Abstract
本发明公开了一种硬壳植物种子的破壳催芽方法,具体涉及农业技术领域,本发明通过使用混合溶液进行浸泡,使得混合溶液中的一些成分的添加能够在短时间内对硬壳种子的表皮进行一定程度的软化,从而加快了种子对水分的吸收和气体间的交换,而混合溶液中其他成分的添加可以合成刺激种子发芽和幼苗生长的活性物,不仅可以加速细胞分裂分化促进种胚的发育,并且提取其中的有效营养成分,增强种子的细胞活性,促进种子对营养成分的吸收转化和利用,诱导种子的萌发,同时双氧水的添加可以促进细胞壁成熟和硬化,与以往方法相比,大大缩短了硬壳种子的催芽处理时间,且发芽率得到了明显的提高,从而有效减少了硬壳种子的浪费。

技术领域
本发明涉及农业技术领域,更具体地说,本发明涉及一种硬壳植物种子的破壳催芽方法。
背景技术
随着农业的发展,种子的快速育苗成为提高农林牧经济作物产量的一个重要举措,而种子因外界环境的不适宜或自身生长不成熟,造成的种子休眠期长、发芽率低、出芽不整齐以及发芽质量低等情况,严重影响了后期作物的生长,并导致了生产周期长、产品质量不高等一系列问题,影响经济效益,在农业的种植过程中,由于硬壳种子表皮较硬,需要的发芽时间较长,发芽率低,有的林木种类播种后有时长达2-3年才能发芽,所以在种植前一般需要对其进行催芽处理,目前传统的硬壳种子的催芽方法如下:
1、用外力使种子硬皮破裂,但不要伤及胚芽等重要发芽点,一般使用摩擦、搓洗、敲砸等方式;
2、用化学原料破皮,一般用稀硫酸浸泡,不同的种皮厚度有着不同的浸泡时间,可以细细观察,或用百分之零点三至百分之零点五左右的高锰酸钾溶液浸泡2小时左右,既可以有利于种皮破裂,又可以起到杀灭病菌、保护种子的作用;
3、用沤制沙藏的方法,硬壳种皮在冬春交替季节之际,用湿沙或其他的基质埋藏后,可于春末夏初时取出播种;
4、热水浸种:热水浸种有各种不同的方法,有些种子只需要放入热水中浸泡,还有些种子需要用水煮法克服种子萌发的阻力,由于种皮构造及生理机制不同等多方面的原因,播种的种子发芽期限也不同。
但是利用以上方法播种后虽然可以在一定程度上降低发芽的时间,但是硬壳种子的发芽率也得不到明显提高低,对硬壳种子造成极大的浪费。
因此,研究一种硬壳植物种子的破壳催芽方法来解决上述问题具有重要意义。
发明内容
为了克服现有技术的上述缺陷,本发明提供了一种硬壳植物种子的破壳催芽方法,本发明所要解决的技术问题是:现有技术中的硬壳种子的破壳催芽方法在实际使用时虽然可以在一定程度上降低发芽的时间,但是硬壳种子的发芽率也得不到明显提高低,对硬壳种子造成极大的浪费的问题。
为实现上述目的,本发明提供如下技术方案:一种硬壳植物种子的破壳催芽方法,包括以下步骤:
S1、将采集后的硬壳种子使用温水进行浸泡,浸泡过程中去除飘浮在水面上的不饱满种子和种子空壳,并对硬壳种子进行超声处理,然后用温水清洗种子两次,清洗后放在通风处,避免阳光直射,并去除硬壳种子中的外部杂质、残缺和形态过小的硬壳种子。
S2、将得到的硬壳种子置于混合溶液中浸泡12-48h后置于冷藏室进行冷藏8-36h,冷藏温度为零下4℃-零下2℃,冷藏完成之后置于烘箱中进行烘干处理,温度为45℃-50℃,取出,并用温水进行清洗后喷洒消毒液进行消毒处理,再用清水清洗2-3次。
S3、然后采用浓度为0.3-0.4%的双氧水,浸泡硬壳种子10-36h,然后将得到的硬壳种子捞出洗净后用湿布包裹,同时在育苗盒上先平铺一层垫料,然后将得到的硬壳种子均匀的撒入,并使用湿布在硬壳种子表面进行覆盖保湿,放置在25-30℃左右温度的地方进行催芽处理,并在硬壳种子表面每天喷洒一次浓度为3-5%的营养液。
S4、选择地势平坦且排灌良好的土地进行苗床铺设,首先去除表面的杂草,并在秋冬季深翻两边土,第二遍整地时,将底肥均匀撒于床面上并进行深翻,并在播种前半个月用杀菌剂对苗床进行处理,然后后铺上一层育苗基质,等待硬壳种子芽破壳时,将种子播种在苗床中,再覆盖一层细土,喷水保湿,保持土壤湿度为80-90%,直至种子发芽。
作为本发明的进一步方案:所述超声处理的过程参数为:超声频率40kHz、水温50-55℃下对硬壳种子进行超声处理1h,然后加入10-15℃水中浸泡,15min后继续于超声频率40kHz、水温50-55℃下超声处理1h。
作为本发明的进一步方案:所述垫料的平铺厚度为8-10cm,所述底肥为农家肥,所述农家肥为牛粪、鸡粪、草木灰与水按照5:6:3:300混合后发酵熟热而成,所述细土的平铺厚度为1-2cm。
作为本发明的进一步方案:所述垫料由如下重量份数的原料混合制成:麦麸干粉10-15份、石棉绒1-2份、0.15-0.2份硫酸钾、0.2-0.3份五氧化二磷、陶瓷微粉3-6份、20-30份珍珠岩、草木灰5-10份和湿泥沙5-8份。
作为本发明的进一步方案:所述混合溶液的制备方法为:
将20-30份双氧水、3-5份植物有机酸、2-3份过氧化钠、20-30份壳寡糖、10-15份维生素C、5-8份维生素E、10-15份复硝酚钠、5-8份茉莉酸、0.5-1份硼砂、3-5份牛磺酸、20-30份壳寡糖、8-10份牡蛎提取液、8-10份紫茎泽兰腐熟粉料和12-15份赤霉素分别融入水中,溶解后全部混合,混合均匀后加入20-30份的无水乙醇和浓度为1%的食盐水,继续均混后得到混合溶液。
作为本发明的进一步方案:所述营养液的制备方法为:
将1-3份螫合微元、3-4份芸苔素、5-8细胞分裂素、20-25复合氨基酸加入到15-20份百分比含量为3-7%稀释沼液中进行分散融合15-20min,即得营养液。
作为本发明的进一步方案:所述消毒液为浓度0.2%的硫酸铜溶液和浓度为0.4%的高锰酸钾的溶液按照1:1的比例混合而成得到的混合溶液。
作为本发明的进一步方案:所述催芽处理中,需要每隔6-8h喷一次清水,并对种子进行翻动处理,使得包内种子发生位移,保证氧气供给。
本发明的有益效果在于:
1、本发明通过先对硬壳种子进行筛选,去除硬壳种子中的杂质和问题种子,然后使用混合溶液进行浸泡,使得混合溶液中的双氧水、食盐水、植物有机酸和过氧化钠的添加能够在短时间内对硬壳种子的表皮进行一定程度的软化,从而加快了种子对水分的吸收和气体间的交换,而混合溶液中壳寡糖赤霉素、维生素C、维生素E、茉莉酸、牛磺酸、牡蛎提取液、和紫茎泽兰腐熟粉料的添加可以合成刺激种子发芽和幼苗生长的活性物,不仅可以加速细胞分裂分化促进种胚的发育,并且提取其中的有效营养成分,能够有效增强种子的细胞活性,促进种子对营养成分的吸收转化和利用,从而诱导种子的萌发,同时双氧水的添加可以促进细胞壁成熟和硬化,与以往方法相比,大大缩短了硬壳种子的催芽处理时间,且发芽率得到了明显的提高,从而有效减少了硬壳种子的浪费;
2、本发明通过在催芽的过程中进行消毒液的喷洒,能够有效杀死种子表面和内部的有害菌,降低后期幼苗的病害率,从而提高种子的成活率,且在对硬壳种子进行处理的过程中,通过超声处理可以加快硬壳种子吸水速度,并通过冷藏的方式,加快营养进入种子,提高胚芽抗逆性,提高种苗成活率,种子经低温烘干时促进种皮表面干燥,容易出现裂缝,从而可以加快营养和水分进入,促进硬壳种子中种子的萌发,从而进一步的降低了硬壳种子的催芽处理时间。

具体实施方式
下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1:
一种硬壳植物(山楂)种子的破壳催芽方法,包括以下步骤:
S1、将采集后的硬壳种子使用温水进行浸泡,浸泡过程中去除飘浮在水面上的不饱满种子和种子空壳,并对硬壳种子进行超声处理,然后用温水清洗种子两次,清洗后放在通风处,避免阳光直射,并去除硬壳种子中的外部杂质、残缺和形态过小的硬壳种子。
S2、将得到的硬壳种子置于混合溶液中浸泡36h后置于冷藏室进行冷藏24h,冷藏温度为零下4℃,冷藏完成之后置于烘箱中进行烘干处理,温度为45℃-,取出,并用温水进行清洗后喷洒消毒液进行消毒处理,再用清水清洗2-3次。
S3、然后采用浓度为0.4%的双氧水,浸泡硬壳种子24h,然后将得到的硬壳种子捞出洗净后用湿布包裹,同时在育苗盒上先平铺一层垫料,然后将得到的硬壳种子均匀的撒入,并使用湿布在硬壳种子表面进行覆盖保湿,放置在30℃左右温度的地方进行催芽处理,并在硬壳种子表面每天喷洒依次浓度为5%的营养液。
S4、选择地势平坦且排灌良好的土地进行苗床铺设,首先去除表面的杂草,并在秋冬季深翻两边土,第二遍整地时,将底肥均匀撒于床面上并进行深翻,并在播种前半个月用杀菌剂对苗床进行处理,然后后铺上一层育苗基质,等待硬壳种子芽破壳时,将种子播种在苗床中,再覆盖一层细土,喷水保湿,保持土壤湿度为90%,直至种子发芽。
超声处理的过程参数为:超声频率40kHz、水温50-55℃下对硬壳种子进行超声处理1h,然后加入10-15℃水中浸泡,15min后继续于超声频率40kHz、水温50-55℃下超声处理1h。
垫料的平铺厚度为8-10cm,底肥为农家肥,农家肥为牛粪、鸡粪、草木灰与水按照5:6:3:300混合后发酵熟热而成,细土的平铺厚度为1-2cm。
垫料由如下重量份数的原料混合制成:麦麸干粉10-15份、石棉绒1-2份、0.15-0.2份硫酸钾、0.2-0.3份五氧化二磷、陶瓷微粉3-6份、20-30份珍珠岩、草木灰5-10份和湿泥沙5-8份。
混合溶液的制备方法为:
将30份双氧水、5份植物有机酸、2-3份过氧化钠、30份壳寡糖、15份维生素C、8份维生素E、15份复硝酚钠、8份茉莉酸、1份硼砂、5份牛磺酸、30份壳寡糖、10份牡蛎提取液、10份紫茎泽兰腐熟粉料和15份赤霉素分别融入水中,溶解后全部混合,混合均匀后加入30份的无水乙醇和浓度为1%的食盐水,继续均混后得到混合溶液。
营养液的制备方法为:
将3份螫合微元、4份芸苔素、8细胞分裂素、25复合氨基酸加入到20份百分比含量为3-7%稀释沼液中进行分散融合15min,即得营养液。
消毒液为浓度0.2%的硫酸铜溶液和浓度为0.4%的高锰酸钾的溶液按照1:1的比例混合而成得到的混合溶液。
催芽处理中,需要每隔6h喷一次清水,并对种子进行翻动处理,使得包内种子发生位移,保证氧气供给。
实施例2:
一种硬壳植物(银杏)种子的破壳催芽方法,包括以下步骤:
S1、将采集后的硬壳种子使用温水进行浸泡,浸泡过程中去除飘浮在水面上的不饱满种子和种子空壳,并对硬壳种子进行超声处理,然后用温水清洗种子两次,清洗后放在通风处,避免阳光直射,并去除硬壳种子中的外部杂质、残缺和形态过小的硬壳种子。
S2、将得到的硬壳种子置于混合溶液中浸泡48h后置于冷藏室进行冷藏36h,冷藏温度为零下2℃,冷藏完成之后置于烘箱中进行烘干处理,温度为45℃,取出,并用温水进行清洗后喷洒消毒液进行消毒处理,再用清水清洗2-3次。
S3、然后采用浓度为0.3%的双氧水,浸泡硬壳种子36h,然后将得到的硬壳种子捞出洗净后用湿布包裹,同时在育苗盒上先平铺一层垫料,然后将得到的硬壳种子均匀的撒入,并使用湿布在硬壳种子表面进行覆盖保湿,放置在30℃左右温度的地方进行催芽处理,并在硬壳种子表面每天喷洒依次浓度为5%的营养液。

S2、将得到的硬壳种子置于混合溶液中浸泡12h后置于冷藏室进行冷藏8h,冷藏温度为零下3℃,冷藏完成之后置于烘箱中进行烘干处理,温度为45℃,取出,并用温水进行清洗后喷洒消毒液进行消毒处理,再用清水清洗2-3次。

营养液的制备方法为:

由上表中的对比可知:本发明通过先对硬壳种子进行筛选,去除硬壳种子中的杂质和问题种子,然后使用混合溶液进行浸泡,使得混合溶液中的双氧水、食盐水、植物有机酸和过氧化钠的添加能够在短时间内对硬壳种子的表皮进行一定程度的软化,从而加快了种子对水分的吸收和气体间的交换,而混合溶液中壳寡糖赤霉素、维生素C、维生素E、茉莉酸、牛磺酸、牡蛎提取液、和紫茎泽兰腐熟粉料的添加可以合成刺激种子发芽和幼苗生长的活性物,不仅可以加速细胞分裂分化促进种胚的发育,并且提取其中的有效营养成分,能够有效增强种子的细胞活性,促进种子对营养成分的吸收转化和利用,从而诱导种子的萌发,同时双氧水的添加可以促进细胞壁成熟和硬化,与以往方法相比,大大缩短了硬壳种子的催芽处理时间,且发芽率得到了明显的提高,从而有效减少了硬壳种子的浪费,且通过在催芽的过程中进行消毒液的喷洒,能够有效杀死种子表面和内部的有害菌,降低后期幼苗的病害率,从而提高种子的成活率,且在对硬壳种子进行处理的过程中,通过超声处理可以加快硬壳种子吸水速度,并通过冷藏的方式,加快营养进入种子,提高胚芽抗逆性,提高种苗成活率,种子经低温烘干时促进种皮表面干燥,容易出现裂缝,从而可以加快营养和水分进入,促进硬壳种子中种子的萌发,从而进一步的降低了硬壳种子的催芽处理时间。
最后应说明的几点是:虽然,上文中已经用一般性说明及具体实施例对本发明作了详尽的描述,但在本发明的基础上,以上各实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述各实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解:其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围。
Claims (8)
1.一种硬壳植物种子的破壳催芽方法,其特征在于:包括以下步骤:
S1、将采集后的硬壳种子使用温水进行浸泡,浸泡过程中去除飘浮在水面上的不饱满种子和种子空壳,并对硬壳种子进行超声处理,然后用温水清洗种子两次,清洗后放在通风处,避免阳光直射,并去除硬壳种子中的外部杂质、残缺和形态过小的硬壳种子;
S2、将得到的硬壳种子置于混合溶液中浸泡12-48h后置于冷藏室进行冷藏8-36h,冷藏温度为零下4℃-零下2℃,冷藏完成之后置于烘箱中进行烘干处理,温度为45℃-50℃,取出,并用温水进行清洗后喷洒消毒液进行消毒处理,再用清水清洗2-3次;
S3、然后采用浓度为0.3-0.4%的双氧水,浸泡硬壳种子10-36h,然后将得到的硬壳种子捞出洗净后用湿布包裹,同时在育苗盒上先平铺一层垫料,然后将得到的硬壳种子均匀的撒入,并使用湿布在硬壳种子表面进行覆盖保湿,放置在25-30℃左右温度的地方进行催芽处理,并在硬壳种子表面每天喷洒一次浓度为3-5%的营养液;
S4、选择地势平坦且排灌良好的土地进行苗床铺设,首先去除表面的杂草,并在秋冬季深翻两边土,第二遍整地时,将底肥均匀撒于床面上并进行深翻,并在播种前半个月用杀菌剂对苗床进行处理,然后后铺上一层育苗基质,等待硬壳种子芽破壳时,将种子播种在苗床中,再覆盖一层细土,喷水保湿,保持土壤湿度为80-90%,直至种子发芽。
2.根据权利要求1所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述超声处理的过程参数为:超声频率40kHz、水温50-55℃下对硬壳种子进行超声处理1h,然后加入10-15℃水中浸泡,15min后继续于超声频率40kHz、水温50-55℃下超声处理1h。
3.根据权利要求1所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述垫料的平铺厚度为8-10cm,所述底肥为农家肥,所述农家肥为牛粪、鸡粪、草木灰与水按照5:6:3:300混合后发酵熟热而成,所述细土的平铺厚度为1-2cm。
4.根据权利要求3所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述垫料由如下重量份数的原料混合制成:麦麸干粉10-15份、石棉绒1-2份、0.15-0.2份硫酸钾、0.2-0.3份五氧化二磷、陶瓷微粉3-6份、20-30份珍珠岩、草木灰5-10份和湿泥沙5-8份。
5.根据权利要求1所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述混合溶液的制备方法为:
将20-30份双氧水、3-5份植物有机酸、2-3份过氧化钠、20-30份壳寡糖、10-15份维生素C、5-8份维生素E、10-15份复硝酚钠、5-8份茉莉酸、0.5-1份硼砂、3-5份牛磺酸、20-30份壳寡糖、8-10份牡蛎提取液、8-10份紫茎泽兰腐熟粉料和12-15份赤霉素分别融入水中,溶解后全部混合,混合均匀后加入20-30份的无水乙醇和浓度为1%的食盐水,继续均混后得到混合溶液。
6.根据权利要求1所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述营养液的制备方法为:
将1-3份螫合微元、3-4份芸苔素、5-8细胞分裂素、20-25复合氨基酸加入到15-20份百分比含量为3-7%稀释沼液中进行分散融合15-20min,即得营养液。
7.根据权利要求1所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述消毒液为浓度0.2%的硫酸铜溶液和浓度为0.4%的高锰酸钾的溶液按照1:1的比例混合而成得到的混合溶液。
8.根据权利要求1所述的一种硬壳植物种子的破壳催芽方法,其特征在于:所述催芽处理中,需要每隔6-8h喷一次清水,并对种子进行翻动处理,使得包内种子发生位移,保证氧气供给。
