肾衰竭、阿尔兹海默病(老年痴呆症)等健康风险。研究证实斑马鱼如暴露于高糖溶液,也会和人类一样加速糖化而产生AGEs。因此斑马鱼适合用作抗糖化测试模型。
专利CN107469066A公开了一种消除98%糖化反应链发生源的源头型抗糖化配方,其通过消除抗糖化反应链的源头,以达到抗糖化的目的,不过,其采用的制备工艺较为繁琐,不利于工业化生产。专利CN113908096A公开了一种美白保湿抗糖化组合物、制备方法及其应用,其体系中加入硫辛酸,巴西番茄荔枝果皮提取物,石榴叶细胞提取物等组分,抑制肌肤发生糖化反应,促进皮肤恢复弹性。
尽管采用α-硫辛酸进行抗糖化反应已经有研究历史,多用于糖尿病类的治疗中,但是对于硫代牛磺酸的抗糖化效果还没有文献公开,本发明通过α-硫辛酸与硫代牛磺酸协同增效可以显著增加抗糖化反应的效果,应用于化妆品,保健品中具有良好的效果。

本发明提供了一种抗糖化硫代组合物的制备方法,包括以下步骤:将硫代磺酸化合物,双硫代有机酸,二甲砜按照重量百分比混合,加入水,配成0.005wt%的抗糖化硫代组合物浓度,即得。
本发明产生了以下有益效果:本发明对硫代磺酸化合物硫代牛磺酸的抗糖化效果进行了研究测试,发现具有显著的抗糖化效果。并且本发明抗糖化硫代组合物,通过硫代牛磺酸与α-硫辛酸的复配增效,对于AGEs的抑制作用大大提升,抗糖化效果显著提高。可以应用于化妆品,食品,保健品中使用,复配组合作用良好,延缓衰老。
具体实施方式
本申请提供了一种抗糖化硫代组合物,所述组合物包括以下物质:硫代磺酸化合物,双硫代有机酸,二甲砜。
在一种优选的实施方式中,所述物质的重量百分比为:硫代磺酸化合物0.01%-0.1%,双硫代有机酸0.05%-0.5%,二甲砜0.3%-7%,余量为水。
优选的,所述物质的重量百分比为:硫代磺酸化合物0.02%-0.06%,双硫代有机酸0.05%-0.3%,二甲砜0.3%-6%,余量为水。
优选的,所述的硫代磺酸化合物包括结构R1,R2,R3,R4中至少包括一个S。
优选的,所述的硫代磺酸化合物选自硫代牛磺酸,硫代苯磺酸,硫代牛黄酸盐,硫代牛磺酸酯中的至少一种。
本体系加入的硫代磺酸化合物使硫代组合物具有显著的抗糖化效果,减少糖基化末端产物AGEs的产生。申请人在实验过程中发现在组合物种加入硫代磺酸化合物可以显著改善组合物的抗氧化效果,使组合物产生显著的抗衰老性能。猜测可能的原因是:人机体的细胞发生老化,除正常的新陈代谢原因外还有很大部分原因是由于细胞发生糖化反应,使细胞出现暗沉,皱纹,弹性下降的问题。当人体摄入过量的糖类物质后,糖类物质分解的葡萄糖与蛋白质发生自由基糖化反应,生成褐色的劣质蛋白AGEs,皮肤中的胶原蛋白和支撑胶原蛋白的弹性蛋白遭到破坏,皮肤出现松弛和下垂,硫代磺酸化合物中含有的共轭双烯键可以捕捉自由基,与自由基结合,达到清除自由基的效果,抑制自由基糖化反应的发生。并且硫代磺酸化合物可以提高抗氧化酶的活性,阻断组织内脂质过氧化链式反应,降低脂质过氧化物对机体的损害,达到延缓衰老的效果。更优选的,所述物质中的硫代磺酸化合物0.05%。
在一种优选的实施方式中,所述的双硫代有机酸,包括消旋α-硫辛酸、R-(+)-硫辛酸、5-[(3S)-二硫环戊-3-基]戊酸中的至少一种。
优选的,所述的双硫代有机酸为消旋α-硫辛酸,所述α-硫辛酸购买自上海高朗化工科技有限公司;本体系中加入α-硫辛酸可以抑制氧化应激反应发生,减少受体表达,抑制糖基化末端产物AGEs的积聚。申请人在实验过程中发现,通过引入双硫代有机酸,优选为α-硫辛酸时,组合物具有良好的抗氧化,抗衰老效果,猜测可能的原因是:人体细胞在高血糖浓度的环境中,蛋白质细胞会发生异常的非酶糖化过程,为氧化应激反应,糖类物质分解的葡萄糖与皮肤细胞中的蛋白质通过非酶类物质发生反应形成AGEs在细胞中聚集,随着AGEs聚集量的增加,细胞的组织功能发生变化,出现衰老迹象。α-硫辛酸中含有的二硫环基团可以清除自由基,阻断氧化应激反应的发生,从而抑制蛋白质的氧化变性,从而达到抗糖化,抗氧化的功效。α-硫辛酸作用于细胞中还可以减少转录序列与受体的结合,抑制AGEs蛋白的生成,从而进一步避免了糖基化末端产物AGEs在细胞中的堆积。申请人在实验过程中进一步发现α-硫辛酸进入人体后可以被还原为双氢硫辛酸,α-硫辛酸与双氢硫辛酸可以促使维生素C,维生素E等物质的产生,进一步发挥抗氧化的功效。
在一种优选的实施方式中,所述的硫代磺酸化合物与双硫代有机酸的重量比为1:(1-5)。
进一步优选的,所述的硫代磺酸化合物与双硫代有机酸的重量比为1:(3-5)。
本体系中硫代磺酸化合物与双硫代有机酸在特定重量比例下使用时,可以进一步提高组合物的糖化效果,增加对AGEs的抑制作用,提高组合物的效果稳定性,减少对表皮的刺激性。申请人在实验过程中发现,硫代磺酸化合物与双硫代有机酸采用1:(1-5)的重量比,更优选的,两者重量比为1:(3-5)时。可以显著提高组合物的抗糖化效果,对AGEs的抑制作用显著提高,并且各个组合物之间的抗糖化效果近似,效果稳定性较高,猜测可能的原因是:硫代磺酸化合物中的共轭硫双键与α-硫辛酸中的双硫环发生协同作用,大大提高了电子云密度,亲电子性明显提高,与自由基反应结合的能力显著增强,进一步减少了葡萄糖与蛋白质之间的自由基反应,避免了肌肤的变色,弹性下降等衰老现象。
更优选的,所述硫代磺酸化合物与双硫代有机酸为硫代牛磺酸与α-硫辛酸时,所述硫代牛磺酸购买自上海遐瑞医药科技有限公司,可以实现抗糖效果的稳定性,各组之间的效果差异不大,猜测原因是,单一硫化物对糖基化反应的作用部位较单一,针对不同类型的糖类分解产物产生的效果差异性较大,采用硫代牛磺酸与α-硫辛酸组合可以抑制多种糖基化反应,降低抑制AGEs的差异化。申请人进一步发现,硫代牛磺酸与α-硫辛酸采用1:(3-5)的重量比,可以使组合物以较温和的方式与皮肤发生作用,超出优选的重量比范围,可能导致组合物的酸性过强,破坏皮肤表面的角质,长期使用可能会导致皮肤角质层变薄,皮肤敏感性升高。更优选的,硫代牛磺酸与α-硫辛酸采用1:4的重量比。
在一种优选的实施方式中,所述的二甲砜与双硫代有机酸的重量比为1:(0.05-0.2)。
优选的,所述的二甲砜与双硫代有机酸的重量比为1:(0.05-0.15)。在本发明体系中,二甲砜与双硫代有机酸在特定重量比例下使用,可以促进糖代谢,减少糖基化反应,促进皮肤胶原蛋白合成,改善组合物与其他产品的协同效果。
更优选的,申请人在实验过程中发现,当双硫代有机酸选择为α-硫辛酸,通过在组合物中引入含硫化合物二甲砜,并且维持二甲砜与α-硫辛酸1:(0.05-0.15)的重量比,可以促进糖类物质在体内的代谢,减少糖基化反应的发生,并且具有促进皮肤胶原蛋白合成的功效。猜测可能的原因是:二甲砜中含有硫元素,进入人体中可以增强体内胰岛素的作用,促进人体糖原的代谢,提高糖的代谢反应,避免糖类物质在体内堆积,与蛋白质发生非酶反应造成皮肤老化。除此之外二甲砜与α-硫辛酸协同作用,可以促进α-硫辛酸在皮肤表皮的渗透,深入皮肤底部抑制糖基化反应,同时二甲砜作为人体胶原蛋白的合成原料,可以促进皮肤胶原蛋白的再生,增加皮肤弹力,改善皮肤衰老状态。申请人进一步发现,当二甲砜与α-硫辛酸采用1:0.1的重量比时,可以改善组合物溶解后的粘度,使组合物在与其他产品复配时具有良好的稳定性,使组合物与产品之间具有良好的相容效果。
本发明提供了一种抗糖化硫代组合物的制备方法,其特征在于,包括以下步骤:将硫代磺酸化合物,双硫代有机酸,二甲砜按照重量百分比混合,加入水,配成0.005wt%的抗糖化硫代组合物浓度,即得。

一种抗糖化硫代组合物的制备方法,包括以下步骤:将硫代磺酸化合物,双硫代有机酸,二甲砜按照重量百分比混合,加入水,配成0.005wt%的抗糖化硫代组合物浓度,即得。

测试方法
将斑马鱼暴露于高糖和丙酮醛诱导高晚期糖基化终末产物(AGEs)模型进行测试。将120尾3天斑马鱼分成6组暴露于1%果糖,1%葡萄糖,1mM丙酮醛和0.05%硫代牛磺酸样品或0.005%硫代组合物样品溶液中,同时设置模型对照组(1%果糖+1%葡萄糖+1mM丙酮醛),暴露4天后将斑马鱼进行冰冻麻醉和蛋白提取,对蛋白质和AGEs含量进行量化及统计分析。

1.一种抗糖化硫代组合物,其特征在于,所述组合物包括以下物质:硫代磺酸化合物,双硫代有机酸,二甲砜;
所述物质的重量百分比为:硫代磺酸化合物0.01%-0.1%,双硫代有机酸0.05%-0.5%,二甲砜0.3%-7%,余量为水;
所述的硫代磺酸化合物选自硫代牛磺酸;所述的双硫代有机酸为消旋α-硫辛酸;
所述硫代磺酸化合物与双硫代有机酸的重量比为1:(3-5);
所述二甲砜与双硫代有机酸的重量比为1:(0.05-0.2)。
2.如权利要求1所述的一种抗糖化硫代组合物,其特征在于,所述二甲砜与双硫代有机酸的重量比为1:(0.05-0.15)。
