在糖化主要指是糖对肌肤真皮层胶原蛋白、弹力蛋白的影响——糖化的胶原蛋白和弹力蛋白将失去它们的生理功能和对皮肤组织的支持作用和弹性,使皮肤出现衰老的表现,主要表现为肤色发黄暗淡、长痘、毛孔粗大以及肌肤失去弹性。当糖分摄入过多的时候,糖分就在体内滞留,而皮肤由真皮层中的胶原蛋白和弹性蛋白构成的,当滞留体内的糖分与这些蛋白质结合,就会形成劣势蛋白质,也就是——晚期糖基化终末产物(AGEs)。在糖化物质AGES的作用下“变色”的蛋白质,不能分解或被人体排出体外,而是沉淀在肌肤底部,肤色暗黄就是糖化变现出的第一信号。 肌肤糖化还表现为肤色暗淡、频发痘痘、毛孔粗大、皮肤松垮、出现皱纹。
所以抗糖化,就是防止皮肤的胶原蛋白和弹力蛋白失去活力,从而使皮肤达到一个更年轻的状态。一般来说,在20岁的时候,我们的皮肤就可以观察到糖化的胶原蛋白,随着年龄的增长皮肤中被观察到的AGEs就会增多,并且糖化的胶原蛋白无法被真皮机制重塑所降解然后再进行更新,所以一旦有所损伤很难修复。
需要我们提起注意的是,并非所有糖化反应都对皮肤有害;盲目“戒糖”,可能没保住皮肤先伤了身体;市面上绝大部分抗糖产品未经专业验证,能否抗糖尚无科学依据。
抗氧化是指抗氧化自由基的简称,英文Anti-Oxidant。人体因为与外界的持续接触,包括 呼吸(氧化反应)、外界污染、放射线照射等因素不断的在人体体内产生自由基。科学研究表明,癌症、衰老或其它疾病大都与过量自由基的产生有关联。研究抗氧化可以有效克服其所带来的危害,所以抗氧化被保健品、化妆品企业列为主要的研发方向之一,也是市场最重要的功能性诉求之一。
抗氧化就是任何以低浓度存在就能有效抑制自由基的氧化反应的物质,其作用机理可以是直接作用在自由基,或是间接消耗掉容易生成自由基的物质,防止发生进一步反应。人体在不可避免地产生自由基的同时,也在自然产生着抵抗自由基的抗氧化物质,以抵消自由基对人体细胞的氧化攻击。研究证明,人体的抗氧化系统是一个可与免疫系统相比拟的、具有完善和复杂功能的系统,机体抗氧化的能力越强,就越健康,生命也越长。
越来越多的研究显示抗氧化是预防衰老的重要步骤,因为自由基或氧化剂会将细胞和组织分解,影响代谢功能,并会引起不同的健康问题。如果能够消除过多的氧化自由基,对于许多自由基引起的及老化相关疾病都能够预防。例如常见的癌症、动脉硬化、糖尿病、白内障、心血管病、老年痴呆、关节炎等,这些疾病都被认为与自由基相关。
AGEs是糖化终产物,它的形成需要多个步骤,通过抑制其中的某个步骤可产生抑制 AGEs 的作用。AGEs的生成与氧化反应密切相关,非酶糖基化生成AGEs的过程涉及大量的氧化反应并伴随着自由基的生成,自由基的存在会显著加快非酶糖基化生成AGEs的速率,因此通过减少糖化过程中自由基的产生,是实现抑制 AGEs 形成的重要方法之一,增强机体抗氧化功能可有效抑制糖基化产物AGEs 的生成。
目前市面上同时抗糖化和抗氧化兼顾的产品比较少,且疗效显著安全性高的产品更是凤毛麟角。而现代生活中,对于同时需要抗糖抗氧化清除自由基的人群越来越多,迫切的需要优秀的产品来满足爱美,爱健康的人士的需求。
发明人提供了一种经过验证的,配比合理的组合物,经试验证实,安全且有非常好的美容效果,抗糖化,抗氧化,清除自由基效果显著,且口感广受赞誉,携带方便,适于长期服用,是市场急需的美容美肤健康产品。
发明内容
在本文发明提供了一种组合物,该组合物是以天然植物成分配合胶原蛋白肽为主要原料,联合酵母抽提物等成分组成。本研究采用野生型AB品系斑马鱼探究得到该组合物对抑制糖基化产物生成,抗氧化有显著效果,能达到抗氧化、祛黄提亮、改善细纹皱纹、延缓衰老等作用。
本发明组合物是由多种成分混合而成,制备工艺简单,因此能最完整地保留原料的营养成分,达到养丰富、口感细腻、易于服用、产品状态稳定的特点。
本发明组合物由以下重量份的原料组成:余甘子粉10-30份,葡萄籽提取物5-15份,红石榴浓缩粉5-15份,野樱莓果汁粉5-15份,酵母抽提物5-15份,胶原蛋白肽0.5-5份,桑葚粉0.5-5份。
本发明组合物优选由以下重量份的原料组成:余甘子粉10份,葡萄籽提取物15份,红石榴浓缩粉5份,野樱莓果汁粉15份,酵母抽提物5份,胶原蛋白肽5份,桑葚粉0.5份。
本发明组合物还可优选由以下重量份的原料组成:余甘子粉30份,葡萄籽提取物5份,红石榴浓缩粉15份,野樱莓果汁粉5份,酵母抽提物15份,胶原蛋白肽0.5份,桑葚粉5份。
本发明组合物亦可优选由以下重量份的原料组成:余甘子粉19.74份,葡萄籽提取物11.05份,红石榴浓缩粉7.90份,野樱莓果汁粉6.32份,酵母抽提物6.32份,胶原蛋白肽1.58份,桑葚粉0.79份。
本发明组合物可根据需要,制备成固体饮料、软体饮料、混合粉、米稀、奶昔、压片糖果、丸剂、胶囊或片剂以供广大消费者服用。
本发明组合物压片糖果中采用的辅料为:山梨糖醇,赤藓糖醇,木糖醇,微晶纤维素,羟丙纤维素、硬脂酸镁,果糖,色缤兰紫色薄膜包衣预混剂,色缤兰透明薄膜包衣预混剂中的一种或几种。
本发明组合物制备压片糖果的工艺为:
(1)原辅料称量:分别将原辅料按照配方量进行称取;
(2)混合:首先将胶原蛋白肽、桑葚粉预混,然后与已称好的余甘子粉、葡萄籽提取物、野樱莓果汁粉、酵母抽提物、微晶纤维素一起投入湿法制粒机内混合均匀;
(3)湿法制粒:加95%乙醇进行湿法制粒;
(4)干燥整粒,得干颗粒备用;
(5)混合、压片:将干颗粒、山梨糖醇与硬脂酸镁投入混合机中混合压片包衣即得。
本发明组合物服用方便、口感好,满足了目标人群改善皮肤,抑制糖基化产物的形成,增加皮肤弹性,保湿锁水;抗氧化、祛黄提亮、改善细纹皱纹、延缓衰老等效果。且便于运输,贮存期长,安全有效,方便有延缓衰老,保健美容需求人士长期服用,适于在人群中推广。

为验证本发明组合物抗糖,抗氧化的作用,特用实施例3所得的压片产物进行了如下产品功效评价试验:
试验例一:氧自由基清除功效评价
1检测器材
1.1样品信息
组合物,棕粉色片剂,临用时用超纯水配制成20mg/mL母液,母液使用后会-20℃储存,等项目完全结束后,客户收到检测报告无疑问后处理。
阳性对照:茶多酚,褐色粉末,批号为Y23M10B83928,阴凉干燥储存,上海源叶生物科技有限公司。临用时用超纯水配制成10mg/mL母液,现配现用。
1.2实验动物
斑马鱼均饲养于28℃的养鱼用水中(水质:每1L反渗透水中加入200mg速溶海盐,电导率为450~550µS/cm;pH为6.5~8.5;硬度为50~100mg/L CaCO3),实验动物使用许可证号为:SYXK(浙)2012-0171,饲养管理符合国际AAALAC认证(认证编号:001458)的要求。
野生型AB品系斑马鱼,以自然成对交配繁殖方式进行,共240尾,每实验组为30尾,年龄为受精后3天(3 dpf)。用于组合物最大检测浓度(MTC)测定。
野生型AB品系斑马鱼,以自然成对交配繁殖方式进行,共180尾,每实验组为30尾,年龄为3 dpf。用于组合物对氧自由基(ROS)清除功效评价。
1.3仪器和试剂
解剖显微镜(SZX7,OLYMPUS,Japan);CCD相机(VertA1,上海土森视觉科技有限公司);精密电子天平(CP214,OHAUS,USA);多功能酶标仪(SPARK,TECAN,Switzerland);紫外线光疗仪(KN-4006,徐州市科诺医学仪器设备有限公司)。
6孔板(Nest Biotech,China);96孔酶标板(Costar,China);CM-H2DCFDA(货号C6827,Life Technologies Corporation,America)。
2检测方法
2.1 MTC
随机选取240尾3 dpf野生型AB品系斑马鱼于六孔板中,每孔(实验组)均处理30尾斑马鱼。分别水溶给予组合物15.6、31.3、62.5、125、250和500µg/mL浓度,同时设置正常对照组(标准稀释水处理斑马鱼)和模型对照组,每孔容量为3mL。各实验组处理2小时后,除正常对照组外,其余组别均采用紫外照射诱导斑马鱼建立光老化模型。模型建立完成后,各实验组继续处理24小时后实验结束,观察统计斑马鱼的毒性表型和死亡情况,确定组合物对光老化模型斑马鱼的MTC。
2.2氧自由基清除功效
随机选取180尾3 dpf野生型AB品系斑马鱼于六孔板中,每孔均处理30尾斑马鱼。分别水溶给予组合物1.73、5.20和15.6µg/mL,阳性对照组茶多酚10.0µg/mL浓度,同时设置正常对照组和模型对照组,每孔容量为3mL。各实验组处理2小时后,除正常对照组外,其余组别均采用紫外照射诱导斑马鱼建立光损伤模型。模型建立完成后,每个实验组均加入活性氧簇特异性检测试剂(CM-H2DCFDA),立即将斑马鱼转移至96孔酶标板中,每孔1尾,每孔容量为100µL,置于28℃培养箱中继续处理20小时后实验结束。应用多功能酶标仪检测各实验组ROS荧光值(S),以ROS荧光值的统计学分析结果评价组合物对光老化模型斑马鱼的氧自由基清除功效。统计学处理结果采用mean±SE表示。氧自由基清除功效计算公式如下:
用方差分析和Dunnett’s T-检验进行统计学分析,p < 0.05表明具有显著性差异。
3检测结果
3.1 MTC
在本实验条件下,组合物对光老化模型斑马鱼的MTC为15.6µg/mL。
3.2氧自由基清除功效
在本实验条件下,组合物有明显的氧自由基清除功效。
试验例二:糖基化改善功效评价
1检测器材
1.1 样品信息

2检测方法:随机选取180尾3 dpf野生型AB品系斑马鱼于六孔板中,每孔均处理30尾斑马鱼。分别水溶给予组合物1.73、5.20和15.6µg/mL,阳性对照组茶多酚10.0µg/mL浓度,同时设置正常对照组和模型对照组,每孔容量为3 mL。各实验组处理2小时后,除正常对照组外,其余组别均采用紫外照射诱导斑马鱼建立光损伤模型。模型建立完成后,各实验组继续处理24小时后实验结束。按照试剂盒说明书收集斑马鱼样品,应用多功能酶标仪测定各实验组糖化血红蛋白含量(C),以糖化血红蛋白含量的统计学分析结果评价组合物对光老化模型斑马鱼糖基化改善功效。统计学处理结果用mean±SE表示,糖基化改善功效计算公式如下:
统计学分析采用方差分析和Dunnett’s T检验,p < 0.05表明具有显著性差异。
检测结果
在本实验条件下,组合物有明显的糖基化改善功效。
具体实施方式
下面通过具体实施例对本发明作进一步详细描述,以助于理解本发明的内容。
实施例1
余甘子粉10g,葡萄籽提取物15g,红石榴浓缩粉5g,野樱莓果汁粉15g,酵母抽提物5g,胶原蛋白肽5g,桑葚粉0.5g
颗粒制备方法为:
(1)原辅料称量:分别将原辅料按照配方量进行称取;
(2)混合:首先将胶原蛋白肽、桑葚粉预混,然后与已称好的余甘子粉、葡萄籽提取物、野樱莓果汁粉、酵母抽提物、一起投入湿法制粒机内混合均匀;
(3)湿法制粒:加95%乙醇进行湿法制粒;
(4)干燥整粒,得颗粒剂。
实施例2
余甘子粉30g,葡萄籽提取物5g,红石榴浓缩粉15g,野樱莓果汁粉5份,酵母抽提物15份,胶原蛋白肽0.5份,桑葚粉5份。
颗粒制备方法为:
(1)原辅料称量:分别将原辅料按照配方量进行称取;
(2)混合:首先将胶原蛋白肽、桑葚粉预混,然后与已称好的余甘子粉、葡萄籽提取物、野樱莓果汁粉、酵母抽提物、一起投入湿法制粒机内混合均匀;
(3)湿法制粒:加95%乙醇进行湿法制粒;
(4)干燥整粒,按常规方式制成固体饮料。
实施例3
山梨糖醇35.14g,余甘子粉19.74g,葡萄籽提取物11.05g,红石榴浓缩粉7.90g,野樱莓果汁粉6.32g,酵母抽提物6.32g,微晶纤维素5.92g,胶原蛋白肽1.58g,桑葚粉0.79g,硬脂酸镁0.48g
压片糖果制备方法为:
(1)原辅料称量:分别将原辅料按照配方量进行称取;
(2)混合:首先将胶原蛋白肽、桑葚粉预混,然后与已称好的余甘子粉、葡萄籽提取物、野樱莓果汁粉、酵母抽提物、微晶纤维素一起投入湿法制粒机内混合均匀;
(3)湿法制粒:加95%乙醇进行湿法制粒;
(4)干燥:50℃干燥后控制水分在2.5%,过筛整粒,得干颗粒备用;
(5)混合、压片:
称量干颗粒重量m1,然后按照0.59*m1称量山梨糖醇m2,按照0.005* (m1+m2)称量硬脂酸镁,将干颗粒、山梨糖醇与硬脂酸镁投入混合机中混合30min压片,即得压片糖果。
Claims (8)
1.一种抗糖化组合物,其特征在于该组合物由以下重量份的原料组成:余甘子粉10-30份,葡萄籽提取物5-15份,红石榴浓缩粉5-15份,野樱莓果汁粉5-15份,酵母抽提物5-15份,胶原蛋白肽0.5-5份,桑葚粉0.5-5份。
2.根据权利要求1所述的组合物,其特征在于该组合物由以下重量份的原料组成:余甘子粉10份,葡萄籽提取物15份,红石榴浓缩粉5份,野樱莓果汁粉15份,酵母抽提物5份,胶原蛋白肽5份,桑葚粉0.5份。
3.根据权利要求1所述的组合物,其特征在于该组合物由以下重量份的原料组成:余甘子粉30份,葡萄籽提取物5份,红石榴浓缩粉15份,野樱莓果汁粉5份,酵母抽提物15份,胶原蛋白肽0.5份,桑葚粉5份。
4.根据权利要求1所述的组合物,其特征在于该组合物由如下重量份的药材组成:余甘子粉19.74份,葡萄籽提取物11.05份,红石榴浓缩粉7.90份,野樱莓果汁粉6.32份,酵母抽提物6.32份,胶原蛋白肽1.58份,桑葚粉0.79份。
5.根据权利要求1-4任一所述的组合物,其特征在于该组合物的制剂可以为固体饮料、软体饮料、混合粉、米稀、奶昔、压片糖果、丸剂、胶囊或片剂。
6.根据权利要求5所述的组合物,其特征在于该组合物压片糖果中采用的辅料为:山梨糖醇,赤藓糖醇,木糖醇,微晶纤维素,羟丙纤维素、硬脂酸镁,果糖,色缤兰紫色薄膜包衣预混剂,色缤兰透明薄膜包衣预混剂中的一种或几种。
7.根据权利要求6所述的组合物,其特征在于该组合物制备压片糖果的工艺为:
(1)原辅料称量:分别将原辅料按照配方量进行称取;
(2)混合:首先将胶原蛋白肽、桑葚粉预混,然后与已称好的余甘子粉、葡萄籽提取物、野樱莓果汁粉、酵母抽提物、微晶纤维素一起投入湿法制粒机内混合均匀;
(3)湿法制粒 :加95%乙醇进行湿法制粒;
(4)干燥整粒,得干颗粒备用;
(5)混合、压片:将干颗粒、山梨糖醇与硬脂酸镁投入混合机中混合压片即得。
8.根据权利要求1-4任一所述的组合物,其特征在于该组合物在制备抗氧化组合物中的应用。
