本发明属于蛋白酶制备技术领域,具体涉及一种兼具界面活性和抗氧化性的植物蛋白酶解物的制备方法。该方法通过以下步骤实现:将高温花生粕进行脱色;取适量高温花生粕粉末,加入去离子水,调节pH和温度,随后向分散液中分别加入酶,酶解,酶解结束后,灭酶离心,将上清液冷冻干燥,‑20ºC保存即可。本发明采用先脱色后酶解的方式制备酶解物,脱色处理工艺简单,便于操作,生产成本低。制备得到的兼具界面活性和抗氧化性的酶解物应用范围更广,可以用在肉制品中减少油脂的析出和油脂的氧化。

背景技术
花生粕是花生制油之后的主要副产品,其蛋白质含量高达40%~50%,是一种丰富的蛋白质资源。目前我国市场上的花生粕主要为高温花生粕,但高温高热条件使蛋白质变性严重、溶解性差、回收率低,其营养价值和功能特性也受到影响。另外,在高温高压条件下发生的化学反应使花生粕呈色较深,故多数高温花生粕被用作饲料,附加值较低。因此,对高温花生粕进行加工,提高蛋白质的回收率,增加高温花生粕的附加值具有重要意义。
酶水解是一种条件温和、不破坏食品中蛋白质等营养物质,可以获得良好蛋白质功能性质并有效提高蛋白质回收率的方法。经过酶解后的水解物易被人体消化吸收,更能促进人体健康,研究表明花生蛋白水解物具有乳化性、起泡性及保水性等生物活性。目前,有关高温花生粕酶解物的研究报道较少,还需继续探讨发掘。

针对现有技术中存在的问题,本发明提供了一种兼具界面活性和抗氧化性的植物蛋白酶解物的制备方法。
本发明为了实现上述目的所采用的技术方案为:
本发明提供了一种兼具界面活性和抗氧化性的植物蛋白酶解物的制备方法,包括以下步骤:
(1)预处理:将高温花生粕置于粉碎机中,粉碎后过筛去除大颗粒和杂质,采用酸溶液洗涤的方式对高温花生粕进行脱色,得高温花生粕粉末;
(2)酶水解:取适量高温花生粕粉末,加入去离子水,调节pH和温度,随后向分散液中分别加入酶,酶解,酶解结束后,灭酶离心,将上清液冷冻干燥,-20ºC保存即可。
进一步的,步骤(1)中,所述酸溶液为浓度为1-4mol/L的盐酸溶液;所述高温花生粕和酸溶液的料液比为1:5;所述脱色为50℃下脱色120min。
进一步的,步骤(2)中,所述高温花生粕和去离子水的料液比为1g:10mL。
进一步的,步骤(2)中,所述调节的pH值为7-8,温度为55℃。
进一步的,步骤(2)中,所述酶为木瓜蛋白酶、碱性蛋白酶、中性蛋白酶、复合蛋白酶或风味蛋白酶;所述酶的加入量为1%(按底物计);所述酶解的时间为360min。
进一步的,所述灭酶的条件为100℃下加热10min;所述离心为4000 rpm下离心30min。
本发明还提供了一种基于上述制备方法得到的动力学模型,所述酶为木瓜蛋白酶时,得到木瓜蛋白酶可控酶解高温花生粕的动力学模型为:DH=0.839ln[1+(1826.859 E0/S0-78.772)t]。
本发明筛选制备兼具功能性和抗氧化性肽的最适用酶,并对其酶解过程进行动力学分析。结果表明,酶水解是提取高温花生粕蛋白的有效手段,最适用酶为木瓜蛋白酶,其水解效率最高,水解度为4.87%,蛋白质回收率为63.77%,起泡性为216.24%,泡沫稳定性为67.19%,EAI为39.7 m2/g,ESI为14.88 min,木瓜蛋白酶水解高温花生粕的动力学模型为DH=0.839ln[1+(1826.859 E0/S0-78.772)t]。
本发明的有益效果为:
(1)本发明提供的方法为高温花生粕的高值化利用提供了方向,制备得到的植物蛋白酶解物起泡性、泡沫稳定性优异。
(2)本发明采用先脱色后酶解的方式制备酶解物,脱色处理工艺简单,便于操作,生产成本低。制备得到的兼具界面活性和抗氧化性的酶解物应用范围更广,可以用在肉制品中减少油脂的析出和油脂的氧化。

具体实施方式
下面通过具体的实施例对本发明的技术方案作进一步的解释和说明。
1、材料与试剂
材料:高温花生粕,购于山东金胜粮油食品有限公司。
试剂:碱性蛋白酶(Alcalase 2.4 L)、中性蛋白酶(Neutrase 0.8 L)、复合蛋白酶(Protamex 2.4AU/g)和风味蛋白酶(Flavourzyme 1000 L)购于北京诺维信生物科技有限公司;木瓜蛋白酶(Papain)购于生工生物工程(上海)股份有限公司;色谱乙腈购于美国Sigma-Aldrich公司; 1,1-二苯基-2-三硝基苯肼(DPPH)试剂购于上海华蓝化学科技有限公司;2,2-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)试剂购于上海源叶生物科技有限公司;其他试剂均为分析纯。
2、仪器与设备
FE28 pH计,梅特勒-托利多仪器(上海)有限公司;TDZ5-WS离心机,湘仪离心机仪器有限公司; K9840自动凯氏定氮仪,山东海能科学仪器有限公司;UV-5100B紫外可见分光光度计,上海元析仪器有限公司;T25高速分散机,德国IKA公司。
实施例1
(1)预处理:将高温花生粕置于粉碎机中,粉碎后过筛去除大颗粒和杂质。采用浓度为4mol/L的盐酸溶液洗涤的方式对高温花生粕进行脱色,脱色条件为:料液比为1:5, 50℃,120 min;
(2)酶水解:取适量高温花生粕粉末,以1:10(w/v)的料液比加入去离子水,并将其调节至酶的最适pH和最适温度(见表1),随后向分散液中分别加入1%(按底物计)的酶,酶解360 min;酶解结束后,将酶解混合物在100ºC下加热10 min灭酶,4000 rpm下离心30 min,取上清液冷冻干燥,-20ºC保存备用。

(一)测定方法说明
1.1 水解度的测定
水解度(DH)的计算采用pH-stat法进行。计算公式如下:
式中B为NaOH的消耗体积(mL),C为NaOH的浓度(mol/L),M为被水解蛋白质的质量(g),α为蛋白质水解时的氨基解离度,htot为每克蛋白中肽键的毫摩尔数(7.13 mmol/g)。
1.2 蛋白质回收率的测定
蛋白质含量测定采用凯氏定氮法进行。蛋白质回收率的测定按照以下公式进行。
按照下列公式计算蛋白质回收率:
1.3 分子量分布测定
采用高效液相色谱(HPLC)测定水解物的分子量分布,检测条件为:色谱柱:TSKgel2000 SWXL(300 mm×7.8 mm);流动相:乙腈﹕水﹕三氟乙酸= 45﹕55﹕0.1(v/v);流速:0.5 mL·min-1;检测波长:214 nm;柱温:30℃;标准分子质量分别为肌红蛋白(Myoglobin,Mw 16951);抑肽酶(Aprotinin,Mw 6511);血管紧张素(Angiotension,Mw 1045);乙氨酸-乙氨酸-酪氨酸-精氨酸(Gly-Gly-Tyr-Arg,Mw 451);乙氨酸-乙氨酸-乙氨酸(Gly-Gly-Gly,Mw 189)。
1.4 起泡性及泡沫稳定性
用20 mM,pH为7的磷酸盐缓冲液配制浓度为2%(w/v)的样品溶液30 mL,用高速分散机机以12000 rpm的速度均质2 min,分别记录均质0 min和30 min后的泡沫体积V1、V2。起泡性(FC)和泡沫稳定性(FS)的计算公式如下:
式中,V1为均质0 min后的泡沫体积,V0为均质前的样品体积。
式中,V2为均质30 min后的泡沫体积。
1.5 乳化活性及乳化稳定性
用20 mM,pH为7的磷酸盐缓冲溶液配制浓度为0.2%(w/v)的样品溶液12 mL,加入4mL大豆油混匀,于10000 rpm均质2 min。均质后0 min和10 min分别从乳化层吸取100 μL样品,与5 mL浓度为0.1% SDS混合,在500 nm处测定吸光值。乳化活性性指数(EAI)、乳化稳定性指数(ESI)的计算公式如下:
式中,∆t为两次测量之间的间隔时间,A10为均质10 min后的吸光值。
1.6 DPPH自由基清除活性的测定
称取一定量的水解物配制成浓度为0.2~2.0 mg/mL的溶液。将2 mL样品溶液与2mL 0.1 mM DPPH 95%乙醇溶液混匀,室温避光反应30 min后在517 nm处测定吸光值A。用等体积乙醇溶液代替DPPH溶液作为空白组,用等体积水代替样品溶液作为对照组,用等体积的水和乙醇混合液调零。DPPH自由基清除活性的计算公式如下:
式中,A1为对照组的吸光值,As为实验组的吸光值,A0为空白组的吸光值。
1.7 酶水解动力模型的构建
以经典的米氏方程为基础,用数学推导结合试验研究的方法,构建蛋白酶水解高温花生粕动力学模型。根据相关数据确定模型参数,并对试验模型进行验证。
蛋白酶酶解蛋白质的反应过程可以表示为:

式中,a、b为动力学参数。
1.8 数据分析
无特殊说明,所有实验平行测定3次,采用IBM SPSS Statistics 25、Origin 2016和Excel软件进行数据处理。
(二)结果与分析
2.1 水解度
不同蛋白酶水解高温花生粕的水解曲线如图1所示。所有蛋白酶水解高温花生粕的水解曲线随时间的增加呈先快速上升,后趋于平稳的趋势。水解30 min后,复合蛋白酶、木瓜蛋白酶、中性蛋白酶和风味蛋白酶的水解进程开始变慢,逐渐达到平稳状态,而碱性蛋白酶的DH一直保持增大趋势,在水解360 min后,高温花生粕经碱性蛋白酶水解的DH最大值为24.87%。值得注意的是,木瓜蛋白酶的水解度在120 min时达到最大值,水解效率最大。
2.2 蛋白质回收率
图2为高温花生粕经不同蛋白酶水解后的蛋白质回收率,由图可知五种蛋白酶均可显著(P<0.05)提高水解物的蛋白质回收率,其中碱性蛋白酶水解产物的蛋白质回收率最高达到82.53%,其次为复合蛋白酶(75.62%)和木瓜蛋白酶(63.77%)。虽然高温花生粕蛋白质变性严重,溶解性差、分散指数低,酶水解的方式可以有效地提取其中的蛋白质,这与酶解后蛋白质分子变小溶解度增大有关。
2.3 分子量分布
分子量大小是影响肽段功能性质和生物活性的重要因素。根据分子量的大小,将色谱峰分为I~V五个区段,分别对应分子量(Mw)>10 kDa、3 ~10 kDa、1 ~3 kDa、500 Da~1kDa 及<500 Da。碱性蛋白酶水解物中小分子量组分(<500 Da及500 Da~1kDa)含量最多,大分子量组分(10 kDa及3~10 kDa)含量最少,而风味蛋白酶水解物中小分子组分含量最少,而大分子量组分最多,说明小分子组分含量与DH呈正相关,大分子组分含量与DH呈负相关。木瓜蛋白酶水解物中>1 kDa组分和<1 kDa组分含量相当。具体结果见表2。

2.4 起泡性和泡沫稳定性
不同蛋白酶水解物的起泡特性如图3所示。木瓜蛋白酶水解物和中性蛋白酶水解物的起泡性最高,优于复合蛋白酶水解物,优于碱性蛋白酶和风味蛋白酶酶解水解物。木瓜蛋白酶水解物有较高的起泡性。为了测定高温粕水解物的泡沫稳定性,对搅拌后的泡沫进行30 min的观察。由图3可知,木瓜蛋白酶水解物的泡沫稳定性最高,可达67.19%。碱性、复合蛋白酶水解物的泡沫稳定性较低,原因可能是水解过程中产生了很多小分子的肽,强度不足以支撑泡沫的稳定。
2.5 乳化活性和乳化稳定性
乳化性用于反映蛋白质在油水界面处的吸附与稳定能力,分别用乳化活性指数(EAI)和乳化稳定性指数(ESI)来表示。图4为不同蛋白酶水解物的EAI和ESI。不同蛋白酶水解物的乳化性不同,木瓜蛋白酶和中性蛋白酶水解物的乳化活性最强,碱性蛋白酶水解物的乳化活性最差,复合蛋白酶水解物的乳化稳定性最强,中性蛋白酶水解物的乳化稳定性最差。
2.6 DPPH自由基清除活性
图5为不同浓度(0.2~2 mg/mL)下水解物的DPPH自由基清除活性,可看出不同的蛋白酶水解物都具有一定的DPPH自由基清除活性,且在一定浓度范围内DPPH自由基清除率与样品浓度呈正相关。用IC50代表DPPH自由基清除率为50%时的样品浓度,IC50越小,DPPH自由基清除能力越强。从表3中可以看出由木瓜蛋白酶水解得到的水解物DPPH自由基清除能力最强(IC50=0.72 mg/mL),其次为复合蛋白酶、碱性蛋白酶、风味蛋白酶水解物,中性蛋白酶水解物DPPH自由基清除能力最差,说明木瓜蛋白酶释放高温花生粕中抗氧化肽段的能力最强。

2.7 酶解动力学模型的构建
2.7.1 模型的拟合
鉴于木瓜蛋白酶水解效率最高,蛋白质回收率较高,起泡性、乳化活性和DPPH自由基清除活性最高,故选择木瓜蛋白酶为最适用酶。采用方程对不同酶浓度(E0)、不同底物浓度(S0)下水解度与水解时间进行回归分析,求出参数a、b,构建酶水解动力模型,结果如图6和表4所示。该方程对数据的拟合度较高(图6),说明方程可以用来描述酶水解过程。b值在不同水解过程中变化不大,趋向于定值1.192。

参数a与E0/S0存在一定线性关系,两者线性关系可表示为a=1532.6E0/S0-66.084(图7)。把上述试验中所得动力学参数a值和b值分别代入方程,得到木瓜蛋白酶可控酶解高温花生粕的动力学模型,即:DH=0.839ln[1+(1826.859 E0/S0-78.772)t]。从该方程可以看出水解度随着初始酶浓度的增大而增大,随初始底物浓度的增大而减小,随着水解时间的延长,水解的反应速率逐渐降低,这与前面的实验结果是一致的,说明该动力模型具有一定的可靠性。
2.7.2 模型的验证
为了验证动力学模型的可靠性,把水解动力学模型的计算结果与实际测定值进行比较。在水解温度为55℃、pH 7.0,初始底物浓度100 g/L、酶浓度0.6 g/L条件下,得到相应水解条件下的理论酶解曲线,结果如图8所示。水解度的理论值与实际值相比,相对平均误差值为7.6%,小于10%,证明所建动力学模型能较好地模拟酶解过程,具有一定的实际应用价值,可以用来描述水解条件与水解度之间的关系。
Claims (7)
1.一种兼具界面活性和抗氧化性的植物蛋白酶解物的制备方法,其特征在于,包括以下步骤:
(1)预处理:将高温花生粕置于粉碎机中,粉碎后过筛去除大颗粒和杂质,采用酸溶液洗涤的方式对高温花生粕进行脱色,得高温花生粕粉末;
(2)酶水解:取适量高温花生粕粉末,加入去离子水,调节pH和温度,随后向分散液中分别加入酶,酶解,酶解结束后,灭酶离心,将上清液冷冻干燥,-20ºC保存即可。
2.根据权利要求1所述的制备方法,其特征在于,步骤(1)中,所述酸溶液为浓度为1-4mol/L的盐酸溶液;所述高温花生粕和酸溶液的料液比为1:5;所述脱色为50℃下脱色120min。
3.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,所述高温花生粕和去离子水的料液比为1g:10mL。
4.根据权利要求1或3所述的制备方法,其特征在于,步骤(2)中,所述调节的pH值为7-8,温度为55℃。
5.根据权利要求1所述的制备方法,其特征在于,步骤(2)中,所述酶为木瓜蛋白酶、碱性蛋白酶、中性蛋白酶、复合蛋白酶或风味蛋白酶;所述酶的加入量为1%(按底物计);所述酶解的时间为360min。
6.根据权利要求1-5任一项所述的制备方法,其特征在于,所述灭酶的条件为100℃下加热10min;所述离心为4000 rpm下离心30 min。
7.一种基于权利要求1-6任一项所述的制备方法得到的动力学模型,其特征在于,所述酶为木瓜蛋白酶时,得到木瓜蛋白酶可控酶解高温花生粕的动力学模型为:DH=0.839ln[1+(1826.859 E0/S0-78.772)t]。
