Abstract
本发明公开了一种3D打印用低生物毒性光敏树脂及制备方法,该树脂由以下重量份的组分组成:双酚A‑甲基丙烯酸缩水甘油酯8‑12份、脂肪族氨基甲酸二甲酯55‑65份、三乙二醇二甲基丙烯酸酯25‑35份、自由基类光引发剂2.5‑3份、天然吸光剂0.1‑0.2份,其中自由基类光引发剂的添加量不超过树脂总量的5%。本发明配方及制备方法制备的光敏树脂在进行3D打印时,除了满足力学性能及耐水性要求外,其固化后无生物毒性,可用于医疗器械的定制打印,扩大使用范围。

技术领域
本发明属于3D打印技术领域,具体涉及一种3D打印用低生物毒性光敏树脂及制备方法。
背景技术
3D打印可连续快速打印三维物体,其大多采用特殊蜡材、光敏树脂、粉末状金属或塑料等可粘合材料作为打印材料介质。部分3D打印机采用DLP数字光处理技术进行工作,打印时光源对光敏树脂进行逐层固化,最终在打印平台下方形成完整的3D打印产品。
目前DLP打印机用光敏树脂大多采用丙烯酸酯树脂、环氧树脂作为主体树脂,采用含羟基的丙烯酸酯、氧杂环丁烷化合物作为活性稀释剂,使用阳离子型光聚合引发剂、自由基型光聚合引发剂作为树脂的光引发剂来提高光敏性,降低临界曝光量,提高打印零件的力学性能和耐水性,以避免由于敏感度较低导致打印失败或者打印出来的模型强度不足的问题。
但是由于以上原料的使用,限制了成品模型在医疗方面的应用。在医疗方面应用时,需要成品模型的材质至少能够通过细胞毒性、致敏性、刺激性的测试,目前多数是采用延长酒精清洗与浸泡的时间来将未完全固化的小分子浸出,从而降低树脂在固化后的小分子释放量;但是以上方法仅适用于树脂分子结构中刺激性官能团含量较低的产品,如果刺激性官能团含量较高,以上的处理方法也无法使树脂满足最终的生物相容性要求。
发明内容
针对现有技术中存在的问题,本发明的目的在于提供一种3D打印用低生物毒性光敏树脂及制备方法,通过特定配方制得的光敏树脂使得3D打印出的最终产品在满足力学性能及耐水性要求的同时,还可应用于医疗器械的定制打印。
本发明通过以下技术方案加以实现:
本发明一方面提供了一种3D打印用低生物毒性光敏树脂,该树脂由以下重量份的组分组成:双酚A-甲基丙烯酸缩水甘油酯8-12份、脂肪族氨基甲酸二甲酯55-65份、三乙二醇二甲基丙烯酸酯25-35份、自由基类光引发剂2.5-3份、天然吸光剂0.1-0.2份,其中自由基类光引发剂的添加量不超过树脂总量的5%。
进一步地,自由基类光引发剂为苯基双(2,4,6-三甲基苯甲酰基)氧化膦和2-羟基-2-甲基-1-苯基-1-丙酮的混合物,天然吸光剂为1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐。
进一步地,树脂由以下重量份的组分组成:双酚A-甲基丙烯酸缩水甘油酯10份、脂肪族氨基甲酸二甲酯60份、三乙二醇二甲基丙烯酸酯30份、苯基双(2,4,6-三甲基苯甲酰基)氧化膦2.2份、2-羟基-2-甲基-1-苯基-1-丙酮0.5份、1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐0.16份。
本发明另一方面提供了一种3D打印用低生物毒性光敏树脂的制备方法,包括以下步骤:
1)用医用酒精清洗所有器具,干燥后备用;
2)将双酚A-甲基丙烯酸缩水甘油酯加热至80℃,称取所需质量于容器内,保温;
3)准确称量脂肪族氨基甲酸二甲酯、三乙二醇二甲基丙烯酸酯于步骤2)的容器内,80℃的温度条件下搅拌30分钟,制得混合物;
4)混合物静置,待温度下将至50℃时,准确称量苯基双(2,4,6-三甲基苯甲酰基)氧化膦、2-羟基-2-甲基-1-苯基-1-丙酮、1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐于容器内,搅拌30分钟;
5)温度降至室温,完成整个生产过程,制得低生物毒性光敏树脂。
进一步地,步骤3)搅拌的线速度为8-10m/s,步骤4)搅拌的线速度为6-8m/s。

本发明主体树脂采用固化后无刺激性基团或刺激性基团含量较低的双酚A-甲基丙烯酸缩水甘油酯,降低固化后树脂刺激性基团的含量;活性稀释剂采用固化后完全不含刺激性基团的脂肪族氨基甲酸二甲酯、三乙二醇二甲基丙烯酸酯,降低可溶出单体的刺激性;采用不同官能度的活性稀释剂互相配合,提高树脂固化后的交联程度,提高成品模型的机械强度,降低成品模型的吸水率;采用高光吸收、低残留的光引发剂苯基双(2,4,6-三甲基苯甲酰基)氧化膦、2-羟基-2-甲基-1-苯基-1-丙酮,降低树脂固化后光引发剂的残留量以及可迁出量;采用天然的吸光剂1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐来控制与调节树脂的光敏感程度,规避添加合成吸光剂带来的风险。

具体实施方式
以下结合具体实施例对本发明技术方案做进一步说明。
实施例
用医用酒精清洗所有器具,干燥后备用;将主体树脂双酚A-甲基丙烯酸缩水甘油酯加热至80℃,称取10g于容器内,保持80℃的温度不变,准确称量脂肪族氨基甲酸二甲酯60g、三乙二醇二甲基丙烯酸酯30g加入主体树脂内,80℃的温度条件下搅拌30分钟,搅拌的线速度为8-10m/s,制得混合物;将混合物静置,待温度下降至50℃时,准确称量苯基双(2,4,6-三甲基苯甲酰基)氧化膦2.2g、2-羟基-2-甲基-1-苯基-1-丙酮0.5g、1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐0.16g于容器内,接着搅拌30分钟,搅拌的线速度为6-8m/s,温度降至室温,完成整个生产过程,制得低生物毒性光敏树脂。
将该实施例制得的光敏树脂用MoonRay-95型DLP 3D打印机制作手术导板,用于以下各试验例。
试验例1
将上述3D打印的手术导板在光功率为4000μW/cm2的ProCure型固化灯箱内60℃环境固化30分钟,弯曲强度可以达到110-120MPa,弯曲模量为2600-2800MPa,一个月吸水率小于40μg/mm3;在经过烘箱130℃烘烤30分钟后机械性能不下降,经过测试热变形温度为135-140℃。
试验例2
手术导板皮肤致敏试验:参照《GB/T 16886.10-2017医疗器械生物学评价》第10部分:刺激与皮肤致敏试验,采用豚鼠最大剂量试验,评价样品潜在的皮肤致敏反应。
1.试验和对照品确定
试验样品:本发明树脂3D打印的手术导板。
对照样品
阴性对照样品:0.9%氯化钠注射液(SC),广西裕源药业有限公司;规格:500mL;批号:H20053015,液体,无色,室温保存
阳性对照样品:2,4-二硝基氯苯,成都艾科达化学试剂有限公司,规格:100g,批号:201904101,诱导浓度0.5%,激发浓度0.1%,溶剂:0.9%氯化钠注射液,液体,浅黄色,室温保存。
2.试验系统鉴别:15只(实验组10只、对照组5只)雄性哈特兰豚鼠,初体重300-500g,健康未使用的动物。
样品浸提:无菌条件下整件取样,按表1的浸提比例(样品:浸提介质)在密闭的惰性容器内振荡浸提,浸提介质为0.9%氯化钠注射液。

浸提前后浸提液状态未发生改变,浸提完成后室温保存,24h内用于测试,浸提液的pH值未经调整,未经过滤、离心、稀释等处理,不加供试品,同条件下制备对照液。
试验步骤
皮内诱导阶段:在试验首日,将对15只豚鼠进行称重和染色,并用电动剃刀除去动物背部区域的毛发,按照图1所示(A、B和C),在每只动物去毛的肩胛骨内侧部位成对皮内注射0.1mL,见图1。
部位A:注射弗氏完全佐剂与选定的溶剂以50:50(V/V)比例混合配制成稳定性乳化剂。
部位B:试验组动物注射试验样品(未经稀释的浸提液);对照组动物仅注射相应溶剂。
部位C:试验组动物按部位B中采用的浸提液浓度,以50:50(V/V)的比例与弗氏完全佐剂与溶剂配制成的乳化剂混合后进行皮内注射,对照组动物注射溶剂与佐剂和溶剂配制成的乳化剂
局部诱导阶段:按注射部位B的最大浓度未产生刺激反应,在局部敷贴应用前(24±2)h,试验区用10%十二烷基苯磺酸钠(溶剂:蒸馏水)进行预处理,按摩导入皮肤。
皮内注射后7±1d,将约8cm2大小的吸收性纱布块用0.5mL供试品浸提液浸透后局部贴敷于每个动物的肩胛骨内侧部位,覆盖诱导注射点。用封闭式包扎带固定样品,并于(48±2)h后除去包扎带和样品。
对照组动物实用对照液同法操作。
激发阶段:于局部诱导后14±1d,用试验样品对全部试验组动物和对照组动物进行激发,用0.5mL供试品浸提液和对照液浸透后2.5cm×2.5cm大小吸收性纱布分别局部贴敷于诱导阶段未试验部位,用封闭式包扎带固定,并于(24±2)h后除去包扎带和贴敷片。
结果观察:除去样品后(24±2)h和(48±2)h,分别观察供试品组和对照组动物激发部位皮肤情况,在全光谱光线下观察皮肤反应,按表2Magnusson和Kligman分级标准对每一激发部位和每一观察时间皮肤红斑和水肿反应进行描述并分级。

结果评价:按Magnusson和Kligman分级标准,如果对照组动物等级小于1,而试验组中等级大于或等于1时一般提示致敏。
如对照组动物等级大于或等于1时,试验组动物反应超过对照组中最严重的反应则认为致敏。
试验结果:激发后皮肤反应结果见表3,试验样品的浸提液在豚鼠皮肤未发现皮肤致敏反应,致敏阳性率为0%。阳性对照组的致敏阳性率为100%,见表4。

试验例3
手术导板细胞毒性试验:依据GB/T 16886.5-2017医疗器械生物学评价第5部分:体外细胞毒性试验,采用MTT法,评价样品潜在的皮细胞毒性。
试验与对照样品确定
试验样品:本发明树脂3D打印的手术导板。
对照样品
阴性对照样品:高密度聚乙烯,美国药典委员会;规格:3片装;批号:K0M357,白色固定,室温保存,浸提介质:含10%胎牛血清的MEM培养液。
阳性对照样品:名称:Zinc diethyldithiocarbamate,制造商:Sigma,规格:25g,批号:MKCB2943V,浓度1%,溶剂:含10%胎牛血清的MEM培养液,白色固体。
空白对照:名称:含10%胎牛血清的MEM培养液,粉红色液体,4±2℃保存。
试验设计
浸提液处理:试验样品灭菌:高压蒸汽灭菌121℃,30min。对照样品灭菌:与试验样品相同。
样品浸提:无菌条件整件取样,并按表5的浸提比例(样品:浸提介质)在密闭的惰性容器内振荡浸提,浸提介质为10%胎牛血清的MEM培养液,浸提完成后,记录浸提液状态和浸提介质的任何变化(提取前和提取后)。提取液立即用于测试。

浸提前后浸提液状态未发生改变,浸提液pH值未经调整,未经过滤,离心,稀释等处理。阳性浸提液经过滤后使用。
试验步骤
试验过程无菌操作:将L929细胞培养在10%胎牛血清和抗生素(青霉素100U/mL,链霉素100微克/mL)的MEM培养液中,置于37℃,5%CO2培养箱中培养,用0.25%胰酶(含EDTA)消化细胞制备成单细胞悬液,细胞悬液离心(200G,3min),然后将细胞重新分散于培养基中,调整细胞密度为1×105个/mL的细胞悬液。
接种上述细胞悬液到1个96孔培养板中,每孔100微升,置37℃培养箱中(培养箱环境:5%CO2,37℃,>90%湿度)培养24h。
待细胞长成单层后,吸出原来的培养液,分别加入100微升不同浓度的试验样品浸提液(100%、75%、50%、25%)、空白对照液、阳性对照(100%)和阴性对照液(100%),37℃,5%CO2培养24h。每组做5个平行样。
培养24h后,取出96孔培养板先做细胞形态学观察,然后吸出原来的培养液,每孔加50微升MTT(1mg/mL),置于37℃,5%CO2培养2h,吸弃上清,加100微升99.9%纯度的异丙醇溶液结晶。
在酶标仪上以570nm为主吸收波长,650nm为参考波长测定吸光度值。
试验结果:试验样品的100%浸提液组细胞活力值为93.5%,具体结果见表6和表7。

质量检查
阴性对照无细胞毒性影响,阳性对照造成细胞毒性影响。
在试验过程中,未经处理的空白对照的光密度绝对值OD570表明了每个孔接种的1×104个细胞以正常倍增时间成指数增长。
空白对照的OD570平均值不能小于0.2。
检查系统的细胞接种误差,96孔培养板的左侧(第2列)和右侧(第11列)作为空白对照(第1列和第12列不用作空白对照)。左右两侧空白对照的平均值不能与所有空白的平均值相差超过15%。
数据分析
使用SPSS 16.0计算各组测定的光密度值的均数±标准差(Mean±SD)。
50%浓度的样品浸提液与100%浓度的样品浸提液相比,细胞活力(%)应至少相同或更高,否则应重复试验。
细胞活力(%)越低,表明受试样品潜在的细胞毒性越大。
评价标准:50%的样品浸提液至少和100%的细胞活力相同或者比100%的细胞活力更高,否则应该重复试验;细胞活力(%)越低,潜在的细胞毒性越大;细胞活力<空白组70%,说明样品具有潜在的细胞毒性;100%试验样品浸提液的细胞活力(%)为最终结果。
在本试验条件下,试验样品的浸提液对L929细胞无潜在毒性影响。
Claims (6)
1.一种3D打印用低生物毒性光敏树脂,其特征在于,该树脂由以下重量份的组分组成:双酚A-甲基丙烯酸缩水甘油酯8-12份、脂肪族氨基甲酸二甲酯55-65份、三乙二醇二甲基丙烯酸酯25-35份、自由基类光引发剂2.5-3份、天然吸光剂0.1-0.2份,其中自由基类光引发剂的添加量不超过树脂总量的5%。
2.如权利要求1所述的一种3D打印用低生物毒性光敏树脂,其特征在于自由基类光引发剂为苯基双(2,4,6-三甲基苯甲酰基)氧化膦和2-羟基-2-甲基-1-苯基-1-丙酮的混合物,所述天然吸光剂为1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐。
3.如权利要求1所述的一种3D打印用低生物毒性光敏树脂,其特征在于,该树脂由以下重量份的组分组成:双酚A-甲基丙烯酸缩水甘油酯10份、脂肪族氨基甲酸二甲酯60份、三乙二醇二甲基丙烯酸酯30份、苯基双(2,4,6-三甲基苯甲酰基)氧化膦2.2份、2-羟基-2-甲基-1-苯基-1-丙酮0.5份、1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐0.16份。
4.如权利要求1-3任一所述的一种3D打印用低生物毒性光敏树脂的制备方法,其特征在于包括以下步骤:
1)用医用酒精清洗所有器具,干燥后备用;
2)将双酚A-甲基丙烯酸缩水甘油酯加热至80℃,称取所需质量于容器内,保温;
3)准确称量脂肪族氨基甲酸二甲酯、三乙二醇二甲基丙烯酸酯于步骤2)的容器内,80℃的温度条件下搅拌30分钟,制得混合物;
4)混合物静置,待温度下将至50℃时,准确称量苯基双(2,4,6-三甲基苯甲酰基)氧化膦、2-羟基-2-甲基-1-苯基-1-丙酮、1-(4-磺酸苯基)-4-(4-磺酸苯基偶氮)-5-吡唑啉酮-3-羧酸三钠盐于容器内,搅拌30分钟;
5)温度降至室温,完成整个生产过程,制得低生物毒性光敏树脂。
5.如权利要求1-3任一所述的一种3D打印用低生物毒性光敏树脂在3D打印中的应用。
6.如权利要求1-3任一所述的一种3D打印用低生物毒性光敏树脂作为3D打印材料的应用。
